Lygus spp. Hahn (Hemiptera: Miridae) are serious pests of a wide variety of economically important crops in North America. A European parasitoid, Peristenus digoneutis Loan (Hymenoptera: Braconidae), released in the northeastern USA for the biological control of Lygus, has successfully established in nine states and in eastern Canada, including southeastern Ontario, southern Quebec and Nova Scotia. To determine the extent to which P. digoneutis has dispersed and established in mirid populations in Ontario, a single-step multiplex PCR assay, designed to differentiate European species P. digoneutis and P. relictus from native North American Peristenus species, was used to identify parasitoid larvae dissected from field-collected Miridae. A total of 222 parasitoid larvae were analysed with the multiplex assay. Most (172) were identified as members of the P. pallipes Curtis complex; however, three from the Niagara region were identified as P. digoneutis. Specimens that did not amplify using the Peristenus multiplex PCR assay were screened with a PCR primer set designed to detect Leiophron (Hymenoptera: Braconidae) species, and most were confirmed to be Leiophron larvae. This study confirms the presence of P. digoneutis species in southern Ontario, and is another example of the utility of molecular methods for the detection of newly introduced or dispersed parasitoids. The presence of exotic P. digoneutis in southern Ontario may expedite future releases to augment already-established populations. Following such releases, the Peristenus multiplex PCR assay will be a useful component of post-release studies to evaluate the success of the biological control programme for Lygus plant bugs in Canada.  相似文献

Lygus lineolaris populations in the northeastern USA have been markedly reduced by the introduced parasitoid Peristenus digoneutis. A 3-year study in NJ where P. digoneutis has been established for many years demonstrated that parasitism of Lygus nymphs in strawberries averaged 30% (mostly P. digoneutis). Peristenus digoneutis Loan and Peristenus stygicus Loan, parasitoids of the European tarnished plant bug Lygus rugulipennis Poppius, are established in the United States for biological control of native North American Lygus species, and are being considered for deliberate release in Canada. High lifetime fecundity of parasitoids is considered a desirable attribute of biological control agents and therefore, an understanding of parasitoid reproductive biology is required. In the present study, the potential lifetime fecundity of both agents was compared under laboratory conditions to estimate the potential impact of Peristenus species on Lygus. Synovigenic P. digoneutis and P. stygicus females oviposited most actively in the first two weeks of their lifetime, with a maximum average daily oviposition rate after five days. The maximum number of eggs laid per day was 83 eggs for P. stygicus, and 36 eggs for P. digoneutis. P. digoneutis has an average potential lifetime fecundity of 385 ± 35 SE eggs produced over 22 ± 3 SE days. In contrast, P. stygicus females have a 50% higher mean potential lifetime fecundity reaching 782 ± 65 SE eggs over 28 ± 1 SE days. A positive correlation between lifetime fecundity and body size was found only for P. stygicus, and both species showed a significant relationship between lifetime fecundity and oviposition period. The present study demonstrates that the fecundity of P. digoneutis and P. stygicus is considerably higher than previously reported. Based on these findings, P. stygicus appears to be the most effective biological control agent for Lygus lineolaris (Palisot de Beauvois) when only fecundity is taken into consideration.  相似文献

Releases of Peristenus digoneutis against Lygus spp. in North America have been conducted for many years; however, no published procedures for mass production of the biological control agent were available. A laboratory rearing method was developed using Lygus lineolaris as the host to enhance establishment efforts and provide large numbers of wasps for inundative releases into high value fruit crops. Experiments were conducted to determine optimum host:parasitoid density and rearing temperature. The effects of nymph:wasp ratios and temperature on parasitism and wasp survival showed a 20:1 ratio at 20°C provided high parasitism (256 parasitized nymphs/wasp over lifetime) and excellent wasp survival of 27 days. Experiments on diapause-inducing conditions for P. digoneutis demonstrated that fluctuating temperatures of 23°C (day) and <16°C (night) and corresponding photo phases of 16 h light, for rearing parasitized nymphs, produced 100% diapausing parasitoids whereas non-diapausing parasitoids were only produced at more than 16 h light. Furthermore, parasitized Lygus nymphs need to be transferred to short day conditions no later than 10 days after parasitism to produce diapausing parasitoids. Critical life stages for exposure to conditions inducing diapause, the egg, first and second instar parasitoid larva, occurred from 0 to 10 days at 24°C constant temperature. Increased time in cold storage reduced the number of days to first emergence of parasitoids from diapausing cocoons when transferred to warm temperatures. The optimum storage time for diapausing P. digoneutis is between 25 and 44 weeks, depending upon the length of time that cocoons remain at warm conditions prior to chilling.  相似文献

Lygus Hahn (Hemiptera: Miridae) are serious pests of agricultural and greenhouse crops throughout North America. In Europe, bivoltine Peristenus Förster (Hymenoptera: Braconidae) species have a significant impact on Lygus populations. Release and establishment of European P. digoneutis Loan in Lygus lineolaris (Palisot de Beauvois) populations in northeastern USA has renewed interest in the intended liberation of European parasitoids for Lygus control in Canada. Accurate identification of natural enemies is the cornerstone of biological control but conventional methods for identifying Peristenus species and estimating parasitism rates rely on tedious and time-consuming dissection and rearing methods. The present study describes species-specific PCR primers for three species of Peristenus, and the use of a multiplex PCR assay to detect P. digoneutis and P. stygicus Loan eggs and larvae from Lygus rugulipennis Poppius nymphs. Results indicate that the primers amplify uniquely sized, species-specific PCR products for the three species and are capable of detecting single eggs in parasitized nymphs within 3 days post-parasitism. Using a multiplex PCR assay, the primers maintain specificity and sensitivity, and allow detection of each of the three species in a single reaction. Although molecular diagnostics have previously been used in the identification of parasitoids and estimation of parasitism rates, this is the first time a single-step multiplex PCR protocol has been described.  相似文献

Several species of Lygus (Heteroptera: Miridae: Mirini) are serious crop pests in North America where their parasitism rate by native nymphal parasitoids is generally lower than in Europe. Peristenus relictus (Ruthe) (formerly P. stygicus Loan) (Hymenoptera: Braconidae: Euphorinae) is the predominant nymphal parasitoid of several Lygus spp. in the warm Mediterranean region and has been a candidate for introduction against Lygus hesperus Knight and L. lineolaris (Palisot de Beauvois) in the southern US. We report a rapid, sensitive, and specific PCR-based assay for diagnosis of P. relictus immature stages within Lygus nymphs that entails three steps: DNA extraction, PCR of the partial mitochondrial COI gene, and agarose gel electrophoresis. The PCR-based methodology is species-specific because the target DNA of other sympatric, congeneric species was not amplified with use of the primers developed for P. relictus diagnosis. The sensitivity of the PCR method, assessed through spike tests, was established by the detection of a ratio of 1:10,000 P. relictus DNA to Lygus DNA. Molecular diagnosis of parasitism of field collected nymphs is achievable in one day, eliminating the need to rear nymphs to obtain adult parasitoids for morphological identification.  相似文献

Peristenus digoneutis Loan (Hymenoptera: Braconidae) was introduced to the US for biological control of the tarnished plant bug, Lygus lineolaris (Palisot de Beauvois) (Hemiptera: Miridae), and has since spread through much of the northeast. The purpose of this study was to determine if P. digoneutis and a native congener, Peristenus pallipes (Curtis), parasitize L. lineolaris in strawberry (where it is a key pest), and what factors relate to parasitism levels. During 1997–1999 we monitored parasitism on 17 strawberry farms in 14 counties in eastern and western New York State. We found that in eastern NY (where P. digoneutis has been established since the early 1990s), overall mean parasitism was 19.7% (ranging from 0 to 70%), mostly by P. digoneutis. Mean parasitism was significantly lower (12.3%, ranging from 0 to 58%) in western NY (where P. digoneutis was first recorded in 1999), and was mostly by P. pallipes. P. pallipes parasitism was significantly lower in eastern than western NY, suggesting the potential for competitive interaction with P. digoneutis. The insecticide regime of a farm was an important factor influencing parasitism rate, which was 5- to 6.5-fold higher on organic or casually sprayed farms than on intensely treated farms, though pest density under these three regimes was not significantly different. L. lineolaris density, and parasitism rate in nearby alfalfa and abandoned fields were also significant factors for parasitism in strawberry.  相似文献

Lygus spp. (Heteroptera: Miridae) are serious pests of numerous field and fruit crops in North America. In an effort to reduce crop damage, parasitoids known to attack these species in Europe were introduced into the USA beginning in the 1970s. Permanent populations of the nymphal endoparasitoid Peristenus relictus (Hymenoptera: Braconidae) were established at two locations in California during the 2000s. Both populations are associated with significant reductions in lygus bugs attacking alfalfa, commercially produced strawberries grown organically and in wild vegetation. Beginning in 2009, in an effort to determine the extent to which P. relictus has spread from the Sacramento Valley and Monterey Bay region, populations of lygus bug were sampled at increasing distances from their original release sites. P. relictus has dispersed at least 213 km in the central region of California and 150 km along the coastal mountains. These respective populations have averaged 16.6 km/year and 17.7 km/year since they were released. While not directly examined, the spread of P. relictus south into the central and major growing region of California, the San Joaquin Valley, where previous releases have failed, suggests this population may be evolving greater heat tolerance, relative to the populations originally introduced into California.  相似文献

We report a long-term survey summarising the seasonal variation in species composition of Peristenus species, and their levels of parasitism in an assemblage of Lygus spp., over more than 10 years in southern Alberta in western Canada. Plants sampled include alfalfa (Medicago sativa L.), canola (Brassica napus L.), and stands of different weed and grass species commonly found on crop field edges. Three native Peristenus species (Peristenus dayi, Peristenus mellipes, and Peristenus howardi) were recorded in 2003–2011 and 2013–2015. In laboratory trials, P. mellipes emerged 13 days earlier than P. howardi. Degree-days for field-collected individuals suggested a sequential occurrence during the growing season: P. dayi appears early in the season, followed by P. mellipes, and later on by P. howardi. Studies of species composition and phenology of native parasitoids of insect pests provide valuable baseline information when considering introduction of exotic agents, and contribute to the management and improvement of conservation biological control by endemic natural enemies in agricultural systems.  相似文献

Lygus spp. (tarnished plant bugs) are generalist herbivores and occur as pests on a wide range of crops. In the development of conservation biological control strategies for Lygus spp. in Sweden, more information is needed on the impact of different natural enemies. In this study, we determined the occurrence and the degree of parasitism on adults and nymphs of the most common Lygus species in alfalfa, barley, red clover and oilseed rape in Uppsala in Central Sweden and in Umeå in Northern Sweden. Nymphs and adults of Lygus spp. were collected by sweep netting for estimation of their parasitism level (by dissection) and identification of parasitoid species (from reared Lygus specimens). The dominant Lygus species in both locations was L. rugulipennis (75–99%). Parasitism by Phasia obesa (Tachnidae) on overwintering Lygus adults was recorded in almost every field sampled at both locations. The parasitism level was low at overwintering sites but increased in arable fields during summer. Lygus nymphs collected in Umeå were parasitised by Peristenus pallipes (Braconidae) and in Uppsala by P. relictus, P. pallipes and P. varisae. The hyperparasite Mesochorus globulator (Ichneumonidae) was recorded in Umeå.  相似文献

High numbers of tarnished plant bugs [Lygus lineolaris (Palisot)], were once common in alfalfa, as was a low level of parasitism (9%) by the native Peristenus pallipes (Curtis). After the bivoltine European parasite Peristenus digoneutis Loan became well established, average parasitism of the first and second generations increased to 64%, and tarnished plant bug numbers dropped by 65%. This reduced host density eventually caused a decline in total parasitism by both parasite species to 22%. A few P. digoneutis also attacked the alfalfa plant bug, Adelphocoris lineolatus (Goeze), but did not reduce this pest or increase its parasitism rate. At another location, where P. digoneutis is not established, parasitism of first generation alfalfa plant bugs, an adventive (accidently introduced) pest, was increased to 21% by the introduced univoltine parasite, Peristenus conradi Marsh, and a slight reduction in the pest may have resulted. P. digoneutis did not parasitize the meadow plant bug, Leptopterna dolabrata (L.), an adventive pest of forage grasses, so did not affect this mirid or its parasite. Neither introduced parasite eliminated the native parasites of the tarnished or alfalfa plant bugs. The narrow host ranges of the braconid parasites of mirid nymphs are contrasted with the broad host range of the native tachinid parasite [Phasia robertsoni (Towns.)] of adult mirids. The major changes in mirid abundance and their mortality by parasites that slowly occurred during this 19-year study demonstrate the need for long-term field research, to adequately document and understand these complex interactions.  相似文献

Lygus Hahn plant bugs (Hemiptera: Miridae) are serious pests of a wide variety of economically important crops in North America. European Peristenus digoneutis Loan and P. relictus Ruthe (Hymenoptera: Braconidae) are being considered for release in Canada as part of a classical biological control program for Lygus. The attractiveness of different host plants to European Peristenus has not been addressed, but may be an important consideration prior to parasitoid release. Lygus rugulipennis Poppius nymphs were collected in the Northern Temperate Atlantic (NTA) ecoregion on red clover (Trifolium pratense L.; Fabaceae) and chamomile (Matricaria recutita L.; Asteraceae), and in the Western European Broadleaf Forest (WEBF) ecoregion on red clover and alfalfa (Medicago sativa L.; Fabaceae). Parasitism levels and parasitoid species were determined using a multiplex PCR assay for P. digoneutis, P. relictus, and P. pallipes Curtis. Mean parasitism levels in L. rugulipennis were 45-49% in the NTA ecoregion and 25-32% in the WEBF ecoregion. However, in neither ecoregion were parasitism levels and parasitoid species compositions significantly different in nymphs from different host plant species. Furthermore, multiparasitism was low despite the fact that P. digoneutis and P. relictus share the same host species.  相似文献

Cotesia flavipes Cameron (Hymenoptera: Braconidae) was first imported into Kenya in 1991 from Pakistan for control of Chilo partellus (Swinhoe) (Lepidoptera: Crambidae). First releases were made at the Kenya coast in 1993 and a few recoveries of the parasitoid were made the following year. Additional foreign exploration for C. flavipes was conducted in central India in 1996 and 1998, which resulted in additional importation of the parasitoid for subsequent releases in eastern and southern Africa. Region-wide releases commenced with releases in Mozambique in 1996, Somalia in 1997 and Uganda in 1998. By 2005, many releases had been made in 10 countries in East and Southern Africa with establishment being reported in all of them except Eritrea but including Ethiopia where releases had never been made. This paper describes the progress made in the release and establishment of C. flavipes in East and Southern Africa and quantifies the rate of spread from the initial release sites in Kenya.  相似文献

  • 1 Sirex noctilio is a woodwasp native to Eurasia and Northern Africa and has recently been found infesting pines in eastern North America. Its pest status in this new range is not yet known, although it is an important pest in other areas where it has been introduced. Pinus spp. in North America are hosts to several native and alien species of subcortical insects. Interactions between the woodwasp and these species may influence its distribution or the characteristics of its life history, thus affecting its population dynamics over time.
  • 2 Sixty S. noctilio‐infested Pinus spp. were felled in Ontario, Canada, and all phloem‐feeding and woodboring insects were collected and identified from each 1‐m section of the tree.
  • 3 Sirex noctilio was in a tree alone 10% of the time but commonly shared the tree with subcortical beetles, such as Tomicus piniperda, Pissodes nemorensis, Ips grandicollis, Gnathotrichus materiarius and Monochamus carolinensis. The woodwasp was distributed throughout the tree stem and this distribution overlapped with that of the beetles.
  • 4 Fewer but larger S. noctilio males emerged from trees with beetles compared with those without, although there was no statistical difference in females.
  • 5 These findings suggest that co‐habiting beetles could negatively affect S. noctilio population dynamics over time. The potential mechanisms for this interaction are discussed.

  1. The Western conifer seed bug Leptoglossus occidentalis, a native insect of North America, was accidentally introduced in Europe in the late 1990s. Since then, it has spread rapidly. Biological control could provide an efficient management option but natural enemies of the pest have been poorly examined in Europe.
  2. In this study, we exposed sentinel egg masses and collected naturally laid egg masses in southern France in 2016 and 2017, to identify the egg parasitoids of L. occidentalis and investigate their potentials.
  3. Three egg parasitoids were detected: Anastatus bifasciatus, Ooencyrtus pityocampae and Ooencyrtus telenomicida. The overall parasitism was low compared to that observed in the native range with 6.4% of all eggs being parasitized, while 17.1% of egg masses carried at least one parasitized egg. The total number of parasitized egg masses was similar between parasitoid species, but the mean number of parasitized eggs per egg mass was highest for A. bifasciatus (5.57 vs. 1.25 for Ooencyrtus spp.).
  4. Sentinel egg masses underestimated the parasitism compared to natural egg masses (respectively, 1.42% and 7.71%).
  5. Our results suggested that the three generalist parasitoids detected can respond in a Leptoglossus egg density-dependent manner, but this requires further investigations.

Meningeal worm (Parelaphostrongylus tenuis), a common nematode parasite in white-tailed deer (Odocoileus virginianus) and pathogenic for several species of ungulates in eastern North America, is not known to occur in the west. Heads of 1,902 white-tailed deer were examined for adult meningeal worm to determine geographic distribution of the parasite in Saskatchewan and Manitoba (Canada) and North Dakota (USA). Finding the parasite in a deer in eastern Saskatchewan near the Manitoba border established the current northern and western limits in Canada. Prevalence of infection was < 1, 18.6, and 8.2% in Saskatchewan, Manitoba, and North Dakota, respectively. Infected deer occurred throughout southern Manitoba and eastern North Dakota. Distribution appears to have changed little since the last published survey for P. tenuis in the region in 1972. We examined precipitation, temperature, deer density, and forest cover as likely correlates to prevalence and distribution of P. tenuis. Deer management units used for hunting purposes were the scale of analysis in the three jurisdictions. Presence of P. tenuis was positively correlated with precipitation during frost-free periods and deer density, and it was negatively correlated with winter and spring temperatures. Landscapes with > 25 and < 75% forest cover were most likely to have infected deer. Low rainfall and low density of white-tailed deer likely influence the westernmost limit of P. tenuis.  相似文献

The parasitoid wasp Ibalia leucospoides is native to the northern hemisphere and has been introduced to the southern hemisphere as a biological control agent for the invasive woodwasp Sirex noctilio. Two subspecies of the parasitoid, Ibalia leucospoides leucospoides (Palearctic distribution) and Ibalia leucospoides ensiger (Nearctic distribution), were introduced and are reported to have hybridized.
  2. Despite extensive records of the numbers and origins of the wasps imported into the southern hemisphere, nothing is known regarding their current population diversity. We investigated the genetic variation of I. leucospoides in its native and introduced ranges using mitochondrial (COI) and nuclear (ITS) markers.
  3. Mitochondrial DNA diversity in the introduced range was limited, with only five haplotypes, although sequence divergence between these haplotypes was high. Similarly, the ITS rDNA sequences revealed multiple clades present in the introduced range.
  4. These results reflect introductions from a wide geographical range but where genetic bottlenecks have possibly reduced the genetic diversity. The data further reflect the origin of the I. leucospoides populations in South America and South Africa from New Zealand or Australia. We found no evidence of hybridization between the two subspecies of the parasitoid in its introduced range, and no evidence that I. leucospoides ensiger has established outside its native range.

Landscape complexity influences patterns of animal dispersal, which in turn may affect both gene flow and the spread of pathogens. White‐nose syndrome (WNS) is an introduced fungal disease that has spread rapidly throughout eastern North America, causing massive mortality in bat populations. We tested for a relationship between the population genetic structure of the most common host, the little brown myotis (Myotis lucifugus), and the geographic spread of WNS to date by evaluating logistic regression models of WNS risk among hibernating colonies in eastern North America. We hypothesized that risk of WNS to susceptible host colonies should increase with both geographic proximity and genetic similarity, reflecting historical connectivity, to infected colonies. Consistent with this hypothesis, inclusion of genetic distance between infected and susceptible colonies significantly improved models of disease spread, capturing heterogeneity in the spatial expansion of WNS despite low levels of genetic differentiation among eastern populations. Expanding our genetic analysis to the continental range of little brown myotis reveals strongly contrasting patterns of population structure between eastern and western North America. Genetic structure increases markedly moving westward into the northern Great Plains, beyond the current distribution of WNS. In western North America, genetic differentiation of geographically proximate populations often exceeds levels observed across the entire eastern region, suggesting infrequent and/or locally restricted dispersal, and thus relatively limited opportunities for pathogen introduction in western North America. Taken together, our analyses suggest a possibly slower future rate of spread of the WNS pathogen, at least as mediated by little brown myotis.  相似文献
