Photoinhibition of photosynthesis was investigated in control (C) and chilling night (CN) leaves of grapevine under natural photoperiod at different sampling time in a day. The degree of photoinhibition was determined by means of the ratio of variable to maximum chlorophyll fluorescence (Fv/Fm) and photosynthetic electron transport measurements. When the potential efficiency of photosystem (PS) 2, Fv/Fm was measured at midday, it markedly declined with significant increase of F0 in CN leaves. In isolated thylakoids, the rate of whole chain and PS2 activity were markedly decreased in CN leaves than control leaves at midday. A smaller inhibition of PS1 activity was also observed in both leaf types. Later, the leaves reached maximum PS2 efficiencies similar to those observed in the morning during sampling at evening. The artificial exogenous electron donors diphenyl carbazide, NH2OH, and Mn2+ failed to restore the PS2 activity in both leaf types at midday. Thus CN enhanced inactivation on the acceptor side of PS2 in grapevine leaves. Quantification of the PS2 reaction centre protein D1 following midday exposure of leaves showed pronounced differences between C and CN leaves. The marked loss of PS2 activity in CN leaves noticed in midday samples was mainly due to the marked loss of D1 protein of the PS2 reaction centre.  相似文献

Photoinhibition under irradiance of 2 000 μmol m−2 s−1 (HI) was studied in detached control (C) and water deficit (WD) leaves of grapevine (Vitis vinifera L.) plants. The degree of photoinhibition was determined by means of the ratio of variable to maximum chlorophyll (Chl) fluorescence (Fv/Fm) and electron transport measurements. The potential efficiency of photosystem (PS) 2, Fv/Fm, marginally declined under HI in WD-leaves without significant increase of F0. In contrast, Fv/Fm ratio declined markedly with significant increase of F0 in C-leaves. In isolated thylakoids, the rate of whole chain and PS2 activity under HI were more decreased in C-than WD-leaves. The artificial exogenous electron donors diphenyl carbazide, NH2OH, and Mn2+ failed to restore the HI-induced loss of PS2 activity in both C-and WD-leaves. Thus HI operates at the acceptor side of PS2 in both leaf types. Quantification of the PS2 reaction centre protein D1 following HI exposure of leaves showed pronounced differences between C-and WD-leaves. The marked loss of PS2 activity under HI of C-leaves was due to the marked loss of D1 protein of the PS2 reaction centre.  相似文献

The degree of photoinhibition was determined by means of the ratio of variable to maximum chlorophyll fluorescence (Fv/Fm) and photosynthetic electron transport measurements. When the photochemical efficiency of PS2, Fv/Fm, markedly declined, F0 increased significantly in leaves of V. berlandieri, while F0 did not increase in V. rupestris leaves. Isolated thylakoids of leaves of V. berlandieri showed significant inhibition of whole chain and PS2 activities at midday. A smaller inhibition was observed for V. rupestris. Later, the leaves reached maximum PS2 efficiencies similar to those observed early in the morning during sampling at evening. The artificial exogenous electron donor Mn2+ failed to restore PS2 activity in both species, while DPC and NH2OH significantly restored PS2 activity in V. rupestris midday leaf samples. Quantification of the PS2 reaction centre protein D1 and 33 kDa protein of water splitting complex following midday exposure of leaves showed pronounced differences between V. berlandieri and V. rupestris leaves. The marked loss of PS2 activity noticed in midday samples was mainly due to the marked loss of D1 protein in V. berlandieri while in V. rupestris it was the 33 kDa protein.  相似文献

The degree of photoinhibition of sun and shade grown leaves of grapevine was determined by means of the ratio of variable to maximum chlorophyll (Chl) fluorescence (Fv/Fm) and electron transport measurements. The potential efficiency of photosystem 2 (PS2), Fv/Fm, markedly declined under high irradiance (HI) in shade leaves with less than 10 % of F0 level. In contrast, Fv/Fm ratio declined with about 20 % increase of F0 level in sun leaves. In isolated thylakoids, the rate of whole chain and PS2 activity in HI shade and sun leaves was decreased by about 60 and 40 %, respectively. A smaller inhibition of photosystem 1 (PS1) activity was also observed in both leaf types. In the subsequent dark incubation, fast recovery was observed in both leaf types that reached maximum PS2 efficiencies similar to non-photoinhibited control leaves. The artificial exogenous electron donors DPC, NH2OH, and Mn2+ failed to restore the HI-induced loss of PS2 activity in sun leaves, while DPC and NH2OH were significantly restored in shade leaves. Hence HI in shade leaves inactivates on the donor side of PS2 whereas it does at the acceptor side in sun leaves, respectively. Quantification of the PS2 reaction centre protein D1 and the 33 kDa protein of water splitting complex following HI-treatment of leaves showed pronounced differences between shade and sun leaves. The marked loss of PS2 activity in HI leaves was due to the marked loss of D1 protein of the PS2 reaction centre protein and the 33 kDa protein of the water splitting complex in sun and shade leaves, respectively.  相似文献

In six dominant species of the Amazonian ‘Bana’ vegetation, leaf blade characteristics, pigment composition, and chlorophyll (Chl) fluorescence parameters were measured in young and mature leaves under field conditions. Leaf δ13C was comparable in the six species, which suggested that both expanding and expanded leaves contained organic matter fixed under similar intercellular and ambient CO2 concentration (C i/C a). High leaf C/N and negative δ15N values found in this habitat were consistent with the extreme soil N-deficiency. Analysis of Chl and carotenoids showed that expanding leaves had an incomplete development of photosynthetic antenna when compared to adult leaves. Dynamic inactivation of photosystem 2 (PS2) at midday was observed at both leaf ages as Fv/Fm decreased compared to predawn values. Adult leaves reached overnight Fv/Fm ratios typical of healthy leaves. Overnight recovery of Fv/Fm in expanding leaves was incomplete. F0 remained unchanged from midday to predawn and Fv tended to increase from midday to predawn. The recovery from midday depression observed in adult leaves suggested an acclimatory down-regulation associated with photo-protection and non-damage of PS2.  相似文献

Infiltration of methyl viologen (MV, source of O2 ) and Na-diethyldithiocarbamate (DDC, inhibitor of SOD) into wheat leaves resulted in the accumulation of active oxygen species and photo-oxidative damage to photosynthetic apparatus under both moderate and high irradiance. Exogenous antioxidants, ascorbate (ASA) and mannitol, scavenged active oxygen efficiently, protected the photosynthetic system from MV and DDC induced oxidative damage, and maintained high Fv/Fm [maximal photochemical efficiency of photosystem 2 (PS2) while all PS2 reaction centres are open], Fm/F0 (another expression for the maximal photochemical efficiency of PS2), PS2 (actual quantum yield of PS2 under actinic irradiation), qP (photochemical quenching coefficient), P N (net photosynthetic rate), and lowered qNP (non-photochemical quenching coefficient) of the leaves kept under high irradiance and oxidative stress. Phenolic compounds used in these experiments, catechol (Cat), resorcinol (Res), and tannic acid (Tan), had similar anti-oxidative activity and protective effect on photosynthetic apparatus as ASA and mannitol. The anti-oxidative activity and the protective effect of phenolic compounds increased with increase in their concentration from 100 to 300 g m–3. The number and the position of hydroxyl group in phenolic molecules seemed to influence their antioxidative activity.  相似文献

Photoinhibition of photosynthesis was studied in young and mature detached sun needles of cypress under high irradiance (HI) of about 1 900 mol m–2 s–1. The degree of photoinhibition was determined by means of the ratio of variable to maximum chlorophyll (Chl) fluorescence (Fv/Fm) and electron transport measurements. Compared with the mature needles, the young needles, containing about half the amount of Chl a+b per unit area, exhibited a higher proportion of total carotenoids (Car) as xanthophyll cycle pigments and had an increased ratio of Car/Chl a+b. The potential efficiency of photosystem (PS) 2, Fv/Fm, markedly declined in HI-treated young needles without significant increase of F0 level. In contrast, the Fv/Fm ratio declined with significant increase of F0 level in mature needles. In isolated thylakoids, the rate of whole chain and PS2 activity markedly decreased in young HI-needles in comparison with mature needles. A smaller inhibition of PS1 activity was observed in both needles. In the subsequent dark incubation, fast recovery was found in both needle Types that reached maximum PS2 efficiencies similar to those observed in non-photoinhibited needles. The artificial exogenous electron donors DPC, NH2OH, and Mn2+ failed to restore the HI-induced loss of PS2 activity in mature needles, while DPC and NH2OH significantly restored it in young needles. Hence, HI-inactivation was on the donor side of PS2 in young needles and on the acceptor side of PS2 in mature needles. Photosynthetic rates of green leaves (GL) and green flower petals (GFP) of the CAM plant Dendrobium cv. Burana Jade and their sensitivities to different growth irradiances were studied in shade-grown plants over a period of 4 weeks. Maximal photosynthetic O2 evolution rates and CAM acidities [dawn/dusk fluctuations in titratable acidity] were higher in leaves exposed to intermediate sunlight [a maximal photosynthetic photon flux density (PPFD) of 500–600 μmol m−2 s−1] than in leaves grown under full sunlight (a maximal PPFD of 1 000–1 200 μmol m−2 s−1) and shade (a maximal PPFD of 200–250 μmol m−2 s−1). However, these two parameters of GFP were highest in plants grown under the shade and lowest in full sun-grown plants. Both GL and GFP of plants exposed to full sunlight had lower predawn Fv/Fm [dark adapted ratio of variable to maximal fluorescence (the maximal photosystem 2 yield without actinic irradiation)] than those of shade-grown plants. When exposed to intermediate sunlight, however, there were no significant changes in predawn Fv/Fm in GL whereas a significant decrease in predawn Fv/Fm was found in GFP of the same plant. GFP exposed to full sunlight exhibited a greater decrease in predawn Fv/Fm compared to those exposed to intermediate sunlight. The patterns of changes in total chlorophyll (Chl) content of GL and GFP were similar to those of Fv/Fm. Although midday Fv/Fm fluctuated with prevailing irradiance, changes of midday Fv/Fm after exposure to different growth irradiances were similar to those of predawn Fv/Fm in both GL and GFP. The decreases in predawn and midday Fv/Fm were much more pronounced in GFP than in GL under full sunlight, indicating greater sensitivity in GFP to high irradiance (HI). In the laboratory, electron transport rate and photochemical and non-photochemical quenching of Chl fluorescence were also determined under different irradiances. All results indicated that GFP are more susceptible to HI than GL. Although the GFP of Dendrobium cv. Burana Jade require a lower amount of radiant energy for photosynthesis and this plant is usually grown in the shade, is not necessarily a shade plant.  相似文献

Cross stress of heat and high irradiance (HI) resulted in the accumulation of active oxygen species and photo-oxidative damage to photosynthetic apparatus of wheat leaves during grain development. Pre-treatment with calcium ion protected the photosynthetic system from oxidative damage by reducing O-. 2 production, inhibiting lipid peroxidation, and retarding electrolyte leakage from cell. Therefore, high Fv/Fm [maximal photochemical efficiency of photosystem 2 (PS2) while all PS2 reaction centres are open], Fm/F0 (another expression for the maximal photochemical efficiency of PS2), ΦPS2 (actual quantum yield of PS2 under actinic irradiation), qP (photochemical quenching coefficient), and P N (net photosynthetic rate) were maintained, and lower qNP (non-photochemical quenching coefficient) of the leaves was kept under heat and HI stress. EGTA (a chelant of calcium ion) and LaCl3 (a blocker of Ca2+ channel in cytoplasmic membrane) had the opposite effect. Thus Ca ion may help protect the photosynthetic system of wheat leaves from oxidative damage induced by the cross stress of heat and HI.  相似文献

The responses to irradiance of photosynthetic CO2 assimilation and photosystem 2 (PS2) electron transport were simultaneously studied by gas exchange and chlorophyll (Chl) fluorescence measurement in two-year-old apple tree leaves (Malus pumila Mill. cv. Tengmu No.1/Malus hupehensis Rehd). Net photosynthetic rate (P N) was saturated at photosynthetic photon flux density (PPFD) 600-1 100 (mol m-2 s-1, while the PS2 non-cyclic electron transport (P-rate) showed a maximum at PPFD 800 mol m-2 s-1. With PPFD increasing, either leaf potential photosynthetic CO2 assimilation activity (Fd/Fs) and PS2 maximal photochemical activity (Fv/Fm) decreased or the ratio of the inactive PS2 reaction centres (RC) [(Fi – Fo)/(Fm – Fo)] and the slow relaxing non-photochemical Chl fluorescence quenching (qs) increased from PPFD 1 200 mol m-2 s-1, but cyclic electron transport around photosystem 1 (RFp), irradiance induced PS2 RC closure [(Fs – Fo)/Fm – Fo)], and the fast and medium relaxing non-photochemical Chl fluorescence quenching (qf and qm) increased remarkably from PPFD 900 (mol m-2 s-1. Hence leaf photosynthesis of young apple leaves saturated at PPFD 800 mol m-2 s-1 and photoinhibition occurred above PPFD 900 mol m-2 s-1. During the photoinhibition at different irradiances, young apple tree leaves could dissipate excess photons mainly by energy quenching and state transition mechanisms at PPFD 900-1 100 mol m-2 s-1, but photosynthetic apparatus damage was unavoidable from PPFD 1 200 mol m-2 s-1. We propose that Chl fluorescence parameter P-rate is superior to the gas exchange parameter P N and the Chl fluorescence parameter Fv/Fm as a definition of saturation irradiance and photoinhibition of plant leaves.  相似文献

Photoinhibition and pigment composition of green stem tissues of field-grown adult Eucalyptus nitens were measured on clear spring days with low morning temperatures—conditions that cause photoinhibition in leaves of many plant species. The sun-exposed (north-facing) bark contained less chlorophyll a+b (531 vs 748 mol m–2), neoxanthin (29 vs 41), and -carotene (54 vs 73), more xanthophyll cycle pigments per unit surface area and per unit chlorophyll (71 vs 53 mol m–2 and 141 vs 66 mmol mol–1 chlorophyll), and less lutein per unit chlorophyll (239 vs 190) than the shaded (southern) side. Maximum electron flow rates were 60 mol m–2 s–1 on the sun-exposed side, and about 10 mol m–2 s–1 on the shaded side. Fv/Fm was always lower than 0.8 on the sun-exposed side and the de-epoxidation state (DEPS) of the xanthophyll cycle was dominated by zeaxanthin in midday samples. Fv/Fm increased quickly after darkening, but DEPS recovered more slowly to 40% overnight. This suggested that rapidly reversible pH-dependent quenching was responsible for the bulk of changes in PS II efficiency. Fv/Fm remained below 0.8 overnight, which may well be indicative of photo-damage to PSII. In contrast, DEPS of the shaded side was lower, and Fv/Fm was higher, than for the sun-exposed side. We conclude that E. nitens chlorenchyma on the sun-exposed stem side has a photosynthetic pigment composition similar to sun leaves and it experiences significant photoinhibition in the field.  相似文献

Photosystem 2 (PS2)-driven electron transfer was studied in primary leaves of barley (Hordeum vulgare L.) seedlings grown under various photon fluxes (0.3–170.0 mol m–2 s–1) of blue (BR) or red (RR) radiation using modulated chlorophyll fluorescence. The Fv/Fm ratio was 0.78–0.79 in leaves of all radiation variants, except in seedlings grown under BR or RR of 0.3 mol m–2 s–1. The extent of the photochemical phase of the polyphasic Fv rise induced by very strong white light was similar in leaves of all radiation treatments. Neither radiation quality nor photon flux under plant cultivation influenced the amount of non QB-transferring centres of PS2 except in leaves of seedlings grown under BR of 0.3 mol m–2 s–1, in which the amount of such centres increased threefold. Both BR and RR stimulated the development of photochemically competent PS2 at photon fluxes as low as 3 mol m–2 s–1. Three exponential components with highly different half times were distinguished in the kinetics of Fv dark decay. This indicates different pathways of electron transfer from QA , the reduced primary acceptor of PS2, to other acceptors. Relative magnitudes of the individual decay components did not depend on the radiation quality or the photon flux during plant cultivation. Significant differences were found, however, between plants grown under BR or RR in the rate of the middle and fast components of Fv dark decay, which showed 1.5-times faster intersystem linear electron transport in BR-grown leaves.  相似文献

The effect of high irradiance (HI, photosynthetically active photon flux density of 1 300 μmol m−2 s−1) on net photosynthetic rate (P N), chlorophyll fluorescence parameters, and xanthophyll cycle components were studied in fruit tree bayberry leaves. HI induced the photoinhibition and inactivation of photosystem 2 (PS2) reaction centres (RCs), which was characterized by decreased P N, maximum yield of fluorescence after dark adaptation (Fm), photochemical efficiency of PS2 (Fv/Fm) and quantum yield of PS2 (ΦPS2), and increased reduction state of QA (1-qP) and non-photochemical quenching (NPQ). Initial fluorescence (F0) showed a decrease after the first 2 h, and subsequently increased from the third hour exposure to HI. Furthermore, a greater increase in the ratio (Fi-F0)/(Fp-F0) which is an expression of the proportion of the QB non-reducing PS2 centres, whereas a remarked decrease in the slope of Fi to Fp which represents the rate of QA reduction was observed in leaves after HI exposure. Additionally, HI caused an increase in the pool size of the xanthophyll cycle pigments and sustained elevated contents of zeaxanthin (Z), antheraxanthin (A), and de-epoxidation state (DES) at the end of the irradiation period. During HI, decreased Fm, Fv/Fm, ΦPS2, NPQ, slope of Fi to Fp, V+A+Z, and DES, and increased F0, 1-qP, ratio (Fi-F0)/(Fp-F0), and V were observed in dithiothreitol (DTT)-fed leaves compared to control ones under the same conditions. Hence photoinhibition caused by HI in bayberry was probably attributed to inactivation of PS2 RCs, and photoprotection from photodamage were mainly related to the xanthophyll cycle-dependent heat dissipation in excess photons.  相似文献

Chlorophyll fluorescence kinetics was used to investigate the effect of 1,4-dithiothreitol (DTT) on the distribution of excitation energy between photosystem 1 (PS1) and photosystem 2 (PS2) in soybean leaves under high irradiance (HI). The maximum PS2 quantum yield (Fv/Fm) was hardly affected by the presence of DTT, however, photon-saturated photosynthesis was depressed distinctly. Photochemical efficiency of open PS2 reaction centres during irradiation (Fv/Fm) was enhanced by about 30–40 % by DTT treatment, whereas photochemical quenching (qP) was depressed by about 40 % under HI. DTT treatment caused a 30 % decrease in allocation of excitation energy to PS1 under HI and a 20 % increase to PS2. An obvious shift in the balance of excitation energy distribution between photosystems was observed in DTT-treated leaves. Though high excitation pressure (1 - qP) resulted from DTT treatment, non-photochemical quenching (qN) was lower. DTT completely inhibited the formation of zeaxanthin and also distinctly depressed the state transition (qT). The shift in the balance of excitation distribution between the two photosystems induced by DTT was mainly due to the enhancement of excitation energy capture by PS2 antenna and the inhibition of state transition. It might be the shift in the balance between the two photosystems that mainly induced the depression of photosynthesis. Thus, to keep high utilization efficiency of absorbed photon energy, it is necessary to maintain the balance of excitation distribution between PS2 and PS1.  相似文献

In canopy shade leaves of grapevine (Vitis vinifera L. cv. Moscato giallo) grown in the field the contents of chlorophyll (Chl), carotenoids (Car), and soluble protein per fresh mass were lower than in sun leaves. RuBPC activity, in vivo nitrate reductase activity (indicator of nitrate utilisation), apparent electron transport rate, and photochemical fluorescence quenching were also significantly reduced in canopy shade leaves. When various photosynthetic activities were followed in isolated thylakoids, canopy shade leaves exerted a marked inhibition of whole chain and photosystem (PS) 2 activity. Smaller inhibition of PS1 activity was observed even in high-level canopy shade (HS) leaves. The artificial exogenous electron donors, DPC and NH2OH, significantly restored the loss of PS2 activity in HS leaves. Similar results were obtained when Fv/Fm was evaluated by Chl fluorescence measurements. The marked loss of PS2 activity in canopy shade leaves was due to the loss of 47, 43, 33, 28–25, 23, 17, and 10 kDa polypeptides.  相似文献

Midday photoinhibition of two newly developed super-rice hybrids   总被引:1,自引:0,他引:1  
Super-rice hybrids are two-line hybrid rice cultivars with 15 to 20 % higher yields than the raditional three-line hybrid rice cultivars. Response of photosynthetic functions to midday photoinhibition was compared between seedlings of the traditional hybrid rice (Oryza sativa L.) Shanyou63 and two super-rice hybrids, Hua-an3 and Liangyoupeijiu. Under strong midday sunlight, in comparison with Shanyou63, the two super-rice hybrids were less photoinhibited, as indicated by the lower loss of the net photosynthetic rate (P N), the quantum yield of photosystem 2 (ΦPS2), and the maximum and effective quantum yield of PS2 photochemistry (Fv/Fm and Fv′/Fm′). They also had a much higher transpiration rate. Hence the super-rice hybrids could protect themselves against midday photoinhibition at the cost of water. The photoprotective de-epoxidized xanthophyll cycle components, antheraxanthin (A) and zeaxanthin (Z), were accumulated more in Hua-an3 and Liangyoupeijiu than in Shanyou63, but the size of xanthophyll cycle pool of the seedlings was not affected by midday photoinhibition. Compared to Shanyou63, the super-rice hybrids were better photoprotected under natural high irradiance stress and the accumulation of Z and A, not the size of the xanthophyll pool protected the rice hybrids against photoinhibition.  相似文献

The role of the xanthophyll cycle in regulating the energy flow to the PS II reaction centers and therefore in photoprotection was studied by measurements of light-induced absorbance changes, Chl fluorescence, and photosynthetic O2 evolution in sun and shade leaves of Hedera canariensis. The light-induced absorbance change at 510 nm (A510) was used for continuous monitoring of zeaxanthin formation by de-epoxidation of violaxanthin. Non-radiative energy dissipation (NRD) was estimated from non-photochemical fluorescence quenching (NPQ).High capacity for zeaxanthin formation in sun leaves was accompanied by large NRD in the pigment bed at high PFDs as indicated by a very strong NPQ both when all PS II centers are closed (F'm) and when all centers are open (F'o). Such Fo quenching, although present, was less pronounced in shade leaves which have a much smaller xanthophyll cycle pool.Dithiothreitol (DTT) provided through the cut petiole completely blocked zeaxanthin formation. DTT had no detectable effect on photosynthetic O2 evolution or the photochemical yield of PS II in the short term but fully inhibited the quenching of Fo and 75% of the quenching of Fm, indicating that NRD in the antenna was largely blocked. This inhibition of quenching was accompanied by an increased closure of the PS II reaction centers.In the presence of DTT a photoinhibitory treatment at a PFD of 200 mol m-2 s-1, followed by a 45 min recovery period at a low PFD, caused a 35% decrease in the photon yield of O2 evolution, compared to a decrease of less than 5% in the absence of DTT. The Fv/Fm ratio, measured in darkness showed a much greater decrease in the presence than in the absence of DTT. The short-term acclimation (10-d) of Norway spruce [Picea abies (L.) Karst] to elevated CO2 concentration (EC) in combination with low irradiance (100 mol m–2 s–1) resulted in stimulation of CO2 assimilation (by 61 %), increased total chlorophyll (Chl) content (by 17 %), significantly higher photosystem 2 (PS2) photochemical efficiency (Fv/Fm; by 4 %), and reduced demand on non-radiative dissipation of absorbed excitation energy corresponding with enhanced capacity of photon utilisation within PS2. On the other hand, at high cultivation irradiance (1 200 mol m–2 s–1) both Norway spruce and spring barley (Hordeum vulgare L. cv. Akcent) responded to EC by reduced photosynthetic capacity and prolonged inhibition of Fv/Fm accompanied with enhanced non-radiative dissipation of absorbed photon energy. Norway spruce needles revealed the expressive retention of zeaxanthin and antheraxanthin (Z+A) in darkness and higher violaxanthin (V) convertibility (yielding even 95 %) under all cultivation regimes in comparison with barley plants. In addition, the non-photochemical quenching of minimum Chl a fluorescence (SV0), expressing the extent of non-radiative dissipation of absorbed photon energy within light-harvesting complexes (LHCs), linearly correlated with V conversion to Z+A very well in spruce, but not in barley plants. Finally, a key role of the Z+A-mediated non-radiative dissipation within LHCs in acclimation of spruce photosynthetic apparatus to high irradiance alone and in combination with EC was documented by extremely high SV0 values, fast induction of non-radiative dissipation of absorbed photon energy, and its stability in darkness.  相似文献

Photoinactivation of Photosystem (PS) II in vivo was investigated by cumulative exposure of pea, rice and spinach leaves to light pulses of variable duration from 2 to 100 s, separated by dark intervals of 30 min. During each light pulse, photosynthetic induction occurred to an extent depending on the time of illumination, but steady-state photosynthesis had not been achieved. During photosynthetic induction, it is clearly demonstrated that reciprocity of irradiance and duration of illumination did not hold: hence the same cumulative photon exposure (mol m–2) does not necessarily give the same extent of photoinactivation of PS II. This contrasts with the situation of steady-state photosynthesis where the photoinactivation of PS II exhibited reciprocity of irradiance and duration of illumination (Park et al. (1995) Planta 196: 401–411). We suggest that, for reciprocity to hold between irradiance and duration of illumination, there must be a balance between photochemical (qP) and non-photochemical (NPQ) quenching at all irradiances. The index of susceptibility to light stress, which represents an intrinsic ability of PS II to balance photochemical and non-photochemical quenching, is defined by the quotient (1-qP)/NPQ. Although constant in steady-state photosynthesis under a wide range of irradiance (Park et al. (1995). Plant Cell Physiol 36: 1163–1169), this index of susceptibility for spinach leaves declined extremely rapidly during photosynthetic induction at a given irradiance, and, at a given cumulative photon exposure, was dependent on irradiance. During photosynthetic induction, only limited photoprotective strategies are developed: while the transthylakoid pH gradient conferred some degree of photoprotection, neither D1 protein turnover nor the xanthophyll cycle was operative. The xanthophyll cycle and the water-water cycle had different functional significance in chilling-sensitive sweet pepper upon exposure to chilling temperature (4 °C) under low irradiance (100 µmol m−2 s−1) for 6 h. During chilling stress, effects of non-photochemical quenching (NPQ) on photosystem 2 (PS2) in dithiothreitol (DTT) fed leaves remained distinguishable from that of the water-water cycle in diethyldithiocarbamate (DDTC) fed leaves. In DTT-fed leaves, NPQ decreased greatly accompanied by visible inhibition of the de-epoxidized ratio of the xanthophyll cycle, and maximum photochemical efficiency of PS2 (Fv/Fm) decreased markedly. Thus the xanthophyll cycle-dependent NPQ could protect PS2 through energy dissipation under chilling stress. However, NPQ had a slighter effect on photosystem 1 (PS1) in DTT-fed leaves than in DDTC-fed leaves, whereas effects of the water-water cycle on PS1 remained distinguishable from that of NPQ. Inhibiting superoxide dismutase (SOD) activity increased the accumulation of , the oxidation level of P700 (P700+) decreased markedly relative to the control and DTT-fed leaves. Both Fv/Fm and NPQ changed little in DDTC-fed leaves accompanied by little change of (A+Z)/(V+A+Z). This is the active oxygen species inducing PS1 photoinhibition in sweet pepper. The water-water cycle can be interrupted easily at chilling temperature.
