Summary The role of D-genome chromosomes in the expression of net photosynthesis in wheats was analysed with the nullitetrasomic and ditelosomic lines of the bread wheat cultivar Chinese Spring. The two arms of chromosome 3 D and the short arm of chromosome 6 D control major mechanisms of photosynthesis. The effect of chromosome 6 D can be thoroughly compensated by that of its homoeologues of genomes A or B, contrary to what can be observed for chromosome 3 D. Chromosome 7 D is responsible for the low photosynthesis of flag leaves developed under high irradiances in genotypes possessing the D-genome, as the likely result of ontogeny or of a loss in adaptability to irradiance.  相似文献

Summary Evolutionary and ontogenetic variation of six seedling esterases of independent genetic control is studied in polyploid wheats and their diploid relatives by means of polyacrylamide gel electrophoresis. Four of them are shown to be controlled by homoeoallelic genes in chromosomes of third, sixth and seventh homoeologous groups.The isoesterase electrophoretic data are considered supporting a monophyletic origin of both the primitive tetraploid and the primitive hexaploid wheat from which contemporary taxa of polyploid wheats have emerged polyphyletically and polytopically through recurrent introgressive hybridization and accumulation of mutations. Ancestral diploids belonging or closely related to Triticum boeoticum, T. urartu, Aegilops speltoides and Ae. tauschii ssp. strangulata are genetically the most suitable genome donors of polyploid wheats. Diploids of the Emarginata subsection of the section Sitopsis, Aegilops longissima s.str., Ae. sharonensis, Ae. searsii and Ae. bicornis, are unsuitable for the role of the wheat B genome donors, being all fixed for the esterase B and D electromorphs different from those of tetraploid wheats.  相似文献

Twenty-five genotypes of early CIMMYT hexaploid wheat were screened for salt tolerance in a glasshouse experiment using photosynthetic capacity and water relation parameters as selection criteria. Under salt stress (150 mM NaCl) the genotypes Frontana, Norin-10, Mayo-54, Noreste-66, and Yaktana-54 excelled all other lines in shoot dry mass, and Na(20)TPP, Penjamo-62, Inia-66, Frontana, Siete Cerros, and Jaral-66 in grain yield per plant in both absolute and relative (percent of control) terms. Although net photosynthetic rate (P N) declined in all genotypes due to salt stress, it was not helpful in discriminating among genotypes according to salt tolerance. Similarly, no positive relationships of salt tolerance of the genotypes with stomatal conductance, leaf water potential, or turgor pressure were found. Every genotype used its own specific mechanism to tolerate salt stress. However, a large amount of variation in salt tolerance observed in 25 early CIMMYT wheat genotypes can be of considerable practical value for improving salt tolerance in the existing commercial hexaploid wheats. Photosynthesis was studied in relation to the carbohydrate status in intact leaves of the C4 plant Amaranthus edulis. The rate of leaf net CO2 assimilation, stomatal conductance and intercellular partial pressure of CO2 remained constant or showed little decline towards the end of an 8-h period of illumination in ambient air (340 bar CO2, 21% O2). When sucrose export from the leaf was inhibited by applying a 4-h cold-block treatment (1°C) to the petiole, the rate of photosynthesis rapidly decreased with time. After the removal of the cold block from the petiole, further reduction in photosynthetic rate occurred, and there was no recovery in the subsequent light period. Although stomatal conductance declined with time, intercellular CO2 partial pressure remained relatively constant, indicating that the inhibition of photosynthesis was not primarily caused by changes in stomatal aperture. Analysis of the leaf carbohydrate status showed a five- to sixfold increase in the soluble sugar fraction (mainly sucrose) in comparison with the untreated controls, whereas the starch content was the same. Leaf osmotic potential increased significantly with the accumulation of soluble sugars upon petiole chilling, and leaf water potential became slightly more negative. After 14 h recovery in the dark, photosynthesis returned to its initial maximum value within 1 h of illumination, and this was associated with a decline in leaf carbohydrate levels overnight. These data show that, in Amaranthus edulis, depression in photosynthesis when translocation is impaired is closely related to the accumulation of soluble sugars (sucrose) in source leaves, indicating feedback control of C4 photosynthesis. Eucalyptus camaldulensis Dehnh. seedlings inoculated with Pisolithus tinctorius (Pers.) Coker & Couch and Thelephora terrestris Ehrl. per Fr. were grown in well watered soil (s –0.03 MPa) or subjected to a long-term soil water stress of up to –1.0 MPa over 13-week period in a glasshouse. After 13 weeks, all seedling containers were watered to field capacity and then water was withheld from the E. camaldulensis seedlings to induce a short-term drought. Diurnal measurements of seedling photosynthesis rate (A), leaf stomatal conductance (g) and leaf water potential (p) were completed before, during, and after the short term drought. Although they were growing in an equal soil volume, photosynthesis rate (A), leaf stomatal conductance and leaf water potential (p) of larger seedlings with P. tinctorius ectomycorrhizae were similar to those of smaller seedlings colonized with T. terrestris during the short-term drought period. Seedlings inoculated with Pisolithus tinctorius maintained higher photosynthesis rates over the course of the short-term drought. Thus, P. tinctorius ectomycorrhizae appear to be more efficient than those of T. terrestris in assisting seedlings to maintain gas exchange and photosynthesis under limited soil moisture conditions.  相似文献

The nitrogen-fixing grey alder,Alnus incana (L.) Moench, has a potential use in forest soil restoration and as part of energy forestry plantations. As a first step to estimate nitrogen fixation byA. incana under field conditions we performed studies on nitrogenase activity and its possible relation to abiotic factors and growth of the alders. Nitrogenase activity was measured as acetylene reduction activity (ARA) on eleven 1-year-old seedlings ofA. incana inoculated with a local source ofFrankia and planted in an experimental plot located in Umeå, northern Sweden. Each alder was planted into an open-ended cylinder which was closed with a gas tight lid around the stem base to serve as cuvette during ARA measurements. Propane served as tracer gas. ARA was measured in the middle of the day at 15 occasions during 26 June to 29 September 1987. Growth was recorded as leaf area and top shoot length at each ARA measurement until the end of August. Weather conditions were recorded for the whole growing season.Maximal ARA was recorded in late July or early August and ranged from 1.86 to 106mol C2H4plant–1h–1. Final leaf area ranged from 0.022 to 0.124 m2. A relationship between ARA and the number of hours of sunshine during the same day was observed. ARA in relation to soil temperature increased during the study period, except for the last measurements. ARA in relation to leaf area was initially high but decreased later on. It is suggested that as leaves got older their contribution to photosynthesis per unit leaf area decreased and their potential to deliver nitrogen for retranslocation within plant increased. Both of these events would cause reduced ARA per unit leaf area. The data on ARA, growth, and abiotic factors taken together supported the view that sunshine and weather conditions affected photosynthesis and thereby delivery of assimilates to the nodules.  相似文献

Four fluorescence parameters [Fv/Fm = the intrinsic efficiency of energy conversion via photosystem 2 (PS2); Fv/Fm= the efficiency of energy conversion via PS2 in the light; P = fraction of absorbed radiant energy utilized for photosynthesis; and D = fraction of absorbed radiant energy dissipated as heat] were measured on leaves of seven species of succulents having epidermal windows. While the function of leaf windows has reportedly been to increase absorption of radiant energy and, hence, the rate of photosynthesis in these species, recent evidence indicates that this translucent portion of epidermal tissue, lacking chlorophyll, may also result in photoinhibition in these species, especially for those with growth habits aboveground. Species with aboveground and belowground growth habits were compared with their leaf windows covered with reflective tape and with windows unobstructed. Results showed no increase in photoinhibition for these species resulting from the radiant energy penetrating the window tissue. Although the efficiency of the photosynthetic mechanism was not significantly influenced by the additional radiant energy provided by the window for individual species, there were significant differences in the efficiencies of radiant energy capture (Fv/Fm) and utilization (P) between the two growth habits. Species with an aboveground growth habit were less efficient in radiant energy utilization compared with the species having a belowground growth habit.  相似文献

Summary Variation in high molecular weight (HMW) glutenin subunit composition among 167 accessions of dicoccum wheat (Triticum turgidum L. var. dicoccum Schrank) of diverse origins was investigated using one-dimensional sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). A total of 20 alleles were identified, and 9 of them were found to be different from those previously detected by Payne and Lawrence (1983 b) in hexaploid wheat (Triticum aestivum L.). The newly discovered alleles enhance the genetic variability available to improve the industrial quality of wheats and some of them may facilitate basic research on the relationship of industrial quality with HMW glutenin subunit number. The novel variants include a GLU-A1 encoded subunit which has higher molecular mass than any other so far described in tetraploid and hexaploid wheats, and a null GLU-B1 allele. Dicoccums containing neither GLU-A1- nor GLU-B1-encoded subunits were also identified. A comparison of the mean number of HMW glutenin subunits contained in various primitive and modern domesticated wheats of different ploidy levels and the identification of wheats containing no HMW glutenin subunits suggest that the occurrence of null GLU-1 alleles in these species depends on chance rather on an inherent tendency on the part of modern polyploid wheats to suppress the activity of redundant GLU-1 genes.  相似文献

Summary The nuclei of 12 common wheats (genome constitution AABBDD) were placed into the cytoplasms of Aegilops kotschyi and Ae. variabilis (both CuCuSvSv) by repeated backcrosses. Using these nucleus-cytoplasm hybrids, male sterility-fertility restoration relationship was investigated. Male sterility was expressed by these cytoplasms only in Slm, Splt and Mch. The other nine common wheat nuclei gave normal fertility against these cytoplasms. These cytoplasms were compared with the Triticum timopheevi cytoplasm that is now widely used in the hybrid wheat breeding program in order to investigate their effects on important agronomic traits of the 12 common wheats: The kotschyi and variabilis cytoplasms were as good as the timopheevi cytoplasm in this respect.The F1 hybrid between (kotschyi)- or (variabilis)-Splt and CS showed normal fertility. Segregation of the fertiles and steriles in their F2 generations followed the simple Mendelian fashion, i.e., 3 fertile1 sterile. Thus, the fertility restoration in this case is mainly controlled by a single dominant gene which will be designated as Rfv1. To determine its location, ditelo-lBS and -lBL of CS were crossed as male parents to male sterile (kotschyi)- and (variabilis)-Splt. The F1 hybrids between the male sterile Spit's and CS ditelo-lBS became male fertile, while those between the male sterile Spit's and CS ditelo-lBL became completely male sterile. Thus, the location of the gene Rfv1 has been determined to be on the short arm of chromosome lB of CS. Furthermore, a close relationship between the fertility-restoring genes and the nucleolus organizer region was pointed out.Finally, the schemes of breeding the male sterile lines of a cultivar with these cytoplasms, and its maintainer line were formulated. The following two points were considered as the advantages of the present male sterility-fertility restoration system over that using the timopheevi cytoplasm in breeding hybrid wheat: (1) easier fertility restoration in F1 hybrids, and (2) no need of breeding the restorer line.This work was supported by a Grand-in-Aid from the Ministry of Education, No. 386002. To elucidate the phylogenetic relationships and cytoplasmic types, restriction endonuclease fragment patterns of chloroplast (cp) and mitochondrial (mt) DNAs isolated from two different accessions of Dasypyrum villosum (L.) candargy were compared with those of tetraploid wheat (Triticum durum Desf., PI265007), hexaploid wheat (Triticum aestivum L., cv Chinese Spring), Aegilops longissimum (S. and M., in Muschli) Bowden and Hordeum vulgare L. T. aestivum and T. durum had identical restriction patterns for their cp and mtDNAs in digestions with four different enzymes. Likewise, no differences were found between the restriction fragment patterns of two accessions of D. villosum. But, there were distinct differences in chloroplast and mitochondrial DNA restriction fragment patterns between D. villosum and tetraploid and hexaploid wheats. A. longissimum (G609) showed a similar pattern to those wheats for PstI digestion of cpDNA. Organellar DNA from Hordeum vulgare (cv Himalaya) showed a distinctly different restriction pattern from those of wheat and D. villosum. The effects of iso-osmotic salinity and drought stresses on leaf net photosynthetic rate (P N) in two wheat (Triticum aestivum L.) cultivars BR 8 and Norin 61, differing in drought tolerance, were compared. In drought-sensitive Norin 61, the decline of P N was larger than that in drought-tolerant BR 8. Under NaCl treatment, P N decreased in two phases similarly in both cultivars. In the first phase, photosynthetic depression was gradual without any photochemical changes. In the second phase, photosynthetic depression was rapid and accompanied with a decline of the energy conversion efficiency in photosystem 2 (PS2). Our observations suggest that the osmotic factor may induce a gradual depression of photosynthesis due to stomatal closure under both stress treatments. However, under NaCl treatment, a ionic factor (uptake and accumulation of excess Na+) may have direct effects on electron transport and cause more severe photosynthetic depression. The drought tolerance mechanism of BR 8 was insufficient to maintain single-leaf photosynthesis under salinity.  相似文献

Summary Intergeneric hybrids between Triticum aestivum cv Chinese Spring (2n=6x=42, AABBDD) and Agropyron michnoi Roshev. (2n=4x=28, PPPP) were obtained by embryo culture. Their spike characteristics were similar to those of common wheat but, unlike their parents, they were long-awned. The average meiotic chromosome pairing at MI of F1 hybrids was: 6.39 I +3.75 rodII+8.64 ringII+0.81 III+0.30 IV+0.04 V, the bivalent and multivalent formation of which was much higher than expected from the genomic formulae. It is especially worthwhile to note that the F1 hybrids were self-fertile, self set being 0.15%, and seeds were easily obtained from the backcross of f1 plants with hexaploid and tetraploid wheats; here the seed set was more than 20.0%. The polyploid taxa and the position of A. Michnoi in Agropyron are discussed.  相似文献

The response of net photosynthesis and apparent light respiration to changes in [O2], light intensity, and drought stress was determined by analysis of net photosynthetic CO2 response curves. Low [O2] treatment resulted in a large reduction in the rate of photorespiratory CO2 evolution. Lightintensity levels influenced the maximum net photosynthetic rate at saturating [CO2]. These results indicate that [CO2], [O2] and light intensity affect the levels of substrates involved in the enzymatic reactions of photosynthesis and photorespiration. Intracellular resistance to CO2 uptake decreased in low [O2] and increased at low leaf water potentials. This response reflects changes in the efficiency with which photosynthetic and photorespiratory substrates are formed and utilized. Water stress had no effect on the CO2 compensation point or the [CO2] at which net photosynthesis began to saturate at high light intensity. Summary Polymorphism and inheritance of wheat storage protein, gliadin, of durum (macaroni) and carthlicum wheats have been studied. Analysis of gliadin in 78 cultivars and in F2 seeds of intercultivar crosses of durum wheat revealed three different chromosome 1A-encoded blocks of components similar to those found in common wheat (GLD1A2, GLD1A18, GLD1A19). Most of the durum cultivars studied had these three blocks; GLD1A2 was also frequent in common wheat. In contrast, all chromosome 1B-encoded blocks of durum clearly differed in component composition from those found in common wheat. Therefore, durum could not be an ancestor or a derivate of recent bread wheat. Analysis of gliadin in the collection of carthlicum wheat (14 accessions) revealed several suspected chromosome 1A, 1B, and 6A-controlled blocks, some of which were similar to those in common wheat, while others were different. Therefore, carthlicum is likely to be an ancestor or a derivate of some forms of bread wheat. There were also chromosome 1A and 6A-, but not 1B-encoded blocks which were identical in durum and carthlicum wheats. The results confirm that all three wheats share the same genome A, but emphasize the heterogeneity of genotypes among donors of this genome. Discovery of identical blocks in tetraploids and hexaploids indicates polyphyletic [from different genotypes of donor (s)] origin of these wheats.  相似文献

Summary Leaves of two field growing co-occuring perennial shrubs (drought-deciduous Diplacus aurantiacus and the evergreen Heteromeles arbutifolia) from the Californian chaparral were exposed to small doses of SO2. During this exposure the leaf environment was manipulated to determine how the presence of SO2 alters the response of gas exchange to other environmental stresses. The data show that no direct changes in stomatal conductance (g) or net assimilation rate (A) could be attributed to short-term (7 h) SO2 (4.2 mol m-3, 0.1 l l-1) exposure. D. aurantiacus leaves possessed features which demonstrate that they were sensitive to changes in environment e.g. light flux and atmospheric relative humidity. The interspecific differences in stomatal sensitivity to water vapour were extremely important, as relative humidity is a major factor influencing carbon fixation and the rate of pollutant absorption. Conditions of high relative humidity and high xylem water potentials are suggested to pre-dispose leaves of D. aurantiacus to greater pollutant doses than the more stomatally-conservative evergreen, H. arbutifolia. In the presence of SO2 there was some indication of increased g for both D. aurantiacus and H. arbutifolia as W became smaller. This SO2-effect was only obvious as increasing atmospheric humidity induced further stomatal opening. In this work, we analyzed 54 domestic cultivars of hexaploid (common) wheat Triticum aestivum L. (AABBDD genome) and accessions of tetraploid wheats of the Timopheevi group (AAGG) and rye Secale cereale (RR) using 21 SNP markers for common wheat. It was demonstrated that application of the SNP markers developed and verified for particular common wheat cultivars in allele-specific PCR analysis of other cultivars with different geographic origins could lead to an incorrect estimation of the similarity between the genotypes tested. The studied SNP markers of common wheat are inappropriate for analyzing genomes of other cereal species, in particular, T. timopheevii wheats and rye S. cereale.  相似文献

Differences between two maize and two triticale genotypes grown in low soil compaction (LSC), moderate soil compaction (MSC) and severe soil compaction (SSC) and with a limited (D) or excess (W) soil water content were observed as a decrease in shoot (S) and root (R) biomass, leaf greening (SPAD) and increase in membrane injury (LI), root and leaf water potential (ψ), photosynthesis (Pn), transpiration (E) and stomata conductance (gS). Close correlations between ψL and ψR, and between differences ψL and ψRψ) were found. Drought or waterlogging with LSC conditions in both maize genotypes resulted in higher WUE than in control plants (LSC C), but under the SSC WUE declined. However, for triticale differences in WUE, between treatments were small and insignificant. In general, changes in markers were greater for genotypes sensitive to the soil compaction (Ankora, CHD-12) than in resistant ones (Tina, CHD-247) and were higher in seedlings grown under SSC conditions.

Summary The relation between daily maximal rates of net photosynthesis and plant water status was studied during a dry season on irrigated and non-irrigated, naturally growing, perennial wild plants.Species were examined which differ in phenology, leaf anatomy and morphology: Hammada scoparia, Artemisia herba-alba, Zygophyllum dumosum, and Reaumuria negevensis. Prumus armeniaca which was growing in the run-off farm at Avdat and which has mosomorphic leaves was included in the comparison. All plants differed in their seasonal change in plant water status, and in their seasonal change in daily maximal net photosynthesis. Rates of CO2 uptake were not uniquely related to simultanously measured leaf water potentials. Daily maximal rates of net photosynthesis of non-irrigated plants, and the difference between maximal CO2 uptake of irrigated and non-irrigated plants were examined in relation to pre-dawn water potential. Maximal net photosynthesis rates decreased very rapidly with decrease in pre-dawn water potential or, for Hammada scoparia, they decreased even with a constant level of pre-dawn water potential. Consequently, it was considered necessary to include both time and water potential in a parameter bar day describing the accumulated drought stress of the plants. All species showed the same relation between relative maximal net photosynthesis and drought experience as determined by cumulative daily addition of pre-dawn water potentials for the non-irrigated plants since the last rain.  相似文献

The effect of leaf water potential () on net CO2 assimilation rate (A), stomatal conductance (g), transpiration (E) and water-use efficiency (WUE) was measured for three cultivars of cacao (Theobroma cacao L.) seedlings during three recurrent drought cycles. Net assimilation varied greatly at high water potentials, but as dropped below approximately -0.8 and -1.0 MPa, A was reduced to less than 1.5 mol CO2 m-2 s-1. The relation between g and A was highly significant and conformed to an asymptotic exponential model, with A approaching maximal values at stomatal conductances of 55–65 mmol H2O m-2 s-1. Net assimilation varied linearly (r=0.95) with transpiration, and the slope of the A-E relation (WUE) was approximately 3.0 mol CO2 mmol-1 H2O throughout the range of stomatal conductances observed. C i was insensitive to water stress, even though both g and A were strongly affected. Under the experimental conditions used here, mesophyll photosynthesis did not appear to control g through changes in C i. As stress intensified within each drying cycle, WUE of nonirrigated seedlings did not decline relative to that of controls even though CO2 and water vapor exchange rates underwent large displacements. Summary The chromosomes of the tetraploid wheats Triticum timopheevi (Genome AAGG) and T. araraticum (Genome AAGG) were C-banded at mitosis. The identity of the banded and unbanded chromosomes was then established by firstly making comparisons with the hexaploid species T. zhukovskyi which has the genome formula AAAAGG. Secondly, the meiotic pairing in F1 hybrids between T. timopheevi and diploid wheats was examined by means of C-banding. The results showed that the banded chromosomes belonged to the G genome, while the unbanded chromosomes belonged to the A genome. Only one of the two pairs of satellited chromosomes had strong heterochromatic bands. The relationship between the genomes of T. timopheevi and T. dicoccum (Genome AABB) was then assessed at meiosis in hybrids between these species, using the techniques of C-banding and in situ hybridisation of a cloned ribosomal RNA gene probe. It was concluded that there were differences both in the amount and distribution of heterochromatin and also translocation differences between the species.  相似文献
