Rice (Oryza sativa L.) plantlets regenerated from callus (rice regenerants) were grownin vitro during the preparation stage either on a 1/4 strength N6 gellan gum (4 g l-1) medium without sucrose (SFM) or with 30 g l-1 sucrose (SCM), and under CO2 concentrations of 0.4, 2, 10, 50 or 100 mmol mol-1, a photoperiod of 24 h and a photosynthetic photon flux density (PPFD) of 125 mol m-2 s-1. Rice regenerants were also grownin vitro on SFM or SCM under CO2 concentration of 50 mmol mol-1, a photoperiod of 12 or 24 h and a PPFD of 80 or 125 mol m-2 s-1. All rice regenerants grew successfully on SFM under CO2 concentrations of 50 or 100 mmol mol-1. Increasing the CO2 concentration increased the survival percentage, shoot length and shoot and root dry weights of rice regenerants grown on SFM. Increasing CO2 concentration had no significant effect on the survival or growth of rice regenerants grown on SCM. Survival percentages of rice regenerants grown on SCM were less than 80% for each of the CO2 concentrations. A photoperiod of 24 h under CO2 enrichment improved the survival and growth of rice regenerants grown on SFM, and increased the survival percentage and shoot dry weight of rice regenerants grown on SCM.  相似文献

Physiological responses to elevated CO2 at the leaf and canopy-level were studied in an intact pine (Pinus taeda) forest ecosystem exposed to elevated CO2 using a free-air CO2 enrichment (FACE) technique. Normalized canopy water-use of trees exposed to elevated CO2 over an 8-day exposure period was similar to that of trees exposed to current ambient CO2 under sunny conditions. During a portion of the exposure period when sky conditions were cloudy, CO2-exposed trees showed minor (7%) but significant reductions in relative sap flux density compared to trees under ambient CO2 conditions. Short-term (minutes) direct stomatal responses to elevated CO2 were also relatively weak (5% reduction in stomatal aperture in response to high CO2 concentrations). We observed no evidence of adjustment in stomatal conductance in foliage grown under elevated CO2 for nearly 80 days compared to foliage grown under current ambient CO2, so intrinsic leaf water-use efficiency at elevated CO2 was enhanced primarily by direct responses of photosynthesis to CO2. We did not detect statistical differences in parameters from photosynthetic responses to intercellular CO2 (A net-C i curves) for Pinus taeda foliage grown under elevated CO2 (550 mol mol–1) for 50–80 days compared to those for foliage grown under current ambient CO2 from similar-sized reference trees nearby. In both cases, leaf net photosynthetic rate at 550 mol mol–1 CO2 was enhanced by approximately 65% compared to the rate at ambient CO2 (350 mol mol–1). A similar level of enhancement under elevated CO2 was observed for daily photosynthesis under field conditions on a sunny day. While enhancement of photosynthesis by elevated CO2 during the study period appears to be primarily attributable to direct photosynthetic responses to CO2 in the pine forest, longer-term CO2 responses and feedbacks remain to be evaluated.  相似文献

The interaction of extreme temperature events with future atmospheric CO2 concentrations may have strong impacts on physiological performance of desert shrub seedlings, which during the critical establishment phase often endure temperature extremes in conjunction with pronounced drought. To evaluate the interaction of drought and CO2 on photosynthesis during heat stress, one-year-old Larrea tridentata[DC] Cov. seedlings were exposed to nine days of heat with midday air temperature maxima reaching 53 °C under three atmospheric CO2 concentrations (360, 550 and 700 mol mol–1) and two water regimes (well-watered and droughted). Photosynthetic gas exchange, chlorophyll fluorescence and water potential responses were measured prior to, during and one week following the high temperature stress event. Heat stress markedly decreased net photosynthetic rate (A net), stomatal conductance (g s), and the photochemical efficiency of photosystem II (F v/F m) in all plants except for well-watered L. tridentata grown in 700 mol mol–1 CO2. A net and g s remained similar to pre-stress levels in these plants. In droughted L. tridentata, A net was ca. 2× (in 550 mol mol–1 CO2) to 3× (in 700 mol mol–1 CO2) higher than in ambient-CO2-grown plants, while g s and F v/F m were similar and low in all CO2 treatments. Following heat stress, g s in all well-watered plants rose dramatically, exceeding pre-stress levels by up to 100%. In droughted plants, g s and A net rose only in plants grown at elevated CO2 following release from heat. This recovery response was strongest at 700 mol mol–1 CO2, which returned to A net and g s values similar to pre-heat following several days of recovery. Extreme heat diminished the photosynthetic down-regulation response to growth at elevated CO2 under well-watered conditions, similar to the action of drought. Ambient-CO2-grown L. tridentata did not show significant recovery of photosynthetic capacity (A \max and CE) after alleviation of temperature stress, especially when exposed to drought, while plants exposed to elevated CO2 appeared to be unaffected. These findings suggest that elevated CO2 could promote photosynthetic activity during critical periods of seedling establishment, and enhance the potential for L. tridentata to survive extreme high temperature events.  相似文献

The relative increase with elevated CO2 of canopy CO2 uptake rate (A), derived from continuous measurements during the day, was examined in full-cover vegetative Lolium perenne canopies after 17 days of regrowth. The stands were grown at ambient (358±50 mol mol-1) and increased (626±50 mol mol-1) CO2 concentration in sunlit growth chambers. Over the entire range of temperature and light conditions (which were strongly coupled and increased simultaneously), A was on average twice as large in high compared to ambient CO2. This response (called M=A in high CO2/A in ambient CO2) could not be explained by changes in canopy conductance for CO2 diffusion (GC). In spite of interaction and strong coupling between temperature and light intensity, there was evidence that temperature rather than light determined M. Further, high CO2 treatment was found to alleviate the afternoon depression in A observed in ambient CO2. Leaf photosynthesis rate of the C4 species Paspalum plicatulum Michx was virtually CO2-saturated at normal atmospheric CO2 concentration but transpiration decreased as CO2 was increased above normal concentrations thereby increasing transpiration efficiency. To test whether this leaf response led growth to be CO2-sensitive when water supply was restricted, plants were grown in sealed pots of soil as miniature swards. Water was supplied either daily to maintain a constant water table, or at three growth restricting levels on a 5-day drying cycle. Plants were either in a cabinet with normal air (340 mol (CO2) mol-1 (air)) or with 250 mol mol-1 enrichment. Harvesting was by several cycles of defoliation.With abundant water supply high CO2 concentration did not cause increased growth, but it did not cause an increase in growth over a wide range of growth-limiting water supplies either. Only when water supply was less than 30–50% of the amount used by the stand with a water-table was there evidence that dry weight growth was enhanced by high CO2. In addition, with successive regrowth, the enhancing effect under a regime of minimal water allocations, became attenuated. Examination of leaf gas exchange, growth and water use data showed that in the long term stomatal conductance responses were of little significance in matching plant water use to low water allocation; regulation of leaf area was the mechanism through which consumption matched supply. Since high CO2 effects operate principally via stomatal conductance in C4 species, we postulate that for this species higher CO2 concentrations expected globally in future will not have much effect on long term growth.  相似文献

Desulfovibrio vulgaris (Marburg) was grown on H2 plus sulfate and H2 plus thiosulfate as the sole energy sources and acetate plus CO2 as the sole carbon sources. Conditions are described under which the bacteria grew exponentially. Specific growth rates () and molar growth yields (Y) at different pH were determined. and Y were found to be strongly dependent on the pH. Highest growth rates and molar growth yields were observed for growth on H2 plus sulfate at pH 6.5 (=0.15h-1; Y SO 4 2- =8.3g·mol-1) and for growth on H2 plus thiosulfate at pH 6.8 (=0.21h-1; Y S 2O 3 2 =16.9g·mol-1).The growth yields were found to increase with increasing growth rates: plots of 1/Y versus 1/ were linear. Via extrapolation to infinite growth rates a Y SO4 2- /max of 12.2g·mol-1 and a YS2O 3 2- /max of 33.5g·mol-1 was obtained.The growth yield data are interpred to indicate that dissimilatory sulfate reduction to sulfide is associated with a net synthesis of 1 mol of ATP and that near to 3 mol of ATP are formed during dissimilatory sulfite reduction to sulfide.  相似文献

Sitka spruce [Picea sitchensis (Bong.) Carr.] seedlings were grown for 3 years in an outside control plot or in ambient (355 mol mol-1) or elevated (ambient + 350 mol mol-1) atmospheric CO2 environments, within open top chambers (OTCs) at the Institute of Terrestrial Ecology, Edinburgh. Sequential harvests were carried out at the end of each growing season and throughout the 1991 growing season, five in all. Plants grown in elevated CO2 had, (i) 35 and 10% larger root/shoot ratios at the end of the first and third season, respectively, (ii) significantly higher summer leader extension relative growth rates, which declined more rapidly in early autumn than ambient grown plants, (iii) after three growing seasons a significantly increased mean annual relative growth rate, (iv) consistently lower foliar nutrient concentrations, and (v) after two growing seasons smaller total projected needle areas. Plants grown inside OTCs were taller, heavier and had a smaller root/shoot ratio than those grown outside the chambers. There was no effect of CO2 concentration on Sitka spruce leaf characteristics, although leaf area ratio, specific leaf area and leaf weight ratio all fell throughout the course of the 3 year experiment.  相似文献

We compared flavonoids in green, mature, and senescing flag leaves of wheat grown under ambient (AC - 370 mol mol-1) and elevated (EC - 550 mol mol-1) concentrations of CO2 in a FACE (Free Air CO2 Enrichment) system. The concentrations of flag leaf flavonoids (e.g., isoorientin and tricin) decreased to one third in mature leaves, and the majoritary isoorientin almost disappeared in senescing leaves. Flavonoid concentrations increased in green well-developed flag leaves under EC (46 % isoorientin and 55 % tricin), whereas the differences disappeared in mature and senescing flag leaves. Predictions of changes in litter phenolic concentrations and their effects on decomposition rates under EC based on changes in green leaves need to be revised.  相似文献

The direct and indirect effects of increasing levels of atmospheric carbon dioxide (CO2) on plant nitrogen (N) content were studied in a shortgrass steppe ecosystem in northeastern Colorado, USA. Beginning in 1997 nine experimental plots were established: three open-top chambers with ambient CO2 levels (approximately 365 mol mol–1), three open-top chambers with twice-ambient CO2 levels (approximately 720 mol mol–1), and three unchambered control plots. After 3 years of growing-season CO2 treatment, the aboveground N concentration of plants grown under elevated atmospheric CO2 decreased, and the carbon–nitrogen (C:N) ratio increased. At the same time, increased aboveground biomass production under elevated atmospheric CO2 conditions increased the net transfer of N out of the soil of elevated-CO2 plots. Aboveground biomass production after simulated herbivory was also greater under elevated CO2 compared to ambient CO2. Surprisingly, no significant changes in belowground plant tissue N content were detected in response to elevated CO2. Measurements of individual species at peak standing phytomass showed significant effects of CO2 treatment on aboveground plant tissue N concentration and significant differences between species in N concentration, suggesting that changes in species composition under elevated CO2 will contribute to overall changes in nutrient cycling. Changes in plant N content, driven by changes in aboveground plant N concentration, could have important consequences for biogeochemical cycling rates and the long-term productivity of the shortgrass steppe as atmospheric CO2 concentrations increase.  相似文献

Norway spruce and red oak trees were planted directly into the soil and enclosed in open-top chambers. For 2 years the trees were exposed to both ambient and elevated CO2 concentrations (700 mol mol-1) and during this time variations in nutrient concentrations were studied. CO2-treated plants had decreases in global leaf concentrations of nitrogen, potassium, calcium and manganese for both species. When different areas of the foliage were analysed however, the response showed much variability between the respective sites and between species. Furthermore the nutrient concentrations changed differently as the plant material aged and this change showed inter-treatment differences. These results show how it may be important to analyse plant material of different ages and at different cell sites when studying nutrient levels.  相似文献

The photoautotrophic micropropagation of potato cv. Russet Burbank was investigated. Single node microcuttings were grown for four weeks on Murashige and Skoog (MS) medium with or without sucrose (30 g l–1) in the growth room at 21/19 °C day/night temperature, with 16-h photoperiod at 150 mol m–2 s–1, with or without supplemental CO2 at 1500 l l–1. A 20% increase in the number of nodes per stem (from 7.5 to 9.4) and a 50% increase in stem dry weight were observed in cultures grown on media with sucrose and in CO2 enriched atmosphere comparing to the conventionally micropropagated cultures or the cultures grown photoautotrophically on media without sucrose but in air supplemented with 1500 l l–1CO2. Stems of these cultures (from media with sucrose in CO2 enriched air) almost doubled in length the stems of cultures from the other two treatments. No significant differences were observed between Control (MS medium supplemented with sucrose, 30 g l–1) and photoautotrophic cultures coming from MS medium with no sucrose grown under 1500 l l–1 of CO2. Photoautotrophic cultures produced stems averaging 43.3 mm, with 7 nodes and weighing 9.2 mg (dry weight), similar to conventionally grown in vitro cultures (47.9 mm with 7.5 nodes, 9.7 mg dry weight). Growers may consider photoautotrophic culturing of potato in areas where the high sterility levels are difficult to maintain. Supplementing air in the growth room with 1500 l l–1 of CO2 could be beneficial for potato plantlet production even on media containing sucrose since it significantly improved quality, size and biomass of produced plantlets, speeding up the multiplication.  相似文献

The relationship between leaf photosynthetic capacity (p n, max), net canopy CO2- and H2O-exchange rate (NCER and E t, respectively) and canopy dry-matter production was examined in Lollium perenne L. cv. Vigor in ambient (363±30 l· l-1) and elevated (631±43 l·l-1) CO2 concentrations. An open system for continuous and simultaneous regulation of atmospheric CO2 concentration and NCER and E t measurement was designed and used over an entire growth cycle to calculate a carbon and a water balance. While NCERmax of full-grown canopies was 49% higher at elevated CO2 level, stimulation of p n, max was only 46% (in spite of a 50% rise in one-sided stomatal resistance for water-vapour diffusion), clearly indicating the effect of a higher leaf-area index under high CO2 (approx. 10% in one growing period examined). A larger amount of CO2-deficient leaves resulted in higher canopy dark-respiration rates and higher canopy light compensation points. The structural component of the high-CO2 effect was therefore a disadvantage at low irradiance, but a far greater benefit at high irradiance. Higher canopy darkrespiration rates under elevated CO2 level and low irradiance during the growing period are the primary causes for the increase in dry-matter production (19%) being much lower than expected merely based on the NCERmax difference. While total water use was the same under high and low CO2 levels, water-use efficiency increased 25% on the canopy level and 87% on a leaf basis. In the course of canopy development, allocation towards the root system became greater, while stimulation of shoot dry-matter accumulation was inversely affected. Summary Elodea canadensis grows over a wide range of inorganic carbon, nutrient, and light conditions in lakes and streams. Affinity for HCO 3 - use during photosynthesis ranged from strong to weak in Elodea collected from seven localities with different HCO 3 - and CO2 concentrations. The response to HCO 3 - was also very plastic in plants grown in the laboratory at high HCO 3 - concentrations and CO2 concentrations varying from 14.8 to 2,200 M. Bicarbonate affinity was markedly reduced with increasing CO2 concentrations in the growth medium so that ultimately HCO 3 - use was not detectable. High CO2 concentrations also decreased CO2 affinity and induced high CO2 compensation points (360M CO2) and tenfold higher half-saturation values (800 M CO2).The variable HCO 3 - affinity is probably environmentally based. Elodea is a recently introduced species in Denmark, where it reproduces only vegetatively, leaving little opportunity for genetic variation. More important, local populations in the same water system had different HCO 3 - affinities, and a similar variation was created by exposing one plant collection to different laboratory conditions.Bicarbonate use enabled Elodea to photosynthesize rapidly in waters of high alkalinity and enhanced the carbon-extracting capacity by maintaining photosynthesis above pH 10. On the other hand, use of HCO 3 - represents an investment in transport apparatus and energy which is probably not profitable when CO2 is high and HCO 3 - is low. Root proliferation into nutrient rich zones is an important mechanism in the exploitation of soil nutrients by plants. No studies have examined atmospheric CO2 effects on cotton (Gossypium hirsutum L.) root distribution as affected by localized phosphorus (P). Cotton plants were grown in a Troup sand (loamy, thermic Grossarenic Kandiudults) using 17.2-l containers placed in open top field chambers (OTC) under ambient (360 mol mol–1) or enriched (720 mol mol–1) atmospheric CO2 concentrations for 40 days. Equivalent amounts of P were added (150 mg P per kg of soil) to 100, 50, 25, 12.5, and 6.25% of the total soil volume; control containers with no added P were also included. Under extremely low P (controls), cotton was unresponsive to CO2 enrichment. In treatments with both fertilized and unfertilized soil volumes, root proliferation was greater in the unfertilized soil under elevated CO2 conditions. Stimulation of root growth occurred in the P-fertilized soil fraction; the pattern of stimulation was similar under both CO2 levels. Under ambient CO2, cotton plant response was positive (shoot mass, and total root mass and length) when soil P was confined to relatively small proportions of the total soil volume (6.25 and 12.5%). Summary Seedlings of five tropical trees, Cecropia obtusifolia, Myriocarpa longipes, Piper auritum, Senna multijuga and Trichospermum mexicanum, were grown both as individuals, and in competition with each other at ambient (350) and two levels of elevated CO2 (525 and 700 l l-1) for a period of 111 days. Growth, allocation, canopy architecture, mid-day leaf water potential and soil moisture content were assessed three times over this period for individually grown plants, and at the end of the experiment for competitively grown plants. In addition, leaf photosynthesis and conductance were assessed for the individually grown plants midway through the experiment, and light profile curves were determined for the competitive arrays at three stages of development. Elevated CO2 did not affect photosynthesis or overall growth of the individually-grown plants but did affect canopy architecture; mean canopy height increased with CO2 in Piper and Trichospermum and decreased in Senna. Stomatal conductance decreased slightly as CO2 increased from 350 to 525 l l-1 but this had no significant effect upon whole plant water use of leaf water potential. Soil moisture content for the individuals increased marginally as CO2 increased, but this did not occur in the competitive arrays. There was a marked effect of CO2 upon species composition of the competitive arrays; Senna decreased in importance as CO2 increased while Cecropia, Trichospermum and Piper increased in importance. Stepwise regression analysis using competitive performance as the independent variable, and the various morphological and physiological parameters measured on the individually grown plants as independent variables, suggested that canopy height was the single most important variable determining competitive ability. Also significant were photosynthetic rate (particularly at low light levels) and allocation to roots early in the experiment. Light profiles in the canopy revealed that less than 15% of incident light penetrated to the level of mean canopy height. Results suggest that competition for light was the major factor determining community composition, and that CO2 affected competitive outcome through its affect upon canopy architecture.This study was supported by a grant from the US Department of Energy  相似文献

Ponderosa pine (Pinus ponderosa Dougl. ex Laws.) seedlings were exposed to near ambient or elevated CO2 (average concentrations during the last growing season 446 versus 699 mol mol–1), combined with low or elevated O3 for three seasons. Ozone exposure during the last growing season (accumulated dose above threshold 0.06 mol mol–1) was 0.05 versus 26.13 mol mol–1 h. Needles of the youngest age class were harvested after the dormancy period. Ozone exposure decreased needle contents of chlorophyll a, chlorophyll b, and ascorbate, and resulted in a more oxidized total ascorbate and a more de-epoxidized xanthophyll cycle pool irrespective of the CO2 level. Trees under elevated CO2 had a more oxidized glutathione pool and lower chlorophyll a content. Contents of glutathione, tocopherol, and carotenoids were not affected by the CO2 or O3 treatments. There were no interactive effects between elevated CO2 and elevated O3 on any of the parameters measured. The results suggest that elevated atmospheric CO2 concentration does not compensate for ozone stress by increasing antioxidative capacity in ponderosa pine.  相似文献

Strawberry (Fragaria ananassaDuch. cv. Fengxiang) plantlets were cultured under two in vitroenvironments for rooting, and then acclimatized under two ex vitroirradiance conditions. At the end of rooting stage plant height, fresh weight and specific leaf area of T1-plants grown under high sucrose concentration (3 sucrose), low photosynthetic photon flux density (30 mol m–2 s–1) and normal CO2 concentration (350–400 l l–1) were significantly higher than those of T2-plantlets grown under low sucrose concentration (0.5), high photosynthetic photon flux density (90 mol m–2 s–1) and elevated CO2 concentration (700–800 l l–1). But T2-plantlets had higher net photosynthetic rate (Pn), effective photochemical quantum yield of PSII (PSII), effective photosynthetic electron transport rate (ETR), photochemical quenching (qP) and ratio of chlorophyll fluorescence yield decrease (Rfd). After transfer, higher irradiance obviously promoted the growth of plantlets and was beneficial for the development of photosynthetic functions during acclimatization. T2-plantlets had higher fresh weight, leaf area, PSII and ETR under higher ex vitroirradiance condition.  相似文献

Atmospheric CO2 levels are expected to exceed 700 mol mol–1 by the end of the 21st century. The influence of increased CO2 concentration on crop plants is of major concern. This study investigated water- and nitrogen-use efficiency (WUE and NUE, respectively, were defined by the amount of biomass accumulated per unit water or N uptake) of spring wheat (Triticum aestivumL.) grown under two atmospheric CO2 concentrations (350 and 700 mol mol–1), two soil moisture treatments (well-watered and drought) and five nitrogen amendment treatments. Results showed that enriched CO2 concentration increased canopy WUE, and more N supply led to higher WUE under the increased CO2. Canopy WUE was significantly lower in well-watered treatments than in drought treatment, but increased with the increased N supply. Elevated CO2 reduced the apparent recovery fraction of applied N by the plant root system (Nr, defined as the ratio of the increased N uptake to N applied), but increased the NUE and agronomic N efficiency (NAE, defined as the ratio of the increased biomass to N applied). Water limitation and high N application reduced the Nr, NUE and NAE, indicating a poor N efficiency. In addition, there was a close relationship between the root mass ratio and NUE. Canopy WUE was negatively related to the root mass ratio and NUE. Our results indicated that CO2 enrichment enhanced WUE more at high N application, but increased NUE more when N application was less.  相似文献

This study reports survival and physiological responses of micropropagated Ceratonia siliqua L. cvs. Galhosa and Mulata plants during ex vitro acclimatization under ambient (AC; 330 mol mol–1) or elevated (EC; 810 mol mol–1) CO2 concentration and a photosynthetic photon flux density of 125 mol m–2 s–1. CO2 enrichment during acclimatization did not improve survival rate that was around 80 % for both treatments. Eight weeks after ex vitro transplantation, photosynthetic capacity and apparent quantum yield in acclimatized leaves were higher in comparison with those in in vitro-grown leaves, without any significant difference between CO2 treatments. Chlorophyll content increased after acclimatization. However, EC led to a decrease in the total amount of chlorophyll in new leaves of both cultivars, compared to those grown at AC. Soluble sugars and starch contents were not markedly affected by growth EC, although starch had significantly increased after transfer to ex vitro conditions. EC induced an increase in the stem elongation and in the effective life of leaves, and a decrease in the number of new leaves.  相似文献
