The denitrification rates in a marine sediment, estimated by using 15N-nitrate, Vmax, Km, and sediment nitrate concentrations, were 12.5 and 2.0 nmol of N2-N cm−3 day−1 at 0 to 1 and 1 to 3 cm, respectively, at 12°C. The total rate was 165 nmol of N2-N m−2 day−1.  相似文献

A method was developed to follow bacterial nitrate reduction in freshwater sediments by using common high-performance liquid chromatographic equipment. The low detection limit (14 pmol) of the method enabled us to study concentration profiles and reaction kinetics under natural conditions. Significant nitrate concentrations (1 to 27 μM) were observed in the sediment of Lake Vechten during the nonstratified period; the concentration profiles showed a successive depletion of oxygen, nitrate, and sulfate with depth. The profiles were restricted to the upper 3 cm of the sediment which is rich in organics and loosely structured. Nitrate reduction in the sediment-water interface followed first-order reaction kinetics at in situ concentrations. Remarkably high potential nitrate-reducing activity was observed in the part of the sediment in which nitrate did not diffuse. This activity was also observed throughout the whole year. Estimates of Km varied between 17 and 100 μM and Vmax varied between 7.2 and 36 μmol cm−3 day−1 for samples taken at different depths. The diffusion coefficient of nitrate ([10 ± 0.4] × 10−6 cm2 s−1) across the sediment-water interface was estimated by a constant-source technique and applied to a mathematical model to estimate the net nitrate reduction during the nonstratified period. In this period, observed nitrate reduction rates by the model, 0.2 to 0.4 mmol m−2 day−1, were lower than those found for oxygen (27 mmol m−2 day−1) and sulfate (0.4 mmol m−2 day−1). During the summer stratification, nitrate was absent in the sediment and reduction could not be estimated by the model.  相似文献

Nitrate and nitrite concentrations in the water and nitrous oxide and nitrite fluxes across the sediment-water interface were measured monthly in the River Colne estuary, England, from December 1996 to March 1998. Water column concentrations of N2O in the Colne were supersaturated with respect to air, indicating that the estuary was a source of N2O for the atmosphere. At the freshwater end of the estuary, nitrous oxide effluxes from the sediment were closely correlated with the nitrite concentrations in the overlying water and with the nitrite influx into the sediment. Increases in N2O production from sediments were about 10 times greater with the addition of nitrite than with the addition of nitrate. Rates of denitrification were stimulated to a larger extent by enhanced nitrite than by nitrate concentrations. At 550 μM nitrite or nitrate (the highest concentration used), the rates of denitrification were 600 μmol N · m−2 · h−1 with nitrite but only 180 μmol N · m−2 · h−1 with nitrate. The ratios of rates of nitrous oxide production and denitrification (N2O/N2 × 100) were significantly higher with the addition of nitrite (7 to 13% of denitrification) than with nitrate (2 to 4% of denitrification). The results suggested that in addition to anaerobic bacteria, which possess the complete denitrification pathway for N2 formation in the estuarine sediments, there may be two other groups of bacteria: nitrite denitrifiers, which reduce nitrite to N2 via N2O, and obligate nitrite-denitrifying bacteria, which reduce nitrite to N2O as the end product. Consideration of free-energy changes during N2O formation led to the conclusion that N2O formation using nitrite as the electron acceptor is favored in the Colne estuary and may be a critical factor regulating the formation of N2O in high-nutrient-load estuaries.  相似文献

Denitrifying activity in a sediment from the freshwater part of a polluted estuary in northwest Europe was quantified using two independent approaches. High-resolution N2O microprofiles were recorded in sediment cores to which acetylene was added to the overlying water and injected laterally into the sediment. The vertical distribution of the rate of denitrification supported by nitrate uptake from the overlying water was then derived from the time series N2O concentration profiles. The rates obtained for the core incubations were compared to the rates predicted by a forward reactive transport model, which included rate expression for denitrification calibrated with potential rate measurements obtained in flowthrough reactors containing undisturbed, 1-cm-thick sediment slices. The two approaches yielded comparable rate profiles, with a near-surface, 2- to 3-mm narrow zone of denitrification and maximum in situ rates on the order of 200 to 300 nmol cm−3 h−1. The maximum in situ rates were about twofold lower than the maximum potential rate for the 0- to 1-cm depth interval of the sediment, indicating that in situ denitrification was nitrate limited. The experimentally and model-derived rates of denitrification implied that there was nitrate uptake by the sediment at a rate that was on the order of 50 (± 10) nmol cm−2 h−1, which agreed well with direct nitrate flux measurements for core incubations. Reactive transport model calculations showed that benthic uptake of nitrate at the site is particularly sensitive to the nitrate concentration in the overlying water and the maximum potential rate of denitrification in the sediment.  相似文献

We estimated rates of denitrification at various depths in sediments known to be affected by submarine discharge of groundwater, and also in the parent aquifer. Surface denitrification was only measured in the autumn; at 40-cm depth, where groundwater-imported nitrate has been measured, denitrification occurred consistently throughout the year, at rates from 0.14 to 2.8 ng-atom of N g−1 day−1. Denitrification consistently occurred below the zone of sulfate reduction and was sometimes comparable to it in magnitude. Denitrification occurred deep (14 to 40 cm) in the sediments along 30 km of shoreline, with highest rates occurring where groundwater input was greatest. Denitrification rates decreased with distance offshore, as does groundwater influx. Added glucose greatly stimulated denitrification at depth, but added nitrate did not. High rates of denitrification were measured in the aquifer (17 ng-atom of N g−1 day−1), and added nitrate did stimulate denitrification there. The denitrification measured was enough to remove 46% of the nitrate decrease observed between 40- and 14-cm depth in the sediment.  相似文献

NO3 concentration profiles were measured in the sediments of a meso-eutrophic lake with a newly developed microelectrode. The depth of penetration of NO3 varied from only 1.3 mm in organic-rich profundal silty sediments to 5 mm in organic-poor littoral sandy sediments. The thickness of the zone of denitrification in the organic-rich sediments was <500 μm. Oxygen profiles measured simultaneously revealed that the zone of denitrification was directly adjacent to the aerobic zone. The results demonstrate high denitrification rates (0.26 to 1.31 mmol m−2 day−1) at in situ nitrate concentrations in the overlying water (0.030 mmol liter−1) and limitation of denitrification by nitrate availability.  相似文献

An investigation of the terminal anaerobic processes occurring in polluted intertidal sediments indicated that terminal carbon flow was mainly mediated by sulfate-reducing organisms in sediments with high sulfate concentrations (>10 mM in the interstitial water) exposed to low loadings of nutrient (equivalent to <102 kg of N · day−1) and biochemical oxygen demand (<0.7 × 103 kg · day−1) in effluents from different pollution sources. However, in sediments exposed to high loadings of nutrient (>102 kg of N · day−1) and biochemical oxygen demand (>0.7 × 103 kg · day−1), methanogenesis was the major process in the mediation of terminal carbon flow, and sulfate concentrations were low (≤2 mM). The respiratory index [14CO2/(14CO2 + 14CH4)] for [2-14C]acetate catabolism, a measure of terminal carbon flow, was ≥0.96 for sediment with high sulfate, but in sediments with sulfate as little as 10 μM in the interstitial water, respiratory index values of ≤0.22 were obtained. In the latter sediment, methane production rates as high as 3 μmol · g−1 (dry weight) · h−1 were obtained, and there was a potential for active sulfate reduction.  相似文献

The effects of three metabolic inhibitors (acetylene, methanol, and allylthiourea [ATU]) on the pathways of N2 production were investigated by using short anoxic incubations of marine sediment with a 15N isotope technique. Acetylene inhibited ammonium oxidation through the anammox pathway as the oxidation rate decreased exponentially with increasing acetylene concentration; the rate decay constant was 0.10 ± 0.02 μM−1, and there was 95% inhibition at ~30 μM. Nitrous oxide reduction, the final step of denitrification, was not sensitive to acetylene concentrations below 10 μM. However, nitrous oxide reduction was inhibited by higher concentrations, and the sensitivity was approximately one-half the sensitivity of anammox (decay constant, 0.049 ± 0.004 μM−1; 95% inhibition at ~70 μM). Methanol specifically inhibited anammox with a decay constant of 0.79 ± 0.12 mM−1, and thus 3 to 4 mM methanol was required for nearly complete inhibition. This level of methanol stimulated denitrification by ~50%. ATU did not have marked effects on the rates of anammox and denitrification. The profile of inhibitor effects on anammox agreed with the results of studies of the process in wastewater bioreactors, which confirmed the similarity between the anammox bacteria in bioreactors and natural environments. Acetylene and methanol can be used to separate anammox and denitrification, but the effects of these compounds on nitrification limits their use in studies of these processes in systems where nitrification is an important source of nitrate. The observed differential effects of acetylene and methanol on anammox and denitrification support our current understanding of the two main pathways of N2 production in marine sediments and the use of 15N isotope methods for their quantification.  相似文献

We measured denitrification and nitrate removal rates in cold seep sediments from the Gulf of Mexico. Heterotrophic potential denitrification rates were assayed in time-series incubations. Surficial sediments inhabited by Beggiatoa exhibited higher heterotrophic potential denitrification rates (32 μ N reduced day−1) than did deeper sediments (11 μ N reduced day−1). Nitrate removal rates were high in both sediment horizons. These nitrate removal rates translate into rapid turnover times (<1 day) for the nitrate pool, resulting in a faster turnover for the nitrate pool than for the sulfate pool. Together, these data underscore the rigorous nature of internal nitrogen cycling at cold seeps and the requirement for novel mechanisms to provide nitrate to the sediment microbial community.  相似文献

Diel and seasonal variations in denitrification were determined in a littoral lake sediment colonized by the perennial macrophyte Littorella uniflora (L.) Aschers. In the winter, the activity was low (5 μmol of N m−2 h−1) and was restricted to the uppermost debris layer at a depth of 0 to 1 cm. By midsummer, the activity increased to 50 μmol of N m−2 h−1 and was found throughout the root zone to a depth of 10 cm. The root zone accounted for as much as 50 to 70% of the annual denitrification. A significant release of organic substrates from the roots seemed to determine the high activities of root zone denitrification in the summer. The denitrification in the surface layer and in the root zone formed two distinct activity zones in the summer, when the root zone also contained nitrification activity, as indicated from the accumulations of NO3. Light conditions inhibited denitrification in both the surface layer and the upper part of the root zone, suggesting that a release of O2 by benthic algae and from the roots of L. uniflora controlled a diel variation of denitrification. In midsummer, the rate of denitrification in both the surface layer and the upper part of the root zone was limited by NO3. In the growth season, there was evidence for a significant population of denitrifiers closely associated with the root surface.  相似文献

An acetylene inhibition method was satisfactorily used for the in situ measurement of denitrification in two sediment-water systems incubated for not more than 22 h. In the presence of added nitrate, denitrification acted as a source of nitrous oxide in a drainage pond, but acted as a sink in its absence. The averaged rates of nitrous oxide accumulation with nitrate enrichment in the absence and presence of acetylene were 0.15 and 0.30 mg of N m−2h−1, respectively. Acetylene reduction at an average rate of 0.07 mmol of C2H4 formed m−2h−1 was simultaneously measured in the absence of added nitrate. In a small eutrophic lake where nitrogen was nonlimiting, the in situ rates of sediment denitrification were 0.09 and 0.11 mg of N m−2h−1 in the presence and absence of macrophytes, respectively, and no acetylene reduction activity was found.  相似文献

Ryan Lake, a 1.6-hectare basin lake near the periphery of the tree blowdown area in the blast zone 19 km north of Mount St. Helens, was studied from August to October 1980 to determine the microbial and chemical response of the lake to the eruption. Nutrient enrichment through the addition of fresh volcanic material and the organic debris from the surrounding conifer forest stimulated intense microbial activity. Concentrations of such nutrients as phosphorus, sulfur, manganese, iron, and dissolved organic carbon were markedly elevated. Nitrogen cycle activity was especially important to the lake ecosystem in regulating biogeochemical cycling owing to the limiting abundance of nitrogen compounds. Nitrogen fixation, both aerobic and anaerobic, was active from aerobic benthic and planktonic cyanobacteria with rates up to 210 nmol of N2 cm−1 h−1 and 667 nmol of N2 liter−1 h−1, respectively, and from anaerobic bacteria with rates reaching 220 nmol of N2 liter−1 h−1. Nitrification was limited to the aerobic epilimnion and littoral zones where rates were 43 and 261 nmol of NO2 liter−1 day−1, respectively. Potential denitrification rates were as high as 30 μmol of N2O liter−1 day−1 in the anaerobic hypolimnion. Total bacterial numbers ranged from 1 × 106 to 3 × 108 ml−1 with the number of viable sulfur-metal-oxidizing bacteria reaching 2 × 106 ml−1 in the hypolimnion. A general scenario for the microbial cycling of nitrogen, carbon, sulfur, and metals is presented for volcanically impacted lakes. The important role of nitrogen as these lakes recover from the cataclysmic eruption and proceed back towards their prior status as oligotrophic alpine lakes is emphasized.  相似文献

The intramolecular distribution of nitrogen isotopes in N2O is an emerging tool for defining the relative importance of microbial sources of this greenhouse gas. The application of intramolecular isotopic distributions to evaluate the origins of N2O, however, requires a foundation in laboratory experiments in which individual production pathways can be isolated. Here we evaluate the site preferences of N2O produced during hydroxylamine oxidation by ammonia oxidizers and by a methanotroph, ammonia oxidation by a nitrifier, nitrite reduction during nitrifier denitrification, and nitrate and nitrite reduction by denitrifiers. The site preferences produced during hydroxylamine oxidation were 33.5 ± 1.2‰, 32.5 ± 0.6‰, and 35.6 ± 1.4‰ for Nitrosomonas europaea, Nitrosospira multiformis, and Methylosinus trichosporium, respectively, indicating similar site preferences for methane and ammonia oxidizers. The site preference of N2O from ammonia oxidation by N. europaea (31.4 ± 4.2‰) was similar to that produced during hydroxylamine oxidation (33.5 ± 1.2‰) and distinct from that produced during nitrifier denitrification by N. multiformis (0.1 ± 1.7‰), indicating that isotopomers differentiate between nitrification and nitrifier denitrification. The site preferences of N2O produced during nitrite reduction by the denitrifiers Pseudomonas chlororaphis and Pseudomonas aureofaciens (−0.6 ± 1.9‰ and −0.5 ± 1.9‰, respectively) were similar to those during nitrate reduction (−0.5 ± 1.9‰ and −0.5 ± 0.6‰, respectively), indicating no influence of either substrate on site preference. Site preferences of ~33‰ and ~0‰ are characteristic of nitrification and denitrification, respectively, and provide a basis to quantitatively apportion N2O.  相似文献

Microbial sulfate reduction rates in acidic peat from a New Jersey Pine Barrens cedar swamp in 1986 were similar to sulfate reduction rates in freshwater lake sediments. The rates ranged from a low of 1.0 nmol cm−3 day−1 in February at 7.5- to 10.0-cm depth to 173.4 nmol cm−3 day−1 in July at 5.0- to 7.5-cm depth. The presence of living Sphagnum moss at the surface generally resulted in reduced rates of sulfate reduction. Pore water sulfate concentrations and water table height also apparently affected the sulfate reduction rate. Concentrations of sulfate in pore water were nearly always higher than those in surface water and groundwater, ranging from 26 to 522 μM. The elevated pore water sulfate levels did not result from the evapotranspiratory concentration of infiltrating stream water or groundwater, but probably resulted from oxidation of reduced sulfur compounds, hydrolysis of ester sulfates present in the peat, or both. The total sulfur content of peat that had no living moss at the surface was 164.64 ± 1.5 and 195.8 ± 21.7 μmol g (dry weight)−1 for peat collected from 2.5 to 5.0 and 7.5 to 10.0 cm, respectively. Organosulfur compounds accounted for 84 to 88% of the total sulfur that was present in the peat. C-bonded sulfur accounted for 91 to 94% of the organic sulfur, with ester sulfate being only a minor constituent. Reduced inorganic sulfur species in peat from 2.5 to 7.5 cm were dominated by H2S-FeS (68%), while pyritic sulfide was the predominant inorganic sulfur species in the peat from depths of 7.5 to 10.0 cm (75%).  相似文献

High-resolution NO3 profiles in freshwater sediment covered with benthic diatoms were obtained with a new microscale NO3 biosensor characterized by absence of interference from chemical species other than NO2 and N2O. Analysis of the microprofiles obtained indicated no nitrification during darkness, high rates of nitrification and a tight coupling between nitrification and denitrification during illumination, and substantial rates of NO3 assimilation during illumination. Nitrification during darkness could be induced by purging the bulk water with O2 gas, indicating that the stimulatory effect on nitrification by illumination was caused by algal production of O2. NH4+ addition did not stimulate nitrification during darkness when O2 was restricted to the upper 1-mm layer, and there was thus a low nitrification potential in the permanently oxic top 1 mm of the sediment.  相似文献

The relationships between various carbohydrate pools of the soybean (Glycine max [L.] Merrill) fruit and growth rate of seeds were evaluated. Plants during midpod-fill were subjected to various CO2 concentrations or light intensities for 7 days to generate different rates of seed growth. Dry matter accumulation rates of seeds and pod wall, along with glucose, sucrose, and starch concentrations in the pod wall, seed coat, and embryo were measured in three-seeded fruits located from nodes six through ten. Seed growth rates ranged from 4 to 37 milligrams·day−1·fruit−1. When seed growth rates were greater than 12 milligrams·day−1·fruit−1, sucrose concentration remained relatively constant in the pod wall (1.5 milligrams·100 milligrams dry weight−1), seed coat (8.5 milligrams·100 milligrams dry weight−1), and embryo (5.0 milligrams·100 milligrams dry weight−1). However, sucrose concentrations decreased in all three parts of the fruit as growth rate of the seeds fell below 12 milligrams·day−1·fruit−1. This relationship suggests that at high seed growth rates, flux of sucrose through the sucrose pools of the fruit was more important than pool size for growth. Starch concentration in the pod wall remained relatively constant (2 milligrams·100 milligrams dry weight−1) at higher rates of seed growth but decreased as seed growth rates fell below 12 milligrams·day−1·fruit−1. This suggests that pod wall starch may buffer seed growth under conditions of limiting assimilate availability. There was no indication that carbohydrate pools of the fruit were a limitation to transport or growth processes of the soybean fruit.  相似文献

Potential rates of nitrification and denitrification were measured in an oligotrophic sediment system. Nitrification potential was estimated using the CO oxidation technique, and potential denitrification was measured by the acetylene blockage technique. The sediments demonstrated both nitrifying and denitrifying activity. Eh, O2, and organic C profiles showed two distinct types of sediment. One type was low in organic C, had high O2 and Eh, and had rates of denitrification 1,000 times lower than the other which had high organic C, low O2, and low Eh. Potential nitrification and denitrification rates were negatively correlated with Eh. This suggests that environmental heterogeneity in denitrifier and nitrifier populations in oligotrophic sediment systems may be assessed using Eh before sampling protocols for nitrification or denitrification rates are established. There was no correlation between denitrification and nitrification rates or between either of these processes and NH4 + or NO3 concentrations. The maximum rate of denitrification was 0.969 nmole N cm–3 hour–1, and the maximum rate of nitrification was 23.6 nmole cm–3 hour–1, suggesting nitrification does not limit denitrification in these oligotrophic sediments. Some sediment cores had mean concentrations of 6.0 mg O2/liter and still showed both nitrification and denitrification activity.  相似文献

Methane concentrations and rates of methane oxidation were measured in intact sediment cores from an inshore marine sediment at Jutland, Denmark. The rates of methane oxidation, determined by the appearance of 14CO2 from injected 14CH4, varied with sediment depth and season. Most methane oxidation was anoxic, but oxygen may have contributed to methane oxidation at the sediment surface. Cumulative rates (0- to 12-cm depth) for methane oxidation at Kysing Fjord were 3.34, 3.48, 8.60, and 17.04 μmol m−2 day−1 for April (4°C), May (13°C), July (17°C), and August (21°C), respectively. If all of the methane was oxidized by sulfate, it would account for only 0.01 to 0.06% of the sulfate reduction. The data indicate that methane was produced, in addition to being oxidized, in the 0- to 18-cm sediment stratum.  相似文献

The acetylene block technique was employed to study denitrification in intertidal estuarine sediments. Addition of nitrate to sediment slurries stimulated denitrification. During the dry season, sediment-slurry denitrification rates displayed Michaelis-Menten kinetics, and ambient NO3 + NO2 concentrations (≤26 μM) were below the apparent Km (50 μM) for nitrate. During the rainy season, when ambient NO3 + NO2 concentrations were higher (37 to 89 μM), an accurate estimate of the Km could not be obtained. Endogenous denitrification activity was confined to the upper 3 cm of the sediment column. However, the addition of nitrate to deeper sediments demonstrated immediate N2O production, and potential activity existed at all depths sampled (the deepest was 15 cm). Loss of N2O in the presence of C2H2 was sometimes observed during these short-term sediment incubations. Experiments with sediment slurries and washed cell suspensions of a marine pseudomonad confirmed that this N2O loss was caused by incomplete blockage of N2O reductase by C2H2 at low nitrate concentrations. Areal estimates of denitrification (in the absence of added nitrate) ranged from 0.8 to 1.2 μmol of N2 m−2 h−1 (for undisturbed sediments) to 17 to 280 μmol of N2 m−2 h−1 (for shaken sediment slurries).  相似文献

One year after impoundment in January 1994, methanotrophic bacteria in Petit Saut Reservoir (French Guiana) were active at the oxic-anoxic interface. This activity was revealed by the sudden extinction of diffusive methane emission (600 metric tons of CH4 · day−1 for the whole lake surface area, i.e., 360 km2). Lifting of inhibition was suspected. After reviewing the potential inhibitors of this physiological guild (O2, NH4+, sulfides) and considering the similarities with nitrifiers, we suggest that sunlight influenced the methanotrophic bacteria. On the basis of phospholipid analysis, only a type II methanotrophic community was identified in the lake. Both growth and methanotrophic activity of an enriched culture, obtained in the laboratory, were largely inhibited by illumination over 150 microeinsteins · m−2 · s−1. These results were confirmed on a pure culture of Methylosinus trichosporium OB3B. In situ conditions showed that water transparency was quite stable in 1994 and 1995 and that the oxycline moved steadily deeper until January 1995. Considering the mean illumination profile during this period, we showed that removal of methanotrophic growth inhibition could only occur below a 2-m depth. The oxycline reached this level in October 1994, allowing methanotrophic bacteria to develop and to consume the entire methane emission 4 months later.  相似文献
