The molecular toxicity of monotertiarybutylhydroquionone (TBHQ) was studied using Tetrahymenapyriformis as a model cell system. TBHQ at 26 ppm in the media inhibited cell growth by 50%. TBHQ inhibited the oxidation of 14C-acetate to 14CO2. In addition, increasing concentrations of TBHQ decreased the incorporation of 14C-acetate into lipids and protein, 14C-amino acids into protein, 3H-uridine into RNA and 3H-thymidine into DNA. The incorporation of 14C-acetate into glycogen increased with concentrations up to 20 ppm TBHQ in the media while glycogen synthesis decreased with 40 ppm TBHQ.  相似文献

Cysteine sulfinic acid decarboxylase activity is not affected when measured in vitro in the presence of pyridoxal phosphate in the brains of vitamin B6-deficient rats. Activity of this enzyme was not detectable in the brains of vitamin B6-deficient animals when assayed in the absence of pyridoxal phosphate. The activity of cysteine sulfinic acid decarboxylase in crude particulate and soluble fractions of rat brain reflects the distribution of the enzyme in vivo and the distribution of endogenous B6 vitamers when assayed in the presence and absence of vitamin B6. This study indicates that virtually no taurine is synthesized by any component of brain when the animal is subjected to a deficiency of vitamin B6.  相似文献

1. The nucleic acid metabolism in the pyridoxine-deficient rat has been investigated through studies on the incorporation of radioactivity from various isotopically labelled compounds into liver and spleen DNA and RNA. 2. In pyridoxine deficiency, the incorporation of radioactivity from sodium [14C]formate was apparently increased. The magnitude of this effect on incorporation into liver RNA and DNA and spleen RNA was approximately the same. The incorporation into spleen DNA was enhanced to a much greater degree. Administration of pyridoxine 24hr. before the rats were killed reversed the changes in incorporation of radioactivity from [14C]formate. 3. In pyridoxine deficiency, the incorporation of radioactivity from dl-[3-14C]serine, [8-14C]adenine, [Me-3H]thymidine and [2-14C]deoxyuridine was decreased. The incorporation of radioactivity from l-[Me-14C]methionine was not affected. No noteworthy differences in the effect of pyridoxine deficiency on the incorporation of radioactivity from dl-[3-14C]serine into DNA and RNA were observed, whereas the effect of the deficiency on the incorporation of radioactivity from [8-14C]adenine into spleen DNA was somewhat greater than that into spleen RNA. Administration of pyridoxine 24hr. before the rats were killed reversed the changes in incorporation of radioactivity from [3-14C]serine and [8-14C]adenine. 4. The adverse effects of pyridoxine deficiency on the biosynthesis of nucleic acids and cell multiplication are discussed in relation to the role of pyridoxal phosphate in the production of C1 units via the serine-hydroxymethylase reaction.  相似文献

Folate deficiency and pancreatic acinar cell function   总被引:1,自引:0,他引:1  
The present study was designed to determine the effect of folate deficiency on pancreatic acinar cell function. In the first series of experiments, three groups of rats were fed ad libitum regular rat feed, folate-deficient diet, or an equivalent amount of folate-sufficient diet. In the second series of experiments, rats were either fed ad libitum or rendered folate deficient by a purified folate-deficient diet; half of the folate-deficient group was replenished with oral folate. Body weight, pancreatic weight, DNA [methyl-14C]thymidine incorporation into DNA, RNA, [8-14C]adenine incorporation into RNA, protein content, synthesis of proteins, amylase content, and basal and bethanechol-stimulated amylase secretion were determined. The parameters were the same in the rats fed a folate-sufficient diet as in those fed a regular rat feed. Feeding a folate-deficient diet resulted in impaired DNA synthesis as evidenced by diminished incorporation of [methyl-14C]thymidine into DNA. There was no change in secretion of amylase. Similar results were obtained in the second series of experiments. These studies indicate that folate deficiency (rather than antibiotic content of the diet) impaired pancreatic function. Folate deficiency may therefore contribute to pancreatic injury in malnutrition and alcoholism.  相似文献

The soluble androgen acceptor has been isolated from 0.35 M NaCl extract of rat prostatic chromatin by affinity chromatography on DNA-cellulose. The acceptor activity was assayed by interaction with 5α-dihydrotestosterone-receptor. Native DNA enhances this interaction. Polyacrylamide gel electrophoresis of the acceptor under denaturing conditions reveals a single polypeptide of molecular weight of 14,000. Amino acid analysis shows that the acceptor protein contains a higher content of acidic amino acid residues than basic amino acid residues. In an invitro RNA synthesizing system catalyzed by rat RNA polymerase II, addition of the acceptor stimulates RNA synthesis. Based on incorporation of [γ-32P]ATP and [γ-32P]GTP, the stimulation by the acceptor is mainly on the initiation of RNA chains.  相似文献

The activities of alternative pathways of glucose metabolism in developing rat brain were evaluated by measurement of the yields of 14CO2 from glucose labeled with 14C on carbons 1, 2, 3 + 4, 6 and uniformly labeled glucose, from the detritiation of [2-3H]glucose and from the incorporation of 14C from specifically labeled glucose into lipids by brain slices from cerebral hemispheres and cerebellum. The glycolytic route and tricarboxylic acid cycle (14CO2 yield from carbons 3, 4, and 6 of glucose) increased during development. The flux through the glutamate-γ-aminobutyric route (14CO2 yield from carbon 2-carbon 6 of glucose) also showed an increase with development. In contrast, the proportion of glucose metabolized via the pentose phosphate pathway was markedly decreased as development progressed. The artificial electron acceptor, phenazine methosulfate, was used as a probe to investigate the effect of alterations in the redox state of NADP+NADPH couple on a number of NADP-linked systems in developing brain. Phenazine methosulfate produced a massive (20- to 50-fold) stimulation of the pentose phosphate pathway, in contrast, the incorporation of glucose carbon into fatty acids and flux through the glutamate-γ-aminobutyrate shunt were sharply decreased. The effects of phenazine methosulfate on the incorporation of glucose into glyceride glycerol, on the flux of glucose through the pyruvate dehydrogenase reaction and tricarboxylic acid cycle, all processes linked to the NAD+NADH couple, appeared to be minimal in the brain at the stages of development studied, i.e., 1, 5, 10, 20 days, and in the adult rat. The significance of the massive reserve potential of the pentose phosphate pathway in the developing brain is discussed.  相似文献

When cycloheximide is added to (B12)-deficient cultures before or after replenishment of the cells with B12, reversion of these cells is inhibited. This inhibition is not caused by interference of the inhibitor in the uptake of B12 as measured by division kinetics. Cycloheximide does not inhibit the initial increase in the rate of DNA synthesis caused by B12 replenishment, but within 30–45 min the rate decreases and DNA synthesis ceases. Cycloheximide added to replenished deficient cells after completion of DNA duplication inhibits cell division. The total cellular protein and RNA in replenished cells treated with cycloheximide does not change. B12 added to deficient cells does not stimulate the incorporation of [14C]leucine into protein during resumption and completion of DNA duplication. However, there is a large increase in [14C]leucine incorporation into the protein of these cells soon after completion of DNA duplication and before resumption of cell division. The addition of cycloheximide to B12-replenished or to nonreplenished deficient cells rapidly inhibits the incorporation. We suggest that the addition of B12 accelerates the rate of DNA synthesis in the deficient cells and that possibly no new protein synthesis is required except for mitosis. However, protein synthesis is needed for continuous DNA synthesis.  相似文献

Isolated axons of Eriocheir sinensis show high ratios of 14CO2 production from glucose-1-14C to 14CO2 production from glucose-6-14C (ratio C1/C6). During osmotic stresses, there is a modification in 14CO production from glucose-6-14C as well as in the ratio C1/C6 while 14CO2 production from glucose-1-14C does not change significantly. These results are interpreted in terms of activity of oxidative and non-oxidative pathways of glucose metabolism.  相似文献

[1-14C]acetate was readily incorporated into unsaturated fatty acids by leaf slices of spinach, barley and whole cells of Chlorellapyrenoidosa and Candidabogoriensis. In these systems the [14C] label in newly synthesized oleate and linoleate was approximately equally distributed in the C1–9 and the C10–18 fragments obtained by reductive ozonolysis of these acids, whereas in a-linolenic acid over 90% of the total [14C] was localized in the C1–9 fragment. While [1-14C]oleic acid was converted by whole cells of Chlorella to [1-14C]linoleic and [1-14C]linolenic acids, [U-14C]oleic acid yielded [U-14C]linoleic acid but a-linolenic acid was labeled only in the carboxyl terminal carbon atoms. When spinach leaf slices were supplied with carboxyl labeled octanoic, decanoic, dodecanoic, tetradecanoic and octadecanoic acids, only the first three acids were converted to a-linolenic acids while the last two acids were ineffective. Thus we suggest that (a) linoleic acid is not the precursor of a-linolenic acid and (b) 12:3(3, 6, 9) is the earliest permissible trienoic acid which is then elongated to a-linolenic acid.  相似文献

14C-Sterigmatocystin isolated from cultures of Aspergillusversicolor supplemented with (1-14C)acetate was shown to be efficiently converted to aflatoxin B1 by the resting mycelium of A. parasiticus. The experimental results may indicate a biosynthetic pathway leading from 5-hydroxysterigmatocystin to sterigmatocystin and then to aflatoxin B1.  相似文献

A doubly labeled 3-ketoceramide, [1-14C] lignoceroyl [1-3H2] 3-ketosphingosine (3H14C ratio, 3.61) was injected into the left ventricle of rat heart. The ceramide isolated from the livers of the animals after 1 hr incubation contained an equal 3H>14C ratio of 3.60. This finding strongly supports the existence for direct conversion of 3-ketoceramide to ceramide in rat liver.  相似文献

Rice callus tissue in suspension culture was labeled invivo with 32PO4?3. Approximately 4% of the total RNA extracted from the tissue could be bound to nitrocellulose filters in the presence of 0.5 M KCl. The base composition of this material was: A, 30.6; U, 32.7; G, 18.0; and C, 18.6%. If the RNA was digested with both T1 and pancreatic ribonucleases prior to binding, the base composition of the bound material was: A, 97.8; U, 0.0; G, 1.6; and C, 0.5%. About 14% of the RNA isolated from purified polyribosomes could be bound to filters in 0.5 M KCl. It is concluded that higher plant RNA contains polyadenylate sequences.  相似文献

A 0.5 × 106Mr RNA found in plastids of the aquatic angiosperm Spirodela, is synthesized at a much higher rate than any other rapidly labeling RNA species about 3–312 h after dark-grown plants are transferred to light. The pulse labeling kinetics of the 0.5 × 106Mr RNA after transfer to light, argue against its involvement in the biogenesis of plant rRNAs. Although poly(A) RNA is found in Spirodela, poly(A) sequences are not detected in the 0.5 × 106Mr RNA; yet a sucrose gradient fraction which includes RNA of this Mr stimulates amino acid incorporation by an E. coli cell free extract more than other RNA fractions. The possible involvement of the 0.5 × 106Mr RNA as a chloroplast messenger is discussed.  相似文献

Isolated livers from fed rats were perfused with a medium containing glucose labeled uniformly with 14C and specifically with 3H. There was considerable formation of glucose from endogenous sources but simultaneously uptake of about half of the 14C in glucose. After 2 hours the 3H14C ratios in perfusate glucose decreased by 55–60% with (2-3H, U-14C), 40–50% with (5-3H, U-14C), 25–30% with (3-3H or 4-3H, U-14C) and by 10–15% with (6-3H, U-14C) glucose. Qualitatively comparable patterns were obtained with rat hepatocytes. These results demonstrate recycling of carbon between glucose and pyruvate. Superimposed upon this there is an extensive futile cycle between glucose and glucose 6-P. There is also futile cycling between fructose 6-P and fructose 1,6 P2 and to a small extent between phosphoenol pyruvate and pyruvate.  相似文献

Biosynthesis of N-methyl-l-glucosamine moiety of streptomycin from d-glucose by Streptomyces griseus was studied. A mixture of d-[1-14C]glucose and d-[6-3H]glucose was given to the culture of S. griseus. The 3H/14C ratio found in N-methyl-d-glucosamine further supports a mechanism that the conversion of d-glucose to l-hexose is carried out without scission of carbon skeleton. When d-[1-14C]glucose and d-[3-3H]glucose were used, the fall of 3H/14C ratio in N-methyl-l-glucosamine showed that the hydrogen atom at C-3 plays a rôle in such a transformation.  相似文献

The phosphorylation invivo of RNA polymerase II after isoproterenol stimulation of confluent rat C6 glioma cell cultures has been investigated. Glioma cells were incubated in the presence of Na2H32PO4 and stimulated for 1 hour with the β-adrenergic agonist isoproterenol. The phosphorylation pattern was analyzed after purification of RNA polymerase II by immunoprecipitation, SDS-polyacrylamide gel electrophoresis and autoradiography. Isoproterenol markedly increased [32P]phosphate incorporation into the 214,000 dalton RNA polymerase subunit. Analysis of the phosphate acceptor amino acid revealed the presence of only [32P]phosphoserine. The data demonstrates an isoproterenol-induced structural modification of RNA polymerase II.  相似文献

O6-Methyl[8-3H]deoxyguanosine in a synthetic DNA polymer, poly(dC, dG, m6dG), is demethylated by cell-free extracts of EscherichiacoliBr adapted by exposure to N-methyl-N′-nitro-N-nitrosoguanidine, as shown by the appearance of 3H-labeled deoxyguanosine in hydrolysates of the recovered DNA. The demethylating activity could not be detected in extracts of nonadapted E. coli. These results provide direct evidence that a previously described inducible repair activity in E. coli acts by demethylating O6-methylguanine at the DNA level.  相似文献
