The diversity of mycorrhizal fungi associated with an introduced weed-like South African orchid (Disa bracteata) and a disturbance-intolerant, widespread, native West Australian orchid (Pyrorchis nigricans) were compared by molecular identification of the fungi isolated from single pelotons. Molecular identification revealed both orchids were associated with fungi from diverse groups in the Rhizoctonia complex with worldwide distribution. Symbiotic germination assays confirmed the majority of fungi isolated from pelotons were mycorrhizal and a factorial experiment uncovered complex webs of compatibility between six terrestrial orchids and 12 fungi from Australia and South Africa. Two weed-like (disturbance-tolerant rapidly spreading) orchids — D. bracteata and the indigenous Australian Microtis media, had the broadest webs of mycorrhizal fungi. In contrast, other native orchids had relatively small webs of fungi (Diuris magnifica and Thelymitra crinita), or germinated exclusively with their own fungus (Caladenia falcata and Pterostylis sanguinea). Orchids, such as D. bracteata and M. media, which form relationships with diverse webs of fungi, had apparent specificity that decreased with time, as some fungi had brief encounters with orchids that supported protocorm formation but not subsequent seedling growth. The interactions between orchid mycorrhizal fungi and their hosts are discussed.  相似文献

Though ≥22 species of Primates consume fungi, most do so at low rates, comprising <5% of their feeding time. Goeldi's monkeys (Callimico goeldii), spend up to 29% of their feeding time year-round consuming fungal sporocarps, the fruiting bodies of fungi. We provide comprehensive data on the nutritional characteristics of 4 species of fungi consumed by Callimico goeldii (Ascopolyporus polyporoides, Ascopolyporus polychrous, Auricularia auricula, and Auricularia delicata). The composition of the fungi is similar to that of other fungi: predominantly fiber (66.2–83.0% dry matter) with small amounts of sugar (2.0–5.6% dry matter) and crude fat (0.9–1.6% dry matter). Though the crude protein content is substantial (5.5–13.4% dry matter), much of the nitrogen in the fungi is not likely to be available to Callimico goeldii because it is associated with indigestible food components or is in nonprotein form. The mineral content of the fungi are within the normal range for fungi generally and the calcium-to-phosphorus ratio is low (0.07–0.25). Fungi appear to be a low-quality food resource for Callimico goeldii and may contribute to their relatively large home ranges and low population density compared to other Callitrichinae. Research on the ability of Callimico goeldii to digest fungi is needed to understand fully the nutritional value of fungi to them. We discuss adaptations Callimico goeldii may have for improving their ability to obtain nutrients from fungi and potential ecological correlates of mycophagy.  相似文献

The effect of three basidiomycetous fungi, Meira geulakonigii, M. argovae and Acaromyces ingoldii on strains of seven phytopathogenic fungal species was assayed in the laboratory. The phytopathogens were placed on dialysis membranes covering media in which the basidiomycetous fungi were formerly cultured. A. ingoldii inhibited the growth of all phytopathogens tested, whereas Meira spp. suppressed their growth to a lesser extent. Upon being returned to extract-free media, all phytopathogens resumed their growth, suggesting a fungistatic effect of the basidiomycetous fungi. These fungi inhibited the growth of Sclerotina sclerotiorum on tomato leaves for a few days and Meira spp. hindered the infection of oranges by Penicillium digitatum, whereas A. ingoldii was less effective. Results of preliminary biochemical assays suggest that neither competition for hydrocarbons nor secreted proteases are responsible for the inhibitory activity of the basidiomycetous fungi, which was probably due to the secretion of unidentified substances by these fungi.  相似文献

About 174 endophytic fungi were isolated from the pharmaceutical plant, Camptotheca acuminata. Of the 18 taxa obtained, non-sporulating fungi (48.9%), Alternaria (12.6%), Phomopsis (6.9%), Sporidesmium (6.3%), Paecilomyces (4.6%) and Fusarium (4.6%) were dominant. ITS rDNA assay indicated that most of the non-sporulating fungi belonged to the Pyrenomycetes and Loculoascomycetes ascomycetes or their anamorph Coelomycetes. The results of the bioactivity test showed that 27.6% of the endophytic fungi displayed inhibition against more than one indicator microorganism. 4.0% and 2.3% of the endophytic fungi showed cytotoxicity and protease inhibition, respectively. The endophytic fungi with bioactivities were distributed in more than 12 taxa including non-sporulating fungi, which are reliable sources for bioactive agents.  相似文献

Most ciids (Ciidae) are strict fungivores specialized on fruit bodies of wood‐rotting fungi. The Ciidae includes both specialist and generalist species. Recent evidence suggests that ciids locate and discriminate their potential hosts based mainly on fungal odours. In this study, we investigated the field distribution of ciids in a local woodland near Bath, U.K. We also evaluated experimentally the behavioural responses of ciids to host and non‐host fungi in an olfactometer, and explored potential differences in putative aroma compounds in host fungi. Our field data showed that Octotemnus glabriculus, Cis boleti and Cis nitidus have preference for one host species, whereas C. bilamellatus has no preference. The experimental evidence was in accordance with the field observations. The specialists O. glabriculus, C. boleti and C. nitidus were specifically attracted to odour compounds of their preferred host fungi: Coriolus versicolor for the first two beetles and Ganoderma adspersum for the latter one. By contrast, the generalist C. bilamellatus was attracted to odours from C. versicolor, G. adspersum and Piptoporus betulinus. The evidence from this and other published studies suggest that ciids depend mainly on fruit body odours for location and discrimination of their host fungi. In accordance with this, we observed diffierential behavioural responses of ciids to aroma compounds of host and non‐host species.  相似文献

Patterns of phenoloxidase activity can be used to characterize fungi of different life styles, and changes in phenoloxidase synthesis were suspected to play a role in the interaction between ectomycorrhizal and two species of Trichoderma. Confrontation between the ectomycorrhizal fungi Amanita muscaria and Laccaria laccata with species of Trichoderma resulted in induction of laccase synthesis, and the laccase enzyme was bound to mycelia of ectomycorrhizal fungi. Tyrosinase release was noted only during interaction of L. laccata strains with Trichoderma harzianum and T. virens. Ectomycorrhizal fungi, especially strains of Suillus bovinus and S. luteus, inhibited growth of Trichoderma species and caused morphological changes in its colonies in the zone of interaction. In contrast, hyphal changes occurred less often in the ectomycorrhizal fungi tested. Species of Suillus are suggested to present a different mechanism in their interaction with other fungi than A. muscaria and L. laccata.  相似文献

We used Pisolithus tinctorius and Cenococcum geophilum to determine the copper (Cu) resistance of ectomycorrhizal (ECM) fungi and their potential for improving phytoremediation of Cu-contaminated soil by Chinese red pine (Pinus tabulaeformis). The results showed that nutrient accumulation in C. geophilum mycelium was significantly lower under higher Cu concentrations in the soil, which was not observed in P. tinctorius. Meanwhile, P. tinctorius exhibited greater Cu tolerance than C. geophilum. Inoculation with ECM fungi significantly improved the growth of pine shoots planted in polluted soil in pot experiments (p < 0.01). The total accumulated Cu in pine seedlings planted in Cu-contaminated soil increased by 72.8% and 113.3% when inoculated with P. tinctorius and C. geophilum, respectively, indicating that ECM fungi may help their host to phytoextract heavy metals. Furthermore, the majority of the total absorbed metals remained in the roots, confirming the ability of ECM fungi to promote heavy metal phytostabilization. There were no differences between the effects of the two fungi in helping the host stabilize and absorb Cu, even though they have different Cu tolerances. Inoculation with ECM fungi can benefit plant establishment in polluted environments and assist plants with phytoremediating heavy-metal-contaminated soils.  相似文献

Among Trichophyton spp. examined for urease production, T. rubrum was negative, whereas T. mentagrophytes appeared to be the most active species. Urease was not detected in cell-free culture fluids of the tested fungi. The endocellular urease of the test fungi was essentially constitutive. Moreover, addition of urea to the growth medium of these organisms markedly inhibited their mycelial biomass and ureolytic yield. Environmental factors showed variable effects on the test fungi and there was no correlation between mycelial growth and urease activity of these fungi.  相似文献

Diversity and heat-adaptation of endophytic fungi (EF) and rhizospheric fungi associated with plants growing in geothermal ecosystems, Southwest China, as well as their benefit in improving host plant thermotolerances were investigated. A total of 1589 culturable fungi belonging to 38 taxa were isolated, in which Curvularia, Acrophialophora, Penicillium, and Aspergillus were the dominant genera. The Shannon indices of EF and rhizospheric fungi ranged from 1.80 to 2.56 and 0.73 to 2.11, respectively. Phylogenetic analysis indicated that the EF have a close relationship with rhizospheric fungi. However, some fungi exhibited apparently species-specific habitat distribution patterns. Growth temperature tests indicated that 60.22% of the tested isolates were thermotolerant fungi and only 39.78% were mesophiles, and the number of heat-adapted fungi increased with increasing environmental temperatures. The strain G1-29, which was isolated from the roots of Hedyotis diffusa and identified as Curvularia crepinii, significantly improved host plant thermotolerance under laboratory conditions: the death rate of endophyte-infected plants was significantly lower than that of endophyte-free plants (t-test, p?=?.0158, df?=?4). Our results suggested that the EF and rhizospheric fungi associated with plants growing in geothermal ecosystems are diverse, and many of them have adapted to the high environmental temperatures. Some fungi have come to be the dominant endemic inhabitants of specific niches, and some played an important role in improving host plant thermotolerances.  相似文献

To examine the relative importance of leaf age and season on the occurrence of phyllosphere fungi, temporal patterns of epiphytic and endophytic phyllosphere fungi of giant dogwood (Swida controversa) were studied with reference to leaf emergence at first occurrence and in the middle of the growing season. A total of 15 and 44 species were isolated from the surface and interior of leaves, respectively. On the leaf surface, detection rate of fungi was consistently 100% and their frequencies increased during the growing season, whereas in the leaf interior, detection rate of fungi and their frequencies were low at leaf emergence and gradually increased during the growing season. Six epiphytic and two endophytic fungi were observed frequently. A white sterile mycelium was frequent only on the surface of newly emerged leaves in the first-order shoot in May. The other 7 species increased during the growing season. The frequencies of Phomopsis sp., Pestalotiopsis sp. 1, and Trichoderma viride were higher on the leaves of first-order shoots than those of higher-order shoots that emerged between July and September, suggesting the possible effects of leaf age on their occurrence. On the other hand, the frequencies of Colletotrichum gloeosporioides, Clonostachys rosea, Cladosporium cladosporioides, and Phoma sp. 1 were not different between the first- and higher-order shoots, suggesting the negligible effect of leaf age. The influence of phenological patterns of leaf emergence of deciduous trees on the diversity and composition of assemblages of phyllosphere fungi is discussed.  相似文献

对我国古老特有植物青檀叶片进行内生和附生真菌的研究,以了解青檀叶片内生和附生真菌的组成特点和探讨内生和附生真菌菌群之间的可能联系,为研究真菌资源多样性、植物附生和内生真菌的相互演化关系及真菌与宿主植物协同进化等提供有益参考资料。研究结果表明,从健康的青檀叶片获得可培养内生真菌839株,附生真菌1857株,共计2696株,鉴定其分属于4目,5科,43属。在目的分类水平上,内生和附生真菌均以丛梗孢目Moniliales为优势菌群,分别占90.23%和92.51%;在科的水平上,内生真菌以暗梗孢科Dematiaceae和丛梗孢科Moniliaceae为优势菌群,分别占47.56%和42.67%,附生真菌以丛梗孢科Moniliaceae和暗梗孢科Dematiaceae为优势菌群,分别占67.04%和25.47%;在属的水平上,内生真菌以黑团孢属Periconia和青霉属Penicillium为优势菌群,分别占31.47%和10.73%,附生真菌以小球霉属Glomerularia、膝葡孢属Gonatobotrys和青霉属Penicillium为优势菌群,分别占20.03%、13.95%和12.22%。青檀叶片内生真菌和附生真菌均存在的菌群数量达到23个属,占53.49%。内生真菌特有的属有6个,共分离19株,占0.70%,附生真菌特有的属有14个,共分离120株,占4.45%。内生真菌的Shannon-Wiener index(H')多样性指数(2.44)和Margalef index(R)丰富度指数(2.88)分别小于附生真菌ShannonWiener index(H')多样性指数(2.57)和Margalef index(R)丰富度指数(3.32),但两者的Evenness index(E)均匀度指数几乎相等。青檀叶片内生和附生真菌菌群组成具有较高的相似性,相似性系数达0.70。通过Fisher's exact test分析表明青檀叶片内生和附生真菌菌群组成无明显差异(P=0.072)。  相似文献

【目的】研究药用植物芍药(Paeonia lactiflora Pall.)内生真菌的种群多样性,同时对其可能存在的聚酮合酶(Polyketide synthase,PKS)和非核糖体多肽合成酶(Non-ribosomal peptide synthetase,NRPS)基因多样性进行评估,预测芍药内生真菌产生活性次生代谢产物的潜力。【方法】采用组织分离法获得芍药根部内生真菌菌株,结合形态学特征和ITS序列分析,进行鉴定;利用兼并性引物对内生真菌中存在的聚酮合酶(PKS)基因和非核糖体多肽合成酶(NRPS)基因进行PCR扩增及序列测定分析,构建系统发育树,明确芍药内真菌PKS基因序列和NRPS基因序列的系统进化地位。【结果】从芍药组织块中共分离得到105株内生分离物,去重复后获得52株内生真菌,菌株ITS基因序列信息显示,52株芍药内生真菌隶属于7目、13科、15属,其中小球腔菌属(Leptosphaeria)、土赤壳属(Ilyonectria)和镰孢属(Fusarium)为优势种群;从52株内生真菌中筛选获得13株含PKS基因片段的菌株,8株含NRPS基因片段的菌株,部分菌株功能基因的氨基酸序列与Gen Bank中已知化合物的合成序列具有一定的同源性,预示芍药根部内生真菌具有合成丰富多样的次生代谢产物的潜力。【结论】药用植物芍药根部具有丰富的内生真菌资源,且具有产生活性次生代谢产物的潜力,值得进一步开发研究和应用。  相似文献

Seventeen species of nematophagous fungi were recorded, three species were endoparasitic and fourteen species were predacious fungi. Among the predacious fungi Arthrobotrys oligospora, Dactylaria brochopaga and Monacrosporium eudermatum were very frequent, whereas others were recorded at lower frequency. Twelve species of nematophagous fungi from compost as well as cow dung manure, 15 species from leaf litter and only eight species from agricultural soils were recorded. In general, substrate colonization by nematophagous fungi was higher in leaf litter, compost and cow dung manure. The agricultural soil amended with FYM (farm yard manure) recorded nine species of nematophagous fungi while unamended soil recorded only seven species. Thirteen species of nematophagous fungi were recorded from soils under banyan tree. Of all these fungi unidentified net-forming fungus, M. eudermatum, A. cladodes, D. brochopaga, S. hadra, A. oligospora and A. dactyloides had higher percentage of soil colonization. In soil collected under pipal tree only eight species were recorded, of which A. oligospora, A. cladode and an unidentified fungus were more predominant as their percentage colonized in soil samples was higher. Few studies have examined root galls as a substratum for colonization of nematophagous fungi. Of all the root gall samples, okra root galls recorded maximum colonization by predacious fungi. Maximum percentage of root gall colonization was recorded for M. eudermatum followed by A. oligospora and M. ellipsosporum. M. eudermatum was also most predominant colonizer of balsam, brinjal and rice root galls.  相似文献

 Thirty-six isolates from 27 species of native ectomycorrhizal fungi collected in northern Spain were tested for ectomycorrhiza formation with Pseudotsuga menziesii seedlings in pure culture syntheses. Thirteen of those species were also tested for ectomycorrhiza formation with six other species of conifers (two native and four introduced) to compare their colonization potential. Twenty-three fungal isolates from 18 species formed ectomycorrhizas with Pseudotsuga menziesii. The colonization level of the root system varied markedly among the different fungal species. Eight fungi colonized over 50% of the short roots. Nine fungi did not form ectomycorrhizas even though some of them were collected in pure stands of Pseudotsuga menziesii. Laccaria laccata, Lyophyllum decastes, Pisolithus tinctorius, and Scleroderma citrinum formed abundant ectomycorrhizas on all the conifers tested. Lactarius deliciosus, Rhizopogon spp., and Suillus luteus showed the greatest host specificity. The success in the introduction of some exotic conifers for reforestation in northern Spain is discussed in relation to their compatibility with native ectomycorrhizal fungi. 本研究从柑橘抗病品种的健康植株不同组织中分离纯化和鉴定内生真菌,并测定其发酵产物对柑橘溃疡病菌的抑制活性,以明确柑橘抗病品种中内生真菌的组成及其产抗柑橘溃疡病菌活性代谢产物的潜力,为柑橘溃疡病抗菌剂的开发奠定基础。该研究通过组织培养法分离内生真菌,采用形态学和分子生物学方法对其进行鉴定; 基于前期的拮抗预试验结果,选取代表性菌株进行发酵培养,通过乙酸乙酯浸提、真空抽滤、旋转蒸发浓缩制备粗提物; 采用带毒平板涂布法测定不同菌株发酵产物乙酸乙酯提取物对柑橘溃疡病菌的抑制活性。结果表明:(1)共分离得到72株内生真菌,归为2门(Ascomycota、Basidiomycota)、14个属,其中优势属为刺盘孢属(Colletotrichum)、球座菌属(Guignardia)、链格孢属(Alternaria)和镰刀菌属(Fusarium)。(2)不同柑橘品种中内生真菌多样性指数为温州蜜柑(桂林)>沙糖桔(桂林)>沙糖桔(梧州)。(3)不同组织中内生真菌多样性变化因地理位置差异而有所不同,采自桂林的温州蜜柑和沙糖桔均为叶片中的内生真菌的多样性高于枝条,而采自梧州的沙糖桔为叶片中的多样性低于枝条,并且采自梧州的柑橘样品与采自桂林的柑橘样品中的内生真菌相似性低。(4)测定了30株内生真菌乙酸乙酯提取物对柑橘溃疡病菌的抑制活性,其中29株菌株表现出不同程度抑制活性。不同柑橘品种中的优势属的MIC介于0.312 5~10 mg·mL-1之间,特有属的MIC介于0.156~5 mg·mL-1,共有属镰刀菌属的MIC介于0.312 5~2.5 mg·mL-1之间。研究结果表明柑橘抗病品种中内生真菌具有丰富多样性,并且其发酵提取物普遍对柑橘溃疡病菌具有抑制作用。特有属抑菌活性总体优于优势属,共有属镰刀菌属在不同柑橘抗病品种中均具有显著抑菌效果。  相似文献

Endophytic fungi are ubiquitously distributed in orchids and have a great impact on the host plant. The diversity of endophytic fungi in the medicinal orchid Dendrobium loddigesii Rolfe was investigated and their bioactivities in microbe and plant growth were explored here. Endophytic fungi were identified by using morphological and molecular biological methods. Antimicrobial activity was determined by a standard disk assay. Activity in promoting plant growth was confirmed by root inoculation of endophytic fungi in seedling tray and pot experiments. Overall, 48 isolates were isolated from D. loddigesii and identified to belong to 18 genera, with Fusarium and Acremonium being the most dominant populations. A total of 17 isolates belonging to 9 genera were screened for their antimicrobial activity, and Fusarium spp., 8 of the 17 isolates, was also the dominant population. In the seedling tray experiment, two isolates, one of Fusarium named DL26 and the other of Pyrenochaeta named DL351, were shown to enhance plant growth in alder bark–humus medium, and the latter displayed weak activity against Bacillus subtilis (As 1.308) and Aspergillus fumigatus (As 3.2910). In the pot experiment, after inoculation of DL26 and DL351, five out of seven media were fit for plant-endophyte symbionts. Medium #1 of red brick fragments and sphagna was optimal in accelerating plant growth. In conclusion, a great diversity of endophytic fungi in D. loddigesii was first confirmed in a considerable proportion of antimicrobial isolates. Furthermore, two endophytes exhibited the ability to enhance plant growth although their activities were influenced by the growth media.  相似文献

Ectomycorrhizal (ECM) fungi contribute significantly to ecosystem respiration, but little research has addressed the effect of temperature on ECM fungal respiration. Some plants have the ability to acclimate to temperature such that long‐term exposure to warmer conditions slows respiration at a given measurement temperature and long‐term exposure to cooler conditions increases respiration at a given measurement temperature. We examined acclimation to temperature and temperature sensitivity (Q10) of respiration by ECM fungi by incubating them for a week at one of three temperatures and measuring respiration over a range of temperatures. Among the 12 ECM fungal isolates that were tested, Suillus intermedius, Cenococcum geophilum, and Lactarius cf. pubescens exhibited significant acclimation to temperature, exhibiting an average reduction in respiration of 20–45% when incubated at 23 °C compared with when incubated at 11 or 17 °C. The isolates differed significantly in their Q10 values, which ranged from 1.67 to 2.56. We also found that half of the isolates significantly increased Q10 with an increase in incubator temperature by an average of 15%. We conclude that substantial variation exists among ECM fungal isolates in their ability to acclimate to temperature and in their sensitivity to temperature. As soil temperatures increase, ECM fungi that acclimate may require less carbon from their host plants than fungi that do not acclimate. The ability of some ECM fungi to acclimate may partially ameliorate the anticipated positive feedback between soil respiration and temperature.  相似文献

The majority of chlorophyllous orchids form mycorrhizal associations with so‐called rhizoctonia fungi, a phylogenetically heterogeneous assemblage of predominantly saprotrophic fungi in Ceratobasidiaceae, Tulasnellaceae, and Serendipitaceae. It is still a matter of debate whether adult orchids mainly associated with rhizoctonia species are partially mycoheterotrophic. Here, we investigated the nutritional modes of green and albino variants of Goodyera velutina, an orchid species considered to be mainly associated with Ceratobasidium spp., by measuring their 13C and 15N abundances, and by molecular barcoding of their mycorrhizal fungi. Molecular analysis revealed that both green and albino variants of G. velutina harbored a similar range of mycobionts, mainly saprotrophic Ceratobasidium spp., Tulasnella spp., and ectomycorrhizal Russula spp. In addition, stable isotope analysis revealed that albino variants were significantly enriched in 13C but not so greatly in 15N, suggesting that saprotrophic Ceratobasidium spp. and Tulasnella spp. are their main carbon source. However, in green variants, 13C levels were depleted and those of 15N were indistinguishable from the co‐occurring autotrophic plants. Therefore, we concluded that the albino G. velutina variants are fully mycoheterotrophic plants whose C derives mainly from saprotrophic rhizoctonia, while the green G. velutina variants are mainly autotrophic plants, at least at our study site, in spite of their additional associations with ectomycorrhizal fungi. This is the first report demonstrating that adult nonphotosynthetic albino variants can obtain their nutrition mainly from nonectomycorrhizal rhizoctonia.  相似文献

Five different species of known ecto-mycorrhizal fungi: Cenococcum geophilum, Amanita muscaria, Tricholoma aurantium, Rhizopogon luteolus and Rhizopogon roseolus were studied for their ability to metabolize the major components of plant cell walls. All strains were able to decompose 14C-labelled plant lignin, 14C-lignocellulose and 14C-DHP-lignin at a rate which was lower than the one observed for the known white rot fungi Heterobasidion annosum and Sporotrichum pulverulentum. Also 14C-(U)-holocellulose was relatively less degradable for the mycorrhizal fungi than for the white rotters. On the other hand, aromatic monomers like 14C-vanillic acid were decomposed to a much higher extent by two species of mycorrhizal fungi compared to the activity observed for Heterobasidion annosum. The results of the experiments reveal that these stains of mycorrhizal fungi are well able to utilize the major components of plant material and thus can contribute to litter decomposition in the forest floor.  相似文献
