Douglas fir seedlings grown under aseptic conditions in a peat-vermiculite substrate were inoculated with four pairs of ectomycorrhizal fungi to assess the relative inoculum dosages needed to establish two mycorrhizal fungi simultaneously in the same root system. The dual fungal combinations tested were: Pisolithus arhizus + Rhizopogon subareolatus, P. arhizus + R. roseolus, Laccaria bicolor + P. arhizus and L. bicolor + R. subareolatus. A total of 12 ml of inocula per plant was applied at the rates: 0+12, 3+9, 6+6, 9+3, 12+0, and 0+0 (v+v) for each combination. After 3 months growth, the number of mycorrhizas and uninfected short roots as well as the total plant biomass produced were recorded. Inoculations were successful with the fungal combinations P. arhizus + R. subareolatus and L. bicolor + P. arhizus. Plants developed P. arhizus and R. subareolatus mycorrhizas only at the rate 9Pa + 3Rs; at other rates tested, only monospecific mycorrhizas were formed. Plants developed L. bicolor and P. arhizus mycorrhizas at the three rates containing both fungi. L. bicolor behaved as an aggressive root colonizer and its level of root colonization remained constant at increasing rates of P. arhizus inoculum. L. bicolor displaced R. subareolatus at all inocula rates. P. arhizus displaced R. roseolus except at the rate 3Pa + 9Rr, with only a low number of mycorrhizas formed by either fungus. Total plant biomass was significantly increased by the presence of any fungal combination up to four times the values for uninoculated controls. P. arhizus and R. subareolatus were more effective in promoting plant growth and stimulating short root formation than either L. bicolor or R. roseolus.  相似文献

Picea sitchensis and Pseudotsuga menziesii seedlings were grown in containers, inoculated with ectomycorrhizal fungi, and planted in British forestry sites. Root samples taken during the year after planting were assessed for mycorrhiza formation. Survival and shoot height were assessed at the end of each year. Observations were made each autumn on the occurrence of sporophores of ectomycorrhizal fungi. Pot experiments were used to assess the colonization potential of soils from the experimental locations. Assessment of mycorrhiza formation by the inoculant fungi both before planting and the following year showed much variation among the fungi used. Similar variation was found among field sites. Inoculation with Laccaria isolates was most successful. Height measurements are reported for the first 2 years after planting, at which time there were few significant effects on growth of Picea sitchensis or Pseudotsuga menziesii seedlings. Experimental assessment of colonization potential was of little value in this work for predicting events in the forest.  相似文献

Ten isolates of six species of ectomycorrhizal fungi were grown in vitro at nine concentrations of three sodium salts (NaCl, Na2SO4, Na3C6H5O7) for 4 weeks. Colony diamater, biomass and protein content of fungi were evaluated. Isolates of Pisolithus tinctorius and Suillus luteus were more tolerant of NaCl and Na2SO4 than of Na3C6H5O7. Fungi in the genera Cenococcum, Laccaria, and Thelephora were highly intolerant of Na3C6H5O7 and Na2SO4 in vitro. Biomass and protein content of fungi generally declined with increasing substrate salinity in solution culture. In situ ectomycorrhizal colonization by Laccara laccata and P. tinctorius and the dry weight of Pinus taeda seedlings were significantly reduced by 80 mM NaCl after 14 weeks. Only select ectomycorrhizal fungi appear capable of growth and symbiosis in saline soils.  相似文献

Two sensitive methods for the enzymatic determination of mannitol are described which were applied to fungal and mycorrhizal extracts. Both methods are based on the oxidation of mannitol by mannitol dehydrogenase from Agaricus hortensis and the fluorometric determination of the NADPH produced in this reaction. The detection limits are 125 pmol for the direct fluorometric assay and 100 fmol, when enzymatic cycling of NADPH is included. The levels of mannitol detected were 123 pmol/g dry wt (mycelia from Cenococcum geophilum, cultivated on malt medium), below 0.3 or about 2.4 pmol/g dry wt (mycelia from Amanita muscaria, dependent on carbon source in the cultivation medium), and between 1.9 and 5.2 pmol/g dry wt in mycorrhizal short roots of Picea abies/Amanita muscaria.  相似文献

In vitro weathering of phlogopite by ectomycorrhizal fungi   总被引:1,自引:0,他引:1  
The ways in which ectomycorrhizal fungi benefit tree growth and nutrition have not been fully elucidated. Whilst it is most probably due to improved soil colonization, it is also likely that ectomycorrhizal fungi could be directly involved in nutrient cycling of soil reserves. This study assessed access by two species of ectomycorrhizal fungi to soil nonexchangeable K+ reserves. The incubation of ectomycorrhizal fungi in bi-compartment Petri dishes with phlogopite led to cation exchange reactions and to crystal lattice weathering. Paxillus involutus COU led to irreversible phlogopite transformations, while Pisolithus tinctorius 441 led to reversible ones. Simultaneous depletion in K+ and Mg2+ led to an enhanced weathering of phlogopite by P. tinctorius 441. The observation of phlogopite evolution shows that some specific Al3+ immobilization occurred under P. tinctorius 441. The data suggest that these bio-weathering mechanisms could be related to the release of fungal organic acids or other complex forming molecules.  相似文献

Th effect of fenvalerate 20% EC (a synthetic pyrethroid) on the growth of two ectomycorrhizal fungi, Thelephora terrestris and Laccaria laccata, was studied in vitro. In the case of T. terrestris, mycelial growth was promoted at lower concentrations of the insecticide but inhibited at higher concentrations; mycelial growth of L. laccata decreased with increase in concentration. The effect of the insecticide at three concentrations (0.25, 0.5, 1.5 ppm) was also studied on ectomycorrhizal formation in 4-month-old seedlings of Pinus patula in nursery trials. At lower concentrations the insecticide had no significant effect on mycorrhizal formation.  相似文献

Seedlings of pine, infected with two different ectomycorrhizal fungi, Suillus bovinus (Fr.) O. Kuntze and an unidentified isolate (BP), were exposed to various external Zn concentrations. An additional strain of S. bovinus, cultured on a Zn-enriched medium, was also used. The effect of mycorrhizal associations on Zn uptake and distribution within the plant was determined by AAS.The results show that nonmycorrhizal seedlings have the capability to control the uptake and translocation of Zn to the shoot until the external Zn concentration reaches a threshold level, where no limitation of uptake is possible. Excess Zn is accumulated in the root system to protect the shoot against toxic tissue concentrations. The effect of an ectomycorrhizal infection on Zn uptake and distribution depends on (1) the fungal species (2) the external concentration and (3) the Zn content of the fungal culture medium. Under conditions of low external Zn supply, especially a mycorrhizal infection with S. bovinus led to an increased Zn uptake in root and needles of Pinus sylvestris. Under high external conditions the mycobionts varied considerably in their capability to reduce the transport of Zn to the shoot. Only by an infection with S. bovinus the plant was able to maintain the shoot tissue concentration on a low level. This effect can be enhanced by pretreatment of S. bovinus with high Zn concentrations.  相似文献

Oxalate accumulation in external medium under hyphal mats of two ectomycorrhizal species is strongly stimulated (1.7 to 35 fold) by a simultaneous depletion of available K+ and Mg2+. Pisolithus tinctorius strain 441 accumulates oxalate both on NH4–N and on NO3–N whereas Paxillus involutus strain COU only accumulates oxalate on NO3–N. On NO3–N, under a simultaneous K+ and Mg2+ deficiency, P. involutus COU is a very active oxalate producer compared to P. tinctorius 441. The present results could explain the various mineralogical evolutions of a phlogopite mica previously recorded under P. involutus COU or P. tinctorius 441 and suggest a key role for fungal oxalic acid during mineral weathering in response to nutrient deficiency.  相似文献

Fruit-body production of two ectomycorrhizal fungi,Hebeloma radicosum andhebeloma sp. (nagaenosugitakedamashi in Japanese), in pure culture was examined. First, nutrients that promote mycelial growth of the fungi when added to the basal medium consisting of barley grains and sawdust were determined. Then the fungi were cultivated to produce fruit-bodies in larger-scale media containing additional nutrients selected for each fungus. Mature fruit-bodies bearing basidiospores were formed after incubation at 22°C for 35–42 d, followed by incubation at 17°C for 21–32 d.  相似文献

Summary Phosphorus-31 nuclear magnetic resonance spectra at 36.4 MHz are presented for intact ectomycorrhizal fungi grown in pure culture. Resonances from polyphosphates and intracellular orthophosphate are identified inCenococcum graniforme, Hebeloma cylindrosporum, andH. crustuliniforme. Comparison of the NMR spectra with phosphorus fractionation of the fungi extracts leads to the statement that the NMR-observed polyphosphaes is a good part of the accumulated polyphosphates. Transfer of 15N between interacting mycelia of a wood-decomposing fungus (Hypholoma fasciculare) and an ectomycorrhizal fungus (Tomentellopsis submollis) was studied in a mature beech (Fagus sylvatica) forest. The amount of 15N transferred from the wood decomposer to the ectomycorrhizal fungus was compared to the amount of 15N released from the wood-decomposing mycelia into the soil solution as 15N-NH4. The study was performed in peat-filled plastic containers placed in forest soil in the field. The wood-decomposing mycelium was growing from an inoculated wood piece and the ectomycorrhizal mycelium from an introduced root from a mature tree. The containers were harvested after 41 weeks when physical contact between the two foraging mycelia was established. At harvest, 15N content was analyzed in the peat (total N and 15NH4 +) and in the mycorrhizal roots. A limited amount of 15N was transferred to the ectomycorrhizal fungus and this transfer could be explained by 15NH4 + released from the wood-decomposing fungus without involving any antagonistic interactions between the two mycelia. Using our approach, it was possible to study nutritional interactions between basidiomycete mycelia under field conditions and this and earlier studies suggest that the outcomes of such interactions are highly species-specific and depend on environmental conditions such as resource availability.  相似文献

Three wheat cultivars with different tolerances against free aluminium were grown monoxenically in association with Azospirillum brasilense. In situ nitrogen fixation, measured with the acetylene reduction assay, was higher by the aluminium-tolerant cultivars than by the sensitive cultivar. The transfer of fixed nitrogen to the host plant, determined by the 15N dilution technique, was also significantly higher in the aluminium-resistant wheat plants. The total accumulation of fixed nitrogen in the host plants due to an A. brasilense inoculation varied from approximately 13% to 17% of the total nitrogen in the root and 2.9% to 3.9% of the nitrogen in the shoot.The quantity and quality of exudates released in liquid nutrient solution were analysed separately for two of the wheat cultivars, one aluminium-tolerant and one aluminium-sensitive. After 29 days of growth the aluminium-tolerant plants exudated significantly higher total amounts of carbon than aluminium-sensitive plants. No differences between the two cultivars existed in the carbon exudation rate per gram dry root.Much higher concentrations of low molecular dicarboxylic acids i.e. succinic, malic and oxalic acid, were found in the exudates of aluminium-tolerant plants. Dicarboxylic acids are potential chelating compounds for positively charged metals such as aluminium and they may play an important role in protecting the plant against aluminium incorporation. They are also very suitable substrates for Azospirillum spp. It is therefore suggested that these factors may be causing the higher associative nitrogen fixation rates which was found in the aluminium-tolerant wheat cultivars.  相似文献

Summary Young pine seedlings were inoculated with mycelial cultures of 8 fungi, and basidiospore cultures of 4 fungi. Four months later distinctive mycorrhizas had been formed by all 4 spore cultures and by 5 of the mycelial cultures.At 10 months one further mycelial culture had given rise to distinctive mycorrhizas. However, by that time contamination between treatments, or from outside, was at a higher level.Survival of outplanted seedlings after 6 months was generally poor, but was slightly greater in some treatments.  相似文献

Ectomycorrhizal fungi (Paxillus involutus, Suillus grevillei and two unidentified basidiomycetes from excised Sitka spruce mycorrhizas) were isolated from stands of Sitka spruce either in monoculture or in a mixture with Japanese larch in an Irish conifer plantation. The growth of these fungi and their mycorrhizal formation in Sitka spruce and Japanese larch were examined after incubation in modified Melin-Norkrans medium containing either KH2PO4, Ca3(PO4)2 or Fe phytate as the phosphorus (P) source. P. involutus and S. grevillei utilized all three P sources. The unidentified basidiomycetes had limited ability to utilize Fe phytate. Basidiomycete 1 showed poor growth on KH2PO4 whereas growth of basidiomycete 2 was low on Ca3(PO4)2. Pure culture synthesis studies confirmed that P. involutus and the two basidiomycetes formed mycorrhizas with both tree species but S. grevillei was mycorrhizal only on Japanese larch. P. involutus formed more mycorrhizas in both conifers than the other fungi. Following inoculation with each of the four fungi, shoot and root dry mass of both Sitka spruce and Japanese larch seedlings was enhanced compared with uninoculated/nonmycorrhizal controls. On Fe phytate, Paxillus-inoculated Sitka spruce seedlings had the lowest primary root length and on KH2PO4, Suillus-inoculated Japanese larch had the greatest number of short roots. The only differences when Sitka spruce seedlings were grown in either monoculture or in a mixture with Japanese larch mycorrhizal with S. grevillei were primary root length and number of short roots after growth on media containing Fe phytate.  相似文献

The ectomycorrhizal fungi Gautieria monticola and Hysterangium setchellii both form dense hyphal mats in coniferous forest soils of the Pacific Northwest. We recently observed that all Douglas-fir seedlings found under the canopy of a maturing 60–75 year stand were associated with mats formed by ectomycorrhizal fungi. The significance of these mat communities in relation to seedling establishment and survival is discussed.  相似文献

The interactions of the collembolan insect Proisotoma minuta with ectomycorrhizal and/or pathogenic fungi was examined in three experiments: (1) in vitro analysis of feeding patterns, (2) in vitro food preference test, and (3) in situ analysis of ectomycorrhizal colonization in relation to population density. The ectomycorrhizal fungi Laccaria laccata, Pisolithus tinctorius, Suillus luteus, Thelephora terrestris and the pathogenic fungi Rhizoctonia solani were employed in all experiments. In vitro and in situ experiments revealed that Pr. minuta consumed all the ectomycorrhizal fungi tested but the feeding pattern and consumption varied with each isolate. In a comparative in vitro feeding preference test, where Pr. minuta was given a choice, R. solani was grazed more heavily than the ectomycorrhizal fungi. Among the ectomycorrhizal fungi examined, Pi. tinctorius was consumed significantly less than L. laccata, S. luteus or T. terrestris in the presence of R. solani. A 10-week in situ analysis of loblolly pine (Pinus taeda L.) seedling root systems inoculated with Pr. minuta revealed that ectomycorrhizal colonization was significantly less than that of control plants (without Pr. minuta). Collectively, these data suggest that mycophagous Collembola may play a major role in the distribution and biomass of ectomycorrhizal fungi in the rhizosphere of tree seedlings.  相似文献

The production of proteolytic enzymes by several strains of ectomycorrhizal fungi i.e., Amanita muscaria (16-3), Laccaria laccata (9-12), L. laccata (9-1), Suillus bovinus (15-4), Suillus bovinus (15-3), Suillus luteus (14-7) on mycelia of Trichoderma harzianum, Trichoderma virens and Mucor hiemalis and sodium caseinate, yeast extract was evaluated. The strains of A. muscaria (16-3) and L. laccata (9-12) were characterized by the highest activity of the acidic and neutral proteases. Taking the mycelia of saprotrophic fungi into consideration, the mycelium of M. hiemalis was the best inductor for proteolytic activity. The examined ectomycorrhizal fungi exhibited higher activity of acidic proteases than neutral ones on the mycelia of saprotrophic fungi, which may imply the participation of acidic proteases in nutrition.  相似文献

The survival, development and mycorrhizal efficiency of a selected strain of Laccaria bicolor along with naturally occurring ectomycorrhizal fungi in a young plantation of Douglas fir was examined. Symbionts were identified and their respective colonization abilities were determined. Eight species of symbiotic fungi, which may have originated in adjacent coniferous forests, were observed on the root systems. Mycorrhizal diversity differed between inoculated (5 taxa) and control (8 taxa) seedlings. Ectomycorrhizal fungi which occurred naturally in the nursery on control seedlings (Thelephora terrestris and Suillus sp.) did not survive after outplanting. Both inoculated and naturally occurring Laccaria species, as well as Cenococcum geophilum, survived on the old roots and colonized the newly formed roots, limiting the colonization by other naturally occurring fungi. Other fungi, such as Paxillus involutus, Scleroderma citrinum and Hebeloma sp. preferentially colonized the old roots near the seedling's collar. Russulaceae were found mainly in the middle section of the root system. Mycorrhizal colonization by Laccaria species on inoculated seedlings (54%) was significantly greater than on controls (13%) which were consequently dominated by the native fungi. Significant differences (up to 239%) were found in the growth of inoculated seedlings, especially in root and shoot weight, which developed mainly during the second year after outplanting. Seedling growth varied with the species of mycorrhizae and with the degree of root colonization. Competitiveness and effectiveness of the introduced strain on improving growth performances of seedlings are discussed.  相似文献

Trends of pollen grain aperture evolution and exine characters as well as characters of leaf venation, petiole and axial vascularization are briefly described and related to geographical distribution and classification ofBuxus. A review of fossil records is given. Three major taxonomic groups can be delimitated within the genus, and aspects of their relationships and chorogenesis are presented. The level of differentiation, the pattern of distribution and the fossil record speak in favour of an ancient origin of the genus.  相似文献

As part of a wider study into the role of soil fungi in the ecology of the winter annual grass, Vulpia ciliata ssp. ambigua (Le Gall) Stace & Auquier, we applied the fungicides benomyl and prochloraz to three natural populations of the grass growing in East anglia, United Kingdom. The rhizosphere and rootinfecting fungi associated with the three populations were analysed each month between February and May 1992 when plants set seed. There were marked differences between the fungal floras associated with each of the three populations of V. ciliata, despite the fact that associated plant species and soil nutrient status were broadly similar between sites. This was attributed to wide differences in soil pH between the three populations. Prochloraz did not affect fungal abundance, but benomyl decreased the isolation frequencies of Fusarium oxysporum from roots and the frequencies of Penicillium and Trichoderma spp. isolated from rhizosphere soil, and increased the frequency of isolation of Mucor hiemalis from the rhizosphere of V. ciliata. There were also significant increases in the isolation frequencies of F. oxysporum from roots and M. hiemalis, Trichoderma spp. and Phoma fimeti from the rhizosphere of V. ciliata as plants matured. The significance of these results for the design of ecological field experiments are discussed in light of a previous study which has shown that asymptomatic root-infecting fungi can affect plant fecundity and hence abundance in natural populations of V. ciliata. We propose that differences in microbial communities between sites, controlled in part by soil chemistry, are a major factor determining plant performance under field conditions.  相似文献
