(1) H+/electron acceptor ratios have been determined with the oxidant pulse method for cells of denitrifying Paracoccus denitrificans oxidizing endogenous substrates during reduction of O2, NO?2 or N2O. Under optimal H+-translocation conditions, the ratios H+O, H+N2O, H+NO?2 for reduction to N2 and H+NO?2 for reduction to N2O were 6.0–6.3, 4.02, 5.79 and 3.37, respectively. (2) With ascorbate/N,N,N′,N′-tetramethyl-p-phenylenediamine as exogenous substrate, addition of NO?2 or N2O to an anaerobic cell suspension resulted in rapid alkalinization of the outer bulk medium. H+N2O, H+NO?2 for reduction to N2 and H+NO?2 for reduction to N2O were ?0.84, ?2.33 and ?1.90, respectively. (3) The H+oxidant ratios, mentioned in item 2, were not altered in the presence of valinomycinK+ and the triphenylmethylphosphonium cation. (4) A simplified scheme of electron transport to O2, NO?2 and N2O is presented which shows a periplasmic orientation of the nitrite reductase as well as the nitrous oxide reductase. Electrons destined for NO?2, N2O or O2 pass two H+-translocating sites. The H+electron acceptor ratios predicted by this scheme are in good agreement with the experimental values.  相似文献

Respiration (O), ammonium (NH4), phosphate (PO4), total nitrogen (NT) and phosphorus (PT) excretions were measured on mixed zooplankton during 3-, 6-, 9-, 12-, 21-, and 24-h incubation periods at 20–23 C. The excretion rates of PO4, NT. and PT decrease during a 21-h period, while rates of respiration and excretion of NH{IN4} are constant. The percentage of inorganic nitrogen excreted increases regularly from 3 h (30–40% of total nitrogen) to 21 h (70–80%) and it could be either due to a bacterial activity which was measured or to a decrease with time of organic nitrogen excreted because of starvation. ONT, OPO4, OPT, and NH4PO4 ratios increase during the first 9 h of incubation; the percentage of inorganic phosphorus excreted is higher at the very beginning and then remains constant from 6 to 24 h. ONH4 and NTPT ratios are constant during a 24-h term, which makes them useful metabolic indexes.  相似文献

We develop equivalent formulation which is valid for absolute abundance data, reduces to the Sanders-Whittaker equality when proportionate abundance data are employed, and is more sensitive to differences in species abundance distributions. The in vitro reaction of bacteriophage T7-DNA with the radioactive diastereomeric benzo(a)pyrene-diol-epoxides, (±) [3H9, 3H10]-7β,8α-dihydroxy-9α,10β-epoxy-7,8,9,10-tetrahydrobenzo(a)pyrene, and (±) [3H9, 3H10]-7β,8α-dihydroxy-9β,19β-epoxy-7,8,9,10-tetrahydrobenzo(1)pyrene, was investigated. Chromatographic analysis of digests of the DNA allowed the distinction of characteristic deoxynucleoside adduct peaks for the two benzo(a)pyrene-diol-epoxides. Our results, together with data from the literature, allow the identification of these adducts as mostly N2-(10-7β,8α,9α-trihydroxy-7,8,9,10-tetrahydrobenzo(a)pyreney1)deoxyguanosine and N2-(10-7β,8α,9β-trihydroxy-7,8,9,10-tetrahydrobenzo(a)pyreney1)deoxyguanosine, respectively. DNA-benzo(a)pyrene adducts with the same chromatographic properties were formed in mouse embryo fibroblasts upon treatment with benzo(a)pyrene.  相似文献

Twelve experiments are reported on naturally occurring groups of N2-fixing blue-green algae in which reduction of acetylene and 15N2 were determined simultaneously. Simple averaging of the C2H2N2 reduction rates yielded a conversion factor of 4.4. Linear and log transform regression analyses yielded values of 4.8 and 4.2, respectively. Reports estimating nitrogen inputs based on acetylene reduction determinations are evaluated.  相似文献

The mean fixation index within subpopulations (FIS) has been defined as F̄IS = ∑wiFISior asF̂IS = ∑wipiqiFISi∑wipiqi. The latter definition is preferred because it can be obtained from the two other fixation indices, FST and FIT and because it is unaffected by the mean gene frequency. The expected frequency of heterozygotes in small subpopulations of dioecious organisms will exceed Hardy-Weinberg expectations and this can be measured by F̂IS. In an isolated subpopulation of constant variance effective size N, F̂IS rapidly tends to 1 − 4N2(N − 1 + [N2 + 1]12)2. In the Island model of population structure, F̂IS is approximately −(1 − m)Nwhere m is the immigration rate.When a sample is drawn from a natural population, the observed FIS will depend upon the genetic structure of the population. The values of FIS expected in three different types of population structure are discussed.  相似文献

In order to test the question if a pool of lipophilic ions may exist in black lipid membranes which cannot be detected by electrical relaxation measurements we have performed simultaneously measurements of the optical absorption of a lipophilic ion. The absorbance of membrane-bound dipicrylamine at 410 nm was measured with a sensitive spectrophotometer which can detect absorbance changes ? 4 · 10?5. A minimal concentration of about 6 · 1011 dipicrylamine ions per cm2 of the membrane could be detected with this instrument. The dipicrylamine concentration in the membrane obtained with the optical method Ntopt is compared with the concentrations Ntel obtained from simultaneous electrical relaxation measurements. Ntopt and Ntel agreed at low dipicrylamine concentrations (10?8–10?7 M in the aqueous phase) and showed saturation at higher concentrations (up to 5 · 10?6 M). In the saturation range Ntopt was maximally four times higher than Ntel. The significance of this difference is discussed together with general aspects of the saturation phenomenon.  相似文献

5-hydroxylysine, an analogue of glutamate and lysine, causes NH4+ production by N2-fixing A. cylindrica; it also reversibly inhibits GS activity in vitro but has no effect on alanine dehydrogenase or GOGAT. On adding 5-hydroxylysine intracellular pools of glutamine, glutamate and aspartate decrease; those of alanine and serine increase. 5-hydroxylysine alleviates the inhibitory effect of NH4+ on heterocyst production and C2H2 reduction and in NH4+-grown cultures results in heterocyst synthesis and in C2H2 reduction. The data suggest that the GS-GOGAT pathway is the sole route of importance in primary NH4+ assimilation in A. cylindrica, that NH4+ alone does not inhibit nitrogenase and heterocyst production, and that GS and/or a product is involved in regulating the production of both.  相似文献

Precipitation of human fibrinogen in 0.15 m NaCl occurred at pH 7.4 (Tris-HCl buffer) when ZnCl2, CuCl2, NiCl2, or CoCl2 were added beyond their respective critical concentrations. The critical concentrations were about 4 × 10?5m ZnCl2, 6 × 10?5m CuCl2, 3 × 10?4m NiCl2 and 1 × 10?3m CoCl2. At pH 5.8 2-(N-morpholino)-ethane sulphonic acid buffer, the critical concentrations were found only for CuCl2 and ZnCl2, and were about 3 × 10?5and 3 × 10?4m, respectively. CaCl2 and MgCl2 were not effective up to 1 × 10?2and 2 × 10?2m at pH 7.4 and 5.8, respectively. At pH 7.4, precipitation was better in 0.015 m NaCl than in 0.15 m NaCl for both CuCl2 and ZnCl2. Little or no conformational change was indicated on binding Cu2+ ions. The fluorescence of tryptophan was quenched only by CuCl2, while other metal ions (ZnCl2, NiCl2, CoCl2 and CaCl2) were ineffective as quenchers.  相似文献

The high-frequency elastic properties of rat-tail tendon collagen have been investigated by means of Brillouin (inelastic) light scattering. Longitudinally and transversely polarised elastic waves of frequency about 1010 Hz have been observed propagating at various angles to the fibre axis of stretched, partially dried tendon. Assuming that the elastic properties of tendon are transversely isotropic, these measurements enable the five elastic constants for such a system to be determined. In particular the ratio of the Young's modulus for strain parallel to the axis to that for strain perpendicular to the axis (EE) is found to be 1.43 and the ratio of the shear modulus to E is 0.28. In wet collagen only the longitudinal branch has been observed and in this case the ratio EE increases to 1.82. The absolute value for E in dry collagen is 11.9 GN m?2 reducing to 5.1 GN m?2 in wet collagen. An interpretation of these results in terms of the expected vibrations of the collagen molecular assembly is given. Possible applications to the determination of the mechanical properties of collagen composite materials such as bone are discussed as well as some measurements on silk and α- and β-keratins, which are fibrous proteins of different molecular conformation to collagen.  相似文献

In many instances the effect of superoxide (O2?) trapping agents in suppressing the net rate of O2 consumption of activated PMN's is not in accordance with theoretical expectations. We offer here an alternate explanation to those previously presented by Segal and Meshulam (FEBS Letters 100, 27–32) and Babior (Biochem. Biophys. Res. Comm. 91, 222–226). The paradoxical results previously presented can be explained by recognizing that shortly after activation of resting cells an O2 diffusion layer is established at or near the outer surface of these cells. The presence of this diffusion layer can markedly alter the anticipated stochiometric relationship between O2? trapped and apparent O2 consumed by these cells when they are exposed to O2? trapping agents.  相似文献

A wide range of concentrated random coil polysaccharide solutions have been assessed for textural attributes by a trained sensory panel. The only textural terms invoked to describe these model systems were ‘thickness’ and ‘stickiness’, which were shown to be highly correlated, and essentially identical numerically, using a ratio scaling technique. Viscosity (η) measurements over a wide range of shear rates (γ) for all these samples gave flow curves (log η versus log γ) of the same form. Differences in flow behaviour between samples could then be characterised completely by two parameters, the maximum viscosity at low shear rates (η0), and the shear rate (γ?0·1) at which η = solη010. A simple linear relationship was demonstrated between these two parameters and perceived thickness (T) or stickiness (S), irrespective of polysaccharide type. For Newtonian liquids, log T (or log S) varied linearly with log η. Hence the effective ‘in-mouth’ thickness of random coil polysaccharide solutions, in normal viscosity units, may be predicted directly from η0 and γ?0·1 by the simple relationship: log ηN = 1·13 log η0 + 0·45 logγ?0·1 ? 1·72 where ηN is the viscosity of a Newtonian solution which would be perceived as identical in thickness (and stickiness) to the polysaccharide solution.  相似文献

N-Phenylhydroxylamine is oxidized in aqueous phosphate buffer to nitrosobenzene, nitrobenzene, and azoxybenzene. Degradation is O2 dependent and shows general catalysis by H2PO4? (k1 = 2.3 M?2 sec?1) and PO4?3 (k2 = 2.3 × 105M?2 sec?1) or kinetically equivalent terms. An investigation of the effect of PGF2α induced parturition alone or in combination with neostigmine (NEO) on subsequent productivity was conducted in a total confinement swine production unit. Forty-five crossbred gilts and 106 sows were randomly assigned to one of six treatments: (1) 0 mg PGF2 α0 mg NEO, (2) 10 mg PGF2 α0 mg NEO, (3) 0 mg PGF2 α5 mg NEO, (4) 10 mg PGF2 α5 mg NEO, (5) 0 mg PGF2 α10 mg NEO, (6) 10 mg PGF2 α10 mg NEO. PGF2α injections (IM) were given 2 days prior to the mean expected farrowing day for this farm (day 112). NEO injections (IM) were given after the fourth pig born in gilt litters and after the fifth pig born in sow litters. Females injected with 0 or 10 mg PGF2α farrowed 71.7 ± 3.7 and 31.4 ± 3.4 hours after treatment, respectively (P<0.01). No other parameters measured were significantly affected by injection of PGF2α. These data show that PGF2α effectively induced parturition and did not adversely affect subsequent litter productivity. NEO was found to be ineffective in reducing total farrowing time (P>0.05).  相似文献

The stoichiometry of free NADPH oxidation in phenobarbital induced rabbit liver microsomes was measured by means of registering the rates of NADPH, H+ and O2 consumption and O2? and H2O2 production. ΔO2?:ΔH2O2 ratio is approximately I indicating that about half H2O2 results from O2? dismutation, the second half being formed directly. ΔNADPH:ΔH2O2 and ΔO2:ΔH2O2 ratios exceed I and therefore another product of the reaction is water. The fact that the ratio (ΔNADPH-ΔH2O2):(ΔO2-ΔH2O2) is 2 allows one to consider direct 4-electron O2 reduction as the major way of water formation rather than endogenous substrate hydroxylation.  相似文献

Oxygenation of heme-mercaptide as well as spectroscopic characteristics of the dioxygen complex formed have been studied. Absorption and magnetic circular dichroism spectra of the O2 complex support the retention of mercaptide in the heme fifth position. A release of O2? in the decomposition of the oxygenated complex and an independent formation of the latter from hemine-dimercaptide and O2? together with electron paramagnetic resonance and Mössbauer data support the O2 presence in the heme coordination sphere. The similarity of optical and magnetic circular dichroism spectra and the closeness of the KCOKO2 ratio for oxy-heme-mercaptide and oxycytochrome P450 unequivocally confirm the presence of an axial cystein mercaptide ligand in oxycytochrome P450.  相似文献

Short, mild treatments of sarcoplasmic reticulum vesicles with aqueous n-alcohols from methanol to n-heptanol caused an inhibition of calcium uptake and an enhancement of ATPase activity. The n-alcohol treatments increased both calcium-dependent (extra) ATPase activity and calcium-independent (basic) ATPase activity of vesicles. The apparent initial reaction rate of ATPase of n-alcohol-treated vesicles was about twice that of control vesicles. With increasing number (n) of carbon atoms of the n-alcohols, the maximum increment of ATPase activity increased, and both the alcohol concentration (NCa) required to inhibit calcium uptake by 50% and the alcohol concentration (NATPase) required to enhance ATPase activity by 50% of the maximum increment of ATPase activity decreased as follows.

The ratio, NATPase to NCa, was constant for all n values. The apparent free energy of binding of the methylene groups of n-alcohols to sarcoplasmic reticulum vesicles was evaluated (?796 cal/mole) and compared with data from the partition of n-alcohols in octanol and water (?670 cal/mole). The effects of n-alcohols on membrane vesicles are discussed on the basis of these data.  相似文献

Infrared spectra of N2O in a variety of solvents and in the brain of a dog under typical conditions of halothane-N2O anesthesia have been determined. The appearance or disappearance of N2O in the brain was readily followed as N2O was administered or withdrawn. The sites in brain were of two major types; one, with ν3 = 2229.8 ± 0.4 cm?1 and Δν12 = 13.0 ± 0.6 cm?1, is rather like the polar site in water and the other, with ν3 = 2216.8 ± 0.8 cm?1 and Δν12 = 9.6 ± 1.0 cm?1, is non-polar and is probably associated with membrane lipid. The significant variations in the antisymmetric stretch (ν3) of N2O as the polarity and other properties of the medium (solvent) vary make possible the characterization of in tissue sites occupied by this anesthetic.  相似文献

Kinetic studies on the RNase T1-catalyzed transesterification of 12 dinucleoside monophosphates, Np1N2 (N1 = A, C, and U; N2 = A, C, G, and U) at pH 5, 25 °C, and 0.2 m ionic strength, revealed that the catalytic efficiency (kcatKm) for GpN substrates (H. L. Osterman, and F. G. Walz, Jr., 1978, Biochemistry, 17, 4142) was ~106-fold greater than corresponding ApNs and at least 108-fold greater than corresponding CpNs and UpNs. The catalytic activity with ApN substrates survives phenol extraction which indicates (along with other criteria) that it is intrinsic to RNase T1 and is not due to trace contamination by other nucleases. Circumstantial evidence is presented which suggests that homologous GpN and ApN substrates bind productively at different sites on the enzyme. The results of steady-state kinetic studies of RNase T1 with IpNs (N = C and U) were compared with those for GpNs and indicated that the primary effect of the guanine 2-NH2 group is to enhance substrate binding at the primary recognition site by ~2.6 kcal/mol. Values of (kcatKm) showed the order NpC > NpU (N = A, G, and I) which evidences the existence of a subsite for the leaving nucleoside group that prefers cytidine: interactions at this subsite are reflected in kcat rather than Km.  相似文献

In the redheaded bunting Emberiza bruniceps, thyroidectomy inhibited premigratory fattening and nocturnal restlessness—two characteristics of avian migration—observed in caged birds during the premigratory period (March/April). Thyroxine (T4) and triiodothyronine (T3) administration in thyroidectomized birds stimulated locomotor activity and restored the loss in body weight. Annual variations in circulating thyroid hormone concentrations revealed a significant rise in T3T4 ratio prior to spring migration in both years studied. This increase in circulating T3T4 ratio may be associated with the development of migratory disposition in this bird. There was no increase in circulating T3T4 ratio prior to autumnal migration, however, plasma T4 increased significantly. Different thyroidal mechanisms are most likely involved in spring and fall migratory periods. While T3 remained low throughout, apart from the characteristic spring rise, high T4 levels in E. bruniceps were associated with periods of reproduction and molting, the latter coinciding partly with autumnal migration.  相似文献
