The clinical picture produced by the feeding of larvae of Ornithodoros aff. puertoricensis on laboratory mice, was studied using different larval infestation protocols that included 30, 40 or 50 larvae per mouse and control uninfested groups. Clinical effects appeared around 72 h of larval feeding, having a first stage characterized by hyperaemia in both nasal and ocular mucosa, followed by respiratory symptoms (96–120 h) and nervous incoordination (120–144 h). No one mouse evidenced paralysis, and nervous symptoms were never observed in animals infested with only 30 larvae. High mortality (commonly up to 70%) was observed in mice with respiratory symptoms, while 100% of animals in the nervous phase died between 168 and 192 h after the beginning of larval feeding. When some infested mice were treated with a solution of Amitraz the larvae were killed and reversion of symptoms was observed. These effects are ascribed to the presence of a toxin in the saliva of the feeding larvae.  相似文献

The susceptibility of larvae of the leek moth, Acrolepiopsis assectella Zeller (Lepidoptera: Acrolepiidae) to different concentrations of an autochthonous strain of Steinernema feltiae (Rhabditida: Steinernematidae) was examined in laboratory experiments using Petri dishes. The efficacy of this strain in pots and field experiments was also evaluated. High mortality (80%–100%) of leek moth larvae was observed when these larvae were exposed to low concentrations (3 × 103 to 1 × 104 IJs/m2) of S. feltiae under laboratory conditions. Foliar application of 30,000 IJs/leek in pot experiments caused a 98% reduction in leek moth larvae. Field experiments showed a 87.7% reduction of leek moth larvae with the nematode treatment, significantly higher than the 22% reduction with the Bacillus thuringiensis treatment. The efficacy of the treatments with S. feltiae in relation to the microhabitat of the leek moth larvae between the interfolded leaves of the leek is discussed.  相似文献

Sexual reproduction of Rhopaloeides odorabile, a common Great Barrier Reef sponge, was quantified using histological sections from samples collected over two reproductive seasons. Rhopaloeides odorabile is viviparous and gonochoristic with a male biased sex ratio (2.5:1). Commencement and cessation of gametogenesis coincides with rising and falling sea surface temperatures (≈24–29°C). Spermatogenesis occurs from October until January. Females initiate oogenesis in September with the asynchronous development of oocytes, embryos and larvae occurring within the brood chambers. A larval release period of 5–6 weeks occurs during January and February. The minimum size of reproductive individuals was 176 cm3 for females and 192 cm3 for males. The total reproductive output for both sexes shows a positive correlation with size. A relative reproductive output index quantified the maximum reproductive investment at approximately 1 and 3% of the total choanoderm for females and males, respectively, which represents a lower range of reproduction in comparison to other viviparous sponges. Low reproductive output in R. odorabile may be offset by an extended spawning period, alleviating the risk of releasing larvae in potentially adverse conditions via one synchronised spawning event and increasing overall larval survival.  相似文献

An increasing number of studies reveal that ticks and their hosts are infected with multiple pathogens, suggesting that coinfection might be frequent for both vectors and wild reservoir hosts. Whereas the examination of associations between coinfecting pathogen agents in natural host–vector–pathogen systems is a prerequisite for a better understanding of disease maintenance and transmission, the associations between pathogens within vectors or hosts are seldom explicitly examined. We examined the prevalence of pathogen agents and the patterns of associations between them under natural conditions, using a previously unexamined host–vector–pathogen system—green lizards Lacerta viridis, hard ticks Ixodes ricinus, and Borrelia, Anaplasma, and Rickettsia pathogens. We found that immature ticks infesting a temperate lizard species in Central Europe were infected with multiple pathogens. Considering I. ricinus nymphs and larvae, the prevalence of Anaplasma, Borrelia, and Rickettsia was 13.1% and 8.7%, 12.8% and 1.3%, and 4.5% and 2.7%, respectively. The patterns of pathogen prevalence and observed coinfection rates suggest that the risk of tick infection with one pathogen is not independent of other pathogens. Our results indicate that Anaplasma can play a role in suppressing the transmission of Borrelia to tick vectors. Overall, however, positive effects of Borrelia on Anaplasma seem to prevail as judged by higher-than-expected BorreliaAnaplasma coinfection rates.  相似文献

The tomato leafminer, Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) is an important pest of tomato crops in South America and it has recently arrived in Europe affecting tomato plantations. The susceptibility of T. absoluta larvae and pupae to three species of entomopathogenic nematodes (Steinernema carpocapsae, Steinernema feltiae and Heterorhabditis bacteriophora) was examined under laboratory conditions. Leaf bioassays were conducted to evaluate the nematode’s capability to reach the larvae and kill them within the galleries. The efficacy of the three nematode species after foliar application to potted tomato plants was evaluated under greenhouse conditions. High larval mortality (78.6–100%) and low pupae mortality (<10%) was determined in laboratory experiments. In the leaf bioassay a high level of larval parasitation (77.1–91.7%) was recorded revealing the nematode’s capacity to kill the larvae inside the galleries. In the pot experiments nematode treatment reduced insect infection of tomato plants by 87–95%. The results demonstrate the suitability of entomopathogenic nematodes for controlling T. absoluta.  相似文献

 A marine fungus was isolated from the black tiger prawn Penaeus monodon at Nha Trang, Vietnam, on March 20, 2001 and named isolate NJM 0131. The fungus was identified as Haliphthoros milfordensis from the characteristics of asexual reproduction, and its physiological characteristics were investigated. Although the optimum temperature for growth of the isolate was 25°–30°C, the fungus grew at a wide range of temperatures (15°–40°C). H. milfordensis grew well in 50%–100% seawater, but poorly in PYG agar containing 1.0%–5.0% NaCl and KCl. The fungus grew at a wide range of pH (4.0–11.0) with the optimum pH value of 7.0–9.0. The isolate also showed pathogenicity to swimming crab larvae (Portunus trituberculatus) by artificial infection, but mortality was not high. The toxicity of para-menthane-3,8-diol (PMD), the main arthropod-repellent compound in the oil of the lemon eucalyptus, Corymbia citriodora, was evaluated against nymphs of Ixodes ricinus using five methods (A–E) of a contact toxicity bioassay. Mortality rates were estimated by recording numbers of dead nymphs at 30 min intervals during the first 5 h after the start of exposure and at longer intervals thereafter. The mortality rate increased with increasing concentration of PMD and duration of exposure with a distinct effect after 3.5 h. From the results obtained by methods A, C and E, the LC50 range was 0.035–0.037 mg PMD/cm2 and the LC95 range was 0.095–0.097 mg PMD/cm2 at 4 h of exposure; the LT50 range was 2.1–2.8 h and the LT95 range was 3.9–4.2 h at 0.1 mg PMD/cm2. To determine the duration of toxic activity of PMD, different concentrations (0.002, 0.01, 0.1 mg PMD/cm2) were tested and mortality was recorded at each concentration after 1 h; thereafter new ticks were tested. This test revealed that the lethal activity of PMD remained for 24 h but appeared absent after 48 h. The overall results show that PMD is toxic to nymphs of I. ricinus and may be useful for tick control.  相似文献

Seventeen isolates of Metarhizium anisopliae (Metschnikoff) Sorokin and two isolates of Beauveria bassiana (Balsamo) Vuillemin were evaluated for their pathogenicity against the tobacco spider mite, Tetranychus evansi Baker & Pritchard. In the laboratory all the fungal isolates were pathogenic to the adult female mites, causing mortality between 22.1 and 82.6%. Isolates causing more than 70% mortality were subjected to dose–response mortality bioassays. The lethal concentration causing 50% mortality (LC50) values ranged between 0.7×107 and 2.5×107 conidia ml−1. The lethal time to 50% mortality (LT50) values of the most active isolates of B. bassiana and M. anisopliae strains varied between 4.6 and 5.8 days. Potted tomato plants were artificially infested with T. evansi and treated with B. bassiana isolate GPK and M. anisopliae isolate ICIPE78. Both fungal isolates reduced the population density of mites as compared to untreated controls. However, conidia formulated in oil outperformed the ones formulated in water. This study demonstrates the prospects of pathogenic fungi for the management of T. evansi.  相似文献

The legume pod borer Maruca vitrata Fabricius (Lepidoptera: Crambidae) is a serious pest of cowpea in West-Africa. The parasitoid Apanteles taragamae Viereck (Hymenoptera: Braconidae) that originates from Taiwan is a potential candidate for biological control of M. vitrata. We investigated under laboratory conditions the functional response of the parasitoid by offering each experienced female 10, 20, 30 and 40 larvae of M. vitrata. We studied the influence of different host larval ages on the development, longevity, sex ratio, lifetime fecundity and parasitization rate of the wasp. In a comparative study, we also investigated the life history of A. taragamae and M. vitrata at different temperatures in the range of 20–30°C. The parasitoid successfully parasitized two- and three-day-old host larvae (first and second instars). Younger larvae (one-day-old) were parasitized to a lesser extent, and only males developed in them. Older larvae were not parasitized, partly because of defensive host behaviour. The success of parasitization was positively correlated with the density of two-day-old M. vitrata larvae. Parasitoid developmental time and longevity decreased with increasing temperature. The intrinsic rate of population increase (r m ) exhibited an optimum curve with a maximum at 24–28°C. For the host M. vitrata, r m was maximal at temperatures of 26–30°C. The data are discussed in the context of the potential of A. taragamae for biological control of M. vitrata.  相似文献

A micropropagation protocol through multiple shoot formation was developed for Thlaspi caerulescens L., one of the most important heavy metals hyperaccumulator plants. In vitro seed-derived young seedlings were used for the initiation of multiple shoots on Murashige and Skoog (MS) medium with combinations of benzylaminopurine (BA; 0.5–1.0 mg dm−3), naphthaleneacetic acid (NAA; 0–0.2 mg dm−3), gibberellic acid (GA3; 0–1.0 mg dm−3) and riboflavin (0–3.0 mg dm−3). The maximum number of shoots was developed on medium containing 1.0 mg dm−3 BA and 0.2 mg dm−3 NAA. GA3 (0.5 mg dm−3) in combination with BA significantly increased shoot length. In view of shoot numbers, shoot length and further rooting rate, the best combination was 1.0 mg dm−3 BA + 0.5 mg dm−3 GA3 + 1.0 mg dm−3 riboflavin. Well-developed shoots (35–50 mm) were successfully rooted at approximately 95 % on MS medium containing 20 g dm−3 sucrose, 8 g dm−3 agar and 1.0 mg dm−3 indolebutyric acid. Almost all in vitro plantlets survived when transferred to pots.  相似文献

Two novel strains of budding bacteria, Z-0071T and Z-0072, were isolated from dystrophic humified waters formed by xylotrophic fungi in the course of spruce wood degradation. The cells of both strains are coccoid (0.95–1.80 μm), nonmotile, single or arranged in pairs. The cells have a complex system of intracellular membranes and are covered with fimbriae and surrounded by a mucous capsule up to 0.3 μm thick. Both strains are aerobic organoheterotrophic, mesophilic, and acid-tolerant microorganisms that are able to grow under microaerobic conditions. They utilize N-acetyl-glucosamine, carbohydrates, and lactate as growth substrates. The strains grow in a pH range of 4.0–7.5 with an optimum at 6.0–6.5. The temperature range for growth is 4–30°C, with an optimum at 25–28°C. Strains Z-0071T and Z-0072, inhabitants of dystrophic low-mineral waters, are NaCl-sensitive: the NaCl content in the media above 0.5 g/l inhibited growth. The main fatty acids of strains Z-0071T and Z-0072 are C16:0, C18:1ω9c, and C18:2ω9c, 12c. The DNA G + C base content is 51.2–51.7 mol %. The sequences of the 16S rRNA gene fragments (1310 bp) of strains Z-0071T and Z-0072 were found to be identical. The obtained sequences showed a 94.3% similarity with the sequences of the type strain of the most closely related species Singulisphaera acidiphila MOB10≅T. The phenotypic and phylogenetic properties of strains Z-0071T and Z-0072 support classification of these strains within the genus Singulisphaera as a new species Singulisphaera mucilagenosa sp. nov., with the type strain Z-0071T (VKM B-2626).  相似文献

Susceptibility of the white grub Cyclocephala signaticollis Burmeister (Coleoptera: Scarabaeidae: Dynastinae) larvae to seven isolates of Beauveria bassiana (Balsamo) Vuillemin, five of Metarhizium anisopliae (Metschnikoff) Sorokin and two of Paecilomyces lilacinus (Thom) Samson (Deuteromycotina: Hyphomycetes) was investigated. Among 14 fungal isolates screened the most virulent was a B. bassiana isolate (Bb 53) that caused 70% mortality of third instar larvae in 40 days after inoculation at 1 × 108 conida/ml. Strains of M. anisopliae and P. lilacinus showed low efficacy or no virulence to the target host.  相似文献

Larval behaviour is important to dispersal and settlement, but is seldom quantified. Behavioural capabilities of larval Lutjanus carponotatus in both offshore pelagic and reef environments at Lizard Island, Great Barrier Reef were observed in situ to determine if they were sufficient to influence dispersal. Offshore, larvae swam with higher directional precision and faster on the windward side of the island (28 cm.s−1) than on the leeward side (16 cm s−1). Most larvae swam directionally. Mean swimming directions were southerly in the windward area and northerly in the leeward area. Larvae avoided the surface and remained mostly between 3–15 m. Larvae released near reefs were 2–3 times faster swimming away from reefs (19 cm s−1) than swimming toward or over them (6–8 cm s−1). Speed swimming away was similar to that offshore. Of 41 larvae released near reefs, 73% reached the reef, 59% settled, and 13% of those reaching the reef were eaten. Larvae settled onto hard and soft coral (58%), topographic reef features (29%) and sand and rubble (13%). Settlement depth averaged 5.5 m (2–8 m). Before settling larvae spent up to 800 s over the reef (mean 231 s) and swam up to 53 m (mean 14 m). About half of the larvae interacted with reef residents including predatory attacks and aggressive approaches by residents and aggressive approaches by settling larvae. Settlement behaviour of L. carponotatus was more similar to a serranid than to pomacentrids. Settlement-stage larvae of L. carponotatus are behaviourally capable, and have a complex settlement behaviour.  相似文献

Bacteria decomposed damaged and moribund Chrysaora quinquecirrha Desor, 1848 releasing a pulse of carbon and nutrients. Tissue decomposed in 5–8 days, with 14 g of wet biomass exhibiting a half-life of 3 days at 22°C, which is 3× longer than previous reports. Decomposition raised mean concentrations of organic carbon and nutrients above controls by 1–2 orders of magnitude. An increase in nitrogen (16,117 μg l−1) occurred 24 h after increases in phosphorus (1,365 μg l−1) and organic carbon (25 mg l−1). Cocci dominated control incubations, with no significant increase in numbers. In incubations of tissue, bacilli increased exponentially after 6 h to become dominant, and cocci reproduced at a rate that was 30% slower. These results, and those from previous studies, suggested that natural assemblages may include bacteria that decompose medusae, as well as bacteria that benefit from the subsequent release of carbon and nutrients. This experiment also indicated that proteins and other nitrogenous compounds are less labile in damaged medusae than in dead or homogenized individuals. Overall, dense patches of decomposing medusae represent an important, but poorly documented, component of the trophic shunt that diverts carbon and nutrients incorporated by gelatinous zooplankton into microbial trophic webs.  相似文献

We tested the combined effect of the fungus Beauveria bassiana and the microsporidium Nosema pyrausta on the European corn borer larvae, Ostrinia nubilalis, in the laboratory. The first instar of O. nubilalis larvae was the most sensitive to the B. bassiana infection followed by the fifth, second, third, and fourth instar (LC50s were 4.91, 6.67, 7.13, 9.15, and 6.51 × 105 conidia/ml for the first to fifth instars, respectively). Mortality of each instar increases positively with concentration of conidia. When B. bassiana and N. pyrausta were used in combination, mortality increased significantly in all instars. Relative to the B. bassiana treatment alone, the B. bassiana + N. pyrausta treatment decreased the LC50s by 42.16%, 37.63%, 21.60%, 27.11%, and 33.95% for the first to fifth instars, respectively. The combined effects of the two pathogens were mostly additive. However, at the two highest concentrations the pathogens interacted synergistically in the first and second instar. Individuals that survived the B. bassiana and B. bassiana + N. pyrausta treatments and developed into adults had significantly shorter lifespans and females oviposited fewer eggs than non-exposed insects. The effects on the longevity and the egg production were most pronounced at high concentration of B. bassiana conidia.  相似文献

Decomposition of air-dried live Typha angustifolia (L) stems and leaves and Phragmites australis (Cav. Trin ex Steud.) leaves and culms were studied in a shallow freshwater lake (Lake Fehér, Fertő-Hanság National Park, Hungary) using the litter bag technique. Samples were analyzed for dry mass, fiber (cellulose, hemicelluloses, lignin) and nutrient (C, N, P, S) contents, litter-associated fungal biomass (ergosterol concentration), potential microbial respiration (electron transport activity: ETS) and cellulolytic bacteria. In terms of mesh size, there were no significant differences in the examined parameters of P. australis leaves and culms and T. angustifolia stems with leaves. P. australis leaves had the highest rate of decomposition and P. australis culms the lowest. Hemicellulose degraded more rapidly than the other fibers, while the lignin had the slowest rate of decomposition. The ETS activity of the examined plant litter types increased from day 91st to 237th while decomposition processes were most active, ergosterol contents were high, and there were few cellulolytic bacteria. The counts of cellulolytic bacteria fluctuated during the decomposition period, they were high at the beginning then they decreased. In each case bacteria were found to be the first colonizers of plant detritus, and were followed by fungal growth.  相似文献

Studying Pneumocystis has proven to be a challenge from the perspective of propagating a significant amount of the pathogen in a facile manner. The study of several fungal pathogens has been aided by the use of invertebrate model hosts. Our efforts to infect the invertebrate larvae Galleria mellonella with Pneumocystis proved futile since P. murina neither caused disease nor was able to proliferate within G. mellonella. It did, however, show that the pathogen could be rapidly cleared from the host.  相似文献

Tomato (Lycopersicon esculentum) is important widely grown vegetable in India and its productivity is affected by bacterial wilt disease infection caused by Ralstonia solanacearum. To prevent this disease infection a study was conducted to isolate and screen effective plant growth promoting rhizobacteria (PGPR) antagonistic to R. solanacearum. A total 297 antagonistic bacteria were isolated through dual culture inoculation technique, out of which forty-two antagonistic bacteria were found positive for phlD gene by PCR amplification using two primer sets Phl2a:Phl2b and B2BF:BPR4. The genetic diversity of phlD + bacteria was studied by amplified 16S rDNA restriction analysis and demonstrated eleven groups at 65% similarity level. Out of these 42 phlD + antagonistic isolates, twenty exhibited significantly fair plant growth promoting activities like phosphate solubilization (0.92–5.33%), 25 produced indole acetic acid (1.63–7.78 μg ml−1) and few strains show production of antifungal metabolites (HCN and siderophore). The screening of PGPR (phlD +) for suppression of bacterial wilt disease in glass house conditions was showed ten isolated phlD + bacteria were able to suppress infection of bacterial wilt disease in tomato plant (var. Arka vikas) in the presence R. solanacearum. The PGPR (phlD +) isolates s188, s215 and s288 was observed to be effective plant growth promoter as it shows highest dry weight per plant (3.86, 3.85 and 3.69 g plant−1 respectively). The complete absence of wilt disease symptoms in tomato crop plants was observed by these treatments compared to negative control. Therefore inoculation of tomato plant with phlD + isolate s188 and other similar biocontrol agents may prove to be a positive strategy for checking wilt disease and thus improving plant vigor.  相似文献
