The capacity for C4 photosynthesis in Panicum milioides, a specieshaving reduced levels of photorespiration, was investigatedby examining the activity of certain key enzymes of the C4 pathwayand by pulse-chase experiments with 14CO2. The ATP$P1 dependentactivity of pyruvate,P1 dikinase in the species was extremelylow (0.14–0.18 µmol mg chlorophyll–1 min–1).Low activity of the enzyme was also found in Panicum decipiensand Panicum hians (related species with reduced photorespiration)and in Panicum laxum (a C3 species). The antibody to pyruvate,P1dikinase caused about 70% inhibition of the ATP$P1 dependentactivity of the enzyme in P. milioides. The activity of NAD-malicenzyme and NADP-malic enzyme in P. milioides was equally low(approximately 0.1–0.2 µmol mg chlorophyll–1min–1) and similar to the activity in P. decipiens, P.hians and P. laxum. Photosynthetic pulse-chase experiments underatmospheric conditions showed a typical C3-like pattern of carbonassimilation including the labelling of glycine and serine asexpected during photorespiration. During the pulse with 14CO2only about 1% of the labelled products appeared in malate and2–3% in aspartate. During a chase in atmospheric levelsof CO2 for up to 6 min there was a slight increase in labellingin the C4 acids. The amount of label in carbon 4 of aspartatedid not change during the chase, indicating little or no turnoverof the C4 acid via decarboxylation. The results indicate thatunder atmospheric conditions P. milioides assimilates carbondirectly through the C3 pathway. Photorespiration as indicatedby the CO2 compensation point may be repressed in the speciesby a more efficient recycling of photorespired CO2. (Received June 8, 1982; Accepted July 22, 1982)  相似文献

A study was made of the incorporation of 14C by intact leavesof Coffea arabica (cultivars Mundo Novo, Catuai, 1130–13,and H 6586–2) and Coffea canephora (cultivar Guarini)supplied with gas mixtures containing 14CO2 under controlledconditions. Samples of the leaves were combusted and the 14Cin the CO2 produced measured using a liquid scintillation counter.The results were used to estimate photosynthetic rates. Theeffects of changing the partial pressures of O2 and CO2 on thephotosynthetic rate were studied and estimates made of the CO2compensation point and photorespiration. The data obtained show differences between the mean net photosyntheticrates of the C. arabica cultivars (6·14 mg CO2 dm–2h–1) and the mean rate for the C. canephora cultivar (3·96mg CO2 dm–2 h–1). The cultivar of the latter speciesphotorespired more rapidly than the cultivar Catuai of C. arabica.Rates of photosynthesis in coffee measured using the 14CO2 methodwere similar to rates obtained by others using an infrared gasanalyser. The 14CO2 method proved to be reliable for photosyntheticmeasurements and the apparatus is suitable for use in fieldconditions.  相似文献

Time-courses of 14CO2-fixation and of enzyme activities involvedin photorespiration and photosynthesis were determined duringthe life span of cotyledons from sunflower seedlings (Helianthusannuus L.). Glycolate formation in vivo was estimated from theresults of combined labelling and inhibitor experiments. NADPH-glyceraldehyde-3-phosphatedehydrogenase, NADPH-glyoxylate reductase and chlorophyll werewell correlated with the time-course of 14CO2-fixation (photosynthesis).There was, however, a considerable discrepancy between the developmentalsequence of photosynthesis and that of both ribulose-l,5-bisphosphatecarboxylase and glycolate oxidase. Furthermore, time-coursesof glycolate oxidase activity in vitro and of glycolate formationin vivo differed significantly. Therefore, the use of glycolateoxidase as a marker for the activity of photorespiration ingreening sunflower cotyledons may be questionable. Results from14CO2-labelling experiments with cotyledons treated with theglycolate oxidase inhibitor 2-hydroxy butynoic acid suggestthat glycolate formation relative to CO2-fixation is reducedin senescent cotyledons. Photosynthetic rates of outdoor-grown soybean (Glycine max L.Merr. cv. Bragg) canopies increased with increasing CO2 concentrationduring growth, before and after canopy closure (complete lightinterception), when measured over a wide range of solar irradiancevalues. Total canopy leaf area was greater as the CO2 concentrationduring growth was increased from 160 to 990 mm3 dm–3.Photosynthetic rates of canopies grown at 330 and 660 mm3 CO2dm–3 were similar when measured at the same CO2 concentrationsand high irradiance. There was no difference in ribulose bisphosphatecarboxylase/oxygenase (rubisco) activity or ribulose 1,5-bisphosphate(RuBP) concentration between plants grown at the two CO2 concentrations.However, photosynthetic rates averaged 87% greater for the canopiesgrown and measured at 660 mm3 CO2 dm–3. A 10°C differencein air temperature during growth resulted in only a 4°Cleaf temperature difference, which was insufficient to changethe photosynthetic rate or rubisco activity in canopies grownand measured at either 330 or 660 mm3 CO2 dm–3. RuBP concentrationsdecreased as air temperature during growth was increased atboth CO2 concentrations. These data indicate that the increasedphotosynthetic rates of soybean canopies at elevated CO2 aredue to several factors, including: more rapid development ofthe leaf area index; a reduction in substrate CO2 limitation;and no downward acclimation in photosynthetic capacity, as occurin some other species. Salsola arbusculiformis is identified as a C3–C4intermediatespecies based on anatomical, biochemical and physiological characteristics.This is the first report of a naturally occurring intermediatespecies in the Chenopodiaceae, the family with the largest numberof C4species amongst the dicots. In the genus Salsola, mostspecies have Salsoloid anatomy with Kranz type bundle sheathcells and C4photosynthesis, while a few species have Sympegmoidanatomy and were found to have non-Kranz type bundle sheathcells and C3photosynthesis. In the cylindrical leaves of C4Salsolawith Salsoloid type anatomy, there is a continuous layer ofdistinct, chlorenchymatous Kranz type bundle sheath cells surroundedby a single layer of mesophyll cells; whereas species with Sympegmoidtype anatomy have an indistinct bundle sheath with few chloroplastsand multiple layers of chlorenchymatous mesophyll cells. However,S. arbusculiformis has intermediate anatomical features. Whileit has two-to-three layers of mesophyll cells, characteristicof Sympegmoid anatomy, it has distinctive, Kranz-like bundlesheath cells with numerous chloroplasts and mitochondria. Measurementsof its CO2compensation point and CO2response of photosynthesisshow S. arbusculiformis functions as an intermediate specieswith reduced levels of photorespiration. The primary means ofreducing photorespiration is suggested to be by refixing photorespiredCO2in bundle sheath cells, since analysis of photosyntheticenzymes (activity and immunolocalization) and14CO2labellingof initial fixation products suggests minimal operation of aC4cycle. Photorespiration rates under air-equilibrated conditions (0.04%CO2 and 21% O2) were measured in Chlamydomonas reinhardtii wild-type2137, a phosphoglycolate-phosphatase-deficient (pgp1) mutantand a suppressor double mutant (7FR2N) derived from the pgp1mutant. In both cells grown under 5% CO2 and adapted air for24 h in the suppressor double mutant, the maximal rate of photorespiration(phosphoglycolate synthesis) was only about half of that ineither the wild type or the pgp1 mutant (18-7F) cells. In theprogeny, the reduced rate of photorespiration was accompaniedby increased photosynthetic affinity for inorganic carbon andthe capacity for growth under air whether accompanied by thepgp1 background or not. Tetrad analyses suggested that thesethree characteristics all resulted from a nuclear single-genemutation at a site unlinked to the pgp1 mutation. The decreasein photorespiration was, however, not due to an increase inthe CO2/O2 relative specificity of ribulose-1,5-bisphosphatecarboxylase/oxygenase of 7FR2N or of any other suppressor doublemutants tested. Inoculating Proctor barley leaves with Erysiphe graminis decreasedrates of photosynthesis, after an initial lag period, and increasedrespiration. Increasing the area inoculated progressively decreased ratesof photosynthesis, but the effects cannot be attributed to asimple loss of leaf area. When less than 30 per cent of a leafwas inoculated, decreases were equivalent to area losses greaterthan those inoculated; when more than 30 per cent was inoculatedthe photo-synthetic losses were equivalent to area losses lessthan those inoculated. Although the relative effects of E. graminis on photosynthesisand respiration were of the same order, the absolute effectson photosynthesis were greater than those on respiration. Inoculating30 per cent of a leaf decreased photosynthesis by 5–6mg CO2/dm2/hr from 12.9 in the uninoculated controls to 7.3.Respiration increased by 0.6 mg CO2/dm/hr, from 1.7 to 2.3-  相似文献

The rates of photosynthetic 14CO2 fixation by Chlorella vulgarisllh, grown under high CO2, were determined between 4 to 37°Cwith air containing from 300 to 13,000 ppm 14CO2. When the CO2level was increased, both the rate of photosynthesis and theoptimum temperature for maximum photosynthesis increased. Themaximum photosynthetic rate was reached at 12°C with 300ppm l4CO2. Among the photosynthetic products fromed at 300 ppm 14CO2, glycolatedecreased greatly when the temperature was raised from 20 to30°C. At 3,000 ppm 14CO2 an insignificant amount of glycolatewas formed at all temperatures, whereas 14C-incorporation intothe insoluble fraction, sucrose, and the lipid fraction wassignificantly higher than at 300 ppm 14CO2. The 14C in sucrosewas greatly increased and the radioactivity in the insolublefraction decreased when the temperature was raised from 28 to36°C. (Received April 8, 1980; )  相似文献

The effects of elevated carbon dioxide (CO2 and ozone (O3) onsoybean (Glycine max (L.) Merr.] photosynthesis and photorespiration-relatedparameters were determined periodically during the growing seasonby measurements of gas exchange, photorespiratory enzyme activitiesand amino acid levels. Plants were treated in open-top fieldchambers from emergence to harvest maturity with seasonal meanconcentrations of either 364 or 726 µmol mol–1 CO2in combination with either 19 or 73 nmol mol–1 O3 (12h daily averages). On average at growth CO2 concentrations,net photosynthesis (A) increased 56% and photorespiration decreased36% in terminal mainstem leaves with CO2 enrichment. Net photosynthesisand photorespiration were suppressed 30% and 41%, respectively,by elevated O3 during late reproductive growth in the ambientCO2 treatment, but not in the elevated CO2 treatment. The ratioof photorespiration to A at growth CO2 was decreased 61% byelevated CO2 There was no statistically significant effect ofelevated O3 on the ratio of photorespiration to A. Activitiesof glycolate oxidase, hydroxypyruvate reductase and catalasewere decreased 10–25% by elevated CO2 and by 46–66%by elevated O3 at late reproductive growth. The treatments hadno significant effect on total amino acid or glycine levels,although serine concentration was lower in the elevated CO2and O3 treatments at several sampling dates. The inhibitoryeffects of elevated O3 on photorespiration-related parameterswere generally commensurate with the O3-induced decline in A.The results suggest that elevated CO2 could promote productivityboth through increased photoassimilation and suppressed photorespiration. 14CO2 assimilation was studied with diploid, tetraploid, hexaploidspecies of the genera Triticum and their wild relatives Aegilops.Attached mature leaves of 3–4 weekold plants were allowedto undergo photosynthesis under air at ambient temperature.The pattern of distribution of 14C was notably similar in Triticumand Aegilops species whatever the level of ploidy. Sucrose wasthe sink for photosynthetic carbon. 14C for sucrose synthesis was supplied either through the glycolatepathway by glycolate, the product of the photorespiration orby the Calvin cycle intermediates exported into the cytoplasm.Depending on the species, the glycolate pathway provided 40to 75%of the sucrose 14C. The higher labeling of sucrose was associated with the greaterparticipation of the glycolate pathway in the wild diploid (DD)A. squarrosa and in the cultivated hexaploid (AABBDD) T. aestivum.The results suggest that the expression of the male D genomeis dominant over the female AB genome in T. aestivum. In T. aestivum under ambient conditions lowering (low temperature)or hindering (1% O2 ) photorespiration, sucrose labeling decreased,but serine and glycine labeling was favoured. Pascopyrum smithii (C3) andBouteloua gracilis (C4) are importantforage grasses native to the Colorado shortgrass steppe. Thisstudy investigated photosynthetic responses of these grassesto long-term CO2enrichment and temperature in relation to leafnonstructural carbohydrate (TNC) and [N]. Glasshouse-grown seedlingswere transferred to growth chambers and grown for 49 d at twoCO2concentrations (380 and 750 µmol mol-1) at 20 and 35°C, and two additional temperatures (25 and 30 °C) at750 µmol mol-1CO2. Leaf CO2exchange rate (CER) was measuredat a plant's respective growth temperature and at two CO2concentrationsof approx. 380 and 700 µmol mol-1. Long-term CO2enrichmentstimulated CER in both species, although the response was greaterin the C3,P. smithii . Doubling the [CO2] from 380 to 750 µmolmol-1stimulated CER ofP. smithii slightly more in plants grownand measured at 30 °C compared to plants grown at 20, 25or 35 °C. CO2-enriched plants sometimes exhibited lowerCER when compared to ambient-grown controls measured at thesame [CO2], indicating photosynthetic acclimation to CO2growthregime. InP. smithii , such reductions in CER were associatedwith increases in TNC and specific leaf mass, reductions inleaf [N] and, in one instance, a reduction in leaf conductancecompared to controls. InB. gracilis , photosynthetic acclimationwas observed more often, but significant changes in leaf metabolitelevels from growth at different [CO2] were generally less evident.Temperatures considered optimal for growth (C3: 20 °C; C4:35 °C) sometimes led to CO2-induced accumulations of TNCin both species, with starch accumulating in the leaves of bothspecies, and fructans accumulating only inP. smithii. Photosynthesisof both species is likely to be enhanced in future CO2-enrichedand warmer environments, although responses will sometimes beattenuated by acclimation. Maize seedlings were grown in pots either with or without preconditionedseeds of the parasitic weed, Striga hermonthica. After between4 and 8 weeks, net photosynthesis in the leaves of maize plantsinfected with Striga decreased compared to leaves of uninfectedcontrol plants. The activities of four enzymes of photosyntheticmetabolism were, however, little affected by infection. A pulse-chaseexperiment using 14CO2 showed that C4 acids were the main earlyproducts of assimilation even when the rate of photosynthesiswas much decreased by infection, but more radio-activity appearedin glycine and serine than in leaves of healthy maize plants.Leaves of infected maize required longer to reach a steady rateof photosynthesis upon enclosure in a leaf chamber than leavesof uninfected plants after similar treatment. Electron microscopy of transverse sections of the leaves ofinfected maize indicated that the cell walls in the bundle sheathand vascular tissue were less robust than in leaves of healthyplants. The results suggest that infection with Striga causesan increase in the permeability of cell walls in the bundlesheath, leakage of CO2 from the bundle sheath cells and decreasedeffectiveness of C4 photosynthesis in host leaves. Light and electron microscopic observations of the leaf tissueof Panicum milioides showed that the bundle sheath cells containeda substantial number of chloroplasts and other organelles. Theradial arrangement of chlorenchymatous bundle sheath cells,designated as Kranz leaf anatomy, has been considered to bespecific to C4 plants. However, photosynthetic 14CO2 fixationand 14CO2 pulse-and-chase experiments revealed that the reductivepentosephosphate pathway was the main route operating in leavesof P. milioides. The interveinal distance of the leaves wasintermediate between C3and C4Gramineae species. Etiolated Avena sativa L. coleoptile sections were used to determinethe influence of C2H4 on in vivo and in vitro rates of CO2 fixation,and to measure the influence of various permutations of C2H4,CO2, and malate on growth. Whereas 1 mM malate or 320 µI-1 CO2 stimulated growth by approximately 100 per cent, inhibitionof growth by 10-8 µ I-1 C2H4 was substantial only in thepresence of malate or CO2 The increase in growth rate in responseto these two agents was eliminated by the simultaneous applicationof C2H4. The in vivo rate of dark [14C]bicarbonate fixationand in vitro enzymic assays of fixation were not measurablyinhibited by C2H4. These results are discussed in the lightof evidence which indicates that CO2-stimulated growth is mediatedby dark fixation. The data do not support the view that C2H4inhibition of growth results from an inhibition of fixation,but suggests that C2H4 may inhibit some step in the processby which malate stimulates growth.  相似文献

Changes in chemical constituents and respiratory metabolismof a long-day duck-weed, Lemna gibba G3 exposed to continuousillumination after short-day cultivation were investigated.The dry weight to fresh weight ratio was constant during thefirst 72 hr of continuous illumination. pH of the crude extractwas constant at 6.6, but pH of the culture medium was raisedwith the Lemna growth. Titratable acidity decreased after about44 hr, whereas malic acid content increased in 18 hr. Therewere no significant changes in total reducing sugar and pentose.Total protein content and lipid showed rhythmical changes withcycles of 48 hr. O2-Uptake gave a damped oscillation with cycles of 24 hr. Itwas low in the first half day and high in the second half. 14CO2-Outputfrom glucose-l-14C showed a similar damped oscillation. 14CO2-Outputfrom glucose-2-14C or glucose-6-14C was almost constant. TheC6/C2 ratio, then, showed damped oscillation in the reverseway to O2- uptake between 0.3–0.5, and the C8/C2 ratiowas constant at 0.9. Accordingly, the diurnal rhythm of O2-uptakewas thought to be brought about by variation in activity ofthe pentose-phosphate pathway. Reproduction of glucose-6-phosphateby the pentose-phosphate pathway was presumably limited in amount. Glyceraldehyde-3-phosphate dehydrogenase activity varied diurnally.The activities of NADP-linked and NAD-linked enzymes increasedand decreased, respectively, in the first half day. Variationsin these enzymatic activities are discussed in correlation withrhythmical changes in O2-uptake and in the C6/C1 ratio. Acidphosphatase activity also followed a diurnal variation. No activitiesof alcohol and formic dehydrogenases were found. Exposure of mature, nodulated plants of white clover (Trifoliumrepens) cv. Blanca to 330 mg dm–3 NO3-N for 8 d causednitrogenase activity per plant to decrease by 80%. Total nodulatedroot respiration was not significantly affected but analysisof its components showed an 81% decrease in nitrogenase-linkedrespiration and a 340% increase in growth and maintenance respiration.Carbon costs of nitrogenase activity (mol CO2 respired per molC2H4 produced) increased by 45% over the exposure period. Sucrosecontent of the nodules decreased, but the pattern of decreasedid not correlate with that of nitrogenase activity. The oxygendiffusion resistance of the nodules was increased by a factorof five. Characterization of this resistance increase suggestsan abnormal modification of the diffusion barrier and it isconcluded that alteration in the oxygen supply to the bacteroidsis involved in the effect of nitrate on nitrogenase activity. The effects of growth at elevated CO2 on the response to hightemperatures in terms of carbon assimilation (net photosynthesis,stomatal conductance, amount and activity of Rubisco, and concentrationsof total soluble sugars and starch) and of photochemistry (forexample, the efficiency of excitation energy captured by openphotosystem II reaction centres) were studied in cork oak (Quercussuber L.). Plants grown in elevated CO2 (700 ppm) showed a down-regulationof photosynthesis and had lower amounts and activity of Rubiscothan plants grown at ambient CO2 (350 ppm), after 14 monthsin the greenhouse. At that time plants were subjected to a heat-shocktreatment (4 h at 45C in a chamber with 80% relative humidityand 800–1000 mol m–2 s–1 photon flux density).Growth in a CO2-enriched atmosphere seems to protect cork oakleaves from the short-term effects of high temperature. ElevatedCO2 plants had positive net carbon uptake rates during the heatshock treatment whereas plants grown at ambient CO2 showed negativerates. Moreover, recovery was faster in high CO2-grown plantswhich, after 30 min at 25C, exhibited higher net carbon uptakerates and lower decreases in photosynthetic capacity (Amax aswell as in the efficiency of excitation energy captured by openphotosystem II reaction centres (FvJFm than plants grown atambient CO2. Net photosynthetic rates per unit ground area for plant standsof Solanum melongena L. var. esculentum (aubergine) and Amaranthuscaudatus L. var. edulis (grain amaranth) were measured over10 min intervals in an airtight, glass, controlled-environmentcabinet for a range of light flux densities provided by thediurnal variation in daylight. Light response curves for photosynthesisof stands, grown at ambient CO2 concentration, were definedat 400, 800 and 1200 vpm CO2. Light compensation points for these stands were around 20-30J m-2 s-1 and decreased slightly at higher CO2 concentrations.For aubergine, a C3 species, the short-term effects of CO2 enrichmentwere to increase the initial slope as well as the asymptoteof the light response curve, reducing light saturation at moderateto high light flux densities; but for amaranthus, a C4 species,saturation was less apparent and CO2 enrichment scarcely increasedphotosynthesis except at light flux densities above 150 J m-2s-1. The canopies intercepted 93-98% of incident light. The efficiencyof utilization of intercepted light in photosynthesis (µgCO2 J-1) increased from zero at the light compensation pointto a maximum at an optimum light flux density of about 100 Jm-2 s-1 (the optimum rose a little with CO2 enrichment) anddecreased slightly with further increase in light. Maximum utilizationefficiencies at 400 vpm CO2 were 8-9 µg CO2 J-1. Enrichmentto 1200 vpm did not affect the peak utilization efficiency ofthe C4 amaranthus, but increased that aubergine to 12·2µg CO2 J-1 (equivalent to some 14% when using the heatof combustion of plant dry matter to convert to the dimensionlessform). The 14C-metabolite distribution pattern following 14C2H4 metabolismin intact pea seedlings (Pisum sativum L.) was determined undervarious conditions. After a 24 hr exposure to 14C2H4, the majorityof 14C-metabolites were water-soluble (60–70%) with lesseramounts in the protein (10–15%), lipid (1%), and insoluble(1–2%) fractions. Ion exchange chromatography of the water-solublecomponents into basic, neutral, and acidic fractions revealeda 50 : 40 : 10 distribution, respectively. Chromatography ofthe neutral fraction revealed two regions of radioactivity (Rf=0.38)and 0.63 which did not cochromatograph with twenty-two knownsugars or neutral metabolites. Chromatograms of the basic fractioncontained 3 regions of radioactivity. Similar distribution patternswere noted when 14C2H4 exposure was followed by a 6 hr air chaseor when 5% CO2, an antagonist of ethylene action, was presentduring the exposure. Marked differences in the 14C-metabolite distribution patternswere obtained when 14CO2 was substituted for 14C2H4.
