During the present study, 360 populations of 174 species belonging to 82 genera and 23 families of dicots have been cytologically worked out from different localities of District Kangra (Himachal Pradesh) between 550 and 3,800 m altitude. As many as 10 species including Berberis ceratophylla (2n = 28), Caltha alba (2n = 32), Corydalis meifolia (2n = 16), C. thyrsiflora (2n = 16), Impatiens reidii (2n = 14), Indigofera hamiltonii (2n = 16), Potentilla thomsonii (2n = 14), Sedum trifidum (2n = 36), Stellaria semivestita (2n = 26) and Viola canescens (2n = 12) have been cytologically worked out for the first time at world level. New intraspecific diploid cytotypes have been recorded in 11 species while new intraspecific tetraploid cytotypes are noticed in 15 species. B-chromosomes have also been reported in 3 species viz. Anemone obtusiloba, Clematis grata and Ranunculus diffusus for the first time at world level. The species of the area are active state of evolution, depicting heterogeneity in chromosome numbers involving polyploidy (48 species, 28.18 %) as well as irregular meiotic behavior. The meiotic abnormalities have been witnessed in 69 species (39.65 %) showing different types of irregularities such as cytomixis, interbivalant connections, unoriented bivalents, chromatin stickiness, chromosomal laggards and bridges as well as abnormal microsprogenesis. All these meiotic abnormalities lead to reduced pollen fertility and formation of variable sized pollen grains in most of these species.  相似文献

Silene vulgaris is an important ethnobotanical species, but so far no thorough meiotic analysis has been performed, despite its wide occurrence in temperate and alpine Himalayas, this prompted us to undertake present study. We investigated original meiotic chromosome number and effect of cytomixis on meiotic course, microsporogenesis and pollen grain size and fertility. The paper also summarizes the previously published chromosome counts in the species and cytogeography of 2x and 4x cytotypes in the Indian Himalayas and elsewhere. Meiocytes preparations were made through standard squashed technique in 1 % acetocarmine. Two ploidy levels have been determined during the cytomorphological surveys, diploid (2n = 2x = 24) and tetraploid (2n = 4x = 48). Most of the presently scored populations were found to be diploid (2n = 2x = 24). Morphologically 2x and 4x individuals were indistinguishable from each other. Meiosis was perfectly normal in 4x cytotype. However, some of the 2x plants showed the phenomenon of cytomixis and associated meiotic abnormalities. Consequent to cytomixis, hypo-, hyperploid and enucleated pollen mother cells (PMCs) were resulted. Such PMCs combined with other meiotic abnormalities yielded sterile and variable sized pollen grains. On the basis of overall information, it is clear that 2x cytotype is the most widely distributed cytotype in the Indian Himalayas as well as elsewhere. On the other hand, the 4x cytotype seems to be less frequently present in the Indian Himalayas and outside of India.  相似文献

Abstract

In the present study 16 populations belonging to 13 species of the genus Vernonia Schreb. were examined cytologically. In total, six different chromosome numbers, which represent three basic numbers: x = 10, x = 16 and x = 17, were found. These results include the first chromosome number reports for the following four species: V. lanifera Cristóbal & Dematt. (2n = 2x = 32), V. membranacea Gardner (2n = 2x = 34), V. salzmannii DC. (2n = 2x = 20) and V. scabrifoliata Hieron. (2n = 2x = 128). Besides, a new chromosome number was found in V. saltensis Hieron. (2n = 2x = 32), for which only tetraploid populations have been previously recorded. The data obtained in this work, along with the information available from the literature, show that the genus Vernonia in South America is heterogeneous with basic chromosome numbers that range between x = 9 and x = 19. These numbers suggest that a combination of polyploidy and aneuploidy has played an important role in the evolution of the genus.  相似文献

The present work includes detailed male meiotic studies on 46 species of grasses falling into 59 accessions from different localities of Parvati Valley in Kullu district of Himachal Pradesh in the altitudinal range of 1,100 to 2,750 m. All the species have been studied cytologically for the first time from the study area. The meiotic chromosome count of n = 14 for Calamagrostis emodensis is the first ever chromosome report. Three species, namely Agrostis alba (n = 21), Avena byzantina (n = 21) and Bromus inermis (n = 14) have been studied cytologically for the first time from India. New intraspecific diploid/polyploid cytotypes have been reported for Arthraxon serrulatus (2n = 4x = 32), Iseilema laxum (2n = 12x = 60), Digitaria albudens (2n = 8x = 72), Festuca kashmiriana (2n = 2x = 14) and Stipa orientalis (2n = 2x = 20). The existence of variable number of B-chromosomes (2n = 60 + 0-5B) has been reported for the first time in the 12x cytotype of Iseilema laxum. Secondary associations of chromosomes in the tetraploid cytotype of Cymbopogon martini (n = 20) indicated its secondary polyploid nature. As many as 18 species showed various meiotic anomalies such as the phenomenon of cytomixis involving inter PMC migration of chromatin material, chromatin stickiness, interbivalent connections, abnormal spindle activity, presence of bridges and laggards during anaphases and telophases and abnormal sporads. These meiotic abnormalities consequently yielded sterile and heterogeneous-sized fertile pollen grains. The polyploidy and aneuploidy have played an active role in the evolution of grasses.  相似文献

The present paper deals with cytological studies on 31 populations covering 17 species belonging to 10 genera of Apiaceae from Western Himalayas. The chromosome numbers in the two species as Chaerophyllum capnoides (n = 11) and Heracleum brunonis (n = 11), along with additional cytotypes for Pimpinella acuminata (n = 9) and Sium latijugum (n = 12) have been reported for the first time on world-wide basis. The genus Pleurospermum, although cytologically worked out earlier from outside India, its species densiflorum (n = 11) makes first representation of the genus from India. Besides, the chromosome number in Chaerophyllum aromaticum (n = 11) have been worked out for the first time from India. The course of meiosis varies from normal to abnormal in different populations of Chaerophyllum villosum, Pimpinella achilleifolia and Sium latijugum while abnormal meiotic course has been observed in all the studied populations of Chaerophyllum acuminatum, C. aromaticum, C. capnoides, Pimpinella acuminata, P. diversifolia, Pleurospermum densiflorum and Vicatia coniifolia. Such taxa are marked with meiotic abnormalities in the form of cytomixis, chromatin stickiness, formation of laggards and bridges resulting into abnormal microsporogenesis. The occurrence of structural heterozygosity has been recorded in the Chaerophyllum acuminatum and C. aromaticum. The effect of these abnormalities is clearly seen on the pollen size and fertility.  相似文献

Detailed male meiosis, critical morphological observations and distribution pattern of diploid as well as tetraploid cytotypes of the Western Himalayan species, Bupleurum lanceolatum have been evaluated at present. A diploid (n = 8) cytotype is reported from Kashmir, whereas, both diploid (n = 8) and tetraploid (n = 16) cytotypes are available from two districts Kangra and Sirmaur of Himachal Pradesh. Out of these, the tetraploid cytotype makes new addition for the species on a worldwide basis. As per behavior, a tetraploid cytotype is characterized by abnormal meiosis leading to high pollen sterility and size variation of the pollen grains. Morphologically, tetraploids are noted to be luxuriant in comparison to the diploids.  相似文献

The various normal and abnormal stages of meiosis and pollen mitosis of Drimiopsis botryoides are described, and a comparison between naturally propagated in vivo and tissue culture derived ex vitro plants in respect to their cytological behaviour presented. We also describe the floral morphology and investigate the relationship between the floral developmental stages and the progression of microgametogenesis. In total, 33 bivalents are observed in diakinesis, which indicate the diploid number 2n = 66 and this number is cross-checked by a haploid set of n = 33 chromosomes in pollen mitosis. Only 6.8% and 4.9% meiotic abnormalities were recorded on in vivo and ex vitro plants, respectively, which led to the formation of non-viable pollen. Finally, the microspores have to develop into tri-cellular male gametophyte. Only 0.2% pollen grains are found with a micro-nucleus. Though the higher pollen viability was recorded on both in vivo (89.3 ± 4.1%) and ex vitro (92.1 ± 4.6%) plants, but surprisingly the pollen germination rate is extremely low with 13.6 ± 1.74% and 21.3 ± 1.55%, respectively. The present study obviously enriches the cytological database of D. botryoides and may help future research on androgenesis and genetic improvement.  相似文献

Campuloclinium macrocephalum DC. is a perennial herb widely distributed in the New World and introduced in South Africa, where it is commonly called “pompom weed”. This species is considered one of the most important weeds of Brazil and one of the problematic invasive plants of South Africa. The meiotic system can be studied to assess the ability of a weed to spread, but only few studies on C. macrocephalum have been realized. In this study, we examined the meiotic behavior and pollen fertility of 14 natural populations of C. macrocephalum from Argentina and Uruguay. Meiotic analysis revealed 2 triploid (2n = 3x = 30), 11 tetraploid (2n = 4x = 40) and 1 mixed population (2n = 2x = 20, 2n = 4x = 40). Both, triploid and tetraploid specimens showed a widely variable meiotic behavior with irregular chromosome pairing showing univalents, bivalents, trivalents (in triploids) and tetravalents (in tetraploids) at diacinesis of first meiotic division. Different abnormalities were observed, such as: laggard chromosomes, chromatin bridges, and out of plate chromosomes at metaphase I. During meiosis I (prophase), some cells showed the phenomenon of cytomixis or chromatin transfer between pollen mother cells. The meiotic indexes suggest that only four populations were normally fertile (over 90 % of fertile pollen), indicating meiotically stable plants. The remaining populations share variable pollen fertility, with triploids ranging from 46.64 to 54.83 % and tetraploids varying from 3.54 to 45.30 %. We suggest that polyploidy seems to be recurrent in C. macrocephalum, promoting partial sterility of pollen grains, generating large numbers of individuals by apomixis promoting invasion of crop fields. This study presents the meiotic behavior of this weed, these could be useful for future studies of biological control in areas with no natural enemies.  相似文献

Tordyliopsis brunonis (Apiaceae) is cytologically investigated here for the first time from India. The chromosome count of 2n = 33, ascertained here, represents a new intraspecific triploid cytotype in the species, supplementing the earlier report of a diploid cytotype with 2n = 22 from Nepal Himalayas. The diploid chromosome count (n = 11) has also been found in some of the presently investigated individuals which showed perfectly normal meiosis with 100 % pollen fertility and normal seed set. However, the individuals with triploid chromosome count showed irregular meiotic behaviour and abnormal microsporogenesis resulting in high pollen sterility (56.26 %) and no seed set. The irregular meiotic behaviour in the triploid individuals is attributed to the occurrence of variable number of univalents (1–7) at diakinesis and metaphase-I. In the subsequent meiotic stages, these univalents lagged at anaphases and constituted micronuclei in sporads. The triploid plants were also observed for natural propagation and it was noticed that no seeds were set. These plants were noticed to propagate vegetatively by rootstocks. Chromosomal pairing in triploid cytotype is typical of an allopolyploid. Based on the characterization of chromosomal pairing during meiosis, we assumed that the triploid individuals are probably alloploid in nature. Hypotheses concerning the possible origin of allotriploid in T. brunonis are also discussed.  相似文献

Abstract

Chromosome number knowledge of the Italian vascular flora is stored in the online database Chrobase.it, which includes 6723 records, referable to 3428 taxa, 2799 accepted species and subspecies (about 35% of the national flora), and 3410 different chromosome countings (cytotypes). Appropriate queries to Chrobase.it allowed us to calculate mean, modal and median chromosome numbers for the Italian vascular flora, for geographical subgroups (islands, south, centre, north) and for selected orders, families and genera. Chromosome number data were available for 41 out of 55 orders (74%) and 107 out of 428 families (67%), represented by 664 out of 1297 genera (51%). The most studied administrative regions are Sicily (844 taxa), Tuscany (592 taxa), and Sardinia (390 taxa), while the most studied families are Asteraceae (465 taxa), Fabaceae (266 taxa), Brassicaceae (158 taxa), and Poaceae (144 taxa). Chromosome numbers range from 2n = 6, occurring in several species of Hypochaeris (Asteraceae), to 2n = 240, occurring in Ophioglossum (Ophioglossaceae), Dryopteris (Dryopteridaceae) and Arenaria (Caryophyllaceae) (mode is 2n = 18, and median is 2n = 24). Chromosome number variability was analyzed by frequencies (linear plots) and ANOVA, resulting in significant differences among geographical groups (mean chromosome number increasing from islands-south to centre-north) and selected taxa. B-chromosomes occur in 5.3% of data (148 taxa) and their number is not significantly different among geographical areas, while they occur only in 14 orders, 17 families, and 56 genera. Cytomorphological studies were made on 322 populations of 150 species/153 taxa belonging to 79 genera and 23 families of flowering plants. Out of these, 14 species were cytologically worked out for the first time on a worldwide basis along with B-chromosomes (0–1B) recorded for the first time in Agrimonia eupatoria. Similarly, 29 species were reported with varied chromosomal reports on a worldwide basis. The chromosome numbers ranged from 2n = 10 to 2n = 96 and posed high amount of intraspecific genetic diversity when seen in relation to other reports for the worked out species in light of previous chromosomal databases on India and world basis. The different levels of ploidy in these species/taxa vary from 2x to 12x with the overall polyploidy being 27.45 % and predominant in members belonging to the genera Ranunculus and Epilobium as well as families Ranunculaceae and Rosaceae. In the taxa showing anomalous meiotic behavior, cytomixis is found to be quite common and associated with various meiotic abnormalities in the meiocytes, which ultimately leads to pollen sterility and formation of variable sized pollen grains.  相似文献

Cytogeographical variability among 564 plants from 26 populations of Turnera sidoides subsp. pinnatifida in mountain ranges of central Argentina was analysed with meiotic chromosome counts and flow cytometry and is described at regional and local scales. Populations were primarily tetraploids (2n = 4x = 28), although diploid (2n = 2x = 14), hexaploid (2n = 2x = 42), and mixed populations of diploids and triploids (2n = 3x = 21) were also found. Diploids, triploids, and hexaploids were fewer in number and restricted to narrow areas, while tetraploids were the most common and geographically widespread cytotype. Diploids grew at higher altitudes and in colder and wet locations; tetraploids had the broadest ecological spectrum, while hexaploids occurred at the lowest altitudes and in drier conditions. The cytotypes were also spatially segregated at a microgeographical scale. Diploids grew in the piedmont, tetraploids were in the adjacent valley, and in the contact zone of both cytotypes, patches of diploids and triploids were found. At a regional scale, the distribution of the cytotypes may be governed by a combination of ecological and historical variables, while segregation in the contact zone may be independent of the selective environment because the cytotypes are unable to coexist as a result of reproductive exclusion. In this study the exact chromosome number, detailed meiotic behavior in pollen mother cells and pollen viability were investigated, which can contribute to a better understanding of the cytological evolution of the species growing in the cold deserts of Lahaul-Spiti (Himachal Pradesh, India). This study is the first such comprehensive attempt to explore the region chromosomally. Chromosome number, meiotic behavior and pollen fertility were analyzed in 301 accessions of 140 species of Polypetalae. Chromosome counts in 14 species are the first ever records, viz., Aquilegia pubiflora (n?=?7), Corydalis govaniana (n?=?8), C. thyrsiflora (n?=?8), Hedysarum astragaloides (n?=?7), H. microcalyx (n?=?7), Oxytropis thomsoni (n?=?8), Rhodiola tibetica (n?=?10), R. wallichianum (n?=?16), Rosularia alpestris (n?=?14), Epilobium chitralense (n?=?18), E. leiospermum (n?=?18), Heracleum brunonis (2n?=?33), H. thomsonii (n?=?11) and Pleurospermum govanianum (n?=?9). New intraspecific diploid or polyploid cytotypes have been recorded in 13 species. The species of these cold deserts are quite active in evolution, depicting heterogeneity in chromosome number involving polyploidy, 51 species (36.43%) and/or aneuploidy (37 species). Various meiotic abnormalities were observed in the majority of the species, causing pollen sterility and pollen grains of variable sizes. We are of the opinion that harsh climatic conditions have caused various meiotic abnormalities in the majority of the plants, which has affected the genetic constitution and viability of male gametes.  相似文献

为探讨国产毛茛科(Ranunculaceae)驴蹄草属(Caltha L.)植物的细胞学特征,对驴蹄草(C.palustris L.)3个居群和花葶驴蹄草(C.scaposa Hook.f.&Thoms.)5个居群进行了细胞学研究。驴蹄草贵州纳雍居群的染色体数目为2n=32(四倍体),两个云南中甸居群的染色体数目均为2n=64(八倍体)。花葶驴蹄草四川红原、康定、石渠居群的染色体数目均为2n=32(四倍体),该数目为首次报道;西藏林芝和云南德钦居群的染色体数目均为2n=64(八倍体)。驴蹄草的染色体比花葶驴蹄草大。这两种植物的32或64条染色体分别以4条或8条为单位大致能够排列为8组同源染色体,但同一组内的染色体经常具有明显的异形性(heteromorphy),不同居群的核型组成多少具有差异。同时,还分析了驴蹄草和花葶驴蹄草的不同倍性细胞型在我国的地理分布式样。  相似文献

Chromosome number, meiotic behaviour and morphological characters related to habit were studied in 10 populations of Scutellaria platystegia Juz. from S. sect. Lupulinaria native to Iran. All populations are diploid and has the chromosome number 2n = 2x = 22, which is consistent with the proposed base number of x = 11. This taxon displayed regular bivalent pairing and chromosome segregation at meiosis. However, some meiotic abnormalities observed included various degrees of fragmented chromosomes, laggards and bridges in anaphase I to telophase II, precocious division of centromeres in metaphase I or II, asynchronous nucleus and cytomixis. We evaluated and determined the population limits within S. platystegia, employing multivariant statistics. We found a striking association between meiotic behaviour and gross morphology. The genus Ziziphora L. (Lamiaceae) consists of annual herbs, except Ziziphora clinopodioides, and is represented by six taxa in the flora of Turkey: Z. clinopodioides, Z. capitata, Z. persica, Z. tenuior, Z. taurica subsp. taurica and Z. taurica subsp. cleonioides. They are strong aromatic herbs and important medicinal plants. This study has been conducted to determine the palynological and karyological features of these taxa. The pollen morphology and exine structure of Ziziphora species were investigated by light microscope and scanning electron microscope. Pollen grains were determined as hexacolpate, medium in size and a circular or elliptic amb. The shape of pollen grains varies from oblate to prolate spheroidal. Ornamentation types of pollen grains are bireticulate, microreticulate or perforate. Somatic chromosome number has been determined and counted as 2n = 16, 18. Results indicate that the pollen characters and chromosome numbers of the genus are valuable for taxonomic applications and may be useful for infrageneric classification.  相似文献

With 600 species Ranunculus is the largest genus in Ranunculaceae, and has a broad global distribution. We studied the karyotypes of R. constantinopolitanus and R. sericeus species of Ranunculaceae and identified their symmetry level. New chromosome numbers of 2n = 21 (Nodeh woods population) and 2n ca. 63 (Javaherdeh population) are reported for R. constantinopolitanus. Two different populations of R. sericeus had two different chromosome results. We investigated morphological and karyological studies along with pollens micromorphology. Different populations of R. constantinopolitanus and R. sericeus, with different chromosome numbers showed morphological and micromorphological differences. Therefore, we considered the two populations of R. sericeus as cytotypes. There was a correlation between the studied morphological characters and pollen size with ploidy levels in the two R. constantinopolitanus populations.  相似文献

Knowledge of the chromosome variation in wild populations is essential to understand the pathways and restrictions of karyotype evolution in plants. The aim of this study is to conduct an intraspecific analysis of the karyotypes by fluorochrome banding and ribosomal DNA (rDNA) loci detection by fluorescent in situ hybridization (FISH) and of the meiotic behaviour in natural populations of Lathyrus nervosus, sect. Notolathyrus. Chromosome banding showed that, despite the high constancy in the karyotype formula and in the rDNA loci among populations, there is intraspecific variation in the amount and distribution pattern of 4’,6-diamidino-2-phenylindole (DAPI+) heterochromatin. However, those changes were not related to the total chromosome length of the haploid complements. This fact demonstrates that structural chromosome changes may be one of the most important mechanisms for karyotype variation among natural populations of L. nervosus. The chromosome number surveyed at the population level revealed the first case of polyploidy in South American species and the first case of uneven polyploidy of the genus. All the chromosome markers analysed indicated that the polyploids found originated by autopolyploidy. The meiotic analysis showed different chromosome abnormalities that may be generating numerical and structural changes in the sporads. The finding of unreduced gametes that are alive at anthesis suggests sexual polyploidization as the most probable mechanism involved in the origin of these 3x and 4x autopolyploid cytotypes in L. nervosus.  相似文献

A total of 44 accessions of Brachiaria decumbens were analysed for chromosome count and meiotic behaviour in order to identify potential progenitors for crosses. Among them, 15 accessions presented 2n = 18; 27 accessions, 2n = 36; and 2 accessions, 2n = 45 chromosomes. Among the diploid accessions, the rate of meiotic abnormalities was low, ranging from 0.82% to 7.93%. In the 27 tetraploid accessions, the rate of meiotic abnormalities ranged from 18.41% to 65.83%. The most common meiotic abnormalities were related to irregular chromosome segregation, but chromosome stickiness and abnormal cytokinesis were observed in low frequency. All abnormalities can compromise pollen viability by generating unbalanced gametes. Based on the chromosome number and meiotic stability, the present study indicates the apomictic tetraploid accessions that can act as male genitor to produce interspecific hybrids with B. ruziziensis or intraspecific hybrids with recently artificially tetraploidized accessions.  相似文献
