Pythium oligandrum was recovered, identified and quantified from air‐dried soil plated on 1.5% water agar containing 0.1% glucose. Isolations of P. oligandrum over 2 years from soils treated with single applications of metalaxyl plus mancozeb were consistently lower than those from untreated soil from the same fields. In three fields in the first year P. oligandrum was reduced from a range of 43.3–115.0 to 17.0–43.2 isolates g‐1 soil. In the second year, results from 11 fields showed reductions from 27.8–141.8 to 2.8–44.5 isolates g‐1 soil. P. oligandrum was sensitive to both metalaxyl and mancozeb, with median effective dose (ED50) values of 0.13 ± 0.02 μg m‐1 and 3.33 ± 0.12 μg ml‐1, respectively. In a pot test with three soils treated with metalaxyl, mancozeb or the combination of fungicides, levels of P. oligandrum declined over 3 months, with effects first recorded 2 weeks after treatment. Levels of P. oligandrum were reducd by differing degrees in the three soils. Isolate counts from untreated soils declined from a mean of 58.0 g‐1 soil at the start of the experiment to 27.5 g‐1 after 3 months, whereas fungicide treatments caused further reductions to 11.1 (metalaxyl), 9.7 (mancozeb) and 4.8 isolates g‐1 (metalaxyl plus mancozeb).  相似文献

A laboratory test was developed to assess the sensitivity of field populations of Phytophthora infestans to metalaxyl. Discs of potato leaf tissue were floated upon solutions of the fungicide at different concentrations and inoculated with spores. The extent of symptom development was noted after incubation under standard conditions for 5–6 days. In preliminary experiments growth of isolates of P. infestans obtained from culture collections was severely inhibited in discs treated at 2 μg/ml. By contrast the development of an isolate obtained from a crop in Eire in which blight control with metalaxyl had failed, and known to be markedly less sensitive in vitro, was unaffected in discs treated at 100 μg/ml. During the summer of 1980, 234 samples of P. infestans were obtained from 20 sites in south-west England, 10 of which had received sprays containing metalaxyl and 10 of which had not. All samples were sensitive to metalaxyl applied at 2 μg/ml. In 1981, 35 sites within the same area, 30 of which had received sprays containing either metalaxyl or ofurace (a related fungicide), were similarly surveyed. Most of the 79 samples of P. infestans examined proved sensitive and at all sites the amount of blight was small. However, at three sites, including one not treated with acylalanine fungicides, strains were found which were unaffected by 100 μg/ml metalaxyl in leaf disc tests. These findings are discussed in relation to the development of resistant blight in other areas and to the use of fungicide mixtures.  相似文献

Over a 3-yr period 261 isolates of 17 species of Pythium were tested for sensitivity to metalaxyl at concentrations of 5, 50 or 100 μ/ml. A wide range of responses was observed, from isolates where growth ceased at 5 μg/ml to those where growth at 100 μg/ml was similar to that of the untreated controls. In further tests isolates of 11 different species had ED50's < 1 μg/ml. A lower sensitivity was detected in isolates of six Pythium spp. where values in the range 1–10 μg/ml were obtained. This lower sensitivity was not related to previous known use of metalaxyl. Three isolates of Pythium dissotocum from sites where the fungicide had been used repeatedly had ED50's > 100 μg/ml and were considered resistant. The resistance was stable over a 2-yr period and isolates were cross-resistant to furalaxyl, benalaxyl, ofurace, cyprofuram and oxadixyl. Increasing concentrations of metalaxyl reduced or prevented the production of zoospores by four species of Pythium, although when zoospores were produced, this was followed by the normal processes of encystment and germination. Culturing P. dissotocum on different sub-lethal concentrations of metalaxyl for 18 wk did not induce a high level of resistance to the fungicide.  相似文献

Pepper Phytophthora blight (PPB), caused by Phytophthora capsici, is an important disease of pepper in China. The extensive application of metalaxyl has resulted in widespread resistance to this fungicide in field. This study has evaluated the activities of several fungicides against the mycelial growth and sporangium germination of metalaxyl‐sensitive and metalaxyl‐resistant P. capsici isolates by determination of EC50 values. The results showed that the novel carboxylic acid amide (CAA) fungicide mandipropamid exhibited excellent inhibitory activity against PPB both in vitro and in vivo, with averagely EC50 values of 0.075 and 0.004 μg/ml in mycelial growth and sporangium germination, respectively, and over 88% efficacy in controlling PPB. The other three CAA fungicides also provided over 70% efficacy in controlling PPB. The mycelial growth was less sensitive to quinone outside inhibitor (QoI) fungicides azoxystrobin and trifloxystrobin than that of sporangium germination in P. capsici isolates. However, azoxystrobin and trifloxystrobin provided over 80% efficacy in controlling PPB. It was noted that propamocarb and cymoxanil did not exhibit activity against the mycelial growth or sporangium germination of P. capsici isolates in the in vitro tests, with over 70% efficacy in controlling PPB. The new fungicide mixture 62.5 g/l fluopicolide + 625 g/l propamocarb (trade name infinito, 687.5 g/l suspension concentrate (SC)) produced over 88% efficacy in controlling PPB caused by both metalaxyl‐sensitive and metalaxyl‐resistant isolates. The data of this study also proved that there was obviously no cross‐resistance between metalaxyl and the other tested fungicides. Therefore, these fungicides should be good alternatives to metalaxyl for the control of PPB and management of metalaxyl resistance.  相似文献

The reactions of isolates of Phytophthora cactorum, P. nicotianae and P. × pelgrandis to metalaxyl, mancozeb, dimethomorph, streptomycin and chloramphenicol were tested to obtain information about the variability of resistance in these pathogens. Distinct genetic groups showed significant differences in resistance to all tested substances except streptomycin. In response to streptomycin, the growth inhibition rates of distinct groups did not differ significantly. The most remarkable differences were detected in the reactions to chloramphenicol and metalaxyl. Discriminant analysis evaluating the effect of all substances confirmed the differences among the groups, which are in agreement with the differences revealed by earlier DNA analyses.  相似文献

Leaf blight caused by Phytophthora colocasiae is the most destructive disease affecting taro (Colocasia esculenta) worldwide including India. Fungicides (primarily metalaxyl) remain as an important strategy to manage taro leaf blight in India over decades. It is important to monitor isolate sensitivity to identify build-up of fungicide resistance and thereby modify fungicide usage strategies. P. colocasiae isolates representing four different geographical regions of India were evaluated for their sensitivity to metalaxyl and three other commercially available fungicides viz. Samarth, Biofight and Akoton by poisoned media technique. All the isolates tested were sensitive to metalaxyl, nevertheless there is an increase in the effective concentration compared to the previous reports. Among the other fungicides, Samarth was found to be superior in completely inhibiting mycelial growth at 0.05% followed by Biofight at 1%. Metalaxyl and Akoton® shared a common inhibitory concentration at 2%. The most effective fungicide determined by the in vitro method was evaluated in vivo for studying the pattern of inhibition before and after the disease development in detached taro leaf. The results of the study revealed that build-up on metalaxyl resistance in P. colocasiae is in its course and caution should be taken while administering against taro leaf blight. Fungicide Samarth could be used as an alternative to metalaxyl for management of taro leaf blight.  相似文献

The sensitivity of Alternaria solani isolates to the fungicides mancozeb and chlorothalonil was evaluated, to determine if inadequate disease management by these fungicides could be attributed to reduced sensitivity of A. solani isolates to these fungicides. The sensitivity of 60 isolates of A. solani was assessed using the inhibition of radial mycelial growth (RG) method, using fungicide concentrations of 0, 1.0, 10, 100, 500 and 1000 μg a.i ml?1 medium. EC50 was calculated for each isolate and fungicide combination. The EC50 values of different A. solani isolates to mancozeb ranged from 9.05 to 712.65 μg ml?1. EC50 values of different isolates to chlorothalonil ranged from 4.25 to 849.4 μg ml?1. The percentage of isolates with reduced sensitivity was 46.7 and 53.3% for mancozeb and chlorothalonil, respectively. Results of the in vivo tests demonstrated decline in disease control by the two fungicides with the reduced-sensitivity isolates compared to the sensitive ones.  相似文献

Sorghum downy mildew, caused by the obligate oomycete Peronosclerospora sorghi, has been controlled through the use of resistant cultivars and seed treatment with metalaxyl. A recent outbreak in fields planted with treated seed revealed the presence of a metalaxyl-resistant variant. Here, PCR-based methods including amplification from RAPD primers and two systems of automated AFLP analysis have been used to detect DNA-level genetic variation among 14 isolates including metalaxyl-resistant and susceptible isolates, as well as representatives of common pathotypes 1 and 3 and a new pathotype. In total, 1708 bands were detected after amplification of EcoRI/MseI fragments with 16 primer combinations. Nearly as many amplified products were observed using eight primer pairs with three-base extensions (LI-COR) as with two-base extensions (ABI-Prism genetic capillary system). Approximately 25 % of the bands were polymorphic across the 14 isolates, with the majority of differences specific to the pathotype P1 isolate. The AFLP banding patterns are consistent with metalaxyl resistance and the new pathotype having evolved from pathotype 3.  相似文献

Phytophthora nicotianae causes black shank, one of the most important diseases of tobacco worldwide. Metalaxyl and dimethomorph are two fungicides which have been used widely for control of this disease in Henan province, China. A study was conducted to determine the level of metalaxyl and dimethomorph sensitivity in isolates of P. nicotianae from tobacco in Henan province and mating type structure of the pathogen population. A total of 32 isolates were isolated from 11 cities in Henan province. Sensitivity of all isolates to metalaxyl and dimethomorph was tested in vitro, and mating types of all isolates were determined by pairing known A1 and A2 testers. For metalaxyl, EC50 values of 32 P. nicotianae isolates ranged from 0.08 to 2.82 mg/L. Sixteen isolates were sensitive, and the rest were intermediate to metalaxyl. None were classified as resistant isolates. For dimethomorph, EC50 values of 32 P. nicotianae isolates ranged from 0.07 to 0.59 mg/L. All isolates were sensitive to dimethomorph. Thirty‐one isolates were A2 mating type, and one isolate was A0 mating type. No isolate was identified as A1 mating type. These results suggested that the P. nicotianae population in Henan province has already exhibited intermediate resistance to metalaxyl and was still sensitive to dimethomorph, and asexual reproduction was the major form of reproduction for the P. nicotianae population.  相似文献

Four fungicides were evaluated for their effectiveness against Phytophthora diseases of peach trees and their ability to absorbed and translocated by roots of peach tree in laboratory. Meta‐laxyl suppressed the development of P. cactorum and P. citwphthora on segments. Fosetyl‐Al showed variable fungicidal activity against P. cactorum and P. citwphthora. Both dimetho‐morph and cyraoxanil were not effective to inhibit the growth of fungi on segments. Section of tree trunks in 2‐yr‐old GF 677 trees were painted with one of the test fungicides. Strips of bark were removed 10 and 20 days after painting within treated area and inoculated with P. cactorum and P. citwphthora. Generally, the results agree with them obtained with excised stem and excised twig methods. Exception is the dimethomorph that reduced the development of P. cartorum and P. citrophthora. This study indicated that application with metalaxyl to the peach tree appears to be an effective procedure to control Phytophthora diseases on peach trees.  相似文献

大豆疫霉Phytophthorasojae易对甲霜灵产生抗性,从大豆疫霉野生型菌株可诱变筛选到对甲霜灵有抗性的菌株Mtr。Mtr抗性菌株的抗性水平可达野生型单游动孢子菌株的870倍以上。Mtr性状在无性后代稳定遗传,在游动孢子后代连续三代未发生抗药性分离。大豆疫霉Mtr性状的保持对甲霜灵没有表现依赖性。Mtr单游动孢子菌株在不含甲霜灵的胡萝卜培养基(CA)平板上培养30d后对甲霜灵的抗性没有下降,其单游动孢子后代也未出现抗药性分离。  相似文献

Abstract: The effect of timing of application on the incompatibility of three fungicides (metalaxyl, mancozeb, copper oxide) and isolate MK2001 of the entomopathogenic fungus Beauveria bassiana (Balsamo) Vuillemin was assessed in vitro . Isolate MK 2001 at 1 × 107 conidia/ml concentration was highly pathogenous to adults of Lygus lineolaris (Hemiptera) (84% mortality at day 4 post-treatment). The fungicides at the manufacturer recommended rate (1X) were strongly fungistatic and inhibited the fungal isolate radial growth on Sabouraud dextrose agar (SDA). Mancozeb, metalaxyl and copper oxide exhibited insecticidal activity of 24, 40 and 48% respectively, on L. lineolaris adults. It is clear that application of the fungal isolate MK2001 2–4 days before applying fungicides metalaxyl, mancozeb or copper oxide synergized the insecticidal effect of the isolate. On the contrary, application of metalaxyl, mancozeb, copper oxide 2–4 days before applying isolate MK2001 antagonized the insecticidal effect. The simultaneous use of each fungicide (metalaxyl, mancozeb or copper oxide) and the isolate gave lesser insect mortality. This study revealed that although some fungicides are incompatible for use alongside fungal isolates, the proper evaluation of their time of use could be beneficial in biological control or IPM programmes. Furthermore, the application of B. bassiana isolate MK2001 followed by fungicide application could synergize insecticidal activity.  相似文献

Bacterial antagonist B8 produced an inhibition zone with each of four Phytophthora cactorum isolates on corn meal agar (CMA) plates. Infections with three P. cactorum isolates were significantly reduced when the soil was simultaneously inoculated with B8. Growth of P. cactorum was completely inhibited on CMA amended with 40–100 per cent 10 fold concentrated B8 extract. Percent oospore germination of P. cactorum was significantly reduced when B8 was present in suspension for 9, 12 and 15 days from inoculation. Survival of oospores was significantly reduced at 60 and 90 mm depths in soil. Bacterial antagonist B8 significantly reduced the population of viable P. cactorum oospores in the top 30 mm of soil where oospores generally survive.  相似文献

The effects of temperature and pH on the production of antifungal compounds and growth in sterilized soil of Enterobacter aerogenes and Bacillus subtilis, antagonists of Phytophthora cactorum, the cause of crown rot of apple trees, were studied. With E. aerogenes maximum amounts of antifungal compounds were produced between 14 and 21°C and at pH levels between 3.5 and 5.0. Bacillus subtilis produced maximum amounts of antifungal compounds between 21 and 28°C andwith pH levels between 5.0 and 8.0. P. cactorum inoculum, produced in the presence of E. aerogenes or B. subtilis at optimum temperature and pH levels, was significantly less virulent compared with controls. The optimum temperature of maximum population growth in sterilized soil for E. aerogenes was 18°C and for B. subtilis 25°C. The population growth of B. subtilis was much slower than that of E. aerogenes. Fosetyl Al stimulated the multiplication of bacteria at lower temperatures while metalaxyl had the same effect at higher temperatures. These results indicate that populations of these antagonistic bacteria increase in sterile soil up to the 33rd day from inoculation and that the fungicides fosetyl, Al and metalaxyl did not limit their multiplication and production of antifungal compounds.  相似文献

Of eleven tomato cultivars ‘Baby’ was the most susceptible to Phytophthora infestans. The sporangium concentration of the fungus and the methods of fungicide application had a significant effect on the efficacy of oxadixyl, mancozeb and cymoxanil when tested singly or in mixture against sensitive and phenylamide resistant strains of P. infestans on tomato. Mancozeb was the least effective fungicide, its activity decreased significantly with increasing numbers of sporangia used for inoculation (increasing disease pressure) and its efficacy was dependent on the method of application. Oxadixyl and cymoxanil showed much stronger antifungal activities (except oxadixyl against resistant strains) which did not depend on the number of sporangia or method of fungicide application. The mixture of oxadixyl, mancozeb and cymoxanil was equally active and was independent of application type, sporangium concentration and level of sensitivity of the fungus. Mixtures of the single components showed synergistic interactions up to levels of 20, depending on the activity of the individual fungicide and the different disease intensities. Three-way mixtures containing oxadixyl, mancozeb, and cymoxanil represent therefore promising possibilities in practice for strong disease pressure, even when resistance problems are expected.  相似文献

The effect of nine different pesticides on the growth of yeasts isolated from the leaves of fruit and forest trees was investigated. Four insecticides (with the active ingredients: thiacloprid, deltamethrin, lambdacyhalothrin, and thiamethoxam) and five fungicides (with the effective substances: bitertanol, kresoxim-methyl, mancozeb, trifloxystrobin, and cupric oxychloride) were tested. The concentrations of chemicals were those recommended by the manufacturers for the spraying of trees. The yeast strains isolated from the leaves of fruit trees were not sensitive to any of the insecticides. The majority of yeast strains isolated from the leaves of forest trees were either not sensitive or only to a small extent. While Rhodotorula mucilaginosa and Pichia anomala were not affected by any insecticide, the strains of Cryptococcus laurentii and Rhodotorula glutinis showed the highest sensitivity. The effects of fungicides on the growth of isolated yeasts were more substantial. The fungicide Dithane DG (mancozeb) completely inhibited the growth of all yeasts. All strains isolated from fruit tree leaves were more resistant to the tested fungicides than those isolated from the leaves of forest trees. The most resistant strains from the leaves of fruit trees belonged to the species Metschnikowia pulcherrima, Pichia anomala, and Saccharomyces cerevisiae, whereas Cryptococcus albidus and C. laurentii, originating from the leaves of forest trees, showed the highest sensitivity to fungicides.  相似文献

Abstract

Phytophthora infestans is one of the most destructive pathogens of potato and causal agents of notorious disease late blight. Different chemicals are used to control the pathogen of late blight but the most commonly used is metalaxyl; its extensive use of has caused decreased sensitivity in the P. infestans population. The metalaxyl sensitivity of the Pakistani population of P. infestans is investigated in the present study. For this purpose, 178 isolates of P. infestans were obtained from the lesions of diseased potato leaves and stems, and samples were collected from the different potato-growing areas of Pakistan, where late blight is a problem. Sensitivity of the isolates of P. infestans was investigated by metalaxyl sensitivity test and with the help of test isolates were divided into three categories, i.e. sensitive, intermediate and resistant, based on their Co-efficient of mycelial growth inhibition (CMGI) values. During the study, highest percentage (50.17%) of resistant isolates was observed in the population of Punjab (zone 2), whereas the lower percentage (33.33%) was observed in the population of Swat valley (zone 6b). In the present study, it was discovered that P. infestans late blight-causing fungus has adopted more resistance against metalaxyl because of its wide use.  相似文献

The study was undertaken to identify and characterize Pythium isolates associated with root rot disease of tobacco seedlings as a first step towards developing management strategies for the pathogen. A total of 85 Pythium isolates were collected from diseased tobacco seedlings during 2015–2016 tobacco growing season. The isolates were identified to species level using sequencing of the internal transcribed spacer region. Thereafter, a subset of the isolates was tested for sensitivity to the commonly used fungicides, metalaxyl, azoxystrobin and a combination of fenamidone/propamocarbby growing isolates on Potato Dextrose Agar plates amended with the fungicides. The sequence analysis of the ITS‐rDNA identified Pythium myriotylum as the dominant Pythium species associated with the root rot of tobacco seedlings in Zimbabwe. Pythium aphanidermatum and P. insidiosum were also identified albeit at lower frequencies. Phylogenetic analyses of the ITS region of the P. myriotylum isolates showed little sequence diversity giving rise to one distinct clade. The fungicide sensitivity tests showed that metalaxyl provided the best control of P. myriotylum in vitro, as compared to other fungicides. To the best of our knowledge, this is the first comprehensive study to determine and characterize Pythium species associated with root rot of tobacco in the float seedling production system in Zimbabwe.  相似文献

Phytophthora capsici is an important oomycete pathogen threatening the vegetable production in China, but very little is known about its population structure. The objective of the present study was to evaluate the genetic diversity of 49 P. capsici isolates obtained from 2007 to 2014 at nine provincial locations in China. Isolates were assessed for mating type, metalaxyl resistance and simple sequence repeat (SSR) genotype. Mating‐type analyses of the isolates showed that both mating types were present in all of the sampled production regions, and the mating‐type frequency in the total Chinese population did not deviate significantly from a 1:1 ratio. Responses of isolates to the fungicide metalaxyl indicated the presence of intermediate resistance to metalaxyl among the field population. A universal fluorescent labelling method was adapted in this study to improve the efficiency of SSR genotyping. Microsatellite genotyping of the isolates using seven SSR markers revealed 44 unique multilocus genotypes. Genetic analyses indicated the existence of two genetic clusters within Chinese P. capsici collection. Clonal reproduction may play a more prominent role in Yunnan Province, but non‐existence of repeated genotypes and existence of both mating types throughout all regions suggest outcrossing and sexual recombination likely play an important role in the overall epidemiology in China. Future studies would include expanded scale sampling at single regions over multiple years to better define the genetic diversity of P. capsici in China.  相似文献

Phytophthora spp. associated with leaf fall, stem canker and black stripe of rubbertree (Hevea brasiliensis Muell.-Arg.) in Xishuangbanna of Yunnan Province were isolated from leaves, bark, fruit and soil in the plantations. Of a total of 50 isolates, 42 were designated as P. citrophthora (R. E. Sm. & E. H. Sm.) Leonian, while the others were identified as P. palmivora (Butl.) Butl., P. colocasiae Rac. and P. cactorum (Leb. & Conn) Schroet. This appears to be the first report of P. citrophthora on Hevea brasiliensis.  相似文献
