Pyruvate fermentation inRhodospirillum rubrum (strains F1, S1, and Ha) was investigated using cells precultured on different substrates anaerobically in the light and than transferred to anaerobic dark conditions. Pyruvate formate lyase was always the key enzyme in pyruvate fermentation but its activity was lower than in cells which have been precultured aerobically in darkness. The preculture substrate also had a clear influence on the pyruvate formate lyase activity. Strains F1 and S1 metabolized the produced formate further to H2 and CO2. A slight production of CO2 from pyruvate, without additional H2-production, could also be detected. It was concluded from this that under anaerobic dark conditions a pyruvate dehydrogenase was also functioning. On inhibition of pyruvate formate lyase the main part of pyruvate breakdown was taken over by pyruvate dehydrogenase.When enzyme synthesis was inhibited by chloramphenicol, propionate production in contrast to formate production was not affected. Protein synthesis was not significant during anaerobic dark culture. The fermatation metabolism ofRhodospirillum rubrum Ha was studied after adaptation of both light-anaerobic and dark aerobic to dark anaerobic conditions.Pyruvate was metabolized to acetate, formate, CO2 and propionate by suspensions of cells adapted to anaerobiosis. Pyruvate cleavage to formate accounted for about two-thirds of the pyruvate decomposed. This process was catalyzed by a coenzyme A dependent pyruvate formate lyase. In carboxylate- and nucleotide-free extracts, the substrate concentrations for half-maximal velocity [S]0.5V were found to be 1.5 mM for pyruvate and 75 M for coenzyme A.Pyruvate formate lyase could practically not be demonstrated in light-anaerobic photosynthesizing cells. Lyase activity was low at a basic level in darkaerobic respiring cells. After adaptation of both types of cells under growth conditions to dark anaerobiosis lyase activity increased about 10-fold. Highest levels could be observed in cells grown aerobically in the dark on pyruvate after transition to dark anaerobic conditions. It is concluded that pyruvate formate lyase is the characteristic key enzyme of the dark-anaerobic fermentative metabolism ofR. rubrum Ha.  相似文献

Rhodospirillum rubrum is able to produce H2 during fermentation anaerobically in the dark in two ways, namely through formate hydrogen lyase and through the nitrogenase. After chemotrophic preculture aerobically in the dark formate hydrogen lyase was synthesized after a lag phase, whilst after phototrophic preculture a slight activity was present from the beginning of the anaerobic dark culture. During fermentation metabolism its activity increased noticeably. Hydrogen production through the nitrogenase occurred if the nitrogenase had been activated during phototrophic preculture. It ceased during fermentation metabolism after about 3 1/2 h anaerobic dark culture. The CO insensitive H2 production by the nitrogenase could be partially inhibited by N2. Potential activity of this system, however, remained and could be increased under conditions of nitrogenase induction. Chlorofluexus aurantiacus OK-70 fl was grown photoautotrophically with hydrogen as the electron source. The lowest doubling time observed was 26 h.The mechanism of CO2 fixation in autotrophically grown cells was studied. The presence of ribulose-1,5-bis-phosphate carboxylase and phosphoribulokinase could not be demonstrated. Carbon isotope fractionation (13C) was small, and alanine and aspartate but not 3-phosphoglycerate were the major labelled compounds in short term 14CO2 labelling. Thus CO2 is not fixed by the Calvin cycle.Fluoroacetate (FAc) completely inhibited protein synthesis in cultures and caused a slight citrate accumulation. However, CO2 fixation continued and increased polyglucose formation occurred. Under these conditions added acetate was metabolized to polyglucose, as were glycine, serine, glyoxylate and succinate, but to a lesser extent; little or no formate or CO was utilised.Glyoxylate inhibited CO2 fixation in vivo, indicating that pyruvate is formed from acetyl-CoA and CO2 by pyruvate synthase. Two key enzymes of the reductive TCA cycle, citrate lyase and -ketoglutarate synthase were not detected in cell free extracts, but pyruvate synthase and phosphoenolpyruvate carboxylase were demonstrated. End-product synthesis and enzyme activities involved in pyruvate catabolism, H2 synthesis, and ethanol production in mid-log (OD600  0.25), early stationary (OD600  0.5), and stationary phase (OD600  0.7) cell extracts were determined in Clostridium thermocellum ATCC 27405 grown in batch cultures on cellobiose. Carbon dioxide, hydrogen, ethanol, acetate and formate were major end-products and their production paralleled growth and cellobiose consumption. Lactate dehydrogenase, pyruvate:formate lyase, pyruvate:ferredoxin oxidoreductase, methyl viologen-dependant hydrogenase, ferredoxin-dependant hydrogenase, NADH-dependant hydrogenase, NADPH-dependant hydrogenase, NADH-dependant acetaldehyde dehydrogenase, NADH-dependant alcohol dehydogenase, and NADPH-dependant alcohol dehydrogenase activities were detected in all extracts, while pyruate dehydrogenase and formate dehydrogenase activities were not detected. All hydrogenase activities decreased (2–12-fold) as growth progressed from early exponential to stationary phase. Alcohol dehydrogenase activities fluctuated only marginally (<45%), while lactate dehydrogenase, pyruvate:formate lyase, and pyruvate:ferredoxin oxidoreductase remained constant in all cell extracts. We have proposed a pathway involved in pyruvate catabolism and end-product formation based on enzyme activity profiles in conjunction with bioinformatics analysis.  相似文献

Brochothrix thermosphacta, a psychrophilic, facultative anaerobe, exhibited homolactic fermentation under anaerobic conditions in the presence of excess glucose. In glucose-limited chemostat culture (on synthetic medium), ethanol, acetate, formate and lactate were formed. Formation of ethanol and acetate was accounted for by the formate concentrations in culture filtrates. Acetate, formate and ethanol formation was enhanced at low growth rates in chemostat culture. O2-limited chemostat studies indicated that formate formation was inhibited by oxygen (<0.2 M) and studies with a variant, strain 301, which lacked pyruvate dehydrogenase activity, showed that cell culture in basal medium did not occur at O2 tensions greater than that preventing formate production in the wild-type strain. The data are consistent with stimulation of pyruvate formate lyase activity by glucose limitation, possibly because of decreased concentrations of glycolytic intermediates.S.P. Singh was and A. Formate was formed in extracts of Chlorogonium elongatum via direct cleavage of pyruvate by a pyruvate formate-lyase (PFL, EC 2.3.1.54). The conversion of PFL to the catalytically active form required S-adenosylmethionine, ferric (2+), photoreduced deazariboflavin as reductant, pyruvate as allosteric effector and strict anaerobic conditions. At the optimum pH (pH 8.0), PFL catalyzed formate formation, pyruvate synthesis and the isotope exchange from [14C]formate into pyruvate with rates of 30.0, 1.5 and 1.2 nmol min-1 mg-1 protein, respectively. Treatment of the active enzyme with O2 irreversibly inactivated PFL activity (half-time 2 min). In addition to PFL, the activities of phosphotransacetylase (EC 2.3.1.8), acetate kinase (EC 2.7.2.1), aldehyde dehydrogenase (CoA acetylating, EC 1.2.1.10) and alcohol dehydrogenase (EC 1.1.1.1) were also detected in extracts of C. elongatum. The active species of CO2 , i.e. CO2 or HCO 3 –(H2CO3) utilized by enzymes catalyzing ferredoxin-linked carboxylation reactions was determined. The enzyme investigated was pyruvate synthase from Clostridium pasteurianum (EC 1.2.7.1; Pyruvate: ferredoxin oxidoreductase). Data were obtained which were compatible with those expected if CO2 is the active species.The dissociation constant (K S) of the enzyme-CO2 complex was measured. Enrichments on L-tartrate from a freshwater lake sediment yielded a pure culture of anaerobic bacterium designated strain 16Lt1. The rod-shaped organism was motile, did not form spores, and had a gram-negative wall structure. No cytochromes were detected. The mol % G+C of the DNA was 58. The new strain was microaerotolerant, and grew optimally at 30°C and neutral pH in freshwater medium. A wide range of carbohydrates was fermented, with formate, acetate, ethanol, lactate and succinate being the end-products detected. L-tartrate and citrate were fermented to formate, acetate and CO2. L-tartrate was fermented by the dehydratase pathway, and glucose by the Embden-Meyerhof-Parnas pathway. Fumarate was reduced, but nitrate, sulfate, sulfur and thiosulfate were not used as terminal electron acceptors. Glucose metabolism was constitutive, whereas L-tartrate-degrading activity was inducible. When glucose and L-tartrate were both present as substrates, growth was diauxic with glucose being metabolized first. The growth rate and growth yield were higher on glucose than on L-tartrate. Strain 16Lt1 has been deposited with the Deutsche Sammlung von Mikroorganismen as Bacteroides sp. DSM6268.  相似文献

Rhodospirillum rubrum (F1) maintained electron balance mainly by producing propionate, formate and H2 during fermentation metabolism. H2 formation was inversely correlated with the production of propionate.In diluted, growing cultures high amounts of H2 and only traces or no propionate were produced from pyruvate. In dense cultures or in resting cultures without (NH4)2SO4, however, propionate was formed from pyruvate in relatively high amounts Cultures always produced much more propionate than H2 from fructose in contrast to cells with pyruvate. Kinetic studies of growth and excretion of fermentation products indicated that the enzyme system for H2 formation is adaptive. Chloramphenicol (3 μg/ml) completely inhibited the formation of H2 if the cells were not adapted to fermentation metabolism. The production of propionate, on the other hand, was not prevented by chloramphenicol after shifting the cells from aerobic dark culture with malate to fermentation conditions with pyruvate.H2 formation was not influenced by sodium ascorbate but it was significantly decreased by K3[Fe(CN)6].Poly(β-hydroxybutyric acid) was also synthesized by the cells during anaerobic dark metabolism especially in dense cultures, probably favoured by the rapid acidification of the medium. Formate can also accumulate in the fermentation metabolism, especially in young growing cultures.These results give an explanation for the differing reports in the literature on the fermentation metabolism of R. rubrum.  相似文献

Zusammenfassung R. rubrum bildet anaerob im Dunkeln aus exogenen und endogenen Substraten hauptsächlich Acetat und Propionat.In Kulturen mit Pyruvat als Substrat wurde in der Regel mehr Acetat als Propionat gebildet (4:1–1:1), mit Fructose dagegen weniger Acetat (1:2–1:3). Ruhende Zellen produzierten aus Pyruvat, im Vergleich zu Kulturen mit (NH4)2SO4 im Medium, relativ mehr Propionat als Acetat.Beim Abbau von gespeicherter PHBS wurde relativ mehr Acetat und weniger Propionat als beim Abbau endogener Polysaccharide produziert.Eine Zugabe von Ascorbat (0,8 u. 1,6%) oder K3[Fe(CN)6] (0,08 u 0,32%) hatte nur einen geringen Effekt auf das Verhältnis von Acetat zu Propionat. Exogenes CO2 war, besonders bei Zugabe von Fructose als Substrat, für die Synthese von Propionat notwendig. Die Wege der Acetat- und Propionatbildung unter anaeroben Bedingungen im Dunkeln werden diskutiert.
The synthesis of volatile acids by fermentation of pyruvate and fructose in anaerobic dark cultures of Rhodospirillum rubrum
Summary Under anaerobic conditions in the dark R. rubrum produced mainly acetate and propionate from exogenous and endogenous substrates.With pyruvate as a substrate usually less propionate than acetate was synthesized, with fructose, however, more propionate than acetate was produced.In resting cells, compared with cultures having (NH4)2SO4 in the medium, more propionate than acetate was synthesized from pyruvate.By degradation of stored poly--hydroxybutyric acid under anaerobic dark conditions relatively more acetate is produced than by degradation of endogenous polysaccharide.Addition of ascorbate (0.8 and 1.6%) or K3[Fe(CN)6] (0.08 and 0.32%) had little influence on the relative concentrations of acetate and propionate.Exogenous CO2 was necessary for the synthesis of propionate, especially when fructose was the substrate. The pathways of acetate and propionate production anaerobically in the dark are discussed.

Nitrosomonas europaea is capable of maintaining an anaerobic metabolism, using pyruvate as an electron donor and nitrite as an electron acceptor; utilization of nitrite depends upon supply of both pyruvate and ammonia. The role of ammonia in this reaction was not determined. N europaea also assimilates CO2 anaerobically into cell material in the presence of nitrite (0.5–1.0 mM), pyruvate and ammonia. This reaction was partially inhibited by nitrite which apparently competed with CO2 for reducing power. The incorporation of 14CO2 by the cell suspensions of an extremely thermophilic, aerobic hydrogen-oxidizing bacterium, Hydrogenobacter thermophilus was studied. After short time incubation of the cell suspensions with 14CO2, the radiactivity was initially present in aspartate, glutamate, succinate, phosphorylated compounds, citrate, malate and fumarate. All of these compounds except phosphorylated compounds were related to the members of the tricarboxylic acid cycle. The proportion of labelled aspartate onglutamate in total radioactivity on each chromatogram decreased with incubation time, while the percentage of the radioactivity incorporated in phosphorylated compounds increased with time up to 10 s. These indicated that aspartate and glutamate is derived from primary products of CO2 fixation.In cell-free extracts of Hydrogenobacter thermophilus, the two key enzymes in the Calvin cycle, ribulose-1,5-bisphosphate carboxylase and phosphoribulokinase could not be detected. The key enzymes of the reductive tricarboxylic acid cycle, fumarate reductase and ATP citrate lyase were present. Activities of phosphoenolpyruvate synthetase and pyruvate carboxylase were also detected. The referse reactions (dehydrogenase reactions) of -ketoglutarate synthase and pyruvate synthase could be detected by using methyl viologen as an electron acceptor.These findings strongly suggested that a new type of the reductive tricarboxylic acid cycle operated as the CO2 fixation pathway in Hydrogenobacter thermophilus.  相似文献

An obligately anaerobic, rod-shaped bacterium was isolated on alanine in co-culture with H2-scavenging Desulfovibrio and obtained in pure culture with glycine as sole fermentation substrate. The isolated strain, al-2, was motile by a polar to subpolar flagellum and stained Gram-positive. The guanine plus cytosine content of the DNA was 44.0 mol%. Strain al-2 grew in defined, reduced glycine media supplemented with biotin. The pure culture fermented 4 mol glycine to 3 mol acetate, 4 mol ammonia and 2 mol CO2. Under optimum conditions (34°C, pH 7.3), the doubling time on glycine was 60 min and the molar growth yield 7.6 g cell dry mass. Serine was fermented to acetate, ethanol, CO2, H2 and ammonia. In addition, betaine, sarcosine or creatine served as substrates for growth and acetate production if H2, formate or e.g. valine were added as H-donors. In pure culture on alanine under N2, strain al-2 grew very poorly and produced H2 up to a partial pressure of 3.6 kPa (0.035 atm). Desulfovibrio species, Methanospirillum hungatei and Acetobacterium woodii served as H2-scavengers that allowed good syntrophic growth on alanine. The co-cultures also grew on aspartate, leucine, valine or malate. Alanine and aspartate were stoichiometrically degraded to acetate and ammonia, whereas the reducing equivalents were recovered as H2S, CH4 or newly synthetized acetate, respectively. Growth of strain al-2 in co-culture with the hydrogenase-negative, formate-utilizing Desulfovibrio baarsii indicated that a syntrophy was also possible by interspecies formate transfer. Growth on glycine, or on betaine, sarcosine or creatine (plus H-donors) depended strictly on the addition of selenite (0.1 M); selenite was not required for fermentation of serine, or for degradation of alanine, aspartate or valine by the co-cultures. Cell-free extracts of glycine-grown cells contained active glycine reductase, glycine decarboxylase and reversible methyl viologen-dependent formate dehydrogenase in addition to the other enzymes necessary for an oxidation to CO2. In all reactions NADP was the preferred H-carrier. Both formate and glycine could be synthesized from bicarbonate. Serine-grown cells did not contain serine hydroxymethyl transferase but serine dehydratase and other enzymes commonly involved in pyruvate metabolism to acetate, CO2 and H2. The enzymes involved in glycine metabolism were repressed during growth on serine. By its morphology and physiology, strain al-2 did not resemble described amino acid-degrading species. Therefore, the new isolate is proposed as type strain of a new species, Eubacterium acidaminophilum.  相似文献

The knowledge of the mechanism of flux distribution will benefit understanding cell physiology and regulation of metabolism. In this study, the measured fluxes obtained under steady-state conditions were used to estimate intracellular fluxes and identify the robustness of branch points of the anaerobic glycerol metabolism in Klebsiella pneumoniae for the production of 1,3-propanediol by metabolic flux analysis. The biomass concentration increased as NADH2/NAD+ decreased at low initial concentration and inversed at high initial glycerol concentration. The flux distribution revealed that the branch points of glycerol and dihydroxyacetonephosphate were rigid to the environmental conditions. However, the pyruvate and acetyl coenzyme A metabolisms gave cells the flexibility to regulate the energy and intermediate fluxes under various environmental conditions. Additionly, it was found that the formation rate of ethanol and the ratio of pyruvate dehydrogenase to pyruvate formate lyase appeared visible fluctuations at high glycerol uptake rate.  相似文献

A native homoethanol pathway (pyruvate-to-acetyl-CoA-to-acetaldehyde-to-ethanol) was engineered in Escherichia coli B. The competing fermentation pathways were eliminated by chromosomal deletions of the genes encoding for fumarate reductase (frdABCD), lactate dehydrogenase (ldhA), acetate kinase (ackA), and pyruvate formate lyase (pflB). For redox balance and anaerobic cell growth, the pyruvate dehydrogenase complex (aceEF-lpd, a typical aerobically-expressed operon) was highly expressed anaerobically using a native anaerobic inducible promoter. The resulting strain SZ420 (ΔfrdBC ΔldhA ΔackA ΔfocA-pflB ΔpdhR::pflBp6-pflBrbs-aceEF-lpd) contains no foreign genes and/or promoters and efficiently ferments glucose and xylose into ethanol with a yield of 90% under anaerobic conditions.  相似文献

Bacteroides succinogenes produces acetate and succinate as major products of carbohydrate fermentation. An investigation of the enzymes involved indicated that pyruvate is oxidized by a flavin-dependent pyruvate cleavage enzyme to acetyl-CoA and CO2. Active CO2 exchange is associated with the pyruvate oxidation system. Reduction of flavin nucleotides is CoASH-dependent and does not require ferredoxin. Acetyl-CoA is further metabolized via acetyl phosphate to acetate and ATP. Reduced flavin nucleotide is used to reduce fumarate to succinate by a particulate flavin-specific fumarate reductase reaction which may involve cytochrome b. Phosphoenolpyruvate (PEP) is carboxylated to oxalacetate by a GDP-specific PEP carboxykinase. Oxalacetate, in turn, is converted to malate by a pyridine nucleotide-dependent malate dehydrogenase. The organism has a NAD-dependent glyceraldehyde-3-phosphate dehydrogenase. The data suggest that reduced pyridine nucleotides generated during glycolysis are oxidized in malate formation and that the electrons generated during pyruvate oxidation are used to reduce fumarate to succinate.  相似文献

Two strains of S-citramalate-fermenting strictly anaerobic non-spore-formers were isolated in pure culture from anoxic mud samples of a creek and from a pond. One of them (strain CreCit 1) was studied in detail. It stained gram-negative, and contained β-hydroxymyristic acid. Nitrate, sulfate and other sulfur compounds were not utilized as electron acceptors. S-citramalate, citrate, mesaconate, and pyruvate were utilized as substrates; but R-citramalate, citraconate, l-glutamate, and carbohydrates not. S-citramalate was fermented to acetate, formate, and hydrogen. Citrate, mesaconate, and pyruvate were fermented to acetate and formate. The DNA base ratio was 59 mol% guanine plus cytosine. Strain CreCit 1 is described as a member of a new genus and a new species in the family Bacteroidaceae, Formivibrio citricus gen. nov., sp. nov.  相似文献
