In order to elucidate the effects of the antibiotic cefotaxime on callus growth and morphogenesis, we incubated embryogenic maize calli (Zea mays L.) of A188 and R91 lines and of their F1 hybrid with 50–500 mg/l cefotaxime throughout several subcultures. Cefotaxime did not affect the induction frequency and growth of the embryogenic callus but enhanced its morphogenesis. In both tested lines and a hybrid, the highest increase in the number of regenerated plants was observed at the antibiotic concentration of 150 mg/l. The degree of morphogenesis stimulation and the range of cefotaxime concentrations effective in stimulation of plant regeneration depended on the properties of calli obtained from tested genotypes.  相似文献

An antibiotic, cefotaxime (Omnatax) has been found to promote somatic embryogenesis and subsequent plant regeneration in vitro in indica-type basmati rice cultures. Response was highly genotype specific. The number, mass and morphology of the calli formed on the scutellar tissues were dependent on the growth medium (with or without cefotaxime). The embryogenic nature of nodular calli was confirmed through histological analysis and their plant regeneration ability. The calli of variety Pusa basmati 1 grown on medium supplemented with cefotaxime (100 mg/L) exhibited up to 70.5% plant regeneration as compared to control (51.51%). Plants regenerated from emryogenic calli were phenotypically normal and identical to seed-derived plants and exhibited normal fertility. A limited humidity and an optimal aeration of the culture tubes further enhanced the frequency of somatic embryogenesis and plant regeneration.  相似文献

In vitro culture ofBrassica alba anthers on a growth medium containing inorganics of KB5 and organics, iron, sucrose and hormones of B5 resulted in a very high response of anthers (93.75%) towards callus induction. All the calli transferred to regeneration media responded favourably even after six months of callus induction. Numerous torpedo-shaped embryoids developed in clusters at many sites from each callus mass. Secondary embryogenesis and multiple shoot formation was also observed in many cases. The number of embryoids and plantlets produced by one embryogenic anther were as high as 169.8 and 17 respectively. 87% of the regenerated plants were haploids.  相似文献

A cephalosporin antibiotic, cefotaxime (Omnatax™) promoted somatic embryogenesis and subsequent shoot regeneration in vitro from spindle in sugarcane irrespective of the genotypes as (CoJ 83, CoJ 88 and CoJ 64) culturered on MS medium with 2,4-D (2.5 mgl−1) and kinetin (0.5 mgl−1). Seven different concentrations of cefotaxime (100, 200, 300, 400, 500, 600 and 700 mgl−1) were tested to find the optimal concentration of cefotaxime for somatic embryogenesis from callus cultures. Among the three varieties, calli of variety CoJ 83 incubated on MS medium with 2,4-D (2.5 mgl−1) + kinetin (0.5 mgl−1) + cefotaxime (500 mgl−1) exhibited maximum somatic embryogenesis. To improve shoot regeneration, the callus was transferred to MS medium with BAP (0.5 mgl−1) + kinetin (0.5 mgl−1) in combination with different levels of cefotaxime. Highest frequency of shoot regeneration was observed in callus of CoJ 83 in the presence of 500 mgl−1 cefotaxime. The plantlets could be successfully hardened in polybags and transferred to soil, where they exhibited normal growth. Our results convincingly demonstrated that cefotaxime improves somatic embryogenesis from spindle and regeneration from embryogenic calli of sugarcane and hence can be strongly recommended for rapid and large scale multiplication of sugarcane.Key words: Saccharum officinarum L., leaf segments, callus, plant regeneration, antibiotic  相似文献

The origin and development of somatic embryos in calli initiated from immature zygotic embryos of Picea abies (L.) Karst. (Norway spruce) and P. glauca (Moench) Voss (white spruce) was studied. Immature zygotic embryos cultured on callus induction medium produced two types of white calli that were phenotypically different from one another. The callus that proliferated from the hypocotyl region was white to translucent, glossy, mucilaginous and embryogenic. The callus mass which originated from the radicle end was reddish-white, nonmucilaginous and nonembryogenic. Whole mount preparations of the entire explant with two different types of calli showed the presence of embryogenic cells in the mucilaginous callus mass derived from the hypocotyl region of the zygotic embryo. Summary Influence of formaldehyde on growth and secondary metabolite production inin vitro grown tissues of pyrethrum, capsicum and carrot were studied. Formaldehyde concentration above 0.025% completely inhibited the growth of addedBacillus andAspergillus in the culture medium. Formaldehyde up to 0.025% level caused slight inhibition in the growth of pyrethrum callus. Callus subjected to 0.05, 0.1 and 0.2% formaldehyde enhanced the level of pyrethrin production in pyrethrum callus, whereas production of phenolics was lower in all the treatments. Growth of carrot callus and anthocyanin production was not inhibited up to a formaldehyde concentration of 0.04%. Similarly, the production of capsaicin in immobilised cell cultures ofCapsicum frutescens was not inhibited up to 0.04% level of formaldeyde. The results demonstrate the usefulness of formaldehyde to control contamination in plant tissue culture.  相似文献

In the following a method for the induction of pistil-like structures in wheat suspension cultures (Triticum aestivum L.) is described. In young influorescences of plants, which were artificially infected with the wheat bunt fungi (Tilletia controversa), organogenic calli with pistil-like structures could be induced on loblolly pine medium + 3 mg/l 3,6-dichloro-2-methoxy benzoic acid. For optimization of the transformation procedure with Pisum sativum L. stern explant callus was used to test the effect of disarmed Agrobacterium tumefaciens strains, cocultivation procedures (preconditioning of explants; use of Nicotiana tabacum L. nurse cultures), duration of cocultivation (2, 3 or 4 days), and agents for selection (kanamycin or hygromycin). The succinamopine strain EHA101(pBI1042) produced the highest percentage of transformed calli (77%) when used in conjunction with tobacco nurse culture during four days of cocultivation. Using this strain, kanamycin (76%) and hygromycin (77%) were equally effective selective agents, but for strain LBA4404(pBI1042) percentage of transformed calli was higher for hygromycin (63%) than for kanamycin (17%). The procedures and strains shown to be optimal for transformation of pea callus will now be complemented by a pea regeneration system.  相似文献

In vitro regeneration of plants from both cotyledon-, and leaf — derived calli of Lotononis bainesii Paker was studied under defined nutritional, hormonal and environmental conditions. Explants from both, cotyledons from seedilings of 4 days old and fully expanded leaves from mature plants, were cultured on MS medium containing 0.8% agar and supplemented with 0.01, 0.1, and 1 mg/1 concentrations of naphthaleneacetic acid (NAA) and 0.1, 1, and 3 mg/1 levels of benzyladenine (BA) in various combinations. Multiple shoot (on an average 4 shoots per callus) regeneration from primary callus occurred within 15 to 35 days of culture in most of the media tested. Although the best medium for shoot regeneration from cotyledon-derived callus contained NAA and BA at 1, and 0.1 mg/1 levels, respectively, maximal shoot regeneration from leaf-derived calli was achieved by using NAA and BA at 0.01 and 0.1 mg/1, respectively. Pollen plants were obtained from anther-derived calli of the indica rice variety Basmati-370. Anther-response (anthers producing pollen derived calli) and plant regeneration frequency from the pollen derived calli. was very low. Donor plants which flowered at the average max/min. temperature of 34.2°/23.3°C gave a significantly higher anther-response to in vitro techniques, than did those which flowered at 29.1°/16.4°C. Somatic callus induction and subsequent plant regeneration was readily obtained from mature seed embryos. The effect of abscisic acid (ABA) on plant regeneration in anther-derived calli of rice Oryza sativa L. varieties Taipei 177, Taipei 309 and Fujisaka 5 was studied.ABA at concentrations up to 4×10–6 M stimulated fresh weight increase in Taipei 309 and Fujisaka 5 while higher concentrations effected corresponding weight decreases in the three varieties tested. Relatively high ABA concentrations resulted in decreased callus size and production of more compact and whitish calli. Plantlets were regenerated from a selected salt-tolerant cell line of Shamouti orange (Citrus sinensis L. Osbeck). Embryogenesis was carried out both in the presence and absence of NaCl, yielding green and white globular embryos, respectively. Greening could be induced subsequently and normal heart shape embryo development was obtained. Plantlet formation required exposure to kinetin prior to the introduction of the root-inducing hormone naphthalene acetic acid. This system differs from the designed protocol for plant regeneration from the salt-sensitive, i.e., unselected callus. It is concluded that NaCl interferes with the regeneration process, with embryogenesis and/or embryo development into plantlets. Its presence during callus growth probably changes the balance of the phytohormones which is later manifested in plant regeneration. Citrus salt-tolerant callus yields salt-tolerant embryos. Salt-tolerant calli derived from regenerated plantlets indicate acquisition of salt tolerance on the whole plant level.  相似文献

Summary Seventeen cultivars of cotton (Gossypium hirsutum L.) were evaluated for callus initiation and maintenance using 3 initiation media and 3 maintenance media. After a series of transfers of a 3% glucose media, calli were placed on a 3% sucrose medium. After several weeks calli were observed for the presence of embryo-like structures. Cultivars Coker 201 and Coker 315 were identified as embryogenic. Embryogenic callus has since been routinely obtained within 6 weeks by initiating callus on glucose media for 3–4 weeks followed by transfer to sucrose media. Histological examination has shown that embryos are derived from isodiametric, densely cytoplasmic cells and follow predictable patterns of development. Upon maturity, transfer to auxin-free media with reduced sucrose levels results in embryo germination. 条对龙胆愈伤组织中龙胆苦苷含量较低,根的分化可以提高龙胆苦苷的含量。培养的离休根中龙胆苦苷的含量则更高。随着继代次数的增加,愈伤组织中龙胆苦苷含量下降快,而分化根的愈伤组织和离体培养根中龙胆苦苷含量下降较慢。在愈伤组织的液体培养中,1/2MS培养基附加低浓度的植物激素,龙胆苦苷含量有所提高。植物激素配比促进愈伤组织中分化根的形成,也促进龙胆苦苷的产生;但促进愈伤组织的生长时,不利于龙胆苦苷产生。在离体根培养中,添加 1mg/L的金雀花碱,促进龙胆苦苷的产生,含量可达1.48%,明显高于愈伤组织(0.52%)和分化根的愈伤组织(0.65%),也高于试管苗根的龙胆苦苷含量(1.01%)。  相似文献

An assay was performed to identify the antibiotics that are most effective againstAgrobacterium tumefaciens strains EHA101 and LBA4404, and to determine if these antibiotics inhibited tobacco callus and shoot formation. We tested ten antibiotics: cefotaxime, carbenecillin, erythromycin, spectinomycin, polymixin B, chloramphenicol, methicillin, Augmentin 500, Augmentin 250, and moxalactam. The effectiveness of each antibiotic against the two strains was determined by measuring the zones of inhibition of bacterial growth in a disk-diffusion assay. The five antibiotics that generated the largest zones of inhibition for each strain were assayed to determine their effects on callus formation. Cefotaxime was the most active antibiotic tested against strain LBA4404, and moxalactam was the most effective antibiotic against EHA101. Both cefotaxime and moxalactam had little or no effect on callus development.  相似文献

Induction of crown galls on 4–6 week old soybean (Glycine max (L.) Merr.) plants cultured in growth chambers was obtained with Agrobacterium tumefaciens strains C58, T37 and ACH5. The crown galls were isolated and cultured in vitro as sterile callus and liquid suspension cultures. Transformation was tested by opine tests and by molecular hybridization of restricted cell DNA with T-DNA fragments. Protoplasts were isolated from suspension cultures. Transformed protoplasts regenerate cell walls, divide and form calli without an exogenous supply of hormones.  相似文献

Callus was induced from the bulb of Allium wakegi Araki on MS semisolid medium supplemented with several growth regulating substances. The calli were subcultured every 40 days. At the time of every subculture the callus was subdivided to be used for chromosome studies, plant regeneration, or continuous callus multiplication. The chromosome constitution of cells in callus and regenerated plants varied over the culture period, and at the 3rd subculture amphidiploid plants were obtained. They appeared even more frequently than amphihaploid plants in the 4th subculture. Hypoamphihaploid regenerants appeared as stumpy shoots but none of these shoots proceeded further to form a normal plant. By Giemsa C-banded karyotype, the chromosome constitution of amphidiploid plants was found to result from exact doubling of the chromosome sets of amphihaploid common species. Amphidiploid plants show better viability and growth than common plants. The possibility and the expectation of new crop plants to be developed from amphidiploid plants will be discussed.  相似文献

The effects of an auxin (NAA) and a cyto kinin (BA), as well as those of peptone, thiamin and coconut water (CW) on callus and protocorm-like body (PLB) induction from root tips of Oncidium varicosum grown in vitro were studied. Relatively high concentrations of NAA favoured callus induction. PLBs were obtained from callus culture either in medium with 15% CW+1.25 mg/1 NAA or in the presence of CW alone. The low regeneration capac ity of buds presented by the calli seems to be the result of an incomplete dedifferentiation of their cells. This seems to keep the callus in a root state.
