Abstract

Addition of amino acids, glycine, alanine, and serine, to poorly soluble copper(II) salts [copper(II) citrate and copper(II) succinate] all increase solubility of the copper(II) salts. Relative increases in solubility follow the polarity trend in the selected amino acids, with serine creating the greatest increase in solubility. Simultaneous equilibria calculations indicate the formation of mixed-ligand complexes in the copper(II) succinate–amino acid systems, the first time such mixed-ligand complexes have been observed. In contrast, mixed-ligand complexes are not predicted in the copper(II) citrate–amino acid systems. Potential bioavailability of copper(II) appears to be increased by the inclusion of amino acids in solution, roughly in parallel with the increase in solubility of the copper(II) salt. Therefore, measurement of the change in solubility caused by addition of amino acids to aqueous solution gives qualitative insight to the potential increase in bioavailability of the metal ion.  相似文献

Abstract

The effect of added amino acid (glycine, alanine and serine) on the solubility of zinc citrate, zinc succinate and zinc oxalate was investigated. Equilibrated solutions were prepared and titrated to endpoint at pH 5.5 with Na2EDTA using Xylenol Orange indicator. In every case, a significant enhancement in solubility was seen as the amino acid increased from 1 to 50 molar excess. The increase was non-linear, and, with zinc succinate, showed saturation effects. In these cases, solution composition may be more complicated than initially thought, including mixed-ligand complexes and increasing percent free metal ion as the proportion of amino acid increases.  相似文献

1. Proline was found to be the major component of CTC-12 (44%) and FSS II (45%) strain.2. The cypermethrin treatment resulted in an increase in most of the amino acids of sixth instar larvae and all amino acids of adult beetles of CTC 12 strain.3. In the susceptible strain (FSS II), however, the tyrosine, phenylalanine and arginine increased, whereas serine, proline, glycine, alanine, valine, isoleucine, leucine and lysine were decreased significantly in the sixth instar larvae.4. In the FSS II adult beetles, only aspartic acid increased, while other amino acids either decreased (threonine, proline, glycine, alanine, valine, methionine, isoleucine, tyrososine, lysine, arginine) or remained unaffected (serine, glutamic acid, leucine, phenylalanine, histidine).  相似文献

Intact spinach (Spinacia oleracea L.) leaf peroxisomes were supplied with glycolate and one to three of the amino acids serine, glutamate, and alanine, and the amount of the respective α-keto acids formed in glyoxylate transamination was assayed. At 1 millimolar glycolate and 1 millimolar each of the three amino acids in combination, the transamination reaction reached saturation; reduction of either glycolate or amino acid concentration decreased the activity. The relative serine, glutamate, and alanine transamination at equal amino acid concentrations was roughly 40, 30, and 30%, respectively. The three amino acids exhibited mutual inhibition to one another in transamination due to the competition for the supply of glyoxylate. In addition to this competition for glyoxylate, competitive inhibition at the active site of enzymes occurred between glutamate and alanine, but not between serine and glutamate or alanine. Alteration of the relative concentrations of the three amino acids changed their relative transamination. Similar work was performed with intact oat (Avena sativa L.) leaf peroxisomes. At 1 millimolar of each of the three amino acids in combination, the relative serine, glutamate, and alanine transamination was roughly 60, 23, and 17%, respectively. Similarly, alteration of the relative concentration of the three amino acids changed their relative transamination. The contents of the three amino acids in leaf extracts were analyzed, and the relative contribution of the three amino acids in glycine production in photorespiration was assessed and discussed.  相似文献

Methylisothiocyanate reacts with the amino group of amino acid in alkaline solution to give methylthiocarbamyl amino acid. This is converted to the methylthiohydantoin derivative (MTH-amino acid) by subsequent acidification.

Gas chromatographical identification of 20 amino acids were examined by making MTH-amino acids. Among them, ten amino acids (glycine, alanine, valine, leucine, isoleucine, proline, methionine, phenylalanine, serine and threonine) were successfully identified. Aspartic acid and glutamic acid could be identified as the methyl esters of the MTH-amino acids.  相似文献

Contact glow discharge electrolyses (CGDE) were carried out relative to the prebiotic formation of amino acids by amination of aliphatic nitrile in aqueous ammoniacal solution, and by cyanization of aliphatic amine by sodium cyanide. The CGDE of propionitrile by amination followed by hydrolysis resulted in the formation of glycine, alanine and β-alanine. The reaction of ethylamine by cyanization, gave glycine, alanine, β-alanine, aspartic acid, and serine. In these reactions, a relatively high ratio of glycine was observed. This could be explained by the cleavage of the α,β-carbon bond, which was broken easily, due to the strong electron-attracting property of the nitrile group of propionitrile and the resulting α-aminopropionitrile.  相似文献

Ammonium acetate in various conditions was irradiated in a reactor to examine the contributions of both the reactor radiations and recoiled14C nucleis to form the biologically interesting molecules. Present investigations demonstrated that several amino acids, glycine, alanine, -alanine and GABA, and may-be aspartic acid, serine and valine by prolonged irradiation, were formed in the aqueous solutions of ammonium acetate.14C-radioactivities were also found distributed in these amino acids. 1) To detect the quantitative changes of amino acid acceptor activity of tRNA's from the posterior and middle silk glands of Bombyx mori at various ages, a relatively simple and rapid method was established using a mixture of radioactive amino acids in Chlorella hydrolysate. 2) The acceptor activities of silk gland tRNA for 15 amino acids tested seemed to be almost on the same level at the end of the 4th moult stage. During the 5th instar, however, characteristic increases were observed in glycine, alanine, and serine acceptor activities in both silk glands. 3) In the posterior silk gland, which produces fibroin, the acceptor activities for glycine and alanine increased more than that for serine. In the middle silk gland, which produces sericine, the acceptor activity for serine increased more than those for glycine and alanine. 4) In the light of observations on the increase of corresponding aminoacyl-tRNA synthetase activities in the silk glands, a functional adaptation of tRNA synthesis in the tissue is discussed.  相似文献

Threonine content of brain decreases in young rats fed a threonine-limiting, low protein diet containing a supplement of small neutral amino acids (serine, glycine and alanine), which are competitors of threonine transport in other systems (Tews et al., 1977). Threonine transport by brain slices was inhibited more by a complex amino acid mixture resembling plasma from rats fed the small neutral amino acid supplement than by mixtures resembling plasma from control rats or from rats fed a supplement of large neutral amino acids. Greater inhibition was seen with mixtures containing only the small neutral amino acids than with mixtures containing only large neutral amino acids. On an equimolar basis, serine and alanine were the most inhibitory; large neutrals were moderately so; and glycine and lysine were without effect. Threonine transport was also strongly inhibited by α-amino-n-butyric acid and homoserine, less so by α-aminoisobutyric acid, and not at all by GABA. The complex amino acid mixtures strongly inhibited α-aminoisobutyric acid transport by brain or liver slices but, in contrast to effects in brain, the extent of the inhibition in liver was not much affected by altering the composition of the mixture. Tryptophan accumulation by brain slices was effectively inhibited by other large neutral amino acids in physiologically occurring concentrations. Threonine, or a mixture of serine, glycine and alanine only slightly inhibited tryptophan uptake; basic amino acids were without effect and histidine stimulated tryptophan transport slightly. These results support the conclusion that a diet-induced decrease in the concentration in brain of a specific amino acid may be related to increased inhibition of its transport into brain by increases in the concentrations of transport-related, plasma amino acids.  相似文献

Two systems mediating the transport of amino acids were studied in vesicles derived from protein-depleted membranes of pigeon erythrocytes. One system (ASC system) catalysed the Na+-dependent exchange of small neutral amino acids, such as alanine, serine and cysteine. The other system, also Na+-dependent, mediated the active transport of glycine. The ASC and glycine systems were distinguished by the sensitivity of the latter to the anion present, by the former's requirement for an exchangeable amino acid and by the inability of alanine to inhibit the transport of glycine. Preliminary results indicated that the influx of glycine was electrically silent. The only major integral protein retained in the vesicles was the band 3 protein, but that could not be unequivocally identified as the transporter.  相似文献

In most other studies the release of amino acid neurotransmitters and modulators in vitro has been studied mostly using labeled preloaded compounds. For several reasons the estimated release may not reliably reflect the release of endogenous compounds. The magnitudes of the release cannot thus be quite correctly estimated using radioactive labels. The basal and K+-evoked release of the neuroactive endogenous amino acids γ-aminobutyrate (GABA), glycine, taurine, glutamate and aspartate was now studied in slices from the striatum from 7-day-old to 3-month-old mice under control (normoxic) and ischemic conditions. The release of alanine, threonine and serine was assessed as control. GABA and glutamate release was much greater in 3-month-old than in 7-day-old mice, whereas with taurine the situation was the opposite. Ischemia markedly enhanced the release of all these three amino acids. The release of aspartate and glycine was markedly enhanced as well whereas no effects were discernible in the release of glutamine, alanine, serine and threonine. K+ stimulation (50 mM) enhanced the release of GABA, glutamate, taurine, aspartate and glycine in most cases, except with taurine in 3-month-old mice under the ischemic conditions and with aspartate in 7-day-old mice under the control conditions. K+ stimulation did not affect the release of glutamine, alanine, serine or threonine. The results on endogenous amino acids are qualitatively similar to those obtained in our earlier experiments with labeled preloaded amino acids. In conclusion, in developing mice only inhibitory taurine is released in such amounts that may counteract the harmful effects of excitatory amino acids in ischemia.  相似文献

Amounts of some metabolites and the incorporation of 14CO2 intophotosynthetic products were measured in the third leaf of wheat,grown with two rates of nitrate supply at two temperatures,to analyse the effects of environmental conditions on the fluxesof carbon. Ribulose bisphosphate and 3-phosphoglyceric acidcontent per unit area were greater under nitrate deficiencyand decreased with leafage, but did not differ consistentlywith temperature. Sucrose content of young leaves was largerin cool than in warm conditions and with low nitrate, and decreasedwith age to similar values in all treatments. Starch accumulatedwith leaf age, slightly more in cool than warm conditions, andwith nitrate deficiency. Glutamate (plus glutamine), aspartate(plus asparagine), glycine and serine content of leaves weregreatest with added nitrate in cool temperature; changes withleaf age and conditions are discussed. The 14C content of assimilationproducts after exposure to 14CO2 (for up to 10 min at 20 ?C)under steady-state conditions was slightly greater in plantsgrown in the warm than in the cool temperature and with additionalnitrate. Additional nitrate increased the proportion of 14Cin, and flux of carbon to, amino acids, particularly serineand glycine, and decreased it in sugar phosphates and sucrose.Cool growth temperatures increased the proportion of 14C inamino acids (pre-dominantly glycine and serine) and decreasedthat in sucrose. Changes in the balance of carbon fluxes betweenamino acids and carbohydrates are discussed in relation to glycolatepathway metabolism and alternative routes of amino acid synthesis. Potentiometric titrations and 1H NMR spectroscopic studies of amino acids binding to the [ZnL]2+‐complex where L = cyclen in aqueous solution provide information concerning complexing species identity, their stability, and coordination mode declaration. The amino acids form stable ternary [ZnL(HLn)]2+ and [ZnL(Ln)]+ complexes. The observations indicate bidentate coordination mode of the deprotonated amino acids, involving both the amine and the carboxylate functions to the [ZnL]2+ complex in pH region of about 7.5–9.5. The determined stability constants indicate that [ZnL]2+ complex is a very efficient receptor for simple amino acids such as glycine and alanine. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

When the amino-acid mixture of an effective chemically defined diet was replaced by single amino acids, keeping the total nitrogen at the same level, the egg production of Dacus oleae was minimal with all the 19 amino acids tested. Male survival was adversely affected by the amino acids : alanine, aspartic acid, glutamic acid, glycine, hydroxyproline, lysine, serine and tyrosine, while female survival was shortened when the amino acids : glycine, hydroxyproline and lysine were added. The creation of amino-acid imbalances, by deleting the 19 amino acids individually, from the complete amino-acid mixture, showed that the amino acids : arginine, histidine, isoleucine, leucine, lysine, methionine, phenylalanine, serine, threonine, tryptophane and valine were indispensable to the adult Dacus oleae flies, as far as egg production is concerned. Survival of the male flies was significantly shortened when the amino acids : alanine, hydroxyproline and tryptophane were omitted. Significant differences in longevity between males and females were scored, when the amino acids : alanine, aspartic acid, cystine, glycine and tryptophane, were omitted.  相似文献

Akaboshi et al. (1990) has found an unexpected protection of the achiral amino acid, glycine, towards ionizing radiation at the expense of the selective destruction of the chiral amino acids, alanine and aspartic acid. The present work examines the mechanism of this protection for the case of alanine. We have developed a computer model for the radiolysis of glycine, alanine and glycine-alanine mixtures in aqueous solution. It is established that this protection is due in part to the reaction of the α-radical of glycine with alanine to regenerate a more stable α-radical, according to the following reaction, $$ \cdot CH(NH_3^ + )CO_2^ - + CH_3 CH(NH_3^ + )CO_2^ - \to CH_2 (NH_3^ + )CO_2^ - + CH_3 \dot C(NH_3^ + )CO_2^ -$$ The rate constant of this reaction was estimated to be ≤104M-1s-1. The implications for this selective protection of glycine are considered for a hypothetical case in which there would be an enrichment of about 10% ofL-alanine in the primitive ocean and taking the glycine/alanine ratios obtained in CH4-and CO2- dominated atmospheres using electric discharge experiments. It is predicted that alanine would be rapidly destroyed and radioracemized in spite of the fact that the concentration of alanine is equal or significantly lower than that of glycine. Assuming that chiral amino acids were a prerequisite for the origin of life, it can be deduced that life could have appeared in a relatively short period of time unless there was a constant supply of optical amino acids from extraterrestrial sources.  相似文献

Seventeen protein-constitutive and free amino acids were isolated and determined from the fruit-bodies of Tricholoma giganteum . Aspartic acid and alanine were the most abundant protein constitutive amino acids. Among the free amino acids, alanine, glutamic acid, serine, aspartic acid and glycine were present in the largest amounts, with aspartic acid and glutamic acids especially dominant.  相似文献

The influence of a microsporidian, Thelohania maenadis Pérez, 1904 (Protozoa: Microspora), on the free amino-acid content of the haemolymph and muscle tissue of Carcinus mediterraneus Czerniavsky, 1884 (Crustacea: Decapoda: Brachyura) is studied.Analysis of free amino acids reveals the presence in affected crabs, whose muscle tissue is 70–80% destroyed, of four additional, non-identified compounds in the haemolymp and eleven in the muscle. Parasitization does not provoke a substantial variation in the total free amino acids of the haemolymph and the muscle. In the latter, methionine sulphoxide, methionine and serine levels increase considerably; glycine, proline and arginine levels fall while alanine and taurine levels rise.The desalination at moderate temperatures of healthy and parasitized crabs alike results in a reduction of the total free amino acids of the haemolymph, mainly due to the lowering of proline, glycine and alanine concentrations. Supersalination leads to only a slight reduction of the total free amino acids in healthy specimens, and has no effect on parasitized crabs. The sharp increase in proline and alanine is offset by a reduction in the levels of other amino acids - arginine, glutamic acid, tryptophan and taurine in healthy crabs; and arginine, tryptophan, taurine and serine in parasitized specimes.The influence of the combined factors of salinity and temperature was studied in conditions comparable to those pertaining in winter and summer in the lagoon which is the crab's natural habitat. At low and high extremes of temperature, the effect of salinity on healthy crabs is reversed: desalination results in increased free amino-acid levels. Parasitization reduces this effect of temperature extremes on the influence exercised by salinity, and may annul it completely at high temperatures.  相似文献

The contents of glycogen, lipid, urea and amino acids, and some enzyme activities in plasma, liver muscle and urine were determined with rats fed 10 to 12 g of 100 g body weight per day of the 10% casein diet (control) and 10% casein diets containing 7% glycine with or without 1.4% l-arginine HC1 and l-methionine for 7 days.

Nine hours after the final feeding, the amount of liver glycogen was high in the order of rats fed 10% casein diet containing 7% glycine, 10% casein diet containing 7% glycine with l-arginine and l-methionine, and the control. The amount of muscle glycogen was decreased only in those fed the control diet. The amount of liver lipid was increased by the addition of l-arginine and l-methionine to the excess glycine diet. Plasma and urinary urea was increased in animals given the excess glycine diets with or without both amino acids. In plasma liver, and muscle of animals given either of both the excess glycine diets 3 and 9 hr after the feeding, in general, glycine and serine were increased, and threonine and alanine were decreased as compared with those of rats given the control diet. However, the increase of glycine in plasma, liver and muscle detected at 9 hr after feeding the excess glycine diet was slightly prevented by the supplementation of both amino acids to the excess glycine diet. The activities of liver glycine oxidase and ornithine δ-aminotransferase of rats given the excess glycine diet with both amino acids were higher than those of other dietary groups. Liver serine dehydratase and glutamate-oxalacetate transaminase activities were high in the order of the animals fed the control, the excess glycine diet and the excess glycine diet containing both amino acids. Glutamate-pyruvate transaminase activity in the liver of rats fed the excess glycine diets with or without both amino acids were markedly higher than that of those fed the control. The activities of phosphopyruvate carboxylase and aconitase in the liver of animals given the excess glycine diet were higher than those of other dietary groups. Liver pyruvate kinase and glutamate dehydrogenase activities were similar among those dietary groups.  相似文献

When 14CO2 was fed to flag leaf laminae at 20 d post-anthesis,the transport organs between the leaf and the grains containedappreciable 14C in glutamine, glutamate, serine, alanine, threonineand glycine. Smaller amounts of 14C were present in gamma-aminobutyricacid (GABA), aspartate and cysteine. Other amino acids whichwere labelled in the source leaf were not labelled in the transportorgans. The export of labelled glutamine, serine, glycine andthreonine from the source leaf was favoured in comparison tothe other amino acids mentioned. Threonine accumulated, andwas subsequently metabolised, in the rachis. [14C]GABA alsoaccumulated in the rachis. In the grains, the relative amountof soluble [14C]alanine increased with chase time. Summary The effects of four lysimeter soil series under three salinity levels were evaluated for grain yield, wt/1000 seeds, protein, and amino acids in Mexican dwarf wheat (Triticum aestivum L. var. Cajeme 71). The soil series consisted of: Holtville clay loam, Greenfield sandy loam, San Emigdio sandy loam, and Altamont clay loam. The irrigation water salinity levels were designated: low –2.2 mmho, medium –4.2 mmho, and high –7.1 mmho.No significant differences were found in the amount of grain harvested or wt/1000 seeds in the 1976 crop produced on the differential soil series. The yield of the 1977 crop was significantly affected by the soil types.Effects of soil type on the protein amino acids in the grain in both years were similar. Significantly higher protein amino acid levels of histidine, arginine, aspartic acid, threonine, serine, glutamic acid, glycine, alanine, cystine, valine, methionine, isoleucine, leucine, tyrosine, and phenylalanine were found in the grain grown on Altamont clay loam soil than the other types.The free amino acids in grain from the 1976 and 1977 crops were similarly affected by the soil types, except that the quantitative values of the free amino acids were substantially lower in 1977 than in 1976. The free amino acids significantly influenced by soil types were tryptophane, lysine, arginine, aspartic acid, threonine, serine, glycine, alanine, valine, isoleucine, tyrosine, and phenylalanine. In both years' crops, the sum of the free amino acid fractions was significantly higher in the grain produced on the Altamont soil than on the other soils.Salinity level in the irrigation water did not affect the 1976 crop yield or wt/1000 seeds. Although yields of the 1977 crop were significantly reduced by salinity, the wt/1000 seeds was not. The sum of protein amino acids was significantly higher in the 1976 and 1977 grain crops irrigated with high salinity water than in low salinity irrigated crops.An increased salinity irrigation water significantly reduced the sum of free amino acid fractions in the 1976 grain crop. Since some of the free amino acids in the 1977 grain crop increased while the others decreased due to the salinity level in the irrigation water, the sum of the free amino acid fractions was not significantly influenced.Significant interactions were found between soil types and salinity levels on free arginine, threonine, serine, glutamic acid, and alanine, and also on the sum of the free amino acids in the 1976 wheat grain. In the 1977 wheat grain, there were significant interactions between soil types and salinity levels on the free glutamic acid, valine, leucine, tyrosine, and phenylalanine, and on protein serine, glutamic acid, glycine, alanine, and the sum of the protein amino acids.The amounts of essential amino acids expressed as mg of amino acid/g of protein were not affected by the soil types or salinity levels. With the exception of lysine, and possibly threonine and methionine plus cystine, the essential amino acids were present in the grain at concentrations equal to or greater than recommended by WHO and FAO.  相似文献
