The pigment-binding proteins of Rhodobacter capsulatus are encoded by the polycistronic puf and puc operons. Both operons show higher expression under low oxygen tension than under high oxygen tension in the wild-type strain. The Tn5 mutant strain AH2 shows only low levels of puf and puc mRNA under high and low oxygen tension, indicating that it lacks a gene product required for stimulation of puf and puc gene expression under low oxygen tension. The formation of wild-type levels of photosynthetic complexes and normal oxygen regulation could be restored by the expression in trans of a 1.7 kb fragment of the R. capsulatus wild-type chromosome or by addition of 10μg I-1 vitamin B12 to the growth medium. An open reading frame of 798 nucleotides containing the Tn5 insertion was identified on the 1.7kb fragment. This open reading frame shows no homology to known genes and has a remarkably high GC content of 76%.  相似文献

The expression of many genes of facultatively photosynthetic bacteria of the genus Rhodobacter is controlled by the oxygen tension. Among these are the genes of the puf and puc operons, which encode proteins of the photosynthetic apparatus. Previous results revealed that thioredoxins are involved in the regulated expression of these operons, but it remained unsolved as to the mechanisms by which thioredoxins affect puf and puc expression. Here we show that reduced TrxA of Rhodobacter capsulatus and Rhodobacter sphaeroides and oxidized TrxC of R.capsulatus interact with DNA gyrase and alter its DNA supercoiling activity. While TrxA enhances supercoiling, TrxC exerts a negative effect on this activity. Furthermore, inhibition of gyrase activity strongly reduces puf and puc expression. Our results reveal a new signaling pathway by which oxygen can affect the expression of bacterial genes.  相似文献

Rhodobacter sphaeroides has been intensively studied and provides an excellent model for studying both photo-synthesis and membrane development. The photosynthetic apparatus (LH2 and LH1-RC complexes) can be synthesized in large scale and integrated into the intracytoplasmic membrane system under specific conditions, which thus provides us insight to utilize the puc or(and) puf operon to heterologously express recombinant proteins in the intracytoplasmic membrane using Rb. sphaeroides as a novel expression system. However, basal level of expression of puc and puf promoter is uncontrolled. We report the construction of LH2 polypeptide expression vector that contains a reengineered lacI q-puc promoter-lac operator hybrid promoter, which allows the puc operon to be regulated by both IPTG and low oxygen level. Synthesis of LH2 complexes was completely repressed in the absence of isopropyl β-D-thiogalactoside (IPTG), and the degree of induction was controlled by varying the concentration of IPTG. The optimal concentration of IPTG was determined. SDS-PAGE and Western blot were employed for further analysis. Our results suggest that the reengineered hybrid promoter is efficient to tightly regulate the expression of the puc operon, and our strategy can open up a new approach in the study of the membrane protein expression system.  相似文献

We quantified the expression of the puf and puc operons, which encode pigment binding proteins, in a number of strains of Rhodobacter capsulatus that have defects in genes affecting different steps of bacteriochlorophyll biosynthesis. Our results show that these mutations have a very similar effect on puf and puc expression. This suggests that the reduced expression of genes encoding pigment-binding proteins is due neither to the accumulation of a specific intermediate of bacteriochlorophyll synthesis nor to the lack of an early intermediate, but is rather the result of the lack of a very late intermediate or the bacteriochlorophyll molecule itself. The effects of light on the accumulation of bacteriochlorophylland carotenoids were investigated in an aerobic photosyntheticbacterium, Roseobacter denitrificans during anaerobic respiration.Accumulation of pigments occurred in darkness but not in whitelight, with the growth rate being similar under both dark andlight conditions. Once pigments had accumulated during growthin darkness, subsequent irradiation with white light did notresult in degradation of the accumulated pigments, an indicationthat the pigments were stabilized in the membranes. The presentresults, therefore, exclude the possibility of inhibition ofthe accumulation of the photosynthetic pigments by the photochemicaldegradation of the pigments in the presence of molecular oxygenand light (blue light). The action spectrum for the inhibitionof the accumulation of the pigments showed that light at 470nm was the most effective and light at wavelengths longer than500 nm had little inhibitory effect. Together with previousresults [Shimada et al. (1992) Plant Cell Physiol. 33: 471],the present data suggest that a signal-transduction system associatedwith an unidentified blue pigment(s) is involved in the inhibitionof the accumulation of the photosynthetic pigments in R. denitrificans. Cytochrome systems in cells of a denitrifying bacterium, Pseudomonasstutzeri (VAN NIEL strain), grown under different atmosphericconditions were compared with reference to the effects of nitrateand nitrite on cytochrome synthesis. When a culture was sufficiently aerated (aerobic conditions),synthesis of all cytochrome components was repressed, regardlessof the presence or absence of nitrate and nitrite. When aeratedmoderately (semi-aerobic conditions), both soluble and paniculatecytochromes c-552 and cytochrome b-558 contents markedly increasedeven in the absence of nitrate and nitrite. Under anaerobic or semi-aerobic conditions, nitrite inducedcytochrome a2–c synthesis. This inductive effect of nitritewas counteracted by nitrate. Photosynthetically active chimeric reaction centers which utilize genetic information from both Rhodobacter capsulatus and Rb. sphaeroides puf operons were isolated using a novel method termed chimeric rescue. This method involves in vivo recombination repair of a Rb. capsulatus host operon harboring a deletion in pufM with a non-expressed Rb. sphaeroides donor puf operon. Following photosynthetic selection, three revertant classes were recovered: 1) those which used Rb. sphaeroides donor sequence to repair the Rb. capsulatus host operon without modification of Rb. sphaeroides puf operon sequences (conversions), 2) those which exchanged sequence between the two operons (inversions), and 3) those which modified plasmid or genomic sequences allowing expression of the Rb. sphaeroides donor operon. The distribution of recombination events across the Rb. capsulatus puf operon was decidedly non-random and could be the result of the intrinsic recombination systems or could be a reflection of some species-specific, functionally distinct characteristic(s). The minimum region required for chimeric rescue is the D-helix and half of the D/E-interhelix of M. When puf operon sequences 3 of nucleotide M882 are exchanged, significant impairment of excitation trapping is observed. In this paper we show the effect of oxygen and light on the expression of the photosynthetic apparatus of a mutant heterologously expressing the puc operon. This mutant was obtained by introducing in trans an expression plasmid, bearing the puc A, B, and C genes of Rhv. sulfidophilum, as well as its own promoter, in an LHII mutant of Rb. capsulatus. The results showed that oxygen and light repressed LHII expression. Even low-light intensities lowered the LHII content to undetectable levels by spectrophotometry or by SDS-PAGE. In high-light grown cells, where the relative ratios of LHI and LHII complexes were significantly diminished, we were able to detect LHII complexes. Under the latter condition, the absorption spectrum showed that some pigment accumulated in the membrane even in the absence of cell division. These pigments were used in a later step to assemble LHII complexes, when the high-light grown cells were transferred to semiaerobiosis in the dark. Transition of high-light grown cells to low-light conditions allowed us to study the adaptability of these heterologous mutant cells. Absorption and fluorescence excitation spectra were measuredfor batch cultures of five species of marine phytoplankton grownunder high and low light. These spectra were examined for propertiescharacteristic of taxonomic position and of photoadaptive response.While regions of absorption and excitation of chlorophyll afluorescence diagnostic of pigment composition were identifiable,photoadaptive response had greater influence on spectral variability.Although reduced growth irradiance caused changes in both theabsorption and fluorescence excitation spectra, the fluorescenceexcitation spectrum appears to be more sensitive to alterationsin the ambient light field for growth than does the absorptionspectrum. For a single species. the fluorescence excitationspectrum for a sample grown at low irradiance showed greaterstructure than that for the sample grown at a high irradiance.Under low light conditions, the excitation of chlorophyll afluorescence by accessory pigments increased relative to theexcitation by chlorophyll a itself The highest fluorescenceyields occur in the blue-green region of the spectrum, correspondingto bands of peak absorption by the accessory pigments. Changesin absorption spectra are less marked, but two features recur.First. in the blue-green region of the spectrum from -500–560nm. absorption is enhanced in the low-light cells relative tothat of the high-light cells. Second, the ratio of absorptionat 435 nm to that at 676 nm was greater for the high-light cells.Correlating changes in pigment concentrations were observed.The influence of photoadaptation on the properties of fluorescenceexcitation spectra is as great or greater than the influenceof pigment complements characteristic of specific algal taxa.  相似文献

L-Pipecolic acid was found to be effective in inducing floweringof Lemna paucicostata 151, 381, 441 and 6746, and of Lemna gibbaG3. When the plants were grown on half-strength Hutner's medium,L-pipecolic acid caused profuse flowering of L. paucicostata151 maintained under 9 and 10 h of light daily. In L. paucicostata441 and 6746, L-pipecolic acid had a strong flower-promotingeffect under a near critical photoperiod. In L. paucicostata381, by contrast, L-pipecolic acid had only a very small effecton flowering. In L. gibba G3 substantial promotion of floweringwas observed under continuous light. When one-twentieth-strengthHutner's medium was used as the basic medium, L-pipecolic acidstimulated flowering in all strains of Lemna examined, evenunder continuous light. When L. paucicostata 151 was grown on one-tenth-strength M mediumor one-twentieth-strength Hutner's medium, the flower-inducingactivity of L-pipecolic acid was greatly enhanced by cytokininunder continuous light. However, when this strain was grownwith 9 h of illumination daily, this synergistic effect of cytokininwas only slight. A short-term (even 1-h) treatment with L-pipecolicacid resulted in flowering, suggesting that L-pipecolic acidis involved in the induction of flowering, rather than its evocation.D-Pipecolic acid also had flower-inducing activity, but itsactivity was 50 times lower than that of the L-isomer. The effect of light on the activity of 3-hydroxy-3-methylglutarylCoenzyme A (HMG-CoA) reductase in Rhodotorula minuta was studiedin cell-free extracts prepared from cells grown under variouslight conditions. HMG-CoA reductase activity in cells grown under continuous illuminationwas higher than that in cells grown in the dark, and dependedon the light intensity used during incubation. The relationshipbetween activity [A (nmol/mg-N/min)] and light intensity [I(erg/cm2/sec)] was expressed by the equation A=0.72 log I$0.80. Illumination at –1.5?C followed by dark incubation at26?C resulted in a rapid increase in HMG-CoA reductase activityimmediately after the beginning of incubation. This photoinducedHMG-CoA reductase activity was regulated by the light dose andfollowed the Roscoe-Bunsen reciprocity law. When cycloheximide was added immediately after the beginningof incubation in the dark, the increase in HMG-CoA reductaseactivity was completely inhibited. The inhibitory effect ofcycloheximide, however, gradually decreased with the delay ofthe addition. On the basis of these results we have postulated that the photoregulationof carotenogenesis in Rh. minuta results from the photoregulationof HMG-CoA reductase synthesis. The electron transport system involved in nitrate reductionand its relationship to photosynthetic cyclic electron transportin a photodenitrifier, Rhodopseudomonas sphaeroides forma sp.denitrificans, were studied. Nitrate oxidized only b-type cytochromein the presence of cyanide, which inhibits nitrite reductase.Heptylhydroxyquinoline-N-oxide (HOQNO) inhibited the oxidationof b-type cytochrome by nitrate, but not the oxidation of b-and c-type cytochrome by nitrite. The inhibition by HOQNO wasovercome by phenazine methosulfate (PMS). Absorption changesof b-type cytochrome induced by illumination were in just theopposite directions for oxygen- and nitrate-oxidized cells;the cytochrome was reduced in oxygen-oxidized cells and oxidizedin nitrate-oxidized cells. Antimycin enhanced the reductionand inhibited the oxidation, but had no inhibitory effect onthe oxidation of b-type cytochrome by nitrate. Dithionite-reducedminus ferricyanide-oxidized difference spectra of cells at 77?Kshowed two b-type cytochrome components with a bands at 556.5and 562 nm. The proportion of the b-562 component decreasedin cells grown under denitrifying conditions. Photosynthetic bradyrhizobia are nitrogen-fixing symbionts colonizing the stem and roots of some leguminous plants like Aeschynomene. The effect of oxygen and light on the formation of the photosynthetic apparatus of Bradyrhizobium sp. C7T1 strain is described here. Oxygen is required for growth, but at high concentration inhibits the synthesis of bacteriochlorophyll (BChl) and of the photosynthetic apparatus. However, we show that in vitro, aerobic photosynthetic electron transport occurred leading to ADP photophosphorylation. The expression of the photosynthetic apparatus was regulated by oxygen in a manner which did not agree with earlier results in other photosynthetic bradyrhizobia since BChl accumulation was the highest under microaerobic conditions. This strain produces photosynthetic pigments when grown under cyclic illumination or darkness. However, under continuous white light illumination, a Northern blot analysis of the puf operon showed that, the expression of the photosynthetic genes of the antenna was considerable. Under latter conditions BChl accumulation in the cells was dependent on the oxygen concentration. It was not detectable at high oxygen tensions but became accumulated under low oxygen (microaerobiosis). It is known that in photosynthetic bradyrhizobia bacteriophytochrome photoreceptor (BphP) partially controls the synthesis of the photosystem in response to light. In C7T1 strain far-red light illumination did not stimulate the synthesis of the photosynthetic apparatus suggesting the presence of a non-functional BphP-mediated light regulatory mechanism.  相似文献

The steady state of photosynthetic electron transport drivenby two photosystems was studied with cells of the cyanophyteSynechocystis PCC 6714 by analyzing the flash-induced oxidation-reductionof Cyt f and P700 under continuous background illumination.We first analyzed the spectra and the kinetics of flash-inducedabsorption changes in the 400 to 440 nm wavelength region anddefined the absorption changes due to oxidation-reduction ofCyt f and P700. Results indicated that the flash-induced absorptionchanges at 420 and 435 nm are due to the oxidation-reductionof Cyt f and P700, respectively. Determination of the steadystate of Cyt f (420 nm) and P700 (435 nm) was made for the cellsgrown under a weak orange light exciting mainly PS II (PS IIlight) and having a high ratio of PS I to PS II (PS I/PS II),and those grown under a weak red light exciting preferentiallyPS I (PS I light) and having a low PS I/PS II. The steady stateof electron transport in cells of the two types were comparedunder PS I and PS II lights. The results indicated that: (1)under the light conditions used for growth (both red and orangelight), the intermediate electron pool between the two photosystemsremained in a redox state so as to keep both photosystems inthe open state. (2) When shifted to PS I light, the intermediatepool and PS I in cells of high PS I/PS II became extremely electron-poor,and so most of the PS I reaction centers were closed. (3) Theintermediate pool in cells of low PS I/PS II became extremelyelectron-rich when shifted to PS II light, and most of the PSII reaction centers were closed. The electron transport stateis released from such biased states by regulation of PS I/PSII. Results supported our previously proposed hypothesis thatthe stoichiometry between PS I and PS II is regulated so asto keep the two photosystems in the open state.
