Microhemodynamics and oxygen tension (pO2) in the brain cortex tissues as well as the heart rate were studied in rodents with different ecological specialization during hypoxia produced by subcutaneous injection of sodium nitrite (3 mg/100 g body mass). It was shown that the blood flow in animals with low (rats) and high (muskrats) resistance to hypoxia decreased by the 30th min of the nitrite action, with its subsequent restoration to 85% and 83% of the initial level by the 60th min. The interspecies difference consisted in an increase of the brain blood flow (by 24%) in muskrats and a decrease (by 33%) in rats 15 min after the injection. In rats, simultaneously with the blood-flow dynamics, a pO2 increase was observed in some brain cortex microareas, while in others—a pO2 decrease 15 min after the NaNO2 injection: meanwhile, in muskrats, at this time period a significant pO2 decrease was observed on the background of a blood flow increase. In both animal species, the pO2 minimal value was reached by the 45th min, while restoration almost to the initial levels—by the 60th min of the nitrite action. Changes in the rats, synchronous and unidirectional with the heart rate frequency, of the brain blood-flow, as well as tachycardia developing throughout the whole experiment in rats allow suggesting that restoration of the oxygen regime in the brain cortex microareas is provided by activation of systemic mechanisms of regulation of circulation.  相似文献

The results of investigation of leukocyte morphology and leukocyte contents of blood and caecum depending on the trematode Quinqueserialis quinqueserialis invasion rate in muskrats from natural population are given. At low trematode invasion rates, there was observed systemic activation of lymphopoiesis and neutrophil granulocytopoiesis with a decrease in the monocyte-macrophage response in caecum (trematode localization organ). At the same time, under high invasion rates, there was detected induction of T cell suppressor activity and the absence of a granulocyte response in the tissues under study. Intensification of B lymphocyte blast transformation in caecum tissues as well as the appearance of blast cells in the blood of infected muskrats was observed.  相似文献

Physiological role of catalase in mechanisms of adaptive reactions to hypoxic influences in diving rodents, muskrats Ondatra zibethica, has been investigated. The myocardial tissues were established to be the most resistant to the oxidative stress produced by hypobaric hypoxia-reoxygenation. An intraperitoneal injection of a specific nonreversible catalase inhibitor, 3-amino-1, 2, 4-triazol (3-AT), at a dose of 1 g/kg body weight, on the background of the diving hypoxia-reoxygenation decreased the catalase activity in liver, kidney, heart, and brain tissues, on average, by 83%. No statistically significant changes of this activity were observed in the femoral muscle tissue. In 56.5 min after the injection, the tissue concentrations of 3-AT in liver, kidney, heart, and femoral muscle were approximately similar to amount to about 1 µg/mg wet tissue. The 3-AT concentration in the brain was three times lower than in other tissues; however, this practically did not affect the extent of catalase inhibition. During the diving hypoxia-reoxygenation the reduction of catalase activity under effect of 3-AT was accompanied by an increase of lipid peroxidation (LPO) only in the heart and liver, which indicates an important role of catalase in protection of these tissues from damaging effects of active oxygen forms at resumed external respiration after the long diving.  相似文献

The intoxication of white mice with sodium nitrite results in the decrease of red cell superoxide dismutase (SOD) and catalase activity. The glutathione peroxidase activity is the same as in the control group. The level of red cell lipid peroxidation in the group of mice that receive sodium nitrite is higher as compared to the control group. After the intoxication the total activity of glucose-6-phosphate dehydrogenase and dehydrogenase of 6-phosphogluconate as well as the activity of glutathione reductase are higher than in the control group. The level of SH-groups and reduced glutathione is higher in the group of mice that receive sodium nitrite in comparison with the control group.  相似文献

The purpose of the study was to asses the effect of mexidol, cudesan, hypoxen and essenciale on the functions of the immune system, liver and muscular tissue in nitrite intoxication. The investigation was carried out on Wistar rats exposed to sodium nitrite. Administration of mexidol, cudesan or hypoxen to the animals did not affect the indices characterizing the functional activity of immunocytes, hepatocytes and myocytes. Mexidol in combination with cudesan reduced the intensity of the decrease of the functional metabolic activity of leukocytes and the development of the humoral immune response. Mexidol in combination with hypoxen increased the physical working cepacity of the animals. Both the combinations increased the rate of the drug biotransformation in the liver. The use of essenciale in combination with mexidol resulted in normalization of the metabolic functions of the liver. Essenciale in combination with cudesan normalized the majority of the immunity functions. The combination of essenciale with hypoxen recovered the lowered physical working capacity in the animals.  相似文献

Effects of natural and artificial hypoxic factors on cardiac chronotropic reactions were studied in the muskrat Ondatra zibethicus naturally adapted to underwater hypoxia under conditions of free behavior. To record cardiac activity, original implanted ECG sensors designed in the laboratory were used. Under observations were muskrats in the states of rest, movement, swimming on the water surface, diving, underwater swimming, forced underwater immersion, and artificial apnea, in the low-pressure chamber during changes of pressure from 100 to 25 kPa (ascent to an altitude of 11 km) and in the atmosphere of hypoxic mixtures with 5–10% O2 as well as under conditions of hemic nitrite hypoxia after injection of 3 mg/kg NaNO2. Heart rate (HR) in muskrats is labile and can change within the limits from 15 to 360 beats/min. A characteristic feature of hypoxic action is development in muskrats of bradycardia that can appear either instantly—both as a conditional reflex and from the nose lobe receptors—or gradually at a decrease of pO2 in inhaled air. Before diving and after coming to the surface a brief tachycardia can also be observed. The gradual development of tachycardia takes place in nitrite hypoxia. Development of bradycardia was eliminated at blockade of M-cholinoreceptors by atropine, and of tachycardia—at blockade of β-adrenoreceptors by propranolol. Blockade of α-adrenoreceptors by phentolamine did not affect cardiac chronotropic reactions, which indicates the absence of their connection with vasoconstriction. Analysis of the cardiac rhythm variability has revealed a large spectrum of slow cardiointerval fluctuations connected with animal functional states. Regulation of cardiac chronotropic reactions in muskrats under effect of hypoxic factors operates along both sympathetic and parasympathetic pathways of the autonomic nervous system, the leading role in these processes being played by vagus influences. Preliminary introduction of nitroxyl radicals to mice decreases methaemoglobin-forming effect of sodium nitrite and diminishes the content of total SH-groups and restored glutathione as well as the activity of glutathione reductase and total activity of dehydrogenases of the pentosophosphate path of erythrocytes. High level of lipids peroxidation in case of sodium nitrite intoxication remains unchanged under preliminary administration of nitroxyl radicals as well. Activity of the key enzymes of antioxidant protection of erythrocytes, superoxide dismutase and catalase, is not recovered with sodium nitrite intoxication in presence of nitroxyl radicals.  相似文献

Cadmium induced lipid peroxidation (LPO) and the activity of antioxidantenzymes after the administration of a single dose of CdCl 2 (0.4 mg kg body wt, ip) was studied in rat erythrocytes.Cd intoxication increased erythrocyte LPO along with a decrease insuperoxide dismutase (SOD) up to three days of Cd treatment. Thedecrease in erythrocyte catalase (CAT) activity was marked within9 h of Cd intoxication. After three days of Cd treatment, LPOdecreased towards normal, along with an increase in erythrocyteSOC and CAT activity. Blood glutathione (GSH) decreased significantlywithin 24 h of Cd treatment, followed by an increase towards normal.Erythrocyte glutathione S-transferase (GST) activity increased up to10 days of Cd intoxication, probably in an attempt to reduce Cd toxicity.Serum glutamate pyruvate transaminase (SGPT), serum alkaline phosphatase(SALP) and serum bilirubin increased up to 10 days of Cd intoxication.Blood urea increased significantly up to three days, followed by a decreasetowards normal. The results show that Cd induced LPO was associated with adecrease in antioxidant enzymes and GSH in erythrocytes; as these antioxidantsincrease in erythrocytes with recovery from Cd intoxication, the Cd inducedLPO reversed towards normal. The increase in the SGPT, SALP and serum bilirubincorrelated with LPO. We examined how oxidative stress and cell damage develop in the liver of rats subjected to water-immersion stress (WIRS). In rats subjected to WIRS for 1.5, 3 or 6 h, serum alanine aminotransferase and aspartate aminotransferase activities increased time-dependently. In the liver tissue, vacuolization and apoptosis occurred at 1.5 h of WIRS and vacuolization further developed without further appearance of apoptosis at 3 h or 6 h. Serum lipid peroxide (LPO) and NOx (nitrite/nitrate) concentrations increased at 3 h of WIRS and these increases were enhanced at 6 h. In liver tissue, increases in LPO and NOx concentrations and myeloperoxidase activity and decreases in ascorbic acid and reduced glutathione concentrations and superoxide dismutase activity occurred at 3 h of WIRS and these changes were enhanced at 6 h, although vitamin E concentration and xanthine oxidase activity were unchanged. These results indicate that oxidative stress in the liver of rats with WIRS develops after the appearance of cell damage in the tissue, and suggests that oxidative stress is caused through disruption of the antioxidant defense system and increases in NO generation and neutrophil infiltration in the liver, which may contribute to the progression of cell damage in the tissue.  相似文献

Dietary nitrite and nitrate are important sources of nitric oxide (NO). However, the use of nitrite as an antihypertensive drug may be limited by increased oxidative stress associated with hypertension. We evaluated the antihypertensive effects of sodium nitrite given in drinking water for 4 weeks in two-kidney one-clip (2K1C) hypertensive rats and the effects induced by nitrite on NO bioavailability and oxidative stress. We found that, even under the increased oxidative stress conditions present in 2K1C hypertension, nitrite reduced systolic blood pressure in a dose-dependent manner. Whereas treatment with nitrite did not significantly change plasma nitrite concentrations in 2K1C rats, it increased plasma nitrate levels significantly. Surprisingly, nitrite treatment exerted antioxidant effects in both hypertensive and sham-normotensive control rats. A series of in vitro experiments was carried out to show that the antioxidant effects induced by nitrite do not involve direct antioxidant effects or xanthine oxidase activity inhibition. Conversely, nitrite decreased vascular NADPH oxidase activity. Taken together, our results show for the first time that nitrite has antihypertensive effects in 2K1C hypertensive rats, which may be due to its antioxidant properties resulting from vascular NADPH oxidase activity inhibition.  相似文献

The effect of mouse interferon on the bactericidal activity of macrophages against pyogenic cocci was examined. Mouse peritoneal macrophages were cultivated with Staphylococcus aureus in vitro and viable Staphylococcus was recovered by treatment of the mixed macrophage-bacteria culture with sodium dodecyl sulphate (SDS) solution. Results showed that S. aureus was phagocytized and killed by the macrophages. Mouse L cell interferon enhanced the bactericidal activity of macrophages. A mouse brain interferon preparation also enhanced this activity. However, heat-inactivated L cell interferon and heterologous rabbit RK-13 cell interferon and human leukocyte interferon did not enhance it. This suggests that interferon enhances the bactericidal activity of macrophages against S. aureus.  相似文献

Investigation ofopioid peptide effect on the production of reactive oxygen species by neutrophils in non-fractionated leukocyte suspension and in purified fraction of peripheral blood neutrophils is disclosed in this work. It was determined that selective delta- and micro-agonists of peptide origin stimulated the spontaneous and suppressed 15 mkg/ml zymosan-induced LDCL (luminol-dependent chemiluminescence) reaction of neutrophils in leukocyte suspension. beta-endorphin was found to render less marked suppressive action on 15 mkg/ml zymosan-induced LDCL, and delta2-agonist deltorphin 2 promoted 15 mkg/ml zymosan-induced LDCL only toward the 25 minutes of the experiment. beta-endorphin and selective d- and m- agonists did not affect the spontaneous and suppressed 15 mkg/ml and 150 mkg/ml zymosan-induced neutrophil LDCL. Therefore, opioid peptides play essential role in the process of direct and indirect regulation of oxygen-dependent system of neutrophil granulocyte bactericidal activity.  相似文献

The interaction of exercise training and ethanol on the myocardial antioxidant enzymes and the oxidative stress markers was investigated in the Wistar strain male albino rats. We also tested the interactive effects of exercise training and ethanol on the age-associated free radical production and antioxidant defense system. We found a significant decrease (p<0.05) in the activity levels of superoxide dismutase (SOD) and catalase (CAT) in the myocardium of old rats when compared to young rats by 26% and 58%, respectively, suggesting the onset of age-dependent decrease in the myocardial antioxidant enzyme system. In contrast to the decreased antioxidant enzyme activity, xanthine oxidase (XOD) and lipid peroxidation (LPO) levels were elevated, suggesting the age-induced oxidative stress. Exercise training significantly (p < 0.05) elevated the activities of SOD, CAT, XOD and LPO levels in both the age groups of animals. Ethanol consumption significantly lowered the SOD and CAT activities in both the age groups, whereas a significant increase was observed in the XOD and LPO levels. In contrast, the combination of exercise training plus ethanol lowered XOD and LPO levels in both the age groups of rats compared to ethanol treated rats. A significant (p < 0.05) increase in the activities of SOD and CAT was reported in the rats treated with the combination of exercise training plus ethanol. This increase was more pronounced in the younger rats than the older rats. The findings of the present investigation on the potential role of antioxidant enzymes to counter the ethanol-induced pro-oxidants showed an increase with the interaction of exercise training. With age, a decrease in the antioxidant enzyme capacity was observed. This reveals that the old age rats were more affected to the pro-oxidants when compared to the young age rats. In conclusion it is demonstrated that two months treadmill endurance exercise training is beneficial to both young and old rats in improving antioxidant defense to challenge the oxidative stress in the myocardial tissue and thereby successfully countering the free radical production due to ethanol intoxication.  相似文献

It is well known that the nervous system has increased susceptibility to methanol intoxication. The present study reveals the effect of methanol intoxication on antioxidant status, lipid peroxidation and DNA integrity in hypothalamic-pituitary-adrenal (HPA) axis organs and spleen. Non-specific and specific immune functions were analyzed. In addition, open field behavior, plasma corticosterone level and blood methanol level were estimated. Male Wistar albino rats were intoxicated with methanol (2.37 g/kg b.wt., i.p.) for 1 day, 15 and 30 days. Administration of methanol showed significant increase in enzymatic (superoxide dismutase, catalase, glutathione peroxidase), non-enzymatic (reduced glutathione and Vitamin C) antioxidants and lipid peroxidation (LPO) in hypothalamus and adrenal gland of day 1 group. However, decrease in enzymatic and non-enzymatic antioxidants with concomitant increase in LPO level were observed in 15 and 30 days groups. Plasma corticosterone level was significantly increased in day 1 and 15 days groups whereas, 30 days methanol intoxication group showed considerable decrease in corticosterone level compared with control animals. Cell-mediated immune response of footpad thickness was significantly decreased with an increased leukocyte migration inhibition. Humoral immune response of antibody titers was elevated in methanol-intoxicated groups. Neutrophil functions, adherence and phagocytic index (PI) were found to be significantly decreases. Furthermore, significant increase in the avidity index and nitro blue tetrozolium reduction was observed in the methanol exposed animals. Day 1 methanol exposed group showed increased PI compared to the control ones. Methanol exposure for 30 days showed an increased DNA fragmentation in the hypothalamus, adrenal glands, and spleen. In conclusion, exposure to methanol-induced oxidative stress disturbs the HPA-axis function altering the level of corticosterone, which lead to varied non-specific and specific immune response in experimental rats.  相似文献

SYNOPSIS. Rats inoculated with 10,000 sporulated oocysts of Eimeria nieschulzi had significantly higher total leukocyte counts on postinoculation days (PI) 1, 5, 6 and 7 when compared to control rats. Relative and absolute neutrophil counts increased concomitantly with a decrease in the relative lymphocyte levels in E. nieschulzi-infected rats on PI day 7. Absolute and relative neutrophil counts in infected rats on PI days 7 and 8 were closely correlated with the host's total oocyst discharge. The E. nieschulzi infection had no significant effect on the relative or absolute levels of monocytes or eosinophils. The described changes in leukocyte levels were not paralleled by a significant change in the erythrocyte count.  相似文献

In anesthetized male rats the arterial blood pressure in femoral artery and electrocardiogram in standard leads were recorded uninterruptedly for 1–1.5 h under conditions of acute nitrite intoxication produced by a subcutaneous injection of water solution of sodium nitrite (donor of nitric oxide) at concentrations of 10, 30, and 50 mg/kg body mass. Results of the study have shown dose-dependent changes of arterial pressure as well as of time and amplitude characteristics of electrocardiogram under effect of NaNO2. At the threshold hypoxic dose, an increase of amplitude of R and S waves was observed by the 30–45th min, while at the maximal NaNO2 dose, amplitude of all waves rose by the 15th min of intoxication. High nitrite doses often caused an elevation of the ST segment above the isoelectric line and a rise of the amplitude of the T wave, on which a notch appeared in some cases. The change of the ECG time parameters was expressed in the dose-dependent development of bradycardia for the first 4–7 min; its level correlated with the progressively decreasing arterial pressure in the beginning (the 2–4th min) of nitrite intoxication. Variation analysis of the heart rate spectral characteristics by Baevskii’s method has revealed a rise of the total spectral power of pulse oscillations. Under effect of nitrite, in the specter of cardiointervals, the slower oscillations have been revealed with frequency of 0.15–0.2 Hz in the LF diapason with subsequent recovery of the normal ECG specter at the end of the experimental period. The maximal nitrite dose produced more pronounced shifts of the heart rate specter towards the LF and VLF diapasons that were not restored for 1 h of experiment. Transitory processes of readjustment of the cardiac rhythm had discrete character. The nitrite dose of 100 mg/kg body mass increased the RR-interval after 4–7 min with amplitude steps of 3–5 imp/s and the time constant of 20–40 s. The revealed ECG changes had the reflex (enhancement of parasympathetic tonus) and metabolic (the hypoxic and histotoxic damage of myocardium) nature.  相似文献

Toxin production by Clostridium botulinum was inhibited by sodium nitrite levels above 50 mug/g of wiener. Sodium ascorbate at levels of 105 and 655 mug/g of product did not decrease the effectiveness of the sodium nitrite inhibition, nor did sodium ascorbate potentiate it. The results indicate that the use of sodium ascorbate in vacuum-packaged wieners does not appreciably alter the inhibition of C. botulinum toxin formation by sodium nitrite.  相似文献

The present study evaluated the protective effect of fish oil in isoproterenol-induced myocardial infarction in rats. The results of the present study indicate that the IPH administration decreases the activities of membrane-bound ATPases compared to control animals. Fish oil pretreatment brought about significant increase in the activity of these membrane-bound ATPases in IPH (isoproterenol hydrochloride)-treated animals. Significant increase in serum potassium level with concomitant decrease in the values of sodium, magnesium, and calcium were observed in IPH-treated rats compared to control rats, fish oil pretreatment reversed these changes to near normal. Significant elevation of sodium and calcium levels with concomitant decrease in the levels of potassium and magnesium were observed in the myocardial tissue of IPH-administered rats compared to control rats, fish oil pretreatment followed by IPH administration brought these levels to near normal. The levels of lipid peroxidation (LPO) in both serum and tissue were increased in IPH-treated rats compared with control rats, whereas pretreatment with fish oil in IPH-treated rats maintained near-normal LPO levels. The results of the present study reveals that the pretreatment of fish maintains the activities of membrane-bound ATPases and the mineral levels at near normal by the inhibition of lipid peroxidation  相似文献

The bactericidal capability of murine peritoneal polymorphonuclear neutrophils against virulent and nonvirulent Salmonella typhimurium was examined in an in vitro system. Although preincubation of the bacteria in specific murine antiserum elicited greater chemiluminescence from phagocytizing neutrophils than did incubation in normal murine serum, antiserum did not enhance ingestion, as less than 5% of the challenge was taken up by neutrophils under any of the conditions studied. Nonvirulent salmonellae showed a transient decrease in viable numbers early during in vitro incubation with or without intact neutrophils. Virulent salmonellae, however, were able to multiply without a lag period except when these bacteria were pretreated with antiserum and incubated in association with intact murine neutrophils. Results of these in vitro studies suggest that the murine polymorphonuclear neutrophil and antisalmonella antibody must act together to effect neutrophil-associated bactericidal activity against virulent salmonellae, and thus, that the neutrophil alone does not play a major role in the protection of unvaccinated, sensitive mice from disease caused by S. typhimurium.  相似文献

A full marathon is the longest running race in official track events and is a form of acute exercise. However, no studies have examined the acute neutrophil function response to a competitive marathon race. Thirty‐six male athletes who had just completed the 42.195 km course of the 50th Beppu‐Oita Mainichi Marathon were enrolled in this study. Neutrophil oxidative burst activity, phagocytic activity and expression of CD11b and CD16 per cell were measured by flow cytometry immediately before and after the marathon. Total leukocyte/neutrophil counts increased significantly (p < 0.001), whereas total oxidative burst activity per neutrophil cell decreased significantly after the race (p < 0.001). Furthermore, total phagocytic activity per neutrophil cell also decreased after the race, although it was not significant (p = 0.08). Although CD11b expression per cell did not change, the expression of CD16 per cell significantly decreased (p < 0.001) after the race. In conclusion, a competitive marathon race decreased neutrophil functions (oxidative burst activity and phagocytic activity), which may be partly due to a decrease in CD16 expression. The increase in total neutrophil counts might reflect a compensatory response to counteract the decrease in neutrophil functions. Copyright © 2003 John Wiley & Sons, Ltd.  相似文献
