Crossfostering was performed using lines selected for increased 6-week body weight (H6) and increased 3-to 6-week postweaning gain (M16) and their reciprocal F1 crosses as nurse dams in the selected crossfostering group, and base population controls (C2, ICR) and their reciprocal F1 crosses in the control group. The offspring suckled were H6, M16 and F2 crosses in the selected group, and C2, ICR and their F2 crosses in the control group. Measurements taken on the individual offspring were body weights at birth (WB) and at 12, 21, 31, 42, and 63 days (W12, W21, W31, W42 and W63, respectively) and weight gains between adjacent ages (GB-12, G12–21, G21–31, G31–42 and G42–63, respectively). Least squares constants fitted to populations of genetic and nurse dams were used to calculate specific linear contrasts. Correlated responses to selection in average direct genetic effects were significant and positive for all traits examined in both H6 and M16, while the correlated responses in average maternal genetic effects were negative in M16 and negligible in H6. Selection response was primarily due to average direct genetic effects while the contribution of average maternal genetic effects was of secondary importance. The response in average direct genetic effects was smaller in M16 than in H6 through weaning (WB, W12 and W21), but was larger in M16 for postweaning weights (W31, W42 and W63). The correlated responses in average maternal genetic effects were consistently smaller in M16 than in H6. Direct heterosis was significant for all traits except for G12–21 and G42–63 in the control group, whereas maternal heterosis was significant for weight gains at early ages and for body weights. Direct heterosis tended to be larger than maternal heterosis in both selected and control crosses. Percent direct heterosis for body weight was larger in the selected crosses relative to the control crosses through 31 days of age, but the trend was reversed by 63 days. Summary Correlated responses to selection for increased growth rate were compared in two mouse populations (M16 and H6) of distinct genetic origin. Traits studied were body composition, feed intake, constituent gains and energetic efficiency. When compared with their respective controls (ICR and C2) at 6 and 9 weeks of age, body weight increased more in M16 (57%and 69 % of the control mean) than in H6 (40 % and 34%). The M16 showed correlated responses in fat percent of 2.6% (P <.05), 8.4% (P <.01) and 11.2% (P <.01) at 3, 6 and 9 weeks, respectively, whereas corresponding values in H6 were –2.4% (P <.05), 3.3% (P <.05) and 2.09 % (P >.05). The correlated responses in fat percent were 2.7 and 4.7 times higher in M16 than H6 at 6 and 9 weeks. The regression of ln fat weight on ln empty body weight was larger in M16 (P <.05) compared to ICR and larger (P <.01) in H6 compared to C2. Both M16 and H8 exhibited positive correlated responses from 3 to 6 weeks of age in feed intake and gain and efficiency in fat, protein, calories and ash; fat and caloric gain and efficiency exhibited higher correlated responses in M16 than H6. During the 6- to 9-week interval, the M16 population continued to evince positive correlated responses in gains and efficiencies of fat, protein and calories, whereas H6 did not. Several possible explanations are presented to account for the differences in correlated responses between the selected populations. Partitioning of correlated response differences between M16 and H6 into average direct and average maternal genetic effects indicated that average direct genetic effects, favoring M16, were responsible for the major difference between the selected populations. A modified crossfostering technique was developed to compare the performance of nurse dams in selected and control populations of mice. The H6 and M16 populations were selected for increased 6-week body weight and 3- to 6-week postweaning gain, respectively, while the C2 and ICR populations were the respective controls. Crossfostering was performed using H6, M16 and their reciprocal F1 crosses as nurse dams in the selected crossfostering group and C2 ICR and their reciprocals in the control group. Measurements recorded for nurse dams included mean body weight of 8 young within a nursed litter at birth (MWB) and 12 days of age (MW12). The latter was used as a measure of postnatal maternal performance. Other traits recorded for nurse dams were number born (NB), body weight at parturition (DWP) and 12 days postpartum (DW12), and weight gain (DWG), feed intake (FED) and efficiency (EFF = DWG/FED) for the first 12 days of lactation. The correlated response in MW12 was negative (P less than .01) for M16 and essentially zero for H6. Both lines exhibited positive (P less than .01) correlated responses in DWP and DW12 and no change in EFF. Only the H6 line increases significantly in DWG and FED as a result of selection. NB increased in M16 and H6, but was significant for the latter population only. Population differences in selection response [(M16-ICR)-(H6-C2)] were significant for FED only, primarily due to average direct genetic effects. Direct comparisons of M16 and H6 indicated that M16 was larger in DWP and DW12 but smaller in DWG and EFF. Average direct genetic effects favored M16 for NB, DWP, and DW12, whereas average maternal genetic effects favored H6 for NB, DWP, DW12 and FED. Percent direct heterosis, in F1 crosses of selected populations was significant for MW12 (13.7%) ,FED (10.8%) and NB (11.4%). Direct heterosis in F1 crosses of the controls was significant for MW12 (9.4%), NB (6.6%), DWP (3.5%), DW12 (3.3%) and FED (4.4%). The effects of MW12, DWG and metabolic body size (MBS) accounted for 47% of the variation in FED, pooled within populations. Of these variables, MW12 accounted for the highest proportion (32%) of variation in total feed intake.  相似文献

Summary A total of 2,457 lifetime performance records of 29 genetic groups of mice was analyzed using multiple regression of records on the proportion of gene contribution from 6 lines (designated as Lines MP, mQ, WP, wQ, CP and cQ). Genetic effects were partitioned into line additive, line maternal, direct heterosis, maternal heterosis and paternal heterosis effects. The line additive and line maternal effects were expressed as deviations from Line cQ. Seventeen of 25 line additive effects differed significantly (P<0.05) from Line cQ whereas only 4 of 25 line maternal effects deviated significantly from Line cQ. Deviations in line additive effects from cQ were negative in all lines examined whereas deviations in line maternal effects from cQ were all positive, indicating a negative relationship between line additive and line maternal effects. Direct heterosis effects were all positive and significant (P < 0.01) except in the MPxWP cross which was produced by mating Lines MP and WP of the same base population (P). Maternal heterosis effects were significant in 10 of 20 cases whereas paternal heterosis effects were significant in 13 of 20 cases. Although direct heterosis is a major component of total heterosis effects (sum of direct, maternal and paternal heterosis), the results suggest that parental heterosis may need to be considered in producing multiple way crosses. The fitting of line additive, line maternal, direct heterosis, maternal heterosis and paternal heterosis effects in the multiple regression model effectively accounted for all genetic effects in lifetime performance.Animal Research Centre Contribution No. 1326  相似文献

Conditions were standardized for the isolation and culture of protoplasts from an embryogenic cell suspension culture of Picea glauca. A combination of 0.5% Cellulase R-10, 0.25% Macerozyme, 0.25% Driselase, 0.25% Rhozyme HP-150 with 0.5M mannitol and 5 mM CaCl2.2H2O produced an average of 4.5 × 106 protoplasts per gram fresh weight of cells. Of the several protoplast culture media tested, von Arnold and Eriksson and Kao and Michayluk (KM8P) media best supported mitotic divisions of protoplasts. A density of 105 protoplasts per ml and the addition of 5 mM glutamine to the culture medium was necessary to induce sustained divisions and microcallus formation. Inbreeding depression is a major driver of mating system evolution and has critical implications for population viability. Theoretical and empirical attention has been paid to predicting how inbreeding depression varies with population size. Lower inbreeding depression is predicted in small populations at equilibrium, primarily due to higher inbreeding rates facilitating purging and/or fixation of deleterious alleles (drift load), but predictions at demographic and genetic disequilibrium are less clear. In this study, we experimentally evaluate how lifetime inbreeding depression and drift load, estimated by heterosis, vary with census (Nc) and effective (estimated as genetic diversity, He) population size across six populations of the biennial Sabatia angularis as well as present novel models of inbreeding depression and heterosis under varying demographic scenarios at disequilibrium (fragmentation, bottlenecks, disturbances). Our experimental study reveals high average inbreeding depression and heterosis across populations. Across our small sample, heterosis declined with He, as predicted, whereas inbreeding depression did not vary with He and actually decreased with Nc. Our theoretical results demonstrate that inbreeding depression and heterosis levels can vary widely across populations at disequilibrium despite similar He and highlight that joint demographic and genetic dynamics are key to predicting patterns of genetic load in nonequilibrium systems.  相似文献

Line crossfostering techniques were used to study differences among selected and control lines of mice in direct genetic and postnatal maternal genetic influences on preweaning (day 12) body weight and composition. The lines were selected for high (H6) and low (L6) 6-week body weight and the control line (C2) was maintained by random selection. There were positive correlated responses to selection in both direct genetic and postnatal maternal genetic effects on body weight and weights of all body components (P less than 0.01) except for water and ash weight in H6. The correlated responses in postnatal maternal genetic effects were of the same order of magnitude as those in direct genetic effects. Correlated responses were greater in L6 than in H6. Correlated responses in direct genetic effects were positive (P less than 0.01) for water percent in H6 and ether extract percent in L6, and negative (P less than 0.01) for water percent and lean percent in L6. Correlated responses in postnatal maternal genetic effects were positive for ether extract percent and negative for water percent (P less than 0.01). Correlated responses were far greater in L6 than in H6 and were greater for postnatal maternal genetic effects than for direct genetic effects. Analyses of covariance results indicated line differences in the relative growth rates of the body components.  相似文献

Because most interspecific Medicago embryos abort before they can be excised and cultured, our objective was to grow young pods in vitro. Various media were used to grow three-day-old pods of annuals [diploids, M. blancheana Boiss., M. disciformis DC., tetraploid M. scutellata (L.) Mill.] and perennials (diploid M. falcata L., tetraploid M. sativa L.).Few pods of perennial species grew to maturity on media containing modified Hoagland's plus 1% glucose or sucrose with or without 5% potato extract. Increasing sucrose to 6% increased the percentage of M. sativa pods that produced mature seeds. On DM (differentiation medium), the best medium, the percentage of pods producing viable seeds was: M. blancheana (82), M. disciformis (81), M. scutellata (48), M. sativa (63), M. falcata (15). Electrophoresis of the proteins of the 50S chloroplast ribosome subunit reveals that there are at least two detectable differencesbetween Nicotiana tabacum and N. glauca. This research was designed to study the effects of sorghum maturity on abundance and damage potential of Banks grass mite,Oligonychus pratensis (Banks). More mites per plant were recorded on the latest maturing line (M100) and the least on the early maturing lines (60M and CK60). Mite densities on B407 (late maturity) did not differ from any of the lines tested. Mite densities were positively correlated with leaf area. In contrast, mite feeding damage was significantly lower on the late maturing lines, demonstrating that sorghum susceptibility to mite feeding increases as plants reach anthesis and caryopsis.  相似文献

Objective: To characterize the phenotypic consequences of long‐term selective breeding for rapid weight gain, with an emphasis on obesity and obesity‐induced diabetes (diabesity). Research Methods and Procedures: M16 is the result of long‐term selection for 3‐ to 6‐week weight gain from an ICR base population. Experiment 1 characterized males from both lines for body weights (3, 6, and 8 weeks), feed (4 to 8 weeks) and H2O (6 to 8 weeks) consumption, and heat loss, body composition, and levels of several plasma proteins at 8 weeks of age. Experiment 2 characterized differences between lines for both sexes at three ages (6, 8, and 16 weeks) and fed two diets (high and normal fat). Body weight, composition, blood glucose, and plasma insulin and leptin levels were evaluated after an 8‐hour fast. Results: At all ages measured, M16 mice were heavier, fatter, hyperphagic, hyperinsulinemic, and hyperleptinemic relative to ICR. M16 males and females were hyperglycemic relative to ICR, with 56% and 22% higher fasted blood glucose levels at 8 weeks of age. Discussion: M16 mice represent an outbred animal model to facilitate gene discovery and pathway regulation controlling early onset polygenic obesity and type 2 diabetic phenotypes. Phenotypes prevalent in the M16 model, with obesity and diabesity exhibited at a young age, closely mirror current trends in human populations.  相似文献

Tissue culture responses to three levels of NaCl (0, 85mM and 170 mM) were evaluated in several Medicago species including: M. dzhawakhetica, M. marina, M. rhodopea, M. rupestris, M. sativa (alfalfa) and M. suffruticosa. The whole plant responses of the same genotypes were evaluated in half-strength Hoagland's solution containing 0, 51.5, and 103 mM NaCl. One or more genotypes of M. dzhawakhetica, M. rhodopea, M. rupestris, and M. sativa exhibited in vitro NaCl tolerance at 85 mM. In addition, one genotype each of M. dzhawakhetica, M. rhodopea, and M. sativa was tolerant of 170 mM NaCl. However, all of the genotypes that demonstrated NaCl tolerance in vitro were NaCl sensitive at the whole plant level. Conversely, M. marina the only species exhibiting whole plant NaCl tolerance, had the most NaCl sensitive genotypes at the in vitro level. Although an in vitro NaCl tolerance mechanism which confers whole plant NaCl tolerance was not observed, a potential NaCl tolerance germplasm source, M. marina, was identified.  相似文献

The effects of CaCO3, Zn sources and levels on the yield of submerged paddy and uptake of Zn, P and N to paddy were studied in green-house at Haryana Agricultural University, Hissar. Powdered CaCO3 was mixed at 0,4 and 8 per cent and Zn was added at 0,5 and 10 ppm through ZnSO4.7H2O, ZnO and Zn EDTA separately. Dry weight at tillering and heading and grain and straw at maturity decreased significantly with 4 and 8 per cent CaCO3 in comparison to the control. Increasing Zn application increased the dry weight and grain yield. Zn EDTA gave highest yield of paddy followed by ZnSO4.7H2O and ZnO.Increasing the application of CaCO3 from 0–8 per cent decreased the concentration and uptake of Zn and increasing Zn application from 0–10 ppm increased concentration and uptake of Zn in paddy at tillering, heading and maturity. Zn EDTA gave the highest concentration and uptake of Zn followed by ZnSO4.7H2O and ZnO. There was interaction between Zn sources and CaCO3.The concentration and uptake of N and P in paddy dry matter at tillering and heading and straw and grain at maturity decreased as compared to control with increasing CaCO3 addition. The concentration and uptake of N increased and that of P decreased in paddy dry matter straw and grain with increasing Zn application. The highest concentration of N was observed with ZnO, followed by ZnSO4.7H2O and Zn EDTA. But highest uptake of N was observed with Zn EDTA followed by ZnSO4.7H2O and ZnO. As regards concentration and uptake of P, it was highest with ZnO followed by ZnSO4.7H2O and Zn EDTA.  相似文献

Summary Two populations of randombred of different origin (P and Q) containing eight lines (MP, WP, BP, CP, MQ, WQ, BQ and CQ) were used to evaluate the growth, feed efficiency and lifetime performance of females from eight pure lines and 16 F1 crosses. Line comparisons within populations (P or Q) revealed that the heaviest line at days 21, 42 and 63 was W, followed by lines B, M and C in both populations, while the highest in feed efficiency between days 21 and 63 was line W, followed by lines B, M and C in population P, and was line B followed by lines W, M and C in population Q. Generally, average body weights and feed efficiencies of crosses within and between populations were similar to those of mid-parents. Selection produced line W superior to the line M in additive direct genetic effects on body weight and feed efficiency in each population, and line WP superior to line WQ in additive maternal genetic effects on body weights at days 21, 42 and 63. In lifetime performance tests, total 20-day weight of litters produced by a dam during 200 days averaged from 442.7 g (WP) to 739.1 g (MP) for the eight lines. Lines M and W of populations P and Q generally did not differ in additive direct and maternal genetic effects on lifetime performance. Crosses excelled lines in the number of litters raised to weaning (5.44 vs. 5.25) and total 20-day litter weight per dam during 200 days (648.5 vs. 589.3 g). For lifetime 20-day litter weight per group, crosses from unselected lines (C) exceeded crosses from lines selected for nursing ability (M), adult weight (W) and both traits (B). Crosses of lines from different populations showed a higher heterosis in lifetime performance than crosses of lines within populations. Heterosis in the number of litters raised to weaning, and total 20-day litter weight per dam was significant in crosses between lines CP and CQ, between lines WP and wQ, and between lines WP and mQ. The effect of prostaglandin E1 (PGE1) on rat anterior pituitary cyclic AMP accumulation and luteinizing hormone (LH) release was studied both in vivo and in vitro. Addition of PGE1 to incubation medium over a concentration range of 10-6 to 10-4 M produced a graded increase in pituitary cyclic AMP. At the lowest concentration (10-6 M) there was no significant increase in LH release, but proportional increments in LH release were seen with increasing concentrations of PGE1.Ten minutes after intravenous administration of 5 μg of PGE1 to adult male rats, pituitary cyclic AMP was substantially increased while serum LH levels were not changed. Administration of a higher dose of PGE1 (20 μg) produced a greater increase in pituitary cyclic AMP; and, at this dose serum LH was significantly increased. These results suggest that the PGE1 effect on LH release is mediated by the adenyl cyclase — cyclic AMP system.  相似文献

Summary Thermophilic degradation of sugar beet pulp was studied in batch cultures at 55°C by different associations of bacteria, includingClostridium thermocellum,Methanobacterium sp. andMethanosarcina MP.C. thermocellum produced acetate, succinate, methanol, ethanol, H2 and CO2. The coculture ofC. thermocellum andMethanobacterium sp. produced trace amounts of ethanol and succinate; acetate concentration was about three times higher than in theC. thermocellum monoculture. Summary Correlated responses to selection for increased 3–6 week postweaning gain in male mice were estimated for seven internal organs (testes, spleen, liver, kidneys, heart, small intestine (S intest) and stomach) weighed at specific degrees of maturity in body weight (37.5, 50.0, 62.5, 75.0, 87.5 and 100%). Correlated responses in organ weights were generally large, but the magnitude and direction of response depended upon whether 1) comparisons were made at the same age, degree of maturity or body weight and 2) absolute or proportional organ weights were used. The selected line (M16) weighed more and had larger organ weights than controls (ICR) when compared at either the same degree of maturity or the same age, indicating positive genetic correlations between body weight and the respective organ weights. Positive correlated responses were found in spleen weight/body weight at all degrees of maturity and in liver and S intest weights as a proportion of body weight at some degrees of maturity. Testes, kidneys, heart and stomach weights as a proportion of body weight had negative correlated responses, though this was consistent only for kidneys across all degrees of maturity. Correlated responses in organ weights adjusted for body weight by covariance analysis were positive for spleen, S intest and stomach and negative for testes and kidneys. Based on the constrained quadratic model, degree of maturity in organ weight relative to degree of maturity in body weight responded positively for testes, kidneys and S intest and negatively for spleen and liver. Summary The effect of the postnatal maternal environment, simulated by rearing mice in litters of three, six or nine, on body weight and body composition was investigated in three lines of mice differing widely in growth rate. The lines were selected for high (H6) and low (L6) 6-week body weight while the control line was maintained by random selection. Body weight and weights and percentages of ether extract, water, ash and protein at 21, 42, 63 and 84 days were recorded. With few exceptions, there were positive correlated responses to selection in body weight and in weights of body components. At 21 and 42 days the correlated responses were larger in L6 mice than in H6 mice. Body weight and weights of body components were larger for mice reared in litters of three than for those reared in litters of nine. Also, mice reared in litters of six were intermediate in body weight and weights of some of the body components between those reared in litters of three and nine. Differences in body weight and weights of body components due to postnatal maternal environment were small by comparison with differences due to genetic line. There were significant line by maternal environment interactions in body weight at 21 days and in ether extract weight at 21 and 63 days. Line and maternal environment differences in percentages of body components did not follow any consistent trend. The results for percentages of body components were further complicated by line x maternal environment interactions. The host-specific toxin from Helminthosporiumcarbonum race 1 was purified from culture filtrates by solvent extraction, gel filtration, and high pressure liquid chromatography. High resolution mass spectrometry of the purified toxin gave a MW of 436.2318 and an elemental composition C21H32N4O6. Amino acid analysis and proton and13C-NMR indicated a peptide containing four amino acids. Their sequence was determined by gas chromatography mass spectrometry. Finally, digestion of the amino acids with D- and L-amino acid oxidases gave the complete structure cyclo[(L-2-amino-9, 10-epoxy-8-oxodecanoyl)-D-prolyl-L-alanyl-L-alanyl].  相似文献

The effects of quinine on the peak amplitude and the decay of calcium currents (ICa) were investigated in nonidentified neurons isolated fromHelix pomatia. A concentration of 1×10–5–5×10–4 M quinine was found to produce a reversible dose-dependent deceleration in the decline of ICa ("lead" effect) and a reversible, slowly evolving dose-dependent reduction in ICa amplitude ("lag" effect). A reduction in amplitude down to half control level is observed at a quinine concentration of 6 ×10–5 M, while the current-voltage relationship of ICa shifts by 5–10 mV towards negative potentials.
