Maize plants were grown in nutrient solution without phosphate,or in which inorganic phosphate (Pi) was maintained at nearlyconstant concentrations of 1 µM, 10µM or 0·5mM. In vivo 31P-NMR measurements showed that there was no discernibledifference in the cytoplasmic Pi content (µmol cm–3root volume) of the mature roots of plants exposed to 1 µM,10µM or 0·5 mM external phosphate for up to 12d. However, the vacuolar Pi content of the mature roots variedabout 10-fold between these three groups. The cytoplasmic Pi content of roots receiving no external phosphatedecreased significantly after about 7 d total growth, and atabout this time the vacuolar pool of Pi became too small foraccurate measurement. The presence of 1 µM Pi in the nutrientsolution completely prevented this decline in cytoplasmic Pi,and there was some evidence that it also raised the Pi contentof the root vacuoles above the almost undetectable level foundin the totally P-starved roots. During the first 7–9 d of growth, the nucleoside triphosphatecontent of the mature roots was unaffected by the concentrationof phosphate in the nutrient solution. The results highlight the close control of cytoplasmic concentrationsof certain important phosphorus metabolites in roots growingin soil of normal agricultural fertility. This paper examines the control of phosphate uptake into Chara corallina. Influxes of inorganic phosphate (Pi) into isolated single internodal cells were measured with 32Pi. Pretreatment of cells without Pi for up to 10 d increased Pi influx. However, during this starvation the concentrations of Pi in both the cytoplasm and the vacuole remained quite constant. When cells were pre-treated with 0.1 mM Pi, the subsequent influx of Pi was low. Under these conditions the Pi concentrations in the cytoplasm was almost the same as that of Pi-starved cells, but vacuolar Pi increased with time. Transfer of cells from medium containing 0.1 mM Pi to Pi-free medium induced an increase of Pi influx within 3 d irrespective of the concentration of Pi in the vacuole.During Pi starvation, neither the membrane potential nor the cytoplasmic pH changed. Manipulation of the cytoplasmic pH by weak acids or ammonium decreased the Pi influx slightly.Pi efflux was also measured, using cells loaded with 32Pi. Addition of a low concentration of Pi in the rinsing medium rapidly and temporarily induced an increase in the efflux.The results show that Pi influx is controlled by factors other than simple feedback from cytoplasmic or vacuolar Pi concentrations or thermodynamic driving forces for H+-coupled Pi uptake. Inorganic phosphate (Pi) homeostasis in multi-cellular eukaryotes depends not only on Pi influx into cells, but also on Pi efflux. Examples in plants for which Pi efflux is crucial are transfer of Pi into the xylem of roots and release of Pi at the peri-arbuscular interface of mycorrhizal roots. Despite its importance, no protein has been identified that specifically mediates phosphate efflux either in animals or plants. The Arabidopsis thaliana PHO1 gene is expressed in roots, and was previously shown to be involved in long-distance transfer of Pi from the root to the shoot. Here we show that PHO1 over-expression in the shoot of A. thaliana led to a two- to threefold increase in shoot Pi content and a severe reduction in shoot growth. (31) P-NMR in vivo showed a normal initial distribution of intracellular Pi between the cytoplasm and the vacuole in leaves over-expressing PHO1, followed by a large efflux of Pi into the infiltration medium, leading to a rapid reduction of the vacuolar Pi pool. Furthermore, the Pi concentration in leaf xylem exudates from intact plants was more than 100-fold higher in PHO1 over-expressing plants compared to wild-type. Together, these results show that PHO1 over-expression in leaves leads to a dramatic efflux of Pi out of cells and into the xylem vessel, revealing a crucial role for PHO1 in Pi efflux.  相似文献

The extent of phosphate uptake measured by the relative changes in cytoplasmic Pi, vacuolar Pi, ATP, glucose-6-phosphate, and UDPG was determined using in vivo31P nuclear magnetic resonance spectroscopy. Maize (Zea mays) root tips were perfused with a solution containing 0.5 or 1.0 millimolar phosphate at pH ~6.5 under different conditions. In the aerated state, phosphate uptake resulted in a significant increase (>80%) in vacuolar Pi, but cytoplasmic Pi only transiently increased by 10%. Under N2, the cytoplasmic Pi increased ~150% which could be attributed to a large extent to the breakdown of ATP, sugar phosphates and UDPG. Vacuolar Pi increased but only to the extent of ~10% of that seen under aerobic conditions. 2-deoxyglucose pretreatment was utilized to decrease the level of cytoplasmic Pi. When pretreated with the 2-deoxyglucose, the excised maize roots absorbed phosphate from the perfusate with a significant increase in the cytoplasmic Pi. The increase could only be traced to external phosphate since the concentrations of other phosphorus containing species remained constant during the uptake period. With 2-deoxyglucose pretreatment, phosphate uptake under anaerobic conditions was substantially inhibited with only the vacuolar phosphate showing a slight increase. When roots were treated with carbonyl cyanide m-chlorophenyl hydrazone, no detectable Pi uptake was found. These results were used to propose a H+-ATPase related transport mechanism for phosphate uptake and compartmentation in corn root cells.  相似文献

The short-lived radio-isotope nitrogen-13 (half-life 10 min)was used as a tracer in studying fluxes of N in the roots ofintact barley plants. After supplying the plants with 13N-nitratefor 30 min, efflux of 13N into an unlabelled (wash) solutionwas followed under steady-state conditions for a further 10min. Tests with ion exchange resins suggested that all of the13N released during this period was in the form of nitrate. In addition to nitrate from a surface film of solution and fromthe free space of the roots, efflux from another compartmentwas detected, tentatively identified as the cytoplasmic nitratepool. In plants grown with nitrate as the only external N-source,efflux from this compartment decreased with a rate constantabout 0·17 min–1 (half-time 4 min). Adding ammoniumsulphate to the wash solution alone did not significantly affecteither the initial rate, or the rate constant, of efflux of13N from these roots. However, 13N efflux decreased more rapidly(rate constant about 0·32 min–1, half-time 2·2min) in roots grown in, and subsequently washed with, solutioncontaining ammonium nitrate. In barley plants grown with 1·5 mol m–3 nitrate,the cytoplasmic nitrate pool was estimated to contain about2% of the total nitrate in the roots, corresponding to a cytoplasmicnitrate concentration 26 mol m–3. Unidirectional fluxes and the cytoplasmic and vacuolar contentsof potassium and sodium in root cells of intact barley seedlings(Hordeum vulgare L., cv. Villa) were determined by use of compartmentalanalysis. In addition, the net vacuolar accumulation Jcv andthe xylem transport øcx of K+ and Na+ were measured.Both of these data were needed for the evaluation of the effluxdata. Fluxes and compartmental contents of K+ and Na+ were comparableto data obtained with excised roots. The effect of the shoot-to-rootratio—as varied by partial excision of the seedlings seminalroots—on the fluxes and contents was investigated. Highershoot-to-root ratios induced an increase in xylem transport,in plasmalemma influx, and also in the cytoplasmic content ofK+ and Na+. With potassium the plasmalemma efflux was almostunaltered while the tonoplast fluxes and vacuolar content weredecreased (in presence of Na+). With sodium, on the other hand,the plasmalemma efflux and the tonoplast fluxes were also increasedin the plants having one root and a high shoot-to-root ratio.These changes occurred even under conditions of low humidity,when transpiration was low and guttation occurred. The latterwas also increased at the high shoot-to-root ratio. The observedchanges could be due to a relieved feedback control of ion fluxesby the shoot and mediated in part by a relatively higher supplyof photosynthates in the plants having one root In addition,hormonal signals were suggested to participate. In particulara possibly decreased level of cytokinins in the plants havingonly one root could contribute to the signal. The observed changesappear to be responses of the plant to an alteration that canoccur under natural conditions when the root system is damaged.  相似文献

The effects of growth and assay temperature on unidirectionalK+ fluxes in excised roots of rye (Secale cereale cv. Rheidol)were studied using 86Rb+ as a tracer. Both K+ influx to thevacuole, estimated as K+ uptake between 3 and 12 h after transferof unlabelled roots to radioactive solution, and movement ofK+ to the xylem were determined directly. Other fluxes weredetermined on excised roots of plants, which had been labelledwith 86Rb+ since germination, by conventional triple exponentialefflux analysis. When assayed at 20°C, roots of plants previously grown at20°C(WG roots) had lower rates of net K+ uptake than rootsof low temperature-acclimated plants, grown with a temperaturediferential between roots (87°C) and shoots (20°C) eithersince germination (DG roots) or for 3 d prior to experiments(DT roots). This resulted from a greater unidirectional K+ effluxacross the plasma membrane and a reduced K+ flux to the xylemin WG roots, compared to DG or DT roots, rather than a decreasein unidirectional K+ influx or a decrease in the net K+ fluxto the vacuole. Indeed, although WG roots had lower rates ofK+ influx and K+ efflux across the tonoplast at 20°C thanDG or DT roots, roots of plants from all growth temperaturetreatments showed an equivalent net K+ flux to the vacuole. Although all unidirectional K+ fluxes in roots from plants grownunder all temperature regimes were reduced by lowering the temperatureof the root, these fluxes were differentially affected in rootsof plants from contrasting growth temperature treatments. Rapidcooling to 8°C of WG roots resulted in a lower rate of K+influx and a transient increase in K+ efflux across both theplasma membrane and tonoplast, compared to DG and DT roots.Furthermore, since the K+ flux to the xylem was lower in WGroots, the net K+ uptake at 8°C into WG roots was considerablyreduced compared to DG and DT roots. These results suggest thatlow temperature-acclimation of K+ fluxes in rye roots may involvea reduction in the temperature sensitivity of K+ influx anda curtailment of K+ efflux across both the plasma membrane andtonoplast at low temperatures. 31P NMR studies of spinach leaves and their chloroplasts   总被引:3,自引:0,他引:3  
An experimental arrangement is described which enables high quality 31P NMR spectra of compressed spinach leaf pieces to be continuously recorded in which all the resonances observed (cytoplasmic and vacuolar Pi, glycerate-3-P, nucleotides) were sharp and well resolved. 31P NMR spectra obtained from intact chloroplasts showed a distinct peak of stromal Pi. An upfield shift of the stromal Pi resonance was associated with a decrease in the external Pi and vice versa. Nucleotides were largely invisible to NMR in intact chloroplasts, whereas the same nucleotides reappeared in a typical 31P NMR spectrum of an acid extract of intact chloroplasts. Perfusion of compressed spinach leaf pieces with a medium containing Pi triggered a dramatic increase in the vacuolar Pi over 12 h. Addition of choline to the Pi-free perfusate of compressed leaf pieces resulted in a steady accumulation of phosphorylcholine in the cytoplasmic compartment at the expense of cytoplasmic Pi. When a threshold of cytoplasmic Pi concentration was attained, Pi was drawn from the vacuole to sustain choline phosphorylation. In spinach leaves, the vacuole represents a potentially large Pi reservoir, and cycling of Pi through vacuolar influx (energy dependent) and efflux pathways is an efficient system that may provide for control over the cytosolic-free Pi and phosphorylated intermediate concentrations. 31P NMR spectra of neutralized perchloric acid extracts of spinach leaves showed well defined multipeak resonances (quadruplet) of intracellular phytate. Orthophosphate (Pi) accumulation by barley (Hordeum vulgareL.) roots was specifically inhibited by NO3 as comparedto Cl and SO42 –, and Pi secretion into the xylemwas stimulated. The inhibition of Pi accumulation by NO3was also observed in roots of intact photosynthesizing horsebean(Vicia faba L.), rice (Oryza sativa L.) and soybean (Glycinemax L.) plants. NO3 effects on Pi transport by rootswere more thoroughly investigated with corn (Zea mays L.). Theywere due to intracellular NO3. Pi secretion was stillstimulated by NO3 after Pi withdrawal from the absorptionsolution. 32Pi influx decreased during Pi accumulation, supportingthe hypothesis that this ion allosterically regulated its owntransport system by feedback control. This control was modulatedby other anions: the decrease was more pronounced in the presenceof nitrate. Chronologically, the depressive effect of NO3on 32Pi influx appeared after the inhibition of Pi accumulation.Furthermore, under conditions where Pi accumulation was notaffected by NO3, 32Pi influx and Pi secretion into thexylem became insensitive to the presence of nitrate. Our hypothesisis that the stimulative effect of NO3 on Pi secretionand the depressive one on 32Pi influx are the repercussionsof an increase in the Pi cytosolic concentration due to an NO3-induced decrease in Pi uptake by the vacuoles. The cytoplasmic pH and the vacuolar pH in root-tip cells ofintact mung bean seedlings under high-NaCl stress were measuredby in vivo 31P-nuclear magnetic resonance (31P-NMR) spectroscopy.When roots were incubated with high levels (100 mM) of NaClat the control external concentration (0.5 mM) of Ca2+ ions,the vacuolar pH increased rapidly from 5.6 to 6.2 within 3 h,while the cytoplasmic pH only decreased by a mere 0.1 pH uniteven after a 24-h incubation under high-NaCl conditions. Theincrease in vacuolar pH induced by the high-NaCl stress wasdiminished by an increase in the external concentration of Ca2+ions from 0.5 mM to 5 mM. The intracellular concentration ofNa+ ions in the root-tip cells increased dramatically upon perfusionof the root cells with 100 mM NaCl, and high external levelsof Ca2+ ions also suppressed the in flow of Na+ ions into thecells. The vacuolar alkalization observed in salt-stressed rootsmay be related to the inhibition of an H+-translocating pyrophosphatasein the tonoplast, caused by the increase in the cytoplasmicconcentration of Na+ ions. It is suggested that, although thevacuolar pH increased markedly under salt stress, the cytoplasmicpH was tightly regulated by some unidentified mechanisms, suchas stimulation of the H+-translocating ATPase of the plasmalemma,in roots of mung bean under salt stress. Short term experiments investigated the effects of potassiumsupply on radiocaesium influx/efflux and the radiocaesium compartmentationin intact spring wheat roots. The efflux of Cl at the plasma membrane of Chara wasstudied in relation to two treatments known to affect the flux:that of removal of external Cl and of light. It is shownthat although removal of external Cl results in a rapidreduction in Cl efflux (consistent with a direct effectof external Cl on the transport system) the magnitudeof this reduction in the dark is greater than the measured darkinflux. Therefore, in the dark at least, it is proposed that1:1 exchange diffusion cannot account for the trans-stimulationof efflux by external Cl. Light induces an inhibition of efflux and a concomitant stimulationof Cl influx at 20 °C, but at 10 °C the responsesto light of the two fluxes can be separated temporally. It istherefore suggested that the fluxes are not reciprocally dependenton the same factor which mediates in the light response. Furtherconsiderations show that it is unlikely that the cytoplasmicCl concentration mediates in the light response of eitherflux, but that changes in cytoplasmic pH may do so.  相似文献

The development of phosphate deficiency (P-stress) was observed in rooted sprouts of Solanum tuberosum L. cv. Desiree growing in solutions without phosphate. Shoot growth was inhibited by P-stress within 3 to 5 days of terminating the phosphate supply, while significant effects on root growth were not recorded until 7 to 9 days. Thus, the shoot:root dry weight ratio decreased from 4.3 to 2.6 over a 10-day period. Growth in the absence of an exogenous phosphate supply progressively diluted the phosphorus in the plant. The proportional decrease in concentration was similar in roots and shoots over a 7-day period, even though the former were growing more quickly. The potential for phosphate uptake per unit weight of root increased rapidly during the first 3 days of P-stress. When the plants were provided subsequently with a labelled, 1 mol m?3 phosphate solution, the absorption rate was 3 to 4-fold greater than that of control plants which had received a continuous phosphate supply. The increased rate of uptake by P-stressed plants was accounted for by an increase (3-fold) in the Vmax of system 1 for phosphate transport and by a marked increase in the affinity of the system for phosphate (decrease in Km). In the early stages of P-stress, before marked changes in growth were measured, the proportion of labelled phosphate translocated to the shoots increased slightly relative to the controls when a phosphate supply was restored. In the later stages of stress a greater proportion was retained in the root system of P-stressed plants than in that of controls. In plants with roots divided between solutions containing or lacking a phosphate supply, the increased absorption rate was determined by the general demand for phosphate in the plant and not by the P-status of the particular root where uptake was measured. By contrast, the poportion translocated was strongly dependent on the P-status of the root. The restoration of a phosphate supply to P-stressed plants was marked by a rapid increase in the P concentration in snoots and roots which returned to levels similar to unstressed controls within 24 h. The enhanced uptake rate persisted for at least 5 days, resulting in supra-normal concentrations of P in both shoots and roots, and in the formation of extensive necrotic areas between the veins of mature leaves. Pea plants (Pisum sativum L.) were supplied with external phosphatefor differing periods of time, so that their phosphorus statusvaried, and the intracellular distribution of inorganic phosphate(P1) in the roots was examined by 31P nuclear magnetic resonance.Over the range of phosphorus nutrition investigated, the quantityof vacuolar P1 per unit fresh weight of root tip changed considerably,whereas the quantity of cytoplasmic P1 per unit fresh weightof root tip did not alter. The relative volumes of the cytoplasmand the vacuole in pea root tips seemed to be little affectedby differences in phosphorus nutrition, and this implied thatthe concentration of P1 in the cytoplasm was kept almost constant,at a level estimated to be 18 mM. The rate of absorption of 32P-labelled phosphate was negativelycorrelated with the vacuolar P1 concentration, but there wasno clear correlation with the concentration of P1 in the cytoplasm. Rapid, Reversible Inhibition of Nitrate Influx in Barley by Ammonium   总被引:18,自引:2,他引:16  
The rate of influx of nitrate into the roots of intact barleyplants was measured over a period of 3–5 min from externalnitrate concentrations of 1–150 mmol m–3, using13N-labelled nitrate as tracer. Ammonium at external concentrationsof 0.005–50 mol m–3 inhibited nitrate influx ina manner which did not conform to a simple kinetic model butincreased approximately as the logarithm of the ammonium concentration.At any particular ammonium concentration, inhibition of nitrateinflux reached its full extent within 3 min of the ammoniumbeing supplied and was not made more severe by up to 17 minpre-treatment with ammonium. On removing the external ammonium,nitrate influx returned to its original rate within about 3min. Potassium at 0.005–50 mol m–3 did not reproducethe rapid effect of ammonium on nitrate influx. Net uptake of nitrate also decreased when ammonium was supplied,over a similar timescale and to a similar extent as nitrateinflux. The decrease in nitrate influx caused by ammonium wassufficient to account for the observed reduction in net uptake,without necessitating any acceleration of nitrate efflux. Excised roots obtained from maize plants, previously treatedfor 24 h with different solutes (K2SO4, Na2SO4, or mannitol)at decreasing osmotic potential values, showed clearly the persistenceof osmo-saline shock in their chloride transport and in somemetabolic processes. In particular, vacuolar accumulation ofchloride was much reduced when the osmotic potential in pretreatmenthad been low; this effect was greater with solutions of electrolytes.Protein synthesis and [14C]leucine uptake were also reduced;thus there appeared to be a correlation between chloride accumulationin the vacuole and protein synthesis. By contrast, accumulationof chloride in the cytoplasm was only slightly modified or evenstimulated. Chloride influx was affected less by osmotic pretreatmentthan accumulation, except in the case of very low osmotic potentialswhere influx was depressed. Chloride efflux was slightly enhancedby osmo-saline shocks and the fluxes at the tonoplast were lessaffected in comparison with those at the plasmalemma. Malateaccumulation was stimulated by shock, but this effect was notpersistent, so that a correlation with the effects on accumulationand fluxes was not possible. Oxygen uptake was affected onlyslightly except that at osmotic potential values of –750kPa, perhaps because of a lowered viability of the roots subjectedto very low osmotic potential for 24 h.  相似文献

Ammonia (pKa 9.25) and methylamine (pKa, 10.65) increase cytoplasmicpH and stimulate Cl influx in Chara corallina, theseeffects being associated with influx of the amine cations ona specific porter. The weak base imidazole (pKa 6.96) has similareffects but diffuses passively into the cell both as an unionizedbase and as a cation. When the external pH is greater than 6.0influx of the unionized species predominates. Imidazole accumulates to high concentrations in the vacuole,where it is protonated. Cytoplasmic pH and vacuolar pH riseby only 0.2–0.3 units, suggesting a large balancing protoninflux across the plasma membrane. Balance of electric chargeis partially maintained by net efflux of K+ and net influx ofCl. Calculation of vacuolar concentrations of imidazole(from (14C] imidazole uptake, assuming that there is no metabolism)plus K+ and Na+ indicates an excess of cations over inorganicanions (Cl). However, although the osmotic potentialof the cells increases, also indicating increased solute concentrations,the increase is less than that predicted by the calculated ionicconcentrations. This discrepancy remains to be resolved. Becausethe osmotic potential also increases when imidazole is absorbedfrom Cl-free solutions it is likely that maintenanceof charge-balance can also involve synthesis and vacuolar storageof organic or amino acids. When young plants of Macroptilium atropurpureum, cv. Siratrowere deprived of external sulphate (-S plants) growth of shootsand roots continued at rates comparable to those in plants wellsupplied with sulphate (control) for 3 d and 5 d respectively.Dilution of internal sulphur therefore took place and redistributionof sulphur occurred between inorganic and organic forms andbetween roots and younger leaves. Even when S-deficiency limitedgrowth, plants contained 16% of their total sulphur as sulphate,but most of this was retained in old leaves and redistributedslowly to growing zones. The capacity for sulphate uptake increased in roots of –Splants very soon after they were deprived of external sulphate;within 24 h the absorption from 0.25 mol m–3 SO42–was more than five times that of control roots. Maximum increasedcapacity was reached after 2–3 d stress when the Vmaxof system 1 was 1948 nmol h–1g–1root fr. wt. in–S plants and 337 nmol h–1g–1root fr. wt.in controls. The Kmfor system 1 did not change significantlywith S-stress being between 5–8 µM in both setsof plants. Absorption of L-cysteine was not stimulated by S-stress. There was a close, positive relationship between plant growthrate and the rate at which sulphate uptake capacity was enhancedby withholding sulphate from culture solutions. When –S plants were replaced in sulphate-containing solutiontheir capacity for SO42– declined to the control levelwithin 24 h. Very marked repression of capacity was also foundwhen –S plants were treated with L-cysteine, but therewas no immediate effect with methionine. Roots of this species appear to have a very active system fordegrading L-cysteine to sulphate, 30% of the label in 35S-cysteineabsorbed by roots was recovered in 35SO42– after 20 minor 2 h incubation. By contrast, roots had a very weak abilityto reduce sulphate. When part of the root system was in solution lacking sulphatethere was enhanced uptake of sulphate by other parts which themselveswere amply supplied with sulphate. This is seen as an exampleof compensatory absorption. The response to S-stress is specific and there were no positiveinteractions between S-stress and the absorption of phosphate,or P-stress and the uptake of sulphate. The results are discussed in relation to the close control ofsulphate uptake by internal sulphate concentration, redistributionof forms of sulphur during stress and mobility of sulphate inthe phloem. Phosphite (Phi, H(2)PO(3)(-)), being the active part of several fungicides, has been shown to influence not only the fungal metabolism but also the development of phosphate-deficient plants. However, the mechanism of phosphite effects on plants is still widely unknown. In this paper we analysed uptake, subcellular distribution and metabolic effects of Phi in tobacco BY-2 cells using in vivo(31)P nuclear magnetic resonance ((31)P-NMR) spectroscopy. Based on the kinetic properties of the phosphate transport system of tobacco BY-2 cells, it was demonstrated that phosphite inhibited phosphate uptake in a competitive manner. To directly follow the fate of phosphate and phosphite in cytoplasmic and vacuolar pools of tobacco cells, we took advantage of the pH-sensitive chemical shift of the Phi anion. The NMR studies showed a distinct cytoplasmic accumulation of Phi in Pi-deprived cells, whereas Pi resupply resulted in a rapid efflux of Phi. Pi-preloaded cells shifted Phi directly into vacuoles. These studies allowed for the first time to follow Phi flux processes in an in vivo setting in plants. On the other hand, the external Pi nutrition status and the metabolic state of the cells had a strong influence on the intracellular compartmentalization of xenobiotic Phi.  相似文献

The effects on leaf age on K (86Rb) efflux, influx and net fluxinto lamina slices from leaf 7 on a tomato plant (Lycopersiconesculentum Mill.) were determined. The ontogenetic trend inK efflux was dependent on the external K concentration. At externalKCI concentrations between 0.5 and 10.0 mM, K efflux rates increasedduring leaf elongation. Only a small increase in efflux occurredin mature leaves with increasing age. It is suggested that thetonoplast retains its structural integrity through the initialstages of leaf senescence. In fully expanded leaves, a zeronet K flux (a balance between influx and efflux) was achievedat external KCI concentrations between 1.0 and 3.5 mM. The Kcontent of lamina slices from leaves 5 and 13 remained constantwhen bathed in a solution containing 2 to 3 mM K. It is suggestedthat the decline in K concentration in mature tomato leaf tissueis due to a decline in leaf free space K concentrations below1 to 3 mM which would result in a net efflux out of leaf cells.
