The growth pattern of the thallus of Lobaria pulmonaria, a foliose lichen, has been described as differentiated into upwards and downwards growing lobes. The former show meristematic properties, whereas the latter, owing to the formation of soralia inactivating apical meristems, become senile lobes. A simple and sensitive histochemical procedure, the TBO test performed in this study, shows co-distribution of zones of active growth and polyphosphates accumulation in L. pulmonaria and gives evidence of the central role of phosphate in lichen metabolism. Upwards and downwards growing lobes mainly differentiate in pattern of accumulation of histochemically detectable polyphosphates. In the former, actively growing zones correspond to the algae, cortical and medullary hyphal cells adjacent to the algal layer of the pseudomeristematic marginal rim and of the adjacent elongation zone. In the downwards sorediate lobes, lacking apical growth, the actively growing zones correspond to medullary hyphae and algae occurring in areas where soredia are formed.  相似文献

Population genetics of the tree‐colonizing lichen Lobaria pulmonaria were studied in the largest primeval beech forest of Europe, covering 10 000 ha. During an intensive survey of the area, we collected 1522 thallus fragments originating from 483 trees, which were genotyped with eight mycobiont‐ and 14 photobiont‐specific microsatellite markers. The mycobiont and photobiont of L. pulmonaria were found to consist of two distinct gene pools, which are co‐existing within small areas of 3–180 ha in a homogeneous beech forest. The small‐scale distribution pattern of the symbiotic gene pools show habitat partitioning of lineages associated with either floodplains or mountain forests. Using approximate Bayesian computation (ABC), we dated the divergence of the two fungal gene pools of L. pulmonaria as the Early Pleistocene. Both fungal gene pools survived the Pleistocene glacial cycles in the Carpathians, although possibly in climatically different refugia. Fungal diversification prior to these cycles and the selection of photobionts with different altitudinal distributions explain the current sympatric, but ecologically differentiated habitat partitioning of L. pulmonaria. In addition, the habitat preferences of the mycobiont are determined by other factors and are rather independent of those of the photobiont at the landscape level. The distinct gene pools should be considered evolutionarily significant units and deserve specific conservation priorities in the future, for example gene pool A, which is a Pliocene relict.  相似文献

In this study, in vitro antimicrobial activity of the physodic acid, usnic acid, atranorin and gyrophoric acid isolated from the lichens Hypogymnia physodes, Parmelia caperata, Physcia aipolia and Umbilicaria polyphylla, has been investigated. An antibiotic assessment was done against six bacteria (three Gram-positive and three Gram-negative) and eight fungi by determining the minimal inhibitory concentration (MIC) by the broth tube dilution method. The tested lichen substances inhibited growth of all the tested microorganisms. The bacteria showed a higher sensitivity against the tested fungi. The highest antimicrobial activity was found in the usnic acid of the Parmelia caperata lichen, where the lowest MIC was 0.0037 mg/ml against the Klebsiella pneumoniae (even lower than the one given by the streptomycin standard). The weakest antimicrobial activity was found in the physodic acid, which inhibited most of the microorganisms in the concentration of 1 mg/ml. Generally, all the components had relatively strong antimicrobial activity against the tested microorganisms, among which were human and animal pathogens. This could be of significance for their use for pharmaceutical purposes.  相似文献

Abstract

The distribution of the epiphytic macrolichen Lobaria pulmonaria (L.) Hoffm. was surveyed in the Regional Forest of Cansiglio, extending over 3,500 ha in the eastern part of the Veneto Pre-Alps (north-east Italy). Data on the main characteristics of the forest sites and on Lobaria trees were collected with the aim to evaluate the main ecological factors, related to forest composition and management, affecting the distribution of Lobaria pulmonaria. Lichen diversity was surveyed on 15 randomly selected Lobaria trees. A good correlation between macrolichens and the total number of species was found. The largest Lobaria populations are concentred in the north-eastern part of the forest, where mixed and multi-plane silver fir – beech formations prevail, and selective cutting is carried out. Our data has enabled us to develop some criteria for the identification of priority conservation sites for Lobaria pulmonaria and lichen diversity in the study area, improving the focus on biodiversity in the management of the forest.  相似文献

Lichen-forming fungi have been assumed to be more or less restricted to the surface of the substrate on which they grow, Conclusive identification of hyphae or an assessment of the fungal diversity inside lichen-covered rock has not been possible using methods based on direct observation. We circumvented this problem by using a DNA sequencing approach. Cores were drilled from a Devonian arcosic sandstone rock harboring the crustose lichen Ophioparma ventosa (L.) Norman on the surface. The cores were cut vertically, and DNA was extracted from the pulverized rock slices. A series of polymerase chain reactions using fungal-specific primers as well as Ophioparma ventosa specific primers were employed to amplify the internal transcribed spacer region of the nuclear ribosomal DNA. The results show that hyphae of O. ventosa penetrate approximately 10–12 mm into the rock. Consequently, the hyphal layer formed by the lichen fungus inside the rock could be 7–12 times as thick as the symbiotic thallus at the surface of the rock. In addition, eight non-lichenized fungal taxa and five that could not be identified to species were encountered. One fungal species in the order Helotiales occurs in six of the eight cores. The significance of these results to the colonization and weathering of rock by lichenized fungi is discussed.  相似文献

Antimicrobial features of acetone, methanol and aqueous extracts of lichens of Cladonia furcata, Parmelia caperata, Parmelia pertusa, Hypogymnia physodes and Umbilicaria polyphylla were investigated by two different methods at the same time. Testing of antimicrobial activities of extracts from five species of lichens was performed by disc diffusion test in relation to Gram-positive and Gram-negative bacteria and fungal organisms, and through determination of minimal inhibitory concentration (MIC) by Broth Tube Dilution method. The obtained results indicated that acetone and methanol extracts of all investigated lichens in different concentrations manifested selective antibacterial and antifungal activity. That activity was more evident in relation to Gram-positive, than Gram-negative bacteria and fungal organisms. Acetone and methanol extracts of lichens Parmelia pertusa, Hypogymnia physodes and Umbilicaria polyphylla inhibited the growth of all tested microorganisms, most of all of lichens Cladonia furcata and Parmelia caperata. Although, the methanol extracts were generally the most active against the test organisms, the lowest MIC value was measured for acetone extract of species Cladonia furcata 0.39 mg/mL in relation to bacterium Bacillus subtilis. Aqueous extracts of investigated lichens were inactive against all tested organisms.  相似文献

Arthopyrenia endobrya from Southern Brazil is illustrated and described as a new species of lichens. The thallus is composed of filamentous green algae loosely surrounded by fungal hyphae. Both symbionts grow endophytically within the leaf cells of two species ofLejeuneaceae (Hepaticae). The algae and hyphae penetrate the cell walls of the host by means of fine perforations. The ascocarps develop between the leaves and perforate them with their apical region. The classification as a member of the genusArthopyrenia is preliminary.

Variations in stable carbon isotope discrimination (δ) were investigated across the thalli of several lichen species possessing different photobiont associations. Lichens containing (i) green algae (phycobiont), (ii) green algae in association with cyanobacteria confined in cephalodia, or (iii) cyanobacteria (cyanobiont) as the photobiont partner were studied. Carbon isotope discrimination was analysed in different thallus sections, which varied in distance from the margin and in age. The marginal thallus region is considered to be youngest, while the central region is thought to be oldest. This analysis showed a clear variation in δ across the thallus related to distance from the growing margin. In most of the species examined, the highest δ values were found in marginal regions (younger), while the central and basal regions (older) showed significantly lower δ. To investigate the effects of the historical increase in atmospheric CO2 concentration and the concurrent decrease in the 13C content of atmospheric CO2 on the δ of lichens, experiments were carried out on herbarium samples of Lobaria pulmonaria collected from the mid 19th Century to 1953. The results obtained showed a pattern of variation of δ consistent with that of freshly collected samples; δ decreased substantially with increasing distance from the thallus margin, irrespective of the collection date. Moreover, no consistent variation of discrimination was found among different collection dates. These results demonstrate that the observed variation in δ is caused by age-related changes in the physiological behaviour of different thallus sections, and that the past 150 years of increasing CO2 concentration have not had significant effects on A in L. pulmonaria. Photosynthesis measurements, chlorophyll analysis and observations using optical microscopy, performed on freshly collected lichens, showed significant changes in morphological and physiological characteristics across the thallus. Particularly, remarkable variations in thickness were found across the thallus. These anatomical changes may be responsible for the variation in δ, through variations in CO2 transfer resistance and, consequently, in CO2 availability across the thallus. The lack of age-dependent variation in δ in cyanobiont lichens is possibly attributable to the operation of a CO2-coneentrating mechanism and, therefore, to a more constant CO2 environment across the thallus in this lichen group.  相似文献

This light and electron microscope study of the liverwort Asterellareveals that, as in other Marchantiales, the cells lining thedorsal air chambers are highly vacuolate with numerous amylochloroplastsin the peripheral cytoplasm. The ventral parenchyma in the midribof the thallus contains aseptate fungal hyphae surrounded byan interfacial matrix and host cytoplasm forming transvacuolarstrands. These are lined with microtubules, rarely seen in otherfungal-hepatic associations or in mycorrhizas. Numerous lipidbodies found in all the thallus cells are thought to be associatedwith perennation during the winter dry season. Elongated, thick walled inner thallus cells, between the dorsalair chambers and the fungus-containing tissue, have a cytologicalorganization not previously recorded in land plants. Initiallyhighly vacuolate, with numerous microtubules of random orientationlining the tonoplast, these cells subsequently show interdigitationof vacuoles and cytoplasm producing a labyrinth of sphericaland elongate tonoplast profiles lined by longitudinal arraysof microtubules. At the same time the cytoplasm becomes increasinglyelectron-lucent and the ribosomes, progressively lost from theER, clump together. At maturity the inner thallus cells arehighly polarized with most of the vacuoles lying nearer thethallus apex. In pits in the end walls, numerous plasmodesmata,with expanded cytoplasmic annuli recall the plamodesmatal fieldsin the mesophyll and phloem of the leaves in vascular plants. Far from being supporting parenchyma or sclerenchyma as assumedhitherto, the inner thallus cells of Asterella are clearly highlydifferentiated. In lichen symbiosis, fungal and algal partners form close associations, often codispersed by vegetative propagules. Due to the particular interdependence, processes such as colonization, dispersal or genetic drift are expected to result in congruent patterns of genetic structure in the symbionts. To study the population structure of an obligate symbiotic system in Europe, we genotyped the fungal and algal symbionts of the epiphytic lichen Lobaria pulmonaria at eight and seven microsatellite loci, respectively, and analysed about 4300 L. pulmonaria thalli from 142 populations from the species' European distribution range. Based on a centroid approach, which localizes centres of genetic differentiation with a high frequency of geographically restricted alleles, we identified the South Italy–Balkan region as the primary glacial refugial area of the lichen symbiosis. Procrustean rotation analysis and a distance congruence test between the fungal and algal population graphs indicated general concordance between the phylogeographies of the symbionts. The incongruent patterns found in areas of postglacial recolonization may show the presence of an additional refugial area for the fungal symbiont, and the impact that horizontal photobiont transmission and different mutation rates of the symbionts have on their genotypic associations at a continental scale.  相似文献

Lichen-forming ascomycetes exhibit often complex morphologies of the vegetative thallus that are usually not found in non-lichenized fungi. This includes the thallus organization and appendical structures associated with the main thallus, such as cilia and rhizines. Such morphological characters are widely employed in the taxonomy of parmelioid lichens, especially at generic level. Within parmelioid lichens, several monophyletic groups can be distinguished, the Hypotrachyna clade being one of them, which includes mostly tropical taxa. In this first molecular study focused specifically on the Hypotrachyna clade, we used maximum parsimony and Bayesian analyses of a combined data set of nuclear ITS and mitochondrial SSU rDNA sequences to (1) test the monophyly of genera presently accepted within the clade and (2) evaluate the phylogenetic value of the morphological characters used to circumscribe genera in parmelioid lichens. Out of the 89 mtSSU and 88 nuITS sequences included in the present study, 121 sequences were newly obtained. Our results show that the taxa within the clade fall into two major groups and that the genus Hypotrachyna is polyphyletic. Everniastrum and Parmelinopsis are nested within Hypotrachyna sensu stricto, the latter being also polyphyletic. Bulbothrix is paraphyletic with Parmelinella nested within and is basal to the second major Hypotrachyna clade. Monophylies of Bulbothrix and Hypotrachyna are significantly rejected. The phylogenetic analysis demonstrates that morphological characters currently used to circumscribe genera in parmelioid lichens, such as cortical anatomy, lobe configuration, cilia, and rhizines have been overestimated and have only minor value in identifying monophyletic groups.  相似文献

The mycobiont of the high-light-susceptible forest lichen Lobaria pulmonaria was shown to deposit brown, melanic compounds in the outer layer of the upper cortex, depending on the long-term level of solar radiation in its natural habitat. Furthermore, pale thalli from a shady habitat produced melanic compounds after transplantation to a sunny habitat. This browning of the cortex appeared to be a physiologically active process, taking place only during periods with frequent hydration. Melanin production was slow. After transplantation, more than 1 year was needed for a shade-adapted thallus to reduce the cortical transmittance (230-1000 nm) to a similar level to that of naturally sun-exposed specimens. Melanic compounds acted as a sun screen, especially reducing UVB and UVA wavelengths, but also visible wavelengths, at the photobiont level. In the near infrared range, there was only a small difference in transmittance between shade- and sun-adapted cortices. A negative correlation was found between the natural light level and the cortical transmittance of wavelengths below 700 nm. However, previous studies have shown that even photobionts of melanic L. pulmonaria thalli are relatively susceptible to high-light exposure. Since melanins also increase the absorbance of solar energy for the whole thallus, it appears that what is gained in terms of UV- and light protection in melanic L. pulmonaria specimens may be offset by increased exposure to excess temperatures for this highly heat-susceptible lichen.  相似文献

Lichens were cultured by attaching a thallus fragment to a nylon monofilament loop with silicone sealer. Two effective methods for adjusting lichen mass to a standard moisture content were developed (the ‘reference-sample’ and ‘sacrificial’ methods). These corrections for moisture content allow detection of very small changes in dry mass without having to oven dry (and kill) all transplants. Average annual biomass growth rates for non-fragmenting species were typically between 5 and 30%. Annual biomass growth rates of healthy, vigorous individuals, as indicated by the 75th percentile, were mostly between 10 and 40%.Alectoria sarmentosawas prone to fragmentation despite the maintenance of healthy thalli. The lichens, Nephroma expallidum (Nyl.) Nyl. and N. arcticum (L.) Torss., consistently have at least two symbionts in a single thallus: a green alga in the algal layer and a blue-green alga in the internal cephalodia. The cephalodia originate from algal cells in contact with the lower surface of the lichen, in the zone of rhizine formation. The rhizines surround the epiphytic algal colony and form a second cortical layer; following dissociation of the original lower cortex, further growth of the two organisms results in the cyanophyte colony being enveloped by a compact layer of fungal tissue and positioned in the lichen medulla. The colony may eventually assume a superior or inferior position in relation to the lichen thallus, depending in part on the lichen species. Nephroma anticum may have two distinct morphological forms of blue-green algae in the same thallus and occasionally in the same cephalodium. It appears that the relationship that exists between the cephalodial algae and the lichen thallus is antagonistic and results, in some cases, in the exclusion of the green algal layer and death to the cephalodial cyanophytes.  相似文献

It is necessary to understand how environmental changes affect plant fitness to predict survival of a species, but this knowledge is scarce for lichens and complicated by their formation of sexual and asexual reproductive structures. Are the presence and number of reproductive structures in Lobaria pulmonaria, a threatened lichen, dependent on thallus size, and is their formation sequential? Does any size-dependence and sequential formation vary along a climate gradient? Generalized linear mixed models were used to explore the effect of environmental predictors on the size and presence/abundance of each reproductive structure and to determine the probability of a given-sized thallus to develop any reproductive structure. The largest individuals are more likely to develop reproductive structures, and the lichen uses a mixed strategy of early asexual reproduction and late sexual. Macro and microclimatic variables also influenced reproductive capacity. Relationships among climate conditions and lichen size and reproductive capacity can compromise the future viability of the species in the most southern populations of Europe.  相似文献

The profound knowledge of the structural and chemical characteristics of the interface between lichen thallus and rock, seems to be indispensable for the process of understanding the lichen symbiosis as well as the significance of the weathering action of lichens. One of the most promising techniques to be used in this investigation is the Scanning Electron Microscopy (SEM) in the back-scattered electron (BSE) emission mode. In the present work thalli of Parmelia conspersa, Aspicilia intermutans and Lecidea auriculata growing in granitic rock were examined by SEM in BSE mode with (Energy Dispersive Spectroscopy) EDS. In the case of the foliose thalli the observation of the interface permits detection of the rhizine/hyphae adherence and determination of the origin of the minerals which adhere to the rhizine/hyphae. In the case of the crustose thalli BSE permits investigation within the ultrastructure of the crustose thallus and crustose lichen-rock contact zone and also allows observations of the penetration and filling of the fissures and cracks of the underlying rock by components of the thallus and other living organisms. The BSE images could contribute to a better knowledge of the cytological state of the rock-inhabiting organisms and also to the understanding of the action of the chemical treatments used in the removal of lichen from building materials.  相似文献

Although water is essential for photosynthetic activation in lichens, rates of vapor uptake and activation in humid air, which likely influence their niche preferences and distribution ranges, are insufficiently known. This study simultaneously quantifies rehydration kinetics and PSII reactivation in sympatric, yet morphologically and functionally distinct cephalolichens (Lobaria amplissima, Lobaria pulmonaria, Lobaria virens). High-temporal resolution monitoring of rehydrating thalli by automatic weighing combined with chlorophyll fluorescence imaging of maximal PSII efficiency (FV/FM) was applied to determine species-specific rates of vapor uptake and photosynthetic activation. The thin and loosely attached growth form of L. pulmonaria rehydrates and reactivates faster in humid air than the thick L. amplissima, with L. virens in between. This flexible hydration strategy is consistent with L. pulmonaria’s wide geographical distribution stretching from rainforests to continental forests. By contrast, the thick and resupinate L. amplissima reactivates slowly in humid air but stores much water when provided in abundance. This prolongs active periods after rain, which could represent an advantage where abundant rain and stem flow alternates with long-lasting drying. Understanding links between morphological traits and functional responses, and their ecological implications for species at risk, is crucial to conservation planning and for modelling populations under various climate scenarios.  相似文献

Compartmentation of heavy metals on or within mycorrhizal fungi may serve as a protective function for the roots of forest trees growing in soils containing elevated concentrations of metals such as Cd and Zn. In this paper we present the first quantitative measurements by X‐ray microanalysis of heavy metals in high‐pressure frozen and cryosectioned ectomycorrhizal fungal hyphae. We used this technique to analyse the main sites of Cd and Zn in fungal cells of mantle and Hartig net hyphae and in cortical root cells of symbiotic Picea abies – Hebeloma crustuliniforme associations to gain new insights into the mechanisms of detoxification of these two metals in Norway spruce seedlings. The mycorrhizal seedlings were exposed in growth pouches to either 1 mM Cd or 2 mM Zn for 5 weeks. The microanalytical data revealed that two distinct Cd‐ and Zn‐binding mechanisms are involved in cellular compartmentation of Cd and Zn in the mycobiont. Whereas extracellular complexation of Cd occurred predominantly in the Hartig net hyphae, both extracellular complexation and cytosolic sequestration of Zn occurred in the fungal tissue. The vacuoles were presumed not to be a significant pool for Cd and Zn storage. Cadmium was almost exclusively localized in the cell walls of the Hartig net (up to 161 mmol kg ? 1 DW) compared with significantly lower concentrations in the cell walls of mantle hyphae (22 mmol kg ? 1 DW) and in the cell walls of cortical cells (15 mmol kg ? 1 DW). This suggests that the apoplast of the Hartig net is a primary accumulation site for Cd. Zinc accumulated mainly in the cell walls of the mantle hyphae (111 mmol kg ? 1 DW), the Hartig net hyphae (130 mmol kg ? 1 DW) and the cortical cells (152 mmol kg ? 1 DW). In addition, Zn occurred in high concentrations in the cytoplasm of the fungal mantle hyphae (up to 164 mmol kg ? 1 DW) suggesting that both the cell walls and the cytoplasm of fungal tissue are the main accumulation sites for Zn in P. abies resulting in decreased Zn transfer from the fungus to the root.  相似文献

By surveying and re-surveying 12 forest sites in southern Sweden for the epiphytic lichen Lobaria pulmonaria with a 9-year interval, and measuring tree-related habitat quality variables, we have investigated whether the local distribution of the lichen is limited by poor dispersal capacity or by habitat quality. Dispersal distances were measured indirectly as the distances between colonised trees and the nearest trees occupied by L. pulmonaria in both 1992 and 2001. To compare habitat quality between trees occupied by L. pulmonaria and neighbouring control trees, we recorded tree species and measured age and growth rate of trees, light conditions, bark structure and bryophyte cover. The estimated mean dispersal distance was 35 m, with a recorded maximum of 75 m. Occupied trees were larger and had a larger cover of bryophytes than unoccupied trees of similar size. The results indicate that dispersal capacity probably is the most important factor in limiting the local distribution of L. pulmonaria, but habitat-quality factors may be important on a smaller spatial scale.  相似文献

Caloplaca erodens is a new species of sect. Pyrenodesmia, characterised by an orbicular, sorediate, bluish-grey thallus which is endolithic but emerges at the periphery with a white, K−, obscurely lobate prothallus. The species is frequent on calcareous outcrops and walls of isolated churches and ruins of the Central Apennines (Sibillini, Gran Sasso), where it may occur in large monospecific populations, from 1000 to 2500 m asl., and is also known from dry sites of the southern Alps. It has been found with apothecia only in the type locality, being predominantly sterile. The reproduction of this lichen is evidently linked to the release of fragments of clusters of photobiont cells and mycobiont hyphae which are continuously exposed with the dissolution of the substratum. Readily distinguished from the apparently similar endolithic C. alociza (which is characterised by numerous apothecia, black, K+ purple prothallus, and esorediate thallus), C. erodens probably belongs to the C. circumalbata complex, whose taxa are always epilithic but have a white, K− prothallus. The phylogenetic position of the new species within sect. Pyrenodesmia as inferred by ITS sequences of the nuclear ribosomal DNA is shortly discussed.  相似文献
