The presence of C37-norcarotenoids (peridinin and probably pyrrhoxanthin, together 87% of total carotenoids) and the carotenoid pattern in general, including dinoxanthin, diatoxanthin and β,β-carotene, but no fucoxanthin, strongly suggest that H. akashiwo is a dinoflagellate and not a chrysophyte.  相似文献

Allelopathic effects of the green macroalgae Ulva lactuca on the growth of three species of red tide microalgae, Heterosigma akashiwo, Alexandrium tamarense, and Skeletonema costatum were tested in laboratory co-cultures precluding the nutrient and light limitation and the effect of high pH. The growth of all three species of microalgae was significantly (p < 0.01) inhibited by fresh U. lactuca. In nutrient replete semicontinuous co-cultures with U. lactuca, H. akashiwo was completely dead in 12 days, and the growth of A. tamarense and S. costatum was reduced by 48 and 46%, respectively by U. lactuca within 12 days. The U. lactuca culture filtrate exhibited a significant (p < 0.05) inhibitory effect on the microalgae in the first 1 or 2 days, but growth resumed in the following days, and S. costatum growth was slightly (p > 0.05) promoted from day 3. The results suggested that the allelopathic compounds are quickly degradable and a long-term inhibition might need the continuous addition of compounds originated from macroalgae. Dried U. lactuca also exhibited inhibitory effects on the microalgae, and the normalized mean growth rates of microalgae decreased with the biomass of dried U. lactuca. The dependent relationships were y = −2.1208x2 + 1.0159x + 0.9752 for H. akashiwo, y = 0.7133x2 − 3.5813x + 1.1665 for A. tamarense, and y = −0.2114x2 − 1.063x + 1.0873 for S. costatum, respectively. The potential feasibility of utilization of dried U. lactuca against red tide microalgae was 2.0 g dry wt L−1. The present study shows that U. lactuca exhibits negative allelopathic effects on harmful bloom-forming microalgae.  相似文献

Growth and dark respiration rates of the marine diatom Leptocylindrus danicus Cleve were measured in axenic batch culture under 49 combinations of temperature (5, 10, 15, 20°C), daylength(15:9, 12:12, 9:15 LD), and irradiance (at least four irradiances per daylength). Cell division rates exhibited a temperature-dependent daylength effect. Optimal temperatures occurred between 15 and 20°C. Both the initial slope () and the growth rate at light saturation (μmax) were strongly influenced by temperature; increased five-fold and μmax by an order of magnitude between 5 and 20°C. The compensation irradiance (Ic) was independent of temperature. μmax was 2.7 div day−1 at 20°C, 2.6 at 15°C, 1.1 at 10°C, and 0.3 at 5 °C. Cells grown under 15:9 and 12:12 LD exhibited similar growth-light curves at 20°C and at 15°C. μmax of cells grown under 9:15 LD at these temperatures were substantially lower than μmax under longer daylengths. Growth at 10 and 5°C was independent of daylength.

Dark respiration rates were a linear function of cell division rates at 10, 15, and 20°C, and support the concept that growth rate is dependent on dark respiration rate. These relationships were not influenced by daylength. A detectable relationship between dark respiration and growth at 5°C was not observed.

Photosynthesis and excretion showed temperature-dependent curvilinear relationships with growth rate, reflecting the lower saturation irradiance for growth compared to light saturation of photosynthesis and excretion. The relationship between Chl a-specific photosynthesis and growth was controlled by the C:Chl a ratio, which showed a positive correlation with cell division rate. At 15 and 20°C, light saturation of growth was associated with C:Chl a ratios of 40 to 60; at 5 and 10°C, cells growing at μmax contained C:Chl a in ratios of 80 to 110.  相似文献

Cell division and chloroplast replication in Heterosigma akashiwo(Hada) Hada occurred as separate synchronous events during thecell cycle when cells were subjected to light-dark regimes.Under three different photoperiodic cycles of 10L/14D (10 hlight/14 h dark), 12L/12D or 16L/8D, cell division began athour 19–20 and finished at hour 23–26 after theonset of the light period, while chloroplast replication beganat hour 20–22 after the onset of the dark period. Almostall the cells divided only once in the 12L/12D cycle. The rateof increase in chloroplast number during one light-anddark cyclewas always equal to that in cell number in every photoperiodexamined. Light was essential for both cell division and chloroplast replication,but the minimum light period necessary for each event differed.When the light period was shorter than 6 h, no cell divisionoccurred; when it was shorter than 3 h, no chloroplast replicationoccurred. Temperature in combination with different feeding frequencies caused rapid changes in the morphology of experimental culture populations of Haliplanella luciae (Verrill) due to exogenous effects on fission rate (k). A temperature of 15°C permitted few fissions, and only when the anemones were fed at frequent intervals. Conditions which maximized k caused significant decreases in diameter, numbers of septa, and numbers of tentacles after only 26 days in culture. Anemones fed every two days at 15°C showed the least morphological change over the culture period, remaining morphologically similar to field specimens. Asexual reproduction in H. luciae is not as well integrated with organismal growth as in hydroids. The importance of k and environmental regulators of k in determining both morphological structure and population size of H. luciae is discussed. In this study H. luciae were of a previously undescribed color variant, with light longitudinal bands.  相似文献

Lactobacillus reuteri shows certain beneficial effects to human health and is recognized as a probiotic. However, its application in frozen foods is still not popular because of its low survival during freezing and frozen storage. Cell immobilization technique could effectively exert protection effects to microbial cells in order to enhance their endurance to unfavorable environmental conditions as well as to improve their viability and cell concentration. Ca-alginate and κ-carrageenan were used to immobilize L. reuteri in this research, and the immobilized cells were exposed to different freezing temperatures, i.e. − 20 °C, − 40 °C, − 60 °C, − 80 °C, and stored at − 40 °C and − 80 °C for 12 weeks. The objectives were to study the protection effects of cell immobilization against the adverse conditions of freezing and frozen storage, and the effects of freezing temperatures to the immobilized cells. Cell immobilization was used to raise the survival of L. reuteri during freezing and frozen storage in order to develop frozen foods with the probiotic effects of L. reuteri. Results indicated that immobilized L. reuteri possessed better survival in both freezing and frozen storage. The survival of immobilized L. reuteri was higher than that of free cells, and the effects of lower freezing temperature were better than higher freezing temperature. The immobilization effects of Ca-alginate were found to be superior to κ-carrageenan. Cell immobilized L. reuteri exhibits potential to be used in frozen foods.  相似文献

Floating Pennywort (Hydrocotyle ranunculoides L. fil.) is a worldwide distributed aquatic plant. The species is native to North America and quite common also in Central and South America. In Europe, Japan and Australia it is known as an alien plant, sometimes causing serious problems for affected ecosystems and human use of water bodies. Starting from Western Europe with an eastwards directed spread, Floating Pennywort was recorded in Germany in 2004 for the first time. Since then, the species spread out and got established in western parts of Central Europe. For a definite prediction of the potential of a further spread, data about biology, in particular growth and photosynthesis are needed. Here, regeneration capacity, growth at different nutrient availabilities and photosynthesis of H. ranunculoides were investigated. In addition biomass samples were taken in the field. Results show an enormous regeneration capacity (e.g., by forming new shoots from small shoot fragments), increasing growth rates under increasing nutrient availability and a maximum increase of biomass reaching 0.132±0.008 g g−1 dw d−1. Dense populations of H. ranunculoides growing in ponds and oxbows were found at high nutrient content of the substrate, the biomass reaching there up to 532.4±14.2 g dw m−2. Gas exchange analysis showed a physiological optimum of H. ranunculoides CO2 uptake at temperatures between 25 and 35 °C and high photon flux densities (PPFD) above 800 μmol photons m−2 s−1. In comparison, native Hydrocotyle vulgaris showed an optimum of net photosynthesis at 20–30 °C and a light saturation of CO2 gas exchange at 350 μmol photons m−2 s−1.  相似文献

Life cycle stages of a new species of the genus Nosema Naegeli, 1857 (Microspora, Nosematidae), were examined by light and electron microscopy. It parasitizes the gut and the uterus of the digenean Monorchis parvus (Monorchiidae), in Diplodus annularis (Pisces, Teleostei). All stages were in close contact with the cytoplasm of the host cell and were probably all diplokaryotic. The divisions of meronts and sporonts were recognizable by the formation of spindle plaques at the surface of the nucleus. Spores were oval, measured 3.2±0.3×2.5±0.2 μm on ultrathin sections, and had a polar filament with 16–17 coils. The polaroplast presented two parts: an anterior region with closely packed lamellae and a posterior part with wider lamellae. This Nosema species is compared with the other microsporidian parasites of digeneans. This new species is named Nosema monorchis n. sp., after the generic name of its host.  相似文献

The effects of the endoparasitic fungus Hirsutella minnesotensis on populations of Meloidogyne hapla from Michigan (MI), Rhode Island (RI), Connecticut (CT), Lyndonville, New York (NYL), Geneva, New York (NYG), and Wisconsin (WI) were studied in the greenhouse. Twenty-day-old tomato (cv. Rutgers) seedlings were inoculated with either 0 or 600 eggs of each nematode population mixed with either 0, 0.02, or 0.1 g of fresh H. minnesotensis mycelium 0.1 L-1 of soil in pots containing 0.5 L of soil, and maintained at 25±2°C for 2 months. No effect of the fungal treatments and nematode treatments on tomato plant heights and shoot dry weights was observed. While all M. hapla populations were suppressed by H. minnesotensis, the degree to which each population was affected varied slightly. Across fungal treatments and nematode populations, the fungus reduced total number of nematodes in roots by 61-98%, with the highest for NYG and RI, intermediate for NYL and CT, and lowest for MI and WI populations. The study demonstrated that H. minnesotensis may be used as a potential suppressor of M. hapla in vegetable production systems in the Great Lakes Region.  相似文献

Diel vertical migration by Heterosigma akashiwo (Hada) Hada (Raphidophyceae) was monitored in a 1.5 in tall microcosm. Vertical stratification, with low salinity and low orthophosphate (Pi) concentration in the upper layer and high salinity and high Pi concentration in the lower layer, was simulated in the tank, analogous to summer stratification in the Seto Inland Sea. The phosphate metabolism of H. akashiwo during this vertical migration was studied using 31P-NMR spectroscopy. At night this species migrated to the lower phosphate-rich layer and took up inorganic phosphate (Pi) which then was accumulated as polyphosphate (PPi) by an increase in the chain length of PPi During the daytime this species migrated to the phosphate-depleted surface water and utilized the accumulated PPi for photophosphorylation by decreasing the chain length of PPi During the first night after the phosphorus was introduced to the previously impoverished waters, the cells took up inorganic phosphate, accumulating the new phosphorus nutrient internally as Pi But the cells did not convert Pi to PPi presumably due to their lack of ATP. After the second day of the experiment, conversion of Pi to PPi at night was much more rapid than on the first day, presumably due to increased ATP availability. Then the cycle continued, with uptake of Pi and conversion to PPi at night at the bottom and its utilization during the day at the surface. These data suggest that the role of PPi in the metabolism of this species appears to be as a phosphate pool which regulates the level of Pi and ATP in the cell. Diel vertical migration allows this red tide species to shuttle between the phosphate-rich lower layer and the photic upper layer in stratified waters. 31P-NMR is shown to be a valuable tool in studying the phosphorus metabolism in migrating organisms.  相似文献

The tube building polychaete Hydroides elegans Haswell was found living attached to colonies of the arborescent bryozoan Bugula neritina (L.) in Port Shelter, Hong Kong. Field data collected during the period of January through May 1996, showed that H. elegans density reached 77.6 individuals of H. elegans per g wet weight of B. neritina. Density of H. elegans on B. neritina at depths from the surface to 0.5 m was lower than that at depths below 1 m. In January–March, when there were no H. elegans settling on PVC plates or found on natural substrata, numbers on B. neritina were ca. 5 per g wet weight. H. elegans settled on B. neritina and grew rapidly as mean diameter of tubes increased from 605 μm in February to 936 μm in March. In laboratory experiments, larvae of H. elegans settled and metamorphosed on branches of B. neritina and on the bottom of dishes containing B. neritina leachate. Compounds extracted from the leachate of B. neritina induced 74% of H. elegans larvae to metamorphose at a concentration of 16 μg/ml seawater, compared to 5% in dishes containing only filtered seawater (controls). Metabolites from the leachate of B. neritina which were bound to amberlite XAD-2, indicating they are lipophilic in nature, induced over 70% metamorphosis in H. elegans larvae at 56 μg/ml seawater. A biofilm from one of four strains of bacterial isolates associated with the surface of B. neritina induced low levels of metamorphosis in H. elegans larvae, while other bacterial isolates were detrimental to the survival of juvenile H. elegans. Field experiments further demonstrated that H. elegans settled preferentially on Phytagel discs embedded with whole extracts of B. neritina over control Phytagel discs. Metabolites from B. neritina deterred feeding on alginate pellets by assemblages of local fishes in field assays. Metabolites originating from B. neritina, bacteria colonizing B. neritina, and the complex structure of B. neritina contributed to the recruitment of H. elegans to B. neritina surfaces. Hydroides elegans may gain a refuge from predation by associating with B. neritina colonies both from its structural and chemical attributes.  相似文献

Deregulated cell turnover in Helicobacter pylori (H. pylori)-colonized gastric mucosa has been suggested to be linked to the gastric carcinogenesis pathway. We previously reported attenuation of apoptosis and enhancement of cellular proliferation in the H. pylori-colonized gastric mucosa of Mongolian gerbils as compared to that in mice, which might reflect a specific link between H. pylori colonization and carcinogenesis in the Mongolian gerbils; the difference between the two strains could be attributable to differences in the host genetic background. Inducible-type nitric oxide synthase (iNOS) is thought to participate in not only the inflammatory response, but also in the regulation of gastric mucosal cell turnover in H. pylori-colonized gastric mucosa. Thus, the present study was designed to examine gastric leukocyte activation and epithelial cell apoptosis in the gastric mucosa following H. pylori inoculation in iNOS-knockout mice. Methods: iNOS-knockout mice (iNOS−/−) and their iNOS+/+ littermates were orally inoculated with the Sydney strain of H. pylori (SS1, 108 colony-forming units [CFU]). H. pylori infection was confirmed by microaerobic bacterial culture. The stomach of each mouse was evaluated 14 weeks and 30 weeks after the inoculation. Gastric mucosal accumulation of polymorphonuclear leukocytes (PMN) was assessed by determining the myeloperoxidase (MPO) activity and histological score based on the updated Sydney system. The level of apoptosis was determined by estimation of the cytoplasmic levels of mono- and oligonucleosomes and by the terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick-end labeling method. Results: The SS1-inoculated mice showed persistent H. pylori colonization for 12 weeks. While gastric mucosal PMN infiltration increased following SS1 inoculation in both iNOS+/+ and iNOS−/−strains, enhanced DNA fragmentation was observed in only SS1-colonized iNOS+/+ mice, and not in the iNOS−/− mice. In conclusion, although the recruitment of PMN in response to H. pylori was evoked even in the gastric mucosa of iNOS−/− mice, epithelial cell apoptosis induced by H. pylori was attenuated in this strain. These data suggest that iNOS may play an important role in promoting apoptosis in the H. pylori-infected inflamed gastric mucosa, and that persistent inflammation without apoptosis in iNOS−/− mice with H. pylori infection may be linked to preneoplastic transformation.  相似文献

An axenic clonal culture of Chattonella antiqua (Hada) Ono was grown on a 12: 12 h LD cycle in a laboratory culture tank containing 1 m3 of f/2 medium. Diel changes in mean cell volume, cellular carbon (carbon content per cell), C/N ratio, cellular Chl a, Chl a/c ratio and carotenoid composition were observed. Mean cell volume and cellular C, N and pigments increased during the light period as a result of photosynthesis and decreased with increase of cell concentration by phased cell division during the dark period. These changes indicated that carbon assimilation and pigment synthesis occurred together during the light period. However, the patterns of increase were not the same since different diel patterns were also found in the ratios of C/N and chl a/c. Photosynthetic pigments were analyzed by reversed-phase high-performance liquid chromatography with ion-pairing solution. This analysis showed that the dominant carotenoids in C. antiqua were fucoxanthin, violaxanthin and β-carotene. Diel patterns of Chls a and c were similar to that of fucoxanthin but different from those of violaxanthin and β-carotene. The cellular contents of Chl a, fucoxanthin and carbon increased in a parallel manner during the light period. On the other hand, the increase of violaxanthin was restricted to only a few hours at the beginning of the light period during cell division cycles.  相似文献

以我国典型赤潮藻赤潮异弯藻(Heterosigma akashiwo)为研究对象,设置了6个浓度梯度(0 mg·L-1、0.001mg·L-1、0.01 mg·L-1、0.1 mg·L-1、1 mg·L-1和10 mg·L-1)的草甘膦异丙胺盐处理,研究了草甘膦异丙胺盐暴露对赤潮异弯藻的生长、叶绿素a含量和可溶性蛋白含量等指标的影响.结果表明,草甘膦异丙胺盐对赤潮异弯藻具有明显的毒性效应,10 mg·L-1浓度处理下,赤潮异弯藻细胞大量死亡,藻细胞密度以及叶绿素a、可溶性蛋白的含量显著降低(p<0.05);当草甘膦异丙胺盐浓度在0.001~1 mg·L-1范围内,在培养的第3 d草甘膦异丙胺盐能够显著促进赤潮异弯藻的细胞密度增加,叶绿素a含量也明显高于对照组(p<0.05),表现出毒物刺激效应;在暴露实验的中后期(第7 d、第9 d和第11 d),赤潮异弯藻的各生长指标均与对照无显著差异,可能是随着培养时间的延长,农药的降解、生物体对农药的适应、进入细胞的农药减少等原因,藻细胞生理状态逐渐恢复到正常水平.  相似文献

This study uses chlorophyll a fluorescence to examine the effect of environmentally relevant (1–4 h) exposures of thermal stress (35–45 °C) on seagrass photosynthetic yield in seven tropical species of seagrasses. Acute response of each tropical seagrass species to thermal stress was characterised, and the capacity of each species to tolerate and recover from thermal stress was assessed. Two fundamental characteristics of heat stress were observed. The first effect was a decrease in photosynthetic yield (Fv / Fm) characterised by reductions in F and Fm′. The dramatic decline in Fv / Fm ratio, due to chronic inhibition of photosynthesis, indicates an intolerance of Halophila ovalis, Zostera capricorni and Syringodium isoetifolium to ecologically relevant exposures of thermal stress and structural alterations to the PhotoSystem II (PSII) reaction centres. The decline in Fm′ represents heat-induced photoinhibition related to closure of PSII reaction centres and chloroplast dysfunction. The key finding was that Cymodocea rotundata, Cymodocea serrulata, Halodule uninervis and Thalassia hemprichii were more tolerant to thermal stress than H. ovalis, Z. capricorni and S. isoetifolium. After 3 days of 4 h temperature treatments ranging from 25 to 40 °C, C. rotundata, C. serrulata and H. uninervis demonstrated a wide tolerance to temperature with no detrimental effect on Fv / Fm′ qN or qP responses. These three species are restricted to subtropical and tropical waters and their tolerance to seawater temperatures up to 40 °C is likely to be an adaptive response to high temperatures commonly occurring at low tides and peak solar irradiance. The results of temperature experiments suggest that the photosynthetic condition of all seagrass species tested are likely to suffer irreparable effects from short-term or episodic changes in seawater temperatures as high as 40–45 °C. Acute stress responses of seagrasses to elevated seawater temperatures are consistent with observed reductions in above-ground biomass during a recent El Niño event.  相似文献

The diel pattern of cell division, cell carbon, adenine nucleotides and vertical migration was determined for laboratory cultures of the photosynthetic marine dinoflagellate, Ceratium furca (Ehr.) Clap. & Lachm., entrained on an alternating 12:12 LD schedule at 20 C. Cell division was initiated during the latter portion of the dark period with ca. 30% of the population undergoing division. Cell C increased during the light period and exhibited a linear decrease with a loss of 33% during the dark period. ATP · cell?1 increased during the light period and decreased by ca. 40–50% during the dark period. The diel patterns of cell C and ATP tended to “buffer” the magnitude of the change in C:ATP ratios around an overall mean value of 89. There was no obvious trend in the concentration of [GTP + UTP] · cell?1 over the cell cycle. The cellular adenylate energy charge was maintained at values between 0.8 to 0.9 throughout the 24 h LD cycle, despite a ca. 40% decrease in total adenylates (AT= ATP + ADP + AMP) during the dark period on 12:12 LD, and over a 68% decrease in ATP during 42 h of continuous darkness. These data lend experimental support to the theory of cellular metabolic control by the adenine nucleotides. With lateral illumination on 12:12 LD cycles, the cells began to concentrate at the surface of the experimental tubes shortly before the lights were turned on, and at the bottom of the tubes shortly before the lights were extinguished. This pattern continued for 6 days in continuous darkness, suggesting that the vertical migration pattern is independent of a phototactic response and may be under the control of an endogenous rhythm.  相似文献

Whole cells of Bacillus halodurans LBK 261 were used as a source of catalase for degradation of hydrogen peroxide. The organism, B. halodurans grown at 55°C and pH 10, yielded a maximum catalase activity of 275 U g-1 (wet wt.) cells. The catalase in the whole cells was active over a broad range of pH with a maximum at pH 8-9. The enzyme was optimally active at 55°C, but had low stability above 40°C. The whole cell biocatalyst exhibited a Km of 6.6 mM for H2O2 and Vmax of 707 mM H2O2 min-1 g-1 wet wt. cells, and showed saturation kinetics at 50 mM H2O2. The cells were entrapped in calcium alginate and used for H2O2 degradation at pH 9 in batch and continuous mode. In the batch process, the immobilized preparation containing 1.5 g (wet wt.) cells could be recycled at least four times for complete degradation of the peroxide in 50 mL solution at 25°C. An excess of immobilized biocatalyst could be used in a continuous stirred tank reactor for an average of 9 days at temperatures upto 55°C, and in a packed bed reactor (PBR) for 5 days before the beads started to deform.  相似文献

Supplementation of culture medium with hemoglobin solution (Erythrogen™) promoted mitotic division, cell colony formation, and plant regeneration from rice (Oryza sativa L. cv. Taipei 309) protoplasts. The mean (± s.e.m., N = 5) final protoplast plating efficiency (FPE) at 28 days following exposure to 1:50 (v:v) Erythrogen™ (0.55 ± 0.06%) was significantly greater (P < 0.01) than in untreated controls (0.26 ± 0.02%). A similar, but less pronounced effect (P < 0.01) also occurred with 1:100 (v:v) (PE 0.53 ± 0.04%) and 1:500 (v:v) (PE 0.46 ± 0.05%) Erythrogen™, respectively. In contrast, there was no corresponding increase in plating efficiency with Erythrogen™ at 1:1,000 (v:v). These beneficial effects were sustained throughout culture. This leads to a 44% increase (P < 0.01) in protoplast-derived calli producing shoots compared to controls. Cytological analyses confirmed the diploid status of plants regenerated from Erythrogen™-treated and control protoplasts.  相似文献

Metal concentrations and population parameters of the seagrass Halodule wrightii were determined at three locations at Rio de Janeiro State, Brazil. The possible increase of metal availability in one of these areas, Sepetiba Bay, as a result of dredging of contaminated bottom sediments which ocurred, was evaluated by analyses of Al, Cd, Cr, Cu, Fe, Ni, Pb and Zn in root, rhizome and shoots. In addition, analyses were carried out in H. wrightii populations from non-contaminated areas located at northwestern (Cabo Frio) and southeastern (Angra do Reis) regions of Rio de Janeiro State. Concurrently, abundance and density data of the seagrass populations were obtained. It was found that concentration from Sepetiba Bay samples up to 1.6 ± 0.4 μg g−1 of Cd, 12 ± 1.0 μg g−1 of Cr, 27 ± 2.4 μg g−1 of Pb, 291 ± 47 μg g−1 of Mn, 128 ± 23 μg g−1 of Zn were significantly higher than that from two other collection sites. An increase in Cd and Zn concentration was observed in H. wrightii from Sepetiba Bay indicating that metal mobilization from contaminated sediments through dredging activities were, at least in part, transferred to the biotic compartment via accumulation by the seagrass. The populations of seagrass within the region demonstrated quite substantial changes in biomass data but not in shoot or rhizome density during the study. Such changes in biomass are to be expected, as these dynamics are typical of the small, isolated monospecific populations of H. wrightii along the Rio de Janeiro coast.  相似文献
