Resistance conferred by the L3 gene is active against most ofthe tobamoviruses, including the Spanish strain (PMMoV-S), aP1,2 pathotype, but not against certain strains of pepper mildmottle virus (PMMoV), termed P1,2,3 pathotype, such as the Italianstrain (PMMoV-I). Both viruses are nearly identical at theirnucleotide sequence level (98%) and were used to challenge Capsicumchinense PI159236 plants harbouring the L3 gene in order tocarry out a comparative proteomic analysis of PR proteins inducedin this host in response to infection by either PMMoV-S or PMMoV-I.PMMoV-S induces a hypersensitive reaction (HR) in C. chinensePI159236 plant leaves with the formation of necrotic local lesionsand restriction of the virus at the primary infection sites.In this paper, C. chinense PR protein isoforms belonging tothe PR-1, β-1,3-glucanases (PR-2), chitinases (PR-3), osmotin-likeprotein (PR-5), peroxidases (PR-9), germin-like protein (PR-16),and PRp27 (PR-17) have been identified. Three of these PR proteinisoforms were specifically induced during PMMoV-S-activationof C. chinense L3 gene-mediated resistance: an acidic β-1,3-glucanaseisoform (PR-2) (Mr 44.6; pI 5.1), an osmotin-like protein (PR-5)(Mr 26.8; pI 7.5), and a basic PR-1 protein isoform (Mr 18;pI 9.4–10.0). In addition, evidence is presented for adifferential accumulation of C. chinense PR proteins and mRNAsin the compatible (PMMoV-I)–C. chinense and incompatible(PMMoV-S)–C. chinense interactions for proteins belongingto all PR proteins detected. Except for an acidic chitinase(PR-3) (Mr 30.2; pI 5.0), an earlier and higher accumulationof PR proteins and mRNAs was detected in plants associated withHR induction. Furthermore, the accumulation rates of PR proteinsand mRNA did not correlate with maximal accumulation levelsof viral RNA, thus indicating that PR protein expression mayreflect the physiological status of the plant. Tobacco plants containing rolA (root locus A) from the Ri TL-DNA (root-inducing, left-hand, transferred DNA) from Agrobacterium rhizogenes have an altered phenotype, including male sterility, abnormal flower morphology, wrinkled leaves and shortened internodes. This phenotype is unstable, reverting to an attenuated form in lateral branches. We show that the pleiotropic effects of rolA are correlated with inhibition of the accumulation of conjugates between polyamines and the hydroxycinnamic acids. The compounds associated with male fertility are primarily affected, particularly the feruloyl derivatives.  相似文献

Oidium heveae, an obligate biotrophic pathogen of rubber trees (Hevea brasiliensis), causes significant yield losses of rubber worldwide. However, the molecular mechanisms underlying the interplay between O. heveae and rubber trees remain largely unknown. In this study, we isolated an O. heveae strain, named HN1106, from cultivated H. brasiliensis in Hainan, China. We found that O. heveae HN1106 triggers the hypersensitive response in a manner that depends on the effector‐triggered immunity proteins EDS1 (Enhanced Disease Susceptibility 1) and PAD4 (Phytoalexin Deficient 4) and on salicylic acid (SA) in the model plant Arabidopsis thaliana. However, SA‐independent resistance also appears to limit O. heveae infection of Arabidopsis, because the pathogen does not produce conidiospores on npr1 (nonexpressor of pr1), sid2 (SA induction deficient 2) and NahG plants, which show disruptions in SA signalling. Furthermore, we found that the callose synthase PMR4 (Powdery Mildew Resistant 4) prevents O. heveae HN1106 penetration into leaves in the early stages of infection. To elucidate the potential mechanism of resistance of Arabidopsis to O. heveae HN1106, we inoculated 47 different Arabidopsis accessions with the pathogen, and analysed the plant disease symptoms and O. heveae HN1106 hyphal growth and conidiospore formation on the leaves. We found that the accession Lag2‐2 showed significant susceptibility to O. heveae HN1106. Overall, this study provides a basis for future research aimed at combatting powdery mildew caused by O. heveae in rubber trees.  相似文献

Somatic embryogenesis of Panax ginseng CA Meyer was initiated from suspension aggregates of an embryogenic callus, in a liquid medium consisting of half strength Murashige and Skoog (1962) supplemented with the synthetic auxin benzoselenienyl-3 acetic acid. The addition of spermidine to this initiation medium significantly increased the production of somatic embryos. In this case, the total polyamine content of the embryogenic mass was higher than that of cultures without spermidine. At day 6 of the culture, a transient accumulation of free polyamines, mainly spermidine, was observed. After this peak, free and conjugated polyamines levels did not show significant variation nor did the polyamine oxidase activity. The results clearly demonstrated that spermidine supplied to the medium was oxidised by polyamine oxidase and partially metabolised into putrescine. The role of spermidine and its interaction with auxin in the initiation of the embryogenic process in Panax ginseng are discussed in relation to embryogenic potential.  相似文献

Summary The obligate parasitic fungus, Erysiphe graminis f. sp. hordei, was found to harbour plasmid-like extrachromosomal DNA. A 1.35-kb fragment of this 9kb plasmid was cloned into the pUC12 vector. No homology was detected to nuclear or mitochondrial DNA. As only about half of the 27 isolates examined contained plasmid-like DNA, this appears to be inessential for fungal survival. The plasmid is frequent in European isolates and is found in both newly collected isolates and in isolates kept under laboratory conditions for many years. No correlation between presence of plasmid and specific avirulence/virulence genes was found. The plasmid appear to be located in the mitochondria.  相似文献

In ungerminated rice seeds, (Japonica rice variety, CV Tapei 309), the content of free amines (putrescine, spermidine, spermine, tyramine) was higher in seed lots having a low germination frequency compared to those with high germination potential. Conversely, amine conjugates (di-feruloylputrescine, di-feruloylspermidine, diferuloyldiaminopropane and feruloyltyramine) decreased with loss of viability. Thus, these compounds appeared to constitute biochemical markers of seed viability. In seeds with high germination potential, conjugates decreased drastically during germination, with an early and rapid increase in free amines (putrescine, spermidine, tyramine). Arginine decarboxylase (ADC) activity was highest during the germination of high germination potential seeds, its activity gradually declining with loss of viability and being closely correlated with agmatine content. The polyamine biosynthetic inhibitors (-DL-difluoromethylarginine, DFMA, a specific and irreversible inhibitor of ADC; -DL-difluoromethylornithine, DFMO, a specific irreversible inhibitor of ornithine decarboxylase (ODC); cyclohexylammonium sulfate, CHA, inhibitor of spermidine synthase) neither depleted putrescine and spermidine levels nor inhibited germination in high germination potential seeds. In low germination potential seeds, the germination process was inhibited by DFMA or CHA. Application of agmatine resulted in a reversal of inhibition. DFMA inhibited ADC activity in both categories of seeds. In low germination potential seeds treated with CHA no ADC activity was found. These results suggest that amines are involved in the germination process of rice seeds. The main free amines identified during growth and development of rice seedlings were agmatine, putrescine, spermidine, diaminopropane and tyramine. Amine composition differed according to tissue and stages of development. Conjugated amines were only found in roots. We present evidence that arginine decarboxylase (ADC) regulates putrescine during the development of rice seedlings. When ADC action was blocked by DFMA (-DL-difluoromethylarginine, a specific irreversible inhibitor of ADC), polyamine titers and seedling development were diminished; when agmatine or putrescine was added, normal polyamine titers and growth were restored. The effects of DFMA were concentration dependent. DFMO (-DL-difluoromethylornithine, a specific irreversible inhibitor of ornithine decarboxylase or ODC) promoted growth and development at concentrations below 2 mM. This effect was probably related to its unexplained, but consistently observed slight enhancement of rice ADC. When the increase in the concentration of spermidine was prevented by CHA (cyclohexylammonium sulfate), the number of roots increased and the increase in length of leaves and roots was strongly inhibited. Flacca, tomato mutant in which the last step of ABA biosynthesis is blocked, displays changes both in polyamine content (especially the ratio between free and conjugated forms) and in ornithine-, arginine- and S- adenosylmethionine decarboxylase activities. Arginine decarboxylase activity is higher compared to that of ornithine decarboxylase especially in older plants (seven internode stage), being on line with its regulatory role. However this activity is not sufficient to justify the polyamine content, in particular of conjugated forms (disappearance of TCA-insoluble conjugates and dramatic decrease of TCA-soluble ones). These results are in favour of a multifunctional competition of many responses to hormone action.  相似文献

A new isosteric charge-deficient spermine analogue, 1,12-diamino-4,9-diaza-5-oxadodecan, and O-(7-amino-4-azaheptyl)oxime of 3-aminopropanal, a stable analogue of the Schiff base intermediate in the enzymatic oxidation of spermine, were synthesized. 为了寻找适合小麦白粉菌基因组DNA微量提取的方法,分别采用改进破壁法,液氮研磨法和溶菌酶消化法进行破壁,提取专性寄生菌小麦白粉菌DNA。结果表明,用改进的破壁方法,仅用3~10 mg的分生孢子粉所获得DNA的收率为(12.23±3.46)~(40.32±5.67)ng/mg,且OD260/OD280比值为1.71~1.92之间,说明该破壁方法获得的DNA收率大且纯度高。通过PCR反应获得了良好的效果。同时该方法也适用于小麦条锈菌和大麦白粉菌专性寄生菌DNA的提取。  相似文献

用小麦白粉病菌11个生理小种的混合菌种,对新疆地区的小麦近缘植物的7个属22个种的47份材料进行接种,除6份免疫外,其余均接种成功.用其中6个属19个种的29份小麦近缘植物产生的白粉病菌,对小麦回接,参试的29份材料全部回接成功.小麦白粉病菌对小麦近缘植物的寄生像在小麦上一样,有明显的寄生专化性.感病的小麦近缘植物的78.0%对小麦白粉病菌的感病性,随生育期增长而急剧下降.文中并对小麦白粉病中间寄主的作用进行了讨论.  相似文献

Powdery mildew, caused by Blumeria graminis is an important disease of cereals in many production regions. Until end of the last century triticale had been regarded as a species characterized by high level of resistance for this disease. However, after several years of intensive production on a big area in Poland, Germany and other European countries it start to be susceptible for many pathogens including B. graminis. Because of this, virulence structure of this pathogen population on triticale in Poland was evaluated across 2008–2010. Leaf samples with symptoms of the powdery mildew disease were collected randomly from nineteen localities. As a total, 1402 B. graminis isolates were collected: 23–25 isolates per locality in each year. Standard differential set of 28 genotypes was used: twenty‐one wheat with known resistance genes and seven triticale. Based on the obtained results it was possible to observe significant differences in virulence structure between years and localities. No virulence's against Pm21 (Yangmai5), and Pm3d + 4b (Kadett) were found in any year. All tested isolates were virulent on Moreno and Lamberto cultivars. In a total, 36% of tested isolates possessed 9, 11 or 12 virulence's per genotypes. Twenty five percent of tested isolates were virulent to 5 triticale cultivars. Correlation between pathotypes frequency and sampling region were not found what suggest that local epidemics play the most important role in triticale growing regions in Poland.  相似文献
