Nitrogen dioxide's rats' inhalations with injections per os of pyrazole, amidopyrine and sodium nitrite lead to considerable increasing of endogenic N-nitrosodimethylamine formation, which had been determined by system gas chromatograph-thermal energetic analyser. This increasing essentially didn't depend on the rats' immunisation by vaccine BCG, which leads to the intensification of NO synthesis by peritoneal macrophages and others manifestations of their metabolic activation: increasing of creatine kinase and adenosine desaminase activities. It hadn't been brought to light the obvious dependent between changes of xanthine oxidase and xanthine dehydrogenase activities in the liver and blood serum and intensification of lipids peroxidation and also the amount of N-nitrosodimethylamine in the rats in the conditions of endogenic and exogenic nitrosation factors' influence.  相似文献

The effect of exogenous ascorbic acid intake on biosynthesis of ascorbic acid in mice has been studied. After the mice were on diets containing added ascorbic acid for two months, the activities of ascorbic acid synthesizing enzymes in the mouse liver homogenates were measured using L-gulono-gamma-lactone as a substrate. Exogenous ascorbic acid intake (0.5, 1 or 5% in the diet) was able to increase the concentration of ascorbic acid in the blood and to decrease the activities of ascorbic acid synthesizing enzymes in mouse liver. The results suggest that ascorbic acid synthesis was controlled by local regulatory mechanism or by the concentration of ascorbic acid in the hepatic portal blood. Ingestion of dietary erythorbic acid, a stereoisomer of ascorbic acid, had no effect on the activities of ascorbic acid synthesizing enzymes.  相似文献

A ninhydrin positive compound (L2) now identified as 2-amino-5-(N-ethylcarboxyamido)-pentanoic acid, from unprocessed tea leaves was a potent inhibitor of thrombin-stimulated thromboxane formation in rabbit whole blood (Ali and Afzal; Prostaglandins, Leukotrienes and Medicine, 27: 9, 1987). In the present study, processed and unprocessed tea leaf extracts were given to rats to consume for a period of eight weeks. Cholesterol and thromboxane levels were measured in the serum obtained from clotting the blood at 37 degrees C. A significant reduction in thromboxane levels was observed in rats taking unprocessed tea extract. This reduction was equally distributed in adult as well as in juvenile rats. However no appreciable changes in the levels of thromboxane were noticed in the serum of rats taking processed tea extracts. This might be due to the presence of a labile component which is destroyed during the processing of green tea leaves. A decreased level of cholesterol was observed in rats consuming unprocessed tea extract. This decrease could be linked to the decrease in thromboxane levels as observed. Processed tea refers to commercially available tea of different brands while unprocessed tea refers to dried green tea leaves.  相似文献

The antioxidant activity of green tea (GT) has been extensively studied; however, the results obtained from dietary intervention studies are controversial. In the present study we investigated the effect of the addition of two cups of GT (containing approximately 250 mg of total catechins) to a controlled diet in a group of healthy volunteers with respect to a group following the same controlled diet but not consuming GT. Antioxidant status and lipid profile in plasma, the resistance from oxidative damage to lipid and DNA, and the activity of glutathione peroxidase (GPX) in isolated lymphocytes were measured at the beginning and the end of the trial. After 42 days, consumption of GT caused a significant increase in plasma total antioxidant activity [from 1.79 to 1.98 micromol Trolox equivalent (TE)/ml, P<.001], significant decreases in plasma peroxides level (from 412 to 288 Carr U, P<.05) and induced DNA oxidative damage in lymphocytes (from 14.2% to 10.1% of DNA in tail, P<.05), a moderate although significant decrease in LDL cholesterol (from 119.9 to 106.6 mg/dL, P<.05) with respect to control. The present study suggests the ability of GT, consumed within a balanced controlled diet, to improve overall the antioxidative status and to protect against oxidative damage in humans.  相似文献

A 2-mo experiment with the white Sprague-Dawley (SD) rats was conducted to investigate the effect of the water extracts of black tea (BTWE) and green tea (GTWE) and the black tea leaves (BTF) and the green tea leaves (GTF) on the metabolism of mineral elements. One hundred eight 12-mo-old white SD rats were randomly divided into 13 groups; 6 of these drank the BTWE or GTWE in which the water extracts concentrations of black tea or green tea were, respectively, 0.6%, 1.2%, and 2.4%, and 6 of these had black tea leaves (BTF) and green tea leaves (GTF) added in which the contents of BTF or GTF were, respectively, 0.5%, 1.0%, and 2.0%, one of these was control. The teas and their water extracts could promote the absorption of manganese and copper. In GTF, BTF, GTWE, and BTWE, the apparent absorption rates of manganese were significantly increased. The manganese contents in the tibia were also elevated, and the differences between GTWE and GTF were significant. The apparent absorption rates of copper and the copper contents in the tibia were increased, but not significantly. The teas and their water extracts inhibited the absorption of calcium (p > 0.05) and iron (p < 0.05). The cerebrum calcium contents were significantly decreased, but the contents of calcium and iron in tibia were not significantly changed. Compared with the control, although the apparent absorption rates of aluminum in all experimental groups were not observed to be different, the aluminum contents in the tibia (p > 0.05) and cerebrum (p < 0.05) were increased. GTF and GTWE decreased the apparent absorption rates of zinc, but BTF and BTWE increased them; the zinc contents in tibia were a little improved, whereas its contents in the cerebrum were gradually decreased with the increase of tea leaves dose and tea concentration.  相似文献

Isolated adrenal cells from Vitamin E-deficient and control rats were prepared by a trypsin digestion method. Cyclic adenosine 3',5'-monophosphate (cyclic AMP) formation was studied in response to adrenocorticotropin (ACTH) in the presence and absence of ascorbate by measuring the conversion of prelabeled adenosine 5'-triphosphate [14C]ATP to cyclic [14C]AMP. Ascorbate (0.5 mM) inhibited ACTH-induced cyclic [14C]AMP formation in adrenal cells isolated from Vitamin E-deficient rats but had no effect in the control cells. The inhibitory effect of ascorbate on ACTH-induced cyclic AMP formation in Vitamin E-deficient rats decreased as the concentration of ACTH increased. In Vitamin E-deficient rats ascorbate inhibited ACTH-induced cyclic [14C]AMP formation after 30 min of incubation. There was no further significant accumulation of cyclic [14C]AMP at 60 min or 120 min although in the absence of ascorbate cyclic [14C]AMP continued to be formed. The in vitro addition of alpha-tocopherol reduced the inhibition of ACTH-induced cyclic [14C]AMP formation by ascorbate in Vitamin E-deficient rats. These studies suggest that alpha-tocopherol and ascorbate may affect ACTH-induced cyclic AMP formation through interaction with the membrane-bound enzyme adenylate cyclase.  相似文献

The effects of dietary powdered green tea (PGT) and theanine on in vivo hepatoma growth and cancerous hyperlipidemia were investigated in rats that had been implanted with a rat ascites hepatoma cell line of AH109A cells. The hepatoma-bearing rats were fed with a 20% casein diet (20C), 20C containing 2% PGT, or 20C containing 0.1% theanine for 14 days. Dietary PGT significantly and time-dependently reduced the solid tumor volume and weight as did dietary theanine. The hepatoma-induced endogenous hyperlipidemia, which was characterized by rises in the serum cholesterol (hypercholesterolemia) and triglyceride (hypertriglyceridemia) levels, was significantly suppressed by PGT and theanine supplementation. Bile acid excretion into the feces was significantly higher in the PGT- and theanine-fed rats than in the control rats. This inhibition of hypercholesterolemia may have resulted from tumor growth suppression as well as increased excretion of steroids from the body. These results suggest that PGT had both anti-proliferative activity toward hepatoma cells and hypolipidemic activity in the hepatoma bearing rats. They also suggest that theanine was, at least in part, responsible for the PGT actions.  相似文献

The antimicrobial effects and biofilm formation inhibition of tea polyphenols (TPP) extracted from Korean green tea (Camellia sinensis L) were evaluated against 12 oral microorganisms. Effective antimicrobial activity against all microorganisms tested, including Lactobacillus spp. (Lactobacillus acidophilus and Lactobacillus plantarum), Streptococcus spp. (Streptococcus mutans, Streptococcus sanguis, Streptococcus sobrinus, Streptococcus mitis, and Streptococcus salivarius), Staphylococcus aureus, Neisseria meningitidis, Escherichia coli, Enterobacter cloacae, Enterococcus faecalis, and Candida albicans, was shown at 2,000 μg/mL TPP within 5 min of incubation. Scanning electron microscopy (SEM) analysis revealed various morphological changes, such as the presence of perforations, the formation of cell aggregates, and the leakage of cytoplasmic materials from cells treated with TPP, depending on the bacteria. The potential role of TPP in biofilm formation inhibition on human teeth was evaluated in BHI broth with 2 mixed strains of S. mutans and S. sanguis. SEM analysis showed biofilm formation on the surface of a tooth shaken only in saline solution, whereas almost no biofilm was observed on a tooth incubated in TPP solution. This result suggests that TPP is effective against adherent cells of S. mutans and S. sanguis. Thus, TPP would be useful for development as an antimicrobial agent against oral microorganisms, and has great potential for use in mouthwash solutions for the prevention and treatment of dental caries.  相似文献

Effect of ascorbic acid on rhinovirus replication in WI-38 cells.   总被引:3,自引:0,他引:3  
Exposure of WI-38 cells to ascorbic acid plus glutathione mixtures in growth medium for 2 days prior to infection with rhinovirus serotype 20 and during virus replication suppressed multicyclic but not single-cycle growth of the virus. Multicyclic growth was suppressed over the range of multiplicity of infection (m.o.i.) of 4 times 10-minus 1 minus 4 times 10-minus 4 PFU/cell. There was some suggestion that, in the presence of ascorbic acid plus glutathione, interferon was produced at the highest m.o.i. tested but at barely detectable levels.  相似文献
