Many different methods have been developed in recent years to gain insight into the structure of proteins, membranes, organelles and cells. Here we demonstrate the application of near-field scanning optical microscopy (NSOM) for analysis of the structures of typical photosynthetic membrane objects such as chloroplasts and thylakoids from spinach and chromatophores from purple bacteria. To our knowledge, this is the first report of application of NSOM to imaging chromatophores from photosynthetic bacteria and intact thylakoids from higher plants. NSOM has the ability to measure optical signals originating from the sample with a spatial resolution better than conventional optical microscopy. The main advantage of near-field optical microscopy, besides the improved lateral optical resolution, is the simultaneously acquired topography. We have applied NSOM to thylakoids obtained by osmotic shock of chloroplasts. Swollen thylakoids had average diameters of 0.8–1 micron and heights of 0.05–0.07 micron. We also describe the use of fluorescent dyes for the analysis of structures resulting from fusion of photosynthetic bacterial chromatophores with lipid impregnated collodion membranes. The structures formed after fusion of chromatophores to the collodion film have diameters ranging from 0.2 to 10 microns and heights from 0.01 to 1 micron. The dual functionality (optical and topographical), high spatial resolution, and the possibility to work with wet samples and under water, make NSOM a useful method for examining the structures, sizes, and heterogeneity of chromatophore and thylakoid preparations.  相似文献

Porous filters and collodion film impregnated with decane solution of phospholipids, were used for measurements of electric potential differences generated by bacteriorhodopsin, chromatophore redox chain, H+-ATPase, pyrophosphatase, and mitochondrial respiratory chain. It was shown that reconstituted proteoliposomes, containing e.g., bacteriorhodopsin or natural coupling membrane vesicles, such as Rhodospirillum rubrum chromatophores, can be attached to a filter surface by means of Ca2+ or Mg2+ ions. Addition of the respective energy source was found to result in electric potential difference being generated across the filter. This effect was measured directly by AgAgCl electrodes immersed into electrolyte solutions on both sides of the filter. Using a phospholipid-impregnated collodion film one can measure electric responses as fast as 300 nsec. The phospholipid-impregnated filters turned out to be sensitive and reliable electrodes for measuring the concentration of synthetic penetrating ions, such as phenyldicarbaundecaborane, tetraphenylborate, tetrapentylammonium, and tetraphenylphosphonium. By measuring changes in the concentration of these ions in the suspension of proteoliposomes, chromatophores, mitochondria, or bacterial cells, one can follow the formation and dissipation of transmembrane potential differences in these systems. It is shown that the phospholipid-impregnated filters are much more reliable and handy than planar phospholipid membranes previously used for studying electrogenic activity of electric current-producing membrane proteins.  相似文献

Direct electrometric measurements of membrane potential changes are a valuable tool for study of vectorial transfer of electrons, protons, and ions. Commonly model membrane systems are created by fusion of lipid/protein vesicles with lipid-coated thin films. We characterized the electric units resulting from this process using chromatophores from the purple bacterium Rhodobacter sphaeroides and either a Mylar film or a planar modified gold electrode as support. Investigation of the shunting activity of the ionophore gramicidin on the flash-induced potential change demonstrates fusion of individual chromatophores to form independent 'blisters', which preserve an interior aqueous compartment. Under current-clamp conditions the photovoltage follows the change of the membrane potential of the individual blisters.  相似文献

Generation of photoelectric potential in chromatophores of Rhodopseudomonas sphaeroides has been measured (i) spectrophotometrically, using electrochromic shift of carotenoid absorption band or (ii) electrometrically, by means of two electrodes separated by a collodion film covered on one side with chromatophores. A 15 ns laser flash was used to induce a single turnover of photosynthetic reaction centers. It was found that results obtained by both methods are similar in (i) direction of electric vector (the chromatophore interior positive) and (ii) redox titration curves (Em = 10mV). The magnitudes of the photopotential were about 60 and 25 mV, when monitored with spectral and electrometric techniques, respectively. In both cases, the rise times of the photopotentials were faster than time resolution of the techniques used. Decay of the response of carotenoids was found to be slower than that in the collodion film system. The addition of ubiquinone Q10 into the decane solution of asolectin used to impregnate the collodion film led to slowing down of the decay. The carotenoid response decay could be accelerated by FCCP or o-phenanthroline. In the latter case, the shape of the decay curve coincides with decay of the photopotential measured in the collodion film system. It is suggested that decane extracts secondary ubiquinone from chromatophores attached to the collodion film. Such an unfavorable effect can be strongly decreased by added ubiquinone  相似文献

1. The cataphoretic P.D. of suspended particles is assumed to be due to an excess in the concentration of one kind of a pair of oppositely charged ions in the film of water enveloping the particles and this excess is generally ascribed to a preferential adsorption of this kind of ions by the particle. The term adsorption fails, however, to distinguish between the two kinds of forces which can bring about such an unequal distribution of ions between the enveloping film and the opposite film of the electrical double layer, namely, forces inherent in the water itself and forces inherent in the particle (e.g. chemical attraction between particle and adsorbed ions). 2. It had been shown in a preceding paper that collodion particles suspended in an aqueous solution of an ordinary electrolyte like NaCl, Na2SO4, Na4Fe(CN)6, CaCl2, HCl, H2SO4, or NaOH are always negatively charged, and that the addition of these electrolytes increases the negative charge as long as their concentration is below M/1,000 until a certain maximal P.D. is reached. Hence no matter whether acid, alkali, or a neutral salt is added, the concentration of anions must always be greater in the film enveloping the collodion particles than in the opposite film of the electrical double layer, and the reverse is true for the concentration of cations. This might suggest that the collodion particles, on account of their chemical constitution, attract anions with a greater force than cations, but such an assumption is rendered difficult in view of the following facts. 3. Experiments with dyes show that at pH 5.8 collodion particles are stained by basic dyes (i.e. dye cations) but not by acid dyes (i.e. dye anions), and that solutions of basic dyes are at pH 5.8 more readily decolorized by particles of collodion than acid dyes. It is also shown in this paper that crystalline egg albumin, gelatin, and Witte''s peptone form durable films on collodion only when the protein exists in the form of a cation or when it is isoelectric, but not when it exists in the form of an anion (i.e. on the alkaline side of its isoelectric point). Hence if any ions of dyes or proteins are permanently bound at the surface of collodion particles through forces inherent in the collodion they are cations but not anions. The fact that isoelectric proteins form durable films on collodion particles suggests, that the forces responsible for this combination are not ionic. 4. It is shown that salts of dyes or proteins, the cations of which are capable of forming durable films on the surface of the collodion, influence the cataphoretic P.D. of the collodion particles in a way entirely different from that of any other salts inasmuch as surprisingly low concentrations of salts, the cation of which is a dye or a protein, render the negatively charged collodion particles positive. Crystalline egg albumin and gelatin have such an effect even in concentrations of 1/130,000 or 1/65,000 of 1 per cent, i.e. in a probable molar concentration of about 10–9. 5. Salts in which the dye or protein is an anion have no such effect but act like salts of the type of NaCl or Na2SO4 on the cataphoretic P.D. of collodion particles. 6. Amino-acids do not form durable films on the surface of collodion particles at any pH and the salts of amino-acids influence their cataphoretic P.D. in the same way as NaCl but not in the same way as proteins or dyes, regardless of whether the amino-acid ion is a cation or an anion. 7. Ordinary salts like LaCl3 also fail to form a durable film on the surface of collodion particles. 8. Until evidence to the contrary is furnished, these facts seem to suggest that the increase of the negative charge of the collodion particles caused by the addition of low concentrations of ordinary electrolytes is chiefly if not entirely due to forces inherent in the aqueous solution but to a less extent, if at all, due to an attraction of the anions of the electrolyte by forces inherent in the collodion particles.  相似文献

Intracytoplasmic membranes of the mutant strain NK3 of Rhodopseudomonas capsulata lacking the lightharvesting complex B800-850 were fused with proteoliposomes containing the B800-850 complex. Fluorescence emission spectroscopy at 77K showed that after fusion the fluorescence of the B850 bacteriochlorophyll disappeared nearly completely and the B870 fluorescence became prominent. This result and control experiments with proteoliposome-chromatophore mixture and with chromatophore and solubilized B800-850 complexes, respectively, indicate that in fused membranes a reorientation of membrane particles took place and excitons migrated from B850 to B870 bacteriochlorophyll.In fused proteoliposome-chromatophore vesicles a light-induced carotenoid band shift was observed, reflecting the building of an electrical membrane potential due to chargeseparation. Chlorophyll a/b light-harvesting complexes (chl a/b LHC) and photosystem II (PSII) cores were isolated from an octyl glucoside-containing sucrose gradient after solubilization of barley thylakoid membranes with Triton X-100 and octyl glucoside. No cation precipitation step was necessary to collect the chl a/b LHC. PAGE under mildly denaturing and fully denaturing conditions showed that the chl a/b LHC fraction contained chlorophyll-protein complexes CP27, CP29, and CP64. The PSII core material contained CP43 and CP47, and little contamination by other nonpigmented polypeptides. Freeze-fracture electron microscopy of the chl a/b LHC after reconstitution into digalactosyldiglyceride (DG) or phosphatidylcholine (PC) vesicles showed that the protein particles (approximately 7.5 +/- 1.6 nm) were approximately 99 and 90% randomly dispersed, respectively, in the liposomes. Addition of Mg++ produced particle aggregation and membrane adhesion in chl a/b LHC-DG liposomes in a manner analogous to that described for LHC-PC liposomes. Reconstitution of PSII cores into DG vesicles also produced proteoliposomes with randomly dispersed particles (approximately 7.5 +/- 1.6 nm). In contrast, PSII-PC mixtures formed convoluted networks of tubular membranes that exhibited very few fracture faces. Most of the protein particles (approximately 7.0 +/- 1.5 nm) were seen trapped between, rather than embedded in, the membranes. The interaction between the zwitterionic head group of the phosphatidyl choline and the negatively charged PSII core may be responsible for the unusual membrane structures observed.  相似文献

The acidic phospholipid cardiolipin was shown to be very efficient in promoting calcium-induced fusion of proteoliposomes. The degree of fusion was dependent on the phosphatidylethanolamine content of the vesicles. Addition of CaCl2 to proteoliposomes containing phosphatidylcholine and cardiolipin but without phosphatidylethanolamine did not induce fusion. Fusion of cytochrome oxidase vesicles, containing less than 50 mol% phosphatidylethanolamine resulted in monolamellar vesicles with a diameter of about 200 nm. The vesicles could be induced to fuse further by establishing an osmotic pressure across their membranes. When proteoliposomes containing more than 50 mol% phosphatidylethanolamine were fused, large vesicles with a diameter exceeding 1 μm were formed. They appeared in the electron microscope as a mixture of multilamellar and monolamellar vesicles. Fusion of corresponding liposomes resulted in formation of even larger structures appearing as dense multilamellar bodies and paracrystalline honeycomb-like lattices.  相似文献

Lipoprotein complexes, containing (1) bacteriochlorophyll reaction centers, (2) bacteriochlorophyll light-harvesting antenna or (3) both reaction centers and antenna, have been isolated from chromatophores of non-sulphur purple bacteria Rhodospirillum rubrum by detergent treatments. The method of reconstituting the proteoliposomes containing these complexes is described. Being associated with planar azolectin membrane, proteoliposomes as well as intact chromatophores were found to generate a light-dependent transmembrane electric potential difference measured by Ag/AgCl electrodes and voltmeter. The direction of the electric field in proteoliposomes can be regulated by the addition of antenna complexes to the reconstitution mixture. The reaction center complex proteoliposomes generate an electric field of a direction opposite to that in chromatophores, whereas proteoliposomes containing reaction center complexes and a sufficient amount of antenna complexes produce a potential difference as in chromatophores. ATP and inorganic pyrophosphate, besides light, were shown to be usable as energy sources for electric generation in chromatophores associated with planar membrane.  相似文献

Bacteriorhodopsin and the nicotinic acetylcholine receptor were biotinylated and reconstituted in lipidic membranes on silicon supports by fusion with proteoliposomes. The presence and distribution of the proteins were studied by binding with streptavidin. Radio-labelled streptavidin was employed for quantifying the amounts of protein remaining in the supported membranes after storage in buffer. The proteins within the membranes remained bound to the surface for weeks. The biological activity of reconstituted unlabelled receptor upon storage showed stability in membranes formed on silicon supports and a reduced stability when formed onto lipid monolayer covered supports. Atomic force microscopy studies on preparations in liquid showed bilayer structures but also attached, partly fused liposomes and membrane particles. In air, the surface was smoother and contained less of liposomes and more of stacked lipid layers. Preparations labelled with streptavidin conjugated to colloidal gold and imaged in air showed the proteins individually distributed, with no protein-rich patches or protein aggregates.  相似文献

The electrochemical behavior of membranes prepared from commercial collodion preparations varies widely, some preparations showing very high, other ones very low electrochemical efficiency ("activity"). 2. The electrochemical activity of a collodion membrane depends entirely upon impurities of an acidic nature contained in the collodion used for casting the membrane. 3. The active acidic impurities are substantially due to partial oxidation which occurs in the manufacturing process. Sulfuric acid compounds; e.g., acid sulfuric acid esters play only a minor rôle, if any. 4. The electrochemical behavior of collodion membranes in solutions of strong electrolytes is decisively dependent upon the acidic groups built permanently into the collodion surfaces. Simple and composite membranes have been prepared from 2% collodion solutions containing different amounts of polystyrenesulfonic acid (PSSA). Various membrane parameters such as water content, electrolyte uptake, exchange capacity, and permselectivity of these membranes have been determined. The resistance and capacitance of simple membranes have been measured as functions of both external electrolyte concentration and internal fixed charge density. The impedance characteristics of composite membranes also have been determined and discussed in terms of the resistance and capacitance characteristics of simple membranes from which the composite structures have been formed.  相似文献

This work reports a technique for the stabilization after storage in air of a lipid film based biosensor for atenolol. Microporous filters composed of glass fibers (nominal pore sizes 0.7 and 1.0 microm) were used as supports for the formation and stabilization of these devices. The lipid film is formed on the filter by polymerization prior to its use. Methacrylic acid was the functional monomer, ethylene glycol dimethacrylate was the crosslinker and 2,2'-azobis-(2-methylpropionitrile) was the initiator. The method for preparation of stabilized lipid film biosensor is studied throughout this work. The response towards atenolol of these stabilized lipid membrane biosensor, for repetitive use, composed of phosphatidylcholine was compared with planar freely suspended bilayer lipid membranes (BLMs). The stabilized lipid membranes provided similar artificial ion gating events as BLMs in the form of transient signals and can function for repetitive uses after storage in air. This will allow the practical use of the techniques for chemical sensing based on lipid films and commercialization of these devices.  相似文献

Layered MoS2 prepared by liquid‐phase exfoliation has been blended with single‐walled carbon nanotubes (SWNTs) to form novel composite thin films for lithium battery applications. The films were formed by vacuum filtration of blended dispersions onto nitrocellulose membranes. The resulting composite films were transferred onto Cu foil electrodes via a facile filtration/wet transfer technique from nitrocellulose membranes. The morphology of the film was characterised by field emission scanning electron microscopy, which suggests that the MoS2‐SWNT composite film shows good adherence to the Cu foil substrate. The MoS2‐SWNT composite thin films show strong electrochemical performance at different charge‐discharge rates. The capacity of a MoS2‐SWNT composite film with thickness of 1 μm is approximately 992 mAh g?1 after 100 cycles. The morphology study showed that the MoS2‐SWNT thin film retains structural integrity after 100 cycles, while the MoS2 thin film without SWNTs displays significant cracking. In addition, the novel composite thin film preparation and transfer protocols developed in this study could be extended to the preparation of various layered‐material‐based composite films, with the potential for new device designs for energy applications.  相似文献

The photochemical cycle and the proton-pumping function of bacteriorhodopsin modified with lanthanum and formaldehyde has been studied. In both preparations, the M412 leads to BR570 transition time has been found to increase considerably. The deceleration of the photochemical cycle has been shown to be accompanied by inhibition of the millisecond phase of the photoelectrical response of bacteriorhodopsin membranes associated with phospholipid-impregnated collodion film. Photoelectrogenic activity measured with permeable ion probe in proteoliposomes was also inhibited. Effects of lanthanum were reversed by EDTA. Formation of M412 was slightly accelerated and the microsecond electrogenic phase was not affected by lanthanum and by formaldehyde. It is concluded that lanthanum, but not formaldehyde, can be used as a specific reversible inhibitor of the second half of the bacteriorhodopsin photocycle and of the associated H+ uptake on the cytoplasmic side of the halobacterial membrane. Possible mechanisms of these effects are discussed.  相似文献

The reponses of oxonol dyes to single and multiple single turnovers of the photosynthetic apparatus of photosynthetic bacteria have been studied, and compared with the responses of the endogenous carotenoid pigments. The absorbance changes of the oxonols can be conveniently measured at 587 nm, because this is an isosbestic point in the 'light-minus-dark' difference spectrum of the chromatophores. The oxonols appear to respond to the light-induced 'energization' by shifting their absorption maxima. In the presence of K+, valinomycin abolished and nigericin enhanced such shifts, suggesting that the dyes, respond to the light-induced membrane potential. Since the dyes are anions at neutral pH values, they probably distribute across the membrane in accordance with the potential, which is positive inside the chromatophores. The accumulation of dye, which is indicated by a decrease in the carotenoid bandshift, poises the dye-membrane equilibrium in favor of increased dye binding and this might be the cause of the spectral shift. The dye response has an apparent second-order rate constant of approx. 2 . 10(6) M-1 . s-1 and so is always slower than the carotenoid bandshift. Thus the dyes cannot be used to monitor membrane potential on submillisecond timescales. Nevertheless, on a timescale of seconds the logarithm of the absorbance change at 587 nm is linear with respect to the membrane potential calibrated with the carotenoid bandshift. This suggests that under appropriate conditions the dyes can be used with confidence as indicators of membrane potential in energy-transducing membranes that do not possess intrinsic probes of potential.  相似文献
