通过分析岷江柏的迁地保护居群和野生居群的遗传多样性、遗传结构及居群间基因流,判断迁地保 护岷江柏居群的遗传多样性水平,为其迁地保护提供理论基础。本研究利用GBS(Genotyping-by-Sequencing) 测序技术获得的SNP位点对四川大渡河双江口岷江柏迁地保护移栽苗、苗圃播种苗及3个野生居群进行主成分分析(PCA分析)、聚类分析、分子进化树、遗传多样性和遗传结构分析。经过GBS测序共获得高质量Clean Data 118 321 514 728 bp,并开发了1947 047个tags,从中鉴定到了1 259 610个SNP位点。系统发育进化树显示大部分移栽岷江柏居群和野生岷江柏聚在一起,居群结构分析结果显示交叉验证错误率的谷值确定最优分群数为1。4个岷江柏居群的观测杂合度(Ho)、期望杂合度(He)、Shannon信息指数I(Shi)、近交系数(Fis)、多态信息含量(Pic)的值分别为0.181 5~0.272 0、0.223 2~0.300 3、0.331 0~0.464 9、0.178 0~0.246 5和0.272 2~0.309 2,说明岷江柏居群的遗传多样性水平较高。移栽岷江柏居群的He=0.300 3,Shi=0.464 9,岷江柏居群迁地保护居群遗传多样性总体水平略高于野生居群。野生岷江柏居群中白湾隧道(BW)_vs_松岗镇(SA)的遗传分化指数(Fst)较大,基因流(Nm)较小(Fst=0.091,Nm=2.496),而迁地保护的岷江柏居群与野生岷江柏居群没有明显的遗传分化,居群间的基因交流频繁(Fst<0.05,基因流Nm>4),说明没有明显的分群现象,岷江柏居群迁地保护居群遗传多样性较高。因此,移栽濒危植物是迁地保护过程中较好的方法,本文为以后野生岷江柏迁地保护提供参考,为其他树木种质资源的保存提供理论依据。  相似文献

Several aspects of the population biology and the population genetic structure of Ctenomys porteousi were studied. Chromosomal and allozyme polymorphisms in two local populations separated by 10 km were used to infer genetic structure. Heterochromatic addition/deletion rearrangements for six autosomal pairs were employed as genetic markers. Allozyme variants were analysed by the study of thirty-one presumptive loci. Thirteen loci (43%) were polymorphic, average observed heterozygosity per locus was 10%, and mean number of alleles per locus was 1.57. Chromosomal and allozyme frequencies were homogeneous across populations. The estimated Fst values were very low, 0.009 and 0.022 for chromosomal and allozyme data respectively. Gene flow was indirectly estimated by the method of Wright, Nm values (mean number of migrants per generation per deme) was very high (27.2 and 11.3 for chromosomal and allozyme data respectively). The Fis values suggest a significant defect of heterozygotes that could be explained by the Wahlund effect. The ecological data indicate the occurrence of sharp changes in density and in distribution pattern in short time. The genetic and ecological data suggest that the population structure of C. porteousi differs from that found in other species of Ctenomys affected by chromosomal mechanisms of speciation.  相似文献

通过RAPD标记法对5类喀斯特小生境中小蓬竹的遗传结构及其多样性进行了分析。RAPD随机引物8个扩增共得87个清晰度高且重复性好的条带,分子量200~3 000 bp,含多态68条,物种PPL=78.16%,Nei’s基因多度H=0.291 3,Shannon多样性指数I=0.431 0。土面(TM)生长的小蓬竹遗传多样性水平最高(PPL=47.13%,H=0.195 3,I=0.283 4),石缝(SF)中的小蓬竹遗传多样性水平最低(PPL=39.08%,H=0.147 4,I=0.219 2)。POPGENE 32分析5类小生境中小蓬竹的遗传分化系数Gst=0.415 1,基因流N_m=0.704 5。5类小生境的小蓬竹间遗传相似系数(I)变化范围0.777 3~0.999 4,平均为0.865 6;遗传距离(D)变化范围0.000 6~0.251 9,平均为0.148 3。各类小生境的遗传多样性水平PPL均值为43.91%,5类小生境间的遗传分化Gst=0.405 4。研究结果说明,小生境间的差异增加了小蓬竹的遗传多样性。  相似文献

Intact chloroplasts isolated from mature leaf tissue of the homosporous fern Athyrium filixfemina were osmotically ruptured and subjected to starch gel electrophoresis in side by side comparisons with whole leaf extracts. The single enzyme activities of reportedly cytosolic [NADP]IDH and [NADP]ME were not expressed in the chloroplast fraction, and these were used as controls ensuring the cytosol-free quality of the chloroplast preparations. Isozymes F1,6DP-1, PGI-1, PGM-1, 6PGDH-1, ALDO-1, TPI-2, [NAD(P)]G3PDH-1, and [NAD(P)]G3PDH-2 are active in the chloroplast fraction, whereas Fl,6DP-2, PGI-2, PGM-2, 6PGDH-2, ALDO-2, and TPI-1 were lacking from the chloroplast fraction and are considered cytosolic. The single enzyme activities observed for AAT and SkDH, are chloroplastic. These data indicate that the two isozymes of certain enzymes in Athyrium filix-femina are not the products of duplicated loci resulting from polyploidy, but are distinct and subcellularly compartmentalized as demonstrated in heterosporous plants. Thus A. filix-femina is functionally diploid in spite of its high chromosome number of 2n = 80.  相似文献

Horizontal starch gel electrophoresis was used to examine genetic diversity within and differences between one population each of two morphologically different species of Haworthia, namely H. pumila and H. herbacea. Twenty-five leaf samples of each species were surveyed for 24 proteins of which 13 were useful for routine analysis, and gene products of 16 protein coding loci revealed genetic variation at 7 (43.8%) thereof in both species. Values of 1.63 (± 0.20) and 1.56 (± 0.18) were obtained for the mean number of alleles per locus and the average heterozygosity per locus was calculated at 0.168 (± 0.058) and 0.144 (± 0.048) for H. herbacea and H. pumila respectively. The malate dehydrogenase-2 locus is a potential genetic marker to identify the species studied electrophoretically. The mean genotypic distance index between the populations studied was 0.184, an indication of large degree of differentiation between the species. These values are much higher than values obtained for other members of the Aloaceae, showing that normal levels of genetic variation can be expected in at least some succulent monocotyledons.  相似文献

Progeny from single pair crosses of Atlantic herring were examined to determine the heritability of genetic variation at seven polymorphic allozyme loci. Mendelian inheritance of codominant autosomal alleles was established for IDH-2, LDH-1, LDH-2, ME-2, PGM-1, and PGI-2. This demonstration of Mendelian inheritance is essential for accurate interpretation of allozymic variation among natural populations of this pelagic species.  相似文献

An analysis of 28 enzyme loci throughout developmental stages ofSepedon fuscipennis fuscipennis Loew indicated that 16 were polymorphic and 4 were monomorphic in all stages. Nine loci were differentially expressed among the stages: EST-1, EST-2, MDH-2, MDH-3 and PGI-1 occurred only in larvae, AK-3 mainly in pupae, and AK-1, AO and HK-1 only in adults. 全球氮沉降通过影响树木叶片的生理过程影响森林生态系统的结构与组成,但目前关于氮沉降对森林生态系统中不同植物叶片非结构性碳水化合物(NSC)含量的影响还不十分清楚.本研究选取黄土丘陵区人工油松林中油松、辽东栎、忍冬、绣线菊、黄刺玫、茜草、披针苔草7种主要高等植物,比较了3年氮添加(0、3、6、9 g N·m-2·a-1,分别用N0、N3、N6、N9表示)对7种植物叶片中NSC的影响.结果表明: 不同植物叶片可溶性糖、淀粉含量变异很大,二者变异最高的均为黄刺玫,最低的分别为绣线菊和披针苔草;不同植物可溶性糖、淀粉对氮添加的响应存在明显差异.N6处理下茜草的可溶性糖和淀粉的变异高于其他物种,N3和N9处理下绣线菊的变异高于其他物种,可溶性糖和淀粉含量变异最小的物种在不同氮添加水平下不同.随着氮添加水平的增加,油松和披针苔草的可溶性糖含量持续升高,绣线菊持续降低,辽东栎、忍冬和黄刺玫的可溶性糖含量先降低后增加,在N6处理达到最小,而茜草的可溶性糖含量呈现较复杂的变化趋势.氮添加对植物叶片中淀粉含量的影响表现为油松、忍冬和披针苔草的淀粉含量持续增加,绣线菊先降低后增加,在N3处理达到最小,而黄刺玫和茜草则呈现较复杂的变化趋势,辽东栎淀粉含量变化趋势平缓.在氮添加后,土壤pH、有机碳、全氮、全磷与植物叶片NSC含量无相关关系,仅N0和N3水平下上述土壤理化指标会显著影响可溶性糖/淀粉.表明林地不同植物叶片中NSC含量对氮添加的响应明显不同,研究全球氮沉降或氮添加对林地生态系统的影响需要考虑不同植物,尤其不同生活型植物的不同响应.  相似文献

为科学认识科尔沁沙地优势固沙灌木的生态适应性和固沙植被演变规律, 该研究对科尔沁沙地流动沙丘、半固定沙丘、固定沙丘和丘间低地的优势固沙灌木小叶锦鸡儿(Caragana microphylla)和盐蒿(Artemisia halodendron)进行野外调查, 研究了这两种固沙灌木的叶片氮(N)、磷(P)化学计量特征、灌丛土壤养分状况以及内稳性特征。结果表明: 1)与盐蒿相比, 灌木小叶锦鸡儿具有较高的叶片N含量及N:P, 而P含量仅为盐蒿的1/2; 2)两种优势固沙灌木灌丛下土壤的全N、全P含量及速效N、速效P含量高于该地区土壤的平均水平, 小叶锦鸡儿灌丛下土壤养分含量显著高于盐蒿灌丛下土壤; 3)盐蒿叶片N、P化学计量内稳性指数(H)表现为HP > HN:P > HN, 说明盐蒿更易受土壤N的限制; 小叶锦鸡儿叶片N、P化学计量内稳性指数表现为HN:P > HN > HP, 意味着小叶锦鸡儿更易受土壤P的限制。在N含量较低的沙化草地, HN较高的固沙灌木小叶锦鸡儿比盐蒿更具生长优势, 对于该地区生态恢复及保护具有不可替代的作用。然而, 小叶锦鸡儿额外吸收的N, 使其生长过程可能易受P的限制, 因此在沙地恢复过程中应注意土壤P的供应。  相似文献

采用13对SSR引物,运用Bioptic Qsep100全自动核酸分析系统,分析了天目山5个海拔檫木群体的遗传多样性和遗传结构及其在不同海拔下的变化模式.结果表明: 天目山檫木群体具有较高的遗传多样性水平,其中期望杂合度和观察杂合度分别为0.84和0.61.根据Shannon指数,天目山檫木中海拔(500~800 m)群体的遗传多样性水平大于低海拔(200 m)和高海拔(1100~1400 m)群体的遗传多样性水平.由基因分化系数和AMOVA分析可知,檫木种群的遗传变异主要存在于群体内.STRUCTURE分析和UPGMA聚类分析表明,中、低海拔被划为一个群体,而高海拔被划为另一个群体.低海拔和中海拔檫木群体遗传距离的差异表明,人为干扰对物种多样性具有负面效应,而自然保护区对物种多样性的保护起到了积极作用.  相似文献

极小种群野生植物云南蓝果树是国家和云南省实施极小种群野生植物保护工程的代表性物种。为有效保护其遗传资源,本研究通过二代测序技术,对其进行简化基因组测序,开发一批特异性高的单核苷酸多态性标记,分析现存群体的遗传结构和遗传多样性。经过遗传变异检测,本次研究中共获得SNP位点98 498个,通过样品最低测序深度>2,样品缺失率<0.5,次要基因型频率(MAF)>0.05筛选以后,得到有效SNP位点6 309个。基于过滤后的SNP,运用生物信息学分析方法,对云南蓝果树完成了群体的遗传分析,其中:系统进化树分析将云南蓝果树划分为3大类,研究分析了云南蓝果树各分类的私人等位基因数目(Private)、平均观测杂合度(Ho)、平均期望杂合度(He)、核苷酸多样性(π)和平均近交系数(FIS)5个遗传多样性参数;群体结构和主成分分析进一步证明了,云南蓝果树现存植株之间亲缘关系较远,遗传多样性差异较大,具有很高的遗传资源保存价值。本研究结果将为基于遗传管理的云南蓝果树就地保护、遗传资源保存和种群重建等保护工程提供科学依据。  相似文献

Nine polymorphic out of 13 isoenzymes (Gdh-A, Lap-A, Mdh-B, Mdh-C, Ndh-A, 6Pgd-B, Pgi-B, Pgm-A and Pgm-B) were used to assess genetic diversity in seven natural Cupressus sempervirens populations in South-western Anatolia. Except for Pgi-B, all loci were in Hardy–Weinberg equilibrium. A high level of genetic diversity was observed within populations. The mean number of alleles was 2.3±0.08, the effective number of alleles was 1.53±0.10, the percentage of polymorphic loci was 82.6±8.7%, expected heterozygosity was 0.350±0.039, observed heterozygosity was 0.326±0.039, and the fixation index was 0.049. Interpopulation differentiation was moderate, with differentiation indices of GST=0.073 and δ=0.104. A UPGMA tree and differentiation (Dj) showed that Kumluca, Kemer and Kas were the most differentiated populations. Pgm-B was one of the loci contributing most to interpopulation differentiation (GST=0.119; Di=0.201), clearly distinguishing Eastern populations, as indicated by a highly significant correlation coefficient between Dj and longitude (R2=0.88**). No other significant correlation was found between genetic measures and geographical traits for the Turkish cypress populations. A metanalysis of these and other cypress populations from Crete and the Aegean islands, that had been characterised previously using the same isoenzymes, showed no significant relationship between altitude and genetic measures (within-population differentiation or δT, observed heterozygosity, fixation index and Dj). Observed heterozygosity and latitude were negatively correlated. Longitude was found to be correlated with heterozygosity, fixation index and differentiation. Genetic and geographic distances were positively correlated over loci. Deviations from selective neutrality by Ewens–Watterson test were observed for Pgm-B, Ndh-A and 6Pgd-B.  相似文献

The genetic structure of populations of the Sigmodontinae rodent Calomys laucha was studied by means of allozyme electrophoresis. This highly opportunistic species is found preferably in periodically perturbed habitats of crop fields in central Argentina, where it can attain very high densities. A total of 17 enzymatic proteins assayed gave information on 25 loci; only four were monomorphic in the seven populations studied. Levels of genetic variability (Ho from 0.144 to 0.171; P95% from 44% to 56%) were higher than mean values reported for mammals and rodents. These high levels of heterozygosity could be maintained by large populations that do not experience great fluctuations in size, or by a social structure consistent in many small breeding groups that are formed and dissappear every breeding season. Genetic differentiation at a macrogeographical scale (θ=0.018) was low but statistically significant, and showed no correlation with geographic distance between pairs of populations. The pattern of population differentiation found is compatible with a relatively recent range expansion.  相似文献

Aims Stoichiometric homeostasis is an important mechanism in maintaining ecosystem structure, function, and stability. The invasion of exotic species, Spartina alterniflora, has largely threatened the structure and function of native ecosystems in the Minjiang River estuarine wetland. However, how S. alterniflora invasion affect plant stoichiometric homeostasis is largely unknown. This could enhance our understanding on wetland ecosystem stability and expand the applications of ecological stoichiometry theory.
Methods Nitrogen (N) and phosphorus (P) contents of plant organs and soils in the S. alterniflora, Cyperus malaccensis var. brevifolius, and S. alterniflora-C. malaccensis var. brevifolius mixture were measured, and the homeostatic index (H) was calculated according to the stoichiometric homeostasis theory.
Important findings Our results showed that the invasion of S. alterniflora significantly increased soil N:P ratio (p < 0.05), but did not affect soil N or P contents. The N and P contents of leaf and stem were the highest for S. alterniflora, and those of the stem were the highest for C. malaccensis var. brevifolius. At the ecosystem level, the average of homeostatic index (H) of N (HN, 25.31) was larger than those of P (HP, 10.33) and N:P (HN:P, 2.50). At the organ level, root HN was significantly larger than stem HN (p < 0.05) and sheath HN:P was greater than root HN:P (p < 0.05), while there was no significant difference for HP among root, stem, leaf, and sheath (p > 0.05). As for species, root HN of S. alterniflora was significantly larger than that of C. malaccensis var. brevifolius in the mixture community (p < 0.05). In the monoculture, stem HN:P of S. alterniflora was significantly higher than that of C. malaccensis var. brevifolius (p < 0.05). Furthermore, root HN, leaf HN and sheath HN of S. alterniflora in the mixed community was significantly larger than that of S. alterniflora in the monoculture (p < 0.05), suggesting that S. alterniflora invasions increased their stoichiometric homeostasis. Meanwhile, the stoichiometric homeostasis of invasive and native plants were influenced by multiple factors, such as nutrients, organs, vegetation, and invasion. However, larger homeostasis was found in S. alterniflora than in C. malaccensis var. brevifolius in some particular organs either in mixture or monoculture communities. Therefore, the successful invasion of S. alterniflora may result from higher homeostatic index than the native species, C. malaccensis var. brevifolius.  相似文献

Variability and genetic divergence of 11 field populations of seven species of theCampanula elatines andC. fragilis rock-plants group from the Mediterranean and pre-Alpine areas have been studied by starch-gel electrophoretic techniques.Campanula isophylla, C. elatines, C. elatinoides, C. fragilis subsp.fragilis and subs.cavolinii, C. garganica were collected in Italy, whilstC. fenestrellata subsp.istriaca andC. portenschlagiana came from W. Jugoslavia. Twelve enzymatic loci for each population were genetically analysed: PGI-1 and 2, PGM-2, IDH-1 and -2, SKDH, ME, ADH, GOT-2, MPI-1 and -2, SOD-1. Reproduction in Chthamalus montagui Southward and Chthamalus stellatus (Poli) has been compared along gradients of wave exposure and tidal height. The proportion of barnacles carrying egg masses was greatest at low shore levels in both species, although the difference between levels was more pronounced in C. stellatus. Both species were found to breed between the beginning of May and the end of September. The mean number of broods released per season in both species ranged from 1.0 to 2.1 at high shore levels, from 2.2 to 3.2 at mid shore levels, and from 2.6 to 4.4 at low shore levels. C. stellatus predominated low on the shore at an intermediately exposed site and on the mid and low shore at the most exposed site. At these places it produced more broods per year than C. montagui. Development of the embryos of the two species was complete after 3 wk in vivo at 15°C. Eggs containing embryos of C. stellatus were larger than those of C. montagui, and within each species, larger barnacles produced larger eggs. Egg masses of C. stellanus included more eggs than those of C. montagui. Brood size in both species was greatest in shelter and least in exposure. Egg production was, however, similar in the two species when adjusted for differences in body mass. Egg production per unit body mass was greatest at an intermediately exposed site.  相似文献

1. The methyl viologen-catalyzed Mehler reaction was investigated in intact cells of five species of blue-green algae and Chlamydomonas reinhardi.

2. In the presence of methyl viologen, all the blue-green algae except Anabaena flos-aquae show a light-dependent O2 consumption as well as a post-illumination O2 evolution. The rate of O2 consumption is stimulated by 1 mM KCN, an inhibitor of catalase, but the dark O2 evolution becomes suppressed.

3. A. flos-aquae shows a light-dependent methyl viologen-catalyzed O2 uptake which is not affected by 1 mM KCN. Furthermore, there is no release of O2 in the dark following illumination.

4. With C. reinhardi, the cells do not show any net O2 exchange during or after illumination. Addition of 1 mM KCN, however, results in an immediate O2 uptake in the light.

5. Based on the mechanism postulated for the Mehler reaction in isolated chloroplasts, it was deduced that the differences in the kinetics of the O2 exchange catalyzed by methyl viologen reflect differences in the endogenous catalase activity in these algae. Cells of A. flos-aquae are deficient in catalase activity whereas those of the other blue-green algae possess catalase, although at low activity. C. reinhardi, on the other hand, has high catalase activity in vivo.

6. These findings are corroborated by results obtained from O2 electrode measurements of catalase activity in cell-free extracts of these algae.

7. The possible roles of catalase in algae and the implications of these results are also discussed.  相似文献

Yeasts similar to Candida kruisii were isolated repeatedly from the digestive tracts of basidioma-feeding beetles, especially nitidulids inhabiting and feeding on a variety of agarics in the southeastern USA and Barro Colorado Island, Panama. Based on the identical sequences of the D1/D2 domains of the LSU rRNA gene (rDNA) and host beetle information, the isolates were grouped into 19 genotypes which varied from C. kruisii by up to 38 nucleotide differences in the D1/D2 region. Phylogenetic analysis of rDNA sequences and phenotypic traits placed the isolates in C. kruisii and in nine undescribed taxa. The new species and type strains are designated as Candida pallodes (NRRL Y-27653T), C. tritomae (NRRL Y-27650T), C. panamensis (NRRL Y-27657T), C. lycoperdinae (NRRL Y-27658T), C. atbi (NRRL Y-27651T), C. barrocoloradensis (NRRL Y-27934T), C. aglyptinia (NRRL Y-27935T), C. stri (NRRL Y-48063T), and C. gatunensis (NRRL Y-48064T). A phylogeny based on analysis of a combined database of sequences of SSU and LSU rDNA and the ITS region showed that the nine new species formed a novel sister clade to C. kruisii that was strongly supported by bootstrap analysis. Candida pallodes, C. tritomae, C. panamensis, and C. lycoperdinae formed one subclade, while C. atbi, C. barrocoloradensis, C. aglyptinia, C. stri, and C. gatunensis formed a second distinct subclade within the larger clade. Candida pallodes and C. atbi showed a strong host specificity to beetle species in the genus Pallodes (Coleoptera: Nitidulidae) collected from a variety of agarics. On the other hand, C. panamensis, C. tritomae, and C. lycoperdinae were associated with several unrelated beetles in Erotylidae, Scarabaeidae, Tenebrionidae, and Curculionidae as well as Lycoperdina ferruginea (Nitidulidae). Candida pallodes, C. tritomae, C. lycoperdinae, and C. atbi have been isolated repeatedly in the USA, while the other five new species have been found only at Barro Colorado Island, Panama.  相似文献

本文采用蛋白电泳技术研究滇南小耳猪血液蛋白多态性。共分析了滇南小耳猪32个遗传位点,其中AKP、CAT、ES、PA、6PGD和TF等6个位点检测到多态性,多态位点百分比P=0.1875, 平均杂合度H=0.0712。结果表明滇南小耳猪的血液蛋白多态程度较高,反映在蛋白质水平上的遗传多样性较为丰富。  相似文献

The effects of light quality on growth, biochemical composition and photosynthetic production in Cyclotella caspia Grunow and Tetraselmis gracilis (Kylin) Butcher were evaluated under controlled laboratory conditions. Cyclotella caspia had the highest values for maximum growth rate in blue-green light, whereas T. gracilis grew faster in red light. The highest cellular contents of chlorophylls [a, (c1 + c2)] and carotenoids of C. caspia were found respectively in red and blue-green light, while protein content did not change in response to spectral quality. Tetraselmis gracilis cells were more stimulated to synthesize pigments and protein when incubated in white light. For both species, pigment ratios showed intermediate values in white regime. The maximum values for photosynthetic rates were obtained in blue-green and red regimes in C. caspia and in red light in T. gracilis. The chromatic adaptive mechanisms shown for both species are compared and discussed in light of recent works presented for different phytoplankters, with emphasis on ecophysiological responses obtained in distinct spectral regimes.  相似文献
