During the year 2008 to 2009, a new disease of stem canker was noticed in most red‐fleshed dragon fruit (Hylocereus polyrhizus) plantations in Malaysia. The symptoms observed were small circular sunken orange spot, black pycnidia and rotted stem. This study was conducted to determine the occurrence of the stem canker on H. polyrhizus in Malaysia, subsequently to isolate, identify and characterize the fungal pathogen based on morphology and molecular characteristics and pathogenicity test. From the surveyed 20 plantations in Malaysia, stem canker was detected in all the plantations. A total of 40 isolates of Scytalidium‐like fungus were isolated and identified as Neoscytalidium dimidiatum based on morphological characteristics and ITS region sequences, which showed 99% similarity to N. dimidiatum (FJ648577). From the phylogenetic analysis using maximum‐likelihood tree, isolates of N. dimidiatum from stem canker of H. polyrhizus were grouped together and did not show any sequence variation. From pathogenicity test, all 40 isolates of N. dimidiatum were pathogenic causing stem canker on H. polyrhizus. To our knowledge, this is the first report of stem canker of H. polyrhizus caused by N. dimidiatum in Malaysia.  相似文献

Polygonatum odoratum (Mill.) Druce (Angular Solomon's seal) is a well‐known ornamental plant and herbal drug, which is used in traditional medicine. Severe leaf spots were observed in wild Angular Solomon's seal plants in a public park in Trento, Italy. A combination of morphological and molecular characteristics, including sequence data of ITS‐rDNA, large subunit (LSU), beta tubulin (TUB) and glyceraldehyde 3‐phosphate dehydrogenase (GAPDH) led to identification of two species, namely Phoma odoratissimi and Colletotrichum dematium s. str., isolated from these leaf spots. Pathogenicity of the isolates was confirmed on wounded leaves of Po. odoratum; indeed, both species induced leaf spot symptoms on inoculated leaves within 10 days of inoculation. This is the first report of leaf spot disease caused by Ph. odoratissimi and C. dematium s. str. on Po. odoratum, which can be considered a new host for both the species examined in this study. In addition, isolation of Ph. odoratissimi represents a new record for the mycobiota of Italy.  相似文献

During 2010–2011, a severe leaf spot disease of sweet potato (Ipomoea batatas) was found in Haikou City, Hainan province of China. The disease is characterized with large, irregular, brown, necrotic lesions on the margin or in the centre of leaves. A species of Stemphylium was consistently recovered from pieces of symptomatic tissues on PDA. Based on morphological characteristics and molecular identification by rDNA‐ITS gene analysis, the fungal species was identified as Stemphylium solani Weber, and its pathogenicity was confirmed by Koch's postulates. This is the first report of leaf spot on sweet potato caused by Ssolani in China.  相似文献

Leaf spot disease was detected on Sansevieria trifasciata in Malaysia. Two Curvularia species namely C. eragrostidis and C. asianensis were isolated as the pathogens. The isolated fungi were identified based on morphology and multigene phylogeny (ITS, GAPDH and TEF1-α). Pathogenicity tests confirmed the roles of both Curvularia isolates in causing leaf spot of S. trifasciata. To our knowledge, this is the first report of C. eragrostidis and C. asianensis causing leaf spot of S. trifasciata in Malaysia.  相似文献

In the past 10 years, there has been a substantial increase in reports, from growers and extension personnel, on bulb and root rots in lily (Lilium longiflorum) in Israel. Rot in these plants, when grown as cut flowers, caused serious economic damage expressed in reduction in yield and quality. In lily, the fungal pathogens involved in the rot were characterized as binucleate Rhizoctonia AG‐A, Rhizoctonia solani, Pythium oligandrum, Fusarium proliferatum (white and purple isolates) and F. oxysporum, using morphological and molecular criteria. These fungi were the prevalent pathogens in diseased plants collected from commercial greenhouses. Pathogenicity trials were conducted on lily bulbs and onion seedlings under controlled conditions in a greenhouse to complete Koch's postulates. Disease symptoms on lily were most severe in treatments inoculated with binucleate Rhizoctonia AG‐A, P. oligandrum and F. proliferatum. Plant height was lower in the above treatments compared with the control plants. The least aggressive fungus was R. solani. In artificial inoculations of onion, seedling survival was significantly affected by all fungi. The most pathogenic fungus was F. proliferatum w and the least were isolates of F. oxysporum (II and III). All fungi were successfully re‐isolated from the inoculated plants.  相似文献

Brown spot, caused by the fungus Bipolaris oryzae, is one of the most destructive diseases of rice. This study investigated the effect of zinc rates on the development of brown spot in rice. Rice plants (cv. ‘Metica‐1′) were grown in hydroponic culture amended with Zn rates (applied as ZnSO4.7H2O) of 0, 0.5, 1, 2 and 4 μm and inoculated with B. oryzae. The foliar concentration of Zn was determined. Leaf samples were assessed for disease severity, and then, area under brown spot progress curve (AUBSPC) was calculated. The relationship between Zn concentrations on leaf tissues and the rates of this micronutrient was best described by a positive linear regression model, while the relationship between the Zn rates and the AUBSPC was best described with a positive quadratic regression model. The correlation between Zn concentrations on leaf tissues and AUBSPC was positive and significant (r = 0.68, P < 0.05). The results from this study showed that high foliar concentration of Zn was associated with increasing rice susceptibility to brown spot.  相似文献

Field resistances against Sclerotinia rot (SR) (Sclerotinia sclerotiorum) were determined in 52 Chinese genotypes of Brassica oleracea var. capitata, 14 Indian Brassica juncea genotypes carrying wild weedy Brassicaceae introgression(s) and four carrying B‐genome introgression, 22 Australian commercial Brassica napus varieties, and 12 B. napus and B. juncea genotypes of known resistance. All plants were individually inoculated by securing an agar disc from a culture of S. sclerotiorum growing on a glucose‐rich medium to the stem above the second internode with Parafilm tape. Mean stem lesion length across tested genotypes ranged from <1 to >68 mm. While there was considerable diversity within the germplasm sets from each country, overall, 65% of the B. oleracea var. capitata genotypes from China showed the highest levels of stem resistance, a level comparable with the highest resistance ever recorded for oilseed B. napus or B. juncea from China or Australia. One Indian B. juncea line carrying weedy introgression displayed a significant level of both stem and leaf resistance. However, the vast majority of commercial Australian oilseed B. napus varieties fell within the most susceptible 40% of genotypes tested for stem disease. There was no correlation between expressions of stem versus leaf resistance, suggesting their independent inheritance. A few Chinese B. oleracea var. capitata genotypes that expressed combined extremely high‐level stem (≤1 mm length) and leaf (≤0.5 mean number of infections/plant) resistance will be particularly significant for developing new SR‐resistant horticultural and oilseed Brassica varieties.  相似文献

Miscanthus spp. are large perennial wetland grasses that are receiving considerable attention as bioenergy crops. In late summer 2011, leaf spot symptoms were observed in a field of Miscanthus sinensis in Jeongseon, Gangwon province, Korea. Bacterial strains that belonged to the γ‐Proteobacteria genus Pseudomonas were isolated from the affected leaves. By phylogenetic analysis and phenotypic characterization, the representative strain MDM‐03 was identified as Pseudomonas lurida. Healthy M. sinensis leaves inoculated with MDM‐03 developed leaf spots similar to those observed in field. Bacteria re‐isolated from the leaf lesions were identical to the original strain MDM‐03 based on their cultural characteristics and 16S rDNA sequencing. This is the first report of bacterial leaf spot in Miscanthus sinensis.  相似文献

During September 2010, Abutilon megapotamicum plants with dark‐brown concentric spots on leaves were observed in a commercial glasshouse located in Beijing, China. This study was carried out to identify the causal agent of this disease based on Koch's postulates and morphological characteristics. Pathogenicity tests in the glasshouse showed that Myrothecium roridum Tode ex Fr. caused the leaf spot on A. megapotamicum plants, which were the same as those observed in naturally infected plants in the field. Moreover, to confirm the pathogen to species, the rDNA internal transcribed spacer (ITS) of isolate was PCR‐amplified using ITS1 and ITS4 primer pairs and sequenced. DNA analysis revealed a 100% species identity index for M. roridum. To the best of our knowledge, this is the first report of M. roridum on A. megapotamicum in China.  相似文献

Pineapple (Ananas comosus) is one the important fruit crops planted in Malaysia, and this study was conducted to determine Fusarium spp. associated with diseases of the fruit crop as Fusarium is prevalent in tropical countries. Our objective was to identify and characterize Fusarium spp. associated with pineapple fruit rot and leaf spot mainly found on the fruits and leaves in Peninsular Malaysia. Fusarium isolates (n = 108) associated with pineapple fruit rot and leaf spot were characterized by morphological, molecular and phylogenetic analyses, a mating study and pathogenicity testing. TEF‐1α sequence analysis identified Fusarium proliferatum, Fusarium verticillioides, Fusarium sacchari and Fusarium sp. Mating was successful only between tester strains of F. proliferatum and F. verticillioides. Sexual crosses with standard tester strains showed that 82 isolates of F. proliferatum produced fertile crosses with mating population D (Gibberella intermedia) and three isolates of F. verticillioides were fertile with the tester strain of mating population A (Gibberella moniliformis). All isolates were pathogenic, causing pineapple fruit rot and leaf spot, thus fulfilling Koch's postulates.  相似文献

The Roegneria kamoji accession ZY 1007 was resistant to the mixed predominant races of Puccinia striiformis f.sp. tritici (Pst) in China based on field tests at adult‐plant stage. The seedling resistance evaluation of ZY 1007 showed that it was resistant to stripe rust physiological strains CYR29, CYR33 and PST‐V26, which were the predominant races of Pst in China. The female parent R. kamoji cv. Gansi No.1 (susceptible to Pst) was crossed with ZY 1007 (resistant to Pst). Parents, F1 and F2 populations were tested in a field inoculated with the mixed urediniospores. ZY 1007 and all the observed 11 F1 hybrid plants were resistant, while plants of Gansi No.1 were susceptible. Among the 221 F2 plants, 168 plants were resistant and 53 were susceptible, and the segregation of resistant and susceptible plants fits 3R:1S ratio (χ2 = 0.074, P > 0.75). It confirmed that the resistance of stripe rust in ZY 1007 was controlled by a single dominant gene and temporarily designated as YrK1007.  相似文献

During 2011–2012, an extensive leaf spot disease caused by Stemphylium lycopersici was observed on vegetable crops including, tomato, eggplant, pepper and lettuce in major vegetable-growing regions of Malaysia. Four isolates of S. lycopersici obtained from each vegetable crop were used to determine cultural and physiological characteristics. The variations were found in colony colour (pale to light grey or light as well as the brown), texture (cottony or mycelium flat), shape (regular with concentric growth rings or irregular) and pigmentation (yellow or deep red) of the cultures. The optimum temperature for the conidial germination and mean radial growth of the isolates was 25?°C, and the radial growth of the isolates was maximal on V-8 juice agar followed by potato carrot agar. The maximum sporulation of S. lycopersici isolates was observed on V-8 juice agar media under 12/12 h light/darkness photoperiod at 25?°C.  相似文献

During August 2010 and January 2011, 10 isolates of Colletotrichum were recovered from stem anthracnose lesions of Hylocereus polyrhizus in the states of Kedah and Penang, Malaysia. Based on the morphological characteristics of colony colour and appearance, and shapes of conidia as well as sequences of internal transcribed spacer regions (ITS), β‐tubulin, actin (ACT) and glyceraldehyde 3‐phosphate dehydrogenase (GAPDH), the fungus was identified as Colletotrichum truncatum. Pathogenicity test showed that C. truncatum isolates were pathogenic to the artificially inoculated H. polyrhizus stem. This is the first report of C. truncatum causing anthracnose on H. polyrhizus stems in Malaysia.  相似文献

Pathogen infection can induce plant volatile organic compounds (VOCs). We infected ‘McNeal’ wheat and ‘Harrington’ barley with a Fusarium spp. blend (F. graminearum,F. avenaceum and F. culmorum). Both cereals had the greatest VOC induction 14 days after pathogen innoculation, only slightly lower induction occurred at 7 days, but displayed no induction at 1 days. The induced VOC bouquet for both cereals included six green leaf volatiles (GLVs; e.g. (Z)‐3‐hexenol and (Z)‐3‐hexenyl acetate), four terpenes (linalool, linalool oxide, (Z)‐β‐ocimene and (E)‐β‐caryophyllene) and benzyl acetate. Neighbouring, uninfected individuals of both cereals had significant VOC induction when exposed to an infected, conspecific plant. The temporal pattern and VOC blend were qualitatively similar to infected plants but with quantitative reductions for all induced VOCs. The degree of neighbouring, uninfected plant induction was negatively related to distance from an infected plant. Plant VOC induction in response to pathogen infection potentially influences herbivore attraction or repellency. Y‐tube tests showed that herbivorous female and male Oulema cyanella Voet. (Chrysomelidae: Coleoptera) were significantly attracted to (Z)‐3‐hexenal and (Z)‐3‐hexenyl acetate at 300 and 1500 ng/h but were repelled by both GLVs as well as (Z)‐β‐ocimene and linalool at 7500 ng/h. These O. cyanella behavioural responses were significantly at higher concentrations than those emitted by single plants with pathogen‐induced VOCs, so adults might only be able to respond to a dense group of infected plants. Also, O. cyanella dose responses differ from the previously tested congeneric O. melanopus (cereal leaf beetle), which was attracted to three VOCs induced by Fusarium infection of maize, barley and wheat. Future behavioural tests may indicate whether different herbivore dose responses measured with each VOC singly can help to predict attraction or repellency to injured and uninjured VOC bouquets from different host plant species.  相似文献

Sedum erythrostictum is a perennial herb in the Crassulaceae family, which is a traditional Chinese medicine used for the treatment of hepatitis, dysentery, herpes zoster and swellings. In 2014, a grey leaf spot disease causing significant damage to plants of S. erythrostictum occurred in the Medicinal Herb Garden of Jilin Agricultural University, Jilin Province, China. The fungus mainly infected the leaves. The necrotic lesions on the leaves were circular or elliptical, amphigenous, greyish brown to brown, slightly concave and surrounded by a dark brown distinct margin. The causal agent from symptomatic tissues was identified as Cercospora cf. pseudokalanchoes based on the symptoms, morphological characteristics, molecular identifications and pathogenicity tests. To our knowledge, this is the first formal report of grey leaf spot of S. erythrostictum caused by C. cf. pseudokalanchoes in China.  相似文献

Brazilian peppertree, Schinus terebinthifolia Raddi (Anacardiaceae), is one of the worst upland exotic weeds in Florida, USA. Foreign exploration for natural enemies led to the discovery of a pit‐galling psyllid, Calophya latiforceps Burckhardt (Hemiptera: Calophyidae), in the state of Bahia, Brazil, in 2010. Crawlers of C. latiforceps stimulate the formation of galls on the leaves of S. terebinthifolia resulting in leaf discoloration and in some cases leaf abscission. To determine whether C. latiforceps is a safe candidate for biological control of Sterebinthifolia, host specificity and the presence of selected plant pathogens were examined. Adult oviposition, gall formation, and adult survival of C. latiforceps were examined on 89 plant species under no‐choice and choice conditions. We found that C. latiforceps laid eggs on plants in seven families; however, crawlers stimulated gall formation and completed development to adult only on S. terebinthifolia. All crawlers on non‐target plants died, likely due to starvation caused either by the absence of a feeding stimulus or by a hypersensitive plant response. Under no‐choice conditions, 10% of adults lived for 19 days on the target weed, but adult survival was reduced to <3 days on non‐target plants. Choice testing revealed that females preferred to oviposit on S. terebinthifolia compared to non‐target plants. Molecular methods and indicator host inoculations did not detect the presence of ‘Candidatus Liberibacter solanacearum’, ‘Ca. L. asiaticus’, ‘Ca. L. americanus’, ‘Ca. L. africanus’, or plant viruses in adult C. latiforceps. We conclude that releasing C. latiforceps in the USA will have extremely low risk to non‐target plants, and provides another tool for the management of S. terebinthifolia.  相似文献

The rust and brown eye spot, caused by Hemileia vastatrix and Cercospora coffeicola, respectively, are the most important fungal diseases on coffee in South America. Their management is mainly by chemical treatment, and there is no genetic resistance to brown eye spot known so far. Considering the need for developing alternative products for their control, the goal of this work was to evaluate the effects of phosphites and by‐products of coffee and citrus industries on rust and brown eye spot. Formulations of coffee and citrus industry by‐products, phosphites and their combination with fungicide were evaluated in field experiments, and their effect on fungal urediniospores and conidia was evaluated in vitro. In the field, treatments were applied individually or in combination and the in vitro assays were performed with manganese phosphite (Reforce Mn), potassium phosphite and citrus industry by‐product (Fortaleza), copper phosphite and coffee industry by‐product (Fitoforce Full), and fungicide. The severity and incidence of rust and brown eye spot on coffee leaves, yield, and leaf retention were evaluated in the field. Percentage of spore germination was evaluated in vitro for both fungi, whereas mycelial growth was evaluated for C. coffeicola only. The treatments Fortaleza, Reforce Mn and Fitoforce Full suppressed both diseases with a reduction in defoliation. In the year 2012, the plants treated with Reforce Mn and Reforce Mn + Fortaleza showed a yield increase of 72 and 88%, respectively, which was similar to the results shown by the fungicide treatment. In vitro inhibition of germination of H. vastatrix urediniospores and of C. coffeicola conidia was observed and suggests that the products exert some toxic effects to both fungi. Finally, the results observed indicate that the combined use of by‐products of plant‐processing industries and phosphites is an alternative and can be added efficiently to the management of coffee diseases.  相似文献

Atractylodes japonica is a perennial herb in Compositae family, which is used for stomach disorders as a traditional Chinese medicine (Guo et al. 2006 ). In 2013, a leaf spot disease was first observed on plants of A. japonica in a production field of Fushun County, Liaoning Province, China. The disease had a speckled appearance initially. Lesions with grey–white centre and brown margin gradually developed and enlarged. Eventually, infections usually caused yellowing of the leaves and premature defoliation. The causal agent of infection on plants was identified as Paraphoma chrysanthemicola based on morphological and cultural characteristics, pathogenicity tests and phylogenetic analysis. To our knowledge, this is the first report of a leaf spot disease on A. japonica caused by P. chrysanthemicola in China.  相似文献

In April 2010, a severe occurrence of Stewart's wilt on Dracaena sanderiana plants was observed in greenhouses in Seongnam, Gyeonggi Province, South Korea, with an incidence of 35‐50%. Being imported plants, little was known about the pathogens associated with D. sanderiana. Symptoms included chlorosis, wilting and leaf blight on the leaf surfaces. Physiological analysis, pathogenicity tests, sequencing and phylogenetic analysis of the 16S rRNA gene revealed that the pathogen was the bacterium Pantoea stewartii. To the best of our knowledge, this is the first report on bacterial wilt caused by P. stewartii on D. sanderiana.  相似文献

Target spot, caused by the fungus Corynespora cassiicola, has become a serious foliar disease in soybean production in the Brazilian Cerrado. Information in the literature regarding the biochemical defence responses of soybean to C. cassiicola infection is rare. Therefore, the objective of this study was to determine the biochemical features associated with soybean resistance to target spot. The activities of chitinases (CHI), β‐1‐3‐glucanases (GLU), phenylalanine ammonia‐lyases (PAL), peroxidases (POX), polyphenol oxidases (PPO) and lipoxygenases (LOX), as well as the concentrations of total soluble phenolics (TSP) and lignin‐thioglycolic acid (LTGA) derivatives, were determined in soybean leaves from both a resistant (FUNDACEP 59) and a susceptible (TMG 132) cultivar. The target spot severity, number of lesions per cm2 of leaflet and area under the disease progress curve were significantly lower for plants from cv. FUNDACEP 59 compared to plants from cv. TMG 132. The GLU, CHI, PAL, POX and PPO activities and the concentration of LTGA derivatives increased significantly, whereas LOX activity decreased significantly on the leaves infected by C. cassiicola. Inoculated plants from cv. FUNDACEP 59 showed a higher PPO activity and concentrations of TSP and LTGA derivatives at 4 and 6 days after inoculation compared to plants from cv. TMG 132. In conclusion, the results of this study demonstrated that the defence‐related enzyme activities increased upon C. cassiicola infection, regardless of the basal level of resistance of the cultivar studied. The increases in PPO activity and concentrations of TSP and LTGA derivatives, but lower LOX activity, at early stages of C. cassiicola infection were highly associated with soybean resistance to target spot.  相似文献
