The presence ofGeotrichum candidum citrus race, the citrus sour rot pathogen, was examined in soils of citrus groves and non-citrus fields of Japan. Soil samples were collected from 223 sites (118 sites in citrus groves, and 105 sites in fields cultivated with 33 species of non-citrus plants and in evergreen broad-leaved forest) in 11 main citrus-growing prefectures, and Hokkaido, a non-citrus-growing area. Of 236 soil samples from citrus groves, 95.76% containedG. candidum citrus race and 0.42% contained the non-citrus race; while of 210 samples from non-citrus fields, 62.85% and 4.76% contained the citrus race and the non-citrus race respectively. All of the citrus race isolates obtained either from citrus groves or non-citrus fields were pathogenic on lemon (Citrus limon) and satsuma mandarin (Citrus unshiu), but some of these isolates failed to infect orange (Citrus sinensis). The non-citrus races were pathogenic on ripe tomato fruit (Lycopersicon esculentum) and ripe muskmelon fruit (Cucumis melo var.reticulatus). Results indicated that citrus sour rot pathogen is widely distributed in citrus groves and non-citrus fields of diverse plant species in Japan.  相似文献

A pectin lyase, poly(methoxygalacturonide) lyase, EC 4.2.2.10, from a culture filtrate of Penicillium expansum was partially purified 33-fold with 7.3% yield. The enzyme was monomeric with a molecular mass of 36.5 kDa. The enzyme did not contain pectate lyase activity and degraded citrus and apple pectin best at pH 7.0 and 40 to 45°C. The K m for citrus pectin was 9 mg ml-1.  相似文献

Field surveys were conducted from 2004 to 2007 to determine the species composition and relative abundance of natural enemies associated with colonies of either the citrus red mite, Panonychus citri, or the two spotted spider mite, Tetranychus urticae, in Valencian citrus orchards (eastern Spain). Fourteen species were recorded, six phytoseiid mites and eight insect predators. Two of them are reported for the first time on citrus in Spain and two more are first reports as predators associated with T. urticae. The community of predators associated with T. urticae and P. citri was almost identical, and the Morisita–Horn index of similarity between both natural enemy complexes was close to one, suggesting that predators forage on both pest species. Quantifying the presence of many known spider mites predators in Valencian citrus orchards is an important first step towards spider mite control. A challenge for future studies will be to establish conservation and/or augmentation management strategies for these predators, especially to improve T. urticae biological control.  相似文献

The parasitoid complex of brown soft scale, Coccus hesperidum L., a multivoltine soft scale, was determined in southern California citrus over the period February 2004–March 2006. The survey was conducted by placing brown soft scale-infested yucca leaves in the canopy of citrus trees and subsequently rearing individually isolated parasitized scales in the laboratory. A total of 14 species parasitized brown soft scale in the field, the most abundant ones belonging to the genus Metaphycus Mercet (75%). The most abundant parasitoid species was Metaphycus angustifrons Compere (38% parasitism), and this is a new record of establishment for this species in California. Coccophagus species accounted for only 11% parasitism. There were important spatio-temporal differences across the parasitoid complex survey locations. We also found that the five most abundant encyrtid parasitoid species showed preferences for scales of different sizes. Our results have implications for biological control of citricola scale, Coccus pseudomagnoliarum (Kuwana), an important pest of citrus in the San Joaquin Valley of central California. Notably, this species is nearly absent in southern California. Brown soft scale is considered to be an alternate host for parasitoids of citricola scale, a univoltine soft scale, at times when the latter species is unavailable for parasitism.  相似文献

Summary Cross-inoculation experiments with 10 pure cultured strains and 17 host species were carried out. The 10 strains were isolated from the root nodules on actinorhizal trees ranging in 9 species, 5 genera and 4 families. The host species belong to 5 genera. The pure cultured strains fromAlnus are of strong ability to infect different species of the same genus. The seedlings inoculated with these strains are able to nodulate normally. These strains can also infect and nodulate the seedlings ofMyrica californica, but not the seedlings of Elaeagnus, Casuarina andMyrica rubra. The pure cultured strains from Elaeagnus can infect and nodulate the host species in the same genus and family with an exception ofE. viridis vardelavayi, which can be only poorly nodulated by a few strains from Elaeagnus. The strains from Elaeagnus cannot infect the seedlings of Alnus andMyrica rubra. The results presented here suggest thatFrankia endophytes can be divided into two groups: Alnus group and Elaeagnus group.  相似文献

Brevipalpus phoenicis (Geijskes) (Acari: Tenuipalpidae) is recognized as the vector of citrus leprosis virus that is a significant problem in several South American countries. Citrus leprosis has been reported from Florida in the past but no longer occurs on citrus in North America. The disease was recently reported in Central America, suggesting that B. phoenicis constitutes a potential threat to the citrus industries of North America and the Caribbean. Besides B. phoenicis, B. obovatus Donnadieu, and B. californicus (Banks) have been incriminated as vectors of citrus leprosis virus and each species has hundreds of host plants. In this study, Brevipalpus mite specimens were collected from different plants, especially citrus, in the States of Florida (USA) and São Paulo (Brazil), and reared on citrus fruit under standard laboratory conditions. Mites were taken from these colonies for DNA extraction and for morphological species identification. One hundred and two Random Amplified Polymorphic DNA (RAPD) markers were scored along with amplification and sequencing of a mitochondrial cytochrome oxidase subunit I gene fragment (374 bp). Variability among the colonies was detected with consistent congruence between both molecular data sets. The mites from the Florida and Brazilian colonies were morphologically identified as belonging to B. phoenicis, and comprise a monophyletic group. These colonies could be further diagnosed and subdivided geographically by mitochondrial DNA analysis.  相似文献

Fermentation kinetics, digestibility, faecal characteristics and bacterial populations (aerobes, anaerobes, lactobacilli, lactic acid bacteria, enterococci, coliforms and clostridia) of dog food mixed with citrus pulp and apple pomace were evaluated. The in vitro gas production of a pre-digested dog food mixed with 0, 30, 50 and 70 g/kg dry matter (DM) of citrus pulp or apple pomace was measured, and also an experiment with dogs fed the same dog food with or without the addition of 70 g/kg of either fresh citrus pulp or apple pomace was conducted. Gas production increased linearly (p < 0.001) and quadratically (p < 0.001) as fibre levels augmented. The inclusion of fibre sources in the diets resulted in higher faecal output (p = 0.005) and defecation frequency (p < 0.001), and lower faecal pH (p < 0.001) and digestibility values (p < 0.01). Faecal consistencies and microbial populations did not differ among treatments. The addition of fresh citrus and apple was effective to stimulate the hindgut fermentation, but slightly depressed the digestion.  相似文献

Chiracanthium mildei, a common sac spider (Clubionidae) in Israel, is an important predator of pest insects in citrus and apple orchards. Field observations indicate that the females of this hunting spider are attracted to traps containing the parapheromone trimedlure and males of the Mediterranean fruit fly (Ceratitis capitata),which were caught in these traps. Laboratory testsshow that C. mildei females are attractedto olfactory cues from male C. capitata, but notto trimedlure. The incidence of predation of eight species of predacious mirids (Hemiptera: Miridae) present in an apple orchard of Québec on the green apple aphid, two-spotted spider mite and European red mite were investigated. The daily consumption rates varied from 1–2 green apple aphids forHyaliodes vitripennis Say andCampylomma verbasci Meyer to 7–9 aphids forDeraeocoris fasciolus Knight andLepidopsallus minisculus Knight.H. vitripennis consumed significantly more mites than the other mirid species with 26 and 18 mites per day for the two-spotted spider mite and the European red mite respectively. The combined use ofH. vitripennis andL. minisculus is suggested for the control of phytophagous mites. There are several examples of successful isolation, selection and development of antagonists of plant pathogens available for commercial use: mild strains of Tristeza virus of citrus for the control of Tristeza virus of citrus through preimmunization; Acremonium alternatum and A. persicinum for the control of tar spot of coconut (Catacauma torrendiella and Coccostroma palmicola); Trichoderma spp. for the control of Phytophthora cactorum on apple after soil treatment with formaldehyde and Dicyma pulvinata for the control of South American leaf blight (Microcyclus ulei) on rubber trees (Hevea brasiliensis) associated with polyclone plantations, are just a few. Citrus rust mite, Phyllocoptruta oleivora (Ashmead), is a major pest of citrus in Israel. Five species of predatory mites: Amblyseius herbicolus Chant, Euseius victoriensis (Womersley), Euseius elinae (Schicha), Typhlodromus rickeri Chant and Euseius stipulatus (Athias-Henriot), were imported to improve the biological control of the pest. Mite rearing and sampling methods were developed and improved, and the predators were released. Recovery in some citrus orchards was recorded, but only E. victoriensis became established in the north of Israel.  相似文献

越来越多的研究表明某些在环境中普遍存在的人与动物的病原微生物能够跨界侵染不同生物界的寄主。本文就Serratia marcescens,Enterobacter cloacae,Pseudomonas aeuriginosa,Klebsiella pneumoniae等动物条件病原细菌环境菌株跨界侵染植物的研究现状进行了综述。这些病原菌在自然界中普遍存在,能够利用与感染人类相同或不同的侵染策略跨界侵染植物,以拓宽其寄主范围。其中,肺炎克雷伯氏菌(Klebsiella pneumoniae)能在自然条件下引起玉米发生顶腐病,揭示了环境中的某些植物可作为各种病原细菌的天然储存库,在条件合适的情况下可能会感染人类和动物,以及在食品生产中的潜在危害。对这些跨界病原菌的研究,在人、动物和植物流行病学上具有非常重要意义,也为环境科学提出了新的研究热点。  相似文献

Allergy to citrus fruits is often associated with pollinosis and sensitization to other plants due to a phenomenon of cross-reactivity. The aims of the present study were to highlight the cross-reactivity among citrus and the major allergenic pollens/fruits, throughout clinical and molecular investigations, and to evaluate the sensitization frequency to citrus fruits in a population of children and adults with pollinosis. We found a relevant percentage of sensitisation (39%) to citrus fruits in the patients recruited and in all of them the IgE-mediated mechanism has been confirmed by the positive response to the prick-to-prick test. RT-PCR experiments showed the expression of Cit s 1, Cit s 3 and a profilin isoform, already described in apple, also in Citrus clementine pollen. Data of multiple sequence alignments demonstrated that Citrus allergens shared high percentage identity values with other clinically relevant species (i.e. Triticum aestivum, Malus domestica), confirming the possible cross-allergenicity citrus/grasses and citrus/apple. Finally, a novelty of the present work has been the expression of two phospholipaseA2 isoforms (PLA2 α and β) in Citrus as well as in Triticum pollens; being PLA2 able to generate pro-inflammatory factors, this enzyme could participate in the activation of the allergenic inflammatory cascade.  相似文献

Understanding the mechanisms responsible for divergence and specialization of pathogens on different hosts is of fundamental importance, especially in the context of the emergence of new diseases via host shifts. Temporal isolation has been reported in a few plants and parasites, but is probably one of the least studied speciation processes. We studied whether temporal isolation could be responsible for the maintenance of genetic differentiation among sympatric populations of Ampelomyces, widespread intracellular mycoparasites of powdery mildew fungi, themselves plant pathogens. The timing of transmission of Ampelomyces depends on the life cycles of the powdery mildew species they parasitize. Internal transcribed spacer sequences and microsatellite markers showed that Ampelomyces populations found in apple powdery mildew (Podosphaera leucotricha) were genetically highly differentiated from other Ampelomyces populations sampled from several other powdery mildew species across Europe, infecting plant hosts other than apple. While P. leucotricha starts its life cycle early in spring, and the main apple powdery mildew epidemics occur before summer, the fungal hosts of the other Ampelomyces cause epidemics mainly in summer and autumn. When two powdery mildew species were experimentally exposed to Ampelomyces strains naturally occurring in P. leucotricha in spring, and to strains naturally present in other mycohost species in autumn, cross‐infections always occurred. Thus, the host‐related genetic differentiation in Ampelomyces cannot be explained by narrow physiological specialization, because Ampelomyces were able to infect powdery mildew species they were unlikely to have encountered in nature, but instead appears to result from temporal isolation.  相似文献

Genetic relationships were studied by means of ten isoenzymatic systems, at the genus and species level, using two distances and four methods of aggregation in a germplasm collection of 198 cultivars and accessions of 54 species belonging to Citrus and 13 related genera. The most consistent results were obtained by the chord distance and the neighbor-joining clustering method. Citrus species were distributed in two main groups: the orange-mandarin group and the lime lemon-citron-pummelo group. The species C. halimii and C. tachibana are not included in these groups. Mandarin species fall into three main subgroups: one includes C. sinensis; the second, C. aurantium, the third, small-fruit species. The citron, the pummelo and the ancient lemon subgroups form a cluster to which the species belonging to subgenus Papeda and the cultivated limes, lemons and bergamots are related. Microcitrus spp, to which Severinia buxifolia and Atalantia ceylanica seem to be related, cluster with the lime lemon-citron-pummelo group while Fortunella is close to the orange-mandarin group. Poncirus trifoliata, the most important species for citrus rootstock improvement is located far from Citrus but connected to it through Fortunella spp. A broad distribution of species has been found that should be taken into account to sample new genotypes in the search of desired characters in order to fully and efficiently use genetic resources for citrus improvement.  相似文献

Novel thermophilic and alkaliphilic bacteria for the processing of bast fibres were isolated using hemp pectin as substrate. The strain PB94A, which showed the highest growth rate (μ = 0.5/h) was identified as Geobacillus thermoglucosidasius (DSM 21625). The strain grew optimally at 60°C and pH 8.5. During growth on citrus pectin, the strain produced pectinolytic lyases, which were excreted into the medium. In contrast to the commercially available pectinase Bioprep 3000 L, the enzymes from G. thermoglucosidasius PB94A converted pectin isolated from hemp fibres. In addition to hemp pectin, the culture supernatant also degraded citrus, sugar beet and apple pectin and polygalacturonic acid. When hemp fibres were incubated with the cell-free fermentation broth of G. thermoglucosidasius PB94A, the fineness of the fibres increased. The strain did not produce any cellulases, which is important in order to avoid damaging the fibres during incubation. Therefore, these bacteria or their enzymes can be used to produce fine high-quality hemp fibres.  相似文献

Host specificity tests carried out in the laboratory in Australia during 1977, showed thatMicrothrix inconspicuella Ragonot could develop on young apple leaves (Harley et al., 1979). Field studies in unsprayed apple orchards in South Africa showed that some feeding occurred, but fewer than 40% of late instar larvae developed to adults when confined in sleeves on apple tree branches. No feeding or survival occurred in large field cages or in the open. Adults which developed from apple fed larvae were smaller, deformed, occasionally mated and laid fertile eggs but their progeny did not feed or develop on apple fruit or leaves. In conclusion,M. inconspicuella larvae did not develop on apple fruit or leaves in the field, damage was mainly limited to apples already injured and feeding on leaves was minimal. Under normal pest control practicesM. inconspicuella populations did not survive on any part of the apple tree or onE. australis growing under the trees.   相似文献

Fruit trees, such as apple (Malus × domestica Borkh.), are woody perennial plants with a long juvenile phase. The biological analysis for the regulation of flowering time provides insights into the reduction of juvenile phase and the acceleration of breeding in fruit trees. In Arabidopsis, LIKE HETEROCHROMATIN PROTEIN1 (LHP1) is involved in epigenetic silencing of the target genes such as flowering genes. We isolated and characterized twin apple LHP1 homolog genes, MdLHP1a and MdLHP1b. These genes may have been generated as a result of ancient genome duplication. Although the putative MdLHP1 proteins showed lower similarity to any other known plant LHP1 homologs, a chromo domain, a chromo shadow domain, and the nuclear localization signal motifs were highly conserved among them. RT-PCR analysis showed that MdLHP1a and MdLHP1b were expressed constantly in developing shoot apices of apple trees throughout the growing season. Constitutive expression of MdLHP1a or MdLHP1b could compensate for the pleiotropic phenotype of lhp1/tfl2 mutant, suggesting that apple LHP1 homolog genes are involved in the regulation of flowering time and whole-plant growth. Based on these results, LHP1 homolog genes might have rapidly evolved among plant species, but the protein functions were conserved, at least between Arabidopsis and apple. The phenology of citricola scale, Coccus pseudomagnoliarum (Kuwana), and its associated parasitoid complex were studied on citrus in the San Joaquin Valley of central California over the period April 1995–March 1997. A total of 10,237 parasitoid specimens of 10 species were collected. Two of these species, Marietta mexicana (Howard) and Encyrtus lecaniorum (Mayr), each recovered from individually isolated scales, represent new parasitoid records for citricola scale. A third species, Encarsia citrinus citrinus (Craw), may represent a new parasitoid record, but this requires further confirmation because a single (male) specimen was recovered from individually isolated scales. The three most dominant parasitoid species, Coccophagus lycimnia (Walker), Metaphycus helvolus (Compere), and Metaphycus luteolus (Timberlake), accounted for the majority (>97%) of the specimens recovered. In contrast to the situation on citrus in southern California, where citricola scale is under effective biological control and is very rarely seen, citricola scale on citrus in the San Joaquin Valley is reemerging as a major pest, especially in groves employing integrated pest management with minimal use of broad-spectrum insecticides. Possible reasons uncovered in this study for the lack of effective biological control of citricola scale in the San Joaquin Valley include: (i) reduced presence of Metaphycus spp. because of hyperparasitism by the heteronomous hyperparasitoid C. lycimnia; (ii) absence of alternate hosts for those species of Metaphycus present; and (iii) absence of hosts of suitable size for Metaphycus at critical times of the year. Recommendations for improving the level of biological control in the San Joaquin Valley are discussed.  相似文献
