Development of perithecia from single, uninucleate ascospores disclosed a homothallic condition for Chaetomium erraticum. This species was found to produce sessile ascogonial coil initials from uninucleate vegetative cells that become enveloped by hyphae formed at the base of the ascogonium. The ascogonium consists of several cells that are uninucleate or binucleate. A perithecium forms from numerous divisions and enlargement of the surrounding uninucleate cells. Differentiation of the perithecial cells results in the formation of a carbonaceous wall, perithecial hairs, and an ostiole lined with periphyses. A convex hymenial cluster of ascogenous cells forms in the lower half of the centrum from which typical croziers develop. Asci push up into the pseudoparenchyma cells of the centrum. The growth of the ascogenous system is in part responsible for increase in perithecial size. The breakdown of the pseudoparenchyma cells around the developing asci results in the formation of a central cavity in which ascospores are released when the asci deliquesce. No paraphyses are present. The type of development and features of the centrum of C. erraticum and other species of Chaetomium indicate a distinct Xylaria-type centrum.  相似文献

Fungicidal and fungistatic effects of biological preparations involving bacteria of the genera Pseudomonas and Bacillusand fungi of the genusChaetomium on phytopathogenic fungi Didymella applanata and Botrytis cinereawere evaluated. All the biological preparations under study inhibited the growth of colonies of the fungi; however, the degree of the inhibition depended on the nature of each particular microorganism and the concentration of each particular preparation. A preparation containing Bacillus subtilis at a concentration of 0.2% effected maximum suppression ofB. cinerea (the diameter of the colonies decreased sevenfold). Preparations containing bacteria of the genus Pseudomonas and fungi of the genus Chaetomium were most efficient in suppressing D. applanata.Preparations containingB. subtilis and Chaetomium spp. showed promise as agents against simultaneous development of spur blight and Botrytis blight.  相似文献

We have previously demonstrated that blue light induces the phosphorylation of a 15-kDa protein in crude membrane fractions of Neurospora crassa mycelia. Here we report the isolation and characterization of a mutant ( psp; phosphorylation of small proteins) that is completely defective for phosphorylation of that protein, as assayed in both crude membrane and soluble fractions. This mutation defines a unique locus that maps to linkage group VR between al-3 and his-6. To elucidate the photobiological significance of the phosphorylation of the protein, we analyzed known photobiological phenomena and discovered that the positioning of beaks on the perithecia, defined as perithecial polarity, was light-dependent in the wild type. In the psp mutant, beaks were phototropic as in the wild type, but their position was random. In a wc-1 mutant, however, beaks were positioned at random and were not phototropic. Thus light-induced perithecial polarity and phototropism of perithecial beaks are controlled differently. A psp; wc-1 double mutant showed the same phenotype as that of wc-1 with respect to these two photomorphogenetic characters. These results indicate that the wc-1 gene is epistatic to psp in the light-signal transduction pathway that controls both phototropism and perithecial polarity. Interest inin vitro study of entomopathogenic fungi, includingCordyceps species, has been increasing due to their valuable bioactive compounds and biocontrol effects. AmongCordyceps species,in vitro stromata ofC. militaris has been successfully produced and cultivated for industrial purposes. However, genetic study onin vitro stromata formation ofC. militaris has not been carried out yet. Here, relationship between mating system and perithecial stromata formation ofC. militaris is reported. Mating system was determined by observing perithecial stromata formation from mono-ascospore cultures and their pair-wise combinations. Certain combinations of mono-ascospore strains produced perithecial club-shaped stromata, whereas other combinations produced either no stromata or only abnormal non-perithecial stromata. Similarly, monoascospore cultures without combination produced either no stromata or only abnormal nonperithecial stromata. Despite obvious heterothallism, self-fertility was occasionally observed in few strains ofC. militaris. These observations indicated thatC. militaris behaves as a bipolar heterothallic fungus and requires two mating compatible strains in order to produce regular clubshaped perithecial stromata, a fundamental requirement for its industrial cultivation.  相似文献

Summary This is the first record of the perithecial state ofSphaerotheca fuliginea onHelianthus annuus L. from India. The perithecial state is collected in the dry cool weather but is absent in wet monsoon season due to the mildew being parasitizing by the hyperparasite,Ampelomyces sp., which thus, appears to influence the development of the perfect state in this mildew.  相似文献

Laboulbenia coneglanensis isolated from Japanese harpaline carabids is described and photographed. It is compared morphologically withL. flagellata from Japan and Europe, which is similar in appearance. The most important difference between the two species is in antheridial character and host range. Other characters such as coloration, perithecial shape, especially the shape of the perithecial apex, and size of ascospores are also useful for distinction.Laboulbenia ophoni var.dilatata described by Maire (1920) is regarded as a synonym ofL. coneglanensis.  相似文献

Laboratory experiments using sterilized apple-leaf discs showed that treatment of leaves with urea during the early stages of perithecial initiation induced a high nitrogen content of the leaves, which prevented further development of perithecia although mycelial growth was unaffected. Treatments applied at a later stage of fungal development inhibited both perithecial and mycelial growth. Some of the bacteria isolated from urea-treated leaves in the field restricted perithecial development, particularly when applied in the first month after inoculation with suspensions of conidia from sexually compatible strains of the fungus. One isolate, a Pseudomonas sp., was shown to be important in the decomposition of apple leaves.  相似文献

An analysis of character variation in species of the Porina rufula aggregate, i.e. P. rufula (Kremp.) Vain., P. limbulata (Kremp.) Vain., P. rubentior (Stirt.) Müll. Arg., and P. pseudofulvella Sérus., showed that the hitherto applied concepts of species delimitation - based on perithecial size and colour - are not appropriate and should be replaced by a concept which primarily regards perithecial colour and shape as specific characters. Revision of type specimens proved the identity of P. rufula and P. pseudofulvella, whereas P. limbulata sensu Santesson had to be split into P. limbulata s. str. and P. leptospermoides Müll. Arg. Evolutionary tendencies are seen in reduction of thallus morphology and perithecial pigments as an adaptation to shady habitats, indicating P. leptospermoides as a more primitive and P. rufula as a more advanced taxon. Judging from species distribution, the phylogenetic origin of the P. rufula aggregate might be in the Neotropics.  相似文献

The biotransformations of (R)-(?)-methyloctalone and (S)-(+)-methyloctalone were investigated using Chaetomium sp.1 KCH 6651 and Didymosphaeria igniaria KCH 6670 as biocatalysts, yielding mostly 6β- and 7β-hydroxy derivatives. During the incubation of (R)-methyloctalone with the Chaetomium sp.1 culture, three products were obtained: the trans-7β-hydroxy derivative in 50% yield, trans-6β-hydroxy derivative in 30% yield and cis-8α-hydroxy derivative in 6% yield. The (S)-enone was hydroxylated mainly in the 6α-position (with 60% yield). 6β- and 7β-hydroxy derivatives are new compounds, not previously described in the literature. The biotransformation of the two enantiomers of methyloctalone with D. igniaria also afforded monohydroxy derivatives. In both cases the main product of transformation was the allylic hydroxy derivative (27–35% yield). D. igniaria transformed the (R)-methyloctalone more rapidly whereas Chaetomium sp. 1 preferred the (S)-methyloctalone.  相似文献

A new ascomycete species,Plagiostoma phragmiticola inhabiting two reed plants,Phragmites australis andP. karka, is described and illustrated. This new species is characterized by having non-stromatic, solitary perithecia, laterally and obliquely protruding perithecial beaks, and relatively small asci and ascospores. Black zone-lines in colonized leaf tissues were not observed. Its anamorph belongs toPhomopsis.  相似文献

Fourteen species of keratinophilic fungi belonging to ten genera (Chrysoporium, Malbranchea, Chaetomium,Sepedonium, Microascus, Scopulariopsis, Curvularia, Fusarium, Aspergillus, Penicillium) were isolated from feathers of about one hundred living poultry birds. The isolated fungi were compared for their keratinase activity after growing them on two different media: (1) basal salts solution containing natural keratin (human hair) as the only source of carbon and nitrogen; (2) the medium was supplemented with a minor amount of readily assimilable source of carbon along with natural keratin. All the test fungi could grow on keratinous material, degrading it and releasing sulphydryl containing compounds detected as cysteine, total proteins and extracellular keratinase. Maximum enzyme release by these fungi occurred in the broth supplemented with glucose and vitamins, thereby indicating a correlation between the mycelial biomass and production of proteolytic keratinases. Chaetomium globosum, the most common species within this genus, produces chaetoglobosins A and C when cultured on building material. Relatively low levels of these compounds have been shown to be lethal to various tissue culture cell lines. This study had two major objectives: (1) to determine the frequency at which Chaetomium species are isolated in water-damaged buildings and (2) to examine the production of chaetoglobosins A and C in isolates of C. globosum obtained from different buildings. Out of 794 water-damaged buildings, Chaetomium species were isolated in 49% of these structures. C. globosum ATCC 16021 was grown on four different media: oatmeal agar (OA), potato dextrose agar (PDA), corn meal agar (CMA), and malt extract agar (MEA). After 4 weeks, fungal growth was evaluated based on colony diameter and the quantity of spores produced on agar plates. In addition, production of chaetoglobosin A and C was monitored using high performance liquid chromatography. Colony diameter, spore production, and mycotoxin production by C. globosum were the highest on OA. Out of 30 C. globosum isolates cultured on OA for 4 weeks, 16 produced detectable amounts of chaetoglobosin A and every isolate produced chaetoglobosin C.  相似文献

Chaetomium is a fungus species that inhabits different cellulose substrates. Some species of this genus destroy books, textile, and wood. This genus is of particular interest due to its antagonistic and enzyme peculiarities. Studies on morphological and cultural features of Chaetomium species have both theoretical and practical output. Growth rates and development of C. globosum, C. funicola, C. elatum, and C. spirale were studied under different temperatures (17–20°C, 25°C, 27°C, 30°C, and 33–35°C) and carbon source media (glucose, saccharose, mannite, lactose, amylum, and cellulose). The optimal growth temperature was 25–27°C for all the studied species, while the temperature range of 33–35°C inhibited the colonies’ growth. Growth dynamics and colony shape and morphology, as well as development of overhead mycelium and ascocarps, varied greatly in different Chaetomium species in regard to the carbon source media. When comparing the destructive effect of four studied species and C. murorum on craft paper, the highest activity was registered for C. globosum, C. funicola, and C. elatum.  相似文献

从蛇足石杉内生菌的次级代谢产物中寻找活性成分,为进一步开发利用蛇足石杉药用植物资源提供了新途径,但至今其内生菌代谢产物的系统性研究较为少见。种类丰富的内生真菌普遍存在于各种植物中,但蕨类植物中内生真菌的研究较少。为了寻找蛇足石杉内生菌中的细胞毒活性成分,该研究从蛇足石杉根部分离得到一株球毛壳属(Chaetomium sp.)真菌M336,对其化学成分进行了研究。对蛇足石杉内生真菌M336采用PDA固体培养基扩大发酵,发酵物经提取及乙酸乙酯萃取后,通过正相硅胶柱色谱法、Sephadex LH-20凝胶柱色谱法、薄层制备、高效液相色谱等色谱手段对其发酵物中的化学成分进行分离纯化,利用理化性质、质谱、核磁等波谱分析技术,并结合相关文献数据鉴定化合物的结构。结果表明:从内生真菌M336发酵提取物的乙酸乙酯萃取部分分离并鉴定出8个化合物,分别为chaetoviridines F、chaetoviridines E、5′-epichaetoviridin A、5′-epichaetoviridin A、xanthoquinodins Al、xanthoquinodins A2、xanthoquinodins B1和毛壳菌素。从M336中分离得到8个化合物,化合物3有一定的抑菌作用,其余化合物有一定的细胞毒活性。该研究结果丰富了蛇足石杉内生真菌球毛壳属中的天然细胞毒活性的化合物。  相似文献

Applications of urea after harvest but before leaf-fall restricted perithecial production by Venturia inaequalis. Immersion of detached leaves in urea appeared to be the most effective method of preventing perithecial formation, although spraying attached leaves was equally effective when leaf abscission occurred within a week of treatment. A high nitrogen content within the leaf was one of the major factors contributing to suppression. Urea-treated leaves decomposed rapidly, thus destroying the overwintering substrate for the fungus. When apple plants (clone M. 111) were sprayed in autumn with 5 % urea, followed by a second (pre-bud-burst) application at 2 %, ascospore production in the spring was suppressed. The second treatment appeared to prevent the release of ascospores from mature perithecia.  相似文献

Endophytic fungi are broadly dispersed residing inside plant tissues and have been demonstrated as a treasure for bioactive natural products. Unexplored harsh and heavy metal contaminant habitat of Avicennia marina may have diverse and potential fungal association. Therefore, this work aimed to isolate the culturable fungal endophytes associated with leaves of A. marina and to evaluate their medical potentialities. Seventeen isolates of endophyte fungi were isolated from healthy leaves and their antimicrobial activities were evaluated. Results showed that isolates had activity against micro-organisms in addition to their antioxidant activity produced a variety of phenolic compounds, besides exhibited a lowest cytotoxicity against ATCC-CCL-81 cell line. Consequently, selected endophytic fungal isolates were identified genetically as Chaetomium sp., Chaetomium madrasense, Chaetomium sp., Chaetomium globosum, Aspergillus hiratsukae, Aspergillus ochraceus, Alternaria tenuissima and Curvularia lunata with gene bank accession numbers MT089951, MT089952, MT089953, MT089954, MT089955, MT089956, MT089957 and MT089958 respectively. The most potent fungus extract was analysed using Gas chromatography–mass spectrometry which verified the presence of numerous bioactive compounds. These findings confirmed that new endophytic fungal strains derived from A. marina thrive in harsh ecosystem produce bioactive metabolites which can be recommended as a novel source for drug discovery.  相似文献

Experiments conducted indicate the mating system in Epichloë typhina to involve simple bipolar heterothallism. Perithecia were formed when condia were transferred to a compatible stroma. Perithecial development followed the growth of mycelium over the surface of the stroma. The perithecial stroma consisted of two layers: an inner layer composed of the original mycelium of the conidial stroma and an outer layer that appears to be composed of mycelium growing from the inner stromal layer. Perithecia were initiated in the outer layer, just above the interface of the two layers. Natural onset of perithecial formation was associated with evidence of visitation by a prasitic fly (Phorbia phrenione).  相似文献
