Ultrastructural changes in chloroplasts of tobacco plants (Nicotiana tabacum L.) with the introduced desC gene for the acyl-lipid Δ9-desaturase from the thermophilic cyanobacterium Synechococcus vulcanus were investigated during plant acclimation to cold. Control plants were transformed with an empty pGA482 binary vector. At optimum growth temperature, a decreased number of grana and thylakoids and an increased number of plastoglobules and their larger area were observed in transgenic plants when compared to control ones. In control plants, acclimation to cold (6 days at 10°C) resulted in the larger areas of chloroplasts and grana. These changes indicated starting cold-induced injuries manifested in swelling of the stroma and a slight decrease in the total number of thylakoids in the chloroplast. In contrast, transgenic plants responded to cold by reducing the chloroplast, granal, and plastoglobule areas. Meantime, the number of thylakoids per granum increased noticeably. The total number of thylakoids in the chloroplast increased from 123 to 203. It was concluded that expression of the acyl-lipid Δ9-desaturase gene in tobacco plants provided for the formation of the cell ultrastructure similar to one characteristic of cold-tolerant plants.  相似文献

The root system of potato (Solanum tuberosum L. cv. Favorita) plants was treated with different O2 and CO2 concentrations for 35 d in aeroponic culture. Under 21 or 5 % O2 in the root zones, the thickness of leaves and palisade parenchyma significantly increased at 3 600 μmol(CO2) mol−1 in the root zone, compared with CO2 concentration 380 μmol mol−1 or low CO2 concentration (100 μmol mol−1). In addition, smaller cells of palisade tissue, more intercellular air spaces and partially two layers of palisade cells were observed in the leaves with root-zone CO2 enrichment. Furthermore, there was a significant increase in the size of chloroplasts and starch grains, and the number of starch grains per chloroplast due to elevated CO2 only under 21 % O2. In addition, a significant decline in the thickness of grana and the number of lamellas, but no significant differences in the number of grana per chloroplast were observed under elevated CO2 concentration. The accumulation of starch grains in the chloroplast under elevated CO2 concentration could change the arrangement of grana thylakoids and consequently inhibited the absorption of sun radiation and photosynthesis of potato plants.  相似文献

Effects of the desA gene from the cyanobacterium Synechocystis sp. encoding Δ12 acyl-lipid desaturase and increasing the level of unsaturated fatty acids (linoleic acid (18:2) primarily) in membrane lipids, which was inserted into potato (Solanum tuberosum L., cv. Desnitsa) plants, on chloroplast ultrastructure and plant tolerance to low temperatures were studied. The main attention was focused on modifications in the chloroplast structure and their possible relation to potato plant tolerance to oxidative and low-temperature stresses under the influence to their transformation with the Δ12 acyl-lipid desaturase gene from cyanobacterium (desA-licBM3-plants). Morphometric analysis showed that, in comparison with wild-type (WT) plants, in desA-licBM3-plants the number of grana in chloroplasts increased substantially. The total number of thylakoids in transformant chloroplasts was almost twice higher than in WT plants. The number of plastoglobules per chloroplast of transformed plants increased by 25%. A marked increase in the number of grana, total number of thylakoids, and the number of plastoglobules in chloroplasts of desA-licBM3-plants indicates their more intense lipid metabolism, as compared with WT plants, and this resulted in the conservation of some part of lipids in plastoglobules. In addition, the expression of heterological desA gene encoding Δ12 acyl-lipid desaturase positively influenced stabilization of not only structure but also functioning of chloroplast membranes, thus preventing a transfer of electrons from the ETR to oxygen and subsequent ROS generation at hypothermia. This was confirmed by the analysis of the rate of superoxide anion generation in tested genotypes.  相似文献

The changes in ultrastructure of leaf cell in waterlogged Kosteletzkya virginica seedlings were observed by transmission electron microscope. During waterlogging, the chloroplasts turned into a round shape and their volume was gradually reduced. Furthermore, the lamellae of thylakoids swelled, and the inclusions in chloroplast decreased. The shape of starch grains slightly changed, but their number and volume decreased, and they even disappeared in the end. On the other hand, plastoglobules enlarged and their amount enhanced gradually. The membrane and inner cristea of mitochondria gradually became unclear, while the mitochondria firstly enlarged but disappeared at the end. Similar to chloroplast, cell nucleus dwindled gradually, concentrated and tended to a round shape. Additionally, the annulate lamellae and multivesicular body occurred on the 20th day of experiment, and the cell wall distorted and twisted at the late stage of waterlogging. Consequently, these changes of organelles are the typical characteristics of the complete cell disintegration or death for Kosteletzkya virginica under long-term waterlogging.  相似文献

The effects of starch accumulation on photosynthesis and chloroplastultrastructure were studied in primary leaves of bean (Phaseolusvulgaris L. cv. Bulgarian). De-topping the shoot above the primaryleaf node, caused over an 8-day period, a considerable increasein the photosynthetic activity of the primary leaves, despitethe fact that a large quantity of starch had accumulated intheir chloroplasts. The accumulation of starch was greater inthe chloroplasts of spongy cells in comparison with that ofthe palisade cells. Initiation of starch grains was observedmainly in the peripheral part of the chloroplast, distant fromthe cell wall. As a result, most of the starch was accumulatedclose to the inner part of the cell, leaving a considerablemass of the chloroplast near the cell wall free of starch. Theaccumulation of starch was accompanied by the destruction, deformationand disorientation of grana and thylakoids. It is concludedthat the accumulation of starch is not inevitably a limitingfactor in photosynthesis and the results cast doubt on the hypothesisthat starch accumulation or dissipation is the main factor involvedin the regulation of photosynthesis. Plants of the Podostemoideae, a subfamily of the unique aquatic angiosperm family Podostemaceae, which are found in rapids and waterfalls of the tropics and subtropics, have two different sizes of chloroplasts in their epidermis. These small and large chloroplasts are located separately in each epidermal cell along its upper and inner tangential walls, respectively. This is the first case of the chloroplast dimorphism in a single epidermal cell of angiosperms. While the large chloroplasts have well developed starch grains, the small chloroplasts have a normal granal ultrastructure but very few starch grains. This suggests that the small chloroplasts mainly function in CO2 uptake for photosynthesis from torrential water.  相似文献

The effects of salinity (300 mM NaCl), putrescine (Put), and the combination of two agents on the structure of chloroplasts and storage deposits were studied in the third leaf pair of a facultative halophyte Mesembryanthemum crystallinum. Within 6 days, the common ice plants responded to NaCl and Put treatments by diminished chloroplast volumes and swollen grana. Different effects of the experimental treatments were primarily manifested in the chloroplast storage inclusions. Under the salinity conditions, the starch content dropped down almost threefold as compared to untreated plants (control), whereas the number of plastoglobules did not change. Put and Put + NaCl treatments further decreased the starch content per unit section area; in contrast, the plastoglobule area per chloroplast section increased eightfold and tenfold in Put and Put + NaCl treatments, respectively. The morphology and electronic density of plastoglobules changed in all treatments. In both Put treatments there ware no destructive changes in the chloroplasts, and therefore the authors presume that the increase in the numbers plastoglobules was related to the redirection of cell metabolism towards the products of the higher reduction potential. The ferritin deposits in the chloroplasts were observed in all treatments they were more abundant in the vascular parenchyma cells, especially under salinity. The ability of the common ice plants to accumulate large Fe quantities in their chloroplasts and the characteristic pectin-filled pockets, which were observed earlier, and intercellular spaces are probably related to the genetically determined traits of plant adaptation to salinity and water deficit.  相似文献

It has been reported in quite a number of literatures that doubled CO2 concentration increased the photosynthetic rate and dry matter production of C3 plants, but substantially affected C4 plants little. However, why may CO2 enrichment promote growth and either no change or decrease reproductive allocation of the C3 species, but havinag no effects on growth characteristics of the C4 plants? So far, there has been no satisfactory explanation on that mentioned above, except the differences in their CO2 compensatory points. In the past, although some studies on ultrastructure of the chloroplasts under doubled CO2 concentration were limitedly conducted. Almost all the relevant experimental materials were only from C3 plants not from C4 plants, and even though the results were of inconsistancy. Thereby, it needs to verify whether the differences in photosynthesis of C3 and C4 plants at doubled CO2 level is caused by the difference in their chloroplast deterioration. Experiments to this subject were conducted at the Botanical Garden of Institute of Botany, Academia Sinica in 1993 and 1994. Both experimental materials from C3 plant alfalfa (Medicago sativa) and C4 plant foxtail millet (Setaria italica) were cultivated in the cylindrical open-top chambers (2.2 m in diameter × 2.4 m in height) with aluminum frames covered by polyethylene film. Natural air or air with 350× 10-6 CO2 were blown from the bottom of the chamber space with constant temperature between inside and outside of the chamber 〈0.2℃〉. Electron microscopic observation revealed that the ultrastructure of the chloroplasts from C3 plant Medicago sativa and C4 plant Seteria italica growing under the same doubled CO2 concentration were quite different from each other. The differential characteristics in ultrastructure of chloro plasts displayed mainly in the configuration of thylakoid membrances and the accumulation of starch grains. They were as follows: 1. The most striking feature was the building up of starch grains in the chloroplasts of the bundle sheath cells (BSCs) and the mesophyll cells (MCs) at doubled CO2 concentra tion. The starch grains appeared centrifugally first in the BSCs and then in the chloroplast of the other MCs. It was worthy to note that the starch grains in the chloroplasts of C4 plant Setaria ira/ica were much more than those of the C3 plant Medicago sativa . The decline of photosynthesis in the doubled CO2-grown C4 plants might be caused by an over accumulation of starch grains, that deformed the chloroplast even demaged the stroma thylakoids and grana. There might exsist a correlation between the comformation of thylakoid system and starch grain accumulation, namely conversion and transfer of starch need energy from ATP, and coupling factor (CF) for ATP formation distributed mainly on protoplastic surface (PSu) of stroma thylakoid membranes, as well as end and margin membranes of grana thylakoids. Thereby, these results could provide a conclusive evidence for the reason of non effectiveness on growth characteristics of C4 plant. 2. Under normal condition , the mature chlolroplats of higher plants usually develop complete and regularly arranged photosynthetic membrane systems . Chloroplasts from the C4 plant Setaria italica, however, exerted significant changes on stacking degree, grana width and stroma thylakoid length under doubled CO2 concentration; In these changes, the grana stacks were smaller and more numerous, and the number of thylakoids per granum was greatly increased, and the stroma thylakoid was greatly lengthened as compared to those of the control chloroplasts. But the grana were mutually intertwined by stroma thylakoid. The integrity of some of the grana were damaged due to the augmentation of the intrathylakoid space . Similarly, the stroma thylakoids were also expanded. In case. the plant was seriously effected by doubled CO2 concentration as observed in C4 plant Setaria italica , its chloroplasts contained merely the stroma (matrix) with abundant starch grains, while grana and stroma thylakoid membranes were unrecognizable, or occasionally a few residuous pieces of thylakoid membranes could be visualized, leaving a situation which appeared likely to be chloroplast deterioration. However, under the same condition the C3 plant Medicago sativa possessed normally developed chloroplasts, with intact grana and stroma thylakoid membranes. Its chloroplasts contained grana intertwined with stroma thylakoid membranes, and increased in stacking degree and granum width, in spite of more accumulated starch grains within the chloroplasts. These configuration changes of the thylakoid system were in consistant with the results of the authors another study on chloroplast function, viz. the increased capacity of chloroplasts for light absorption and efficiency of PSⅡ.  相似文献

Tobacco (Nicotiana tabacum L.) cells growing heterotrophically in the light on supplied sucrose (S0) have previously been adapted to grow in 428 mM NaCl (S25). Among the changes occurring in salinity-adapted cell cultures are (a) elevated levels of chlorophyll compared to unadapted cells; (b) decreased levels of starch; (c) alterations in chloroplast ultrastructure, including loss of starch grains, increased thylakoid membrane structure, and the presence of plastoglobules; and (d) increased rates of O2 evolution, CO2 fixation, and photophosphorylation relative to S0 cells. These latter changes apparently derive from the fact that thylakoid membranes in S25 cells contain higher levels of photosystem I- and II-associated proteins as well as thylakoid ATPase components. S25 chloroplasts contain immunologically detectable levels of ribulose-1,5-bisphosphate carboxylase/oxygenase, whereas S0 completely lack the enzyme. These changes taken together suggest that even in the presence of sucrose, S25 cells have acquired a significant degree of salt-tolerant photosynthetic competence. This salt-tolerant photoysynthetic capability manifests itself in plants backcrossed with normal plants for three generations. These plants contain chloroplasts that demonstrate in vitro more salt-tolerant CO2 fixation, O2 evolution, and photophosphorylation than do backcross progeny of plants regenerated from S0 cultures.  相似文献

Leaf samples of Mn-deficient and Mn-sufficient (control) ‘Navelate’ orange plants grown in a greenhouse were taken to investigate the effects of Mn deficiency in leaf structure and chloroplast ultrastructure. Total leaf chlorophyll concentration was significantly lower in Mn-deficient plants than in control ones. Entire lamina thickness was not altered due to Mn deficiency. However, Mn deficiency resulted in disorganization of mesophyll cells, mainly of palisade parenchyma cells. The number of mesophyll chloroplasts per cellular area and their length were both affected negatively. The membranous system of chloroplasts was also disorganized. The percentages of starch grains and plastoglobuli per chloroplast of Mn-deficient leaves were significantly greater than those of control leaves.  相似文献

Plant growth, photosynthetic parameters, chloroplast ultrastructure, and the ascorbate-glutathione cycle system in chloroplasts of self-grafted and rootstock-grafted cucumber leaves were investigated. Grafted plants were grown hydroponically and were exposed to 0, 50, and 100 mM NaCl concentrations for 10 days. Under NaCl stress, the hydrogen peroxide (H2O2) content in cucumber chloroplasts increased, the chloroplast ultrastructure was damaged, and the gas stomatal conductance, intercellular CO2 concentration, as well as shoot dry weight, plant height, stem diameter, leaf area, and leaf relative water content were inhibited, whereas these changes were less severe in rootstock-grafted plants. The activities of ascorbate peroxidase (APX; EC 1.11.1.11), glutathione reductase (GR; EC 1.6.4.2), and dehydroascorbate reductase (DHAR EC 1.8.5.1) were higher in the chloroplasts of rootstock-grafted plants compared with those of self-grafted plants under 50 and 100 mM NaCl. Similar trends were shown in leaf net CO2 assimilation rate and transpiration rate, as well as reduced glutathione content under 100 mM NaCl. Results suggest that rootstock grafting enhances the H2O2-scavenging capacity of the ascorbate–glutathione cycle in cucumber chloroplasts under NaCl stress, thereby protecting the chloroplast structure and improving the photosynthetic performance of cucumber leaves. As a result, cucumber growth is promoted.  相似文献

Utilization of storage starch in the cells of cotyledon mesophyll and root meristem in the course of alfalfa (Medicago sativa L.) seed germination on the solutions of NaCl, Na2SO4, and mannitol at different concentrations and identical osmotic pressure was investigated using the method of transmission electron microscopy. Ultrastructural analysis showed changes in the number of starch grains and deceleration of chloroplast development depending on the osmotic component of salt influence. At low concentrations corresponding to osmotic pressure of 202.6 kPa, Na2SO4 did not affect the formation of the photosynthetic machinery and utilization of starch inclusions; mannitol contributed to the preservation of considerable reserve of starch without disturbing the development of chloroplasts; NaCl did not inhibit the development of the photosynthetic machinery and induced an increase in the number of starch grains presumably at the expense of newly produced starch. When the concentration of the investigated substances increased up to the values corresponding to the osmotic pressure of 607.8 kPa, NaCl did not suppress transformation of amyloplasts into chloroplasts and utilization of starch; Na2SO4 inhibited the development of chloroplasts and starch utilization; mannitol decelerated transformation of amyloplasts and inhibited mobilization of starch grains. The obtained results make it possible to propose a method of preliminary estimation of tolerance of dicotyledons to abiotic stresses based on the cytological analysis of utilization of starch grains and formation of photosynthetic compartments of chloroplasts in the mesophyll of cotyledons.  相似文献

Photosynthetic activity, the content of various photosynthetic pigments, and the chloroplast ultrastructure were examined in the leaves of cucumber (Cucumis sativus L.) and pea (Pisum sativum L.) plants of different ages grown under red light (600–700 nm, 100 W/m2). In pea leaves tolerant to red-light irradiation, chloroplast ultrastructure did not essentially change. In the first true leaves of cucumber plants susceptible to red-light irradiation, we observed a considerable increase in the number and size of plastoglobules, the appearance of chloroplasts lacking grana or containing only infrequent grana, and stromal thylakoids. In the upper leaves of 22-day-old cucumber plants, the chloroplast structure was essentially similar to that of the control chloroplasts in white light, and we therefore suppose that these plants have acclimated to red light.  相似文献

The chloroplast is one of the most important organelles in plants. Proteomic investigations of chloroplasts have been undertaken for many herb plant species, but to date no such investigation has been reported for woody plant chloroplasts. In the present study we initiated a systematic proteomic study of Populus chloroplasts using a shotgun proteomic method. After isolation of chloroplasts and tryptic digestion of the proteins, the protein fragments were separated via HPLC using an SCX column, and the peptides were analyzed by LC-MS/MS; 119 proteins were successfully identified. Based on annotation information in the UniProtKB/Swiss-Prot database, these proteins were identified as being localized in the chloroplast thylakoid membrane, chloroplast stroma, chloroplast thylakoid lumen, and plastoglobules. Over 50% of all identified proteins were confirmed as chloroplast thylakoid proteins, and 85 are encoded by the chloroplast genome with the remaining proteins encoded by the nuclear genome. Based on functional annotation, these proteins were classified into four functional categories, including photosynthesis, redox regulation and stress, primary and secondary metabolism, transport and signaling. These data provide a valuable basis for further studies on photosynthesis in poplar species.  相似文献

Exposure of plants to chilling (low temperatures above freezing) limits growth and development in all environments outside the lowest latitudes. Cell ultrastructure and morphometric studies may allow associations to be made between chilling-induced changes at the ultrastructural level, molecular events and their physiological consequences. We examined changes in the shape, size and membrane organization of the organelles of mesophyll cells in Arabidopsis thaliana (Col 0), a cold-resistant species, after subjecting 6-week-old plants grown at normal growth temperatures to chilling (2.5–4°C; 14-h dark/10-h light cycle) for 6, 24 and 72 h and after a re-warming period of 50 h. No ultrastructural differences were seen in the first 6 h of chilling but after 24 h we observed swollen and rounded chloroplasts with larger starch grains and dilated thylakoids compared to control plants. By 72 h, chilling had resulted in a large accumulation of starch in chloroplasts, an apparent crowding of the cytosol and a lower abundance of peripheral reticulum than in the controls. The average area per chloroplast in cell sections increased after 72-h chilling while the number of chloroplasts remained the same. Ring-shaped and other morphologically aberrant mitochondria were present in significantly higher abundance in plants given 72 h chilling than in the controls. Plant re-warming for 50 h reduced chloroplast size to those of the controls and returned mitochondria to standard morphology, but peripheral reticulum remained less abundant than in plants never given a cold treatment. The near full return to normal ultrastructure upon plant re-warming indicates that the morphological changes may be part of acclimation to cold.  相似文献

The chloroplast disintegration during zygote maturation inSpirogyra verruculosa was investigated by electron microscopy. In the seven-day-old zygote about half of the chloroplasts commenced to disintegrate and to turn yellow, losing starch grains, and, then, were torn into fragments of various sizes, which had mostly vesiculated thylakoids and plastoglobules increasing in both size and number. At about two weeks after conjugation, in the cytoplasm, electron-dense structures, linear in section, appeared and vacuoles of various sizes developed. Each of the dense linear structures lying around a fragment seemed to form a cavity of crescent shape in section, and these cavities fused mutually into a large one, leading to the separation of the fragment from the bulk of cytoplasm. The vacuoles seemed to be, involved in the sequestration of the fragments by their fusion with the cavities and by the invagination, of tonoplast. The fragments entrapped by the vacuoles were rapidly broken down into the aggregation of residual membrane pieces, plastoglobules, and undigested starch grains. The maintained chloroplasts changed little in structure compared with the chloroplast of the vegetative cell, and were transmitted to the germling. It is suggested that the eliminated chloroplasts are derived exclusively from the male gamete.  相似文献

When following low-temperature acclimation (5 days at 2°C) of cold-resistant plants of Arabidopsis (Arabidopsis thaliana Heynh. (L.), ecotype Columbia) in relation to the changes in chloroplast ultrastructure, we registered the high efficiency of hardening and the ability of hardened plants to lower a threshold of frost damage by about 3°C. During hardening, the area of grana in the chloroplasts more than doubled, with considerably increased numbers of thylakoids per granum and thylakoids per chloroplast. The rate of apparent photosynthesis decreased to lesser extent than the rate of dark respiration, as a result the content of soluble sugars increased fourfold, ensuring an adaptive reorganization of metabolism, which enabled the hardened plants to survive even at below-zero temperatures (up to ?7°C). The authors conclude that a considerable increase in the number of thylakoids in the chloroplasts helps maintain photosynthesis at low above-zero temperatures and is a prerequisite for the accumulation of soluble sugars in Arabidopsis leaves.  相似文献

This study examines the effects of doubled CO2 concentration on the ultrastructure and function of chloroplasts from cycads and, for control from two other herbaceous angiosperms. Under a doubled CO2 concentration condition, the chloroplast ultrastructure of the two cycads (Cycas multipinnata with a shade-type chloroplast andC. panzhihuaensis with a sun-type chloroplast) changed little: The conformation of the thylakoid membrane system kept well, and almost no starch grains accumulated. In contrast, under the same conditions the chloroplast ultrastructure of soybean and foxtail millet changed considerably, with starch grains accumulating in their chloroplasts and some of thylakoids (especially stroma thylakoid) membranes being destroyed to some degree by the more numerous and larger starch grains that accumulated in the chloroplasts. Interestingly, the changes in the ultrastructure of the chloroplasts from the two cycads was correlated with the 77K fluorescence emission spectra of their chlorophyll; i.e., the F685/F734 (PS II / PS I) ratio within the chloroplasts, which were minimal. The absorption spectrum showed decreases in the red and blue peaks. These changes in the absorption spectrum may be related to changes in the structural arrangement of the thylakoid membranes. Preliminarily, this experimental result shows that the cycads may adapt themselves to environmental changes under doubled CO2 concentration in the coming centuries. However, more studies on this aspect are necessary.  相似文献

Aims In recent years, intercropping system has become one of the major practice of peanut (Arachis hypogaea) cultivation in northern China because of the high land and energy utilization efficiency, to some extent compensating for the production loss caused by decreasing area of cultivation land. Intercropped peanut plants often have a lower pod yield compared with monoculture due to constraint on light availability. This study was conducted to explore the shade-tolerance mechanism in two peanut cultivars, ‘Huayu 22’ and ‘Baisha 1016’, that grew in an intercropping system, by studying chloroplast ultrastructure and rubisco activity under different levels of shading.
Methods A field experiment was conducted with three levels of light treatments, including full natural light (CK), 50% natural light indensity (NLI), and 15% NLI. The ‘Huayu 22’ was used as a shade-tolerant cultivar and the ‘Baisha 1016’ as a shade-susceptible cultivar based on previous studies. Experimental plants of both cultivars were shaded for 40 days from emergency in 2006. Rubisco activity, the number and shapes of chloroplasts and starch grains, and number of grana and granum lamella were investigated in functional leaves of plants in all treatments.
Important findings The functional leaves of peanut plants in the 50% and 15% NLI treatments had a lower rubisco activity than those in the CK treatment. In the ‘Baisha 1016’, the reduction in rubisco activity was 40.1% in the 50% NLI treatment and 59.4% in the 15% NLI treatment, respectively, compared to the CK treatment;whereas no significant differences were found among treatments in the ‘Huayu 22’ in the rubisco activity. Compared with the CK, the number of chloroplasts remained unchanged, the number of grana and lamella in grana increased, and the individual chloroplast was longer and in perfect development in the functional leaves of plants of the ‘Huayu 22’ grown in the 50% NLI treatment. In contrast, the number of chloroplasts, grana and starch grains of the ‘Huayu 22’ plants decreased significantly, the chloroplast membrane and grana lamella were damaged, the number of granum lamella increased, and the individual chloroplast became longer in the 15% NLI treatment. The number and ultrastructure of chloroplasts in the ‘Baisha 1016’ plants followed similar patterns of changes as those of the ‘Huayu 22’ in the 50% NLI treatment. For plants of the ‘Baisha 1016’ in the 15% NLI treatment, their chloroplasts became more roundly shaped, with decreasing number of grana lamella and increasing number of starch grains, compared with the CK. There were a greater decrease in the grana number and more damage in the grana lamella in plants of the ‘Baisha 1016’ than those of the ‘Huayu 22’. In conclusion, the shade tolerance of the ‘Huayu 22’ resulted from lack of changes in rubisco activity and less damage in the ultrastructure of chloroplasts when under low light stress compared with the ‘Baisha 1016’.  相似文献

Ultrastructural investigations of cells and organelles by transmission electron microscopy (TEM) usually lead to two-dimensional information of cell structures without supplying exact quantitative data due to the limited number of investigated ultrathin sections. This can lead to misinterpretation of observed structures especially in context of their three-dimensional (3D) assembly. 3D investigations and quantitative morphometric analysis are therefore essential to get detailed information about the arrangement and the amount of subcellular structures inside a cell or organelle, respectively, especially when the plant sample was exposed to environmental stress. In the present research, serial sectioned chloroplasts, mitochondria, and peroxisomes from first year spruce needles (Picea abies (L.) Karst.) were 3D reconstructed and digitally measured using a computer-supported image analysis system in order to obtain a detailed quantitative characterization of complete cell organelles including precise morphological data of drought-induced fine structural changes. In control plants, chloroplast volume was composed of 56% stroma, 15% starch, 27% thylakoids, and 2% plastoglobules. In drought-stressed chloroplasts, the relative volume of both the thylakoids and the plastoglobules significantly increased to 37% and 12%, respectively. Chloroplasts of stressed plants differed from control plants not only in the mean thylakoid and plastoglobules content but also in the complete lack of starch grains. Mitochondria occurred in variable forms in both control and stressed samples. In stressed plants, mitochondria showed a significant smaller mean volume which was only 81% when compared with the control organelles. Peroxisomes were inconspicuous in both samples and their volume did not differ between control and drought-stressed samples. The present study shows that specific subcellular structures are subject to significant quantitative changes during drought stress of spruce needles giving a detailed insight in adaptation processes of the investigated cell organelles.  相似文献
