After the exposure to air, the crystalline nitrogenase MoFe protein from Azotobacter vinelandii was resulted in the remarkable increase in its absorption (ABS) and the significant decrease in its activity and circular dichroism (CD). However, when the aerated MoFe protein was incubated with the reconstituting solution which consisted of Na2MoO4, ferric citrate, Na2S and dithiothreitol, the ABS and CD of the aerated. MoFe protein both were completely restored, simultaneously with the significant restoration of acetylene reduction. It is shown that the P-cluster and other parts related to the protein activity which was damaged by O2 are able to be repaired to a certain extent by the reconstituting solution. 相似文献

测定了自马槟榔(Capparis masaikai Levl.)种子分离的二种甜味蛋白MaⅠ和MaⅡ的远紫外区域圆二色谱,并按Yang和Chen的方法用最小二乘法计算了它们的构象单元含量,结果表明α螺旋含量最多:对MaⅠf_H=0.43、f_β=0.24、f_R=0.33;对MaⅡ,f_H=0.37、f_β=0.33、f_R=0.30;其相关系数均为0.9876;计算所得理论曲线与实验曲钱其本吻合。二种甜蛋白的激光拉曼光谱测定结果也表明其主要构象单元为α螺旋,此外,均无SH谱带:MaⅠ的Tyr残基暴露于分子表面;与MaⅡ相比,MaⅠ有明显的545cm~(-1)和1101cm~(-1)谱带,这可能从构象上说明MaⅠ与MaⅡ对热和变性剂处理表现不同的原因。 相似文献

It has often been assumed that the role of aromatic side chains in the far-ultraviolet region of protein circular dichroism (CD) is negligible. However, some proteins have positive CD bands in the 220–230 nm region which are almost certainly due to aromatic side chains. The contributions to the CD of interactions between tryptophan side chains and the nearest neighbor peptide groups have been studied, focusing on the indole Bb transition which occurs near 220 nm. Calculations on idealized peptide conformations show that the CD depends strongly on both backbone and side-chain conformation. Because of the low symmetry of indole, rotation about the CC bond (dihedral angle 2) by 180° generally leads to large changes in the CD, often causing the Bb band to reverse sign. When side-chain conformational preferences are taken into account, there is no strong bias for either positive or negative Bb rotational strengths. The observation that simple tryptophan derivatives such as N-acetyl-L-tryptophan methylamide have positive CD near 220 nm implies either that these derivatives prefer the R region over the region, or that there is little preference for 2 < 180° over 2 > 180°. Nearest-neighbor-only calculations on individual tryptophans in 15 globular proteins also reveal a small bias toward positive Bb bands. Rotational strengths of the Bb transition for some conformations can be as large as 1.0 Debye-Bohr magnetons in magnitude, corresponding to maximum molar ellipticities greater than 105 degcm2/dmol. Although a substantial amount of cancellation occurs in most of the examples considered here, such CD contributions could be significant, especially in proteins of low helix content. 相似文献

Circular dichroism (CD) is an important spectroscopic technique for monitoring chirality and biological macromolecule conformation. However, during a CD measurement, absorbance, light scattering/turbidity, and fluorescence can also be detected. The simultaneous measurement of these different spectral features for a single sample is the basis of a multi-mode optical spectrometer. This allows time-efficient gathering of complementary information and provides a scheme to ensure that CD measurements are reliable. Aspects of circular polarization differential light scattering, pH, and temperature variation of a protein (antibody) solution are described. Chitosan, a naturally occurring cationic polyelectrolyte, restores the adsorption of the clinical lung surfactant Survanta to the air-water interface in the presence of albumin at much lower concentrations than uncharged polymers such as polyethylene glycol. This is consistent with the positively charged chitosan forming ion pairs with negative charges on the albumin and lung surfactant particles, reducing the net charge in the double-layer, and decreasing the electrostatic energy barrier to adsorption to the air-water interface. However, chitosan, like other polyelectrolytes, cannot perfectly match the charge distribution on the surfactant, which leads to patches of positive and negative charge at net neutrality. Increasing the chitosan concentration further leads to a reduction in the rate of surfactant adsorption consistent with an over-compensation of the negative charge on the surfactant and albumin surfaces, which creates a new repulsive electrostatic potential between the now cationic surfaces. This charge neutralization followed by charge inversion explains the window of polyelectrolyte concentration that enhances surfactant adsorption; the same physical mechanism is observed in flocculation and re-stabilization of anionic colloids by chitosan and in alternate layer deposition of anionic and cationic polyelectrolytes on charged colloids. 相似文献

Bacteriorhodopsin has been reconstituted into lipid vesicles with dipalmitoyl and dimyristoyls phosphatidylcholine. Circular dichroism (CD) measurements show that the proteins are in a monomeric state above the main lipid phase transition temperature (Tc), 41 and 23°C for dipalmitoyl and dimyristoyl phosphatidylcholine, respectively. Below Tc, the CD spectrum is the same as that found for the purple membrane. The latter result implies that the orientation of the chromophore at these temperatures is most likely the same as in the purple membrane ( from the normal to the membrane plane).Transient dichroism measurements show that below Tc the proteins are immobile, while above this temperature protein rotation around an axis normal to the plane of the membrane is occurring. In addition, from the data the angle of the chromophore for the rotating proteins with respect to the rotational diffusion axis can be calculated. This angle is found to be and in dimyristoyl phosphatidylcholine and dipalmitoyl phosphatidylcholine, respectively. This is considerably smaller than the value of for the natural biomembrane. A reversible reorientation of the chromophore above and below the respective main Tc transition temperature could explain the change of angle observed provided that all the molecules rotate above Tc. 相似文献

Summary In order to analyze the evolutionary behavior of the cobra venom cytotoxins, their probable tertiary structure was predicted using computer graphics. The 41 amino acid sequences known show that the major evolutionary changes have taken place in two particularly exposed areas of the molecular surface. In each area, neighboring residue positions seem to have evolved interdependently, but there is no obvious interdependence between the two areas. Indeed, the relative evolution of these two areas prompts a subdivision of the sequence set into four groups. According to the known cytotoxin circular dichroism spectra, one of these four groups could be characterized by a difference in molecular secondary structure. Sine the two variable areas have functional associations, it is suggested that their evolution may be governed by a target with several similar binding sites. 相似文献

Autotransporters are the most common virulence factors secreted from Gram-negative pathogens. Until recently, autotransporter folding and outer membrane translocation were thought to be self-mediated events that did not require accessory factors. Here, we report that two variants of the autotransporter plasmid-encoded toxin are secreted by a lab strain of Escherichia coli. Biophysical analysis and cell-based toxicity assays demonstrated that only one of the two variants was in a folded, active conformation. The misfolded variant was not produced by a pathogenic strain of enteroaggregative E. coli and did not result from protein overproduction in the lab strain of E. coli. Our data suggest a host-specific factor is required for efficient folding of plasmid-encoded toxin. 相似文献
