Genetic variation was assessed in a range of populations of Amaranthus retroflexus using isoenzyme analysis. Population genetic diversity was measured by evaluating patterns of variation at six putatively neutral isoenzyme loci (comprising 24 putative alleles) within and among 20 populations of A. retroflexus collected in different habitats: ruderal habitats, cereal fields and hop gardens. Amaranthus retroflexus is a noxious weed of North American origin that infests various crops. Overall, A. retroflexus displayed moderate levels of genetic diversity in comparison with other herbaceous plants. The percentage of polymorphic loci was 50.0%, with mean values of 2.01, 0.142 and 0.227 for the average number of alleles per polymorphic locus ( A), observed heterozygosity ( Ho) and expected heterozygosity ( He), respectively. A discrepancy between observed and expected heterozygosity and significant differences from H-W expectation indicate that there is an excess of homozygotes in many populations. As a result, there is strong evidence of inbreeding within populations ( FIS = 0.382) and significant population differentiation ( FST = 0.270). Even though the species is partly autogamous, inbreeding does not lead to strong inbreeding depression resulting from self-pollination, as inbreeding has no effect on the success of the species in today's countryside. Moreover, allele frequencies detected in agricultural habitats (i.e., cereal fields and hop gardens) differed from those detected in populations collected from ruderal habitats, which is probably caused by systematic application of herbicides in agricultural ecosystems. 相似文献

Species introductions often coincide with loss of genetic diversity and natural enemies. Anthropogenic translocation of the North-American pumpkinseed Lepomis gibbosus (L., 1758) (Centrarchidae) and its further spread have resulted in recent species establishment in most European countries. This study determines genetic differentiation of non-native European pumpkinseed populations and identifies how their genetic structure relates to the distribution and abundance of parasite species. Microsatellite analysis indicated presence of three genetic lineages, which were well supported by discriminant analysis based on parasite abundance data. The first lineage clustered pumpkinseed populations from northern and southern France and showed high allelic richness, heterozygosity and parasite richness. The second included populations along the “Southern invasion corridor” connecting the rivers Rhine, Main and Danube. The fish exhibited low to high genetic and parasite diversity and generally high parasite abundance. The third lineage clustered populations with low genetic and parasite diversity, located in Portuguese reservoirs and water bodies along the upper Elbe. Parasite species richness was significantly associated with host microsatellite heterozygosity and allelic richness, a trend partially affected by richness of North-American parasites. Furthermore, our results indicate that parasite community composition may serve as a useful biological tool to discriminate non-native fish populations and their inter-relationships. 相似文献

Human polymorphonuclear neutrophil (PMN) responses to G protein-coupled chemoattractants are highly dependent upon store-operated Ca(2+) entry (SOCE). Recent research suggests that SOCE currents can be mediated by a variety of related channel proteins of the transient receptor potential superfamily. SOCE has been regarded as a specific response to depletion of cell calcium stores. We hypothesized that net SOCE might reflect the contributions of more than one calcium entry pathway. SOCE was studied in normal human PMN using Ca(2+) and Sr(2+) ions. We found that PMN SOCE depends on at least two divalent cation influx pathways. One of these was nonspecific and Sr(2+) permeable; the other was Ca(2+) specific. The two pathways show different degrees of dependence on store depletion by thapsigargin and ionomycin, and differential sensitivity to inhibition by 2-aminoethyoxydiphenyl borane and gadolinium. The inflammatory G protein-coupled chemoattractants fMLP, platelet-activating factor, and IL-8 elicit unique patterns of Sr(2+) and Ca(2+) influx channel activation, and SOCE responses to these agonists displayed differing degrees of linkage to prior Ca(2+) store depletion. The mechanisms of PMN SOCE responses to G protein-coupled chemoattractants are physiologically diverse. They appear to reflect Ca(2+) transport through a variety of channels that are independently regulated to varying degrees by store depletion and by G protein-coupled receptor activation. 相似文献

Population genetic theory predicts that the self-incompatible and perennial herb, Arabidopsis lyrata, will have a genetic structure that differs from the self-fertilizing, annual Arabidopsis thaliana. We quantified the genetic structure for eight populations of A. lyrata ssp. petraea in historically nonglaciated regions of central Europe. Analysis of 20 microsatellite loci for 344 individuals demonstrated that, in accordance with predictions, diploid populations had high genome-wide heterozygosity (H(O) = 0.48; H(E) = 0.52), high within-population diversity (83% of total) compatible with mutation-drift equilibrium, and moderate differentiation among populations (F(ST) = 0.17). Within a single population, the vast majority of genetic variability (92%) was found at the smallest spatial scale (< 3 m). Although there was no evidence of biparental inbreeding or clonal propagation at this scale (F(IS) = 0.003), significant fine-scale spatial autocorrelation indicated localized gene flow presumably due to gravity dispersed seeds (Sp = 0.018). Limited gene flow between isolated population clusters (regions) separated by hundreds of kilometres has given rise to an isolation by distance pattern of diversification, with low, but significant, differentiation among regions (F(ST) = 0.05). The maintenance of geographically widespread polymorphisms and uniformly high diversity throughout central Europe is consistent with periglacial survival of A. lyrata ssp. petraea north of the Alps in steppe-tundra habitats during the last glacial maximum. As expected of northern and previously glaciated localities, A. lyrata in Iceland was genetically less diverse and highly differentiated from central Europe (H(E) = 0.37; F(ST) = 0.27). 相似文献

Summary The investigations on the structure of TBE foci were carried out in the localities Jívová (northern Moravia), Topolianky and írany (Tribe Mountains), and environs of Debreczén (Pannonian lowland, Hungary). The biphasic activity curve of Ixodes ricinus, characteristic for the central European conditions was confirmed in all three types.In some localities two or three tick species as vectors come into consideration and the unequal seasonal occurrence of individual species results in a permanent presence of virus-harbouring ticks in nature throughout the year.The average population density of ticks per hectare in a focus near topolianky in mid-April, 1969, reached 46,000 nymphae and 50,000 adults. About 1% of the tick population were found to be viruliferous.Great differences were found in the composition of plant communities, mammalian synusies and in the population density of small mammals and ticks within the regions studied.Each elementary focus consists of many microfoci where the territories of individual host species overlap in space and time. Nests in tree holes and underground probably also belong to these microfoci. The maintenance hosts and indicators in each type of focus are mentioned. Free-living Carnivora can be considered as incidental hosts in each type of focus. 相似文献

Deciphering the colonization processes by which introduced pests invade new areas isessential to limit the risk of further expansion and/or multiple introductions.We here studied the invasion history of the maritime pine bast scale Matsucoccusfeytaudi. This host-specific insect does not cause any damage in its nativearea, but it devastated maritime pine forests of South-Eastern France where it wasdetected in the 1960s, and since then reached Italy and Corsica. We used populationgenetic approaches to infer the populations'' recent evolutionary history frommicrosatellite markers and Approximate Bayesian Computation. Consistent with previousmitochondrial data, we showed that the native range is geographically stronglystructured, which is probably due to the patchy distribution of the obligate host andthe limited dispersal capacity of the scale. Our results show that the invasionhistory can be described in three successive steps involving different colonizationand dispersal processes. During the mid-XXth century, massive introductions occurredfrom the Landes planted forest to South-Eastern France, probably due totransportation of infested wood material after World War II. Stepping-stoneexpansion, consistent with natural dispersal, then allowed M. feytaudi toreach the maritime pine forests of Liguria and Tuscany in Italy. The island ofCorsica was accidentally colonized in the 1990s, and the most plausible scenarioinvolves the introduction of a limited number of migrants from the forests ofSouth-Eastern France and Liguria, which is consistent with an aerial dispersal due tothe dominant winds that blow in spring in this region. 相似文献

Understanding the genetic structure of the European population is important, not only from a historical perspective, but also for the appropriate design and interpretation of genetic epidemiological studies. Previous population genetic analyses with autosomal markers in Europe either had a wide geographic but narrow genomic coverage [1] and [2], or vice versa [3], [4], [5] and [6]. We therefore investigated Affymetrix GeneChip 500K genotype data from 2,514 individuals belonging to 23 different subpopulations, widely spread over Europe. Although we found only a low level of genetic differentiation between subpopulations, the existing differences were characterized by a strong continent-wide correlation between geographic and genetic distance. Furthermore, mean heterozygosity was larger, and mean linkage disequilibrium smaller, in southern as compared to northern Europe. Both parameters clearly showed a clinal distribution that provided evidence for a spatial continuity of genetic diversity in Europe. Our comprehensive genetic data are thus compatible with expectations based upon European population history, including the hypotheses of a south-north expansion and/or a larger effective population size in southern than in northern Europe. By including the widely used CEPH from Utah (CEU) samples into our analysis, we could show that these individuals represent northern and western Europeans reasonably well, thereby confirming their assumed regional ancestry. 相似文献

Given the general pattern of invasions with severe ecological consequences commonly resulting from multiple introductions of large numbers of individuals on the intercontinental scale, we explored an example of a highly successful, ecologically significant invader introduced over a short distance, possibly via minimal propagule pressure. The Sacramento pikeminnow ( Ptychocheilus grandis) has been introduced to two coastal rivers in northern California where it poses a risk to threatened and endangered fishes. We assayed variation in seven microsatellite loci and one mitochondrial DNA gene to identify the source populations and estimate founder numbers for these introductions. Our analysis suggests that successful invasion of the Eel River was likely the result of a single transfer of 3–4 effective founders from nearby within the species’ native range: Clear Lake or its outflow Cache Creek. The other introduced population (Elk River), known from only seven individuals, likely represents secondary expansion from the introduced Eel River population. Our findings highlight the threat posed by close-range invaders and the ability of some fishes to rapidly invade ecologically suitable areas despite small effective founding numbers. 相似文献

The wasp Vespula germanica is a highly successful invasive pest. This study examined the population genetic structure of V. germanica in its introduced range in Australia. We sampled 1320 workers and 376 males from 141 nests obtained from three widely separated geographical areas on the Australian mainland and one on the island of Tasmania. The genotypes of all wasps were assayed at three polymorphic DNA microsatellite markers. Our analyses uncovered significant allelic differentiation among all four V. germanica populations. Pairwise estimates of genetic divergence between populations agreed with the results of a model-based clustering algorithm which indicated that the Tasmanian population was particularly distinct from the other populations. Within-population analyses revealed that genetic similarity declined with spatial distance, indicating that wasps from nests separated by more than approximately 25 km belonged to separate mating pools. We suggest that the observed genetic patterns resulted from frequent bottlenecks experienced by the V. germanica populations during their colonization of Australia. 相似文献

The study focuses on geographical patterns of genetic variation at allozyme loci common for four main tree species of Central Europe (Norway spruce, silver fir, common beech and sessile oak). Moving-window averaging of four indicators of allelic richness and diversity (proportion of polymorphic loci, mean number of alleles per locus, effective number of alleles and expected heterozygosity) with window size of 50 × 50 km was used to identify the patterns. Moreover, local genetic divergence was assessed using the G ST (Nei, Molecular population genetic and evolution, Amsterdam and Oxford, North-Holland, 1975) and D j (Gregorius and Roberds, Theor Appl Genet 71:826–834, 1986) statistics for common beech and silver fir, where raw genotype data were available. Spatial patterns of diversity and allelic richness were quite similar. Romanian Carpathians were identified as the most important hotspot of genetic diversity and evolutionary divergence in Central Europe. Anthropogenic habitat fragmentation — ubiquitous in modern ecosystems — has strong impacts on gene flow and genetic population structure. Reptiles may be particularly susceptible to the effects of fragmentation because of their extreme sensitivity to environmental conditions and limited dispersal. We investigate fine-scale spatial genetic structure, individual relatedness, and sex-biased dispersal in a large population of a long-lived reptile (tuatara, Sphenodon punctatus) on a recently fragmented island. We genotyped individuals from remnant forest, regenerating forest, and grassland pasture sites at seven microsatellite loci and found significant genetic structuring ( RST = 0.012) across small distances (< 500 m). Isolation by distance was not evident, but rather, genetic distance was weakly correlated with habitat similarity. Only individuals in forest fragments were correctly assignable to their site of origin, and individual pairwise relatedness in one fragment was significantly higher than expected. We did not detect sex-biased dispersal, but natural dispersal patterns may be confounded by fragmentation. Assignment tests showed that reforestation appears to have provided refuges for tuatara from disturbed areas. Our results suggest that fine-scale genetic structuring is driven by recent habitat modification and compounded by the sedentary lifestyle of these long-lived reptiles. Extreme longevity, large population size, simple social structure and random dispersal are not strong enough to counteract the genetic structure caused by a sedentary lifestyle. We suspect that fine-scale spatial genetic structuring could occur in any sedentary species with limited dispersal, making them more susceptible to the effects of fragmentation. 相似文献

SUMMARY 1. Biological invasions often involve close taxonomic relatives either as native/invader pairs or as invader/invader pairs. Precise identification and differentiation of species is therefore of paramount importance to reconstruct the invasion history. Genetic studies are indispensable in the case of morphologically conservative taxonomic groups. 2. We analysed the Pontocaspian freshwater amphipods Dikerogammarus that have successfully invaded the benthos of large Central European rivers. Taxonomic uncertainties were clarified by phylogenetic analyses of mitochondrial 16S and COI genes. The three-way partitioning of allozyme genotypes in a syntopic population further corroborated the taxonomic status of the three species Dikerogammarus haemobaphes , D. villosus and D. bispinosus . Dikerogammarus bispinosus had been prior misidentified as a subspecies of D. villosus . The conspicuous colour types of D. villosus , however, appeared to be conspecific. 3. The genetic identification of the previously more abundant D. haemobaphes individuals in old samples supported the 'successive invasion wave' hypothesis with D. haemobaphes as the first invader displaced by the second invader D. villosus . Dikerogammarus bispinosus could be a potential future invader. 4. Haplotype differentiation was apparent between two invasion lines of D. haemobaphes , but the occurrence of a single widespread haplotype indicates genetic impoverishment during rapid colonisation. 相似文献

The house sparrow is one of the most widely introduced vertebrate species around the world, making it an important model species for the study of invasion ecology. Population genetic studies of these invasions provide important insights into colonisation processes and adaptive responses occurring during invasion. Here we use microsatellite data to infer the population structure and invasion history of the introduced house sparrow ( Passer domesticus) in Australia and New Zealand. Our results identify stronger population structure within Australia in comparison to New Zealand and patterns are consistent with historical records of multiple introduction sites across both countries. Within the five population clusters identified in Australia, we find declines in genetic diversity as we move away from the reported introduction site within each cluster. This pattern is consistent with sequential founder events. Interestingly, an even stronger decline in genetic diversity is seen across Australia as we move away from the Melbourne introduction site; secondary historical reports suggest this site imported a large number of sparrows and was possible the source of a single range expansion across Australia. However, private allele numbers are highest in the north, away from Melbourne, which could be a result of drift increasing the frequency of rare alleles in areas of smaller population size or due to an independent introduction that seeded or augmented the northern population. This study highlights the difficulties of elucidating population dynamics in introduced species with complex introduction histories and suggests that a combination of historical and genetic data can be useful. 相似文献

Maize (Zea mays L.) is one of the most important cereal crops and a model for the study of genetics, evolution, and domestication. To better understand maize genome organization and to build a framework for genome sequencing, we constructed a sequence-ready fingerprinted contig-based physical map that covers 93.5% of the genome, of which 86.1% is aligned to the genetic map. The fingerprinted contig map contains 25,908 genic markers that enabled us to align nearly 73% of the anchored maize genome to the rice genome. The distribution pattern of expressed sequence tags correlates to that of recombination. In collinear regions, 1 kb in rice corresponds to an average of 3.2 kb in maize, yet maize has a 6-fold genome size expansion. This can be explained by the fact that most rice regions correspond to two regions in maize as a result of its recent polyploid origin. Inversions account for the majority of chromosome structural variations during subsequent maize diploidization. We also find clear evidence of ancient genome duplication predating the divergence of the progenitors of maize and rice. Reconstructing the paleoethnobotany of the maize genome indicates that the progenitors of modern maize contained ten chromosomes. 相似文献

We used 30 genetic markers of 6 different classes to describe hierarchical genetic structure in introduced populations of the fire ant Solenopsis invicta. These included four classes of presumably neutral nuclear loci (allozymes, codominant random amplified polymorphic DNAs (RAPDs), microsatellites, and dominant RAPDs), a class comprising two linked protein-coding nuclear loci under selection, and a marker of the mitochondrial DNA (mtDNA). Patterns of structure revealed by F statistics and exact tests of differentiation were highly concordant among the four classes of neutral nuclear markers, although the microsatellites were the most effective markers for detecting structure. The results from the mtDNA complemented those from the neutral nuclear markers by revealing that strong limitations to female-mediated gene flow were the cause of the local structure registered by the nuclear markers. The pattern of structure inferred from the selected nuclear loci was markedly different from the patterns derived from the other sets of markers but was predictable on the basis of the presumed mode of selection acting on these loci. In general, the results for all six classes of markers can be explained by known features of the social and reproductive biology of fire ants. Thus, the results from these diverse sets of markers, combined with detailed natural history data, provide an unusually complete picture of how the fundamental evolutionary forces of gene flow, drift, and selection govern the distribution of genetic variation within and between fire ant populations. 相似文献

Phylogeographic analyses can help to reveal the refugial structure of plants during and after the ice ages, but the detailed history of regional refugial isolation and differentiation in Central Europe is still poorly understood. A recent study of Meum athamanticum in its total range of occurrences revealed persistence of this temperate montane plant species in Central Europe north of the Alps, without going into details. We therefore aimed to study differentiation and migration processes of this plant species in more detail throughout Central Europe. We used high resolution amplified fragment length polymorphisms (AFLP) markers and analyzed 210 plant individuals of 14 Central European populations with three pairs of primer combinations (128 loci, 111 of which polymorphic). The data show genetic differentiation and varying levels of molecular diversity within populations and groups of populations. Altogether, the studied populations did not show a gradient in molecular variation along presumptive postglacial migration routes across Central Europe. Rather, they reveal a genetic division into seven major groups. Four of them are characterised by high genetic diversity, private fragments and higher than average number of rare and sparse fragments, leading to the assumption that they are descendants of independent populations which survived in glacial refugia. In combination with information from paleoclimate and paleovegetation, it is likely that microclimatically favoured habitats at (i) the eastern flank of the Black Forest, (ii) the southern margin of the Cologne basin, (iii) the foothills of the Erzgebirge, and (iv) the foothills of the Jura Mountains acted as sources for the postglacial recolonisation of this species to the other mountains of Central Europe. As some of the populations analysed show intermixed gene-pools (i.e. including genetic information from different groups) and partly have exceptionally high genetic diversity, but no private and only relatively few rare or sparse fragments, they might represent contact zones. On the other hand, genetic pauperization and isolation of two other populations in connection with extremely small population sizes and unfavourable habitat conditions seem to reflect recent bottlenecks. Consequently, the genetic structure of M. athamanticum in Central Europe is shaped by (i) extra-Mediterranean glacial refugia in situ, (ii) following postglacial hybridization along emerging contact zones and (iii) genetic bottlenecks in thereafter isolated small populations. All results provide evidences for small scale migration of the species between Central European valleys and surrounding highlands. Therefore, our study provides molecular evidence for both climate dependent wide ranging periglacial tabula rasa, but some small refugia in locally buffered areas. We hereby show that the environmental heterogeneity of cold stage landscapes in Central Europe is generally underestimated. 相似文献

BackgroundUnderstanding the dimensions of pathways of introduction of alien plants is important for regulating species invasions, but how particular pathways differ in terms of post-invasion success of species they deliver has never been rigorously tested. We asked whether invasion status, distribution and habitat range of 1,007 alien plant species introduced after 1500 A.D. to the Czech Republic differ among four basic pathways of introduction recognized for plants. Principal FindingsPathways introducing alien species deliberately as commodities (direct release into the wild; escape from cultivation) result in easier naturalization and invasion than pathways of unintentional introduction (contaminant of a commodity; stowaway arriving without association with it). The proportion of naturalized and invasive species among all introductions delivered by a particular pathway decreases with a decreasing level of direct assistance from humans associated with that pathway, from release and escape to contaminant and stowaway. However, those species that are introduced via unintentional pathways and become invasive are as widely distributed as deliberately introduced species, and those introduced as contaminants invade an even wider range of seminatural habitats. ConclusionsPathways associated with deliberate species introductions with commodities and pathways whereby species are unintentionally introduced are contrasting modes of introductions in terms of invasion success. However, various measures of the outcome of the invasion process, in terms of species'' invasion success, need to be considered to accurately evaluate the role of and threat imposed by individual pathways. By employing various measures we show that invasions by unintentionally introduced plant species need to be considered by management as seriously as those introduced by horticulture, because they invade a wide range of seminatural habitats, hence representing even a greater threat to natural areas. 相似文献

Summary Oranizational forms and the current status of genetic counseling within the health care system of 15 European countries were evaluated by questionnaire and at a symposium, with individuals present from Austria, Belgium, Czechoslovakia, Denmark, the Federal Republic of Germany, the German Democratic Republic, Finland, France, Hungary, Italy, the Netherlands, Norway, Switzerland, the United Kingdom, and the Soviet Union. In spite of wide differences between these countries, certain similarities with respect to the delivery of genetic counseling services could be observed: (i) most genetic counseling is done within university institutions or closely linked to it; (ii) governmental support of genetic counseling is developing slowly, and genetic counseling is usually not yet fully integrated into the health care system; (iii) there is lack of qualified personnel; (iv) no guide lines for formal education have been developed, but a postgraduate training period of no less than four years is considered a minimum; (v) without appropriate support, genetic counseling is a burden for research in human genetics; yet, a strict separation of genetic counseling and research activities is not recommended; (vi) on the average, a team providing genetic counseling for about 1–2 million people should consist of 3–4 physicians, 5–10 technicians, 2–3 secretaries, and other supportive personnel.This study was supported in part by the Deutsche Forschungsgeneinschaft 相似文献
