Degrading high-strength phenol using aerobic granular sludge   总被引:1,自引:0,他引:1  
Aerobic granules were adopted to degrade high-strength phenol wastewater in batch experiments. The acclimated granules effectively degraded phenol at a concentration of up to 5,000 mg l−1 without severe inhibitory effects. The biodegradation of phenol by activated sludge was inhibited at phenol concentrations >3,000 mg l−1. The granules were composed of cells embedded in a compact extracellular matrix. After acid or alkaline pretreatment, the granules continued to degrade phenol at an acceptable rate. The polymerase chain reaction-denaturing gradient gel electrophoresis technique was employed to monitor the microbial communities of the activated sludge and the aerobic granules following their being used to treat high concentrations of phenol in batch tests.  相似文献

A spiral packed-bed bioreactor inoculated with microorganisms obtained from activated sludge was used to conduct a feasibility study for phenol removal. The reactor was operated continuously at various phenol loadings ranging from 53 to 201.4 g m−3 h−1, and at different hydraulic retention times (HRT) in the range of 20–180 min to estimate the performance of the device. The results indicated that phenol removal efficiency ranging from 82.9 to 100% can be reached when the reactor is operated at an HRT of 1 h and a phenol loading of less than 111.9 g m−3 h−1. At an influent phenol concentration of 201.4 g m−3, the removal efficiency increased from 18.6 to 76.9% with an increase in the HRT (20–120 min). For treatment of phenol in the reactor, the maximum biodegradation rate (V m) was 1.82 mg l−1 min−1; the half-saturation constant (K s), 34.95 mg l−1.  相似文献

Results of this study describe the feasibility of anaerobic treatment of highly concentrated phenol synthetic wastewater using an anaerobic fluidized bed reactor (AFBR) in both continuous and batch modes. Wastewater with a maximum load of 2,100 mg C·l−1 was prepared using phenol (maximum concentration of 1,600 mg C·l−1) as substrate and a mixture of acetic, propionic and butyric acids (500 mg C·l−1) as co-substrate. AFBR reached total organic carbon (TOC) and phenol removal efficiency over 95% treating the highest organic loading rate (OLR) containing phenol studied for this kind of reactor (5.03 g C·l−1·d−1). The phenol loading rate rise caused volumetric biogas rate increase up to 4.4 l·l−1·d−1 (average yield of 0.28 l CH4·g−1 CODremoved) as well as variation in the biogas composition; the CO2 percentage increased while the CH4 percentage decreased. Morphological examination of the bioparticles at 4.10 g C·l−1·d−1, revealed significant differences in the biofilm structure, microbial colonization and bacterial morphological type development. The five batch assays showed that phenol degradation may be favoured by the presence of volatile fatty acids (VFAs) (co-metabolism), whereas VFAs degradation may be inhibited by phenol. The activated sludge process (ASP) has high operational costs due to the need for aeration at dissolved O2 (DO) levels of ≥2 mg l−1 and high capital costs to construct large reactors due to a low organic loading [typically 1 kg chemical oxygen demand (COD) m−3 day−1]. A novel method for improving the energy use and treatment efficiency of the ASP via limited oxygenation (0.4 mg DO l−1) and high organic loading (6.2 kg COD m−3 day−1) is proposed based on a laboratory-scale ASP for ammonia-rich industrial wastewaters. The sludge blanket phenomenon and granulation occurred simultaneously in the upflow microaerobic reactor.  相似文献

Methyl iso-butyl ketone (MIBK) is a widely used volatile organic compound (VOC) which is highly toxic in nature and has significant adverse effects on human beings. The present study deals with the removal of MIBK using biodegradation by an acclimated mixed culture developed from activated sludge. The biodegradation of MIBK is studied for an initial MIBK concentration ranging from 200–700 mg l−1 in a batch mode of operation. The maximum specific growth rate achieved is 0.128 h−1 at 600 mg l−1of initial MIBK concentration. The kinetic parameters are estimated using five growth kinetic models for biodegradation of organic compounds available in the literature. The experimental data found to fit well with the Luong model (R 2 = 0.904) as compared to Haldane model (R 2 = 0.702) and Edward model (R 2 = 0.786). The coefficient of determination (R 2) obtained for the other two models, Monod and Powell models are 0.497 and 0.533, respectively. The biodegradation rate found to follow the three-half-order kinetics and the resulting kinetic parameters are reported.  相似文献

A sequencing batch reactor (SBR) system is demonstrated to biologically remove nitrogen, phosphorus and chemical oxygen demand (COD) to very low levels from abattoir wastewater. Each 6 h cycle contained three anoxic/anaerobic and aerobic sub-cycles with wastewater fed at the beginning of each anoxic/anaerobic period. The step-feed strategy was applied to avoid high-level build-up of nitrate or nitrite during nitrification, and therefore to facilitate the creation of anaerobic conditions required for biological phosphorus removal. A high degree removal of total phosphorus (>98%), total nitrogen (>97%) and total COD (>95%) was consistently and reliably achieved after a 3-month start-up period. The concentrations of total phosphate and inorganic nitrogen in the effluent were consistently lower than 0.2 mg P l−1 and 8 mg N l−1, respectively. Fluorescence in situ hybridization revealed that the sludge was enriched in Accumulibacter spp. (20–40%), a known polyphosphate accumulating organism, whereas the known glycogen accumulating organisms were almost absent. The SBR received two streams of abattoir wastewater, namely the effluent from a full-scale anaerobic pond (75%) and the effluent from a lab-scale high-rate pre-fermentor (25%), both receiving raw abattoir wastewater as feed. The pond effluent contained approximately 250 mg N l−1 total nitrogen and 40 mg P l−1 of total phosphorus, but relatively low levels of soluble COD (around 500 mg l−1). The high-rate lab-scale pre-fermentor, operated at 37°C and with a sludge retention time of 1 day, proved to be a cheap and effective method for providing supplementary volatile fatty acids allowing for high-degree of biological nutrient removal from abattoir wastewater.  相似文献

Combining chemical and biological treatments is a potentially economic approach to remove high concentration of recalcitrant compounds from wastewaters. In the present study, the biodegradation of 1,4-benzoquinone, an intermediate compound formed during phenol oxidation by chlorine dioxide, was investigated using Pseudomonas putida (ATCC 17484) in batch and continuous bioreactors. Batch experiments were conducted to determine the effects of 1,4-benzoquinone concentration and temperature on the microbial activity and biodegradation kinetics. Using the generated data, the maximum specific growth rate and biodegradation rate were determined as 0.94 h−1 and 6.71 mg of 1,4-benzoquinone l−1 h−1. Biodegradation in a continuous bioreactor indicated a linear relationship between substrate loading and biodegradation rates prior to wash out of the cells, with a maximum biodegradation rate of 246 mg l−1 h−1 observed at a loading rate of 275 mg l−1 h−1 (residence time: 1.82 h). Biokinetic parameters were also determined using the steady state substrate and biomass concentrations at various dilution rates and compared to those obtained in batch cultures.  相似文献

Biotreatment of bagasse effluent using Phanerochaete chrysosporium (white rot fungus) is investigated. This study confirmed that lignin is the major pollutant component in this effluent followed by different carbohydrates. The treatment conditions must be very proper, especially in terms of biomass culture to achieve a successful treatment. The best conditions of temperature, biomass concentration, pH and duration for biotreatment of this effluent were 35°C, 552 mg l−1, 6 and 5 to 9 days, respectively. Under these conditions, a 9 days long treatment reduced by 98.7% the original biochemical oxygen demand (of 2,780 mg l−1) and by 98.5% the dissolved chemical oxygen demand (initial 4,200 mg l−1). Moreover, fungal treatment reduced total dissolved solids from 3,950 to 575 mg l−1 and color from 560 mg l−1 PtCo to 111 mg l−1 PtCo.  相似文献

The longer start-up period of the Anammox process is due to the very low cellular yield and growth rates of Anammox bacteria. Nitrite inhibition is considered to be the key factor in the instability of the Anammox process during the operation. However, little attention was paid to the inhibitory effect of pH and free ammonia. This paper presents start-up and inhibition analysis of an Anammox biofilm reactor seeded with anaerobic granular sludge. Results showed that the start-up period could be divided into the sludge lysis phase, lag phase, propagation phase, stationary phase and inhibition phase. Optimization control could be implemented correspondingly to accelerate the start-up of Anammox bioreactors. Effluent pH increased to 8.7–9.1 when the nitrogen removal rate was higher than 1,200 mg l−1 day−1. The free ammonia concentration was accompanied with a higher level of 64–73 mg l−1. Inhibitory effects of high pH and free ammonia on Anammox bacteria contributed to the destabilization of the Anammox bioreactor during the first 125 days with influent KHCO3 of 0.5 g l−1. Increasing the suffering capacity in the inlet by dosing 1.25 g KHCO3 l−1 effectively reduced the pH variation, and the nitrogen removal performance of the reactor was further developed.  相似文献

The denitrification performance of a lab-scale anoxic rotating biological contactor (RBC) using landfill leachate with high nitrate concentration was evaluated. Under a carbon to nitrogen ratio (C/N) of 2, the reactor achieved N-NO3 removal efficiencies above 95% for concentrations up to 100 mg N-NO3  l−1. The highest observed denitrification rate was 55 mg N-NO3  l−1 h−1 (15 g N-NO3  m−2 d−1) at a nitrate concentration of 560 mg N-NO3  l−1. Although the reactor has revealed a very good performance in terms of denitrification, effluent chemical oxygen demand (COD) concentrations were still high for direct discharge. The results obtained in a subsequent experiment at constant nitrate concentration (220 mg N-NO3  l−1) and lower C/N ratios (1.2 and 1.5) evidenced that the organic matter present in the leachate was non-biodegradable. A phosphorus concentration of 10 mg P-PO4 3− l−1 promoted autotrophic denitrification, revealing the importance of phosphorus concentration on biological denitrification processes.  相似文献

An efficient protocol for high frequency in vitro regeneration of multiple shoots and somatic embryos from the embryonic axis of common bean (Phaseolus vulgaris) was developed. Ten common bean cultivars representing a wide range of diversity among current commercial market classes were used for in vitro regeneration evaluation in our study. These cultivars were tested on 63 different media formulations consisting of combinations of cytokinins, namely benzyladenine (BA) and thidiazuron (TDZ) at concentration levels of 0.0, 1.0, 2.5, 5.0 and 10.0 mg l−1 and auxin, namely naphthalene acetic acid (NAA) and indole-3-acetic acid (IAA) at concentration levels of 0.0, 0.05, 0.1 and 1.0 mg l−1. P. vulgaris cv. Olathe pinto bean performed the best producing over 20 multiple shoots per explant while cv. Condor black bean was the poorest with nine multiple shoots per explant. The optimum media for regeneration of multiple shoots was 4.4 mg l−1 Murashige and Skoog (MS) containing 2.5 mg l−1 BA and 0.1 mg l−1 IAA supplemented with 30 mg l−1 silver nitrate. Adventitious shoots and somatic embryos were regenerated on 4.4 mg l−1 MS medium containing 1 mg l−1 TDZ and 0.05 mg l−1 NAA supplemented with 30 mg l−1 silver nitrate or activated charcoal. Efficient and effective rooting of plantlets was achieved by dipping the cut end base of in vitro regenerated shoots in 1.0 mg l−1 indole-3-butyric acid (IBA) solution and culturing on media containing 4.4 mg l−1 MS supplemented by 0.1 mg l−1 IAA, NAA or IBA.  相似文献

Beauvericin (BEA) is a cyclic hexadepsipeptide mycotoxin with notable phytotoxic and insecticidal activities. Fusarium redolens Dzf2 is a highly BEA-producing fungus isolated from a medicinal plant. The aim of the current study was to develop a simple and valid kinetic model for F. redolens Dzf2 mycelial growth and the optimal fed-batch operation for efficient BEA production. A modified Monod model with substrate (glucose) and product (BEA) inhibition was constructed based on the culture characteristics of F. redolens Dzf2 mycelia in a liquid medium. Model parameters were derived by simulation of the experimental data from batch culture. The model fitted closely with the experimental data over 20–50 g l−1 glucose concentration range in batch fermentation. The kinetic model together with the stoichiometric relationships for biomass, substrate and product was applied to predict the optimal feeding scheme for fed-batch fermentation, leading to 54% higher BEA yield (299 mg l−1) than in the batch culture (194 mg l−1). The modified Monod model incorporating substrate and product inhibition was proven adequate for describing the growth kinetics of F. redolens Dzf2 mycelial culture at suitable but not excessive initial glucose levels in batch and fed-batch cultures.  相似文献

The fungal strain, Aspergillus niger SA1, isolated from textile wastewater sludge was screened for its decolorization ability for four different textile dyes. It was initially adapted to higher concentration of dyes (10–1,000 mg l−1) on solid culture medium after repeated sub-culturing. Maximum resistant level (mg l−1) sustained by fungal strain against four dyes was in order of; Acid red 151 (850) > Orange II (650) > Drimarene blue K2RL (550) > Sulfur black (500). The apparent dye removal for dyes was seen largely due to biosorption/bioadsorption into/onto the fungal biomass. Decolorization of Acid red 151, Orange II, Sulfur black and Drimarine blue K2RL was 68.64 and 66.72, 43.23 and 44.52, 21.74 and 28.18, 39.45 and 9.33% in two different liquid media under static condition, whereas, it was 67.26, 78.08, 45.83 and 13.74% with 1.40, 1.73, 5.16 and 1.87 mg l−1 of biomass production under shaking conditions respectively in 8 days. The residual amount (mg l−1) of the three products (α-naphthol, sulfanilic acid and aniline) kept quite low i.e., ≤2 in case AR 151 and Or II under shaking conditions. Results clearly elucidated the role of Aspergillus niger SA1 in decolorizing/degrading structurally different dyes into basic constituents.  相似文献

Toxic at low concentrations, phenol is one of the most common organic pollutants in air and water. In this work, phenol biodegradation was studied in extreme conditions (80°C, pH = 3.2) in a 2.7 l bioreactor with the thermoacidophilic archaeon Sulfolobus solfataricus 98/2. The strain was first acclimatized to phenol on a mixture of glucose (2000 mg l−1) and phenol (94 mg l−1) at a constant dissolved oxygen concentration of 1.5 mg l−1. After a short lag-phase, only glucose was consumed. Phenol degradation then began while glucose was still present in the reactor. When glucose was exhausted, phenol was used for respiration and then for biomass build-up. After several batch runs (phenol < 365 mg l−1), specific growth rate (μX) was 0.034 ± 0.001 h−1, specific phenol degradation rate (qP) was 57.5 ± 2 mg g−1 h−1, biomass yield (YX/P) was 52.2 ± 1.1 g mol−1, and oxygen yield factor ( \textY\textX/\textO 2 ) \left( {{\text{Y}}_{{{\text{X}}/{\text{O}}_{ 2} }} } \right) was 9.2 ± 0.2 g mol−1. A carbon recovery close to 100% suggested that phenol was exclusively transformed into biomass (35%) and CO2 (65%). Molar phenol oxidation constant ( \textY\textO 2 /\textP ) \left( {{\text{Y}}_{{{\text{O}}_{ 2} /{\text{P}}}} } \right) was calculated from stoichiometry of phenol oxidation and introducing experimental biomass and CO2 conversion yields on phenol, leading to values varying between 4.78 and 5.22 mol mol−1. Respiratory quotient was about 0.84 mol mol−1, very close to theoretical value (0.87 mol mol−1). Carbon dioxide production, oxygen demand and redox potential, monitored on-line, were good indicators of growth, substrate consumption and exhaustion, and can therefore be usefully employed for industrial phenol bioremediation in extreme environments.  相似文献

Constant medium feeding rate and intermittent fed-batch fermentation strategies were investigated aiming to increase the yields of γ-decalactone production by Yarrowia lipolytica, using methyl ricinoleate as substrate and ricinoleic acid source. The accumulation of another compound, 3-hydroxy-γ-decalactone, was also analyzed since it derives from the direct precursor of γ-decalactone thereby providing information about the enzymatic activities of the pathway. Both strategies were compared with the traditional batch mode in terms of overall productivity and yield in respect to the substrate. Although the productivity of γ-decalactone was considerably higher in the batch mode (168 mg l−1 h−1), substrate conversion to lactone (73 mg γ-decalactone g−1) was greater in the intermittent fed-batch giving 6.8 g γ-decalactone l−1. This last strategy therefore has potential for γ-decalactone production at an industrial level.  相似文献

In the present study, the bioremoval of Cr(VI) and the removal of total organic carbon (TOC) were achieved with a system composed by an anaerobic filter and a submerged biofilter with intermittent aeration using a mixed culture of microorganisms originating from contaminated sludge. In the aforementioned biofilters, the concentrations of chromium, carbon, and nitrogen were optimized according to response surface methodology. The initial concentration of Cr(VI) was 137.35 mg l−1, and a bioremoval of 85.23% was attained. The optimal conditions for the removal of TOC were 4 to 8 g l−1 of sodium acetate, >0.8 g l−1 of ammonium chloride and 60 to 100 mg l−1 of Cr(VI). The results revealed that ammonium chloride had the strongest effect on the TOC removal, and 120 mg l−1 of Cr(VI) could be removed after 156 h of operation. Moreover, 100% of the Cr(VI) and the total chromium content of the aerobic reactor output were removed, and TOC removals of 80 and 87% were attained after operating the anaerobic and aerobic reactors for 130 and 142 h, respectively. The concentrations of cells in both reactors remained nearly constant over time. The residence time distribution was obtained to evaluate the flow through the bioreactors.  相似文献

The effect of increasing concentration of polycyclic aromatic hydrocarbon (PAH) fluoranthene (FLT; 0.1, 1 and 5 mg l−1) on the growth, ethylene production and anatomy of stems of 21-day-old pea plants cultivated in vitro in MS medium, with or without FLT, enriched with 0.1 mg l−1 indole-3-acetic acid (IAA) or with combination of 0.1 mg l−1 IAA + 0.1 mg l−1 N6-benzyladenine (BA) were investigated. The low concentration of 0.1 mg l−1 FLT, in both IAA- and IAA + BA-treated plants, significantly stimulated the growth of pea callus, while higher concentrations 1 mg l−1 and especially 5 mg l−1 FLT significantly inhibited it. Pea shoots were significantly influenced only after application of 5 mg l−1 FLT in IAA treatment. Significantly increased production of ethylene was found in IAA + BA treatments in all concentrations of FLT, whereas in IAA treatments in 1 and 5 mg l−1 FLT. The lysigenous aerenchyma formation in the cortex of pea stems significantly increased in all FLT treatments and its highest proportion was found in plants exposed to 1 mg l−1 FLT.  相似文献

A bacterial consortium that can degrade chloro- and nitrophenols has been isolated from the rhizosphere of Phragmitis communis. Degradation of 4-chlorophenol (4-CP) by a consortium attached to granular activated carbon (GAC) in a biofilm reactor was evaluated during both open and closed modes of operation. During the operation of the biofilm reactor, 4-CP was not detected in the column effluent, being either adsorbed to the GAC or biodegraded by the consortium. When 4-CP at 100 mg l−1 was fed to the column in open mode operation (20 mg g−1 GAC total supply), up to 27% was immediately available for biodegradation, the rest being adsorbed to the GAC. Biodegradation continued after the system was returned to closed mode operation, indicating that GAC bound 4-CP became available to the consortium. Biofilm batch cultures supplied with 10–216 mg 4-CP g−1 GAC suggested that a residual fraction of GAC-bound 4-CP was biologically unavailable. The consortium was able to metabolise 4-CP after perturbations by the addition of chromium (Cr VI) at 1–5 mg l−1 and nitrate at concentrations up to 400 mg l−1. The development of the biofilm structure was analysed by scanning electron microscopy and confocal laser scanning microscopy (CLSM) techniques. CLSM revealed a heterogeneous structure with a network of channels throughout the biofilm, partially occupied by microbial exopolymer structures. The fungal strain A. niger SA1 isolated from textile wastewater pond proved to be an important source of remediation (decolorization/degradation) for textile dye, AR 151 (Reactive diazo dye) under different physicochemical conditions. Decolorization assays of AR 151 were carried out in Simulated textile effluent under shake flask condition for 8 days. Decolorization (at 20 mg l−1 of dye) and related biomass production overall decreased with increase in pH from 5 to 9, at 30°C. It was maximum (95.71%) at pH 5 with highest amount of three residual products (36.91 (α-naphthol = 5.72) (sulfanilic acid = 24.81) (aniline = 6.38)) besides 2.05 mg ml−1 of biomass production at an optimum concentration 6 and 0.1 mg l−1 of glucose and urea respectively. The formation of the three products followed a quite different pattern at different pH values, however, it was considerably low (Total = 2.81 mg l−1) compared to the amount of decolorization (67.26%) at pH 8. Decolorization (95–97%) was most favored under mesophilic temperature (25–45°C). It increased i.e., 90–98% with subsequent increase in dye from 10 to 100 mg l−1, kept ≥50% below 400 mg l−1 and drastically declined to 17% at 500 mg l−1 of dye. Apparently, decolorization is found to be associated with fungal growth and hyphal uptake mechanism (Biosorption/Bioadsorption), however, mineralization of AR 151 and related products under different operational conditions also suggested a metabolically mediated decolorization/degradation.  相似文献

The aim of this study was to investigate the potential of the green microalga Chlorella saccharophila as a source of oil for biodiesel production. We evaluated for the first time, the effect of salinity and/or nitrogen depletion (ND) on cell growth, lipid accumulation and lipid profile in this microalga. The fatty acid methyl esters (FAME) identified for C. saccharophila in this study consisted of C-16:0, C-18:0, C-18:1 cis, and C-18:1 trans. Among these, C-18:1 (indicator of biodiesel quality) was the main FAME found, representing approximately 76 and 80% of total FAME under normal and ND growing conditions, respectively. Under a normal growing condition this microalga showed 154.63 mg l−1 d−1, 63.33 mg l−1 d−1, and 103.73 mg l−1 of biomass productivity, lipid productivity, and FAME yield, respectively. The higher biomass productivity (159.58 mg l−1 d−1), lipid productivity (99.33 mg l−1 d−1), and FAME yield (315.53 mg l−1) were obtained under the ND treatment. In comparison to other related studies, our results suggest that C. saccharophila can be considered as a suitable source of oil for biodiesel production.  相似文献
