The results of the experimental investigation of the biological activity of a new biinsecticide in the form of emulsifiable concentrate containing 400 g/1 of pirimiphos-methyl and 10 g/1 of bifenthrin with respect to adults of Tribolium confusum are described. One and 7 days after treatment with biinsecticide applied directly onto insects, the LD-99.9 values were 1.45 ml/m2 and < 0.01 ml/m2, respectively. For insects contacting with treated concrete surfaces, LD-99.9 was 0.27 ml/m2 and 0.1 ml/m2 1 and 7 days after treatment, respectively.  相似文献

Signal peptidase complexes (SPCs) are conserved from bacteria to human beings, and are typically composed of four to five subunits. There are four genes encoding SPC proteins in the red flour beetle, Tribolium castaneum. To understand their importance to insect development, double‐stranded RNA for each SPC gene was injected into red flour beetles at the early larval and adult stages. Knockdown of all four signal peptidase genes was lethal to larvae. Moreover, larvae had difficulty with old cuticle ecdysis. Knockdown of TcSPC12 alone did not affect pupal or adult development. When TcSPC12, TcSPC18, and TcSPC25 were knocked down in larvae, the melanization of hemocytes and midguts was observed. When knocked down in larvae and adults, TcSPC18 induced severe cell apoptosis in midguts, and the adult midgut lost the ability to maintain crypts after knockdown of TcSPC18, indicating its importance to midgut cell proliferation and differentiation. Knockdown of TcSPC22 or TcSPC25 also resulted in many apoptotic cells in the midguts. However, TcSPC12 appeared to be unimportant for midgut development. We conclude that TcSPC18 is essential for maintaining the adult midgut crypts.  相似文献

The results of experimental investigations of adult mortality of six harmful beetle species on wheat grain treated with emulsifiable concentrate of biinsecticide containing 400 g/L of pirimiphos-methyl and 10 g/L of bifenthrin are described. The period of complete protection of grain from infestation with Laemophloeus ferrugineus Steph., Rhizopertha dominica F., Sitophilus oryzae L., Oryzaephilus surinamensis L., Sitophilus granarius L., and Tribolium confusum Duv. at a dosage of 2 mL/t was 12, 4, 1, 1, 1, and 0 months, respectively; at 3 mL/t, 12, 9, 4, 2, 2, and 1 months; at 4 mL/t, 12, 9, 8, 7, 2 and 1 months; at 6 mL/t, 12, 12, 12, 12, 8, and 1 months; and at 8 mL/t, 12, 12, 12, 12, 12, and 3 months.  相似文献

Vector population control using insecticides is a key element of current strategies to prevent malaria transmission in Africa. The introduction of effective insecticides, such as the organophosphate pirimiphos-methyl, is essential to overcome the recurrent emergence of resistance driven by the highly diverse Anopheles genomes. Here, we use a population genomic approach to investigate the basis of pirimiphos-methyl resistance in the major malaria vectors Anopheles gambiae and A. coluzzii. A combination of copy number variation and a single non-synonymous substitution in the acetylcholinesterase gene, Ace1, provides the key resistance diagnostic in an A. coluzzii population from Côte d’Ivoire that we used for sequence-based association mapping, with replication in other West African populations. The Ace1 substitution and duplications occur on a unique resistance haplotype that evolved in A. gambiae and introgressed into A. coluzzii, and is now common in West Africa primarily due to selection imposed by other organophosphate or carbamate insecticides. Our findings highlight the predictive value of this complex resistance haplotype for phenotypic resistance and clarify its evolutionary history, providing tools to for molecular surveillance of the current and future effectiveness of pirimiphos-methyl based interventions.  相似文献

Laboratory toxicity studies were conducted in southeastern Queensland, Australia, to determine the acute lethal effects of a 1-h pulse exposure of selected insecticides to adult and juvenile (<72 h old) crimson-spotted rainbowfish, Melanotaenia duboulayi (Castlenau). In addition, to its ecological significance, this native fish is a predator of mosquitoes. Two organophosphate (OP) compounds (temephos and pirimiphos-methyl), an entomophathogenic bacterium (Bacillus thuringiensis variety israelensis [Bti]) and two insect growth regulators (IGRs) (s-methoprene and pyriproxyfen) were evaluated. Although none of the five insecticides were acutely toxic to adult M. duboulayi under the test conditions, temephos and pirimiphos-methyl were found to be toxic to juveniles, with 24 h pulse exposure LC50 values of 27 and 15 microg/liter (ppb), respectively. Of the two OPs, pirimiphos-methyl was the most toxic, with a lethal dose ratio (pulse exposure LC50 temephos/ pulse exposure LC50 pirimiphos-methyl) of 1.8 (95% CL 0.5-6.4). These pulse exposure LC50 values represented 40 and 4.5% of the estimated environmental concentrations (EEC) for a 15 cm deep water body, respectively. Bti and the two IGRs had no acute toxic effects at up to 10 and 12.5 times the EEC, respectively. Accordingly, in the interests of environmental conservation and integrated pest management (IPM), preference should be given to the latter three insecticides for control of mosquito larvae in juvenile M. duboulayi habitat.  相似文献

Observations on behavioural activity involved in the host selection by secondary pests of stored grains, Oryzaephilus surinamensis (Linnaeus), Tribolium castaneum (Herbst) and Tribolium confusum J. du Val, with respect to intact and mechanically or naturally damaged kernels are reported. Our results indicate that the attraction of secondary pests is facilitated by broken grain kernels, which resulted from either mechanical damage during harvesting and/or binning procedures, or the feeding activity of primary insect pests. Insect damaged kernels were more attractive to O. surinamensis, T. castaneum and T. confusum than whole kernels; in addition insect damaged kernels elicit more attractiveness than mechanically split kernels. The damage caused by primary pests, such as Rhyzopertha dominica (Fabricius) and Sitophilus oryzae (Linnaeus), on whole kernels may facilitate colonization by secondary pests, which continue damaging the cereals. O. surinamensis, T. castaneum and T. confusum utilize the grain volatile odours to distinguish whether the grain kernels of the stored cereals are damaged mechanically or by insects.  相似文献

目的:研究线粒体分裂蛋白1(Mitochondrial fission protein 1,FIS1)介导的线粒体分裂对肝癌细胞侵袭与迁移的调控作用与机制。方法:采用免疫组化实验比较10对肝癌原发灶与转移灶组织中FIS1表达,以明确FIS1与肝癌转移的关系。通过si RNA干扰FIS1的表达后,用Transwell实验检测肝癌细胞迁移与侵袭能力的变化,q PCR与Western Blot检测上皮间质转化标志分子上皮型钙黏蛋白(epithelia cadherin,E-cadherin)、紧密连接蛋白(zonula occludens-1,ZO-1)、神经型钙黏蛋白(neural cadherin,N-cadherin)、波形蛋白Vimentin的表达。结果:肝癌转移灶组织中FIS1的表达显著高于原发灶组织。干扰FIS1表达后,肝癌细胞迁移和侵袭能力均明显下降,细胞上皮间质转化标志蛋白E-cadherin和ZO-1的表达上调,而N-cadherin和Vimentin的表达下调。结论:线粒体分裂蛋白FIS1在肝癌转移灶组织中高表达,并可能通过调节细胞上皮间质转化促进肝癌细胞转移。  相似文献

The results of an experimental study of voracity of adults and larvae of seven main stored-product insect pests are described. One adult of Sitophilus oryzae, S. granarius, Rhizopertha dominica, Tribolium castaneum, T. confusum, and Oryzaephilus surinamensis consumed 0.38, 0.48, 0.89, 0.15, 0.16, and 0.11 mg of wheat grain per day, respectively. During their development, larvae of these insects consumed 7.60, 13.77, 7.92, 3.32, 3.60, and 2.32 mg of wheat grain, respectively. One larva of Sitotroga cerealella consumed 17.46 mg of wheat grain. The coefficients of harmfulness of these species are calculated, and a method for estimating the “total infestation density” of grain is proposed.  相似文献

Complementary DNA overexpression and short hairpin RNA interference approaches were evaluated for decreasing expression of primordial germ cell (PGC) marker genes and thereby sterilizing channel catfish, Ictalurus punctatus, by delivering knockdown constructs driven by a constitutive promoter from yeast and a copper transport protein gene into fish embryos by electroporation. Two PGC marker genes, nanos and dead end, were the target knockdown genes, and their expressions, along with that of an off-target gene, vasa, were evaluated temporally using real-time polymerase chain reaction. Copper sulfate was evaluated as a repressor compound. Some of the constructs knocked down PGC marker gene expression, and some of the constructs were partially repressed by application of 0.1-ppm copper sulfate. When the rate of sexual maturity was compared for three-year-old broodfish that had been exposed to the sterilizing constructs during embryologic development and controls that had not been exposed, several treatments had reduced sexual maturity for the exposed fish. Of two promoter systems evaluated, the one which had been designed to be less sensitive to copper generally was more effective at achieving sterilization and more responsive to repression. Knockdown constructs based on 3′ nanos short hairpin RNA interference appeared to result in the best repression and restoration of normal sexual maturity. We conclude that these copper-based systems exhibited good potential for repressible transgenic sterilization. Optimization of this system could allow environmentally safe application of transgenic technology and might be applicable to other applications for aquatic organisms.  相似文献

The Akt signaling pathway is well known to regulate cell proliferation and growth. Girdin, a novel substrate of Akt, plays a crucial role in organization of the actin cytoskeleton and cell motility under the control of Akt. We here identified a novel Girdin-like protein in Drosophila (dGirdin), which has two isoforms, dGirdin PA and dGirdin PB. dGirdin shows high homology with human Girdin in the N-terminal and coiled-coil domains, while diverging at the C-terminal domain. On establishment of transgenic fly lines, featuring knockdown or overexpression of dGirdin in vivo, overexpression in the wing disc cells induced ectopic apoptosis, implying a role in directing apoptosis. Knockdown of dGirdin in the Drosophila wing imaginal disc cells resulted in reduction of cell size. Furthermore, this was enhanced by half reduction of the Akt gene dose, suggesting that Akt positively regulates dGirdin. In the wing disc, cells in which dGirdin was knocked down exhibited disruption of actin filaments. From these in vivo analyses, we conclude that dGirdin is required for actin organization and regulation of appropriate cell size under control of the Akt signaling pathway.  相似文献

Trehalose is proposed to serve multiple physiological roles in insects. However, its importance remains largely unconfirmed. In the present paper, we knocked down either a trehalose biosynthesis gene (trehalose-6-phosphate synthase, LdTPS) or each of three degradation genes (soluble trehalases LdTRE1a, LdTRE1b or membrane-bound LdTRE2) in Leptinotarsa decemlineata by RNA interference (RNAi). Knockdown of LdTPS decreased trehalose content and caused larval and pupal lethality. The LdTPS RNAi survivors consumed a greater amount of foliage, obtained a heavier body mass, accumulated more glycogen, lipid and proline, and had a smaller amount of chitin compared with the controls. Ingestion of trehalose but not glucose rescued the food consumption increase and larval mass rise, increased survivorship, and recovered glycogen, lipid and chitin to the normal levels. In contrast, silencing of LdTRE1a increased trehalose content and resulted in larval and pupal lethality. The surviving LdTRE1a RNAi hypomorphs fed a smaller quantity of food, had a lighter body weight, depleted lipid and several glucogenic amino acids, and contained a smaller amount of chitin. Neither trehalose nor glucose ingestion rescued these LdTRE1a RNAi defects. Silencing of LdTRE1b caused little effects. Knockdown of LdTRE2 caused larval death, increased trehalose contents in several tissues and diminished glycogen in the brain-corpora cardiaca-corpora allata complex (BCC). Feeding glucose but not trehalose partially rescued the high mortality rate and recovered glycogen content in the BCC. It seems that trehalose is involved in feeding regulation, sugar absorption, brain energy supply and chitin biosynthesis in L. decemlineata larvae.  相似文献

Glutathione (GSH) and GSH-dependent enzymes play a key role in cellular detoxification processes that enable organism to cope with various internal and environmental stressors. However, it is often not clear, which components of the complex GSH-metabolism are required for tolerance towards a certain stressor. To address this question, a small scale RNAi-screen was carried out in Caenorhabditis elegans where GSH-related genes were systematically knocked down and worms were subsequently analysed for their survival rate under sub-lethal concentrations of arsenite and the redox cycler juglone. While the knockdown of γ-glutamylcysteine synthetase led to a diminished survival rate under arsenite stress conditions, GSR-1 (glutathione reductase) was shown to be essential for survival under juglone stress conditions. gsr-1 is the sole GSR encoding gene found in C. elegans. Knockdown of GSR-1 hardly affected total glutathione levels nor reduced glutathione/glutathione disulphide (GSH/GSSG) ratio under normal laboratory conditions. Nevertheless, when GSSG recycling was impaired by gsr-1(RNAi), GSH synthesis was induced, but not vice versa. Moreover, the impact of GSSG recycling was potentiated under oxidative stress conditions, explaining the enormous effect gsr-1(RNAi) knockdown had on juglone tolerance. Accordingly, overexpression of GSR-1 was capable of increasing stress tolerance. Furthermore, expression levels of SKN-1-regulated GSR-1 also affected life span of C. elegans, emphasising the crucial role the GSH redox state plays in both processes.  相似文献

Nilaparvata lugens harbors yeast-like symbionts (YLSs). In present paper, a genome-wide analysis found 115 genes from Ni. lugens and 90 genes from YLSs that were involved in the metabolic degradation of 20 proteinogenic amino acids. These 205 genes encoded for 77 enzymes. Accordingly, the degradation pathways for the 20 amino acids were manually constructed. It is postulated that Ni. lugens can independently degrade fourteen amino acids (threonine, alanine, glycine, serine, aspartate, asparagine, phenylalanine, tyrosine, glutamate, glutamine, proline, histidine, leucine and lysine). Ni. lugens and YLSs enzymes may work collaboratively to break down tryptophan, cysteine, arginine, isoleucine, methionine and valine. We cloned a lysine-ketoglutarate reductase/saccharopine dehydrogenase gene (Nllkr/sdh) that encoded a bifunctional enzyme catalyzing the first two steps of lysine catabolism. Nllkr/sdh is widely expressed in the first through fifth instar nymphs and adults, and is highly expressed in the fat body, ovary and gut in adults. Ingestion of dsNllkr/sdh by nymphs successfully knocked down the target gene, and caused nymphal/adult mortality, shortened nymphal development stage and reduced adult fresh weight. Moreover, Nllkr/sdh knockdown resulted in three defects: wings were shortened and thickened; cuticles were stretched and thinned; and old nymphal cuticles remained on the tips of legs and abdomen and were not completely shed. These data indicate that impaired lysine degradation negatively affects the survival and development of Ni. lugens.  相似文献

The soil contact effects of several commonly used agricultural insecticides on 2nd instar and adultForficula auricularia (L.) were studied in the laboratory. The earwigs were individually confined on soil surfaces sprayed under a track sprayer. Pirimicarb had no effect on either adult or 2nd instar earwigs. The LD50,s of 2nd instar earwigs for γ HCH, fenitrothion, DDT and dimethoate were all lower than their normal field rates. Second instars were more susceptible to dimethoate and cypermethrin than adults. The pyrethroids cypermethrin and deltamethrin had LD50,s above normal field rates. However, high levels of knockdown were induced by both insecticides at concentrations below normal application rates. The effects of this in the field remain unknown.  相似文献

Cereals are staple food for many countries and are grown on millions of hectares of land, but much of the harvest is wasted due to losses by pests. To minimize these losses, many pesticides are used which are damaging to the environment and human health. There are debates to get rid of these chemicals but they are still in use at large scale. An alternative control strategy for insect pests in storage houses is the use of botanicals. In this study, four plant essential oils, two plant extracts, two herbicides, and two insecticides were used against Tribolium confusum and the comparison of toxicity was made by calculating LC50 and LT50 values. LC50 values were higher for abamectin (2.09–10.23 mg/L) and cypermethrin (3.41–11.78 mg/L) insecticides followed by neem essential oil (7.39–19.24 mg/L) and citrus extract (10.14–24.50 mg/L). However, LC50 values were maximum in case of jaman plant extract (22.38–176.42 mg/L) followed by two herbicides, Logran (19.66–39.72 mg/L) and Topik (29.09–47.67 mg/L) However, LC50 values were higher for topic herbicide (24.098 ppm) and jaman essential oil (16.383 ppm) after four days of treatment. Abamectin and cypermethrin insecticides, neem essential oil and citrus plant extract also killed adults of T. confusum quicker as compared other essential oils, extracts and herbicides. Results revealed that botanical formulations being environmentally safe could be used instead of highly hazardous pesticides for stored products’ pests. This study also elaborates the non-host toxicity of herbicides commonly applied in our agroecosystem.  相似文献

This study examines the biological efficacy of four mosquito mat vaporizers each containing different active ingredients: prallethrin with PBO, dimefluthrin, prallethrin, and d-allethrin. The glass chamber assay was used to evaluate their efficacy on Aedes albopictus (Skuse) (Diptera: Culicidae) from nine districts in Selangor, Malaysia. Aedes albopictus exhibited different knockdown rates, with 50% knockdown times, KT50, varying from 1.19 to 2.00 min, 1.22 to 2.20 min, 1.39 to 5.85 min, and 1.39 to 1.92 min for prallethrin with PBO, dimefluthrin, prallethrin and d-allethrin, respectively. In general, all populations of Ae. albopictus were completely knocked down after exposure to all active ingredients except Hulu Selangor population, which showed 96.00% knockdown against d-allethrin. On the contrary, mortality rates were observed from 84.00–100.00%, 84.00–100.00%, 90.67–100.00% and 90.67–100.00% in populations tested with prallethrin with PBO, dimefluthrin, prallethrin and d-allethrin, respectively. Moreover, significant correlations between mortality rates of prallethrin with PBO vs dimefluthrin (r = 0.836, P = 0.003), prallethrin with PBO vs prallethrin (r = 0.760, P = 0.011), and prallethrin vs d-allethrin (r = 0.694, P = 0.026) were also observed, suggesting cross-resistance among pyrethroids. d-allethrin was found to be high in insecticidal activity, followed by prallethrin, prallethrin with PBO, and dimefluthrin. In consistent with mortality due to insecticide exposure, elevated levels of enzyme activities were also demonstrated in Sabak Bernam, Hulu Selangor, Gombak, Petaling, Hulu Langat and Klang populations.  相似文献

不同杀虫剂选育对家蝇抗药性水平及kdr基因频率的影响   总被引:4,自引:0,他引:4  
采用杀虫剂(溴氰菊酯和甲基嘧啶磷)筛选及不接触药物自然衰退的方法,研究了家蝇Musca domestica氯氟氰菊酯高抗品系(Cyh-R)对杀虫剂的抗性变化,探讨蝇类抗药性治理的方法。用点滴法测定氯氟氰菊酯对不同家蝇品系的毒力,比较抗药性的变化,结合特异性等位基因PCR扩增(PASA)技术检测了不同家蝇品系的kdr基因频率,探讨kdr基因频率与抗性水平之间的关系。结果表明:甲基嘧啶磷筛选后,氯氟氰菊酯对第2~8代Cyh-R品系的LD50呈递减趋势,从F0的2.8434 μg/蝇降为F8的0.4404 μg/蝇,但第8~18代Cyh-R品系的LD50呈逐代递增趋势;溴氰菊酯筛选后,氯氟氰菊酯对Cyh-R品系第2~16代的LD50呈上升趋势,从F0的2.8434 μg/蝇升至F16的24.3249 μg/蝇;表明了施用有机磷杀虫剂可降低其对氯氟氰菊酯的抗药性,而施用拟除虫菊酯药剂则有助于其对氯氟氰菊酯抗药性的增长;不使用任何杀虫剂也能降低其对氯氟氰菊酯的抗药性,但下降速率低于甲基嘧啶磷。PASA技术检测表明Cyh-R品系的kdr抗性基因频率为88.8%,不经过任何药剂筛选其kdr抗性基因频率下降程度最大,达到69.7%;甲基嘧啶磷筛选后其结果降为78.8%,而经溴氰菊酯筛选后kdr抗性基因频率则明显升高,达到98.9%。通过对kdr抗性基因频率和抗性水平进行相关和回归分析表明kdr抗性基因频率与家蝇对氯氟氰菊酯的LD50呈对数关系,即LD50值高的品系其kdr抗性基因频率相应的也较高。建议在家蝇防治中考虑轮换用药。  相似文献

The confused flour beetle, Tribolium confusum, is a common and severe pest of stored products. Here, using fumigation tests during four different exposure times, we evaluated the toxicity of different doses of essential oil of spike lavender, Lavandula spica and one of its major constituents, linalool, on different life stages of T. confusum under laboratory conditions. The toxicity of the L. spica oil and linalool varied as a function of the developmental stage and treatment duration. Young larvae (L1) were the most susceptible to toxic effects, with LC50 = 19.535 μl/L of air for L. spica oil and LC50 = 14.198 μl/L of air for linalool after 24 h of exposure, whereas older larvae (L8) were affected only very little by fumigation. Linalool caused higher egg mortality than L. spica oil at equivalent doses, but lower mortality in pupae and adults. Emergence of intact adult insects from surviving eggs, larvae and pupae was further reduced as a function of dose and exposure time to both L. spica oil and linalool compared to control-treated insects. Our results show that L. spica oil and linalool might be suitable for biological control of T. confusum, but tests at a larger scale are necessary to confirm our results.  相似文献
