β-glucosidase (BG) was believed to take part in abscisic acid (ABA) synthesis via hydrolysis of ABA glucose ester to release active ABA during plant growth and development. However, there is no genetic evidence available to indicate the role of genes during fruit ripening. Here, the expression patterns of three genes (VvBG1, VvBG2, and VvBG3) encoding β-glucosidase were analyzed during grape fruit development, and it was found that β-glucosidase activity increased in grape fruit in response to various stresses. Furthermore, to verify the function of β-glucosidase during fruit ripening, heterogeneous expression of the VvBG1 gene in strawberry fruit was validated, and the results showed that the VvBG1 over-expression increased β-glucosidase and promoted the fruit ripening process in strawberry. In addition, we found that ABA contents increased in the VvBG1 over-expression of strawberry fruit, which induced fruit anthocyanin, soluble solid accumulation, and fruit softening. Moreover, genes related to coloring (CHS, CHI, F3H, and UFGT), softening (PG1, PL1, and EXP1), and aroma (SAAT, and QR) were up-regulated. This work will elucidate the specific roles of VvBGs in the synthesis of ABA and provide some new insights into the ABA-controlled grape ripening mechanism.  相似文献

Strawberry is one of the most economically important fruit crops in the world. Cytokinins (CKs) play a critical role in plant growth and development, as well as the stress response, and the level of CKs in plants is regulated by synthesis and degradation pathways. The key synthetic enzymes of CKs are isopentenyl transferases (IPTs) and LONELY GUYS (LOGs). We surveyed the strawberry genome and identified seven FvIPT genes and nine FvLOG genes. We analyzed gene structures, conserved domains, and their phylogenetic relationships with rice and Arabidopsis. The isoelectric points and glycosylation sites of the proteins were predicted. We also analyzed tissue- or organ-specific expression patterns of the FvIPT and FvLOG genes. The FvIPT and FvLOG genes showed different expression profiles in different organs. Most FvIPT and FvLOG genes were down-regulated in response to osmotic stress, high-temperature treatment, and exogenous abscisic acid (ABA) application, suggesting possible roles of these genes in the plants’ resistance to abiotic stresses. In addition, we found that the results of bioinformatics analyses to identify cis-regulatory elements may not be consistent with experimental expression data; thus, computer-predicted putative cis-elements need to be confirmed by experiments. Our systematic analyses of the FvIPT and FvLOG families provide a foundation for characterizing the function of these genes in the regulation of growth, development, and stress tolerance in Fragaria vesca, as well as a reference for improving stress tolerance by manipulating CK content.  相似文献

A family of carotenoid cleavage dioxygenases (CCDs) produces diverse apocarotenoid compounds via the oxidative cleavage of carotenoids as substrates. Their types are highly dependent on the action of the CCD family to cleave the double bonds at the specific position on the carotenoids. Here, we report in vivo function of the AtCCD4 gene, one of the nine members of the Arabidopsis CCD gene family, in transgenic rice plants. Using two independent single-copy rice lines overexpressing the AtCCD4 transgene, the targeted analysis for carotenoids and apocarotenoids showed the markedly lowered levels of β-carotene (74 %) and lutein (72 %) along with the changed levels of two β-carotene (C40) cleavage products, a two-fold increase of β-ionone (C13) and de novo generation of β-cyclocitral (C10) at lower levels, compared with non-transgenic rice plants. It suggests that β-carotene could be the principal substrate being cleaved at 9–10 (9′–10′) for β-ionone and 7–8 (7′–8′) positions for β-cyclocitral by AtCCD4. This study is in planta report on the generation of apocarotenal volatiles from carotenoid substrates via cleavage by AtCCD4. We further verified that the production of these volatiles was due to the action of exogenous AtCCD4 and not the expression of endogenous rice CCD genes (OsCCD1, 4a, and 4b).  相似文献

Carotenoid cleavage dioxygenases (CCDs) in plant species is one of the most important enzymes in the carotenoid metabolism. In this study, we performed a comprehensive analysis for the CCDs family in Solanum lycopersicum based on the whole tomato genome sequences and explored their expression pattern. At least seven CCD genes were discovered in the tomato genome sequence. Two pairs of them were arranged in tandem. The tandem duplication events could be dating to approximately 14 and 21 Mya, and the tandem duplication genes experienced a purifying selection during the course of evolution after diversification. Additionally, subcellular localization revealed that four members were predicted to be cytoplasm-localized and the three remaining members plastids-localized. Subsequently, a number of cis-regulatory elements, which were involved in light responsiveness, hormone regulation, and abiotic and biotic stresses, were identified in the promoter sequences of SlCCD genes. Phylogenetic tree revealed that the CCDs from Solanaceae crops have a closer genetic relationship. The difference in abundance and distinct expression patterns during the vegetative and reproductive development suggests different functions for these seven SlCCDs. Our findings suggest that SlCCDs family play important roles throughout the whole life course and will lay the foundation for further elaborating the regulatory mechanism of each member in tomato.  相似文献

Polyphenols are secondary metabolites widely present in plants which benefit to human health. In the present study we analyzed the changes of polyphenol contents during strawberry fruit development as well as changes of polyphenol oxidase (PPO). The results depicted that the polyphenol content showed a decreasing trend with the fruit development. The pH value impacts the PPO activity, and in strawberry fruit the optimal pH for the PPO activity was 4.5. Meanwhile, PPO activity kept decreasing with the development of the fruit flesh and achenes. The damaged fruit enhanced the PPO activity. We found four PPO genes encoding the PPO in the strawberry that had different expression levels in tissues. The overexpression of the FaPPO1 genes improved the PPO activity in strawberry fruit and delays the fungus infection process. The FaPPO1 gene expression changes had affected the pathogen-related gene expression, such as PAL, SOD, POD, BG, and Chitinase genes. The fruit damage induced the FaPPO1 gene expression, and the abscisic acid and methyl jasmonic were also involved in the regulation of FaPPO1 gene expression. The FaPPO1 and FaPPO2 gene expressions were regulated both by abiotic stresses of low temperature, NaCl, and H2O2 and biotic stresses of powdery mildew and gray mold. Understanding the regulation mechanism of PPO will be helpful and provide meaningful ideas in future for strawberry breeders.  相似文献

SIMILAR TO RCD ONE (SRO) is a small plant-specific gene family, which play essential roles in plant growth and development as well as in abiotic stresses. However, the function of SROs in maize is still unknown. In our study, six putative SRO genes were isolated from the maize genome. A systematic analysis was performed to characterize the ZmSRO gene family. The ZmSRO gene family was divided into two groups according to the motif and intron/exon analysis. Phylogenetic analysis of them with other plants showed that the clades of SROs along with the divergence of monocot and dicot and ZmSROs were more closely with OsSROs. Many abiotic stress response and hormone-induced cis-regulatory elements were identified from the promoter region of ZmSROs. Furthermore, RNA-seq analysis indicated that SRO genes were widely expressed in different tissues and development stages in maize, and the expression divergence was also obviously observed. Analyses of expression in response to PEG6000 and NaCl treatment, in addition to exogenous application of ABA and GA hormones showed that the majority of the members display stress-induced expression patterns. Taken together, our results provide valuable reference for further functional analysis of the SRO gene family in maize, especially in abiotic stress responses.  相似文献

The cold shock domain proteins (CSDPs) are small group of nucleic acid-binding proteins that act as RNA chaperones in growth regulation, development, and stress adaptation in plants. The functions of CSDPs have been studied in Arabidopsis (Arabidopsis thaliana), rice (Oryza sativa), wheat (Triticum aestivum), and Chinese cabbage (Brassica rapa). To gain insight into the function of CSDPs in tomato (Solanum lycopersicum), we performed a genome-wide analysis of CSDPs through in silico characterization and expression profiling in different organs and in response to different abiotic stress and phytohormone treatments. We identified five non-redundant SlCSDP genes. The evolutionary analysis and phylogenetic classification indicated that tomato CSDPs are more closely related to potato than those of others. The five SlCSDP genes are distributed on four of the 12 tomato chromosomes and no segmental or tandem duplication events are detected among them. Expression analysis showed broad expression patterns with strong expression in fruit development and ripening. Expression of individual SlCSDP genes was significantly altered by stress and phytohormone treatments. SlCSDP2, SlCSDP3, and SlCSDP4 were highly induced by all four abiotic stresses and by phytohormone treatment in tomato. These findings provide a foundation for future research towards functional biological roles of CSDP gene in particular to develop tomato cultivars with large size, early ripening, and abiotic stress tolerance.  相似文献

Abscisic acid plays a crucial role in the regulation of fruit development and ripening, however, its role in the floral development and the fruit set is still unclear. In the present study, the ABA accumulation and the expression patterns of genes related to ABA metabolism and signalling in sweet cherry were investigated. The results showed that ABA accumulation increased and peaked at stage V in ovary, at stage VI in stamen, and in young fruit it peaked at 7 days after full bloom. The expression pattern of ABA synthetase PaNCED1 was consistent with the changes of ABA accumulation. Among four ABA degradation enzymes PaCYP707As, PaCYP707A4 was highly expressed in ovary, PaCYP707A1 was mainly in stamen, and PaCYP707A2 was in young fruit, and their expressions were reversed to the trend of PaNCED1. With regard to ABA signalling genes, among three ABA receptors PaPYLs, PaPYL2 and PaPYL3 were high expression genes in ovary and in young fruit with similar expression patterns, while PaPYL3 was the high expression gene in stamen. Within six PaPP2Cs, PaPP2C1/2/3 were highly expressed in ovary and young fruit, while PaPP2C3/4 were mainly in stamen. The six PaSnRK2s showed different expression patterns: PaSnRK2.1/2.2/2.4 were highly expressed in ovary and young fruit, while PaSnRK2.1/2.3 were highly expressed in stamen. In situ hybridization results showed that PaPYL3, PaPP2C3 and PaSnRK2.4 were expressed in seed, pulp and fruit peel during fruit set. In conclusion, ABA and its signaling may play an important role in the regulation of floral development and fruit set.  相似文献

Stress-associated proteins (SAPs) are a novel class of zinc finger proteins that extensively participate in abiotic stress responses. To date, no overall analysis and expression profiling of SAP genes in woody plants have been reported. Populus euphratica is distributed in desert regions and is extraordinarily adaptable to abiotic stresses. Thus, it is regarded as a promising candidate for studying abiotic stress resistance mechanisms of woody plants. In this study, 18 non-redundant SAP genes were identified from the genome of P. euphratica using basic local alignment search tool algorithms and functional domain verification. Among these 18 PeuSAP genes, 15 were intronless. To investigate the evolutionary relationships of SAP genes in P. euphratica and other Salicaceae plants, phylogenetic analyses were performed. Subsequently, the expression profiles of the 18 PeuSAP genes were analyzed in different tissues and under various stresses (drought, salt, heat, cold, and abscisic acid (ABA) treatment) using quantitative real-time PCR. Tissue expression analysis indicated that PeuSAPs showed no tissue specificity. PeuSAPs were induced by multiple abiotic stresses, especially drought, salt, and heat stresses, perhaps because of abundant cis-acting heat shock elements and drought-inducible elements in the promoter regions of the PeuSAPs. Moreover, single nucleotide polymorphisms (SNPs) variant analysis revealed many synonymous and non-synonymous SNPs in PeuSAP genes, but the zinc finger structure was conserved during evolution. These results provide an overview of the SAP gene family in P. euphratica and a reference for further functional research on PeuSAP genes.  相似文献

Phytohormones regulate numerous aspects of plant growth and development. Green-mature banana fruit were treated with deionized water (control), abscisic acid (ABA), indole-3-acetic acid (IAA) and ABA + IAA, respectively, to investigate the role of ABA and IAA in fruit ripening. Results showed that ABA accelerated fruit ripening, but IAA delayed the process. However, treatment of ABA + IAA showed little difference in fruit color and firmness. The acceleration of ABA and delay of IAA on banana ripening process seems to be neutralized by ABA + IAA. Digital gene expression revealed that ABA + IAA treated fruit maintained the similar color phenotype with the control by regulating the expression of chlorophyll degradation-related gene PaO (GSMUA_Achr6G25590_001), and carotenoid biosynthesis-related genes DXR (GSMUA_Achr3G20790_001) and PSY (GSMUA_Achr2G12480_001, GSMUA_Achr4G17270_001, GSMUA_Achr4G17290_001). Moreover, ABA + IAA treated fruit maintained the similar softening phenotype with the control by adjusting the expression of pectin degradation-related genes PME (GSMUA_Achr3G05740_001) and PL (GSMUA_Achr6G28160_001, GSMUA_Achr7G04580_001). ABA + IAA treatment nearly abolished the action of individual ABA or IAA through equilibrating the expression of specific genes involved in chlorophyll degradation, carotenoid biosynthesis and pectin degradation pathways in the postharvest ripening of banana. The interaction between ABA and IAA might exercise as an antagonistic mechanism of neutralizing the specific gene expression either induced by ABA or reduced by IAA in the postharvest ripening of banana.  相似文献
