Summary Rates of O2 uptake across isolated perfused skin of bullfrogs (Rana catesbeiana) were measured in relation to blood flow at three levels of ambient O2 tension: normoxia (O2 tension=152 torr), hypoxia (12% O2, 87 torr) and hyperoxia (42% O2, 306 torr). At bulk perfusion rates ranging from 3.4 to 10.1 l·cm-2·min-1, O2 uptake was positively correlated with hemoglobin delivery rate in both normoxia and hyperoxia, but was independent of delivery rate in hypoxia. Mean O2 uptake in normoxia was 3.8 nmol O2·cm-2·min-1 at a delivery rate of 9.8 nmol·cm-2·min-1 and 6.5 nmol O2·cm-2·min-1 at a delivery rate of 28.3 nmol·cm-2·min-1. At any given bulk perfusion rate, oxygen uptake averaged about 49% lower in hypoxia than in normoxia, decreasing in proportion to the reduction of O2 tension difference between medium and blood. In hyperoxia, O2 uptake did not increase proportionally with the difference in O2 tension between blood and medium, averaging only 50% higher at a 2.4-fold greater O2 tension difference. Cutaneous diffusing capacity for O2 averaged 0.041 nmol O2·cm-2·torr-1·min-1 during the first hour of perfusion in normoxia, and was not affected by reduction of ambient O2 tension. The results indicate that cutaneous O2 uptake in hypoxia is highly diffusion limited, and consequently, increases in cutaneous perfusion can not effectively compensate for reduction of ambient O2 tension. The profundal zone of Lake Esrom, Denmark has a dense population of Chironomus anthracinus, which survives 2–4 months of oxygen depletion each summer during stratification. The metabolism of 3rd and 4th instar larvae was examined in regard to variation in biomass and temperature. Respiration at air saturation was described by a curvilinear multiple regression relating oxygen consumption to individual AFDW and temperature. At 10 °C and varying oxygen regimes the O2 consumption and CO2 production of 4th instar larvae were almost unaltered from saturation to about 3 mg O2 l–1, but decreased steeply below this level. The respiratory quotient increased from 0.82 at saturation to about 3.4 at oxygen concentrations near 0.5 mg O2 l–1. This implied a shift from aerobic to partially anaerobic metabolism. At 0.5 mg O2 l–1 the total energy production equalled 20% of the rate at saturation of which more than one third was accounted for by anaerobic degradation of glycogen. This corresponded to a daily loss of 12 µg mg AFDW–1 or approximately 5% of the body reserves. At unchanged metabolic rate the glycogen store would last three weeks, but long term oxygen deficiency causes a further suppression of the energy metabolism in C. anthracinus.  相似文献

The role played by a bacterial community composed ofPseudomonas putida, strain 21;Pseudomonas stutzeri, strain 18; andPseudomonas sp., strain 5, and by physical and chemical factors in the degradation of CN and SCN was studied. It was shown that the degradation of CN is determined both by the action of bacteria and by abiotic physical and chemical factors (pH, O2, temperature, the medium agitation rate, etc.). The contribution of chemical degradation was found to increase drastically at pH below 9.0; when air was blown through the medium (irrespective of the pH value); under active agitation of the medium; and when the medium surface interfacing air was increased. Even at elevated pH values (9.0-9.2), suboptimal for bacterial growth, the microbial degradation could account for at most 20–25 mg/1 of CN, regardless of its initial concentration. When CN and SCN were concurrently present in the medium, the former compound was the first to be degraded by microorganisms. The rate of bacterial degradation of SCN under continuous cultivation in a chain of reactors was found to depend on its concentration, the medium flow rate, agitation rate, and the pattern of carbon source supply and could exceed 1 g/(l day). CN and SCN are utilized by bacteria solely as nitrogen sources. The mechanism of CN and SCN degradation by the microbial community is discussed. Deceased.  相似文献

Growth of the green algae Chlamydomonas reinhardtii and Chlorella sp. in batch cultures was investigated in a novel gas-tight photobioreactor, in which CO2, H2, and N2 were titrated into the gas phase to control medium pH, dissolved oxygen partial pressure, and headspace pressure, respectively. The exit gas from the reactor was circulated through a loop of tubing and re-introduced into the culture. CO2 uptake was estimated from the addition of CO2 as acidic titrant and O2 evolution was estimated from titration by H2, which was used to reduce O2 over a Pd catalyst. The photosynthetic quotient, PQ, was estimated as the ratio between O2 evolution and CO2 up-take rates. NH4 +, NO2 , or NO3 was the final cell density limiting nutrient. Cultures of both algae were, in general, characterised by a nitrogen sufficient growth phase followed by a nitrogen depleted phase in which starch was the major product. The estimated PQ values were dependent on the level of oxidation of the nitrogen source. The PQ was 1 with NH4 + as the nitrogen source and 1.3 when NO3 was the nitrogen source. In cultures grown on all nitrogen sources, the PQ value approached 1 when the nitrogen source was depleted and starch synthesis became dominant, to further increase towards 1.3 over a period of 3–4 days. This latter increase in PQ, which was indicative of production of reduced compounds like lipids, correlated with a simultaneous increase in the degree of reduction of the biomass. When using the titrations of CO2 and H2 into the reactor headspace to estimate the up-take of CO2, the production of O2, and the PQ, the rate of biomass production could be followed, the stoichiometrical composition of the produced algal biomass could be estimated, and different growth phases could be identified.  相似文献

A mass spectrometric 16O2/18O2-isotope technique was used to analyse the rates of gross O2 evolution, net O2 evolution and gross O2 uptake in relation to photon fluence rate by Dunaliella tertiolecta adapted to 0.5, 1.0, 1.5, 2.0 and 2.5 M NaCl at 25°C and pH 7.0.At concentrations of dissolved inorganic carbon saturating for photosynthesis (200 M) gross O2 evolution and net O2 evolution increased with increasing salinity as well as with photon fluence rate. Light compensation was also enhanced with increased salinities. Light saturation of net O2 evolution was reached at about 1000 mol m-2s-1 for all salt concentrations tested. Gross O2 uptake in the light was increased in relation to the NaCl concentration but it was decreased with increasing photon fluence rate for almost all salinities, although an enhanced flow of light generated electrons was simultaneously observed. In addition, a comparison between gross O2 uptake at 1000 mol photons m-2s-1, dark respiration before illumination and immediately after darkening of each experiment showed that gross O2 uptake in the light paralleled but was lower than mitochondrial O2 consumption in the dark.From these results it is suggested that O2 uptake by Dunaliella tertiolecta in the light is mainly influenced by mitochondrial O2 uptake. Anabaena doliolum and Chlorella vulgaris immobilized on chitosan were more efficient at removing NO3 , NO2 p–, PO4 3– and CR2O7 2– from wastewaters than cells immobilized on agar, alginate, carrageenan or even free cells. Carrageenan-immobilized cells, however, were better at removing NH4 + and Ni2+. The PO4 3– uptake capacity was significantly increased in cells starved of PO4 3– for 24 h. Agar-immobilized cells, though having good metal and nutrient uptake efficiency, had only a slow growth rate. The effects of glycerol and the oxygen transfer rate on the xylitol production rate by a xylitol dehydrogenase gene (XYL2)-disrupted mutant of Candida tropicalis were investigated. The mutant produced xylitol near the almost yield of 100% from d-xylose using glycerol as a co-substrate for cell growth and NADPH regeneration: 50 g d-xylose l−1 was completely converted into xylitol when at least 20 g glycerol l−1 was used as a co-substrate. The xylitol production rate increased with the O2 transfer rate until saturation and it was not necessary to control the dissolved O2 tension precisely. Under the optimum conditions, the volumetric productivity and xylitol yield were 3.2 g l−1 h−1 and 97% (w/w), respectively.  相似文献

The nonenzymatic reaction of glyoxylate and H2O2 was measured under physiological conditions of the pH and concentrations of reactants. The reaction of glyoxylate and H2O2 was secondorder, with a rate constant of 2.27 l mol-1 s-1 at pH 8.0 and 25° C. The rate constant increased by 4.4 times in the presence of Zn2+ and doubled at 35°C. We propose a mechanism for the reaction between glyoxylate and H2O2. From a comparison of the rates of H2O2 decomposition by catalase and the reaction with glyoxylate, we conclude that H2O2 produced during glycolate oxidation in peroxisomes is decomposed by catalase but not by the reaction with glyoxylate, and that photorespiratory CO2 originates from glycine, but not from glyoxylate, in C3 plants. Simulation using the above rate constant and reported kinetic parameters leads to the same conclusion, and also makes it clear that alanine is a satisfactory amino donor in the conversion of glyoxylate to glycine. Some serine might be decomposed to give glycine and methylene-tetrahydrofolate; the latter is ultimately oxidized to CO2. A novel bacterial species identified as Exiguobacterium sp. RD3 degraded the diazo dye reactive yellow 84A (50 mg l−1) within 48 h at static condition, at 30°C and pH 7. Lower salinity conditions were found to be favorable for growth and decolorization. Enzymatic activities of an H2O2 independent oxidase along with laccase and an azoreductase suggest their prominent role during the decolorization of reactive yellow 84A. Presence of an H2O2 independent oxidase in Exiguobacterium sp. RD3 was confirmed and hydrogen peroxide produced was detected by a coupled iodometric assay. Azoreductase activity was prominent in presence of cofactors NADH and NADP in mineral salt medium. Considerable depletion of COD of the dye solution during degradation of dye was indicative of conversion of complex dye into simple oxidizable products. Products of degradation were analyzed by HPLC, FTIR and GCMS. A possible product of the degradation was identified by GCMS. Degradation of dye resulted with significant reduction of phytotoxicity, confirming the environmentally safe nature of the degradation metabolites.  相似文献

Oxygen and CO2 exchange were measured concurrently in leaves of shade-grownAlocasia macrorrhiza (L.) G. Don during lightflecks consisting of short periods of high photon flux density (PFD) superimposed on a low-PFD background illumination. Oxygen exchange was measured with a zirconium-oxide ceramic cell in an atmosphere containing 1 600 bar O2 and 350 bar CO2. Following an increase in PFD from 10 to 500 mol photons·m-2·s-1, O2 evolution immediately increased to a maximum rate that was about twice as high as the highest CO2-exchange rates that were observed. Oxygen evolution then decreased over the next 5–10 s to rates equal to the much more slowly increasing rates of CO2 uptake. When the PFD was decreased at the end of a lightfleck, O2 evolution decreased nearly instantaneously to the low-PFD rate while CO2 fixation continued at an elevated rate for about 20 s. When PFD during the lightfleck was at a level that was limiting for steady-state CO2 exchange, then the O2-evolution rate was constant during the lightfleck. This observed pattern of O2 evolution during lightflecks indicated that the maximum rate of electron transport exceeded the maximum rate of CO2 fixation in these leaves. In noninduced leaves, rates of O2 evolution for the first fraction of a second were about as high as rates in fully induced leaves, indicating that O2 evolution and the electron-transport chain are not directly affected by the leaf's induction state. Severalfold differences between induced and noninduced leaves in O2 evolution during a lightfleck were seen for lightflecks longer than a few seconds where the rate of O2 evolution appeared to be limited by the utilization of reducing power in CO2 fixation.Abbreviation PFD photon flux density (of photosynthetically active radiation)  相似文献

1-Hydroxy-2-naphthoate is formed as an intermediate in the bacterial degradation of phenanthrene. A monooxygenase which catalyzed the oxidation of 1-hydroxy-2-naphthoateto 1,2-dihydroxynaphthalene was purified from the phenanthrene- and naphthalene-degrading Pseudomonas putida strain BS202-P1. The purified protein had a molecular weight of45 kDa and required NAD(P)H and FAD as cofactors. The purified enzyme also catalysed the oxidation of salicylate and various substituted salicylates. The comparison of the Kmand Vmax values for 1-hydroxy-2-naphthoate and salicylate demonstrated a higher catalytic efficiency of the enzyme for salicylate as a substrate. A significant substrate-inhibition was detected with higher concentrations of 1-hydroxy-2-naphthoate.The aminoterminal amino acid sequence of the purified enzyme showed significant homologies to salicylate 1-monooxygenases from other Gram negative bacteria. It was therefore concluded that during the degradation of phenanthrene the conversion of 1-hydroxy-2-naphthoate to 1,2-dihydroxynaphthalene is catalysed by a salicylate1-monooxygenase. Together with previous studies, this suggested that the enzymes of the naphthalene pathway are sufficient to catalyse also the mineralization of phenanthrene.  相似文献

A flue gas originating from a municipal waste incinerator was used as a source of CO2 for the cultivation of the microalga Chlorella vulgaris, in order to decrease the biomass production costs and to bioremediate CO2 simultaneously. The utilization of the flue gas containing 10–13% (v/v) CO2 and 8–10% (v/v) O2 for the photobioreactor agitation and CO2 supply was proven to be convenient. The growth rate of algal cultures on the flue gas was even higher when compared with the control culture supplied by a mixture of pure CO2 and air (11% (v/v) CO2). Correspondingly, the CO2 fixation rate was also higher when using the flue gas (4.4 g CO2 l−1 24 h−1) than using the control gas (3.0 g CO2 l−1 24 h−1). The toxicological analysis of the biomass produced using untreated flue gas showed only a slight excess of mercury while all the other compounds (other heavy metals, polycyclic aromatic hydrocarbons, polychlorinated dibenzodioxins and dibenzofurans, and polychlorinated biphenyls) were below the limits required by the European Union foodstuff legislation. Fortunately, extending the flue gas treatment prior to the cultivation unit by a simple granulated activated carbon column led to an efficient absorption of gaseous mercury and to the algal biomass composition compliant with all the foodstuff legislation requirements.  相似文献

Ferryl compounds [Fe(IV)=O] in living organisms play an essential role in the radical catalytic cycle and degradation processes of hemeproteins. We studied the reactions between H2O2 and hemoglobin II (HbII) (GlnE7, TyrB10, PheCD1, PheE11), recombinant hemoglobin I (HbI) (GlnE7, PheB10, PheCD1, PheE11), and the HbI PheB10Tyr mutant of L. pectinata. We found that the tyrosine residue in the B10 position tailors, in two very distinct ways, the reactivity of the ferryl species, compounds I and II. First, increasing the reaction pH from 4.86 to 7.50, and then to 11.2, caused the the second-order rate constant for HbII to decrease from 141.60 to 77.78 M−1 s−1, and to 2.96 M−1 s−1, respectively. This pH dependence is associated with the disruption of the heme–tyrosine (603 nm) protein moiety, which controls the access of the H2O2 to the hemeprotein active center, thus regulating the formation of the ferryl species. Second, the presence of compound I was evident in the UV–vis spectra (648-nm band) in the reactions of HbI and recombinant HbI with H2O2, This band, however, is completely absent in the analogous reaction with HbII and the HbI PheB10Tyr mutant. Therefore, the existence of a hydrogen-bonding network between the heme pocket amino acids (i.e., TyrB10) and the ferryl compound I created a path much faster than 3.0×10−2 s−1 for the decay of compound I to compound II. Furthermore, the decay of the heme ferryl compound I to compound II was independent of the proximal HisF8 trans-ligand strength. Thus, the pH dependence of the heme–tyrosine moiety complex determined the overall reaction rate of the oxidative reaction limiting the interaction with H2O2 at neutral pH. The hydrogen-bonding strength between the TyrB10 and the heme ferryl species suggests the presence of a cycle where the ferryl consumption by the ferric heme increases significantly the pseudoperoxidase activity of these hemeproteins.  相似文献

Low production rates and sensitivity to O2 are two major obstacles which prevent the technical exploitation of the ability of green algae to produce H2 from water. Both problems were addressed in the present work. The inhibitory effect of O2 on the hydrogen photoproduction of the green alga Chlorella fusca could be minimized by using algal cells which had not yet fully restored their oxygen evolving capacities after an artificially induced chloroplast de/regeneration cycle (de-/regreening). The H2 photoproductivity peaked after 30 h of greening light while the O2 evolution at this time reached only 59% of its normal capacity. The H2PP yields could be further increased if NH4Cl was added to the reaction medium at the beginning of the anaerobic preincubation period. No stimulatory effect was observed when NH4Cl was added just before illumination, i.e. at the end of the 5-h-preincubation period. It is assumed that NH4Cl inhibited the photosynthetic reduction of nitrite, which competed with hydrogen photoproduction indirectly by feedback repression of the NO 2 - /NO 3 - -reductive system. Pseudomonas putida and Pseudomonas fluorescens present as a coculture were studied for their abilities to degrade benzene, toluene, ethylbenzene, and xylenes (collectively known as BTEX) under various growth conditions. The coculture effectively degraded various concentrations of BTEX as sole carbon sources. However, all BTEX compounds showed substrate inhibition to the bacteria, in terms of specific growth, degradation rate, and cell net yield. Cell growth was completely inhibited at 500mgl–1 of benzene, 600mgl–1 of o-xylene, and 1000mgl–1 of toluene. Without aeration, aerobic biodegradation of BTEX required additional oxygen provided as hydrogen peroxide in the medium. Under hypoxic conditions, however, nitrate could be used as an alternative electron acceptor for BTEX biodegradation when oxygen was limited and denitrification took place in the culture. The carbon mass balance study confirmed that benzene and toluene were completely mineralized to CO2 and H2O without producing any identifiable intermediate metabolites.  相似文献

To investigate harmful effects of the dinoflagellate Alexandrium species on microzooplankton, the rotifer Brachionus plicatilis was chosen as an assay species, and tested with 10 strains of Alexandrium including one known non-PSP-producer (Alexandrium tamarense, AT-6). HPLC analysis confirmed the PSP-content of the various strains: Alexandrium lusitanicum, Alexandrium minutum and Alexandrium tamarense (ATHK, AT5-1, AT5-3, ATCI02, ATCI03) used in the experiment were PSP-producers. No PSP toxins were detected in the strains Alexandrium sp1, Alexandrium sp2.Exposing rotifer populations to the densities of 2000 cells ml−1 of each of these 10 Alexandrium strains revealed that the (non-PSP) A. tamarense (AT-6) and two other PSP-producing algae: A. lusitanicum, A. minutum, did not appear to adversely impact rotifer populations. Rotifers exposed to these three strains were able to maintain their population numbers, and in some cases, increase them. Although some increases in rotifer population growth following exposures to these three algal species were noted, the rate was less than for the non-exposed control rotifer groups.In contrast, the remaining seven algal strains (A. tamarense ATHK, AT5-1, AT5-3, ATCI02, ATCI03; also Alexandrium sp1 and Alexandrium sp2) all have adverse effects on the rotifers. Dosing rotifers with respective algal cell densities of 2000 cells ml−1 each, for Alexandrium sp1, Alexandrium sp2, and A. tamarense strains ATHK and ATCI03 showed mean lethal time (LT50) on rotifer populations of 21, 28, 29, and 36h, respectively. The remaining three species (A. tamarense strains AT5-1, AT5-3, ATCI02) caused respective mean rotifer LT50s of 56, 56, and 71 h, compared to 160 h for the unexposed “starved control” rotifers. Experiments to determine ingestion rates for the rotifers, based on changes in their Chlorophyll a content, showed that the rotifers could feed on A. lusitanicum, A. minutum and A. tamarense strain AT-6, but could graze to little or no extent upon algal cells of the other seven strains. The effects on rotifers exposed to different cell densities, fractions, and growth phases of A. tamarense algal culture were respectively compared. It was found that only the whole algal cells had lethal effects, with strongest impact being shown by the early exponential growth phase of A. tamarense. The results indicate that some toxic mechanism(s), other than PSP and present in whole algal cells, might be responsible for the adverse effects on the exposed rotifers.  相似文献

The objectives of this study were to clarify the influences of nitrogen (N) load on the growth and photosynthetic responses of Quercus serrata seedlings to O3 and to obtain basic data for evaluating the critical levels of O3 for protecting Q. serrata forests in Japan. The effects of O3 and/or N load on growth and photosynthetic activity of Q. serrata seedlings were investigated during the two growing seasons. Two-year-old seedlings were assigned to 12 experimental treatments, which were comprised of the combination of four gas treatments (charcoal-filtered air and three levels of O3 at 1.0, 1.5 and 2.0 times ambient concentration) and three N treatments (0, 20 and 50 kg ha−1 year−1). During the second growing season, no significant interactive effects of O3 and N load on the growth and net photosynthetic rate of the seedlings were detected. Threrfore, we concluded that N supply to the soil at ≤50 kg ha−1 year−1 does not significantly influence the growth and photosynthetic responses of Q. serrata seedlings to O3. Based on the O3 exposure-response relationships for the whole-plant growth of the seedlings, the critical level of O3 for Q. serrata was estimated to be approximately 36 nmol mol−1 as the average 15-h O3 concentration during the one growing season.  相似文献

Through use of a recently developed technique that can measure CO2 exchange by individual attached roots, the influences of soil O2 and CO2 concentrations on root respiration were determined for two species of shallow-rooted cacti that typically occur in porous, well-drained soils. Although soil O2 concentrations in the rooting zone in the field were indistinguishable from that in the ambient air (21% by volume), the CO2 concentrations 10 cm below the soil surface averaged 540 μLL−1 for the barrel cactusFerocactus acanthodes under dry conditions and 2400 μLL−1 under wet conditions in a loamy sand. For the widely cultivated platyopuntiaOpuntia ficus-indica in a sandy clay loam, the CO2 concentration at 10 cm averaged 1080 μLL−1 under dry conditions and 4170 μLL−1 under wet conditions. For both species, the respiration rate in the laboratory was zero at 0% O2 and increased to its maximum value at 5% O2 for rain roots (roots induced by watering) and 16% O2 for established roots. Established roots ofO. ficus-indica were slightly more tolerant of elevated CO2 than were those ofF. acanthodes, 5000 μLL−1 inhibiting respiration by 35% and 46%, respectively. For both species, root respiration was reduced to zero at 20,000 μLL−1 (2%) CO2. In contrast to the reversible effects of 0% O2, inhibition by 2% CO2 was irreversible and led to the death of cortical cells in established roots in 6 h. Although the restriction of various cacti and other CAM plants to porous soils has generally been attributed to their requirement for high O2 concentrations, the present results indicate that susceptibility of root respiration to elevated soil CO2 concentrations may be more important.  相似文献

Both photogeneration and quenching of singlet oxygen by monomeric and aggregated (dimeric and oligomeric) molecules of bacteriochlorophyll (BChl) d have been studied in solution and in chlorosomes isolated from the green photosynthetic bacterium Chlorobium vibrioforme f. thiosulfatophilum. The yield of singlet-oxygen photogeneration by pigment dimers was about 6 times less than for monomers. Singlet oxygen formation was not observed in oligomer-containing solutions or in chlorosomes. To estimate the efficiency of singlet oxygen quenching an effective rate constant for 1O2 quenching by BChl molecules (kq M) was determined using the Stern-Volmer equation and the total concentration of BChl d in the samples. In solutions containing only monomeric BChl, the kq M values coincide with the real values for 1O2 quenching rate constants by BChl molecules. Aggregation weakly influenced the kq M values in pigment solutions. In chlorosomes (which contain both BChl and carotenoids) the kq M value was less than in solutions of BChl alone and much less than in acetone extracts from chlorosomes. Thus 1O2 quenching by BChl and carotenoids is much less efficient in chlorosomes than in solution and is likely caused primarily by BChl molecules which are close to the surface of the large chlorosome particles. The data allow a general conclusion that monomeric and dimeric chlorophyll molecules are the most likely sources of 1O2 formation in photosynthetic systems and excitation energy trapping by the long wavelength aggregates as well as 1O2 physical quenching by monomeric and aggregated chlorophyll can be considered as parts of the protective system against singlet oxygen formation.Abbreviations BChl bacteriochlorophyll - MBpd methyl bacteriopheophorbide - Chl chlorophyll - TPP meso-tetraphenylporphyrin - TPPS meso-tetra (p-sulfophenyl) porphyrin  相似文献
