Bio-microarray fabrication techniques--a review   总被引:1,自引:0,他引:1  
Microarrays with biomolecules (e.g., DNA and proteins), cells, and tissues immobilized on solid substrates are important tools for biological research, including genomics, proteomics, and cell analysis. In this paper, the current state of microarray fabrication is reviewed. According to spot formation techniques, methods are categorized as "contact printing" and "non-contact printing." Contact printing is a widely used technology, comprising methods such as contact pin printing and microstamping. These methods have many advantages, including reproducibility of printed spots and facile maintenance, as well as drawbacks, including low-throughput fabrication of arrays. Non-contact printing techniques are newer and more varied, comprising photochemistry-based methods, laser writing, electrospray deposition, and inkjet technologies. These technologies emerged from other applications and have the potential to increase microarray fabrication throughput; however, there are several challenges in applying them to microarray fabrication, including interference from satellite drops and biomolecule denaturization.  相似文献

ObjectivesAfter a century of spectacular advances, healthcare systems are facing unprecedented crisis, linked to shortage of health human resources and health technologies. In fact, availability of care depends on both technological and human resources of health. The objective of this study is to develop indicators that can measure qualitatively human resources and technologies of health in healthcare facilities, in order to assess availability of care in sub-Saharan African countries.Materials and MethodsRegarding “health technology” related to “medical devices”, an indicator called “TechSan” for “Technologies de Santé” was previously developed and published (Ndione FB et al. (2019) [6]). To address the deficiencies in usual indicators related to health human resources, a second indicator called “RhSan” for “Ressources humaines de santé” in French is proposed. This indicator assigns a weight to each health worker taking into account his specific “level of medical knowledge” and “experience”. In order to correlate “RhSan” with “TechSan”, a third indicator called “RhTech” is also developed to assess matches between “health technologies” and “health human resources” and establish realistic availability of care. These indicators have the advantage to be consolidated by specialty such as laboratory, imaging, surgery, and “mother and child care”.ResultsThe application of TechSan, RhSan and RhTech to data collected in Senegal in 2016, enabled to assess the distribution of “health technology” and “health human resources” in this country. They also permit the mapping of care availability per specialty in Senegal. The results show a strong oversupply of Dakar in terms of both human resources and technologies of health compared to other Senegalese regions. Oppositely, Sedhiou, Kaffrine, Matam and Kédougou are poorly endowed showing limits of the Senegalese health pyramid system.ConclusionTechSan, RhSan and RhTech can provide reliable decision-making tools in order to elaborate health policies in sub-Saharan African countries on more rigorous basis.  相似文献

Background and purposeThe use of cone beam computed tomography (CBCT) for performing dose calculations in radiation therapy has been widely investigated as it could provide a quantitative analysis of the dosimetric impact of changes in patients during the treatment. The aim of this review was to classify different techniques adopted to perform CBCT dose calculation and to report their dosimetric accuracy with respect to the metrics used.Methods and materialsA literature search was carried out in PubMed and ScienceDirect databases, based upon the following keywords: “cone beam computed tomography”, “CBCT”, “cone beam CT”, “dose calculation”, “accuracy”. Sixty-nine peer-reviewed relevant articles were included in this review: thirty-one patient studies, fifteen phantom studies and twenty-three patient & phantom studies. Most studies were found to have focused on head and neck, lung and prostate cancers.ResultsThe techniques adopted to perform CBCT dose calculation have been grouped in six categories labelled as (1) pCT calibration, (2) CBCT calibration, (3) HU override, (4) Deformable image registration, (5) Dose deformation, and (6) Combined techniques. Differences between CBCT dose and reference dose were reported both for target volumes and OARs.ConclusionsA comparison among the available techniques for CBCT dose calculations is challenging as many variables are involved. Therefore, a set of reporting standards is recommended to enable meaningful comparisons among different studies. The accuracy of the results was strongly dependent on the image quality, regardless of the methods used, highlighting the need for dose validation and quality assurance standards.  相似文献

ABSTRACT

Mysticete (baleen) whales produce a variety of vocalizations and sounds, but relatively few of these have been well described with accompanying behavior. This review concentrates on the vocalizations consistently associated with behavioral interactions or acoustic exchanges between or among conspecifics. These communication “signals” have been categorized for this review as contact calls of single animals outside of the breeding season (including cow-calf pairs), vocalizations reported during the breeding season (often designated as “songs”), and calls produced by active groups of whales that may or may not have a reproductive function. While much remains unknown, the data obtained thus far indicate that the social vocalizations of baleen whales have structural/functional similarities with those of other mammals and birds.  相似文献

Stem cells in vivo are housed within a functional microenvironment termed the “stem cell niche.” As the niche components can modulate stem cell behaviors like proliferation, migration and differentiation, evaluating these components would be important to determine the most optimal platform for their maintenance or differentiation. In this review, we have discussed methods and technologies that have aided in the development of high throughput screening assays for stem cell research, including enabling technologies such as the well-established multiwell/microwell plates and robotic spotting, and emerging technologies like microfluidics, micro-contact printing and lithography. We also discuss the studies that utilized high throughput screening platform to investigate stem cell response to extracellular matrix, topography, biomaterials and stiffness gradients in the stem cell niche. The combination of the aforementioned techniques could lay the foundation for new perspectives in further development of high throughput technology and stem cell research.  相似文献

Purpose

The social aspects of municipal solid waste management (MSWM) systems are underpinning their sustainability and effectiveness. The assessment of these systems from a life cycle perspective is widespread throughout environmental life cycle assessment (LCA), but few studies have used social life cycle assessment (S-LCA). The present study is an innovative review with the objective to analyse and describe the current level of development of S-LCA applications in MSWM, and to identify the main methodological challenges and best practices, aiming at recommending approaches to harmonise future S-LCA applications in MSWM.

Materials and methods

A systematic review of the literature found 36 relevant scientific articles. These were submitted to bibliometric and content analysis, which includes an analysis of how methodological aspects of the four phases of S-LCA were applied in comparison with best practice and existing guidelines.

Results and discussion

There was a predominance of case studies in developing countries (59%) and evaluation of the stages of collection/transportation, pre-processing (sorting) and landfilling (55%). There were more studies focusing on stakeholders, “workers” and “local communities” and in the impact subcategories “employment”, “working hours”, “health and safety/working conditions”, “community involvement/participation” and “health and safety/living conditions of community”. There was great variability in the application of the method (47% of the studies included methodological developments). However, the 39% based on UNEP guidelines were closer to a methodological consensus.

Conclusion

In general, studies need more detail and clarity in describing the methodological decisions used. Improvements are needed for issues that limit the S-LCA method, including the difficulties of covering the entire life cycle, relating impacts to the functional unit, standardizing impact assessment methods, addressing allocation and data quality issues and interpretation of results and their limitations. Improvements can be achieved by using participatory methods in the selection of categories, subcategories and impact indicators, as well as by clarifying the definition of a product system and detailing “cut-off criteria” of processes/organizations and the impact of these decisions on results.

AimA systemic review and analysis of evolution journey of indices, such as conformity index (CI), homogeneity index (HI) and gradient index (GI), described in the literature.BackgroundModern radiotherapy techniques like VMAT, SRS and SBRT produce highly conformal plans and provide better critical structure and normal tissue sparing. These treatment techniques can generate a number of competitive plans for the same patients with different dose distributions. Therefore, indices like CI, HI and GI serve as complementary tools in addition to visual slice by slice isodose verification while plan evaluation. Reliability and accuracy of these indices have been tested in the past and found shortcomings and benefits when compared to one another.Material and methodsPotentially relevant studies published after 1993 were identified through a pubmed and web of science search using words “conformity index”, “Homogeneity index”, “Gradient index”,” Stereotactic radiosurgery”,” stereotactic Body radiotherapy” “complexity metrics” and “plan evaluation index”. Combinations of words “plan evaluation index conformity index” were also searched as were bibliographies of downloaded papers.Results and conclusionsMathematical definitions of plan evaluation indices modified with time. CI definitions presented by various authors tested at their own and could not be generalized. Those mathematical definitions of CI which take into account OAR sparing grant more confidence in plan evaluation. Gradient index emerged as a significant plan evaluation index in addition to CI whereas homogeneity index losing its credibility. Biological index base plan evaluation is becoming popular and may replace or alter the role of dosimetrical indices.  相似文献

ObjectivesThe aim of this study was to determine the association of “ABO” and “Rhesus” blood groups with incidence of breast cancer.MethodsIn this study, we identified 70 research documents from data based search engines including “PubMed”, “ISI-Web of Knowledge”, “Embase” and “Google Scholar”. The research papers were selected by using the primary key-terms including “ABO blood type”, “Rhesus” blood type and “breast cancer”. The research documents in which “ABO” and “Rhesus” blood types and breast cancer was debated were included. After screening, we reviewed 32 papers and finally we selected 25 research papers which met the inclusion criteria and remaining documents were excluded.ResultsBlood group “A” has high incidence of breast cancer (45.88%), blood group “O” has (31.69%); “B” (16.16%) and blood group “AB” has (6.27%) incidence of breast cancer. Blood group “A” has highest and blood group “AB” has least association with breast cancer. Furthermore, “Rhesus +ve” blood group has high incidence of breast cancer (88.31%) and “Rhesus –ve” blood group has least association with breast cancer (11.68%).ConclusionBlood group “A” and “Rhesus +ve” have high risk of breast cancer, while blood type “AB” and “Rhesus –ve” are at low peril of breast cancer. Physicians should carefully monitor the females with blood group “A” and “Rh +ve” as these females are more prone to develop breast cancer. To reduce breast cancer incidence and its burden, preventive and screening programs for breast cancer especially in young women are highly recommended.  相似文献

BackgroundMost recalcitrant infections are associated to colonization and microbial biofilm development. These biofilms are difficult to eliminate by the immune response mechanisms and the current antimicrobial therapy.AimTo describe the antifungal of micafungin against fungal biofilms based in the scientific and medical literature of recent years.MethodsWe have done a bibliographic retrieval using the scientific terms “micafungin”, “activity”, “biofilm”, “Candida”, “Aspergillus”, “fungi”, “mycos”*, susceptibility, in PubMed/Medline from the National Library of Medicine from 2006 to 2009.ResultsMost current antifungal agents (amphotericin B and fluconazole) and the new azole antifungals have no activity against fungal biofilms. However, micafungin and the rest of echinocandins are very active against Candida albicans, Candida dubliniensis, Candida glabrata, and Candida krusei biofilms but their activities are variable and less strong against Candida tropicalis and Candida parapsilosis biofilms. Moreover, they have not activities against the biofilms of Cryptococcus y Trichosporon.ConclusionsThe activity of micafungin against Candida biofilms gives more strength to its therapeutic indication for candidaemia and invasive candidiasis associated to catheter, prosthesis and other biomedical devices.  相似文献

Abstract

The paper discusses a series of new technologies and new strategic factors entering into naval power and their analytical consequences. The argument is made that (1) traditional ways of identifying “naval power”; no longer have any utility or carry real meaning; (2) current high‐value surface naval forces of declining utility are extremely vulnerable to attack, which will force a radical revaluation in the West of “sea control”; and the costs of such control; and (3) the Soviet Union, by virtue of its geography and its capacity to innovate in “sea denial”; naval force, may be better positioned that the West to take advantage of new antinaval technologies. Western countermeasures may be expensive, involve a radical restructuring of naval force, and mean the acceptance of permanent strategic vulnerability at sea. The “new era”; of naval politics cannot be divorced, analytically and operationally, from emergent space systems and from the strategic nuclear balance.  相似文献

The conventional methods of using autografts and allografts for repairing defects in bone, the osteochondral bone, and the cartilage tissue have many disadvantages, like donor site morbidity and shortage of donors. Moreover, only 30% of the implanted grafts are shown to be successful in treating the defects. Hence, exploring alternative techniques such as tissue engineering to treat bone tissue associated defects is promising as it eliminates the above-mentioned limitations. To enhance the mechanical and biological properties of the tissue engineered product, it is essential to fabricate the scaffold used in tissue engineering by the combination of various biomaterials. Three-dimensional (3D) printing, with its ability to print composite materials and with complex geometry seems to have a huge potential in scaffold fabrication technique for engineering bone associated tissues. This review summarizes the recent applications and future perspectives of 3D printing technologies in the fabrication of composite scaffolds used in bone, osteochondral, and cartilage tissue engineering. Key developments in the field of 3D printing technologies involves the incorporation of various biomaterials and cells in printing composite scaffolds mimicking physiologically relevant complex geometry and gradient porosity. Much recently, the emerging trend of printing smart scaffolds which can respond to external stimulus such as temperature, pH and magnetic field, known as 4D printing is gaining immense popularity and can be considered as the future of 3D printing applications in the field of tissue engineering.  相似文献

The technology of “Lab-on-a-Chip” allows the synthesis and analysis of chemicals and biological substance within a portable or handheld device. The 3D printed structures enable precise control of various geometries. The combination of these two technologies in recent years makes a significant progress. The current approaches of 3D printing, such as stereolithography, polyjet, and fused deposition modeling, are introduced. Their manufacture specifications, such as surface roughness, resolution, replication fidelity, cost, and fabrication time, are compared with each other. Finally, novel application of 3D printed channel in biology are reviewed, including pathogenic bacteria detection using magnetic nanoparticle clusters in a helical microchannel, cell stimulation by 3D chemical gradients, perfused functional vascular channels, 3D tissue construct, organ-on-a-chip, and miniaturized fluidic “reactionware” devices for chemical syntheses. Overall, the 3D printed fluidic chip is becoming a powerful tool in the both medical and chemical industries.  相似文献

Introduction: Renal tract malformations (RTMs) are congenital anomalies of the kidneys and urinary tract, which are the major cause of end-stage renal disease in children. Using immunoassay-based approaches (ELISA, western blot), individual urinary proteins including transforming growth factor β, tumor necrosis factor and monocyte attractant proteins 1 were found to be associated to RTMs. However, only mass spectrometry (MS) based methods leading to the identification of panels of protein-based markers composed of fragments of the extracellular matrix allowed the prediction of progression of RTMs and its complications.

Areas covered: In this review, we summarized relevant studies identified in “Pubmed” using the keywords “urinary biomarkers” and “proteomics” and “renal tract malformations” or “hydronephrosis” or “ureteropelvic junction obstruction” or “posterior urethral valves” or “vesicoureteral reflux”. These publications represent studies on potential protein-based biomarkers, either individually or combined in panels, of RTMs in human and animal models.

Expert commentary: Successful use in the clinic of these protein-based biomarkers will need to involve larger scale studies to reach sufficient power. Improved performance will potentially come from combining immunoassay- and MS-based markers.  相似文献

In this review, recent advances in DNA microarray technologies and their applications are examined. Various DNA microarray platforms are described along with their methods for fabrication and their use. The second International Conference on Accelerating Biopharmaceutical Development was held in San Diego, California. The meeting was organized by the Society for Biological Engineering (SBE) and the American Institute of Chemical Engineers (AIChE); SBE is a technological community of the AIChE. Bob Adamson (Wyeth) and Chuck Goochee (Centocor) were co-chairs of the event, which had the theme “Delivering cost-effective, robust processes and methods quickly and efficiently.” The first day focused on emerging disruptive technologies and cutting-edge analytical techniques. Day two featured presentations on accelerated cell culture process development, critical quality attributes, specifications and comparability, and high throughput protein formulation development. Biomolecules and living cells can be printed in high‐resolution patterns to fabricate living constructs for tissue engineering. To evaluate the impact of processing cells with rapid prototyping (RP) methods, we modeled the printing phase of two RP systems that use biomaterial inks containing living cells: a high‐resolution inkjet system (BioJet) and a lower‐resolution nozzle‐based contact printing system (PAM2). In the first fabrication method, we reasoned that cell damage occurs principally during drop collision on the printing surface, in the second we hypothesize that shear stresses act on cells during extrusion (within the printing nozzle). The two cases were modeled changing the printing conditions: biomaterial substrate stiffness and volumetric flow rate, respectively, in BioJet and PAM2. Results show that during inkjet printing impact energies of about 10?8 J are transmitted to cells, whereas extrusion energies of the order of 10?11 J are exerted in direct printing. Viability tests of printed cells can be related to those numerical simulations, suggesting a threshold energy of 10?9 J to avoid permanent cell damage. To obtain well‐defined living constructs, a combination of these methods is proposed for the fabrication of scaffolds with controlled 3D architecture and spatial distribution of biomolecules and cells. © 2012 American Institute of Chemical Engineers Biotechnol. PurposeTo propose a “staggered overlap” technique in volumetric modulated arc therapy (VMAT) for craniospinal irradiation (CSI) and compare the dose distribution and plan robustness with “overlap” technique and “gradient optimization” approach.Methods and Materials6 patients previously treated in our clinic were retrospectively selected. 9 VMAT plans of each patient were optimized with “staggered overlap”, “overlap” and “gradient optimization” in overlapping region of 3 cm, 6 cm, and 9 cm separately. For the “staggered overlap” plan, adjacent field sets were intentionally overlapped by staggering field edges in an appropriate step size to avoid sharp dose gradient. Evaluation metrics including V95%, D2%, D98%, conformity number (CN) and homogeneity index (HI) were employed to evaluate the dose distribution. Moreover, shifts of the upper spinal field isocenter in each direction were performed to simulate junction errors for robustness analysis.ResultsThe CN and HI of VMAT plans with “staggered overlap” were 0.82 (0.811–0.822) and 0.113 (0.112–0.114), while they were 0.778 (0.776–0.782) and 0.131 (0.130–0.131) for plans with “gradient optimization”. In the robustness study, <3% dose deviations were found for 5 mm shifts in lateral and vertical directions with all techniques. In cranial-caudal direction, “overlap” technique created hot spots (D2% > 170%) and cold spots (D98% < 44%) in the junction region with 10 mm shifts. The dose deviations were decreased to 22% for plans with “staggered overlap” and 9 cm overlapping region.Conclusion“Staggered overlap” technique provides better plan quality as compared to “gradient optimization” approach and makes the plan more robust against junction errors as compared to “overlap” technique.  相似文献
