The antiradical properties of three samples of ginger (Zingiber officinale R.)—juice from fresh rhizome, essential oil, and extracts (oleoresin)—were studied and compared with the properties of synthetic antioxidant ionol (butylatedhydroxy-toluene, BHT). Reaction antioxidants with stable free 2,2-diphenyl-1-picrylhydrozyl radicals were used as model systems. DPPH equivalents per gram of ginger sample, EC50, and antiradical efficiency (AE) were determined. The EC50 and AE values for ginger oleoresin and BHT were similar. They were the same as those of highly active natural antioxidants, and the values for essential oil and ginger juice were lower by two orders of magnitude. On the base of kinetic parameters, the ginger samples may belong to antiradical compounds with prolonged action.  相似文献

In Brazil, there is a large diversity of species of small edible fruits that are considered sources of nutrients and functional properties. They present a high innovation domain for the pharmaceutical, cosmetic and food industries due to their health-promoting properties. Edible fruits from Brosimum gaudichaudii (Moraceae) are widely consumed and used in folk medicine and in feed by the population of the Brazilian Cerrado. Nevertheless, detailed information on the chemical fingerprint, antiradical activity and safety aspects of these fruits is still unknown. Thus, the aim of this work was to investigate the bioactive compounds of hydroethanolic extracts of fruits from Brosimum gaudichaudii using high-performance liquid chromatography combined with mass spectrometry using electrospray ionization (HPLC ESI-MS). Eighteen different compounds, including flavonoids, coumarins, arylbenzofurans, terpenoids, stilbenes, xanthones and esters, were detected. Moreover, the study indicated that the hydroethanolic extract of fruits from B. gaudichaudii presented low scavenging activity against 2,2-diphenyl-1-picrylhydrazyl radicals (IC50>800 μg mL−1) and was cytotoxic (IC50<30 μg mL−1) in Chinese hamster ovary cells (CHO−K1) by an in vitro assay. This is the first report of the chemical profile, antioxidant activity and cytotoxic properties of the hydroethanolic extract of fruits from B. gaudichaudii.  相似文献

Polyphenols are the predominant ingredients in apple seeds. However, few data are available on the phenolic profile or antioxidant activity in apple seeds in previous researches. In this study, low-molecular-weight phenolic compounds and antioxidant activity in seeds, peels, and flesh of seven apple cultivars grown in northwest China were measured and analyzed using HPLC and FRAP, DPPH, ABTS assays, respectively. HPLC analysis revealed phloridzin as the dominant phenolic compound in the seeds with its contents being 240.45–864.42 mg/100 gDW. Total phenolic content (TPC) measured by the Folin–Ciocalteu assay in apple seed extracts of seven cultivars ranged from 5.74 (Golden Delicious) to 17.44 (Honeycrisp) mgGAE/gDW. Apple seeds showed higher antioxidant activity than peels or flesh; antioxidant activity in seeds varied from 57.59 to 397.70 μM Trolox equivalents (TE)/g FW for FRAP, from 37.56 to 64.31 μM TE/g FW for DPPH, and from 220.52 to 708.02 μM TE/g FW for ABTS. TPC in apple seeds was significantly correlated with all three assays. Principal component analysis (PCA) indicated that Honeycrisp was characterized with high contents of total polyphenols and phloridzin. Our findings suggest that phenolic extracts from apple seeds have good commercial potential as a promising antioxidant for use in food or cosmetics.  相似文献

Antioxidant activity in α- and β-chitosan at a wide range of molecular weight (Mw) and chitosan concentration (CS) was determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, reducing ability, chelating ability, and hydroxyl radical scavenging activity. The form of chitosan (FC) had significant (P <0.05) effect on all measurements except DPPH radical scavenging activity, and antioxidant activity was dependent on Mw and CS. High Mw (280–300 kDa) of β-chitosan had extremely lower half maximal effective concentrations (EC50) than α-chitosan in DPPH radical scavenging activity and reducing ability. The 22–30 kDa of α- and β-chitosan showed significantly (P <0.05) higher activities in DPPH radical scavenging, reducing ability, and hydroxyl radical scavenging than samples at other Mw, while chelating ability was the highest in 4–5 kDa chitosan. CS had significant effect on all measurements and the effect was related to Mw. The antioxidant activity of 280–300 kDa chitosan was affected by coil-overlap concentrations (C1) in the CS range of 4–10 mg/mL, forming entanglements. Reducing ability and hydroxyl radical scavenging activity were more predominant action in antioxidant activity of chitosan as shown by the lower EC50 values than those in other antioxidant measurements.  相似文献

Leaf extracts of Bryophyllum pinnatum (BPEs) are used in several countries. Contextually, evaluation of the therapeutic potential of these was carried out in this study to explore antioxidant and antityrosinase potential through different in vitro methods. The radical scavenging properties of BPEs were studied using various techniques, based on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) dot blot thin-layer chromatography (TLC) method, electron paramagnetic resonance (EPR) spectroscopy, metal chelation, β-carotene bleaching, inhibition of DNA breakage on agarose gel, and lipid peroxidation inhibition using liver and brain microsomes. EC50 values of the results reflected that aqueous-methanolic BPE was the most active one. Antibrowning potential of the fresh leaf extract showed an antityrosinase property, with EC50 values of enzymatic assay of tyrosinase inhibitory activity further advocating the findings.  相似文献

Chitosans were prepared by H2O2 oxidative depolymerization from squid pens with low molecular weights (LMW) of 13,025, 7011, 4169, 2242 and 963 Da. The bile acid binding capacities and antioxidant properties of LMW chitosans were studied in vitro. LMW chitosans exhibited stronger bile acid binding capacities than that of chitosan. The scavenging ability of LMW chitosans against DPPH radicals improved with increasing concentration, and EC50 values were below 1.3 mg/mL. The EC50 values of LMW chitosans against hydroxyl radicals ranged from 0.93 to 3.66 mg/mL. All LMW chitosans exhibited a strong ferrous ion chelating effect and reducing power. At 1 mg/mL, the scavenging ability of chitosan-963 towards superoxide radicals was 67.76%. These results indicated that LMW chitosans which have stronger bile acid binding capacity and antioxidant activities may act as potential antioxidants in vitro.  相似文献

Extracts from 48 marine macroalgae species (17 Chlorophyta, 8 Phaeophyta and 23 Rhodophyta) from the coasts of Yucatan and Quintana Roo (Mexico) were evaluated for antioxidant activity. The antioxidant activity was measured with the DPPH (2,2-diphenyl-1-picrylhydrasyl) method, and the phenolic content of each extract were also evaluated. All species exhibited a DPPH radical scavenging activity, and three species (Avrainvillea longicaulis, Chondria baileyana and Lobophora variegata) demonstrated great antioxidant potential with very low oxidation index EC50 (1.44 ± 0.01, 2.84 ± 0.07 and 0.32 ± 0.01 mg mL−1, respectively), significantly equivalent to EC50 of some commercial antioxidants such as α-tocopherol, ascorbic acid, BHA and BHT. Moreover, extracts of the most active species exhibited reducing activities, superoxide anion radical scavenging and inhibition of lipid peroxidation. These results suggest that some macroalgae from the Yucatan peninsula have a great antioxidant potential which could be considered for future applications in medicine, food production or cosmetic industry.  相似文献

Bioactive peptides have been defined as specific protein fragments that have numerous biological activities. The aim of this study was to introduce three multifunctional peptides. Hence, we used rabbit lung angiotensin converting enzyme (ACE) inhibitor peptide AFKDEDTEEVPFR to prepare two analogous peptides KDEDTEEVP and KDEDTEEVH. ACE inhibitory, antioxidant, and antimicrobial activities of three synthetic peptides were investigated. Among the three peptides, KDEDTEEVP exhibited the highest ACE inhibitory activity with IC50 value of 69.63 ± 2.51 μM. Furthermore, the results of fluorescence spectroscopy and molecular modeling showed that KDEDTEEVP had more affinity to ACE than other peptides. The peptide of KDEDTEEVH showed the strongest antioxidant scavenging capacity on DPPH radicals (EC50 = 135 ± 9.62 μM), hydroxyl radicals (EC50 = 144 ± 8.73 μM), and ABTS radicals (EC50 = 62 ± 4.52%). Moreover, it showed the highest activity in iron-chelating test (EC50 = 226 ± 14.13 μM) and could also effectively inhibit the peroxidation of linoleic acid. The antimicrobial activity results showed that KDEDTEEVH had higher efficiency against Gram-positive than Gram-negative bacteria with MIC values of higher than 205 ± 10.75 μM. Although there was not a direct correlation between ACE inhibitor and antioxidant activity for analogous peptides, both analogous peptides exhibited more efficiency than the mother peptide. Thus, they can be considered as multifunctional peptides and would be beneficial ingredient to be used in food and drug industry.  相似文献

Antioxidant activities and phenolic contents of 26 species extracts from 20 botanical families grown in north-western Himalaya were investigated. Antioxidant activities were determined using DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging and ferric reducing antioxidant power (FRAP) assays. Total phenolic content (TPC) was determined using a Folin-Ciocalteu assay. Quantitative and qualitative analysis of phenolic compounds was also carried out by reverse phase high performance liquid chromatography (RP-HPLC) using diode array detector (DAD). Major phenolics determined using RP-HPLC in analyzed species were gallic acid, chlorogenic acid, p-hydroxy benzoic acid, caffeic acid, vanillic acid, syringic acid, p-coumaric acid and ferulic acid. Antiradical efficiency (1/EC50) determined using DPPH radical scavenging assay ranged from 0.13 to 5.46. FRAP values ranged from 8.66 to 380.9 μmol Fe(II)/g dw. Similarly, the total phenolic content in the analyzed species varied from 3.01 to 69.96 mg of gallic acid equivalents (GAE)/g dry weight. Gallic acid was found in the majority of the samples, being most abundant compound in Syzygium cumini bark (92.64 mg/100 g dw). Vanillic acid was the predominant phenolic compound in Picrorhiza kurroa root stolen (161.2 mg/100 g dry weight). The medicinal plants with highest antioxidant activities were Taxus baccata and Syzygium cumini. A significant positive correlation, R 2?=?0.9461 and R 2?=?0.9112 was observed between TPC determined using Folin-Ciocalteu method and antiradical efficiency and FRAP values respectively, indicating that phenolic compounds are the major contributor of antioxidant activity of these medicinal plants.  相似文献

The effect of UV-C irradiation on jasmonates, abscisic acid (ABA), polyamines and antioxidant activities was investigated in apple plants (Malus domestica Borkh.). The EC50 values of O2 -scavenging activity in the UV-C treated leaves were lower than those in the untreated control. In addition, total ascorbic acid and polyphenolic concentrations in the UV-C treated leaves were generally higher than those in the untreated control. Although the endogenous ABA concentrations were not significantly different between the untreated control and UV-C treatment, the endogenous jasmonic acid, putrescine, and spermine concentrations in the UV-C treated plants were higher than those in the untreated control. However, the application of PDJ before UV treatment inhibited the rise of endogenous jasmonate and putrescine concentrations. The increase of ascorbic acid was also inhibited by PDJ application before UV treatment. These facts suggest that the increase of jasmonates and polyamines may be associated with UV-C stimulation, and UV-C irradiation may be effective for increasing antioxidant activity in apple leaves.  相似文献

A new natural flavonoid patuletin 3′-β-xylofuranoside was isolated from Leuzea carthamoides leaves. The antioxidant activity of this compound was evaluated by the DPPH radical assay and ferric reducing antioxidant power (FRAP) assay, and the results were compared with those for trolox and quercetin. DPPH radical scavenging activity of the tested compounds was expressed by the parameter EC50: patuletin 3′-β-xylofuranoside (56.0 μM), trolox (27.8 μM), and quercetin (25.3 μM). The ferric reducing activity of the compounds was demonstrated as FRAP values at 4 and 60?min: patuletin 3′-β-xylofuranoside (28.4 μM, 35.8 μM), trolox (19.3 μM, 20.2 μM), and quercetin (54.3 μM, 79.9 μM). The structure/activity relationship of the flavonoid is also discussed. The results indicate significant antioxidant potency of patuletin 3′-β-xylofuranoside.  相似文献

The aim of this study was to evaluate the antioxidant and anti-acetylcholinesterase properties and phytochemical constituents of the latex from Euphorbia dendroides L. (Euphorbiaceae) growing wild in Sicily. Phytochemical analysis revealed that into E. dendroides latex the triterpenoids were the most abundant among the identified compounds. Furthermore, a high content of polyphenols mainly as phenolic acids, was found. The antioxidant and free-radical scavenging properties, by several in vitro assays such as DPPH, TEAC and FRAP, have been evaluated. The results showed that E. dendroides latex has significant antioxidant activity, as measured by DPPH assay (2927.01?±?98.03 µmols of Trolox equivalent (TE)/100g FW). Reactivity towards ABTS radical cation and ferric-reducing antioxidant power (FRAP) values were 7580.95?±?97.65 µmols of TE/100g FW and 4383.13?±?95.30?μmol of TE/100g FW, respectively. The latex exhibited also significant inhibition of acetylcholinesterase activity with an IC50 value of 4.46 µg/mL (C.L.?=?2.002–9.947). Furthermore, Brine shrimp (Artemia salina) cytotoxicity bioassay showed that the larvae viability was significantly affected at higher concentrations than those capable to induce significant antioxidant and anti-acetylcholinesterase effects (LD50 25 µg/mL). The results suggest that polyphenols and terpenoids can contribute significantly to antioxidant and anti-acetylcholinesterase activities of E. dendroides latex.  相似文献

We investigated the antioxidant and radical scavenging activity of polyphenolic isochromans. To assess the relation between structure and scavenging properties the natural occurring 1-(3′-methoxy-4′-hydroxy)phenyl-6,7-dihydroxy-isochroman (ISO-3, three OH groups) was compared with three newly synthesized derivatives that differ in their degree of hydroxylation by substitution with methoxy-groups (ISO-4: four OH groups; ISO-2: two OH groups and ISO-0: fully methoxylated). We found that ISO-4 is a 2-fold better scavenger for the artificial radical 1,1-diphenyl-2-picrylhydrazyl (DPPH, 100?μM) with an EC50=10.3?μM compared to the natural ISO-3 (EC50=22.4?μM) and to ISO-2 (EC50=25.1?μM), while ISO-0 did not react with DPPH. The scavenging capacity for superoxide enzymatically generated in a hypoxanthin-xanthinoxidase reaction was the highest for ISO-4 (EC50=34.3?μM) compared to those of ISO-3 (EC50=84.0?μM) and ISO-2 (EC50=91.8?μM), while ISO-0 was inactive. In analogy, ISO-4 scavenged peroxynitrite (ONOO?, EC25=23.0?μM) more effective than ISO-3, ISO-2 and ISO-0.

When C6 rat glioma cells loaded with the reactive oxygen/nitrogen (ROS/RNS)-sensitive fluorochrome 2,7-dichlorodihydrofluorescein, were exposed to hydrogen peroxide, the lowest stress level as indicated by the fluorescence signal was detected when the cells were pretreated with ISO-4 or ISO-2 but to a much lesser extent with ISO-3, while ISO-0 did not show any effect. All tested hydroxyisochromans superceded the scavenging effect of trolox.

The excellent radical and ROS/RNS scavenging features of the hydroxy-1-aryl isochromans and their simple synthesis let these compounds appear to be interesting candidates for pharmaceutical interventions that protect against the deleterious action of ROS/RNS.  相似文献

A novel biopolymer-based antioxidant, chitosan conjugated with gallic acid (chitosan galloylate, chitosan-GA), is proposed. Electron paramagnetic resonance (EPR) demonstrates a wide range of antioxidant activity for chitosan-GA as evidenced from its reactions with oxidizing free radicals, that is, 1,1-diphenyl-2-picryl-hydrazyl (DPPH), horseradish peroxidase (HRP)-H2O2, carbon-centered alkyl radicals, and hydroxyl radicals. The EPR spectrum of the radical formed on chitosan-GA was attributed to the semiquinone radical of the gallate moiety. The stoichiometry and effective concentration (EC50) of the DPPH free radical with chitosan-GA show that the radical scavenging capacity is maintained even after thermal treatment at 100 °C for an hour. Although the degree of substitution of GA on chitosan was about 15%, its antioxidant capacity, that is, the reaction with carbon-centered and hydroxyl radicals, is comparable to that of GA.  相似文献

Hardy kiwifruit (Actinidia arguta) is a highly appreciated exotic fruit endowed with outstanding bioactive compounds. The present work proposes to characterize the pulp from A. arguta organic fruits, emphasizing its radicals scavenging capacity and effects on intestinal cells (Caco-2 and HT29-MTX). The physicochemical properties and phenolic profile were also screened. The total phenolic and flavonoid contents (TPC and TFC, respectively) of pulp were 12.21 mg GAE/g on dry weight (DW) and 5.92 mg CE/g DW, respectively. A high antioxidant activity was observed (FRAP: 151.41 μmol FSE/g DW; DPPH: 12.17 mg TE/g DW). Furthermore, the pulp did not induce a toxic effect on Caco-2 and HT29-MTX cells viability up to 1000 μg/mL. Regarding in vitro scavenging capacity, the pulp revealed the highest scavenging power against NO. (IC50=3.45 μg/mL) and HOCl (IC50=12.77 μg/mL). These results emphasize the richness of A. arguta fruit pulp to be used in different food products.  相似文献

Macroalgae are currently being explored as novel and sustainable sources of bioactive compounds for both pharmaceutical and nutraceutical applications arising from their antioxidant, anticancer, and antimicrobial activity. In the present study, the antitumoral and antioxidant activities of crude methanolic extracts of the freshwater macroalga Cladophora surera Parodi & Cáceres, harvested from Napostá Creek (Argentina), were investigated in vitro. The antioxidant activity was assessed by DPPH method and polyphenol content using Folin-Ciocalteu phenol reagent. Antitumoral activity was evaluated on the human breast adenocarcinoma cell line MCF-7 by measuring proliferation, migration, and cell adhesion. The algal extract (AE) showed a total phenol content of 1.62?±?0.17 μg GAE mg?1 dry alga and DPPH scavenging activity of 25.03?±?1.99% (10 mg)?1 dry alga. The trypan blue assay after 48 h of treatment indicated that the AE significantly inhibits proliferation in a dose-dependent manner (1–100 μg mL?1), being more effective the highest dose employed, with a concomitant increment in dead cells. However, the colorimetric MTS assay only showed a significant decrease in cell viability at 100 μg mL?1 AE. Using the wound healing assay, we demonstrated that AE inhibits cell migration. Through a cell adhesion assay, we found that AE affects considerably the cell adhesion capacity at all doses probed. Analysis of cell spreading indicated that cell morphology was also affected by AE treatment. These results indicate that C. surera could be a source of valuable bioactive compounds usable as antitumoral preventive therapy for their effects on the regulation of processes involved in metastasis in cells derived from human mammary cancer.  相似文献

We investigated the antioxidant and enzyme inhibitory activities and chemical composition of the hydro-distilled essential oil (0.35% yield) from aerial parts of Thymus spathulifolius. Antioxidant capacity of the oil was assessed by different methods including free radical scavenging (DPPH and ABTS), reducing power (FRAP and CUPRAC) and phosphomolybdenum assay. Inhibitory activities were analyzed against acetylcholinesterase (AChE), butyrylcholinesterase (BChE), α-amylase, α-glucosidase, and tyrosinase. Twenty-one constituents were identified representing 97.2% of the total oil with thymol (50.5%), borneol (16.7%) and carvacrol (7.7%) as the major components. The essential oil exhibited good antioxidant activity with IC50 values of 3.82 and 0.22?mg/mL determined by free radical scavenging DPPH and ABTS, respectively. EC50 values of FRAP and CUPRAC were found to be 0.12 and 0.34?mg/mL, respectively. The results of the present study support the uses of T. spathulifolius essential oil as a source of natural antioxidants and bioactivities for functional foods and phytomedicines.  相似文献

A versatile synthesis of novel 5-hydroxylaminoisoxazoles bearing adamantane moieties has been accomplished using the heterocyclization reactions of readily available unsaturated esters by the treatment with tetranitromethane in the presence of triethylamine and subsequent reduction of resulting 5-nitroisoxazoles by SnCl2 with the participation of THF. A number of obtained isoxazole derivatives were evaluated for their antioxidative activity, inhibition of lipoxygenases and impact on the rat liver mitochondria. The majority of tested compounds demonstrated moderate antiradical activity in DPPH test (up to EC50 16 μM). The same compounds strongly inhibited soybean lipoxygenase (up to IC50 0.4 μM) and Fe2+- and Fe3+-induced lipid peroxidation (LP) of rat brain cortex homogenate (up to IC50 0.3 μM). All tested isoxazole derivatives promoted the phosphorylating respiratory activity simultaneously with maximal stimulated respiratory activity of mitochondria and do not reveal any toxicity towards the primary culture of rat cortex neurons.  相似文献

Proanthocyanidins in Chinese bayberry leaves (PCBLs) were qualitatively analyzed. NMR data suggest that PCBLs are mostly composed of (epi)gallocatechin gallate units. Matrix-assisted laser desorption time-of-flight MS data indicate 95 possible prodelphinidin structures, ranging from dimers to tridecamers. Preparative normal-phase HPLC and further analysis by reverse-phase HPLC together with electrospray ionization MS enabled detection of 20 compounds, including seven newly identified compounds in Chinese bayberry leaves. The antioxidant capacity of PCBLs was evaluated by (1,1-diphenyl-2-picryl-hydrazyl), ferric-reducing antioxidant power, and oxygen radical absorption capacity assays. The EC50 of DPPH radical scavenging activities (as 50% decrease in the initial DPPH concentration) were 7.60 µg. The FRAP and ORAC values were 8859.33±978.39 and 12991.61±1553.34 µmol Trolox equivalents per gram, respectively. The results indicate the high antioxidant potency of PCBLs.  相似文献

In the current study, the effect of glycerol -as a green solvent- addition into solvent mixture (50:50 ethanol:distilled water), on some biofunctional properties of Origanum onites L. was investigated. Response surface methodology (RSM) was used to detect the optimum conditions for the extraction process. Three variables namely glycerol concentration (X1: 1–9 g), extraction temperature (X2: 25–75 °C) and time (X3:10–30 min) were selected and also total phenolic content, total flavonoid content, antioxidant capacity and antiradical activity of O. onites extracts were determined. Analysis of variance (ANOVA) showed that glycerol incorporation significantly increased the total phenolic content and antioxidant activity (p < 0.05) of the samples. Maximum levels to obtain the highest bioactive properties (highest total phenolic content and antioxidant activity) were determined as to be 9 g of glycerol addition for the extraction conditions as 45.4 °C and 75 min. This study reports the effect of glycerol on bioactive properties of O. onites and suggests that glycerol can be used to produce hydroalcoholic extracts having higher bioactivity from Origanum genus.  相似文献
