The hypothesis of the present study was that bacterial communities would differentiate under Eucalyptus camaldulensis and that an enhancement of arbuscular mycorrhizal (AM) density would minimize this exotic plant species effect. Treatments consisted of control plants, preplanting fertilizer application and AM inoculation. After 4 months of culture in autoclaved soil, E. camaldulensis seedlings were either harvested for growth measurement or transferred into containers filled with the same soil but not sterilized. Other containers were kept without E. camaldulensis seedlings. After 12 months, effects of fertilizer amendment and AM inoculation were measured on the growth of Eucalyptus seedlings and on soil microbial communities. The results clearly show that this plant species significantly modified the soil bacterial community. Both community structure (assessed by denaturing gradient gel electrophoresis profiles) and function (assessed by substrate-induced respiration responses including soil catabolic evenness) were significantly affected. Such changes in the bacterial structure and function were accompanied by disturbances in the composition of the herbaceous plant species layer. These results highlight the role of AM symbiosis in the processes involved in soil bio-functioning and plant coexistence and in afforestation programmes with exotic tree species that target preservation of native plant diversity.  相似文献

We investigated in two tobacco (Nicotiana tabacum) plant lines (wildtype or antisense mutant) whether impairment in expression of the plasma membrane aquaporin gene (NtAQP1) affects the arbuscular mycorrhizal (AM) fungal colonisation pattern or the symbiotic efficiency of AM fungi. These two objectives were investigated under well-watered and drought stress conditions. Both plant lines had a similar pattern of root colonisation under well-watered and drought stress conditions. In contrast, under drought stress, AM wildtype plants grew faster than mycorrhizal antisense plants. Plant gas exchange also appeared to depend on the expression of NtAQP1 and parallelled the determined growth increments. The implications of enhanced symplastic water transport via NtAQP1 for the efficiency of the AM symbiosis under drought stress conditions are further discussed.  相似文献

采用分室培养系统,模拟正常水分和干旱胁迫两种环境条件,探讨不同丛枝菌根真菌(arbuscular mycorrhizal fungi,AMF)对紫花苜蓿(Medicago sativa L.)生长和土壤水稳性团聚体的影响.试验条件下,Glomus intraradices对苜蓿根系的侵染率均显著高于Acaulospora scrobiculata和Diversispora spurcum接种处理.正常水分条件下,供试AM真菌均能显著提高植株生物量及磷浓度.干旱胁迫显著抑制了植株生长和菌根共生体发育,总体上菌根共生体对植株生长没有明显影响,接种D.spurcum甚至趋于降低植株生物量;同时,仅有G.intraradices显著提高了植株磷浓度.AM真菌主要影响到>2mm的水稳性团聚体数量,以G.intraradices作用效果最为显著.在菌丝室中,G.intraradices显著提高了总球囊霉素含量.研究表明AM真菌对土壤大团聚体形成具有积极作用,而菌根效应因土壤水分条件和不同菌种而异,干旱胁迫下仅有G.intraradices对土壤结构和植物生长表现出显著积极作用.在应用菌根技术治理退化土壤时,需要选用抗逆性强共生效率高的菌株,对于不同AM真菌抗逆性差异的生物学与遗传学基础尚需进一步研究.  相似文献

Serpentine soils represent a unique environment that imposes multiple stresses on vegetation (low Ca/Mg ratios, macronutrient deficiencies, elevated heavy metal concentrations and drought). Under these conditions, a substantial role of arbuscular mycorrhizal (AM) symbiosis can be anticipated due to its importance for plant nutrition and stress alleviation. We tested whether serpentine and non-serpentine populations of Knautia arvensis (Dipsacaceae) differ in the benefits derived from native AM fungal communities. Four serpentine and four non-serpentine populations were characterised in terms of mycorrhizal colonisation and soil characteristics. The serpentine populations showed significantly lower mycorrhizal colonisation than their non-serpentine counterparts. The mycorrhizal colonisation positively correlated with soil pH, Ca and K concentrations and Ca/Mg ratio. Seedlings from each population were then grown for 3 months in their sterilised native substrates, either uninoculated or reinoculated with native AM fungi. Two serpentine and two non-serpentine populations responded positively to mycorrhizal inoculation, while no significant change in plant growth was observed in the remaining populations. Contrary to our hypothesis, serpentine populations of K. arvensis did not show higher mycorrhizal growth dependence than non-serpentine populations when grown in their native soils and inoculated with native AM fungi.  相似文献

测定分析了接种丛枝菌根(AM)真菌摩西管柄囊霉Funneliformis mosseae对正常供水与干旱处理的盆栽枳Poncirus trifoliata实生苗生长、活性氧代谢及抗氧化酶基因表达量的影响。结果表明,7周干旱处理显著降低了根系菌根侵染率。接种摩西管柄囊霉显著促进了干旱处理的枳植株生长,增加了根系体积和叶片相对含水量,显著降低了叶片脯氨酸含量,同时也上调了干旱处理的枳叶片精氨酸脱羧酶基因(PtADC1PtADC2)和超氧化物歧化酶基因(PtFe-SODPtMn-SOD)、过氧化物酶基因(PtPOD)和过氧化氢酶基因(PtCAT1)的表达,因而维持了一个相对更低的活性氧水平(如过氧化氢),有利于增强植株的抗旱性。  相似文献

We studied the effect of inoculation with a mixture of three arbuscular mycorrhizal (AM) fungi (Glomus intraradices Schenck & Smith, Glomus deserticola (Trappe, Bloss. & Menge) and Glomus mosseae (Nicol & Gerd.) Gerd. & Trappe) and addition of a composted organic residue on plant growth, nutrient uptake, mycorrhizal colonisation and superoxide dismutase (SOD, EC 1.15.1.1) and total peroxidase (POX, EC 1.11.1.7) activities in shoots of Juniperus oxycedrus seedlings after well-watered, drought and recovery periods. The mycorrhizal inoculation and composted residue addition significantly increased the growth, foliar nutrients (N, P, K) and shoot water content of the plants, independent of the water regime. POX activity in control plants increased during drought (about 250% higher than under well-watered conditions) and returned to initial levels after re-watering. The seedlings inoculated with AM fungi showed the highest values of POX activity, followed by the plants grown in the amended soil, which varied little during the drought and recovery periods. Drought decreased the SOD activity in shoots of both J. oxycedrus seedlings inoculated with AM fungi and those grown with composted residue, but did not affect that of control plants. After re-watering, the SOD activity in mycorrhizal or residue-amended plants increased, showing values similar to control plants.  相似文献

Medicago arborea can be used for re-vegetationpurposes under semiarid conditions. These woody legumes have the ability toforman association with arbuscular mycorrhizal (AM) fungi and rhizobial bacteria,which can be maximised by microorganisms producing certain stimulatingmetabolites acting as plant growth promoting rhizobacteria (PGPR). The effectsof single and combined inoculations using microorganisms with different andinteractive metabolic capacities, namely three Glomusspecies, two Rhizobium meliloti strains (a wild type, WTand its genetically modified derivative GM) and a plant growth promotingrhizobacterium, (PGPR), were evaluated. All three inoculated AM fungi affectedMedicago growth in different ways. Differences weremaintained when soil was co-inoculated with each of the rhizobial strains (WTorGM) and the PGPR. Mycorrhizal fungi were effective in all cases, but the PGPRonly affected plant growth specific microbial situations. PGPR increased growthof G. mosseae-colonised plants associated withRhizobium WT strain by 36% and those infected byG. deserticola when associated with the rhizobial GMstrainby 40%. The most efficient microbial treatments involved mycorrhizalinoculation, which was an indication of the AM dependency of this plantspecies.Moreover, PGPR inoculation was only effective when associated with specificmycorrhizal endophytes (G. mosseae plus WT andG.deserticola plus GM rhizobial strain). The reduced root/shoot (R/S)ratio resulting from PGPR inoculation, was an indication of more effective rootfunction in treated plants. AM colonisation and nodule formation wereunaffectedby the type of AM fungus or bacteria (rhizobial strain and/or PGPR). AM fromnatural soil were less infective and effective than those from the collection.The results supported the existence of selective microbial interactionsaffecting plant performance. The indigenous AM fungi appeared to be ineffectiveand M. arborea behaved as though it was highly dependentonAM colonisation, which implied that it must have a mycorrhizal association toreach maximum growth in the stressed conditions tested. Optimum growth ofmycorrhizal M. arborea plants was associated with specificmicrobial groups, accounting for a 355% increase in growth overnodulatedcontrol plants. The beneficial effect of PGPR in increasing the growth of awoody legume, such as M. arborea under stress, was onlyobserved with co-inoculation of specific AM endophytes. As a result of theinteraction, only shoot biomass was enhanced, but not as a consequence ofenhancing of the colonising abilities of the endophytes. The growthstimulation,occurring as a consequence of selected microbial groups, may be critical anddecisive for the successful establishment of plants under Mediterraneanclimaticand soil conditions.  相似文献

针对西北干旱半干旱地区土壤贫瘠与水分缺乏的问题,利用微生物与作物形成互惠互利的共生关系,本研究设置两个水分梯度:干旱胁迫(供试土壤最大持水量的35%)和正常水分(供试土壤最大持水量的75%),两个覆膜方式:无覆膜(NM)和覆膜(FM),4个接种微生物水平:单接AM真菌(AM)、单接解磷细菌(PSB)、联合接种AM真菌与解磷细菌(AM+PSB)以及对照(CK),研究不同水分和覆膜条件下4个接种微生物对玉米生长特性、地上养分吸收与水分利用效率的影响.结果表明: 与正常水分处理相比,干旱胁迫能够显著提高接种AM真菌处理的侵染率,但正常水分处理下土壤根外菌丝密度、总球囊霉素(T-GRSP)与易提取球囊霉素(EE-GRSP)含量明显提高.干旱胁迫下,单接AM真菌处理的促生作用和菌根效应表现最好,能够提高玉米生物量、水分利用效率和土壤有机碳含量,促进土壤N、P、K的吸收与运输,从而增加玉米地上部分N、P、K吸收量;而正常水分下,联合接种AM+PSB处理表现要好于单接AM和PSB处理,且其与覆膜的互作效果最好.相关分析结果表明,玉米生物量、叶片SPAD值和地上部分N、P、K吸收量均与土壤根外菌丝密度呈显著正相关,玉米水分利用效率与其呈显著负相关.  相似文献

Biochar may alleviate plant water stress in association with arbuscular mycorrhizal (AM) fungi but research has not been conclusive. Therefore, a glasshouse experiment was conducted to understand how interactions between AM fungi and plants respond to biochar application under water-stressed conditions. A twin chamber pot system was used to determine whether a woody biochar increased root colonisation by a natural AM fungal population in a pasture soil (‘field’ chamber) and whether this was associated with increased growth of extraradical AM fungal hyphae detected by plants growing in an adjacent (‘bait’) chamber containing irradiated soil. The two chambers were separated by a mesh that excluded roots. Subterranean clover was grown with and without water stress and harvested after 35, 49 and 63 days from each chamber. When biochar was applied to the field chamber under water-stressed conditions, shoot mass increased in parallel with mycorrhizal colonisation, extraradical hyphal length and shoot phosphorus concentration. AM fungal colonisation of roots in the bait chamber indicated an increase in extraradical mycorrhizal hyphae in the field chamber. Biochar had little effect on AM fungi or plant growth under well-watered conditions. The biochar-induced increase in mycorrhizal colonisation was associated with increased growth of extraradical AM fungal hyphae in the pasture soil under water-stressed conditions.  相似文献

Two greenhouse experiments were focused on the application of arbuscular mycorrhizal fungi (AMF) and plant growth promoting rhizobacteria (PGPR) in planting of high-biomass crops on reclaimed spoil banks. In the first experiment, we tested the effects of different organic amendments on growth of alfalfa and on the introduced microorganisms. While growth of plants was supported in substrate with compost amendment, mycorrhizal colonization was suppressed. Lignocellulose papermill waste had no negative effects on AMF, but did not positively affect growth of plants. The mixture of these two amendments was found to be optimal in both respects, plant growth and mycorrhizal development. Decreasing doses of this mixture amendment were used in the second experiment, where the effects of microbial inoculation (assumed to compensate for reduced doses of organic matter) on growth of two high-biomass crops, hemp and reed canarygrass, were studied. Plant growth response to microbial inoculation was either positive or negative, depending on the dose of the applied amendment and plant species.  相似文献

The effects of bacterial inoculation (Bacillus sp.) on the development and physiology of the symbiosis between lettuce and the arbuscular mycorrhizal (AM) fungi Glomus mosseae (Nicol. and Gerd.) Gerd. and Trappe and Glomus intraradices (Schenck and Smith) were investigated. Plant growth, mineral nutrition and gas-exchange values in response to bacterial inoculation after PEG-induced drought stress were also evaluated. In AM plants, inoculation with Bacillus sp. enhanced fungal development and metabolism, measured as succinate dehydrogenase (SDH) and alkaline phosphatase (ALP) activities, more than plant growth. Under non-stressed conditions, G. intraradices colonization increased all plant physiological values to a higher extent when in dual inoculation with the bacterium. Under stress conditions, the bacterium had an important stimulatory effect on G. intraradices development. Under such conditions, the effects of the bacterium on photosynthetic rate, water use efficiency (WUE) and stomatal conductance of lettuce plants differed with the fungus species. Plant-gas exchange was enhanced in G. intraradices- and reduced in G. mosseae-colonized plants when co-inoculated with Bacillus sp. Thus, the effects of each fungus on plant physiology were modulated by the bacterium. Stress was detrimental, particularly in G. intraradices-colonized plants without the bacterium, reducing intra and extraradical mycelium growth and vitality (SDH), as well as plant-gas exchange. Nevertheless, Bacillus sp. inoculation improved all these plant and fungal parameters to the same level as in non-stressed plants. The highest amount of alive and active AM mycelium for both fungi was obtained after co-inoculation with Bacillus sp. These results suggest that selected free-living bacteria and AM fungi should be co-inoculated to optimize the formation and functioning of the AM symbiosis in both normal and adverse environments.  相似文献

The influence of Glomus etunicatum colonization on plant growth and drought tolerance of 3-month-old Pistacia vera seedlings in potted culture was studied in two different water treatments. The arbuscular mycorrhiza (AM) inoculation and plant growth (including plant shoot and root weight, leaf area, and total chlorophyll) were higher for well-watered than for water-stressed plants. The growth of AM-treated seedlings was higher than non-AM-treatment regardless of water status. P, K, Zn and Cu contents in AM-treated shoots were greater than those in non-AM shoots under well-watered conditions and drought stress. N and Ca content were higher under drought stress, while AM symbiosis did not affect the Mg content. The contents of soluble sugars, proteins, flavonoid and proline were higher in mycorrhizal than non-mycorrhizal-treated plants under the whole water regime. AM colonization increased the activities of peroxidase enzyme in treatments, but did not affect the catalase activity in shoots and roots under well-watered conditions and drought stress. We conclude that AM colonization improved the drought tolerance of P. vera seedlings by increasing the accumulation of osmotic adjustment compounds, nutritional and antioxidant enzyme activity. It appears that AM formation enhanced the drought tolerance of pistachio plants, which increased host biomass and plant growth.  相似文献

The objective of this study was to investigate the effects of arbuscular mycorrhizal fungus (AMF) inoculation on plant growth and drought tolerance in seedlings of a promising oilseed crop, Sacha Inchi (Plukenetia volubilis L.), under well-watered or drought conditions. AMF inoculation was applied in four treatments: without AMF inoculation, Glomus versiforme, Paraglomus occultum, or combination of both microorganism inoculations. The results showed that AMF colonization significantly enhanced the growth of Sacha Inchi seedlings regardless of soil water conditions, and the greatest development was reached in plants dually inoculated under well-watered conditions. G. versiforme was more efficient than P. occultum. Plants inoculated with both symbionts had significantly greater specific leaf area, leaf area ratio and root volume when compared with the uninoculated control, G. versiforme, and P. occultum treatments alone, indicating a synergistic effect in the two AMF inoculation. Photosynthetic rate and water-use efficiency were stimulated by AMF, but not stomatal conductance. Inoculation with AM fungus increased antioxidant enzymes activities including guaiacol peroxidase and catalase, thus lowering hydrogen peroxide accumulation and oxidative damage, especially under drought stress conditions. However, proline content showed little change during drought stress and AMF colonization conditions, which suggested that proline accumulation might not serve as the main compound for osmotic adjustment of the studied species. These results indicate that AMF inoculation stimulated growth and enhanced drought tolerance of Sacha Inchi seedlings, through alterations in morphological, physiological and biochemical traits. This microbial symbiosis might be an effective cultivation practice in improving the performance and development for Sacha Inchi plants.  相似文献

The aim of this research was to investigate the effect of arbuscular mycorrhizal (AM) colonisation on root morphology and nitrogen uptake capacity of carob ( Ceratonia siliqua L.) under high and low nutrient conditions. The experimental design was a factorial arrangement of presence/absence of mycorrhizal fungus inoculation ( Glomus intraradices) and high/low nutrient status. Percent AM colonisation, nitrate and ammonium uptake capacity, and nitrogen and phosphorus contents were determined in 3-month-old seedlings. Grayscale and colour images were used to study root morphology and topology, and to assess the relation between root pigmentation and physiological activities. AM colonisation lead to a higher allocation of biomass to white and yellow parts of the root. Inorganic nitrogen uptake capacity per unit root length and nitrogen content were greatest in AM colonised plants grown under low nutrient conditions. A better match was found between plant nitrogen content and biomass accumulation, than between plant phosphorus content and biomass accumulation. It is suggested that the increase in nutrient uptake capacity of AM colonised roots is dependent both on changes in root morphology and physiological uptake potential. This study contributes to an understanding of the role of AM fungi and root morphology in plant nutrient uptake and shows that AM colonisation improves the nitrogen nutrition of plants, mainly when growing at low levels of nutrients.  相似文献

Grassland productivity is often primarily limited by water availability, and therefore, grasslands may be especially sensitive to climate change. Fungal symbionts can mediate plant drought response by enhancing drought tolerance and avoidance, but these effects have not been quantified across grass species. We performed a factorial meta-analysis of previously published studies to determine how arbuscular mycorrhizal (AM) fungi and endophytic fungal symbionts affect growth of grasses under drought. We then examined how the effect of fungal symbionts on plant growth was influenced by biotic (plant photosynthetic pathway) and abiotic (level of drought) factors. We also measured the phylogenetic signal of fungal symbionts on grass growth under control and drought conditions. Under drought conditions, grasses colonized by AM fungi grew larger than those without mycorrhizal symbionts. The increased growth of grasses conferred from fungal symbionts was greatest at the lowest soil moisture levels. Furthermore, under both drought and control conditions, C3 grasses colonized by AM fungi grew larger than C3 grasses without symbionts, but the biomass of C4 grasses was not affected by AM fungi. Endophytes did not increase plant biomass overall under any treatment. However, there was a phylogenetically conserved increase in plant biomass in grasses colonized by endophytes. Grasses and their fungal symbionts seem to interact within a context-dependent symbiosis, varying with biotic and abiotic conditions. Because plant–fungal symbioses significantly alter plant drought response, including these responses could improve our ability to predict grassland functioning under global change.  相似文献

Background and Aims

Plants growing on serpentine bedrock have to cope with the unique soil chemistry and often also low water-holding capacity. As plant-soil interactions are substantially modified by arbuscular mycorrhizal (AM) symbiosis, we hypothesise that drought tolerance of serpentine plants is enhanced by AM fungi (AMF).

Methods

We conducted a pot experiment combining four levels of drought stress and three AMF inoculation treatments, using serpentine Knautia arvensis (Dipsacaceae) plants as a model.

Results

AMF inoculation improved plant growth and increased phosphorus uptake. The diminishing water supply caused a gradual decrease in plant growth, accompanied by increasing concentrations of drought stress markers (proline, abscisic acid) in root tissues. Mycorrhizal growth dependence and phosphorus uptake benefit increased with drought intensity, and the alleviating effect of AMF on plant drought stress was also indicated by lower proline accumulation.

Conclusions

We documented the role of AM symbiosis in plant drought tolerance under serpentine conditions. However, the potential of AMF to alleviate drought stress was limited beyond a certain threshold, as indicated by a steep decline in mycorrhizal growth dependence and phosphorus uptake benefit and a concomitant rise in proline concentrations in the roots of mycorrhizal plants at the highest drought intensity.  相似文献

Five species of stoloniferous plants originating from the same field site (Galeobdolon montanum, Glechoma hederacea, Potentilla anserina, Ranunculus repens and Trifolium repens) were studied with respect to their interaction with arbuscular mycorrhizal (AM) fungi. More specifically, the question was addressed whether mycorrhizal growth response of host plant species could be related to their vegetative mobility. The roots of all the species examined were colonised with AM fungi in the field, with the percentage of colonisation varying among species from approximately 40% to 90%. In a subsequent pot experiment, plants of all the species were either left non-inoculated or were inoculated with a mixture of three native AM fungi isolated from the site of plant origin (Glomus mosseae, G. intraradices and G. microaggregatum). AM fungi increased phosphorus uptake in all the plant species; however, plant growth response to inoculation varied widely from negative to positive. In addition to the biomass response, AM inoculation led to a change in clonal growth traits such as stolon number and length or ramet number in some species. Possible causes of the observed differences in mycorrhizal growth response of various stoloniferous plants are discussed.  相似文献

The effect of vesicular-arbuscular mycorrhizal (VAM) fungi on growth and drought resistance of Acacia nilotica and Leucaena leucocephala seedlings was studied in a glasshouse experiment. The experimental design was a 2·2·2 factorial: ± mycorrhizal inoculation, ± application of phosphorus fertilizer and ± repeated drought treatment. The growth promoting effect of VAM fungi equalled the effect of phosphorus fertilization after 12 weeks. The drought treatment reduced seedling biomass and nodulation. Differences between the plant species were found with respect to growth improvements due to VAM inoculation and/or phosphorus fertilization under drought stress conditions. The results are discussed in relation to plant drought resistance and reforestation in the subhumid to arid tropics.  相似文献

Both arbuscular mycorrhizal (AM) fungi and root hairs play important roles in plant uptake of water and mineral nutrients. To reveal the relative importance of mycorrhiza and root hairs in plant water relations, a bald root barley (brb) mutant and its wild type (wt) were grown with or without inoculation of the AM fungus Rhizophagus intraradices under well-watered or drought conditions, and plant physiological traits relevant to drought stress resistance were recorded. The experimental results indicated that the AM fungus could almost compensate for the absence of root hairs under drought-stressed conditions. Moreover, phosphorus (P) concentration, leaf water potential, photosynthetic rate, transpiration rate, stomatal conductance, and water use efficiency were significantly increased by R. intraradices but not by root hairs, except for shoot P concentration and photosynthetic rate under the drought condition. Root hairs even significantly decreased root P concentration under drought stresses. These results confirm that AM fungi can enhance plant drought tolerance by improvement of P uptake and plant water relations, which subsequently promote plant photosynthetic performance and growth, while root hairs presumably contribute to the improvement of plant growth and photosynthetic capacity through an increase in shoot P concentration.  相似文献
