An Exact theoretical expression for the apparent diffusion coefficient Dapp(K) of a thin rigid rod with arbitrary anisotropy of its translational diffusion diffusion coefficient is derived from the first cumulant of its dynamic structure factor. Dapp(K) is predicted to reach a limiting plateau value at extermely large values of KL, where K is the scattering vector and L the rod length. Howerver, that limiting plateau value is approached only very slowly along a quasi-plateau with a very gradual slope. Dynamic light-scattering studies have been performed on tobacco mosaic virus from K2 = (0.4–20) × 1010 cm?2 using 632-8-nm laser radiation. The present data yield D0 = (4.19 ± 0.10) × 10?8 cm2/s (corrected to 20,w conditions) and, with literature data to establish L = 2980 Å and the rotational diffusion coefficient DR = 318s?1, yield also Δ ≡ D ? D = (1.79 ± 0.38) × 10?8 cm2/s. The experimental data closely follow the curve of Dapp(K) vs K2 calcuated for these parameters. The present value of D0 substantially exceeds all previous dynamic light-scattering values, but is in good aggreement with previous sedimentation data, which were confirmed for the presemt sample. The anisotropy ratio Δ/D0 = 0.43 ± 0.09 is in accord with theoretical predictions based on the modified Kirkwood algorithm, despite the fact the D0 lies significantly below its corresponding theoretical value. The present data largely predlude the possibility that both D0 and Δ/D0 could simultaneously match their theoretical predictions. We present a detailed comparison of the experimental data with the calculations of Tirado and Garcia de la Torre based on the modified Kirkwood algorithm and with the Broersma formulas.  相似文献

The structure of thermally denatured Type I collagen has been studied using laser light scattering. The results indicate that the diffusion coefficients of α-chains and β- and γ-components are 1.550 ± 0.08 × 10?7, 1.000 ± 0.05 × 10?7, and 0.835 ± 0.04 × 10?7 cm2/sec, respectively, at temperatures between 20 and 40°C. It is concluded from diffusion data that these species have hydrodynamic radii of about 13.8 nm (α-chain), 21.5 nm (β-component), and 25.7 nm (γ-component), consistent with previous studies of thermal denaturation by light scattering. It is also concluded, based on volume calculations, that a large volume increase occurs when the triple helix unfolds. Homodyne correlation functions for two component mixtures of α-chains and β-and γ-components appeared to decay exponentially. In all but one case discussed the correlation function could be fitted with a single component having a translational diffusion coefficient which was an intensity weighted average of the diffusion coefficient of each component present.  相似文献

The garfish Belone belone euxini (Günther, 1866) is a commercially important pelagic fish species in Sinop artisanal fishery, which is showing a decreasing trend in catch results. As a basis for fisheries management a sampling program was carried out between October 2000 and September 2001 along the Turkish coast of the Black Sea, to study the population structure, growth, and reproduction cycle of garfish in the area, and to achieve a rough estimate of exploitation. The length–weight relationship and von Bertalanffy growth parameters were estimated as W = 0.00076 L3.137, L = 74.64 cm, K = 0.13 year?1, to = ?3.67, respectively. First sexual maturity was estimated at age 2 and at a total length of 38.8 cm for females. The spawning period was from May to September. The total fecundity–length relationship was estimated as F = 0.0041 L4.1086 (r2 = 0.92). Mortality rates were Z = 1.24 year?1, M = 0.23 year?1and F = 1.01 year?1 for total, natural, and fishing mortality, respectively. The exploitation ratio E = 0.81 indicates that the population is heavily exploited.  相似文献

A Malvern laser light-scattering instrument has been modified for use at scattering angles down to 5° and both total intensity and quasi-elastic scattering experiments. A sample of sheared, length-fractionated calf-thymus DNA was characterized by sedimentation, viscosity and electron microscopy. Quasi-elastic scattering and absolute intensity determinations were performed with the laser instrument and intensity determinations only with a Fica conventional light-scattering photometer. The total intensity experiments gave M?w = (3.75 ± 0.15) × 106 and 〈R21/2z = (206.9 ± 10.3) nm which yielded a value for the persistence length, allowing for polydispersity, of 66 ± 6nm. The quasi-elastic experiments at scattering angles below 20° gave D020, w = (2.23 ± 0.06) × 10?8 cm2/sec which combined with S020, w = 15.6 in the Svedberg equation gave M?w = (3.73 ± 0.18) × 106. In addition, from the higher angle data we extracted a value of the longest intramolecular relaxation time, τ1 of 17.5 msec. This is not in particularly good agreement with τ1 predicted by the Zimm–Rouse theory using our other experimental parameters. The disagreement may be due to the restricted applicability of the Zimm–Rouse spring-bead model as a quantitative representation of DNA molecules. Alternatively, it may be due to present difficulties in the unambiguous interpretation of molecular motions from the experimental autocorrelation functions.  相似文献

Internal Brownian motions of clean ?29 and λ-DNAs have been studied using photon-correlation techniques at both visible (λ0 = 632.8 nm) and uv (λ0 = 363.8 nm) wavelengths. The present dynamic light scattering data, which extend to K2 = 19 × 1010 cm?2, can in every case be satisfactorily simulated by a Rouse-Zimm model polymer with an appropriate choice of the three model parameters. The effects of pH, salt concentration, single-strand breaks, and molecular weight on those model parameters have also been investigated. Intact clean DNAs exhibit surprisingly little variation with pH from 7.85 to 10.25, with salt concentration from 0.01 NaCl to 5.4M NH4Cl, or with molecular weight or GC content. The single-strand breaks have no effect at pH 9.46, but produce dramatic changes in the model parameters at pH 10.0 and 10.25, indicating the introduction of titratable joints at those pHs. The failure of either single-strand breaks or a large change in GC content to alter the model parameters in the neutral pH range is a strong indication that local denaturation is not required for those flexions and torsions that dominate the relaxation of fluctuations in the scattered light. The Langevin relaxation time for the slowest internal mode of a particular Rouse-Zimm model derived from the dynamic light scattering data is compared with pertinent literature data extrapolated to the same molecular weight. The present algorithm for determining model parameters from the light-scattering Dapp vs K2 curve actually yields a Langevin time in fairly good agreement with the literature value. For unknown reasons the light-scattering D0 values generally exceed those obtained from the molecular weight and sedimentation coefficient by about 20%.  相似文献

We have measured the percentages of cis and trans Gly-Pro and X-Hyp peptide bonds in thermally unfolded type I collagen. 13C-nmr solution spectra show that 16% of the Gly-Pro and 8% of the X-Hyp bonds are cis in unfolded chick calvaria collagen. These results support the hypothesis that cistrans isomerization is that rate-limiting step in the propagation of the collagen triple helix. We have used multinuclear solid-state nmr to study the molecular dynamics of the collagen backbone in tendon, demineralized bone, and intact bone as a function of temperature, hydration, and pH. These studies show that collagen backbone motions are characterized by a broad distribution of correlation times, τ, covering the range from 10?4 to 10?9 s. In the case of nonmineralized collagen, the root-mean-square fluctuations in azimuthal angle, γrms, range from ca. 10° when τ ~ 10?9 s to ca. 30° when τ < 10?4 s; in the case of bone collagen, γrms values are about half as large as those found in nonmineralized collagen. Backbone motions are negligible at temperatures below ?25°C. This is also the case at 22°C when demineralized bone collagen is lyophilized. In contrast, flexibility of hydrated demineralized bone collagen greatly increases as pH is lowered from 7 to 2. The more limited flexibility observed at neutral pH is a consequence of the intermolecular interactions that contribute to fibril organization and strength. However, the fibrils retain significant flexibility at physiological pH, enabling them to distribute stress and dissipate mechanical energy.  相似文献

Oxygen transfer in Streptomyces fermentation broths   总被引:1,自引:0,他引:1  
The oxygen transfer coefficient has been investigated in S. noursci and S. lavendulae fermentation broths obtained from fermentors of different operating volumes (61., 30001., 20,0001.). Fermentors had KLas values ranging from 1.0 to 17.0 min?1, calculated from sulphite oxidation rates. The dynamic measurement of the volumetric oxygen transfer coefficient. (KLa) has been performed in the different fermenting systems. As the fermentation progressed, especially in the first stages, KLa values have decreased in both fermentations and in each system of fermentors. In order to characterise the whole fermenting system an average KLas was calculated from the obtained KLa values. The average KLa grew with increasing KLas values and ranged from 0.03 to 3.72 min?l. Some factors possibly having an influence on the, change of KLa have been studied. The oxygen transfer coefficients of the broths have been measured in falling films and ranged from 0.05 to 0.4 cm min?1. The flow conditions have been characterized by Reynolds numbers of broths varying between 1.0 and 60.0. The average thickness of the falling films have been measured and plotted against Reynolds number. The Re+ which is the breaking-point of the plot increased as the fermentation proceeded. In the region of Re+ the values of the oxygen transfer coefficient increased rapidly. An approximate correlation could be established between the Re+ and the physical properties of fermentation broth.  相似文献

In this study, the biohydrogen (bioH2) production of a microbial consortium was optimized by adjusting the type and configuration of two impellers, the mixing regimen and the mass transfer process (KLa coefficients). A continuous stirred-tank reactor (CSTR) system, with a nonstandard geometry, was characterized. Two different mixing configurations with either predominant axial (PB4 impeller) or radial pumping (Rushton impeller) were assessed and four different impeller configurations to produce bioH2. The best configuration for an adequate mixing time was determined by an ANOVA analysis. A response surface methodology was also used to fully elucidate the optimal configuration. When the PB4 impellers were placed in best configuration, c/Dt?=?0.5, s/Di?=?1, the maximum bioH2 productivity obtained was 440?mL?L?1?hr?1, with a bioH2 molar yield of 1.8. The second best configuration obtained with the PB4 impellers presented a bioH2 productivity of 407.94?mL?L?1?hr?1. The configurations based on Rushton impellers showed a lower bioH2 productivity and bioH2 molar yield of 177.065?mL?L?1?hr?1 and 0.71, respectively. The experiments with axial impellers (PB4) showed the lowest KLa coefficient and the highest bioH2 production, suggesting that mixing is more important than KLa for the enhanced production of bioH2.  相似文献

Leaf water gets isotopically enriched through transpiration, and diffusion of enriched water through the leaf depends on transpiration flow and the effective path length (L). The aim of this work was to relate L with physiological variables likely to respond to similar processes. We studied the response to drought and vein severing of leaf lamina hydraulic conductance (Klamina), mesophyll conductance for CO2 (gm) and leaf water isotope enrichment in Vitis vinifera L cv. Grenache. We hypothesized that restrictions in water pathways would reduce Klamina and increase L. As a secondary hypothesis, we proposed that, given the common pathways for water and CO2 involved, a similar response should be found in gm. Our results showed that L was strongly related to mesophyll variables, such as Klamina or gm across experimental drought and vein‐cutting treatments, showing stronger relationships than with variables included as input parameters for the models, such as transpiration. Our findings were further supported by a literature survey showing a close link between L and leaf hydraulic conductance (Kleaf = 31.5 × L?0.43, r2 = 0.60, n = 24). The strong correlation found between L, Klamina and gm supports the idea that water and CO2 share an important part of their diffusion pathways through the mesophyll.  相似文献

Six amylose‐2‐acetyl‐3,6‐bis(phenylcarbamate) (AAPC) samples ranging in weight‐average molar mass Mw from 1.8 × 104 g mol?1 to 1.1 × 106 g mol?1 have been prepared from enzymatically synthesized amylose samples. Static light scattering, small‐angle X‐ray scattering, sedimentation equilibrium, and viscosity measurements were made for the samples in 1,4‐dioxane (DIOX), 2‐ethoxyethanol (2EE), and 2‐butanone (MEK) all at 25°C to determine particle scattering functions, z‐average radii of gyration, intrinsic viscosities, as well as Mw. The data were analyzed in terms of the wormlike cylinder model mainly to yield the helix pitch per residue h and the Kuhn segment length λ?1, which corresponds to twice of the persistence length. The latter parameters (λ?1) in 2EE (11 nm) and MEK (12 nm) are quite smaller than those for amylose tris(phenylcarbamate) (ATPC) in the same solvent (16 nm in 2EE and 18 nm in MEK) whereas those for AAPC (21 nm) and ATPC (22 nm) in DIOX are essentially the same as each other. This indicates that the chain stiffness of AAPC is more strongly influenced by the solvents since the number of intramolecular H‐bonds of AAPC is more changeable than that for ATPC. © 2012 Wiley Periodicals, Inc. Quasielastic and static light-scattering measurements were made on DNA isolated from chicken erythrocyte mononucleosomes as a function of ionic strength between 6 × 10?4 and 1.0M. A transition from single-exponential autocorrelation functions to markedly non-single-exponential decays was observed around 10?2M ionic strength and was accompanied by a large decrease in the excess light-scattering intensity. Autocorrelation functions recorded below 10?2M salt were well fit by the sum of two exponential relaxation which differed by as much as 100-fold in time constants. Apparent diffusion coefficients for the fast and slow processes plateaued around 10?3M with numerical values approximately 10-fold and 1/10, respectively, of the translational diffusion coefficient for mononucleosome DNA at high ionic strength. This behavior is similar to that observed with poly(L -lysine), for which the slow decay has been associated with a transition to an extraordinary phase. The strong and complex salt dependence observed here illustrates potential difficulties in deriving structural information from scattering by polyions at low ionic strength.  相似文献

(S)-1-Phenylethanol derivatives, which are the precursors of many pharmacological products, have also been used as anti-Alzheimer drugs. Bioreduction experiments were performed in a batch and packed-bed bioreactor. Then, the kinetics constants were determined by examining the reaction kinetics in the batch system with free and immobilized carrot cells. Also, the effective diffusion coefficient (De) of acetophenone in calcium alginate-immobilized carrot cells was investigated. Kinetics constants for free cells, which are intrinsic values, are reaction rate Vmax?=?0.052?mmol?L?1?min?1, and constants of the Michaelis–Menten KM?=?2.31?mmol?L?1. Kinetics constants for immobilized cells, which are considered apparent values, are Vmax, app?=?0.0407?mmol?L?1 min?1, KM, app?=?3.0472?mmol?L?1 for 2?mm bead diameter, and Vmax, app?=?0.0453?mmol?L?1 min?1, KM, app?=?4.9383?mmol?L?1 for 3?mm bead diameter. Average value of effective diffusion coefficient of acetophenone in immobilized beads was determined as 1.97?×?10?6?cm2?s?1. Using immobilized carrot cells in an up-flow packed-bed reactor, continuous production of (S)-1-phenylethanol through asymmetric bioreduction of acetophenone was performed. The effects of the residence time and concentrations of substrate were investigated at pH 7.6 and 33°C. Enantiomerically pure (S)-1-phenylethanol (ee?>?99%) was produced with 75% conversion at 4-hr residence time.  相似文献

Age and growth rates of bull shark Carcharhinus leucas[n = 255; 555–2230 mm fork length (LF)] from the northern Gulf of Mexico were estimated from ring counts on vertebral sections collected from fishery‐dependent and ‐independent surveys. Two growth models were fitted to observed data: the von Bertalanffy growth model (VBGM) with t0 as the third parameter and a modified version of the VBGM using a fixed size‐at‐birth intercept as the third parameter. To address the variability in size‐at‐birth, a Monte Carlo simulation was incorporated into the size‐at‐birth intercept. The sex‐specific growth models were not significantly different, allowing a sexes combined model to be generated. The traditional VBGM predicted a theoretical maximum size (L) of 3007·1 mm LF, a growth coefficient (K) of 0·042 year?1 and a theoretical age at zero length (t0) of –6·844 years. The modified VBGM with a fixed size‐at‐birth intercept of 565 mm LF predicted an L of 2289·2 mm LF and a K value of 0·089 year?1. When comparing model estimates to previously published information, the traditional VBGM predicted a significantly lower theoretical maximum size and a higher growth coefficient than those produced using data collected during the 1980s. Overall, results obtained using the VBGM with a fixed size‐at‐birth produced more biologically realistic parameters than that of the VBGM with t0. The Monte‐Carlo simulation incorporating variability in size‐at‐birth produced similar results to the VBGM using a fixed size‐at‐birth. This study provides the first attempt to incorporate variability at size‐at‐birth and provide measurements of variability around the individual parameter estimates for an elasmobranch.  相似文献

The aim of the study was to compare the physico‐chemical parameters of milt from sea trout (Salmo trutta m. trutta), brook trout (Salvelinus fontinalis) and rainbow trout (Oncorhynchus mykiss). Milt was collected by stripping and spermatozoa concentrations, were determined and compared with sperm motility and spermatocrit values along with seminal plasma indices (pH, osmolality, sodium, potassium, chlorine, calcium, magnesium, glucose and protein concentrations). The highest spermatozoa concentration of 22.3 ± 6.7 × 109 ml?1 was found in the sea trout milt, and was significantly different of those observed in brook trout (11.9 ± 3.3 × 109 ml?1) and rainbow trout (10.7 ± 4.4 × 109 ml?1). The values for pH and K+ did not differ significantly among species. The mean pH was 8.0 in the milt of each species and the K+ concentrations ranged from 24.8 ± 7.2 to 30.5 ± 7.6 mm L?1. Considerable differences were determined for the Ca2+ ions concentrations. The highest value was found in sea trout (1.7 ± 0.3 mm L?1), while in the rainbow trout it was 0.7 ± 0.5 and in the brook trout 0.4 ± 0.1 mm L?1. The most pronounced differences were found in the glucose concentration cause of its unnaturally low concentration in rainbow trout of the mean value of 6.0 ± 15.2 mg L?1. The mean value in sea trout and brook trout was 185.0 ± 172.4 and 231.2 ± 148.4 mg L?1 respectively. For all species, protein mean values were below 1.3 g L?1. The mean osmolality was between 230.6 ± 98.6 and 272.0 ± 26.4 mOsm kg?1 in the species studied. No correlation was found between any components determined in milt and the spermatozoa motility (P > 0.05). The sperm concentration was positively correlated with the protein content in the milt of the three species studied, other less exhibited correlation was found.  相似文献

The physical properties of type I collagen were studied by electron microscopy of rotary shadowed collagen molecules and laser light scattering techniques. The physical properties, molecular structure and flexibility of type I collagen molecules from two structurally and functionally different connective tissues, cornea and sclera, were similar when measured in HCl, pH 2.0. The molecular weights were 328 and 298 × 102 for corneal and scleral type I collagen, respectively, while the values of TM were 33.7°C for both preparations. These values were in agreement with those obtained for other type I collagens. The higher level of glycosylation in corneal versus scleral type I collagen did not significantly modify the physical properties of type I collagen in acid solution or the charge distribution along the molecule as determined from the positively stained SLS banding patterns. Our morphological studies indicated that the collagen molecule, although relatively flexible based on electron microscopy, behaved as a long thin rod in solution. The mean end-to-end distances measured from electron micrographs were 253 and 256 nm for corneal and cler type I collagen, respectively, while the molecular contour lengths were 298 and 305 nm. The translational diffusion coefficients (0.849 and 0.857 × 10?7cm2s?1) were consistent with the contour lengths while the reported values in the literature for the rotational diffusion coefficient of type I collagen were consistent with the end-to-end distances. The intermediate value for molecular length obtained from the particle scattering factor (277 nm) reflects contributions from all possible molecular configurations.  相似文献

The α-methylserine aldolase gene from Variovorax paradoxus strains AJ110406, NBRC15149, and NBRC15150 was cloned and expressed in Escherichia coli. Formaldehyde release activity from α-methyl-L-serine was detected in the cell-free extract of E.coli expressing the gene from three strains. The recombinant enzyme from V. paradoxus NBRC15150 was purified. The V max and K m of the enzyme for the formaldehyde release reaction from α-methyl-L-serine were 1.89 μmol min?1 mg?1 and 1.2 mM respectively. The enzyme was also capable of catalyzing the synthesis of α-methyl-L-serine and α-ethyl-L-serine from L-alanine and L-2-aminobutyric acid respectively, accompanied by hydroxymethyl transfer from formaldehyde. The purified enzyme also catalyzed alanine racemization. It contained 1 mole of pyridoxal 5′-phosphate per mol of the enzyme subunit, and exhibited a specific spectral peak at 429 nm. With L-alanine and L-2-aminobutyric acid as substrates, the specific peak, assumed to be a result of the formation of a quinonoid intermediate, increased at 498 nm and 500 nm respectively.  相似文献

Abstract

We have calculated the variance of equilibrium distribution of a circular wormlike polymer chain over the writhing number, ?(Wr)2?, as a function of the number of Kuhn statistical segments, n, For large n these data splice well with our earlier results obtained for a circular freely jointed polymer chain. Assuming that ?(ΔLk)2? = ?(ΔTw)2? + ?(Wr)2? we have compared our results with experimental data on the chain length dependence of the ?(ΔLk) 2? value recently obtained by Horowitz and Wang for small DNA rings. This comparison has shown an excellent agreement between theory and experiment and yielded a reliable estimate of the torsional and bending rigidity parameters. Namely, the torsional rigidity constant is C = 3.0·10?19 erg cm, and the bending rigidity as expressed in terms of the DNA persistence length is a = 500 A. The obtained value of C agrees well with earlier estimates by Shore and Baldwin as well as by Horowitz and Wang whereas the a value is in accord with the data of Hagerman. We have found the data of Shore and Baldwin on the chain length dependence of the ?(ΔLk) 2? value to be entirely inconsistent with our theoretical results.  相似文献

This study presents some traits of the Aphanius ginaonis life history in the Geno hot spring and explains the potential risks of its extinction. Sampling was from March 2009 to February 2010. A total of 61 males and 71 females were measured (total length) and weighed, with data on reproductive biometry also taken. Growth parameters were determined in addition to weight–length relationships: W (t) = 0.012TL3.42, R2 = 0.96 for females and W (t) = 0.0101TL3.38, R2 = 0.94 for males. A. ginaonis females showed an asymptotic total length (TL) of 53.03 mm; the growth coefficient, K (year?1) 0.15, t0 (year) 1.01; and natural mortality coefficient M (year?1) 0.62. In males the value for TL was 48.83 mm; for K (year?1) 0.2, t0 (year) 0.44; and M (year?1) 0.49. The relationship between absolute fecundity and fish size (total length, body weight or age) showed a strong correlation to body weight. The A. ginaonis population is threatened with extinction – and a co‐management in cooperation with the local population for strong protection measures is urgently needed.  相似文献

A literature review of data on nitrate uptake by phytoplankton suggests that nitrate levels above 20 μmol N·L?1 generally stimulated uptake rates in cultured unicellular algae and natural phytoplankton communities. This phenomenon indicates that phytoplankton cells acclimate to elevated nitrate levels by increasing their uptake capacity in a range of concentrations previously considered to be saturating. Cyanobacteria and flagellates were found to present a considerable capacity for acclimation, with low (0.1–2 μmol N·L?1) half‐saturation values (Ks) at low (5–20 μmol N·L?1) substrate levels and high (1–80 μmol N·L?1) Ks values at high (30–100 μmol N·L?1) substrate levels. However, some diatom genera (Rhizosolenia, Skeletonema, Thalassiosira) also appeared to possess a low affinity nitrate uptake system (Ks between 18 and 120 μmol N·L?1), which can help resolve the paradox of their presence in enriched seas. It follows that present models of nitrate uptake can severely underestimate the effects of high nitrate concentrations on phytoplankton dynamics and development. A more adequate approach would be to consider the possibility of multiphasic uptake involving several phase transitions as nitrate concentrations increased. Because it is a nonlinear phenomenon featuring strong thresholds, this effect appears to override that of other variables, such as irradiance, temperature, and cell size. Within the present context of eutrophication and for a range of concentrations that is becoming more and more ecologically relevant, equations are tentatively presented as a first approach to estimate Ks from ambient nitrate concentrations.  相似文献
