The process of daughter colony formation in Pediastrum biradiatumMeyen, P. tetras (Ehren-berg) Ralfs, P. duplex Meyen, and Hydrodictyonreticulatum (L.) Lagerheim has been investigated using lightmicroscopy and electron microscopy of thin sections. The precisedisposition of the organelles confers upon the zoospores ofthese algae a special symmetry of a type unusual for the Chlorophyta.Apart from this, the detailed ultrastructure of the zoosporeand young vegetative cells resembles that of other green algaepreviously investigated. A common sequence of events has beenfound to occur during colony formation in both Pediastrum andHydrodictyon, with a correlation between zoospore symmetry andthat of the colony produced. The changes involved in the transitionfrom a motile to a non-motile cell are considered, and appearto have features in common with this process in some other greenalgae. The early stages in wall deposition involve the simultaneousformation of two distinct layers and serve to stick togetherthe cells which by this time have collectively assumed the basicmorphological characteristics of the mature coenobia.  相似文献

The freshwater green algal family Hydrodictyaceae (Sphaeropleales, Chlorophyta) has traditionally consisted of four coenobial genera, Pediastrum Meyen 1829, Hydrodictyon Roth 1797, Sorastrum Kützing 1845, and Euastropsis Lagerheim1894. Two recent molecular phylogenetic studies demonstrated the need for reevaluation of the generic and species boundaries in this morphology‐rich family. This study expands the previous work to include phylogenetic analyses of 103 ingroup isolates representing North America, Europe, and Australia, with an emphasis on the common and geographically widespread species Pediastrum duplex. Nucleotide sequence data were collected from the nuclear LSU (26S rDNA) and the chloroplast RUBISCO LSU (rbcL) genes, totaling >3,000 aligned characters. The 26S and rbcL data sets were analyzed using maximum‐likelihood (ML) and Bayesian phylogenetic methods. In addition, SEM was used to examine the wall morphology of a majority of the isolates. The results supported previous indications that the P. duplex Meyen 1829 morphotype is nonmonophyletic and resolved some previously ambiguous relationships recovered in earlier phylogenetic estimations using fewer isolates. These new data allowed testing of the recent taxonomic revisions of the family that split Pediastrum into five genera. Some of the previous revisions by Buchheim et al. (2005) were well supported (erection of Stauridium and Monactinus), while others were not (Pediastrum, Pseudopediastrum, Parapediastrum).  相似文献

Variations in the levels of triglycerides, wax esters and polarlipids were determined in adults of the calanoid copepod Acanthodiaptomusdenticornis when freshly caught, starved or fed on the followingalgae: Anabaena spiroides, Cyclotella pseudostelligera and Pediastrumduplex. Over 7 days starvation, triglycerides and wax esterswere almost entirely used up by the copepods. Subsequent feedingover 20 days partially restored triglycerides but restored onlya relatively small fraction (<20%) of wax esters in the animals.Differences in the lipid restoration were found: the restoredtriglyceride level was higher in animals feeding on Cyclotellapseudostelligera or Pediastrum duplex than in those feedingon Anabaena spiroides. Fatty acid composition of neutral lipidswas closely linked to fatty acid composition of algae. The resultssuggest that lipid and fatty acid contents of Acanthodiaptomusdenticornis are good indices of the copepod's nutritional statusand short-term (0–20 days) feeding history.  相似文献

The morphologies of 15 rare chlorococcalean algae, i.e. three species of the Characiaceae [Hydrianum crassiapex Korshikov, H. viride (Scherffel) H. Ettl, Characiellopsis skujae (Fott) Komárek], four taxa of the genus Pediastrum Meyen [Pediastrum privum (Printz) E. Hegewald, P. duplex Meyen var. rugulosum Raciborski, P. angulosum (Ehrenberg) ex Meneghini, Pediastrum biradiatum Meyen], a radiococcalean alga Phacomyxa sphagnicola Skuja, Pseudodictyosphaerium fluviatile (Hindák) Hindák from the Dictyosphaeriaceae, three representatives of the family Oocystaceae (Amphikrikos minutissimus Korshikov, Gloeotaenium loitlesbergianum Hansgirg, Chlorolobion obtusum Korshikov) and three species of the genus Scenedesmus Meyen (S. ginzbergeri Kammerer, S. incrassatulus Bohlin, S. parisiensis Deflandre), are documented by drawings and micrographs and their taxonomy is discussed. Of these, Hydrianum viride, Pediastrum duplex var. rugulosum, Scenedesmus ginzbergeri and S. parisiensis are recorded for the first time in Slovakia. Chloroplast division in Nannochloris bacillaris Naumann (Chlorophyta) was examined by electron microscopy after preparation of samples by freeze-substitution. A pair of belts appeared on the surface of the outer and inner envelope membranes at the middle of the chloroplast. These belts seemed to be constructed of thin fibrils that run parallel to the longitudinal direction of the belts. The outer fibrillar belt increased in width as the constriction of the chloroplast advanced. It appears that the fibrillar belt is the division apparatus of the chloroplast. It encircles the chloroplast and finally divides the chloroplast in two as the diameter of the belt decreases.  相似文献

The dividing and the constricted regions of mitochondria ina green alga, Scenedesmus acutus , were studied by electronmicroscopy. Electron-dense substances were always visible inthese specific regions of mitochondria that had been fixed byfreeze-substitution. Thin fibres, which seemed to surround theconstricted regions, were also seen. Accurately defining species boundaries in the green algae (Chlorophyta) is integral for studies of biodiversity and conservation, water‐quality assessments, and the use of particular species as paleoindicators. Recent molecular phylogenetic and SEM analyses of the family Hydrodictyaceae (Chlorophyta) resolved three phylogenetic lineages of isolates with the Pediastrum duplex Meyen 1829 phenotype. The present study employed analyses of cell shape and cell wall ultrastructure to determine if the three lineages possessing the P. duplex morphotype were distinguishable. Only one of the groups, containing isolates with the P. duplex var. gracillimum West et G. S. West phenotype, was shown to be morphologically distinct from the other two P. duplex groups. The erection of a new genus, Lacunastrum, is proposed to recognize this group as a separate taxon.  相似文献

The amount of total chlorophyll, chlorophylls a and b as wellas the ratio of a to b decreased in chloroplasts isolated fromQuercus rotundifolia leaves, kept for 17 d in a solution of35.5 µM evernic acid in 1 mM Na HCO3, when compared withthe chloroplasts of control leaves (kept in NaHCO3). The chloroplastsin the spongy parenchyma were smaller and the amount of starchand plastoglobuli lower. The number of grana per chloroplastsection, the number of thylakoids per grana and the height ofgrana stacks were also less in the chloroplasts of leaves treatedwith evernic acid. Grown under saline conditions, Suaeda maritima accumulates Na+ and Cl- into its leaves, where individual mesophyll cells behave differently in their compartmentation of these ions. Measurements of ion concentrations within selected subcellular compartments show that freeze-substitution with dry sectioning is a valuable preparative technique for analytical electron microscopy of highly vacuolate plant material. Using this approach, absolute estimates were made of Na+, K+ and Cl- concentrations in the cytoplasm, cell walls, chloroplasts and vacuoles of leaf mesophyll cells.Abbreviation TAEM transmission analytical electron microscopy  相似文献

The basis for the resistance of certain algae to microbial decomposition in natural waters was investigated using Pediastrum duplex, Staurastrum sp., and Fischerella muscicola as test organisms. Enzyme preparations previously found to convert susceptible algae into spheroplasts had no such effect on the resistant species, although glucose and galacturonic acid were released from P. duplex walls. Little protein or lipid but considerable carbohydrate was found in the walls of the refractory organisms, but resistance was not correlated with the presence of a unique sugar monomer. A substance present in Staurastrum sp. walls was characterized as lignin or lignin-like on the basis of its extraction characteristics, infrared spectrum, pyrolysis pattern, and content of an aromatic building block. Sporopollenin was found in P. duplex, and cellulose in Staurastrum sp. Cell walls of the algae were fractionated, and the fractions least susceptible to microbial degradation were the sporopollenin of P. duplex, the polyaromatic component of Staurastrum sp., and two F. muscicola fractions containing several sugar monomers. The sporopollenin content of P. duplex, the content of lignin or a related constituent of Staurastrum sp., and the resistance of the algae to microbial attack increased with age. It is suggested that resistance results from the presence of sporopollenin in P. duplex, a lignin-like material in Staurastrum sp., and possibly heteropolysaccharides in F. muscicola.  相似文献

Ochna pulchra Hook. is a deciduous broad-leaved tree in theMixed Bushveld vegetation of the Northern Transvaal. The growthand development of leaves taken from trees in the field werestudied from a stage shortly before bud break, in late spring,until they were fully expanded and at the peak of photosyntheticactivity. Leaf area was measured by photographing the leaf against a transparentmm2 grid. Finally a constant relationship between leaf area(A) and the linear dimension of length (L) and breadth (B) wasestablished: A = b x LB, where coefficient b = 0.72. Transverse sections of the lamina of the youngest leaves showeda five-layered plate meristem with a few functional conductingelements in the midrib. During further leaf development, celldivision was followed by means of autoradiography using [3H]thymidine.It was most active during the week after bud break. Leaf cell increment following on cell division made the majorcontribution to leaf growth resulting in a lamina that was atleast 90% expanded 4 weeks after bud break. The histologicalchanges accompanying cell division were observed using lightand electron microscopy. Even in late stages of leaf development mature and differentiatingstomata occurred together, limited to the abaxial epidermisand the midrib. Scanning electron microscopy showed stomataldistribution, their increasing density and gradual opening.The structure of these sunken stomata could reduce the outwarddiffusion of water vapour and increase the diffusion resistanceto carbon dioxide. Carbon assimilation rates of the developing leaves were measuredusing an IRGA (infra-red gas analyser) and their chlorophyllvalues were calculated. Photosynthesis was first measured amonth after bud break when the leaves were fully expanded, over50 % of the stomata exposed and leaf mesophyll tissue differentiatedwith mature chloroplasts. Illus. 1963.-An electron microscopic study of cell wall development in P. birudiatum is described. Micrographs were taken of thin sections of cells from several stages involved in the transformation of the motile zoospore into the 4-pronged adult cell type during asexual reproduction. The cell wall begins as a thin membrane which does not change noticeably during the transformation of the zoospore to the adult cell type. In a subsequent period of not more than 6 hr, a definitive cell wall arises accompanied or followed shortly by the appearance of a globular network on the underside of the cell wall proper. During all of the developmental stages osmiophilic globuli are found in the cytoplasm, frequently at the cell surface. Similar, though smaller, globuli are found in the chloroplasts of Pediastrum and other plants, indicating that these bodies may have a plastid origin. It is suggested that whole osmiophilic globuli, or parts thereof, may be transferred to the cell wall proper, giving rise, ultimately, to the globular network.  相似文献

Plastid-dividing rings (PDs) are described for the first timein the ferns Ophioglossum, Gleichenia, Hymenophyllum, Trichomanes,Athyrium, Dryopteris, Ceratopteris and Pteridium. They are foundin the constricted isthmuses of undifferentiated plastids, amyloplasts,amylochloroplasts and chloroplasts in gametophytes and in arange of sporophytic tissues including apices, differentiatingleaf mesophyll and vascular parenchyma from roots, stems andleaves. Fern PDs comprise two concentric structures; a densegranular ring 60-100 nm wide adhering to the stromal face ofthe constriction and a smaller cytoplasmic annulus only 20-40nm wide. The diameters of the PD-containing constrictions, particularlythose in vascular parenchyma, where the outer component is usuallylacking, are extremely uniform (means of 177-188 nm in differentspecies and tissues) and closely in line with those in angiosperms.PDs in ferns differ from those previously described in othergroups of lands plants in that they are far more persistentand in multilobed plastids one is present at each constriction.The occurrence of multiple PDs in the giant plastids found inup to 30% of vascular parenchyma cells may be associated withorganelle fusion rather than division.Copyright 1993, 1999 AcademicPress Chloroplast division, division cycle, ferns, plastid-dividing rings, pteridophytes, transmission electron microscopy  相似文献

Wharton D. A. 1982. Observations on the coiled posture of trichostrongyle infective larvae using a freeze-substitution method and scanning electron microscopy. International Journal for Parasitology12: 335–343. A method for the preparation of the infective larvae of Trichostrongylus colubriformis for scanning electron microscopy is described. This involves freeze-substitution with methanol followed by critical-point drying. The method resulted in good preservation and the retention of the coiled posture. The morphology of ensheathed and exsheathed larvae is described and the mechanisms of coil formation discussed. Chloroplast division comprises a sequence of events that facilitatesymmetric binary fission and that involve prokaryotic-like stromaldivision factors such as tubulin-like GTPase FtsZ and the divisionsite regulator MinD. In Arabidopsis, a nuclear-encoded prokaryoticMinE homolog, AtMinE1, has been characterized in terms of itseffects on a dividing or terminal chloroplast state in a limitedseries of leaf tissues. However, the relationship between AtMinE1expression and chloroplast phenotype remains to be fully elucidated.Here, we demonstrate that a T-DNA insertion mutation in AtMinE1results in a severe inhibition of chloroplast division, producingmotile dots and short filaments of FtsZ. In AtMinE1 sense (overexpressor)plants, dividing chloroplasts possess either single or multipleFtsZ rings located at random intervals and showing constrictiondepth, mainly along the chloroplast polarity axis. The AtMinE1sense plants displayed equivalent chloroplast phenotypes toarc11, a loss-of-function mutant of AtMinD1 which forms replicatingmini-chloroplasts. Furthermore, a certain population of FtsZrings formed within developing chloroplasts failed to initiateor progress the membrane constriction of chloroplasts and consequentiallyto complete chloroplast fission in both AtMinE1 sense and arc11/atminD1plants. Our present data thus demonstrate that the chloroplastdivision site placement involves a balance between the opposingactivities of AtMinE1 and AtMinD1, which acts to prevent FtsZring formation anywhere outside of the mid-chloroplast. In addition,the imbalance caused by an AtMinE1 dominance causes multiple,non-synchronous division events at the single chloroplast level,as well as division arrest, which becomes apparent as the chloroplastsmature, in spite of the presence of FtsZ rings.  相似文献

Fourier transform infra-red (FTIR) spectroscopy was used to assess carbon allocation in selected phytoplankton (Pediastrum duplex, Ceratium hirundinella, Anabaena flos-aquae) in a eutrophic lake. In contrast to bulk-analysis techniques, FTIR spectroscopy can provide information on carbon allocation at the species level within natural mixed populations. Carbon allocation was determined using lipid/protein (L/P) and carbohydrate/protein (C/P) ratios and multivariate analysis of complete spectra and showed considerable intraspecific heterogeneity within samples dates, and clear seasonal changes in the spectra of Pediastrum and Anabaena, with both alga showing increased L/P and C/P ratios on selected sampling dates. Although increased ratios can indicate nutrient deficiency, the alga were not nutrient deficient during these periods, as indicated by continued population growth and high (>0.1%) internal phosphorus concentrations. FTIR spectra from Ceratium did not show any clear seasonal change. In all three algae, the transition from population growth to decline was not accompanied by any significant change in ratios, nor were low epilimnetic nutrients correlated with increased ratios, marking a significant contrast to laboratory batch cultures (previous studies) where low nutrients and the transition to stationary phase led to marked spectral changes.  相似文献

Chloroplast development was followed in intact bean leaves illuminatedwith far-red light by extracting chloroplasts at various timesto assay photosynthetic activities. Photochemical activity wasdetected in isolated chloroplasts prior to the times which werepreviously reported for intact leaf discs. Cyclic phosphorylationwas observed in isolated chloroplasts after 8 h of far-red illuminationwhile non-cyclic electron transport and phosphorylation weremeasurable after 12 and 16 h of illumination respectively. TheP/2e ratios were less than 0.5 after 24 h of far-red exposurebut approached a value of 1.0 by 60 h of illumination. Ammoniumchloride (10–3 M) had little effect on electron transportin isolated chloroplasts until after 24 h of far-red illumination.Chlorophyll a accumulated slowly from the onset of far-red illuminationwhile chlorophyll b was not detected until after 48 h of far-redexposure. Leaf fresh weight increased four-fold over the 60h illumination period. Electron microscopy of isolated chloroplasts from far-red-illuminatedleaves indicated the presence of unfused primary thylakoidsby 12 h of exposure and prolamellar bodies throughout the entire60 h illumination period. Grana were not observed in isolatedchloroplasts nor were they induced by a 2 min exposure of thechloroplasts to 172 000 lx of white light. O2 evolution in leaf discs of far-red-illuminated plants wasmeasurable after 16 h of illumination, attained a maximum valueby 36 h of far-red exposure, and then declined. Net CO2 fixationwas observed in leaf discs after 8 h of far-red illuminationand the rates remained constant for an additional 16 h, beforeincreasing at least two-fold.  相似文献
