Karyological characteristics during basidiosporogenesis of Rhizopogon roseolus, a member of the hypogeous Agaricomycetes, were studied by light and transmission electron microscopy. More than 1000 tissue fragments of young basidiomata were stained with HCl-Giemsa and observed by a light microscopy to evaluate nuclear behavior. Basidium morphology in the hymenium was observed by transmission electron microscopy. Meiosis and post-meiotic mitosis took place in the center of the basidium. Sterigmata appeared when the first meiotic division occurred, and the center of the basidium became constricted when the second meiotic division occurred. Asynchronous nuclear migration from the basidium into the basidiospores occurred after post-meiotic mitosis, producing eight uninucleate basidiospores. The nucleus migrated patchily into basidiospores. The pattern of post-meiotic mitosis of R. roseolus, in which post-meiotic mitosis took place in the center of the basidium, is reported for the first time.  相似文献

Agaricus brasiliensis is a medicinal mushroom native to Brazil. It was first identified as Agaricus blazei and its scientific name continues to be debated. We examined the cytology of different Brazilian commercial strains of A. brasiliensis and the nuclear behavior of strain CS1 during basidiospore development using fluorescent microscopy. All strains have multinucleate hyphae and no significant differences in nuclei numbers were observed between them. Basidia from A. brasiliensis strain CS1 are typically tetrasporics and produce binucleate basidiospores, demonstrating that a postmeiotic mitosis occurs during basidiospore development. This result suggests that A. brasiliensis is primarily a heterothallic species.  相似文献

The meiotic spindles of Phallus impudicus had a similar ultrastructure to those seen in other basidiomycetes. After meiosis the basidal apex differentiated to form a number of broadly based protuberances which subsequently developed into basidiospores (up to 9 per basidium). The nuclei then underwent a post-meiotic mitosis within the cytoplasm of each basidiospore. After this mitosis one nucleus remained in the basidiospore and a complete septum developed at the base. The significance of this arrangement of meiosis, spore production and post-meiotic mitosis is discussed with respect to the taxonomy and life cycle of this unusual basidiomycete.  相似文献

In the genus Laccaria, basidial formation, dikaryotic basidia, karyogamy, and meiosis were generally similar to structures and phenomena reported for other Agaricales. The haploid nuclei of dikaryotic basidia resided side by side in the basidium prior to karyogamy. Following karyogamy a single nucleolus was observed in L. montana (four-spored species); several nucleoli remained in the nucleus of L. tortilis (two-spored species). The haploid number of chromosomes for L. montana appeared to be n = 9. Postmeiotic mitosis typically occurred in the basidiospores resulting in binucleate basidiospores for four-spored species and quadrinucleate basidiospores for two-spored species. Postmeiotic mitosis sometimes occurred in the sterigmata and basidia proper. In instances where postmeiotic mitosis occurred in basidia, mature basidiospores were not formed and the basidia were collapsed, and contained up to eight nuclei.  相似文献

The process of basidiospore formation in a mutant strain Fisc of Coprinus macrorhizus, a heterothallic species of Basidiomycete, which forms monokaryotic fruiting bodies was examined. A single nucleus in a young basidium divided mitotically and two daughter nuclei were fused subsequently. The fused nucleus then divided meiotically forming four basidiospores on a basidium. The typical chromosome behaviours in the first meiotic prophase were observed. Synaptonemal complexes were observed in a basidium at the first meiotic prophase. A continuous illumination of fruiting bodies was effective to arrest meiosis in monokaryotic fruiting bodies at the particular stage of meiotic division.  相似文献

Summary The cytology of teliospore germination and basidiospore formation inUromyces appendiculatus var.appendiculatus was characterized with light and fluoroscence microscopy. Meiosis of the diploid nucleus occurred in the metabasidium. The four haploid daughter nuclei migrated into the basidiospore initials where they underwent a post meiotic mitosis. Each basidiospore was delimited from the meatabasidium by a septum at the apex of the sterigma. Seventy-five percent of mature basidiospores were binucleate, 24.5% uninucleate, and 0.5% trinucleate. Mature, released basidiospores measured 16×9 m, were smooth-surfaced, and reniform to ovate-elliptical in shape.This study represents portion of a dissertation submitted by the senior author to the Faculty of Biology of the University of Constance in March, 1983, in partial fulfillment of the requirements for the degree of Doctor of Natural Sciences (Dr. rer. nat.).  相似文献

Meiotic division in Fomes annosus is similar to that reported for other higher fungi. Nuclei in dikaryotic cells prior to fusion in the basidia are long, thin, and double-stranded with paired heterochromatic areas. Various stages of prophase are similar to those in higher plants. At metaphase I and II seven pairs of chromosomes are aligned in a circle and the chromatids migrate to opposite poles established by two centrioles. The centrioles function in the movement of the nuclei in the basidium, nuclear alignment prior to fusion, establishment of poles for division, and the migration of the nuclei into the basidiospore. After nuclei migrate into the basidiospore, they soon divide, producing a binucleate spore.  相似文献

The ultrastructure of developing basidiospores in Rhizopogon roseolus is described. When viewed in the fruiting body chamber using scanning electron microscopy, basidiospores appear narrowly ellipsoid and have smooth walls. Eight basidiospores are usually produced on the apex of each sterigma on the basidium. Transmission electron micrographs showed that basidiospores formed by movement of cytoplasm (including the nuclei) via the sterigmata, and then each basidiospore eventually became separated from its sterigma by an electron-lucent septum. The sterigma and basidium subsequently collapsed, resulting in spore release. Freshly released spores retained the sterigmal appendage connected to the collapsed basidium. After spore release, the major ultrastructural changes in the spore concerned the lipid bodies and the spore wall. During maturation, lipid bodies formed and then expanded. Before release, the spore wall was homogeneous and electronlucent, but after release the spore wall comprised two distinct layers with electron-dense depositions at the inner wall, and the dense depositions formed an electron-dense third layer. The mature spore wall complex comprised at least four distinct layers: the outer electron-lucent thin double layers, the mottled electron-dense third layer, and the electron-lucent fourth layer in which electron-lucent granular substances were dispersed.  相似文献

The production of even a limited number of heterokaryotic spores would be advantageous for establishing new individuals after long distance dispersal. While Suillus and Laccaria species are known to produce binucleate, heterokaryotic spores, this condition is poorly studied for most ectomycorrhizal fungi. To begin addressing this matter the number of nuclei in basidiospores was recorded from 142 sporocarps in 63 species and 20 genera of ectomycorrhizal (EM) fungi. The mean proportion of binucleate basidiospores produced by sporocarps within a species ranged from 0.00 to 1.00, with most genera within a family showing similar patterns. Basidiospores from fungi in Amanita, Cortinariaceae and Laccaria were primarily binucleate but were likely still homokaryotic. Basidiospores from fungi in Boletaceae, Cantharellus, Rhizopogonaceae, Russulaceae, Thelephorales and Tricholoma were primarily uninucleate, but binucleate basidiospores were observed in many genera and in high levels in Boletus. Further research is needed to relate basidiospore nuclear number to reproductive potential in ectomycorrhizal species.  相似文献

Prototrophic strains recovered from crosses between auxotrophic strains of the lignin-degrading basidiomycete Phanerochaete chrysosporium were induced to fruit. The progeny of most of these self-crosses were prototrophic, indicating that the nuclei of the original prototroph were wild-type recombinants rather than complementary heterokaryons and that the binucleate basidiospores of this organism are homokaryotic. Various wild-type strains were shown to have multinucleate cells lacking clamp connections and to possess a variable number of sterigmata per basidium. Colonies arising from single conidia of various wild-type strains were all capable of producing fruit bodies and basidiospores. In addition, single basidiospores from three wild-type strains all produced fruit bodies and basidiospores. Nonfruiting as well as fruiting isolates were obtained from single basidiospores of five other wild-type strains. Basidiospores from these fruiting isolates always yielded colonies that fruited, again indicating that the spores are homokaryotic. Nonfruiting isolates from the same strain did not produce basidiospores when allowed to form a heterokaryon, implying that these isolates do not represent mating types. All this evidence indicates that P. chrysosporium has a primary homothallic mating system. In addition to fruiting and nonfruiting phenotypes, basidiospores from strain OGC101, a derivative of ME-446, gave rise to colonies which did not grow on cellulose (Cel). The fruiting, nonfruiting, and Cel phenotypes differed from each other and from the parental wild-type strain in a variety of characteristics, including growth, conidiation, and evolution of 14CO2 from 14C-side chain-labeled lignin, indicating that strain OCG101 is a heterokaryon.  相似文献

Summary

The nucleoli of Oudemansiella mucida and Nolanea cetrata become disassociated from the parent chromatin during both mitosis and meiosis. Disassociation of the nucleoli from the post-meiotic nuclei at the onset of migration is taken, along with other evidence, to indicate that the third nuclear division is initiated in the basidium and completed in the basidiospores. It is postulated that the coincidence of migration and the third nuclear division arises from the inability of nucleoli to pass through the sterigmata. This view is supported by observations made by the authors on other members of the Homobasidiomycetidae.  相似文献

Coprinopsis asiaticiphlyctidospora (Basidiomycota, Psathyrellaceae), is a new Coprinoid ammonia fungus from Amami and Okinawa, Japan. In macro- and micro-morphology, this species is similar to C. phlyctidospora and C. austrophlyctidospora, but differs in having smaller basidiospores and spiny basidiospore ornamentation. Phylogenetic analysis with the nuclear ribosomal internal transcribed spacer (ITS) data also supports this new taxon.  相似文献

食用外共生菌根菌是一类可食用并与植物共生的大型真菌,其在纯培养条件下菌丝生长缓慢,不会扭结发育成原基,不能完成生活史。因此关于食用菌根菌纯培养条件下生活史的研究报道极少。本研究的兰茂牛肝菌Lanmaoa asiatica已能在纯培养条件下诱导出原基,这使生活史的研究得以完成。利用扫描电子显微镜、透射电子显微镜及荧光显微镜对兰茂牛肝菌子实体担子、担孢子、纯培养菌丝和原基的核相进行了观察。结果表明,子实体菌丝细胞为双核,担子经过减数分裂形成4个单核担孢子;担孢子萌发时长轴端长出芽状突起伸长为菌丝,原孢子中的细胞核不分裂,直接进入菌丝,形成单核初生菌丝;初生菌丝5d后变为双核菌丝;纯培养菌丝及原基细胞均为双核,其菌丝表面光滑,未观察到锁状联合。  相似文献

There is controversy surrounding the described life cycle of the rust fungus Puccinia psidii sensu lato, which causes disease on several plant species in the family Myrtaceae. The objective of this study was to determine whether P. psidii s.l. is autoecious by performing basidiospore inoculations, and microscopically examining the fate of basidiospores on the leaf surface and nuclear condition at different stages of rust development. No spermogonia developed on leaves of Agonis flexuosa inoculated either with a teliospore suspension or basidiospores naturally discharged from telia. Uredinial sori that developed in all three inoculations with teliospore suspensions and in one of the five inoculations with naturally-discharged basidiospores from telia were most likely the result of urediniospore infections. Microsatellite analysis revealed that isolates made from these uredinial sori had the same multilocus genotype as that of the original isolate. No signs of penetration of plant cells by basidiospores were observed on A. flexuosa and Syzygium jambos. The nuclear condition of mycelia of uredinial sori, urediniospores, teliospores, and four-celled metabasidia was typical of that in many rust fungi. Our study could not provide unequivocal proof that P. psidii s.l. is autoecious. While it is possible that it could be heteroecious, with an unknown alternate aecial host, it is also possible that basidiospores have lost the ability to infect Myrtaceae or are infrequently operational.  相似文献

Light and electron microscope observations were made on the hyphal anastomosis, basidia and basidiospores ofTomentella bombycina andT. fuscoferruginosa. Three different types of anastomosis were observed and the presence of nuclei at the anastomosis area was seen in most cases. The probasidial cell appeared tetranucleate and nuclear migration to the sterigmata took place when basidiospore genesis was initiated. The basidiospores showed from 1 to 4 nuclei, but the nuclear identification on the electron microscope was difficult in mature basidiospores.T. fuscoferruginosa basidiospore cell wall appeared to be formed by one single layer, while theT. bombycina basidiospore wall showed three well-delimited layers with different electron density.
Zusammenfassung Licht- und elektronmikroskopische Beobachtungen sind an der Hyphalanastomose, Basidia und Basidiosporen vonTomentella bombycina undT. fuscoferruginosa gemacht worden. Drei verschiedene Typen von Anastomose sind festgestellt worden und die Gegenwart der Kerne war in der Fläche der Anastomose in den meisten Fällen sichtbar. Die probasidiale Zelle erschien vierkernig und eine Kernwanderung zu den Sterigmata fand statt, wenn Basidiosporengenese eingeleitet war. Die Basidiosporen zeigten 1 bis 4 Sporen, aber die Identifikation der Kerne war durch Elektronmikroskopie in reifen Basidiosporen schwierig.T. fuscoferruginosa formed die Zellwand der Basidiospore durch eine einzige Schicht, während die Zellwand der Basidiospore vonT. bombycina drei gut begrenzte Schichten mit verschiedenen Elektrondichtigkeit zeigte.

Filobasidiella neoformans, the teleomorph of Cryptococcus neoformans, was observed using electron microscopy to confirm its phyletic status. No septa were found in the basidium of F. neoformans. Mature basidiospores were encapsulated by a relatively thick cell wall. Basidiospores directly budded from the basidium without sterigma formation, and no hilar appendix was noted. A lateral component of a synaptonemal complex was observed. As for mitotic apparatus, a spherical nucleus associated organelle (NAO), 0.12 m in diameter, was found in the hemispherical depressive area of the nuclear envelope. Therefore, this fungus seems to have both characteristics of the Holobasidiomycetidae and of the primitive Basidiomycotina such as the Teliomycetes.  相似文献

Sistotrema, typified by S. confluens, is characterized mainly by its urniform basidia mostly with 6–8 sterigmata as well as by a monomitic hyphal system, oil rich hyphae and smooth basidiospores. The species of this genus have various hymenophore configurations and basidiospore shapes. During a field trip in 2011, two specimens with urniform basidia were collected from Changbaishan Nature Reserve, northeastern China. After careful morphological and molecular studies, they are described and illustrated here as a new species, S. subconfluens. The new species shares a terrestrial habit, stipitate basidiocarps and poroid hymenophores with S. confluens, besides urniform basidia. These characters make the two species different from all other species of Sistotrema with resupinate basidiocarps on wood. Sistotrema subconfluens differs from the type by having larger basidiocarps, shorter basidiospores and consistent poroid hymenophores. In phylogeny inferred from nuclear large subunit rDNA, the two species were sister taxa but clearly separated. The difference of internal transcribed spacer sequences between the two species was 3.6%.  相似文献

以3个不同的金针菇菌株为材料,研究了其担孢子的核相及遗传属性。荧光染色观察显示,担孢子核相以双核为主,双核孢子、单核孢子和无核孢子分别占80.2%、7.5%和12.3%。源于单孢分离物的菌丝为有隔膜、无锁状联合的多核菌丝。在交配试验中,源于不同菌株单孢分离物的菌丝原生质体的配对形成具锁状联合的菌落,而源于同一单孢分离物的菌丝原生质体的配对则形成无锁状联合的菌落,暗示担孢子中的两个核具有相同的交配型。RAPD分析显示,源于同一单孢分离物的菌丝原生质体为10个随机引物所扩增的图谱彼此完全相同,印证了担孢子中的双核是同质的。此外,观察表明,一个担子上着生有4个担孢子。因此,金针菇是一种具4个含同质双核担孢子的四极性蕈菌。  相似文献

When basidia of Cronartium asclepiadeum (Uredinales) develop basidiospores, nuclei migrate from the basidial cells into the basidiospores. A mitotic nuclear division yields two nuclei in the basidiospore. One of these nuclei degenerates if the basidiospore develops a secondary ballistospore. Several stages in the degeneration of the nuclei can be recognized: (1) Condensation of chromatin, (2) separation of the nucleus into a portion containing the chromatin and a portion containing the nucleoplasm by an invagination of the nuclear envelope, (3) reduction of nuclear volume, (4) enveloping and (5) twisting of nuclear membranes around the degenerating nucleus, (6) homogenization of the chromatin with reduction of the nuclear envelope, and reduction of the enclosing membrane complex from a multilamellar structure to a single membrane layer, and (7) invagination or splitting off of the spiralled membranes. The nuclear behavior during early developmental stages of secondary spore formation is similar to that of budding basidiomycetous yeasts. Basidiomycetes producing ballistospores may possibly have arisen from those whose reproducing phase is yeast-like.  相似文献
