Assisted reproductive technologies can be an important part of programs directed for maintenance and protection of genetic variability. The objective of this study was to develop methods for liquid storage and cryopreservation of semen capercaillie and black grouse semen. Our results provide for the first time evidence for successful short-term storage and cryopreservation of capercaillie and black grouse semen using criteria of sperm motility characteristics as quality indices. Sperm motility could be protected up to 48 h liquid storage; however, 24-h storage should be preferable. Cryopreservation secured 40–60% post-thaw motility (as compared with control), both for freshly collected and 24-h-stored semen. In conclusion, a procedure for short-term and cryopreservation of capercaillie and black grouse semen are now available. These assisted reproductive technologies can be implemented into captive breeding programs for these species.  相似文献

The objective of this retrospective field study was to determine the effects of storing, for up to 62 h, heterospermic and homospermic semen in the short-term extender Beltsville thawing solution (BTS), on reproductive performance in Norwegian swine of four different breed combinations. The study was based on fertility records after single or double inseminations with semen collected at an AI station in Norway from January 1998 to June 2001. Increasing the duration of storage of homospermic semen, but not heterospermic semen, from an interval of 4-14 h to an interval of 52-62 h, was associated with a 0.5 piglet reduction in litter size. There were differences in reproductive performance among breed combinations that appeared to be associated with duration of semen storage. In conclusion, prolonged semen storage decreased reproductive performance; the extent varied among breeds and was prevented by the use of heterospermic versus homospermic semen.  相似文献

In this comparative study, reproductive parameters and semen characteristics of cloned bulls (n = 3) derived from somatic cell nuclear transfer (SCNT) were compared to their original cell donor Holstein-Friesian (n = 2) bulls from the same enterprise to assess the differences in reproductive potential between a donor bull and its clones. The parameters evaluated included motility of fresh, frozen-thawed and Percoll-treated frozen-thawed spermatozoa, as well as in vitro fertilization (IVF) ability, embryo quality, birth and survival of calves following IVF and embryo transfer with frozen-thawed semen. With fresh semen, spermatozoa from one cloned bull had lower motility than its donor. Cloned bulls had higher velocity parameters in fresh semen, but those effects were not obvious in frozen-thawed or frozen-thawed semen selected with a Percoll gradient. Semen collected from cloned bulls had significantly higher IVF rates compared to donors; however, embryo development per cleaved embryo or quality of blastocysts did not differ between donors and cloned bulls. Pregnancy and live offspring rates from one donor and its cloned bull did not differ between fresh (40%, 16/40 versus 46%, 17/37) and vitrified/thawed (13%, 2/16 versus 25%, 4/16) embryo transfer following IVF. A total of 26 calves were obtained from genotypically identical donor and cloned bulls with no signs of phenotypical abnormalities. These preliminary results suggested that the physiology of surviving postpubertal cloned bulls and quality of collected semen had equivalent reproductive potential to their original cell donor, with no evidence of any deleterious effects in their progeny.  相似文献

It is difficult to maintain turkey semen quality after in vitro liquid storage and the problem is worsened by animal aging. Little is currently known about the effects of both reproductive period and strain on the preservability of qualitative characteristics of turkey semen during liquid storage. The purpose of this study was to evaluate the effect of the reproductive period of two commercial turkey strains on semen quality changes during in vitro storage for upto 48 h at 5 degrees C. Two different periods were considered: first period from 32 to 40 weeks of age and the second one from 44 to 52 weeks. Turkey males from either British United Turkeys (BUT) Big-6 line and Hybrid Large White line (Hybrid) were used. Semen pools of each tom strain were diluted with Beltsville Poultry Semen Extender (BPSE) and the motility, viability and membrane integrity of sperm were evaluated at 3, 24 and 48 h of liquid storage at 5 degrees C. The sperm concentration was significantly affected by period (P<0.01) and strain (P<0.05), with best values in first period and in the Hybrid semen. Besides also the motility, viability and membrane integrity during 48 h of storage were better (P<0.05) in the first period compared to the second one for both strains, particularly in Hybrid semen. During storage it was clearly shown in the first period that Hybrid sperm worsened more than the BUT one: in spite of the motility and viability values were at first (3h) higher (P<0.05) in Hybrid semen, after 48 h of storage the motility did not show any significant difference between strains while the viability resulted even better (P<0.05) in BUT semen. In the second period, although the semen quality decreased during the storage with a similar trend for both strains, better (P<0.05) values were found in BUT semen. Our results indicated that the reproductive period affected the quality of turkey semen in a different manner according to the strain. Moreover BUT semen showed a better in vitro storage ability compared to the Hybrid one.  相似文献

Porcine reproductive and respiratory syndrome virus (PRRSV) has been reported to be shed in the semen of infected boars. To determine whether the reproductive tissues could be a persistent source of virus and the possible origin of PRRSV found in semen of infected boars, 20 PRRSV-seronegative boars were intranasally inoculated with 5 x 10(6) median tissue culture infective doses (TCID50) of PRRSV and necropsied at different times post-inoculation (p.i.) from Day 2 to Day 37 p.i. Blood samples were collected before experimental inoculation, at necropsy and at different times p.i. At necropsy, epididymal semen and reproductive tissues were collected and the presence of the virus determined by virus isolation. The infection of the boars was demonstrated by the isolation of the virus from the sera of all inoculated boars and by seroconversion. PRRSV was detected in serum samples from Day 2 to Day 23 p.i., although the viremic period was largely dependent on the individual response to infection. Viral replication was proven within different reproductive tissues from Day 2 to Day 23 p.i., being most consistently found in the epididymus. In addition, PRRSV was isolated in semen from Day 4 to Day 10 p.i. The correlation of a diminished viremia and the inability to isolate PRRSV from semen or reproductive tissues may be due to one of two possibilities. First, viremia is responsible for most of the virus isolated from reproductive tissues due to the movement of PRRSV-infected cells out of the blood and into the tissues. Second, viremia may initially seed the reproductive tissues with PRRSV, and then the virus is produced into the reproductive tract and shed into semen at low levels.  相似文献

Although the potential use of reproductive biotechnology for safeguarding of endangered wildlife species is undoubted, initial evaluation of the genetic and reproductive relationship between the endangered mammals and those closely related species is indispensable. Isfahan mouflon Ovis orientalis isphahanica is now considered as a threatened species by International Union for the Conservation of Nature. Therefore, little is known about the biology of this species. This study was carried out to investigate the possible reproductive potential of domestic sheep for ex situ conservation of the Isfahan mouflon. Somatic cell cultures were taken from ear biopsies of the wild and domestic sheep and were used for karyotype analysis. Semen samples were collected by electroejaculator from the wild and domestic rams. The spermatological characteristics of the collected semen samples were determined and used for both cryopreservation and cross-insemination of the synchronized wild and domestic female sheep. To establish a cryobank for the threatened species biomaterials, freezed samples of the somatic cells and semen were transferred to a cryotank. The result suggested that Isfahan mouflon has conserved its chromosomal integrity as previously observed and contains the same chromosomal number as the domestic sheep (2n = 54). The semen samples of both species revealed similar cryoviability (>35% gross motility postthawing). Cross-insemination of both species resulted in successful pregnancy. It was suggested that domestic sheep possesses the required biological characteristics to be considered for safeguarding of the Isfahan mouflon.  相似文献

White rhinoceroses suffer from a low rate of reproduction in captivity. This study examines the role of male reproductive function as a contributing factor. We used ultrasonography to image accessory sex glands, testis and epididymis. Electroejaculation provided 36 ejaculates from 21 rhinoceroses. Based on the percentage of progressively motile spermatozoa, semen was categorized in three groups, high (I: >75%), intermediate (II: 50-70%) or low (III: <50%) quality. Only 52% of the males showed high semen quality. Ejaculates in the high motility category also had the highest proportion of morphologically intact spermatozoa. Both semen parameters, sperm motility and morphology, were found to positively correlate with size of the accessory sex glands. The semen category was associated with group size suggesting that the social status influenced functional reproductive parameters. Change of territorial status (n = 1) improved semen quality. Testicular fibrosis was characterized as a sign of reproductive ageing in all males older than 15 years of age (n = 13); although, this ageing process did not notably affect semen parameters. Furthermore, for the benefit of assisted-reproduction and genetic banking programs protocols for the storage of cooled semen and the cryopreservation of spermatozoa were designed using different cryodiluents. This report provides basic data for the evaluation of reproductive components and of breeding management in male rhinoceros. Our results indicate that low rate of reproduction in captivity can be attributed to reduced male reproductive fitness. Changes in management of white rhinoceroses may positively affect male reproductive function.  相似文献

Hypothyroidism was induced in goats using thiourea as the goitrogen for 90 days and the reproductive behaviour and semen characteristics were studied. The reversability of the changes was also observed for 90 days on discontinuation of treatment with thiourea. Loss of libido in males and anoestrum in females were the important clinical features observed in hypothyroid animals. There was a decrease in ejaculate volume, sperm concentration, motility and viability with an increased incidence of abnormal spermatozoa. There was significant improvement in the quality and quantity of semen when the animal became euthyroid after withdrawal of thiourea.  相似文献

Bovine virus diarrhea (BVD) virus was inoculated into 9 bulls. Semen samples were collected to determine if BVD virus could be isolated from semen and to determine if the virus affected semen quality. The reproductive tracts of 6 bulls were recovered following slaughter and subjected to virus isolation procedures and histologic examination.BVD virus was detected in 4 of 98 semen samples following inoculation. The virus was also recovered from the testicle of one bull following slaughter. Inoculation of BVD virus into bulls did not affect semen quality and did not cause gross or microscopic lesions in the reproductive tract. This experiment indicates that following inoculation of BVD virus into bulls, the virus may be shed in the semen. However, the probability of this occurring is low.  相似文献

The chamois of Abruzzi (Rupicapra pyrenaica ornata) has been classified as endangered by the World Union for Conservation. The objective of this study was to analyze seasonal differences in the characteristics of various male reproductive organs and in semen quality. The study was conduced during 2004 in the reserve of Lama dei Peligni (Italy) on three chamois males aged between 2 and 5 years. Males were captured during March-May months and October-December months. Various testicular and scrotal measurements were taken and semen was collected using an electroejaculator. Sperm motility pattern was evaluated using computer assisted sperm analyzer, membrane integrity using differential staining and morphology with phase contrast microscopy. Testicular size, sperm motility membrane integrity and the percent of morphological normal spermatozoa were greater during October-December. The authors concluded that sperm characteristics are strongly influenced by season and that semen collected in this period (breeding season) has suitable quality for manipulation and long-term preservation.  相似文献

Despite the wide geographical distribution of coati (Nasua nasua) from the south of Canada to the north of Argentina, studies regarding the reproductive characteristics of this species are extremely limited. The objective of this study was to describe the various characteristics of coati semen by morphometric and ultrastructural analysis. Five mature males were anesthetized and electroejaculated for the collection of semen. Semen was immediately evaluated for color, volume, pH, sperm motility, vigor, morphology, acrosomal integrity, percentage of live cells and hypo-osmotic response by light microscopy. Sperm cell morphometry and ultrastructural analyses were also performed. Observations of seminal characteristics determined by electroejaculation in captive coatis represent a valuable baseline dataset for establishing fertility standards and provide background information that may be useful for assisted breeding programmes in members of the Procyonidae family.  相似文献

Fumonisins, a group of toxic metabolites produced by the genus Fusarium, are known to be consumed by farm animals and are the causative agent or a suspected contributing factor in farm animal diseases. Pigs are particularly susceptible to fumonisins. Reproductive inefficiency is recognized as the most costly limiting constraint to efficient animal production. To account for potential reproductive effects of fumonisin in boars, dietary fumonisin B1 (FB1) was fed to 24 male Large White weanling pigs, 8 to 9 weeks of age. The animals were randomly assigned to four diets containing 5.0, 10.0, 15.0 and 0.2 mg FB1/kg constituting diets 1, 2, 3 and control diet, respectively. After 6 months, semen samples were collected from the pubertal boars and analysed. After the semen collection, all the pubertal boars were killed by decapitation, their reproductive systems dissected and the weights of the testes and epididymides as well as the volumes of the testes recorded. Dietary FB1 did not influence both the relative weights of the testes and epididymides as well as the volumes of the testes. Except for the semen volume and spermatozoa morphological abnormalities, all the semen characteristics studied decreased in a dose-dependent manner and the decrease was statistically significant (P < 0.05). The sperm concentration, total sperm and motile sperm per ejaculate of the animals on diet 3 were 83.3%, 79.1% and 59.6% of the controls, respectively. The dietary FB1 levels influenced the mass activities of the semen, which ranged from very turbulent motion for animals on the control diet to absence of wave motion for those on diet 3. The study revealed that male weanling pigs for breeding should not be exposed to dietary FB1 higher than 5 mg/kg for optimum reproductive performance. The results of this present study suggest that the recommendation of 10 mg/kg by the United States Food and Drug Administration as the maximum level of total dietary fumonisins was above the no-observable-effect level of dietary fumonisin for swine.  相似文献

Bull breeding soundness parameters, semen characteristics and sexual behavior were evaluated for effects on reproductive performance in single-sire beef herds. A total of 155 cow herds (Angus, 50 herds; Hereford, 40 herds; Brahman, 46 herds; and Senepol, 19 herds) bred to bulls of the same breed were observed for 8 yr. All bulls produced adequate quality semen and had scrotal circumference (SC) >/= 30 cm. Reproductive performance was evaluated by the conception rate (CON), conception rate during the first 21 d of the breeding season (21dCON), mean calving date (MCD), and mean calving date of the first half of the herd to calve (HHCD). Correlations were determined between breeding soundness parameters and reproductive performance for all bulls combined, by breed, and by age. The Cp statistic was used to select models for the effects of parameters on CON, 21dCON, MCD and HHCD. Breeding season length and breed had significant effects. The percentages of normal cells, proximal droplets, detached heads and the semen score (motility plus percentage of normal cells) had a significant effect on CON when all bulls were considered. After the effect of season was deleted, the most significant parameter affecting CON in the Brahman was the percentage of detached sperm heads. In the Angus, motility was significantly correlated with all reproductive performance indices. In the Hereford, breeding soundness examination score (BSE) was positively correlated with 21dCON.  相似文献

Soy and derivative diets deliver large doses of isoflavones to human and animals throughout their lifespan, including gestation. Epidemiologic and experimental data suggest that the consumption of soybean containing foods may protect against cardiovascular disease and decrease breast, prostate and endometrial cancer risk. Based on animal and in vitro studies, however, concerns have been raised that consumption of isoflavones may cause potential adverse effects on the reproductive tract and behavior. The aim of this study was to investigate the effects of chronic consumption of a soy meal containing diet or soy isoflavones supplement on the morphology of reproductive organs, semen quality, age that males reached puberty, and sexual behavior of male rabbits. With this purpose, 16 female rabbits were randomly assigned to receive: (1) a soy- and alfalfa-free diet; (2) a soy- and alfalfa-free diet supplemented with 5mg/kg body wt./day of soy isoflavones; (3) a soy- and alfalfa-free diet supplemented with 20mg/kg body wt./day of soy isoflavones; (4) a diet containing 18% of soy meal, throughout the gestation and lactation. After weaning, male offspring received the same diet, which was given to the respective mother. The age that males reached puberty, semen characteristics and sexual behavior were evaluated in these animals. At 33 weeks of age, the reproductive organs were submitted to histological evaluation. Rabbits, which received large amounts of isoflavones (20mg/kg body wt./day) had a lesser food intake, body weight and semen volume. Spermatogenesis, morphology of male genital organs and sexual behavior did not differ significantly from the control group. We conclude that chronic dietary treatment with soy based diet or soy isoflavones have no adverse effects on the observed reproductive patterns of male rabbits.  相似文献

A method for in vitro storage of Atlantic halibut Hippoglossus hippoglossus L. semen can facilitate seed production. The study aimed at determining the effect of male spermiation advancement on viability of chilled stored spermatozoa. The use of catheterization of semen from the sperm duct was examined. Also, large volumes of semen were stored under sub-optimal production-like conditions in order to determine the suitability of the method into practical use. Semen was collected from two broodstocks: natural photoperiod males, being at the first phase of the reproductive season and 3-month advanced photoperiod males, being at the end of the reproductive season. Semen samples were diluted 1:5 (v/v) with modified Hanks' Balanced Salt Solution supplemented with antibiotics, and stored in Ziploc bags filled with air. Sperm motility parameters, assessed by computer-assisted sperm analysis (CASA), were assessed weekly. Experimental and production-scale fertilization trials were performed. Sperm samples from natural photoperiod males showed significantly longer viability under in vitro storage conditions than sperm from advanced photoperiod males. In the natural photoperiod group, the decrease in spermatozoa motility, curvilinear velocity and straight-line velocity occurred on day 50, 14, and 28 of storage, respectively. Spermatozoa from one of five males were still motile on day 80 of storage, and fertilization rates and embryo survival rates obtained using semen stored for 70 days did not differ from control values and they were significantly higher than values obtained with the use of fresh semen of the same male, but being at the end of reproductive season. Catheterization of semen showed no advantage to stripping the semen without a catheter, even for samples stored undiluted for 1 day of collection, before dilution. Under sub-optimal conditions, spermatozoa stored in large volumes (10-100mL of diluted semen) without any special treatments except for weekly swirling, remained viable for more than 1 month. Production-scale fertilizations with samples stored for 5-21 days resulted in high survivals of embryos and hatchlings. Because of its simplicity and efficiency, the method shows a high potential for use in commercial Atlantic halibut farming. It has already been applied to a halibut breeding programme for the next reproductive season at our research station.  相似文献

Thirty five females and 15 males of New Zealand White mature rabbits about 6 months of age, were assigned to 1–5 dietary treatments (7 does+3 bucks for each): uncontaminated control diet, naturally aflatoxin contaminated diet without or with 1,2 and 3% bentonite. Rabbit fed with the aflatoxin-diet had a decreased (P<0.01 or 0.05) physical semen characteristics of bucks and a reproductive performance traits of does. The values of conception rate (%), gestation length (days), litter size (n) and litter weights (g) at birth and viability (%) of litters of doe rabbits, fed with the aflatoxin-diet, recorded, respectively: 64.5; 31.0; 4.4; 275.0 and 57.1 versus 85.6; 30.3; 7.9; 508.0; and 100 for those fed with the uncontaminated diet. Addition of bentonite to the aflatoxin contaminated diet improved in general the physical semen characteristics of buck and reproductive performance traits of doe rabbits. The results of the study demonstrate that adding 1% of Egyptian raw bentonite to the naturally aflatoxin contaminated rabbit diets can provide an effective, cheap and safe practical technique for preventing the aflatoxicosis in mature rabbits.  相似文献

There is still controversy as to how body mass index (BMI) affects male reproduction. We investigated how BMI is associated with semen quality and reproductive hormones in 166 men, including 38 severely obese men. Standard semen analysis and sperm DNA integrity analysis were performed, and blood samples were analysed for reproductive hormones. Adjusted for age and time of abstinence, BMI was negatively associated with sperm concentration (B = -0.088, P = 0.009), total sperm count (B = -0.223, P = 0.001), progressive sperm motility (B = -0.675, P = 0.007), normal sperm morphology (B = -0.078, P = 0.001), and percentage of vital spermatozoa (B = -0.006, P = 0.027). A negative relationship was observed between BMI and total testosterone (B = -0.378, P < 0.001), sex hormone binding globulin (B = -0.572, P < 0.001), inhibin B (B = -3.120, P < 0.001) and anti-Müllerian hormone (AMH) (B = -0.009, P < 0.001). Our findings suggest that high BMI is negatively associated with semen characteristics and serum levels of AMH.  相似文献

Compared to other domestic bird species, geese exhibit the lowest reproductive efficiency (poor semen quality, low egg production, and poor fertility and hatchability rates). From an economic perspective, it is a necessity of improve these reproductive traits. Studies have demonstrated that the essential trace element—selenium—plays key roles in testicular development and the maintenance of spermatogenesis. The aim of the present study was to determine the effect of feed supplementation with organic selenium and vitamin E on ganders’ response to manual semen collection and semen quality. Sixteen 3-year-old White Koluda ganders were randomly divided into two groups. The control group was provided commercial feed while the experimental group was provided with the same commercial feed supplemented with selenium (0.3 mg/kg) and vitamin E (100 mg/kg). The response of individual ganders from both groups to manual semen collection and the quality of the semen collected were evaluated. The supplements increased (P?≤?0.05) the frequency and decreased the time interval of a complete ejaculatory response of the ganders to manual semen collections (82.7 % supplement vs. 73.5 % control). Males from the supplemented group had significantly higher (P?≤?0.01; P?≤?0.05) ejaculate volumes, sperm concentrations, and percentages of viable sperm and lower percentages of immature sperm (spermatids). Lipids peroxidation, expressed in terms of the malondialdehyde concentration, was lower (P?≤?0.01) in semen of the supplemented group (0.172 nmol/50?×?106) as compared to the controls (0.320 nmol/50?×?106). Moreover, the duration of the reproductive period of the ganders in the experimental group was 1 week longer. The results show that supplemental dietary selenium and vitamin E improved both the ganders’ response to manual semen collection and semen quality. We conclude that such feed supplementation could lead to greater economic benefits through increased reproductive efficiency within the goose production industry.  相似文献
