Potted plants of Ceratonia siliqua L., growing in a greenhouse,were used to detect xylem cavitation (in terms of ultrasoundacoustic emissions AE) in internodes and node-to-petiole (N-P)junctions, after different periods of drought (9, 16 and 23d). Diurnal AE were only 100 in internodes of watered (W) plantsbut 320, 1250 and 2460 in 9-, 16- and 23-d stressed ones. InN-P junctions, AE were only 15 to 20% with respect to internodes. Stem perfusion with dye allowed measurement of the percentageof xylem conduit transverse area blocked by cavitation. Thiswas 2% in internodes of W-plants and 5.2, 13.8 and 40.4% inthose of 9-, 16- and 23-d stressed ones. In N-P junctions, 18.5%of the xylem conduit transverse area was blocked in the 23-dstressed plants only. The major resistance to cavitation exhibitedby the N-P junctions is interpreted in terms of their greaternumber of narrow xylem conduits. The percentage of blocked xylemconduits within a range of diameters showed that the narrowera xylem conduit, the less likely it was that cavitation wouldoccur. After rewatering, the release of the xylem blockage causedby cavitation occurred within 2 h. Our data suggest that C.siliqua can be considered to be a cavitation avoider, especiallyin its stem-to-leaf transition zones. Measurements of xylem conduit length and width and the distribution of xylem conduit ends were made in inter-nodes (I), nodes (N) and twig junctions (J) of 1-, 2- and 3-year-old twigs of plants of Quercus cerris L. Parallel measurements were also made of the loss of hydraulic conductivity of twigs subjected to pressure differentials across conduit pit membranes, equalling the leaf water potential at the turgor loss point. The loss of theoretical hydraulic conductivity was calculated as the ratio of i esivr4 (where r is the conduit radius) of the non-conducting conduits to that of all the conduits in the outermost wood ring of I, N and J. Stem zones such as 1-year-old nodes and junctions were localized with narrower and shorter xylem conduits and with higher percentages of conduit ends than internodes. Such ‘constricted zonesrsquo; were less vulnerable to embolism than internodes. Latewood conduits were consistently narrower, shorter and less vulnerable to embolism than earlywood ones. A positive relation therefore existed between conduit diameter and length and vulnerability to embolism. The overall vulnerability to embolism of Q. cerris plants is discussed in terms of xylem conduit width and length and of the distribution of conduit ends.  相似文献

Xylem conduction and cavitation in Hevea brasiliensis   总被引:2,自引:0,他引:2  
Clones of Hevea were studied in an attempt to discover the reasonsfor differences in the hydraulic performance of xylem. Differencesbetween clones were determined, including hydraulic conductivityand conduit width and length distributions. However, it hasproved difficult to reconcile anatomical differences with physiologicalperformance for use in future plant breeding programmes. When leaf relative water content (RWC) had been reduced fromabout 95% to 85%, the hydraulic conductivity of petioles decreasedsharply to about 40% of the initial value. This value correspondedwith xylem sap tensions of 1.8–2.0 MPa. Acoustic detectionexperiments revealed that this reduction in hydraulic conductivitycoincided with the greatest occurrence of cavitation. It seemsinescapable that the reduction in hydraulic conductivity wascaused by embolization; thereafter gas bubbles blocked the flowof water inside many of the conduits. There was some indicationthat eventually such bubbles might be dissolved, because thehydraulic conductivity increased again if specimens were fullyrehydrated. Apparently, the incidence of cavitation coincides with the entryof gas bubbles via ultramicroscopic pores into the conduitsthrough the walls according to the air-seeding hypothesis. Whena petiolate leaf is tested in a pressure chamber it is impossibleto make satisfactory measurements of a balancing pressure beyondc. 1.8–2.0 MPa, because air bubbles, mixed with sap andescaping from the conduits, form a persistent froth. Chemical Composition of Bleeding Xylem Sap from Kiwifruit Vines   总被引:5,自引:0,他引:5  
A study of the chemical composition and charge balance was madeof bleeding xylem sap collected from excised one-year-old extensionshoots of healthy, Mn-deficient, Mn-toxic and Zn-deficient kiwifruitvines (Actinidia deliciosa (A. Chev.) C. F. Liang et A. R. Ferguson)immediately prior to leafburst. The exudates were analysed formacronutrient cations and anions, trace elements, amino acids,organic acids and sugars. Major charged species measured wereCa (13.3 mM), K (8.9 mM), Mg (5.6 mM), malate (12.5 mM) andphosphate (5.8 mM). Glutamine (12 mM) was the predominant Ncarrier identified, accounting for 58 per cent of the totalN followed by NO2-N (4.5 per cent), NH4+-N (3.5 per cent)and arginine-N (2.9 per cent). Approximately 22 per cent ofthe N was in a hydrolysable proteinaceous fraction comprisingmainly glutamine and glutamate. Eighteen free proteinaceousamino acids were idetified in sap, the most abundant being glutamine,glutamic acid, valine, isoleucine and phenylalanine. Computersimulation of the chemical composition predicted that in additionto hydrated cations, ion pairs formed between inorganic components(SO42–, HPO42–, H2PO4) and cations (Ca2+,Mg2+, Mn2+), plus metal-organic ligand complexes (Ca Malate,Zn Malate, FeCit, CuHis, CuGln) are important species involvedin translocation. The solubility product of hydroxyapatite wasexceeded in all exudates although in vitro precipitation wasnot observed. To achieve electroneutrality with the componentsmeasured, however, formation of precipitate precursors (hydroxyapatitenuclei) had to be assumed. Irregularities in Mn nutrition (butnot Zn) were clearly indicated by the elemental compositionof exudate suggesting the use of sap analysis as a possiblepre-season indicator of nutritional status for this species. The possible role of water expelled from cavitated xylem conduits in the rehydration of water-stressed leaves has been studied in one-year-old twigs of populus deltoides Bartr. Twigs were dehydrated in air. At desired values of leaf water potential (Ψl) (between near full turgor and -1.62 MPa), twigs were placed in black plastic bags for 1–2h. Leaf water content was measured every 3–5 min before bagging and every 10 min in the dark. Hydraulic conductivity and xylem cavitation were measured both in the open and in the dark. Cavitation was monitored as ultrasound acoustic emissions (AE). A critical Ψl value of -0.96 MPa was found, at which AE increased significantly while the leaf water deficit decreased by gain of water. Since the twigs were no longer attached to roots, it was concluded that water expelled from cavitated xylem conduits was transported to the leaves, thus contributing to their rehydration. Xylem cavitation is discussed in terms of a ‘leaf water deficit buffer mechanism’, under not very severe water stress conditions.  相似文献

Myrothamnus flabellifoliusWelw. is a desiccation-tolerant (‘resurrection’)plant with a woody stem. Xylem vessels are narrow (14 µmmean diameter) and perforation plates are reticulate. This leadsto specific and leaf specific hydraulic conductivities thatare amongst the lowest recorded for angiosperms (ks0.87 kg m-1MPa-1s-1;kl3.28x10-5kg m-1MPa-1s-1, stem diameter 3 mm). Hydraulic conductivitiesdecrease with increasing pressure gradient. Transpiration ratesin well watered plants were moderate to low, generating xylemwater potentials of -1 to -2 MPa. Acoustic emissions indicatedextensive cavitation events that were initiated at xylem waterpotentials of -2 to -3 MPa. The desiccation-tolerant natureof the tissue permits this species to survive this interruptionof the water supply. On rewatering the roots pressures thatwere developed were low (2.4 kPa). However capillary forceswere demonstrated to be adequate to account for the refillingof xylem vessels and re-establishment of hydraulic continuityeven when water was under a tension of -8 kPa. During dehydrationand rehydration cycles stems showed considerable shrinking andswelling. Unusual knob-like structures of unknown chemical compositionwere observed on the outer surface of xylem vessels. Uptake and partitioning through the xylem and phloem of K+,Na+, Mg2+ , Ca2+ and Cl were studied over a 9 d intervalduring late vegetative growth of castor bean (Ricinus communisL.) plants exposed to a mean salinity stress of 128 mol m–3NaCl. Empirically based models of flow and utilization of eachion within the whole plant were constructed using informationon ion increments of plant parts, molar ratios of ions to carbonin phloem sap sampled from petioles and stem internodes andpreviously derived information on carbon flow between plantsparts in xylem and phloem in identical plant material. Salientfeatures of the plant budget for K+ were prominent depositionin leaves, high mobility of K+ in phloem, high rates of cyclingthrough leaves and downward translocation of K+ providing theroot with a large excess of K+ . Corresponding data for Na+showed marked retention in the root, lateral uptake from xylemby hypocotyl, stem internodes and petioles leading to low intakeby young leaf laminae and substantial cycling from older leavesback to the root. The partitioning of the anionic componentof NaCl salinity, Cl, contrasted to that of Na+ in thatit was not substantially retained in the root, but depositedmore or less uniformly in stem, petiole and leaf lamina tissues.The flow pattern for Mg2+ showed relatively even depositionthrough the plant but some preferential uptake by young leaves,generally lesser export than import by leaf laminae, and a returnflow of Mg2+ from shoot to root considerably less than the recordedincrement of the root. Ca2+ partitioning contrasted with thatof the other ions in showing extremely poor phloem mobility,leading to progressive preferential accumulation in leaf laminaeand negligible cycling of the element through leaves or root.Features of the response of Ricinus to salinity shown in thepresent study were discussed with data from similar modellingstudies on white lupin (Lupinus albus L.) and barley (Hordeumvulgare L.) Nodulated 1-1.5-year-old plants of Acacia littorea grown inminus nitrogen culture were each partnered with a single seedlingof the root hemiparasite Olax phyllanthi. Partitioning of fixedN between plant organs of the host and parasite was studiedfor the period 4–8 months after introducing the parasite.N fluxes through nodules of Acacia and xylem-tapping haustoriaof Olax were compared using measured xylem flows of fixed Nand anatomical information for the two organs. N2 fixation duringthe study interval (635 µg N g FW nodules–1 d–1)corresponded to a xylem loading flux of 0.20 µg N mm–2d–1 across the secretory membranes of the pencycle parenchymaof the nodule vascular strands. A much higher flux of N (4891µg mm–2 d–1) exited through xylem at the junctionof nodule and root. The corresponding flux of N from host xylemacross absorptive membranes of the endophyte parenchyma of Olaxhaustorium was 1.15 µg N mm–1 d–1, six timesthe loading flux in nodules. The exit flux from haustorium toparasite rootlet was 20.0 pg N mm–1 d–1, 200-foldless than that passing through xylem elements of the nodule.Fluxes of individual amino compounds in xylem of nodule andhaustorium were assessed on a molar and N basis. Amino acid composition of xylem (tracheal) sap and ethanolicextracts of shoots of mistletoes (Amyema spp. and Lysiana casuarinae)and their hosts were compared, using material collected in theirnative habitats. Data indicated that certain host xylem soluteswere transferred directly to the parasite xylem, while otherswere either not absorbed or were metabolized prior to transfer.Certain solutes were major constituents of parasite xylem, butundetected or only in trace amount in the host. Shoot aminoacid pools of parasites differed markedly from those of hosts.The mistletoe, Amyema preissii, exhibited differential storageand transport of arginine when parasitizing three differentspecies, but accumulated proline on only two of these hosts.Host- specific amino acids (djenkolic acid in Acacia saligna,and tyramine in Acacia acuminata) were transported and accumulatedin relatively large amounts by the parasite, but were not detectedin other associations. Proline was the major solute of Amyemalinophyllum parasitizing Casuarina obesa, but arginine predominatedin Lysiana casuarinae on the same host. However, when L. csuarinaeparasitized A. linophyllum, in turn parasitic on C. obesa, theLysiana accumulated equal amounts of proline and arginine andmore asparagine than when directly on the Casuarina. Xylem feedingof 15N-labelled aspartic acid or 13N-(amide labelled) asparagineto cut shoots or whole haustoria-bearing plants of the mistletoeA. preissii resulted in 68–73% of the 15N of aspartateand 24–30% of that of asparagine appearing in ethanol-solubleshoot amino compounds other than the fed solute. 15N labellingpatterns of detached shoots were not noticeably different fromthat of whole plants suggesting that the haustorium had relativelylittle effect on processing incoming solutes. Alanine, glutamine,and arginine were principal recipients of 15N from aspartate,alanine and glutamine in the case of fed asparagine. It is estimatedthat 24% of the carbon requirements for dry matter accumulationin Amyema linophyllm were met by intake of xylem sap solutesfrom its host Casuarina obesa. Hordeum vulgare cv. California Mariout was established in sandculture at two different NaCl concentrations (0.5 mol m–3‘control’ and 100 mol m–3) in the presenceof 6.5 mol m–3 K +. Between 16 and 31 d after germination,before stem elongation started, xylem sap was collected by useof a pressure chamber. Collections were made at three differentsites on leaves 1 and 3: at the base of the sheath, at the baseof the blade, i.e. above the ligule, and at the tip of the blade.Phloem sap was collected from leaf 3 at similar sites throughaphid stylets. The concentrations of K +, Na+, Mg2+ and Ca2+were measured. Ion concentrations in xylem sap collected at the base of leaves1 and 3 were identical, indicating there was no preferentialdelivery of specific ions to older leaves. All ion concentrationsin the xylem decreased from the base of the leaf towards thetip; these gradients were remarkably steep for young leaves,indicating high rates of ion uptake from the xylem. The gradientsdecreased with leaf age, but did not disappear completely. In phloem sap, concentrations of K+ and total osmolality declinedslightly from the tip to the base of leaves of both controland salt-treated plants. By contrast, Na+ concentrations inphloem sap collected from salt-treated plants decreased drasticallyfrom 21 mol m–3 at the tip to 7.5 mol m–3 at thebase. Data of K/Na ratios in xylem and phloem sap were used to constructan empirical model of Na+ and K+ flows within xylem and phloemduring the life cycle of a leaf, indicating recirculation ofNa+ within the leaf. Abstract

The xylem conduit dimensions (i.e. their width and length) have been measured in 1-year-old internodes, nodes and node-to-petiole (N-P) junctions of three species with diffuse-porous wood, namely Ceratonia siliqua L., Laurus nobilis L. and Olea europaea L. as well as of three species with ring-porous wood, namely Quercus ilex L., Q. suber L. and Q. pubescens Willd‥ The xylem conduit diameter and length distributions have been related to the drought resistance strategies adopted by the six species. C. siliqua and Q. ilex (drought avoiding water spenders) showed the widest xylem conduits (each species within its characteristic pattern of wood anatomy). This is consistent with their high demand of efficient water transport to leaves. L. nobilis (drought avoiding water saver) showed relatively narrow xylem conduits, efficient enough, however, to assure water supply to leaves at the reduced transpiration rate exhibited by the species. O. europaea, Q. suber and Q. pubescens (drought tolerants) showed the narrowest xylem conduits but also the longest ones. The xylem system of C. siliqua and Q. ilex represented a good compromise between efficiency and safety of the water transport, the former as due to wide xylem conduits, the latter to the reduced xylem conduit length as well as to the strong «hydraulic constrictions» at their nodes and N-P junctions. The ecological interpretation of such hydraulic architecture is discussed.  相似文献

Quercus ilex L. growing in the southern Mediterranean Basin region is exposed to xylem embolism induced by both winter freezing and summer drought. The distribution of the species in Sicily could be explained in terms of the different vulnerability to embolism of its xylem conduits. Naturally occurring climatic conditions were simulated by: (1) maintaining plants for 3h at ambient temperatures of 0, -1.5, -2.5, -5.0 and -11°C; and (2) allowing plants to dry out to ratios of their minimum diurnal leaf water potentials (Ψ1) to that at the turgor loss point (Ψtlp) of 0.6, 0.9, 1.05, 1.20 and 1.33. The loss of hydraulic conductivity of one-year-old twigs reached 40% at -1.5°C and at Ψ1tlP= 1.05. Recovery from these strains was almost complete 24 h after the release of thermal stress or after one irrigation, respectively. More severe stresses reduced recovery consistently. The percentages of xylem conduits embolized following application of the two stresses, were positively related to xylem conduit diameter. The capability of the xylem conduits to recover from stress was positively related to the conduit diameter in plants subjected to summer drought, but not in the plants subjected to winter freezing stress. The ecological significance of the different vulnerabilities to embolism of xylem conduits under naturally occurring climatic conditions is discussed.  相似文献

The economy of carbon, nitrogen, water and mineral elementsin fruits of Lupinus albus L. was studied by measuring accumulationof these quantities in the developing fruit and estimating itstranspirational losses and CO2 exchanges. Combining this informationwith data on levels of mineral elements in the xylem sap andphloem sap supplying the fruit, it was possible to test whethertransport based on mass inflow through xylem and phloem wouldaccount for the observed intake of elements. A model of transportbased on water and carbon intake suggested that vascular intakeduring the fruit's life matched the recorded increment for mineralsto within ± 15 per cent for N, Na, Zn, Fe and Cu, andto within ± 23 per cent for P, K and S. However, estimatedvascular intake of Ca, Mg and Mn accounted for less than one–thirdof the recorded intake by the fruit, inadequacy of vascularintake being especially great early in growth. Transport inphloem accounted for more than 80 per cent of the fruit's vascularintake of C, N and S, and 70–80 per cent of its P, K,Mg and Zn. Xylem contributed 68 per cent of the vascular inputof Ca, 59 per cent of the Na, and 34–38 per cent of theFe, Mn and Cu. Enclosure and darkening of fruits reduced levelsof Ca and Fe but increased levels of N, P, K and Zn in fruitdry matter relative to unenclosed, illuminated fruits. Wolterbeek, H. Th. 1987. Relationships between adsorption, chemicalstate and fluxes of cadmium applied as Cd(NO3)2 in isolatedxylem cell walls of tomato.—J. exp. Bot. 38: 419–432. Isolated xylem cell wall pieces were applied as membranes inion diffusion experiments. The cell walls were isolated fromtomato internodes (Lycopersicon esculentum Mill, cv. Tiny Tim)and sealed in a two-compartment diffusion system. In flux andadsorption calculations, the cell wall was regarded as a leakymembrane with parallel fluxes through Donnan Free Space (DFS)and Water Free Space (WFS). During the experiments absorptioninto and diffusion across the walls was determined of Cd2 +, applied as 115Cd(NO3)2. Flux experiments with 82Brindicated that excluded volume effects and path tortuosity resultedin apparent WFS diffusion coefficients in the walls which were0·012 times as high as in water. The free proton concentration in the DFS was shown to be relatedto a complex formation between fixed charges and Cd2 +. Thecell wall permeability for Cd2 + and NO3 varied withapplied and absorbed concentrations, and the Cd2 + flux curveshowed an inflexion point coinciding with a buffered degreeof dissociation of fixed charges in the DFS. The necessary couplingof fluxes of opposite charges resulted in relatively high NO3and small Cd2 + permeability of the DFS for strongly dilutedsolutions (P = 10–4 m s–1 and 10–11 m s–1for NO3 and Cd2 + respectively). The results demonstratethe possible regulatory effects of the cell wall in processesof ion transfer from xylem vessels, or ion uptake in plant tissues. Guttation was used as a non-destructive way to study the flowof water and mineral ions from the roots and compared with parallelmeasurements of root exudation. Guttation of the leaves of barley seedlings depends on age andon the culture solution. Best rates of guttation were obtainedwith the primary leaves of 6- to 7-day-old seedlings grown onfull mineral nutrient solution. The growing leaf tissue becomessaturated with K+ below 1.5 mM K+ in the medium, whereas K+concentration in the guttated fluid still increases furtheras K+ concentration in the medium is raised. At 3 mM K+ averagevalues of guttation were 1.4–2.4 mm3 h–1 per plantwith a K+ concentration of 10–20 mM; for exuding plantsthe flow was 4.2–7.6 mm3 h–1 per plant and K+ concentration35–55 mM. Abscisic acid (ABA) at 10–6 to 10–4 M 0–2h after addition to the root medium increased volume flow ofguttation and exudation and the amount of K+ exported. Threeh after addition of ABA both volume and amount of K+ were reduced.There was an ABA-dependent increase in water permeability (Lp)of exuding roots shortly after ABA addition. Later Lp was decreasedby 35 per cent and salt export by 60 per cent suggesting aneffect of ABA on salt transport to the xylem apart from itseffect on Lp. Benzyladenine (5 x 10–8 to 10–5 M)and kinetin (5 x 10–6 M) progressively reduced volumeflow and K+ export in guttation and exudation and reduced Lp. The composition of xylem sap and exudate from stem incisionsof Nicotiana glauca Grah. was compared in detail. Exudationfrom stem incisions occurred over a 5 min period in certainplants, enabling collection of 5–30 µl of sap. Therate of exudation showed an exponential decline. Exudate hada high dry matter content (170–196 mg ml–1) andhigh sugar (sucrose) levels. Xylem sap had a low pH (5.8) andexudate a pH of 7.9. Glutamine dominated the amino compoundsin xylem sap and exudate, and K+ was the major cation. Totalamino compounds in stem exudate reached 10.8 mg ml–1 whereasxylem sap contained much lower levels (0.28 mg ml–1).All mineral elements and amino compounds with the exceptionof calcium were more concentrated in stem exudate than in xylemsap. Sucrose was labelled heavily in stem exudate following pulsingof an adjacent leaf with 14CO2. A concentration gradient ofsugar (2.1 bar m–1) was recorded for stems. Levels ofsucrose, amino compounds and K+ ions in stem exudate showeda diurnal periodicity. Each commodity reached maximum concentrationat or near noon and minimum concentration about dawn. The evidencesuggests that exudate from stem incisions of N. glauca is arepresentative sample of solutes translocated in the phloem. Solute composition of root xylem sap of common native hostsof quandong (Santalum acuminatum) was compared with that ofcorresponding xylem sap and ethanolic extracts of endophytictissues of haustoria of the hemiparasite. Each host transporteda characteristic set of organic nitrogenous solutes, but littleor no nitrate, and the data indicated only limited direct flowof amino compounds between xylem streams of hosts and parasite.Proline predominated in the haustorium and xylem ofSantalum,but was at negligible levels in the xylem of most hosts. Sucrose,fructose, glucose, malate and citrate were at high levels inall saps, and fructose especially prominent inSantalum. Chloride,sulphate and phosphate were the principal inorganic anions ofthe xylem. Based on C:N ratios of xylem and dry matter ofSantalumandassuming a 70% or more dependence on the host for N, it wasestimated thatSantalumwould gain approximately one third ofits C requirement for dry matter production heterotrophicallyfrom the xylem of its hosts. Infiltration of xylem of haustoria-bearingroot segments of a major host (Acacia rostellifera) with a rangeof15N labelled substrates resulted in 40–80% of the15Nof endophytes of the attached haustoria being received as proline.Nitrate reductase activity was induced in haustoria followinghost xylem feeding of nitrate. The study concludes that haustoriaofSantalumact as a major site of synthesis and export of prolineand might therefore play an important role in osmotic adjustmentof the parasite and its related acquisition of water from hosts.
