A study on 12 entomopathogenic fungi for controlling broad mite (Polyphagotarsonemus latus (Banks)) in mulberry found that Metarhizium anisopliae CKM-048 was the most virulent strain in controlling both larvae and adult broad mites at the concentration of 2 x 10(8) conidia/ml. There was no ovicidal effect when tested with broad mite eggs. Median lethal concentrations (LC(50)) of M. anisopliae in killing larvae and adults were 8.7 x 10(6) and 1.3 x 10(7 )conidia/ml, respectively. Median lethal times (LT(50)) of larvae and adults were 2.4 and 3.8 days, respectively, at the concentration of 2 x 10(8) conidia/ml. The fungus was found to produce protease and chitinase. Scanning electron microscope (SEM) studies were done to monitor the infection steps of the fungus on broad mites. A greenhouse test on mulberry trees revealed that M. anisopliae could reduce the broad mite population within 4 days after treatment. However, after 7 days, its efficacy was decreased significantly.  相似文献

The pathogenicity of 32 fungal isolates from the genera of Beauveria, Verticillium, Paecilomyces, Metarhizium, Mariannaea, and Hirsutella to second-instar tarnished plant bug, Lygus lineolaris (Palisot de Beauvois), was tested under laboratory conditions. These isolates originated from various insect hosts and substrates from France, Denmark, Austria, Italy, Turkey, Syria, and the United States. A single exposure concentration (1 x 10(7) conidia/ ml) assay for each isolate was first conducted by immersing the insects in 10 ml of a fungal suspension for 5s. These were followed by concentration-mortality assays on five of the most pathogenic isolates using four test concentrations ranging from 2 x 10(4) to 2 x 10(7) conidia/ml. B. bassiana 726 (Bemisia-passaged GHA strain) was used as a standard for comparison in all of the assays. Among the test isolates, three produced mortality not significantly different from the water control. Mortality ranged from 35 to 98% among the other 29 isolates. The LC50 values of the five most pathogenic isolates ranged from 0.8 to 5.0 x 10(5) conidia/ml. The LT50 values for these isolates ranged from 6.0 to 6.9, 3.1 to 5.1, and 2.5 to 4.0 d for concentrations of 2 x 10(5), 2 x 10(6), and 2 x 10(7) conidia/ml, respectively. Two strains of B. bassiana (ARSEF 1394,5665) and one M anisopliae (ARSEF 3540) were more pathogenic to the nymphs than the standard, having significantly lower LC50 and LT50, values. Our results demonstrated that several genera of entomopathogenic fungi have promise as microbial control agents against L. lineolaris.  相似文献

The virulence of Hirsutella thompsonii (Fischer) to Brevipalpus phoenicis (Geijskes) was evaluated in laboratory, grown on complete and solid culture media (MC-S); complete and liquid culture media (MC-L); rice (APC) and powdered rice (APC-SM). Adults were confined to arenas prepared with citrus leaves in acrylic dishes containing water-agar. Conidial suspensions were prepared at different concentration (3.2 x 10(5) to 1 x 10(7) spores/ml) and applied on mites to establish the table curve-response on fourth day. For field evaluation, adults were maintained in arenas prepared with fruits which were placed in plants. In this test, four treatments were tried: H. thompsonii cultured on rice (APC) at two concentrations (20 kg/ha and 10 kg/ha), H. thompsonii produced by liquid fermentation (MC-L) (5 L/ha) and control (sterile water). Adult survival, number of eggs and nymphs per fruit were observed 10 and 20 days after the fungus application. The lowest LC25 value calculated was from pathogen produced in MC-S (1.9 x 10(5) conidia/ml).The LC25 values calculated to APC and APC-SM did not differ statistically. The LC25 values to MC-L and MC-S were 1.9 x 10(6) infective cells/ml and 2.2 x 10(5) conidia/ml. In the field, concentration and time to death differed between treatments and control. The applications resulted in reduction of adult survival and number of eggs.  相似文献

Pathogenicity of the hyphomycete, Verticillium lecanii (DAOM 198499), was investigated to aphid, Myzus persicae under laboratory conditions. Analysis of lethal effect of six various concentrations 0, 10(4), 10(5), 10(6), 10(7) and 10(8) conidia/ml of V. lecanii against third nymphal stage of M. persicae, indicated significant mortality on aphids. Mean comparisons showed that there was significant difference between treatments. Three days after treatment, aphid mortality observed and after 12 days minimal mortality was 17.77 in control and maximum was 100 percent related to 10(7) and 10(8) conidia/ml. LC50 and LT50 values were estimated by probit analysis and life test. LC50 value for aphid mortality was 1.4 x 10(4) conidia/ml, LT50 values for concentrations 10(4), 10(6), 10(6), 10(7) and 10(8) conidla/ml was 10, 10, 9, 8 and 6 days, respectively. In this experiment, net reproduction rate of aphid's (R0) decreased significantly when concentration increased. These observations showed that V. lecanii (DAOM 198499) can be an active biological agent against aphids.  相似文献

An emulsifiable concentrate (EC) and granular (G) formulation of the entomopathogenic fungus, Metarhizium brunneum strain F52 (formerly Metarhizium anisopliae strain F52) were tested against unfed adults and nymphs of Ixodes scapularis Say in the laboratory. Three exposure methods; dip, surface contact, and direct spray application, and three exposure time intervals (3, 30, and 300 min) were used to evaluate the EC formulation. Application rates ranged from 2.6 x 10(2) to 2.6 x 10(8) conidia/cm2. The surface treatment was used for granular formulation with concentrations ranging from 2.3 x 10(5) to 2.3 x 10(7) conidia/cm2 for same three exposure times. Both the EC and G formulations of this fungus were highly pathogenic against I. scapularis adults and nymphs. Logistic regression analysis found formulation, spore concentration, time of exposure, and observation period were significant or highly significant factors influencing tick mortality. For adult I. scapularis, the spray application with the EC formulation of M. brunneum F52 resulted in a lower LC50 (5.9 x 10(4) conidia/cm2) at 30 min than surface exposure to the EC (LC50 = 1.3 x 10(6) conidia/cm2) or G formulation (LC50 = 8.1 x 10(5) conidia/cm2). At higher concentrations, fungal activity was evident in adult I. scapularis held at 5 degrees C suggesting the fungus may provide control in the cooler fall season. While the observed pathogenicity of a fungus against ticks can be dependent upon the bioassay assessment, we found nymphs and adults of I. scapularis to be highly susceptible to M. brunneum F52, regardless of the exposure method used.  相似文献

Bioassays were carried out under controlled conditions (27 +/- 2 degrees C, 80 +/- 5% RH, and a photoperiod of 12:12 [L:D] h) to evaluate the effect of eight strains of the entomopathogenic fungus Beauveria bassiana upon larvae, pupae, and adult females of the Mexican fruit fly, Anastrepha ludens (Loew). Mortality of the immature stages was low, 2-8% in larvae and 0% in pupae. However, very high levels of mortality were obtained for adult flies, with values of 100, 98, and 98% for the strains Bb16, Bb24, and Bb26, respectively. LC50 values for these three strains ranged from 3.12 x 10(6) to 9.07 x 10(6) conidia/ml. Lethal time 50 (LT50) was 2.8, 3.7, and 4.2 d for Bb16, Bb26, and Bb24 strains, respectively, with an average LT50 of 4.4 d across all strains. The fungal mycelium emerged through the soft parts of the exoskeleton, such as the wing bases, mouth, intersegmental regions of the legs, and membranous regions of the abdomen, coxae, and neck. Maximum percentage sporulation ranged from 66.4 to 74.7% for the three most virulent strains.  相似文献

Background

Dengue is the most prevalent arboviral disease transmitted by Aedes aegypti worldwide, whose chemical control is difficult, expensive, and of inconsistent efficacy. Releases of Metarhizium anisopliae—exposed Ae. aegypti males to disseminate conidia among female mosquitoes by mating represents a promising biological control approach against this important vector. A better understanding of fungus virulence and impact on reproductive parameters of Ae. aegypti, is need before testing auto-dissemination strategies.

Methodology/Principal Findings

Mortality, mating competitiveness, sperm production, and the capacity to auto-disseminate the fungus to females up to the 5thcopulation, were compared between Aedes aegypti males exposed to 5.96 x 107 conidia per cm2 of M. anisopliae and uninfected males. Half (50%) of fungus-exposed males (FEMs) died within the first 4 days post-exposure (PE). FEMs required 34% more time to successively copulate with 5 females (165 ± 3 minutes) than uninfected males (109 ± 3 minutes). Additionally, fungus infection reduced the sperm production by 87% at 5 days PE. Some beneficial impacts were observed, FEMs were able to successfully compete with uninfected males in cages, inseminating an equivalent number of females (about 25%). Under semi-field conditions, the ability of FEMs to search for and inseminate females was also equivalent to uninfected males (both inseminating about 40% females); but for the remaining females that were not inseminated, evidence of tarsal contact (transfer of fluorescent dust) was significantly greater in FEMs compared to controls. The estimated conidia load of a female exposed on the 5th copulation was 5,200 mL-1 which was sufficient to cause mortality.

Conclusion/Significance

Our study is the first to demonstrate auto-dissemination of M. anisopliae through transfer of fungus from males to female Ae. aegypti during mating under semi-field conditions. Our results suggest that auto-dissemination studies using releases of FEMs inside households could successfully infect wild Ae. aegypti females, providing another viable biological control tool for this important the dengue vector.  相似文献

Abstract:  The effect of diamondback moth (DBM), Plutella xylostella (Lep., Plutellidae) male and female multiple mating on fecundity, fertility, and longevity was studied. Males could mate for five times with virgin females during scotophase. The successful copulation rates, fecundity of female, and longevity of both females and males decreased when male mating times increased, whereas copulation duration increased. Correlation coefficient between copulation duration and male mating times was significant ( r  = 0.7358, P = 0.0001, spearman rank-order correlation). There were linear relationships between mating history of males and longevities of males and females, and regression relationships between them were significant. Mated females had similar daily reproductive pattern, which laid the most eggs on the first day after mating in spite of their mates' mating history. Virgin females laid some infertile eggs before they died. Most of the females mated once during their lifespan but 19.9% of females mated twice when one female kept with one male during scotophase. There were no significant differences in the fecundity, fertility and longevity between the single- and twice-mated females. Correlation coefficient between copulation duration and female mating times was not significant ( r  = 0.0860, P = 0.8575). Results suggested that DBM females may be monandrous. Multiple mating did not increase male or female mating fitness.  相似文献

Previous research showed that exposure to ginger root, Zingiber officinale Roscoe, oil increased the mating success of mass-reared, sterile males of the Mediterranean fruit fly, Ceratitis capitata (Wiedemann). This work, however, involved the exposure of small groups of males (n = 25) in small containers (volume 400 ml). Several sterile male release programs use plastic adult rearing containers (so-called PARC boxes; hereafter termed storage boxes; 0.48 by 0.60 by 0.33 m) to hold mature pupae and newly emerged adults before release (approximately = 36,000 flies per box). The objective of the current study was to determine whether the application of ginger root oil to individual storage boxes increases the mating competitiveness of sterile C. capitata males. Irradiated pupae were placed in storage boxes 2 d before adult emergence, and in the initial experiment (adult exposure) ginger root oil was applied 5 d later (i.e., 3 d after peak adult emergence) for 24 h at doses of 0.0625, 0.25, 0.5, 1.0, and 2.0 ml. In a second experiment (pupal-adult exposure), ginger root oil was applied to storage boxes immediately after pupal placement and left for 6 d (i.e., 4 d after peak adult emergence) at doses of 0.25 and 1.0 ml. Using field cages, we conducted mating trials in which ginger root oil-exposed (treated) or nonexposed (control) sterile males competed against wild-like males for copulations with wild-like females. After adult exposure, treated males had significantly higher mating success than control males for all doses of ginger root oil, except 2.0 ml. After pupal-adult exposure, treated males had a significantly higher mating success than control males for the 1.0-ml but not the 0.25-ml dose of ginger root oil. The results suggest that ginger root oil can be used in conjunction with prerelease, storage boxes to increase the effectiveness of sterile insect release programs.  相似文献

Twenty isolates of the fungus Metarhizium anisopliae (Metsch). Sorkin (Ma) were evaluated to determine their virulence against last instar and adult emergence of Mexican fruit fly, Anastrepha ludens (Loew). Larvae were exposed by immersion in a conidial suspension at a concentration of 10(8) UFC/ml under laboratory conditions. Larvae and pupae cumulative mortality rates ranged from 37.9 to 98.75%. Thirteen isolates caused mortality rates > 83.7%, and their LT50 values ranged from 1.8 to 6.2 d. The Ma2, Ma8, and Ma16 isolates were evaluated at seven different concentrations ranging from 10(1) to 10(7) UFC/ml, showing LC50 values from 3.7 to 4.8 x 10(5) UFC/ml. In a field-cage experiment, 200 ml of a conidial suspension of Ma2, at a concentration of 2.5 x 10(6) UFC/ml, was applied on 2,500 cm2 soil surface (2 x 10(5) UFC/cm2). The fungus reduced adult emergence, 22% fewer adults emerging in a sandy loam soil, and 43% fewer in loam soil, compared with the controls. M. anisopliae may offer a preferable alternative to chemicals as a biological control agent against A. ludens.  相似文献

Mating more than once is extremely costly for females in many species, making the near ubiquity of polyandry difficult to understand. However, evidence of mating costs for males is much rarer. We investigated the effects of copulation on longevity of male and female flies (Saltella sphondylli). We also scrutinized potential fecundity and fertility benefits to females with differing mating history. Copulation per se was found to decrease the longevity of males but not that of females. However, when females were allowed to lay eggs, females that mated died earlier than virgin females, indicating costs of egg production and/or oviposition. Thus, although longevity costs of copulation are higher for males, reproduction is nevertheless costly for females. We also found no differences in fecundity or fertility relative to female mating history. Results suggest that polyandry may be driven by minor costs rather than by major benefits in this species.  相似文献

The potential of Hirsutella thompsonii Fisher and Metarhizium anisopliae (Metschinkoff) as biological control agents of the parasitic mite, Varroa destructor Anderson and Trueman was evaluated in the laboratory and in observation hives. In the laboratory, time required for 90% cumulative mortality of mites (LT(90)) was 4.16 (3.98-4.42) days for H. thompsonii and 5.85 (5.48-7.43) days for M. anisopliae at 1.1 x 10(3) conidia mm(-2). At a temperature (34+/-1 degrees C) similar to that of the broodnest in a honey bee colony, Apis mellifera L., H. thompsonii [LC(90)=9.90 x 10(1) (5.86-19.35) conidia mm(-2) at Day 7] and M. anisopliae [LC(90)=7.13 x 10(3) (2.80-23.45) conidia mm(-2) at Day 7] both showed significant virulence against V. destructor. The applications of H. thompsonii to observation hives resulted in significant mortality of mites, and reduction of the number of mites per bee 21 and 42 days post-treatments. The treatments did not significantly affect the mite population in sealed brood. However, the fungus must have persisted because infected mites were still observed [82.97+/-(0.6)%] 42 days post-treatment. In addition, the fungus was found to sporulate on the host. A small percentage [2.86+/-(0.2)%] of dead mites found in the control hives also showed fungal infection, suggesting that adult bees drifted between hives and disseminated the fungus. H. thompsonii was harmless to the honey bees at the concentrations applied and did not have any deleterious effects on the fecundity of the queens. Microbial control with fungal pathogens provides promising new avenues for control of V. destructor and could be a useful component of an integrated pest management program for the honey bee industry.  相似文献

用“孢子浴”法将人工培养的安徽虫瘟霉(Zophthoraanhuiensis)对桃蚜(Myzuspersi cae)接种而进行定量生物测定(18℃,光周期为L∶D12∶12),包括10个剂量(1.5~197.7个分生孢子/mm2),每剂量处理蚜虫64~120头,逐日观察记载死亡数至第7d。接种后第3d在高剂量处理中始见少量死蚜,第4~6d为死蚜盛期。所获数据很好地拟合时间 剂量 死亡率模型,由模型参数估计出该菌作用于桃蚜的时间效应随剂量增大而减小,在37.1~197.7个分生孢子/mm2的剂量范围内LT50值为4.5~6.7d;剂量效应在接种后随时间递减,第5~7d的LD50分别为86.8、43.7和34.1个分生孢子/mm2。结果表明,安徽虫瘟霉是一种较为理想的杀蚜微生物,在寄主体内的潜伏期为4~5d。  相似文献

2005年3月至2006年3月,采取所有事件取样法,对成都动物园5只(2雄,3雌)圈养金钱豹(Panthera pardu)进行观察,旨在了解圈养金钱豹的交配情况。记录交配行为1 174次。结果显示,圈养金钱豹全年皆可发情。具有明显的交配模式,交配姿势仅有一种,为背腹式。平均交配持续天数为(4.75±1.26)d。昼夜都有交配行为,但白天交配次数较夜间多。日交配的高峰发生在08:00~10:00时,不同的雄性个体出现的交配高峰日不同。平均交配持续时间为(7.48±1.22)s。在交配持续时间(P=0.000)、总交配次数(P=0.04)上,不同的雄性个体间存在显著性差异;而同一雄性在与不同雌性交配时,其持续时间无显著差异。金钱豹交配的特点为,交配的频次多,但每次交配持续时间短。  相似文献

草地贪夜蛾Spodoptera frugiperda(J.E Smith)是我国新发现的一种重要外来入侵害虫。为了寻找对草地贪夜蛾有致病性的昆虫病原真菌,本实验采用浸虫法研究了玫烟色虫草Cordyceps(=Isaria)fumosorosea SCAU-IFCF01对草地贪夜蛾1~6龄幼虫的致病力,通过体视镜观察了幼虫感菌后侵染症状和体表病变过程。结果表明:菌株IFCF01可侵染草地贪夜蛾6个龄期的幼虫。4龄幼虫接种后48 h,虫体出现缩短、强直等形态变化;接种后72 h,幼虫死亡且体外形成菌丝层;接种后120 h,虫体被浅玫烟色的分生孢子覆盖。随孢子浓度的升高,幼虫的感病死亡率增加,当浓度达到1×109孢子/mL时,1~3龄幼虫的累计死亡率均为100%,4龄幼虫和5龄幼虫也达到了98.9%和50%,6龄幼虫仅21.1%。5 d后1~5龄幼虫的LC 50值分别为3.74×104、1.17×105、1.86×105、8.09×105和3.03×108孢子/mL。幼虫LT 50值随孢子浓度增加而递减,在浓度为1.0×105~1.0×109孢子/mL的范围内,1龄幼虫和2龄幼虫的LT 50值分别为3.62~1.37 d和4.51~1.65 d;在1.0×106~1.0×109孢子/mL时,对3龄幼虫的LT 50为4.03~1.82 d;浓度为1.0×107~1.0×109孢子/mL时,对4龄幼虫的LT 50为3.47~2.52 d。浓度为1.0×109孢子/mL时,5龄幼虫的LT 50为4.74 d。本研究结果表明,玫烟色虫草菌株IFCF01对草地贪夜蛾幼虫具有较强的致病力,为今后草地贪夜蛾微生物防治提供重要的有效真菌。  相似文献

Abstract.  Adult diet is an important determinant of sexual activity in many tephritid fruit flies. Whether availability of protein (hydrolysed yeast) in addition to sucrose influences sexual activity or longevity of male and female Queensland fruit flies ( Bactrocera tryoni Froggatt, 'Q-flies'), and whether irradiation of flies as pupae modifies their dietary needs, is investigated. Previous studies on groups of flies suggest that protein is required for sexual maturation of females but not males. By contrast, this study of individual flies demonstrates that protein in the adult diet provides a massive boost to sexual activity of both males and females. Mating probability increases with age from 4-14 days as the flies began to mature. However, mating probability reaches much higher levels when the flies are provided with protein. Although males and females mate at similar rates when provided with protein, females suffer a greater reduction in mating probability than males when deprived of protein. In addition to increased mating probability, access to dietary protein is also associated with reduced latency from onset of dusk until copulation. Furthermore, young male flies with access to dietary protein have longer copula duration than males fed only sucrose. Irradiation of flies as pupae has no apparent effect on mating probability, the latency to copulate or copula duration. However, when deprived of protein, sterile flies (especially males) suffer a greater reduction in longevity compared with fertile flies. Overall, access to dietary protein increases longevity for both males and females, although females live longer than males on both diets. These findings suggest that prerelease provision of dietary protein has the potential to greatly enhance the efficacy of Q-flies used in the sterile insect technique.  相似文献

Second and third instar Chilo partellus larvae were infected with Beauveria bassiana and Metarhizium anisopliae (both at 1x10(8)conidia/ml) and daily consumption of maize leaves was measured. Infection by the fungi was associated with reduced mean daily food consumption. Reduction in food consumption became evident 3-4 days after treatment with the fungi for second instar larvae and 4-5 days for third instar larvae. Four conidial concentrations, 1x10(5), 1x10(6), 1x10(7), and 1x10(8)conidia/ml, were tested against second instar larvae. Food consumption dropped by 70-85% when the second instar larvae were treated with the fungi at 1x10(8)conidia/ml. Reduction in food consumption by C. partellus larvae infected with B. bassiana and M. anisopliae may offset the slow speed of kill of the fungi. The effect of artificial versus natural diets on mortality and mycoses of second instar larvae treated with the fungi at 1x10(8)conidia/ml was determined. Larvae provided with artificial diet suffered little mortality and mycoses than larvae provided with maize leaves. The LT(50) was longer for larvae provided with artificial diet.  相似文献

Abstract The effects of delayed mating on the copulation duration, female fertility, fecundity, egg fertility, longevity and the number days alive after mating of females of diamondback moth (DBM), Plutella xylostella, were studied. When male mating was delayed, the female fertility, fecundity, egg fertility, longevity and number days alive after mating of DBM decreased, and there was a negative correlation between the age of the moth with those variables except copulation duration. When female mating was delayed, the female fertility, fecundity, percent egg fertility and number days alive after mating of DBM also decreased, but the longevity increased, which also showed a negative relationship between the age of the moth with the variables except copulation duration and longevity. When both males and females delayed mating, the female fertility and fecundity decreased; egg fertility was affected marginally, and the longevity of females increased. The moth age was negatively correlated with those variables.  相似文献

Background  

In a number of species males damage females during copulation, but the reasons for this remain unclear. It may be that males are trying to manipulate female mating behaviour or their life histories. Alternatively, damage may be a side-effect of male-male competition. In the black scavenger or dung fly Sepsis cynipsea (Diptera: Sepsidae) mating reduces female survival, apparently because males wound females during copulation. However, this damage does not seem to relate to attempted manipulation of female reproduction by males. Here we tested the hypothesis that harming females during mating is an incidental by-product of characters favoured during pre-copulatory male-male competition. We assessed whether males and their sons vary genetically in their ability to obtain matings and harm females, and whether more successful males were also more damaging. We did this by ranking males' mating success in paired competitions across several females whose longevity under starvation was subsequently measured.  相似文献

We determined the temporal pattern of female remating in the Mediterranean fruit fly, Ceratitis capitata, and how mating with sterile males affects remating. In addition, we examined the hypotheses that sterile male nutrition and age affect the subsequent receptivity of their mates. Temporally, female receptivity varied significantly throughout the experimental period. Relatively high levels of remating (14%) on the days following the first copulation were followed by a decline, with a significantly low point (4.1%) 2 weeks after mating. Subsequently, receptivity is gradually restored (18%) 3 and 4 weeks after the initial copulation. When females were first mated to sterile males, significantly higher remating percentages were recorded. The ability of sterile males to inhibit receptivity of both wild and laboratory reared females on the day of first mating was significantly improved when they were fed a nutrient rich diet. Male age at first mating also affected female receptivity: sterile males of intermediate age (11 days old) inhibited female remating significantly more than younger or older flies. Although further studies are needed to determine the relative roles of natural and sexual selection in modulating patterns of female sexual receptivity, the Sterile Insect Technique may be improved by releasing well nourished, older sterile males.  相似文献
