Six 1,3-diphenylpropanes exhibiting inhibitory activities against both the monophenolase and diphenolase actions of tyrosinase were isolated from the methanol (95%) extract of Broussonetia kazinoki. These compounds, 16, were identified as kazinol C (1), D (2), F (3), broussonin C (4), kazinol S (5) and kazinol T (6). The latter two species (5 and 6) emerged to be new 1,3-diphenylpropanes which we fully spectroscopically characterized. The IC50 values of compounds (1, 35) for monophenolase inhibition were determined to range between 0.43 and 17.9 μM. Compounds 1 and 35 also inhibited diphenolase significantly with IC50 values of 22.8, 1.7, 0.57, and 26.9 μM, respectively. All four active tyrosinase inhibitors (1, 35) were competitive inhibitors. Interestigly they all mainfested simple reversible slow-binding inhibition against diphenolase. The most potent inhibitor, compound 4 diplayed the following kinetic parameters k3 = 0.0993 μM?1 min?1, k4 = 0.0048 min-1, and Kiapp = 0.0485 μM.  相似文献

Two new tirucallane triterpenoids, 21-methoxy-21,23-epoxy-tirucalla-7,24-dien-3α-ol (1) and 21-methoxy-21,23-epoxy-tirucalla-7,24-diene-1α,3α-diol (2), together with thirteen known compounds were isolated from the CH2Cl2 extract of the stem bark of Araliopsis synopsis. The structures of the compounds were determined by comprehensive analyses of their 1D and 2D NMR, mass spectral (EI and ESI) data and comparison with previously known analogs. Compounds 110 were tested against bacteria, fungi and plant pathogen oomycetes by the paper disk agar diffusion assay resulting in missing to low activities corresponding with MICs > 1 mg/mL. However, compounds 510 exhibited high cytotoxic activity against the human Caucasian prostate adenocarcinoma cell PC-3 line, with IC50 8.5–12.5 μM compared to the standard Doxorubicin with IC50 = 0.9 μM, while compounds 1, 3 and 4 showed low activity.  相似文献

Five new C15 eight-membered cyclic ethers (1, 3–6) with a characteristic terminal cis ene–yne moiety, along with the previously reported acetylenic chloro diol (2) were isolated from the organic extract of the red alga Laurencia glandulifera, collected at Crete island in South Greece. Full assignment of all 1H and 13C resonances were carried out by extensive analysis of their NMR spectra. All metabolites were tested for their antistaphylococcal activity and the minimum inhibitory concentrations (MICs) of 2–5 were in the range of 8–256 μg/ml.  相似文献

Phytochemical research on the dry flower of Carthamus tinctorius L. led to the isolation of two new epimeric tetrahydro-β-carboline alkaloids (1 and 2), together with three known analogues (35). Their planar structures were determined by comprehensive 1D, 2D-NMR, and HR-ESI–MS spectroscopic data analyses. The absolute configurations of alkaloids 1 and 2 were assigned by comparing their experimental electronic circular dichroism (ECD) curves with the calculated ECD data. To investigate their impact on melanogenesis, all of the alkaloids isolated were tested for their tyrosinase inhibitory activity and alkaloid 4 was found to inhibit tyrosinase with an IC50 value of 0.17 mM, compared with 0.18 mM for arbutin. The putative binding interactions between the compounds and tyrosine were studied by molecular docking to provide an explanation for their inhibitory activities, and the results obtained indicated that hydrophobic interactions played a very significant role in the combination.  相似文献

Five new phenolic compounds, designated candenatenins G–K (15), along with four known compounds, were isolated from the heartwood of Dalbergia candenatensis. The structures of these compounds were elucidated by HR-EI-MS, 1H and 13C NMR, COSY, HMQC, HMBC, and NOESY spectra. Compound 5 exhibited potent activity against DPPH radical scavenging with IC50 value of 25.7 μM, whereas compound 2 showed cytotoxicity against NCI-H187 cell line with IC50 value of 14.8 μM.  相似文献

Chemical investigation of the ethanol extract of the stalks and infructescence of Sibiraea leavigata led to the isolation of two new monoterpenes named (4R)-2-(2-hydroxy-4-methyl-3-pentenyl)furan-2(5H)-one (1) and (2R,4R)-2-(2-hydroxyethyl)-4-(2-methyl-1-propenyl)furan-5H-2-one (2) along with eight known phenylpropanoids (3–10). Their structures were established on the basis of the interpretation of spectroscopic data and electronic circular dichroism (ECD) calculations. In addition, all of these isolates were evaluated for their cytotoxic activity. The results showed that compound 3 displayed moderate cytotoxicity with IC50 values ranging from 10.8 to 49.2 μg mL−1 against five cell lines. While 1 showed selective promotion effects on proliferation of gastric cancer MGC803 and RSC96 cell lines.  相似文献

Bioassay-guided fractionation of an ethanol extract of a Madagascar collection of Elaeodendron alluaudianum led to the isolation of two new cardenolide glycosides (1 and 2). The 1H and 13C NMR spectra of both compounds were fully assigned using a combination of 2D NMR experiments, including 1H–1H COSY, HSQC, HMBC, and ROESY sequences. Both compounds 1 and 2 were tested against the A2780 human ovarian cancer cell line and the U937 human histiocytic lymphoma cell line assays, and showed significant antiproliferative activity with IC50 values of 0.12 and 0.07 μM against the A2780 human ovarian cancer cell line, and 0.15 and 0.08 μM against the U937 human histiocytic lymphoma cell line, respectively.  相似文献

Two new hydroxychavicol analogs nudibaccatumin A (1) and B (2), together with twenty known compounds were isolated from the methanol extract of Piper nudibaccatum. Their structures were elucidated by extensive spectroscopic analyses (1D and 2D NMR, HRESIMS, UV, IR and polarimetry). Hydroxychavicol is a known inhibitor of xanthine oxidase (XO). In the present study, hydroxychavicol and 5 natural analogs (15) were evaluated for their XO inhibitory activity. Neotaiwanensol B (3) (IC50 = 0.28 μM) showed a greater inhibitory effect than hydroxychavicol and allopurinol (the positive control). Two new compounds 1 and 2 showed a moderate inhibition activity with an IC50 value of 62.94 μM and 70.67 μM, respectively.  相似文献

Seven eremophilane-type sesquiterpenes (1–7), six cycloartane derivatives (813) and α-amyrin acetate (14) were isolated from the leaves of the far-eastern plant Ligularia alticola Worosch. (Family Asteraceae). (4S,5R,8S,10R)-8-Ethoxyeremophil-7(11)-en-12(8)-olide (1), 8α,11-epidioxy-8β-methoxyeremophil-6-ene (2) and 29-norcycloartan-3α-ol (8) have not been previously reported. Fukinone α-epoxide (3) was isolated for the first time from a natural source. The structures of all the compounds were established by the extensive analysis of their 1D and 2D NMR spectra and HR ESI mass spectrometry. The absolute stereochemistry of 1 was determined by comparison of theoretical and experimental ECD spectra with the application of B3LYP-TDDFT and B3LYP-GIAO calculations as well as by NMR spectroscopy. Compound 1 showed cytotoxic action against human cancer HL-60, Raji, and THP-1 cell lines (IC50 12.6, 6.0 and 6.9 μM, respectively). Compounds 2 and 4 demonstrated significant cytotoxic activities against HL-60 (IC50 2.8 and 5.8 μM, respectively) and Raji cells (IC50 2.9 and 4.2 μM, respectively). Compound 6 was cytotoxic against Raji cells (IC50 4.6 μM). None of tested compounds were cytotoxic against RAW 264.7 cells. Compounds 1 and 4–7 significantly decreased intracellular ROS levels, induced by endotoxic LPS from Escherichia coli in RAW 264.7 murine macrophages.  相似文献

Investigation of the n-BuOH extract of the whole plant of Beesia calthaefolia led to the isolation of three new cycloartane triterpenoids (1–3) and two known compounds (4, 5). Their structures were elucidated by 1D and 2D NMR, HRESIMS and optical rotation spectral data. All of the isolates were investigated for their inhibitory effects on the classical pathway of the complement system. Among them, compound 3 showed stronger inhibitory activity (IC50 148.0 μM) than the positive control (rosmarinic acid, IC50 181.8 μM), while other compounds (1, 2, 4 and 5) showed moderate activity. From the CH2Cl2/CH3OH (1:1) extract of the root bark of Millettia micans, a new pterocarpan, (6aR,11aR)-3-hydroxy-7,8,9-trimethoxypterocarpan (1), named micanspterocarpan, was isolated. Similar investigation of the CH2Cl2/CH3OH (1:1) extract of the root bark of Millettia dura gave a further new pterocarpan, (6aR,11aR)-8,9-methylenedioxy-3-prenyloxypterocarpan (2), named 3-O-prenylmaackiain, along with six known isoflavones (3-8) and a chalcone (9). All purified compounds were identified by NMR and MS, whereas the absolute configurations of the new pterocarpans were established by chriptical data analyses including quantum chemical ECD calculation. Among the isolated constituents, calopogonium isoflavone B (3) and isoerythrin A-4′-(3-methylbut-2-enyl) ether (4) showed marginal activities against the 3D7 and the Dd2 strains of Plasmodium falciparum (70–90% inhibition at 40 μM). Maximaisoflavone B (5) and 7,2′-dimethoxy-4′,5′-methylenedioxyisoflavone (7) were weakly cytotoxic (IC50 153.5 and 174.1 μM, respectively) against the MDA-MB-231 human breast cancer cell line. None of the tested compounds showed in-vitro translation inhibitory activity or toxicity against the HEK-293 human embryonic kidney cell line at 40 μM.  相似文献

One new α-pyrone derivative (S)-3-(4-methoxy-3-methyl-2-oxo-2H-pyran-6-yl)butyl acetate (1) and one new fatty acid derivative (4S,5S)-5-acetoxy-4-hydroxyhexanoic acid (2), along with six known compounds (3–8) were obtained from the mangrove-derived fungus Phomopsis sp. HYP11. Their structures and absolute configurations were determined by detailed 1D and 2D NMR, HR-ESI-MS spectral, and quantum chemical electronic circular dichroism (ECD) calculations. Compounds 2, 3, 5, 6 and 8 showed antioxidant activity with the IC50 values range of 0.09–0.67 mM, especially compounds 5 and 6 showed stronger antioxidant activity than the positive control trolox (IC50 = 0.29 mM). Compound 8 showed weak antagonistic effect on Ralstonia solanacearum with the MIC value of 50 μg/mL.  相似文献

A novel series of 2-(5-methyl-1,3-diphenyl-1H-pyrazol-4-yl)-5-phenyl-1,3,4-oxadiazoles 7(am) were synthesized either by cyclization of N′-benzoyl-5-methyl-1,3-diphenyl-1H-pyrazole-4-carbohydrazide 4a using POCl3 at 120 °C or by oxidative cyclization of hydrazones derived from various arylaldehyde and (E)-N′-benzylidene-5-methyl-1,3-diphenyl-1H-pyrazole-4-carbohydrazide 5(ad) using chloramine-T as oxidant. Newly synthesized compounds were characterized by analytical and spectral (IR, 1H NMR, 13C NMR and LC–MS) methods. The synthesized compounds were evaluated for their antimicrobial activity and were compared with standard drugs. The compounds demonstrated potent to weak antimicrobial activity. Among the synthesized compounds, compound 7m emerged as an effective antimicrobial agent, while compounds 7d, 7f, 7i and 7l showed good to moderate activity. The minimum inhibitory concentration of the compounds was in the range of 20–50 μg mL−1 against bacteria and 25–55 μg mL−1 against fungi. The title compounds represent a novel class of potent antimicrobial agents.  相似文献

Chemical investigations of the MeOH extract of air dried flowers of the Australian tree Angophora woodsiana (Myrtaceae) yielded two new β-triketones, woodsianones A and B (1, 2) and nine known β-triketones (311). Woodsianone A is a β-triketone-sesquiterpene adduct and woodsianone B is a β-triketone epoxide derivative. The structures of the new and known compounds were elucidated from the analysis of 1D/2D NMR and MS data. The relative configurations of the compounds were determined from analysis of 1H–1H coupling constants and ROESY correlations. All compounds (111) had antiplasmodial activity against the chloroquine sensitive strain 3D7. The known compound rhodomyrtone (5) and new compound woodsianone B (2) showed moderate antiplasmodial activities against the 3D7 strain (1.84 µM and 3.00 µM, respectively) and chloroquine resistant strain Dd2 (4.00 µM and 2.53 µM, respectively).  相似文献

A new eudesmane sesquiterpenoid (1), and a new homologue of virginiae butanolide E (2) along with butyl isobutyl phthalate (3) were isolated from, actinomycete-Lentzea violacea strain AS08 isolated from north western Himalayas by stressing on modified one strain-many compounds (OSMAC) method. The structures of the new compounds were elucidated by extensive spectroscopic analyses including 1D, 2D NMR along with HR-ESI-MS and FT-IR data. Herein, a distinctive method was added for inspecting secretory profile of the strain by quantification of extract value of cell free supernatant in different types of culture media fallowed by HPLC profiling of respective extracts, which revealed a highly altered metabolic profile of the strain and formed the base for the selection of media. The compounds 1 and 2 showed moderate activity against Gram negative (MIC ~32–64 µg ml?1) in comparison to Gram positive bacterial pathogens. Eleven prenylated C6–C3 compounds, illihenryifunones A, B (1, 2), illihenryifunol A (3), illihenryipyranol A (4), illihenryiones A−G (511), and three known prenylated C6–C3 compounds (1214), were isolated from the roots of Illicium henryi. Structures of 111 were elucidated by spectroscopic methods including NMR, HRESIMS, and CD. The absolute configuration of the 11,12-diol moiety in 5 was determined by observing its induced circular dichroism after addition of Mo2(OAc)4 in DMSO. The absolute configuration of C-11 in 4 was determined as S based on the Rh2(OCOCF3)4-induced CD data; the absolute configuration of 3 was determined as R by comparison of its experimental and calculated electronic circular dichroism (ECD). The antioxidant activities of compounds 114 were also evaluated. Compound 4 exhibited strong antioxidant activity with an IC50 value of 2.97 ± 1.30 μM, whereas compounds 3 and 8 showed antioxidant activities with IC50 values of 44.36 ± 0.30 and 48.00 ± 2.01 μM, respectively.  相似文献

Hepatocellular carcinoma (HCC) is the most common type of liver cancer, and treatment options for HCC are limited. In addition, the discovery of new natural compounds with anti-hepatocarcinoma activity is attracting increasing attention. For this reason, phytochemical investigation of Croton crassifolius led to the isolation of 17 diterpenoids, including three new clerodane diterpenoids, named crassifolius A-C (13), along with 14 known ones (417). Their structures were established by 1D, 2D NMR, HR-ESI-MS, detailed calculated electronic circular dichroism (ECD) spectra and the assistance of quantum chemical predictions (QCP) of 13C NMR chemical shifts. The cytotoxicities of all these compounds against human liver cancer lines (HepG2 and Hep3B) were determined. Among them, compound 1 exhibited good cytotoxicity with IC50 value of 17.91 μM against human liver tumor cells Hep3B. Following further studies of the anti-tumor mechanism of compound 1-induced cell growth inhibition, we found that compound 1 caused apoptotic cell death in Hep3B cells by detecting morphologic changes and Western blotting analysis.  相似文献

Two new 1,4-naphthoquinone derivatives, balsaminone D (1), balsaminone E (2) along with two known compounds (3 and 4) were discovered from Impatiens balsamina L. flowers. Their structures were identified with spectroscopic methods including HR–EI–MS, 1D and 2D NMR, as well as the absolute configuration was determined by ECD calculation. In addition, new compounds 1 and 2 with IC50 value of 30.54 and 40.67 μg/mL exhibited better activities against activated t-HSC/Cl-6 cells than positive control Silymarin and Fufang Biejia Ruangan Pian, of which the IC50 value were 202.34 and 231.56 μg/mL, respectively.  相似文献

Various chromatographic separations of the methanolic extract of Paramignya scandens stem and leaves resulted in the isolation of two new tirucallane derivatives, paramignyols A and B (1 and 2). Their structures were established by HR-ESI-MS, 1D and 2D NMR experiments, as well as, comparison with literature data. Compounds 1 and 2 showed significant cytotoxic activity (IC50s  3.55–10.50 μM) on four tested human cancer cell lines: KB (epidermoid carcinoma), SK-Mel-2 (melanoma), LU-1 (lung adenocarcinoma), and MCF7 (breast cancer).
