We assessed the fertility (reproductive success, litter size, testis weight, spermatocyte-to-spermatid ratio) of F1s and backcrosses between different wild-derived outbred and inbred strains of two mouse subspecies, Mus musculus domesticus and M. m. musculus . A significant proportion of the F1 females between the outbred crosses did not reproduce, suggesting that female infertility was present. As the spermatocyte-to-spermatid ratio was correlated with testis weight, the latter was used to attribute a sterile vs. fertile phenotype to all males. Segregation proportions in the backcrosses of F1 females yielded 11 (inbred) to 17% (outbred) sterile males, suggesting the contribution of two to three major genetic factors to hybrid male sterility. Only one direction of cross between the inbred strains produced sterile F1 males, indicating that one factor was borne by the musculus X-chromosome. No such differences were observed between reciprocal crosses in the outbred strains. The involvement of the X chromosome in male sterility thus could not be assessed, but its contribution appears likely given the limited introgression of X-linked markers through the hybrid zone between the subspecies. The central coastal region of Tunisia harbours two chromosomal races of the house mouse (2 n  = 22, 22Rb; 2 n  = 40, 40Std), which are genetically differentiated and show a high level of reproductive isolation. This study presents an allozyme analysis of house mice from the inland city of Kairouan 75 km from the coastal region. Results showed that the 22Rb and 40Std mice from Kairouan shared the same high degree of variability, and were not genetically differentiated. This contrasts with the genetic data registered for the two chromosomal races from the coastal towns, in which a particularly low genic diversity was observed in the 22Rb populations. As the two races in Kairouan show the same low hybridization rates as the populations in the coastal region, these results argue in favour of a local differentiation of the Rb race in Tunisia, most likely originating in Kairouan. The data further suggest that the decrease in variability observed in the coastal 22Rb populations is related to a founder effect or a bottleneck following the introduction of mice from Kairouan into one of these localities, prior to their dispersion throughout the coastal region.  © 2005 The Linnean Society of London, Biological Journal of the Linnean Society , 2005, 84 , 515–521.  相似文献

We investigated the hybrid origin of × Crepidiastrixeris denticulato-platyphylla using RAPDs and ITS sequence data. The putative parents Paraixeris denticulata and Crepidiastrum platyphyllum represent separate species, irrespective of geographical origin. The occurrence of species specific RAPD markers from P. denticulata and C. platyphyllum in × C. denticulato-platyphylla established unambiguously a hybrid origin between the two taxa. This was in line with the occurrence of a combination of morphological characters such as plant habit and floret numbers. The parent taxa differed from each other by 7 nucleotide substitutions and 2 indel events in the ITS region. The hybrids showed sequence additivity and most likely represent F1 plants, with the exception of two plants which were of possible F2 origin, possessing either the ITS sequences of one parent only, or one predominant ITS type. The hybrids occurred in two out of three localities where the parents occurred sympatrically. Hybridization between divergent lineages often results in reduced hybrid viability. Here we report findings from a series of independent molecular analyses over several seasons on four life stages of F1 hybrids between the newts Triturus cristatus and T. marmoratus . These two species form a bimodal hybrid zone of broad overlap in France, with F1 hybrids making up about 4% of the adult population. We demonstrate strong asymmetry in the direction of the cross, with one class ( cristatus -mothered) making up about 90% of F1 hybrids. By analyzing embryos and hatchlings, we show that this asymmetry is not due to prezygotic effects, as both classes of hybrid embryos are present at similar frequencies, implicating differential selection on the two hybrid classes after hatching. Adult F1 hybrids show a weak Haldane effect overall, with a 72% excess of females. The rarer marmoratus -mothered class, however, consists entirely of males. The absence of females from this class of adult F1 hybrids is best explained by an incompatibility between the cristatus X chromosome and marmoratus cytoplasm. It is thus important to distinguish the two classes of reciprocal-cross hybrids before making general statements about whether Haldane's rule is observed.  相似文献

Adult roach, bream and their presumed F1 hybrid from an Anglian Water reservoir were identified on the basis of morphological and meristic characteristics. The hybrid was clearly intermediate. Four hybrid breeding crosses were induced to spawn by hypophysis. A bream × roach cross (female named first) failed to produce fertile eggs, whereas F1 hybrid × roach, roach × F1 hybrid and F1 hybrid × F1 hybrid all produced fry. Fertility (defined as survival of eggs to hatching) was high for the F1 hybrid × roach back-cross (56%) but low for the others (<2%), in comparison to the pure species controls (roach 69%, bream 76%). Progeny from these crosses were reared until anal fin rays could be counted. These counts indicated intermediacy between the parents and back-crossed individuals, and similarity between F1 hybrids and their F2 progeny.  相似文献

The chrysomelid beetles Chrysochus auratus and C. cobaltinus form a narrow hybrid zone in western North America. We used a combination of direct and indirect analyses to examine the fitness of Chrysochus hybrids. For the direct analyses, we compared the mating frequency, longevity, fecundity and fertility of hybrid females and parentals. For the indirect approach, we tested predictions of multilocus genotype frequencies at a focal site in the hybrid zone, based on the frequencies of mating combinations during the previous generation. Hybrid females produced fewer eggs than did parentals and the eggs they produced in the lab failed to hatch, in contrast to those of parental females. In addition, contrary to predictions that 15.8% of the individuals at the focal site would have multilocus genotypes other than those expected of parentals or F1 individuals, we found no such genotypes at this site. This hybrid zone appears to be an example of a classic tension zone, with endogenous selection against hybrid individuals. We discuss the implications of low hybrid fitness for the evolution of premating barriers in this system, and argue that the integration of direct and indirect approaches is a powerful means of assessing the relative fitness of hybrids, particularly for species in which mate choices are easy to observe in the field.  © 2005 The Linnean Society of London, Biological Journal of the Linnean Society , 2005, 84 , 273–286.  相似文献

A cross between queen butterflies of the Palaeotropical species Danaus chrysippus and the Neotropical D. gilippus was achieved with difficulty in both directions. Only one progeny ( N  = 70) was reared comprising sterile males and inviable females in a precisely 1 : 1 ratio. Both prezygotic and postzygotic barriers to gene flow are strong. The result supports Haldane's Rule, to which we propose a minor amendment. The F1 hybrids were intermediate for background colour between the brown (genotype BB ) of gilippus and orange (genotype bb ) of chrysippus . Most F1 pattern characters were also intermediate. In polymorphic chrysippus populations, because Bb heterozygotes are brown, or nearly so, we suggest the B allele may have evolved towards dominance in sympatry. Hybrid males show positive heterosis for body size. The close similarity of male genitalia between the allopatric, genetically distant species chrysippus  and gilippus , compared to their divergence between gilippus and its largely sympatric sister species eresimus , suggest that reinforcement of sexual isolation or reproductive character displacement have evolved in sympatry.  © 2002 The Linnean Societyof London, Biological Journal of the Linnean Society , 2002, 76 , 535–544.  相似文献

Supernatant malate dehydrogenase (MDH) isozymes (as visualized by starch gel electrophoresis) are encoded by two distinct gene loci in both the largemouth and smallmouth bass. When an interspecific F1 hybrid is formed between these two fish, a unique MDH isozyme is generated. The results of freeze-thaw molecular hybridization (which is the first application of this technique to MDH) indicate that this unique isozyme in the F1 hybrid is a heterodimer composed of one subunit of each parental type. The F1 hybrids produced F2 hybrids which in turn formed the F3 hybrid population. The inheritance of alleles at the MDH-B locus is consistent with a single Mendelian autosomal locus. Furthermore, there is no evidence of linkage between the lactate dehydrogenase-E locus and the MDH-B locus.  相似文献

To characterize the genetic basis of voluntary calcium consumption, we tested C57BL/6J mice (B6; with low avidity for calcium), PWK/PhJ mice (PWK; with high avidity for calcium) and their F1 and F2 hybrids. All mice received a series of 96-h two-bottle preference tests with a choice between water and the following: 50 m m CaCl2, 50 m m calcium lactate, 50 m m MgCl2, 100 m m KCl, 100 m m NH4Cl, 100 m m NaCl, 5 m m citric acid, 30 μ m quinine hydrochloride and 2 m m saccharin. Most frequency distributions of the parental and F1 but not F2 groups were normally distributed, and there were few sex differences. Reciprocal cross analysis showed that B6 × PWK F1 mice had a non-specific elevation of fluid intake relative to PWK × B6 F1 mice. In the F2 mice, trait correlations were clustered among the divalent salts and the monovalent chlorides. A genome screen involving 116 markers showed 30 quantitative trait loci (QTLs), of which six involved consumption of calcium chloride or lactate. The results show pleiotropic controls of calcium and magnesium consumption that are distinct from those controlling consumption of monovalent chlorides or exemplars of the primary taste qualities.  相似文献

Three species of Darwin's Finches hybridize on the Galapagos island of Daphne Major. We examined mating patterns to determine if hybrids exhibit mate preferences. Geospiza fortis x G. scandens F1 hybrids backcrossed to both of the parental species, whereas all backcrosses bred with the parental species to which they were most related, or with hybrids. Paternal song was shown to be the crucial factor determining the mating pattern of G. fortis x G. scandens F1 hybrids and their offspring. Song is culturally inherited, transmitted faithfully from father to son (with few exceptions) as a result of an imprinting-like process. Size also contributes to the choice of mates. G. fortis x G. scandens F1 hybrid females paired with large G. scandens -like G. fortis males. G. fortis x G. fuliginosa F1 hybrids paired negatively assortatively with respect to the size of their G. fortis mates. Non-random mating of hybrids based on song, a non-generic trait, has interesting evolutionary consequences. Song characteristics and nuclear and mitochondrial genes flow from G. fuliginosa into the G. fortis population, whereas the direction of transfer of genetic and song information between G. fortis and G. scandens depends on which song was sung by the father of the hybrids.  相似文献

Two chromosomal races of house mice are present in Tunisia, one represented by mice carrying the 40-acrocentric standard karyotype and the other by a Robertsonian race (2re = 22) homozygous for nine centric fusions (Rb). A comparative summary on allozyme divergence, geographical distribution and level of reproductive isolation in the Tunisian and European Rb races is presented, to which new data on mitochondrial DNA and morphological divergence are added. The Tunisian 22Rb race revealed unique features not matched by the European chromosomal races, such as a decrease in allozymic variability, a higher level of genetic and morphological differentiation and a mosaic geographical distribution. The mtDNA analysis argued in favour of a local origin of the chromosomal divergence suggesting that the higher level of differentiation between the Tunisian races resulted from the older age of the 22Rb race and/or from a severe botdeneck. The decrease in fertility of chromosomal hybrids between the Tunisian races was compatible with the limited genetic introgression between diem. Furthermore, data on the restricted distribution of hybrid populations suggested that premating reproductive barriers may be evolving. The Tunisian 22Rb race is thus an appropriate model to investigate a chromosomally-mediated speciation event.  相似文献

Abstract.— We analyzed the rate at which postzygotic incompatibilities accumulate in birds. Our purposes were to assess the role of intrinsic F1 hybrid infertility and inviability in the speciation process, and to compare rates of loss of fertility and viability between the sexes. Among our sample more than half the crosses between species in the same genus produce fertile hybrids. Complete loss of F1 hybrid fertility takes on the order of millions of years. Loss of F1 hybrid viability occurs over longer timescales than fertility: some viable hybrids have been produced between taxa that appear to have been separated for more than 55 my. There is strong support for Haldane's rule, with very few examples where the male has lower fitness than the female. However, in contrast to Drosophila , fertility of the homogametic sex in the F1 appears to be lost before viability of the heterogametic sex in the F1. We conclude that the time span of loss of intrinsic hybrid fertility and viability is often, but not always, longer than the time to speciation. Premating isolation is an important mechanism maintaining reproductive isolation in birds. In addition, other factors causing postzygotic reproductive isolation such as ecological causes of hybrid unfitness, reduced mating success of hybrids, and genetic incompatibilities in the F2s and backcrosses may often be involved in the speciation process.  相似文献

From 25 populations of Pseudorasbora in Japan, polymorphism at 22 allozyme loci indicated that the level of genetic differentiation between Pseudorasbora parva and Pseudorasbora pumila ( D  = 0·421–0·517) was greater than that between the two subspecies of P. pumila , P. pumila pumila and P. pumila subsp. ( D  = 0·164), consistent with morphological differences. While P. parva displayed genetic variation ( H  = 0·003–0·100) similar to other freshwater fishes, P. pumila pumila and P. pumila subsp. populations showed no genetic variation. In five of the 15 populations collected from the contact zone between P. parva and P. pumila pumila , hybrids were detected by allozyme analyses. All hybrids were presumed to represent the F1 generation, because they were heterozygous at all 12 loci diagnostic between P. parva and P. pumila pumila . Although four populations were characterized by high frequencies of F1 hybrids ( c . 40%), only one of the parental species was observed in each case. The results indicated that the two species have hybridized easily under natural conditions, but cannot coexist in the long-term. It is suggested that continued invasion of P. parva would hasten the extinction of P. pumila pumila .  相似文献

Phylogenetic analyses based on mtDNA cytochrome  b were performed in 42 lizards of the Gymnodactylus darwinii complex from three regions within Brazil's Atlantic Forest. Mainland regions and continental shelf islands in the south-eastern range and mainland areas from the north-east were sampled. The criteria of maximum parsimony (MP), maximum likelihood (ML) and Bayesian methods were explored, with the robustness for nodes assessed by bootstrapping (MP and ML) and posterior probabilities (Bayesian searches). By all methods, three distinctive phylogroups were recovered: a south-eastern clade (SE) and two clades from northern regions (NE1 and NE2). The pattern of genetic structure of the major clades coincided with the presence of river systems in the Atlantic Forest, and based on corrected genetic distances between those clades, divergence times were tentatively estimated using mtDNA rates calibrated for squamate reptiles. The putative role of Atlantic Forest rivers in generating differentiation is discussed. We present a hypothesis of species limits for G. darwinii , based on concordant lines of evidence including cytogenetic and mtDNA analyses. Two chromosome races (cytotype A, 2n = 38; and cytotype B, 2n = 40) had distributions concordant with clades SE and NE1 + NE2, respectively. The effect of hybridization on morphological variation was investigated in 120 western house mice, Mus musculus domesticus , from the hybrid zone between the Barcelona and standard chromosomal races. The incidence of 37 non-metric cranial traits was calculated for standard mice (2 n  = 40) and Barcelona-standard hybrids (2 n  = 27–39). Subsequent analyses were conducted on several karyological subgroups, established by grouping the animals according to either their diploid number or their degree of chromosomal heterozygosity. Results revealed no significant difference by sex, asymmetry, or geographical distance. Significant phenetic divergences were found between the karyotypes studied in relation to several variants. Differences were especially substantial between the standard race and hybrid mice, even with respect to those hybrids with karyotypes close to that of the standard race. Within the hybrids, the maximum divergence corresponded to the 28-chromosome homozygotes, chromosomally close to the Barcelona race, and to the heterozygotes with more than two fusions. Since differences in non-metric trait frequencies are generally considered a measure of genetic divergence, the results suggest the occurrence of a barrier to gene flow in the Barcelona hybrid zone. Transgenic oilseed rape ( Brassica napus ) plants can interbreed with nearby weedy Brassica rapa , potentially enhancing the weediness and/or invasiveness of subsequent hybrid offspring. We have previously demonstrated that transgenic mitigation effectively reduces the fitness of the transgenic dwarf and herbicide-resistant B. napus volunteers. We now report the efficacy of such a tandem construct, including a primary herbicide-resistant gene and a dwarfing mitigator gene, to preclude the risks of gene establishment in the related weed B. rapa and its backcross progeny. The transgenically mitigated and non-transgenic B. rapa  ×  B. napus interspecific hybrids and the backcrosses (BC1) with B. rapa were grown alone and in competition with B. rapa weed. The reproductive fitness of hybrid offspring progressively decreased with increased B. rapa genes in the offspring, illustrating the efficacy of the concept. The fitness of F2 interspecific non-transgenic hybrids was between 50% and 80% of the competing weedy B. rapa , whereas the fitness of the comparable T2 interspecific transgenic hybrids was never more than 2%. The reproductive fitness of the transgenic T2 BC1 mixed with B. rapa was further severely suppressed to 0.9% of that of the competing weed due to dwarfism. Clearly, the mitigation technology works efficiently in a rapeseed crop–weed system under biocontainment-controlled environments, but field studies should further validate its utility for minimizing the risks of gene flow.  相似文献

Ecological selection against hybrids, the reduction in hybrid fitness attributed solely to environmental factors, was tested by introducing young-of-the-year benthic, limnetic and F1 hybrid sticklebacks Gasterosteus aculeatus to divided experimental ponds and lake enclosures. The frequency of hybrids in samples taken at the end was significantly lower than their frequency at introduction. Hybrid survival was significantly lower in pond-sides in which they were initially the most common cross type than in pond-sides in which they were initially rare, suggesting that hybrid survival may be frequency-dependent. Growth rate of F1 hybrids was marginally lower than benthic growth rates, being significantly lower than in ponds and not different in lake enclosures. The diet of hybrids overlapped with both parent species in ponds and with benthic diets in lake enclosures. The results suggest that ecological selection is acting against young-of-the-year hybrid sticklebacks.  相似文献

The Robertsonian phenomenon in house mice (Mus musculus domesticus) from Tunisia consists in the presence of only one 22-chromosome Robertsonian race (22Rb) carrying the maximum number of fusions observed until now. The 22Rb populations exclusively occupy urban centers in the Eastern-Central region of Tunisia where standard population with 40-all acrocentric chromosomes (40Std) occur in surrounding neighborhoods and rural environments. In addition to the habitat partition, allozyme and mitochondrial DNA analyses showed that the 22Rb populations were genetically differentiated from the 40Std ones. This differentiation mostly stemmed from an important decrease in genetic variability in the 22Rb populations from the Sahel towns. The extent of morphological, ecological and genetical divergence observed between these chromosomal races in Tunisia is in agreement with the predictions of the chromosomal speciation model of White which advocates that karyotypic differentiation between taxa can lead to their reproductive isolation and independent evolution. Such a process is verified if the Rb process in Tunisia results from local differentiation which is supported by both the genetic and morphological data. However, the hypothesis of an origin by introduction of these mice from another region of Tunisia or from another country cannot be totally dismissed. In this study, an allozymic analysis of mice (22Rb and 40Std) from the geographically distant city of Kairouan was performed. Results showed that 22Rb and 40Std mice from Kairouan shared the same high degree of variability, and were not genetically differentiated. This contrasts with the results registered in the two chromosomal races in the Sahel towns. Such data argue in favor of a local differentiation of the Robertsonian process in Tunisia and suggest that the decrease in variability of the structural nuclear genes in the Sahel 22Rb populations can be related to an introduction from Kairouan into a Sahel locality resulting in a founder effect or followed by a severe bottleneck prior to its dispersion throughout the Sahel region.  相似文献
