Aims

We assessed and quantified the cumulative impact of 20 years of biomass management on the nature and bioavailability of soil phosphorus (P) accumulated from antecedent fertiliser inputs.

Methods

Soil (0–2.5, 2.5–5, 5–10 cm) and plant samples were taken from replicate plots in a grassland field experiment maintained for 20 years under contrasting plant biomass regimen- biomass retained or removed after mowing. Analyses included dry matter production and P uptake, root biomass, total soil carbon (C), total nitrogen (N), total P, soil P fractionation, and 31P NMR spectroscopy.

Results

Contemporary plant production and P uptake were over 2-fold higher for the biomass retained compared with the biomass removed regimes. Soil C, total P, soluble and labile forms of inorganic and organic soil P were significantly higher under biomass retention than removal.

Conclusions

Reserves of soluble and labile inorganic P in soil were significantly depleted in response to continued long-term removal of P in plant biomass compared to retention. However, this was only sufficient to sustain plant production at half the level observed for the biomass retention after 20 years, which was partly attributed to limited mobilisation of organic P in response to P removal.

Background and aims

Single superphosphate (SSP) is a major source of phosphorus (P) used in grazing systems to improve pasture production. The aim of this experiment was to determine the fate of fertiliser P in clover pastures under field conditions.

Methods

A procedure was developed to radiolabel SSP granules with a 33P radiotracer, which was then applied to the soil surface (equivalent to ~12 kg P ha?1) of a clover pasture. Recovery of fertiliser P was determined in clover shoots, fertiliser granules and soil fractions (surface layer: 0–4 cm and sub-surface layer: 4–8 cm).

Results

The P diffusion patterns of the 33P-labelled SSP granules were not significantly different to those of commercial SSP granules (P?>?0.05). Recovery of fertiliser P in clover shoots was 30–35 %. A considerable proportion of the fertiliser P (~28 %) was recovered in the surface soil layer and was largely inorganic P.

Conclusions

Recovery of fertiliser P by clover plants was up to 35 % in the year of application. Much of the fertiliser P in soil fractions was inorganic P, which highlights the importance of inorganic P forms and dynamics in soils under clover pasture on a single season timeframe at these sites.

Background and aim

Recycled sources of phosphorus (P), such as struvite extracted from wastewater, have potential to substitute for more soluble manufactured fertilisers and help reduce the long-term threat to food security from dwindling finite reserves of phosphate rock (PR). This study aimed to determine whether struvite could be a component of a sustainable P fertiliser management strategy for arable crops.

Methods

A combination of laboratory experiments, pot trials and mathematical modelling of the root system examined the P release properties of commercial fertiliser-grade struvite and patterns of P uptake from a low-P sandy soil by two different crop types, in comparison to more soluble inorganic P fertilisers (di-ammonium phosphate (DAP) and triple super phosphate (TSP)).

Results

Struvite had greatly enhanced solubility in the presence of organic acid anions; buckwheat, which exudes a high level of organic acids, was more effective at mobilising struvite P than the low level exuder, spring wheat. Struvite granules placed with the seed did not provide the same rate of P supply as placed DAP granules for early growth of spring wheat, but gave equivalent rates of P uptake, yield and apparent fertiliser recovery at harvest, even though only 26 % of struvite granules completely dissolved. Fertiliser mixes containing struvite and DAP applied to spring wheat have potential to provide both optimal early and late season P uptake and improve overall P use efficiency.

Conclusions

We conclude that the potential resource savings and potential efficiency benefits of utilising a recycled slow release fertiliser like struvite offers a more sustainable alternative to only using conventional, high solubility, PR-based fertilisers.

Aims

Intercropping can improve plant yields and soil phosphorus (P) use efficiency. This study compares inter- and intra-species intercropping, and determines whether P uptake and shoot biomass accumulation in intercrops are affected by soil P availability.

Methods

Four barley cultivars (Hordeum vulgare L.) and three legume species (Trifolium subterreneum, Ornithopus sativus and Medicago truncatula) were selected on the basis of their contrasting root exudation and morphological responses to P deficiency. Monocultures and barley-barley and barley-legume intercrops were grown for 6 weeks in a pot trial at very limiting, slightly limiting and excess available soil P. Above-ground biomass and shoot P were measured.

Results

Barley-legume intercrops had 10–70% greater P accumulation and 0–40% greater biomass than monocultures, with the greatest gains occurring at or below the sub-critical P requirement for barley. No benefit of barley-barley intercropping was observed. The plant combination had no significant effect on biomass and P uptake observed in intercropped treatments.

Conclusions

Barley-legume intercropping shows promise for sustainable production systems, especially at low soil P. Gains in biomass and P uptake come from inter- rather than intra-species intercropping, indicating that plant diversity resulted in decreased competition between plants for P.

Aims

A better understanding of how plant growth, N nutrition and symbiotic nitrogen fixation (SNF) are influenced by soil inorganic N availability, for a wide range of legume species, is crucial to optimise legume productivity, N2 fixation, while limiting environmental risks such as N leaching.

Methods

A comparative analysis was performed for ten legume crops, grown in a field experiment and supplied with four N fertiliser rates. Dry matter, N concentration and SNF were measured. In parallel, root elongation rates were studied in a greenhouse experiment.

Results

For most species, N fertilisation had little effect on plant growth and N accumulation. SNF was reduced by soil inorganic N available at sowing but with large differences in the magnitude of the response among species. The response varied according to plant N requirements for growth and plant ability to retrieve inorganic N. Accordingly, root lateral expansion rate measured in RhizoTubes was highly correlated with plant ability to retrieve inorganic N measured in the field experiment.

Conclusion

Combining SNF response to soil inorganic N, shoot N and plant ability to retrieve inorganic N, allowed a robust evaluation of differential response to soil inorganic N among a wide range of legume species.

Aims

The aim of our study was to clarify whether inoculating a soil with Pseudomonas sp. RU47 (RU47) bacteria would stimulate the enzymatic cleavage of organic P compounds in the rhizosphere and bulk soil, promoting plant growth. Adding either viable or heat treated RU47 cells made it possible to separate direct from indirect effects of the inoculum on P cycling in soil and plants.

Methods

We performed a rhizobox experiment in the greenhouse with tomato plants (Solanum lycopersicum) under low P soil conditions. Three inoculation treatments were conducted, using unselectively grown soil bacteria (bacterial mix), heat treated (HT-RU47) and viable RU47 (RU47) cells, and one not inoculated, optimally P-fertilized treatment. We verified plant growth, nutrient availability, enzyme activities and microbial community structure in soil.

Results

A plant growth promotion effect with improved P uptake was observed in both RU47 treatments. Inoculations of RU47 cells increased microbial phosphatase activity (PA) in the rhizosphere.

Conclusions

Plant growth promotion by RU47 cells is primarily associated with increased microbial PA in soil, while promotion of indigenous Pseudomonads as well as phytohormonal effects appear to be the dominant mechanisms when adding HT-RU47 cells. Thus, using RU47 offers a promising approach for more efficient P fertilization in agriculture.

Aims

The objectives of this study were to evaluate (1) the fertilizer potential of bone char, (2) the effects of wood biochar on plant-available phosphorus (P), and (3) the role of root-mycorrhizae-biochar interactions in plant P acquisition from a P-fixing soil.

Methods

Incubation and pot experiments were conducted with a P-fixing soil and maize with or without root hairs and arbuscular mycorrhizae (AM) inoculation. Olsen-, resin-P and plant P accumulation were used to estimate P availability from bone char, co-pyrolyzed bone char-wood biochar, and separate bone char and wood biochar additions produced at 60, 350 and 750 °C, and Triple Superphosphate (TSP).

Results

Maize inoculated with AM showed similar P accumulation when fertilized with either 750 °C bone char or TSP. Pyrolyzing bone did not increase extractable P in soil in comparison to unpyrolyzed bone, apart from a 67 % increase in resin-extractable P after additions of bone char pyrolyzed at 350 °C. Despite greater Olsen-P extractability, co-pyrolysis of bone with wood reduced maize P uptake. Wood biochars reduced resin-P from bone char by 14–26 %, whereas oven-dried wood increased resin-P by 23 %.

Conclusions

Bone char is an effective P fertilizer, especially if root-AM interactions are simultaneously considered. Biochar influences plant access to soil P and requires careful management to improve P availability.

Background and aims

Variation in fire intensity within an ecosystem is likely to moderate fire effects on plant and soil properties. We tested the effect of fire intensity on grassland biomass, soil microbial biomass, and soil nutrients. Additional tests determined plant-microbe, plant-nutrient, and microbe-nutrient associations.

Methods

A replicated field experiment produced a fire intensity gradient. We measured plant and soil microbial biomasses at peak plant productivity the first growing season after fire. We concurrently measured flux in 11 soil nutrients and soil moisture.

Results

Fire intensity positively affected soil nitrogen, phosphorus (P), and zinc but did not appreciably affect plant biomass, microbial biomass, and other soil nutrients. Plant biomass was seemingly (co-)limited by boron, manganese, and P. Microbial biomass was (co-)limited mainly by P and also iron.

Conclusions

In the Northern Great Plains, plant and soil microbial biomasses were limited mainly by P and some micronutrients. Fire intensity affected soil nutrients, however, pulsed P (due to fire) did not result in appreciable fire intensity effects on plant and microbial biomasses. Variable responses in plant productivity to fire are common and indicate the complexity of factors that regulate plant production after fire.

Aims

Reclamation following oil sands mining in northeastern Alberta (Canada) creates adverse reforestation soil conditions, including extreme pH values. We elucidated pH tolerance limits of boreal plant species and how pH affects nutrient uptake in these plants.

Methods

We measured growth, gas exchange, and foliar nutrient concentration of 15 common northern boreal forest plants after eight weeks exposure to root zone pH ranging from 5.0 to 9.0. Cluster analyses were used to group these species based on their pH responses.

Results

Based on their growth and gas exchange responses to pH, the 15 plant species could be divided into five groups, each of which contained species that commonly co-occur in particular boreal forest site types. For the foliar nutrient responses to pH, the 15 species could be grouped into only two categories; both showed decreases in foliar N, P, Fe and Zn concentration with increasing pH, with a more pronounced effect on the group that included trembling aspen, paper birch and chokecherry.

Conclusions

The evidence of differential adaptation to pH by habitat type suggests the importance of soil pH as a factor affecting boreal plant species distribution and could be helpful for selection of species suitable for reclamation of sites with altered soil pH.

Aims

The mechanisms by which rhizosphere bacteria increase the availability of mineral P precipitates for plant use are understudied. However, Paraburkholderia bryophila Ha185 is known to solubilize inorganic phosphate in vitro via a novel process. Therefore, this study aimed to demonstrate P solubilization by Ha185 in association with roots of perennial ryegrass (Lolium perenne L.).

Methods

We developed a gnotobiotic plant assay to assess P solubilization by Ha185 on ryegrass roots under various nutrient conditions. A green fluorescent protein (GFP)-tagged derivative of Ha185 was used in conjunction with fluorescent microscopy and confocal microscopy to visualize colonization of ryegrass roots.

Results

Ha185 solubilized mineral P (hydroxyapatite) in association with ryegrass roots and increased ryegrass growth by 20% under P-limited conditions. The GFP-tagged Ha185 strain colonized the rhizoplane and penetrated the primary root of ryegrass, possibly through “crack entry” at the point of lateral root emergence, but also by entering the epidermal cells via root hairs.

Conclusions

Ha185 supported ryegrass growth under P-limited conditions, indicating this strain may improve availability of soil P for uptake by ryegrass. Tools developed in this study have broad application in the study of rhizobacteria-plant interactions.

Background and aims

Identification of organic P species is important to understand their origin, turnover in soils and their effects on soil fertility. Attention has been recently devoted to microbial inocula, referred to as Bioeffectors, that are capable to increase P bioavailability and plant uptake. Nevertheless, little is known on the effect of Bioeffectors on soil P forms and their dynamics in agricultural soils upon different P fertilization.

Methods

We investigated the effects of the application of different commercial inocula strains (Trichoderma harzianum T 22, Pseudomonas sp., and Bacillus amyloliquefaciens) alone or in combination with different P fertilizers (triple superphosphate, rock phosphate, and both composted cow- and horse-manure) on soil organic P forms. P forms were characterized by liquid-state 31P–NMR spectroscopy, while plant P uptake from P-treated soil was followed in a greenhouse pot experiment under maize cultivation.

Results

NMR spectra showed that the type of P fertilizer and bioeffectors inoculation, affected the abundance and the composition of organic P forms. The specific capacity of all bioeffectors, and especially Pseudomonas, was related to an increased content of diesters P forms. Pseudomonas, and, to a lesser extent, B. amyloliquefaciens showed the largest increase in combination with organic P amendments, which also provided the largest plant P uptake. This suggests a key role of Diester-P forms in determining P availability in agroecosystems.

Conclusions

Microbial inoculation plays an important role in the dynamics of soil P, inducing a rapid P cycling that prevents P fixation and losses from soils, thus enhancing the P fertilizer use efficiency in agricultural soils.

Aims

Progress in improving the internal phosphorus utilisation efficiency of crops has been limited, which may be due to poor screening methods that allow differences in P uptake among genotypes grown in soil to mask genotypic differences in shoot biomass produced per unit of shoot P (PUE). We investigated alternative soil and hydroponic screening methods for their capacity to produce a consensus ranking of genotypes with regard to PUE.

Methods

Six rice genotypes previously identified in hydroponic screening studies as being high, intermediate or low in PUE were screened using multi P rate hydroponic and soil-based experiments.

Results

Comparisons made at each rate of soil-P supply produced estimates of PUE strongly biased by P uptake differences among genotypes. Using multiple-rate data to derive response functions per genotype showed that similar P content was achieved at different rates of P supply but that high-PUE genotypes clearly separated from intermediate- and low-PUE genotypes if equal P content was used. Ranking analysis suggested that results obtained from soil agreed well with those from the hydroponic study.

Conclusions

PUE was significantly influenced by genotype and P supply, but there was no significant genotype x P supply interaction. Hence, we conclude that screening genotypes using hydroponics at one or two P supply levels is the most cost- and time effective means to screen large numbers of rice genotypes for PUE.

Backgrounds and aims

Soybean yield depends on total N uptake, N use efficiency, and harvest index. Nitrogen uptake relays on biological fixation (BNF) and soil absorption. Usually, BNF is considered a yield-related process. However, there is limited information on whether maximizing percent BNF (%BNF) is actually required to maximize N uptake and yield.

Methods

Seventy cultivars were evaluated for total N uptake, N use efficiency, and harvest index. Biological N fixation was determined in a subset of cultivars. The harvest index of N derived from atmosphere and from soil was also assessed.

Results

Yield was positively associated with total N uptake. Highest N uptake was not linked to increased %BNF. An inverse relationship between the amount of BNF (kgBNF) and soil N absorption was observed. Harvest index of N derived from BNF was 85%, while it was 77% for N derived from soil.

Conclusions

Highest total N uptake was attained by different combinations of kgBNF and mineral soil N absorption. This showed that maximizing %BNF is not required to maximize yield. High %BNF played a pivotal role in determining neutral soil N balance. This is so even though N derived from BNF was more partitioned to seeds than N derived from soil.

Background and aims

Macropores may be preferential root pathways into the subsoil. We hypothesised that the presence of macropores promotes P-uptake from subsoil, particularly at limited water supply in surface soil. We tested this hypothesis in a rhizotron experiment with spring wheat (Triticum aestivum cv. Scirocco) under variation of fertilisation and irrigation.

Methods

Rhizotrons were filled with compacted subsoil (bulk density 1.4 g cm?3), underneath a P-depleted topsoil. In half of these rhizotrons the subsoil contained artificial macropores. Spring wheat was grown for 41 days with and without irrigation and 31P–addition. Also, a 33P–tracer was added at the soil surface to trace P-distribution in plants using liquid scintillation counting and radioactive imaging.

Results

Fertilisation and irrigation promoted biomass production and plant P-uptake. Improved growing conditions resulted in a higher proportion of subsoil roots, indicating that the topsoil root system additionally promoted subsoil nutrient acquisition. The presence of macropores did not improve plant growth but tended to increase translocation of 33P into both above- and belowground biomass. 33P–imaging confirmed that this plant-internal transport of topsoil-P extended into subsoil roots.

Conclusions

The lack of penetration resistance in macropores did not increase plant growth and nutrient uptake from subsoil here; however, wheat specifically re-allocated topsoil-P for subsoil root growth.

Aims

Phosphorus (P) is frequently limiting crop production in agroecosystems. Large progress was achieved in understanding root traits associated with P acquisition efficiency (PAE, i.e. P uptake achieved under low P conditions). Most former studies were performed in controlled environments, and avoided the complexity of soil-root interactions. This may lead to an oversimplification of the root-soil relations. The aim of the present study was, therefore, to identify the dominant root and rhizosphere-related traits determining PAE, in contrasting soil conditions in the field.

Methods

Twenty-three maize hybrids were grown at two contrasting P levels of a long-term P-fertilizer trial in two adjacent soil types: alkaline and neutral. Bulk soil, rhizosphere and root parameters were studied in relation to plant P acquisition.

Results

Soil type had robust effect on PAE. Hybrids’ performance in one soil type was not related to that in the other soil type. In the neutral soil, roots exhibited higher specific root length, higher root/shoot ratio but lower PAE. Best performing hybrids in the neutral soil were characterized by top soil exploration, i.e., greater root surface and topsoil foraging. In contrast, in the alkaline soil, PAE and foraging traits were not correlated, P availability in the rhizosphere was greater than the bulk soil and phosphatase activity was higher, suggesting a ‘mining strategy’ in that case (i.e. traits that facilitate elevated P availability).

Conclusions

These results indicate the key role of environmental factors for roots traits determining high PAE. The study highlights the need to consider soil properties when breeding for high PAE, as various soil types are likely to require different crop ideotypes.

Background and aims

Most investigations of fungi as nematode antagonists have focused on their interactions with nematodes in the soil. This study tested a foliar-isolated endophytic Phialemonium inflatum for its effects against the root-knot nematode as an endophyte in cotton using a seed treatment inoculation.

Methods

Cotton seeds were inoculated with P. inflatum spore suspensions prior to planting. Nematode infection and reproduction were quantified at Day 12 and 6 weeks after nematode egg inoculation, respectively. To establish whether the observed negative effects on nematodes were due to P. inflatum in the soil or as an endophyte in the plant, we also applied a soil fungicide treatment at the seedling stage to kill the fungi outside the plant.

Results

Persistent suppression of nematode penetration and galling, as well as subsequent reproduction, were observed in endophyte-treated plants independent of fungicide treatment, consistent with an endophytic mode of nematode suppression; and these negative effects did not depend on the concentration of fungal inoculum used to treat to the seed.

Conclusions

Our study highlights a novel role for P. inflatum as part of a plant-fungal defensive symbiosis in cotton, as well as the need for a broader understanding of endophyte-plant-nematode ecological interactions.

Background and aims

Root hair growth and development are important features of plant response to varying soil conditions and of nutrient and water uptake. Most current methods of characterizing root hairs in the field are unreliable or inefficient. We describe a method to quantify root hair area in digital images, such as those collected in situ by minirhizotron systems.

Methods

This method uses ImageJ and R open source software and is partially automated using code presented here. It requires manual tracing of a subset of root hair images (training data set) to which a multivariate logistic regression is fit with each color channel in the image as an independent variable. Thereafter the model is applied to complete sets of selected root hair sections to estimate total root hair area.

Results

There was good agreement between the training data sets and the predictions of the regression models in castor (Ricinus communis L.), maize (Zea mays L.), and papaya (Carica papaya L.).

Conclusion

This method enables time-efficient and consistent quantification of root hairs using in situ root imaging systems that are already widely in use.

Background

The fungus Colletotrichum is a plant pathogen that causes the anthracnose disease, resulting in huge losses in various crops including the rose-scented geranium (Pelargonium graveolens). Although the bacterial community associated with plants has an important role in the establishment of plant diseases, little is known about what happens in P. graveolens.

Aims

To increase the knowledge about the bacterial community associated with P. graveolens and its relationship with anthracnose disease symptoms.

Methods

Quantitative PCR and high-throughput sequencing were combined to determine the presence of the fungus Colletotrichum and to reveal the bacterial communities associated with different plant parts – root, stem and leaf – and in the rhizosphere and bulk soil, and also to determine the respective bacterial communities associated with P. graveolens leaves symptomatic and asymptomatic for anthracnose disease.

Results

The fungus Colletotrichum was detected in all plant parts and in the surrounding soil. Bacterial communities varied spatially in plants, and the disease symptoms also influenced the composition of the bacterial community. Abundances of operational taxonomic units (OTUs) assigned to the phylum Actinobacteria and to the genus Streptococcus were greatly increased in asymptomatic leaves.

Conclusions

The bacterial community associated to geranium leaves responds to anthracnose symptoms.

Aims

This study aimed to determine the capacity of Si to mitigate Al toxicity in upland rice plants (Oryza sativa L.) by evaluating plant growth and the Si and Al uptake kinetics.

Methods

Plants were grown for 40 days, after which the Si and Al uptake kinetics (Cmin, Km and Imax) were analyzed. Then, the shoots and roots were separated, and the dry matter, root morphology and Si and Al concentration and accumulation in the plant were evaluated.

Results

Aluminum decreased plant growth and the Si uptake capacity by decreasing the root growth and Si transport system efficiency in the upland rice roots (> Km and > Cmin). Silicon mitigated Al toxicity in the upland rice plants by decreasing Al transport to the plant shoots, although it did not reduce the Al uptake rate (Imax). Si treatment increased the growth of upland rice plant shoots grown in the presence of Al without influencing the root growth. The alleviation of Al toxicity by Si is more evident in the susceptible upland rice cultivar Maravilha.

Conclusions

Silicon mitigated Al toxicity in the upland rice plants by decreasing Al transport to the plant shoots but did not reduce the Al uptake rate by roots.
