A variety of obstacles have hindered the ultrastructural localization of hyaluronan (HA). These include a lack of adequate fixation techniques to prevent the loss of HA, the lack of highly sensitive and specific probes, and a lack of accessibility due to the masking of HA by HA-binding macromolecules such as proteoglycans and glycoproteins. Despite these problems, a number of studies, both biochemical and histochemical, have been published indicating that HA is not restricted to the extracellular milieu, but is also present intracellularly. This review focuses on the possible functions of intracellular HA, its potential relationships to extracellular HA structures, and implications for inflammatory processes.  相似文献

At the beginning of the 21st century, biology will try to address the function of a large number of new genes. From the perspective of technologies applied today to functional genomics, this task appears to be more complex than the effort invested in the sequencing of the human genome. Conceptually, a high-throughput approach permitting correlation between newly discovered genes and functional properties of their protein products has yet to be developed. To address relationships between tens of thousands of genes and their cognate proteins, novel interdisciplinary technologies need to emerge. In this paper, a new idea of immunomics is presented and an experimental strategy is outlined to circumvent some of the restrictions associated with methodologies currently in use. It is proposed that cloned segments of genomic DNA are used for genetic immunization to obtain a large collection of antibodies, and to generate microarrays of these antibodies for tracing differentially expressed cellular proteins.  相似文献

The envelope protein of the human T-cell leukemia virus type I (HTLV-I) is highly conserved among the isolates sequenced so far, as opposed to what is observed for the human immunodeficiency virus (HIV) envelope. By linker insertion scanning, we have produced 33 random mutations along the HTLV-I envelope gene, cloned into a eukaryotic expression vector. The resulting envelope products were analysed by immunoprecipitation and syncytia formation after transfection into COS-1 cells. We show here that 25 out of 33 mutations result in a non-functional envelope product as assessed by the lack of ability to form syncytia. In the majority of these mutants, the processing of the envelope gp61 precursor into the mature gp45 and gp20 proteins was affected. We propose that conformational constraints for processing and fusion abilities tend to limit the variability of the HTLV-I envelope. In three mutants, processing was observed but no syncytia were formed. These mutations might affect regions important for HTLV-I envelope functions, such as the receptor binding region.  相似文献

The interactions between herbivores and plants are of general interest in ecology. Even though the extensive research carried out during the last decades has culminated in many theories, additional studies are necessary to validate these findings. In particular, the hypotheses dealing with the complex interrelations of plant defense mechanisms and herbivores continue to be debated.In this paper, we develop a new indicator value that quantifies the defense mechanisms of Central European woody plants against large mammalian herbivores. The indicator value is based on three plant-specific traits: chemical defense (toxic compounds, digestion inhibitors), mechanical defense and leaf size. Our validation of the newly established indicator shows that evergreen woody plants have a significantly higher indicator value than deciduous woody plants. Moreover, plant defense is correlated with growth height: woody plants growing in the browsing zone preferred by large mammalian herbivores have significantly higher levels of defense compared with woody plants capable of growth high above the reach of large herbivores.We conclude that the new plant defense indicator value is a valuable tool for the validation of existing hypotheses and habitat calibration on a statistical basis. The quantification of plant mechanisms of defense against large herbivores produces a significantly better understanding of the multifaceted nature of plant–animal interactions and should contribute positively to future studies.  相似文献

 Sexual reproduction in plants is intimately connected to the activity of the cytoskeletal apparatus in reproductive cells. Because of the ease with which the pollen tube can be studied, it has become a model for studying many aspects of cell physiology related to the cytoskeleton, such as movement of organelles and vesicles and cell division. However, information about cytoskeletal proteins is still insufficient for determining cytoskeletal functions during reproduction, especially in terms of cell-cell interactions. One reason may be that cytological and biochemical research on the cytoskeleton of pollen and the embryo sac has not been complemented by sufficient research activity at genetic and molecular levels, and few laboratories are currently involved in this work. This might be because of problems in identifying appropriate applied applications of the work that might attract more investigation. Some herbivores deliberately consume a mixed diet, either to obtain a superior mix of nutrients or to avoid consuming too much of any one toxin. Few studies have examined diet mixing in parasitic plants, which typically have very broad host ranges. We offered the parasitic plant Cuscuta indecora (dodder), a range of mixtures of two hosts (Iva frutescens and Borrichia frutescens) in the greenhouse, and observed correlations between the host community and Cuscuta infection in the field. In the greenhouse, Cuscuta performed better on mixtures with a higher relative abundance of Iva. Cuscuta selectively foraged on whichever host was more abundant (diet switching), the exact opposite of the behavior that would be expected if diet mixing was advantageous. In the field, the intensity of Cuscuta infections was decreased by the presence of non-hosts (grasses), not strongly affected by the presence of intermediate hosts, and increased by the presence of Borrichia. We conclude that Cuscuta does not obtain nutritional benefits from a broad diet, but instead is constrained by its relative lack of mobility to attack hosts of intermediate value. In general, the lack of mobility of parasitic plants compared to herbivores probably selects for broad host ranges in parasitic plants.  相似文献

Velvetbean (Mucuna spp.), a vigorous climbing legume of Asian origin, is prominent among the plants currently promoted for use as a green manure and cover crop in the humid tropics. What is not so well known, however, is that the development and diffusion of cropping systems using velvetbean is the result of experimentation by numerous farmers and scientists spanning four centuries and at least eight countries. This article traces the movement of velvetbean and knowledge of its uses, with a view to identifying some of the conditions under which the crop has waxed and waned in the United States and Mesoamerica. Climatic factors, land and labor constraints, and market forces are discussed. The velvetbean story shows that agricultural innovation is neither static nor the purview of a privileged class of innovators. It also suggests that sustainable cropping practices such as green manuring should not be promoted as static models but rather as dynamic systems responsive to the changing conditions of farmers and the broader environment.  相似文献

A major challenge in formulating an effective immunotherapy is to overcome the mechanisms of tumor escape from immunosurveillance. We showed that hypoxia-induced autophagy impairs cytotoxic T-lymphocyte (CTL)-mediated tumor cell lysis by regulating phospho-STAT3 in target cells. Autophagy inhibition in hypoxic cells decreases phospho-STAT3 and restores CTL-mediated tumor cell killing by a mechanism involving the ubiquitin proteasome system and SQSTM1/p62. Simultaneously boosting the CTL-response, using a TRP-peptide vaccination strategy, and targeting autophagy in hypoxic tumors, improves the efficacy of cancer vaccines and promotes tumor regression in vivo. Overall, in addition to its immunosuppressive effect, the hypoxic microenvironment also contributes to immunoresistance and can be detrimental to antitumor effector cell functions.  相似文献

Patients with systemic lupus erythematosus (SLE) have a greatly increased risk of cardiovascular disease. There is growing interest in the link between vascular damage and lupus-specific inflammatory factors. Impaired endothelial repair could account for the endothelial dysfunction in this patient group. This review describes the contribution that endothelial progenitor cells could play in the pathogenesis of premature vascular damage in this disease. The methods of isolation, detection, and characterization of endothelial progenitor cells, together with their potential role in repair of the endothelium and as a therapeutic target in SLE, are discussed.  相似文献
