The claw closer muscle of Neohelice granulata (Grapsoidea, Varunidae): a morphological and histochemical study. —Acta Zoologica (Stockholm) 92 : 126–133. The claw closer muscle of Neohelice granulata was studied according to histological, histochemical, and morphometrical criteria. Adult male crabs in intermoult stage were collected from Mar Chiquita Lagoon (Buenos Aires, Argentina). Muscle fibers show evident striations and oval‐elongated nuclei with loose chromatin. The loose connective tissue among muscle fibers consists of cells and fibers embedded in an amorphous substance. Muscle histochemistry reveals two slow fiber types: ‘A’ and ‘B’. Prevailing A fibers are larger, and they usually show, with respect to B type, a weaker reaction to whole techniques. Fibers with short (SS), intermediate (IS), and long sarcomeres (LS) appear in the claw closer muscle, being the LS fibers predominant. Concluding, the histochemical and morphometrical characteristics of the claw closer muscle fibers of N. granulata are indicative of slow fibers. The slow A type (low resistant to fatigue) prevails.  相似文献

The fibres of superficial and deep abductor muscles of the pectoral fins of the stripped weakfish, Cynoscion guatucupa have been studied using histochemical techniques: succinic dehydrogenase (SDH) for mitochondria, periodic acid–Schiff (PAS) for glycogen, myosin‐adenosintriphosphatase (mATPase) to identify different fibre types based on the contraction speed and modified ATPase to identify capillaries. The fibre diameters were measured, and the capillaries of the main fibre types – red, pink and white— were counted. The two muscles showed both macroscopically and microscopically two well‐differentiated zones with predominant white fibres. The area of insertion of muscles into the fin rays had red, pink and white fibres. The origin zone of the muscle into the bone was composed by white fibres only. Both zones of white muscle evidenced a mosaic of small, medium and large polygonal white fibres. Red, pink and white muscles showed a wide histochemical diversity of fibre subtypes. The area per peripheral capillary increased from the red to the white muscles. Due to the predominance of white fibres, the pectoral fins of C. guatucupa were mainly involved in rapid movements to stop/discontinue and stabilize the body during swimming.  相似文献

The abundance of Aulacoseira granulata (Ehrenburg) Simonsen and Gloeocystis planctonica (West & G.S.West) Lemmermann was assessed during the summers of 2005 and 2010 in the eutrophic Fox River, Wisconsin, USA. In both years, a mid‐summer bloom of G. planctonica was followed by the rapid growth of A. granulata. Laboratory experiments in which A. granulata was grown in cell‐free filtrate of a G. planctonica culture revealed that the growth of A. granulata was stimulated in the G. planctonica‐treated medium relative to controls. This effect was detected when dormant A. granulata cells were used as the source culture for the experiment but not when actively growing cells were used. Dormant A. granulata also grew more rapidly in river water collected after the 2010 G. planctonica bloom relative to river water collected before the bloom. These results suggest that the summer bloom of A. granulata in the river was stimulated by G. planctonica. This relationship can be described as stimulated rejuvenation, an interaction where the transition of an algal resting stage into active growth is triggered by exposure to another species.  相似文献

The estuarine grapsid crabs Chasmagnathus granulata and Cyrtograpsus angulatus belong to the most typical and dominant inhabitants of brackish coastal lagoons in southeastern South America. In a combined laboratory and field investigation of juvenile growth, we measured the increase in body size in these species under controlled conditions as well as in field experiments (in Mar Chiquita lagoon, Argentina), seasonal changes in size frequency distribution of a natural population, and growth related changes in selected morphometric traits of male and female juveniles (relations between carapace width, carapace length, propodus height and length of the cheliped, and pleon width). At 24°C, Cy. angulatus grew faster than Ch. granulata; it reached the crab-9 instar (C9; 13 mm carapace width) after 92 days, while Ch. granulata required 107 days to reach the C8 instar (7.4 mm). At 12°C, growth ceased in both species. The pleon begins to show sexual differences in the C5 (Cy. angulatus) and C8 instar (Ch. granulata), respectively, while the chelae differentiate earlier in Ch. granulata than in Cy. angulatus (in C4 vs C6). In the field, growth was maximal in summer, and was generally faster than in laboratory cultures. However, there is great individual variability in size (about 25% even in the first crab instar) and in size increments at ecdysis, increasing throughout juvenile growth. Our data indicate that, in the field, small-scale and short-term variations in feeding conditions, temperature, and salinity account for an extremely high degree of variability in the absolute and relative rates of growth as well as in the time to sexual differentiation. Profilicollis chasmagnathi Holcman-Spector, Mañé-Garzón & Dei-Cas, 1977 (Acanthocephala: Polymorphidae) has been reported to parasitise different grapsid species as intermediate hosts along the South Atlantic shores, i.e. Cyrtograpsus angulatus (Dana) and Neohelice granulata (Dana) in Uruguay and Cyrtograpsus altimanus (Rathbun) in Argentina. Larvae of a similar acanthocephalan described as Profilicollis antarcticus Zdzitowiecki, 1985 were recorded in the crab Hemigrapsus crenulatus (Milne-Edwards) from an estuarine habitat on the Southeast Pacific shore in Chile. Earlier studies have questioned the specific assignation of the Chilean estuarine populations of Profilicollis Meyer, 1931. The aim of this study was to re-examine the identification of these acanthocephalans by means of morphological and molecular analyses of cystacanths of Profilicollis spp. gathered from C. angulatus, N. granulata, C. altimanus and H. crenulatus. Our analyses showed that a single species of Profilicollis, P. chasmagnathi, parasitises these four crab species. The assessment of specimens from the South Shetlands Islands, the type-locality of P. antarcticus, is needed before formally proposing that P. antarcticus is a junior subjective synonym of P. chasmagnathi.  相似文献

Summary Fibre types in the costal region of the diaphragm muscle of several mammalian species with widely different respiratory rates were examined microphotometrically for succinate dehydrogenase (SDH) activity. Mean activities indicated no significant (p>0.05) difference between the type I and IIA fibres for any of the species examined. SDH activities in type IIB fibres were significantly lower (p<0.05) than either the type I or type IIA fibres in the cat, guinea pig, rat and rabbit whereas in the mouse no difference was found. The dog had no classical type 1B fibres. Analysis of the distribution of SDH activities by fibre type indicated a wide scattering of scores with no distinct separation between fibre types. Large differences in SDH activity were noted between species. Mean SDH activities were highest in the mouse and rat, intermediate in the rabbit and guinea pig and lowest in the cat and dog. These data suggest an association between respiratory rate and aerobic oxidative potential of the various fibre types in diaphragms of the species examined.  相似文献

 Myofibrillar ATPase (mATPase), succinate dehydrogenase (SDH) and α-glycerophosphate dehydrogenase (GPD) activities and cross-sectional area (CSA) were measured in fibres of rat medial gastrocnemius muscle using quantitative histochemistry. The same fibres were typed immunohistochemically using monoclonal antibodies specific to selected myosin heavy chain (MHC) isoforms. The values of mATPase, SDH, GPD and CSA formed a continuum, but significant differences in mean values were observed among fibre types of presumed homogeneous MHC content. Type I fibres had the lowest mATPase activity, followed in rank order by type IIA<type IID/X<type IIB. Type IIA fibres had the highest SDH activity, followed in rank order by type IID/X>type I>type IIB. The mean GPD activity was consistently ranked according to fibre type such that type IIB>type IID/X >type IIA>type I. Type IIA fibres were the smallest, type IIB fibres were the largest and types I and IID/X were of intermediate size. Significant interrelationships between mATPase, SDH, GPD and CSA values were found on a fibre-to-fibre basis. Consequently, discrimination of fibres according to their MHC content was possible on the basis of their mATPase, SDH, GPD and CSA profiles. These intrafibre interrelationships suggest that the MHC isoform is associated with phenotypic differences in contractile, metabolic and size properties of muscle fibre types. Summary Some histochemical parameters of the three main fibre types of rat vastus lateralis muscle were studied. Succinic dehydrogenase (SDH), creatine kinase (CK), sarcotubular ATPase (SR-ATPase) and mitochondrial ATPase activities were demonstrated in serial sections. The three fibre types, recognised by the distribution pattern of SDH activity, all show high CK activity. However, red Type I oxidative fibres when examined for ATPase and ATPase dependent CK activity, show distinct heterogeneity revealing sub-populations within the same homogeneous fibre type. Three distinct patterns were recognised in the red Type I fibres depending on the distribution of the final reaction product. The present histochemical evidence confirms the fact that subdivision of mammalian skeletal muscle into three fibre types is only approximate and probably more than three types exist.  相似文献

Burcucumbers (Sicyos angulatus) showing necrotic leaf spots were found in several locations in Korea during 2008–2012. The causal agent was a fungus that was identified as Cercospora citrullina, based on morphological characteristics as well as sequences of the ribosomal DNA internal transcribed spacer region. Although C. citrullina has been known to attack various cucurbitaceous plants, this is the first report of this fungus on S. angulatus.  相似文献

Summary Cryostat sections incubated for myofibrillar ATPase, SDH, LDH, and -GPDH as well as p-phenylene-diamine stained semithin sections were used to define muscle fibre types in the trunk musculature of the cod (Gadus morhua, L.).Three zones (superficial, intermediate, deep) containing different muscle fibre types are present within both epaxial and hypaxial parts of each myomere subjacent to the lateral line.Atypical relations concerning myofibrillar ATPase activity probably reflects instability of myosin during storage of frozen tissue. The histochemical reaction does not distinguish between myofibrillar and mitochondrial ATPase in cod muscle.Based on ATPase and SDH activities, seven different histochemical profiles of muscle fibres can be identified in trunk musculature of this teleost fish. Attempts to homologize these fibre types with those in cyclostomes or those in higher animals proved futile. The higher number of histochemically defined muscle fibre types in cod might be explained by developmental processes and an admixture of immature fibres throughout life.  相似文献

Cotton provides us the most important natural fibre. High fibre quality is the major goal of cotton breeding, and introducing genes conferring longer, finer and stronger fibre from Gossypium barbadense to Gossypium hirsutum is an important breeding strategy. We previously analysed the G. barbadense fibre development mechanism by gene expression profiling and found two homoeologous fibre‐specific α‐expansins from G. barbadense, GbEXPA2 and GbEXPATR. GbEXPA2 (from the DT genome) is a classical α‐expansin, while its homoeolog, GbEXPATR (AT genome), encodes a truncated protein lacking the normal C‐terminal polysaccharide‐binding domain of other α‐expansins and is specifically expressed in G. barbadense. Silencing EXPA in G. hirsutum induced shorter fibres with thicker cell walls. GbEXPA2 overexpression in G. hirsutum had no effect on mature fibre length, but produced fibres with a slightly thicker wall and increased crystalline cellulose content. Interestingly, GbEXPATR overexpression resulted in longer, finer and stronger fibres coupled with significantly thinner cell walls. The longer and thinner fibre was associated with lower expression of a number of secondary wall‐associated genes, especially chitinase‐like genes, and walls with lower cellulose levels but higher noncellulosic polysaccharides which advocated that a delay in the transition to secondary wall synthesis might be responsible for better fibre. In conclusion, we propose that α‐expansins play a critical role in fibre development by loosening the cell wall; furthermore, a truncated form, GbEXPATR, has a more dramatic effect through reorganizing secondary wall synthesis and metabolism and should be a candidate gene for developing G. hirsutum cultivars with superior fibre quality.  相似文献

Neohelice granulata (previously known as Chasmagnathus granulata and C. granulatus) is a burrowing semiterrestrial crab found in the intertidal zone of estuaries, salt marshes and mangroves of the South-western Atlantic Ocean. Beginning in the late 1989s, an explosion of publications appeared in international journals dealing with its ecology, physiology, toxicology and behavior. A bibliometric analysis using the Scopus database allowed detecting 309 papers that deal with this species during the period 1986–2009. The number of papers per year increased continuously, reaching a mean annual value of 22.6 during the last 5 years; a great majority of them were authored by researchers from Argentina and Brazil. Neohelice granulata has become now one of the most studied crab species, after Carcinus maenas, Callinectes sapidus, Scylla serrata and Cancer pagurus and C. magister, and it can be considered as an emergent animal model for biochemical, physiological and ecological research.  相似文献

Cyrtograpsus angulatus andChasmagnathus granulata (Grapsidae) are the two dominant decapod crustacean species in the outer parts of Mar Chiquita Lagoon, the southernmost in a series of coastal lagoons that occur along the temperate Atlantic coasts of South America. Distribution and habitat preferences (water and sediment type) in these crab species were studied in late spring. There is evidence of ontogenetic changes in habitat selection of both species. Recruitment ofC. angulatus takes place mainly in crevices of tube-building polychaete (Ficopomatus enigmaticus) “reefs” and, to a lesser extent, also in other protected microhabitats (under stones). In the latter, mostly somewhat larger juveniles were found, suggesting that these are used as a refuge for growing individuals. Adults are most frequently found on unprotected muddy and sandy beaches.C. angulatus was found in all parts of Mar Chiquita Lagoon, including freshwater, brackish, and marine habitats.C. granulata, in contrast, was restricted to the lower parts of the lagoon, where brackish water predominates and freshwater or marine conditions occur only exceptionally. It showed highest population density on “dry mud” flats and inSpartina densiflora grassland, where it can build stable burrows and where high contents of organic matter occur in the sediment. Such habitats are characterized by mixed populations of juveniles (including newly settled recruits) and adults, males and females (including a high percentage of ovigerous). Unstable “wet mud” as well as stony sand were found to be inhabited by chiefly adult populations, with only few ovigerous females. In “dry mud” flats, the proportion of males increased vertically with increasing level in the intertidal zone, showing a significantly increasing trend also in their average body size. These observations may be explained by higher resistance of males, in particular of large individuals, to desiccation, salinity, and temperature stress occurring in the upper intertidal. However, an opposite, or no such, tendency was found in the distribution of ovigerous and non-ovigerous females, respectively. With increasing distance from the water edge, salinity increased and pH decreased significantly inC. granulata burrows, whereas temperature showed no consistent tendency within the intertidal gradient. A highly significant linear relationship (r=−0.794; P<0.001) between salinity and pH in water from crab burrows is described. This regression line is significantly different from one that had been observed in water from the lagoon, indicating consistently lower pH values at any salinity level in burrow water. This is interpreted as a result of crab and/or microbial respiration.  相似文献

Brown cotton fibres are the most widely used naturally coloured raw materials for the eco‐friendly textile industry. Previous studies have indicated that brown fibre pigments belong to proanthocyanidins (PAs) or their derivatives, and fibre coloration is negatively associated with cotton productivity and fibre quality. To date, the molecular basis controlling the biosynthesis and accumulation of brown pigments in cotton fibres is largely unknown. In this study, based on expressional and transgenic analyses of cotton homologs of ArabidopsisPA regulator TRANSPARENT TESTA 2 (TT2) and fine‐mapping of the cotton dark‐brown fibre gene (Lc1), we show that a TT2 homolog, GhTT2‐3A, controls PA biosynthesis and brown pigmentation in cotton fibres. We observed that GhTT2‐3A activated GhbHLH130D, a homolog of ArabidopsisTT8, which in turn synergistically acted with GhTT2‐3A to activate downstream PA structural genes and PA synthesis and accumulation in cotton fibres. Furthermore, the up‐regulation of GhTT2‐3A in fibres at the secondary wall‐thickening stage resulted in brown mature fibres, and fibre quality and lint percentage were comparable to that of the white‐fibre control. The findings of this study reveal the regulatory mechanism controlling brown pigmentation in cotton fibres and demonstrate a promising biotechnological strategy to break the negative linkage between coloration and fibre quality and/or productivity.  相似文献

The shikimate pathway synthesizes aromatic amino acids essential for protein biosynthesis. Shikimate dehydrogenase (SDH) is a central enzyme of this primary metabolic pathway, producing shikimate. The structurally similar quinate is a secondary metabolite synthesized by quinate dehydrogenase (QDH). SDH and QDH belong to the same gene family, which diverged into two phylogenetic clades after a defining gene duplication just prior to the angiosperm/gymnosperm split. Non‐seed plants that diverged before this duplication harbour only a single gene of this family. Extant representatives from the chlorophytes (Chlamydomonas reinhardtii), bryophytes (Physcomitrella patens) and lycophytes (Selaginella moellendorfii) encoded almost exclusively SDH activity in vitro. A reconstructed ancestral sequence representing the node just prior to the gene duplication also encoded SDH activity. Quinate dehydrogenase activity was gained only in seed plants following gene duplication. Quinate dehydrogenases of gymnosperms, represented here by Pinus taeda, may be reminiscent of an evolutionary intermediate since they encode equal SDH and QDH activities. The second copy in P. taeda maintained specificity for shikimate similar to the activity found in the angiosperm SDH sister clade. The codon for a tyrosine residue within the active site displayed a signature of positive selection at the node defining the QDH clade, where it changed to a glycine. Replacing the tyrosine with a glycine in a highly shikimate‐specific angiosperm SDH was sufficient to gain some QDH function. Thus, very few mutations were necessary to facilitate the evolution of QDH genes.  相似文献

Summary Activity levels of succinate dehydrogenase (SDH) were determined kinetically by means of comparative microphotometric measurements in situ. Activities were correlated with fibre types classified histochemically according to Brooke and Kaiser (1970). Analyses of tibialis anterior muscles in the mouse, rat, guinea pig, rabbit, cat and the human showed pronounced variations in the activity profiles of type I, type IIA and IIB fibres of these muscles. Large scattering of enzyme activity existed in the three fibre populations. Overlaps of varying extent were found for the SDH profiles between the different muscles. Type I fibres reveal species diffeences in aerobic oxidative capacity. Whereas the majority of the IIB fibres in rabbit muscle tended to be low in SDH activity, the main fraction of this fibre population was characterized by high activities in mouse muscle. Similarly, the IIA fibre populations revealed opposite properties in mouse and rabbit muscles. These extremes as well as intermediate activity patterns indicate that no general scheme exists according to which the histochemically assessable myosin ATPase is correlated with the aerobic oxidative capacity of muscle fibres in various mammalian muscles.  相似文献

Mar Chiquita, a brackish coastal lagoon in central Argentina, is inhabited by dense populations of two intertidal grapsid crab species,Cyrtograpsus angulatus andChasmagnathus granulata. During a preliminary one-year study and a subsequent intensive sampling programme (November–December 1992), the physical properties and the occurrence of decapod crustacean larvae in the surface water of the lagoon were investigated. The lagoon is characterized by highly variable physical conditions, with oligohaline waters frequently predominating over extended periods. The adjacent coastal waters show a complex pattern of semidiurnal tides that often do not influence the lagoon, due to the existence of a sandbar across its entrance. Besides frequently occurring larvae (exclusively freshly hatched zoeae and a few megalopae) of the two dominating crab species, those of three other brachyurans (Plathyxanthus crenulatus, Uca uruguayensis, Pinnixa patagonica) and of one anomuran (the porcellanidPachycheles haigae) were also found occasionally. Caridean shrimp (Palaemonetes argentinus) larvae occurred in a moderate number of samples, with a maximum density of 800·m−3. The highest larval abundance was recorded inC. angulatus, with almost 8000°m−3. Significantly moreC. angulatus andC. granulata zoeae occurred at night than during daylight conditions, and more larvae (statistically significant only in the former species) during ebb (outflowing) than during flood (inflowing) tides. In consequence, most crab zoeae were observed during nocturnal ebb, the least with diurnal flood tides. Our data suggest that crab larvae do not develop in the lagoon, where the adult populations live, but exhibit an export strategy, probably based upon exogenously coordinated egg hatching rhythms. Zoeal development must take place in coastal marine waters, from where the megalopa eventually returns for settlement and metamorphosis in the lagoon. Significantly higher larval frequency ofC. granulata in low salinities (≤12‰) and at a particular sampling site may be related to local distribution patterns of the reproducing adult population. Unlike crab larvae, those of shrimp (P. argentinus) are retained inside the lagoon, where they develop from hatching through metamorphosis. They significantly prefer low salinity and occur at the lagoon surface more often at night. These patterns cannot be explained by larval release rhythms like those in brachyuran crabs, but may reflect diel vertical migrations to the bottom. It is concluded that osmotic stress as well as predation pressure exerted by visually directed predators (small species or life-cycle stages of estuarine fishes) may be the principal selection factors for the evolution of hatching and migration rhythms in decapod larvae, and that these are characteristics of export or retention mechanisms, respectively.  相似文献

Phylogenetic relationships of Oceanian staple yams (species of Dioscorea section Enantiophyllum) were investigated using plastid trnL‐F and rpl32‐trnL(UAG) sequences and nine nuclear co‐dominant microsatellites. Analysis of herbarium specimens, used as taxonomic references, allowed the comparison with samples collected in the field. It appears that D. alata, D. transversa and D. hastifolia are closely related species. This study does not support a direct ancestry from D. nummularia to D. alata as previously hypothesized. The dichotomy in D. nummularia previously described by farmers in semi‐perennial and annual types was reflected by molecular markers, but the genetic structure of D. nummularia appears more complex. Dioscorea nummularia displayed two haplotypes, each corresponding to a different genetic group. One, including a D. nummularia voucher from New Guinea, is closer to D. tranversa, D. alata and D. hastifolia and encompasses only semi‐perennial types. The second group is composed of semi‐perennial and annual yams. However, some of these annual yams also displayed D. alata haplotypes. Nuclear markers revealed that some annual yams shared alleles with D. alata and semi‐perennial D. nummularia, suggesting a hybrid origin, which may explain their intermediate morphotypes and the difficulty met in classifying them.  相似文献

Cotton fibres, the single‐celled trichomes derived from the ovule epidermis, provide the most important natural material for the global textile industry. A number of studies have demonstrated that regulating endogenous hormone levels through transgenic approaches can improve cotton fibre qualities. Phytosulfokine‐α (PSK‐α) is a novel peptide hormone in plants that is involved in regulating cell proliferation and elongation. However, its potential applications in crop genetic improvement have not been evaluated. In this study, we describe how exogenous PSK‐α application promotes cotton fibre cell elongation in vitro. Chlorate, an effective inhibitor of peptide sulfation, suppressed fibre elongation in ovule culture. Exogenously applied PSK‐α partly restored the chlorate‐induced suppression. A putative PSK gene (GhPSK) was cloned from Gossypium hirsutum. Expression pattern analysis revealed that GhPSK is preferentially expressed in rapidly elongating fibre cells (5–20 days postanthesis). Overexpression of GhPSK in cotton increased the endogenous PSK‐α level and promoted cotton fibre cell elongation, resulting in longer and finer fibres. Further results from electrophysiological and physiological analyses suggest that GhPSK affects fibre development through regulation of K+ efflux. Digital gene expression (DGE) profile analysis of GhPSK overexpression lines indicates that PSK signalling may regulate the respiratory electron‐transport chain and reactive oxygen species to affect cotton fibre development. These results imply that peptide hormones are involved in cotton fibre growth and suggest a new strategy for the biotechnological improvement of cotton fibre quality.  相似文献

Little is known about the natural history, biology, and population genetic structure of the Hardhead Silverside, Atherinomorus stipes, a small schooling fish found around islands throughout the Caribbean. Our field observations of A. stipes in the cays of Belize and the Florida Keys found that populations tend to be in close association with the shoreline in mangrove habitats. Due to this potential island‐based population structuring, A. stipes represents an ideal system to examine questions about gene flow and isolation by distance at different geographic scales. For this study, the mitochondrial gene nd2 was amplified from 394 individuals collected from seven different Belizean Cays (N = 175) and eight different Floridian Keys (N = 219). Results show surprisingly high haplotype diversity both within and between island‐groups, as well as a high prevalence of unique haplotypes within each island population. The results are consistent with models that require gene flow among populations as well as in situ evolution of rare haplotypes. There was no evidence for an isolation by distance model. The nd2 gene tree consists of two well‐supported monophyletic groups: a Belizean‐type clade and a Floridian‐type clade, indicating potential species‐level differentiation.  相似文献
